Claims (1)
- 【特許請求の範囲】 1.自律増殖ベクター(autonomously replicating vector)(2)に組込まれた 形で少なくとも1つのプロモータヌクレオチド配列(3)と毒素として作用する 融合たんぱくを暗号化する少なくとも1つのヌクレオチド配列(4)とを含み、 いくつかのクローニング部位を含む暗号ヌクレオチド配列(5)とたんぱく毒を 暗号化するヌクレオチド配列(6)とを融合することによって前記ヌクレオチド 配列(4)を得ることを特徴とするクローニング及び/または配列ベクター。 2.自律増殖ベクター(2)が組換えウィルスであることを特徴とする請求の範 囲第1項に記載のベクター。 3.自律増殖ベクター(2)が組換えプラスミドであることを特徴とする請求の 範囲第1項に記載のベクター。 4.自律増殖ベクター(2)が組換えpUCプラスミドであることを特徴とする 請求の範囲第3項に記載のベクター。 5.プロモータヌクレオチド配列(3)がLacオペロンプロモータから成るこ とを特徴とする請求の範囲第1項から第4項までのいずれかに記載のベクター。 6.たんぱく毒を暗号化するヌクレオチド配列(6)と融合するヌクレオチド配 列(5)の特定クローニング部位(unique cloning sites)がクローニングベクタ ーのヌクレオチド 配列の残り部分に存在しないことを特徴とする請求の範囲第1項から第5項まで のいずれかに記載のベクター。 7.たんぱく毒を暗号化するヌクレオチド配列(6)がたんぱくCcdBを暗号 化する野性型遺伝子のヌクレオチド配列のすべてまたは一部から成ることを特徴 とする請求の範囲第1項から第6項までのいずれかに記載のベクター。 8.たんぱく毒を暗号化するヌクレオチド配列(6)に制限酵素SmaIの切断 部位(cleavage site)が存在しないことを特徴とする請求の範囲第7項に記載の ベクター。 9.寄託No.LMBP2781に相当することを特徴とする請求の範囲第1項 から第8項までのいずれかに記載のベクター。 10.請求の範囲第1項から第9項までのいずれかに記載のクローニングベクタ ーで形質転換される原核細胞。 11.好ましくは染色体Iq及び高い形質転換効率を有し、融合たんぱくの毒性 に対する耐性を授ける突然変異を有し、及び/または融合たんぱくに対する抗毒 素であるたんぱくを暗号化する遺伝子を有することを特徴とする請求の範囲第1 項から第9項までのいずれかに記載のクローニングベクターのための原核宿主細 胞。 12.ジャイレースのサブユニットAを暗号化する遺伝子またはジャイレースの サブユニットBを暗号化する遺伝子中に突然 変異を有し、融合たんぱくに対する耐性を授け、及び/または融合たんぱくに対 する抗毒素であるたんぱくCcdAを暗号化する遺伝子を有することを特徴とす る請求の範囲第11項に記載のクローニングベクターのための原核宿主細胞。 13.ジャイレースのGyrAポリペプチドのアミノ酸配列中でアルギニン46 2をシステインに代える突然変異を有し、それによって融合たんぱくに対する耐 性を授けることを特徴とする請求の範囲第11項または第12項に記載の大腸菌 細胞。 14.LacIq突然変異を有することを特徴とする請求の範囲第11項から第 13項までのいずれかに記載の細胞。 15.No.LMGP−12601として寄託されていることを特徴とする請求 の範囲第11項から第14項までのいずれかに記載の細胞。 16.いくつかの特定クローニング部位を含むヌクレオチド配列(5)の両側に 位置する配列とハイブリッド化する10ないし30個のヌクレオチドの配列から 成ることを特徴とする請求の範囲第1項から第9項までのいずれかに記載のベク ターの断片。 17.組換えクローンの選択を目的とする請求の範囲第1項から第9項までのい ずれかに記載のクローニングベクターの利用。
