Abstract

1. A process for obtaining surface or membrane-associated antigens from whole cells or culture media in which they are grown, characterized in that it comprises: a) destroying and fixing said cells; (b) separating said cells from the medium; (c) extracting antigen from said cells or culture medium with an alkaline aqueous solution of ph regulated alkali metal salt having a ph of about 6.0 to about 11.0, wherein the concentration of said ph regulator and salt is at least 0.01 mole; d) concentrating said antigen; (e) acid precipitating said concentrated antigen with an aqueous acid or an acid buffer solution having a ph of from about 1.0 to about 6.0 wherein the concentration of said acid or regulator is at least 0, 01 molar; f) solubilizing said antigen precipitated with acid with an aqueous alkaline solution of regulated ph uqe has a ph of about 6.0 to about 11.0 where the concentration of said regulator is at least 0.01 molar and, if wishes to digest the antigen solubilized with a proteolytic enzyme for about 1/4 hr to about 4hr between about 0ºC and about 45º c; and stop the enzyme digestion of the antigen.
