・水溶性ポリマーは口の中で迅速に溶解/分散し、歯の表面上に、高濃度の生体活性ガラス、従ってヒドロキシカーボネートアパタイトを形成するために必要とされるイオンを供給する。
一般的に言えば、本発明は、1本以上の歯に付着しうるフィルムの形態での再石灰化組成物の提供に属し、歯の表面上に再石灰化成分の高く持続的な局所濃度を生じさせる簡便で目立たない方法を提供する。水和すると、フィルムは、再石灰化イオンを放出して、歯の表面上に安定した結晶性ヒドロキシカーボネートアパタイト層を形成し、歯の過敏性の長期的な低減をもたらす。フィルムはまた、齲蝕発達および歯周病に関連する口腔細菌の増殖を阻害する能力をも有する。
表面微小硬さ試験の結果を図1に示す。結果は、白斑病変の相対的な硬化として示す(すなわち、すべてのデータは病変化エナメル質の測定硬度に対して正規化されている)。本発明の16重量%NovaMinフィルムおよび7.5重量%NovaMin練り歯磨き(SootheRx、陽性対照)による処理は、人為的な白斑病変の有意な硬化をもたらした。対照BioFilmサンプル(陰性対照)およびMI Paste(競合製品、Recaldentを含有する)による処理は、病変の表面硬度を変化させなかった。
図2a-bは、ウシのエナメル質上に事前に形成された白斑病変の走査型電子顕微鏡写真(SEM)画像である。図2aは、病変形成によって生じた重度の表面損傷の低倍率画像である。図2bは、白斑の縁にそって、また、図2aに見られるエナメル質の「帯」内において無傷のままである、エナメル質の柱状構造の高倍率画像である。対照BioFilmサンプルで処理した表面の画像(図3a-b)およびMI Pasteで処理した表面の画像(図4a-b)は、同様の特徴を示す。
NovaMin粒子を含有するフィルムのin-vitro試験の結果および口腔細菌の増殖を阻害するそれらの能力を図10に詳述する。15%NovaMinを含有する吸収性ヒドロキシプロピルメチルセルロースポリマーのストリップから、1.0%および0.5%NovaMin水溶液を調製した。各溶液から、6セットの100%、50%、および25%の段階希釈液を調製し、5mlの分割量に約0.02mlのストレプトコッカス・ミュータンス(Streptococcus mutans) ATCC 25175、ストレプトコッカス・サングイヌス(Streptococcus sanguinus) ATCC 10556、および黄色ブドウ球菌(Staphylococcus aureus) NCTC 10418を接種した。陽性対照および陰性対照もまた調製した。すべての試験希釈液および対照を35℃で一晩培養し、その後トリプトンソイ寒天(TSA)プレート上に画線した。その後、寒天プレートを35℃で一晩培養し、増殖を調べた。完全な濃度(100%)で、1.0%および0.5%溶液は、3つの細菌種すべての増殖を完全に阻害した。溶液の希釈によって阻害効果が若干低下したが、これは対照と比較するとごくわずかなものであった。
本明細書に示した結果から、本発明の16%NovaMin粒子を含有する吸収性ポリマーストリップ(試験生成物1)が、エナメル質表面に形成した白斑病変を効果的に治療する能力を有することを示される。NovaMin含有BioFilmサンプルによる処理は、NovaMin含有練り歯磨き(試験生成物3‐比較対照)による処理と同様の効果を有し、このモデルでは、ポリマーのみの吸収性ストリップ(試験生成物2)およびRecaldent含有練り歯磨き(試験生成物4)よりも格段に優れていた。
Claims (20)
- 口腔内で使用するためのフィルムであって、前記フィルムが:
水溶性ポリマーフィルム形成剤;および
生体活性ガラス
を含有しており;前記フィルムが溶解または実質的に溶解する前に、最大約60分間、口腔内の少なくとも1本の歯に付着することができ、また前記フィルムが歯の再石灰化を行うことができることを特徴とするフィルム。 - 前記フィルムが、水和した際、歯の表面上に安定した結晶性ヒドロキシカーボネートアパタイト層を形成し得る再石灰化イオンを放出することができ、それにより歯の過敏症の長期的な減少をもたらすことを特徴とする、請求項1に記載の口腔内で使用するためのフィルム。
- 前記フィルムが、水和した際、ヒドロキシカーボネートアパタイト鉱物形成に最適な、約7.5〜9.5のpH範囲をもたらすことができる、請求項1に記載の口腔内で使用するためのフィルム。
- 前記水溶性ポリマーフィルム形成剤が、ヒドロキシプロピルセルロースのような食用の非イオン性セルロース誘導体である、請求項1に記載の口腔内で使用するためのフィルム。
- 前記生体活性ガラスが再石灰化組成物を放出し得る、請求項1に記載の口腔内で使用するためのフィルム。
- 前記生体活性ガラスが、その主成分としてケイ素の酸化物を有し、生理的液体と接触した際、成長組織と接着し得る無機ガラスまたはセラミック材料である、請求項1に記載の口腔内で使用するためのフィルム。
- 前記生体活性ガラスが、下記の酸化物および/またはイオン:すなわち、カルシウム(例えばCaO);リン(例えばP2O5);ケイ素(例えばSiO2)およびナトリウム(例えばNa2O)の任意の1つまたはその組合せを含有する、請求項1に記載の口腔内で使用するためのフィルム。
- 前記フィルムが、水溶性ポリマーフィルム形成剤をフィルムの重量にして約1〜90%、約20〜80%、約30〜70%、約40〜80%または約50〜60%の量で含有する、請求項1に記載の口腔内で使用するためのフィルム。
- 前記フィルムが、生体活性ガラスをフィルムの重量にして約1〜60%、約1〜30%、約2〜25%、約5〜20%、または約8〜16%の量で含有する、請求項1に記載の口腔内で使用するためのフィルム。
- 前記フィルムが、約40〜80重量%の二酸化ケイ素(SiO2)、約0〜35重量%の酸化ナトリウム(Na2O)、約4〜46重量%の酸化カルシウム(CaO)および約1〜15重量%の酸化リン(P2O5)を含有する、請求項1に記載の口腔内で使用するためのフィルム。
- 前記水溶性ポリマーフィルム形成剤が、約1,000〜1,500,000、約1,000〜500,000、約50,000〜500,000、約100,000〜200,000、約140,000、約1,000〜100,000、約1,000〜50,000、約1,000〜20,000または約10,000〜20,000の平均分子量範囲を有する、請求項1に記載の口腔内で使用するためのフィルム。
- 前記生体活性ガラスが:生体活性材料の粒子;繊維;および/またはプレートレットを含む、請求項1に記載の口腔内で使用するためのフィルム。
- 生体活性ガラスを形成している粒子が、約100ミクロン未満、約75ミクロン未満、約50ミクロン未満、約20ミクロン未満、約10ミクロン未満、または約5ミクロン未満の平均粒径を有する、請求項1に記載の口腔内で使用するためのフィルム。
- 前記水溶性ポリマーフィルム形成剤が、天然由来の化学修飾されたフィルム形成剤である、請求項1に記載の口腔内で使用するためのフィルム。
- 口腔内で使用するための、歯の再石灰化を行うことができるフィルムを形成する方法であって、前記方法が:
水溶性ポリマーフィルム形成剤を用意すること;
生体活性ガラスを用意すること;および
水溶性ポリマーフィルム形成剤と生体活性ガラスを混合すること
を含んでおり;水溶性ポリマーフィルム形成剤と生体活性ガラスとの混合物が、歯の再石灰化を行うことができるフィルムに形成され得ることを特徴とする方法。 - 前記生体活性ガラスが、水溶性ポリマーフィルム形成剤を含有する水性混合物に添加されることを特徴とする、請求項15に記載の口腔内で使用するための、歯の再石灰化を行うことができるフィルムを形成する方法。
- 水溶性ポリマーフィルム形成剤と生体活性ガラスとの混合物が、均一な分散液を調製するために高せん断ミキサーにかけられ、次に、担体紙上に被覆する前に脱気され、室温以上で風乾されることを特徴とする、請求項15に記載の口腔内で使用するための、歯の再石灰化を行うことができるフィルムを形成する方法。
- 前記フィルムが口腔内で膨潤または溶解し、約5〜10分間にわたって再石灰化組成物を放出することを特徴とする、請求項1に記載のフィルムの使用。
- 前記フィルムが歯列矯正器の周囲に置かれ、必要な場所でのヒドロキシカーボネートアパタイトの生成を助けるために必要とされる高濃度の再石灰化イオンを送達し得る、請求項1に記載のフィルムの使用。
- 口腔内で使用するためのフィルムであって、前記フィルムが:
水溶性ポリマーフィルム形成剤;および
生体活性ガラス
を含有しており;前記フィルムが最大約60分以内に溶解または少なくとも実質的に溶解することができ、また前記フィルムが歯の再石灰化を行うことができることを特徴とするフィルム。
