Abstract
L'invention se rapporte à un procédé de détection quantitative de traces d'un analyte dans un échantillon. Le procédé, dans le mode préféré de réalisation, consiste à produire un fragment Fab' d'une immunoglobuline marquée au niveau d'un groupe sulfhydryle réactif avec une couleur fluorescente; à combiner le fragment Fab' marqué avec un échantillon qui peut contenir l'analyte; à concentrer les éléments du mélange obtenu dans un champ électrique; à séparer le complexe marqué analyte/agent obtenu à partir de n'importe quel agent marqué non transformé à l'aide des procédés électrophorétiques capillaires; et à détecter le signal fluorescent du complexe marqué analyte/agent. L'invention se rapporte également à un procédé de production d'un fragment Fab' marqué qui consiste à dissocier une molécule d'immunoglobuline pour obtenir un fragment F(ab')2; à réduire les liaisons de dislufure du fragment F(ab')2 pour obtenir deux fragments Fab', chacun ayant au moins un groupe sulfhydryle réactif, libre; et à mélanger un fragment Fab' ayant au moins un grupe sulfhydryle libre avec une couleur libre pour former un fragment Fab' marqué. De préférence, avant l'étape finale de mélanger, des liaisons de disulfure intrabrins sont formées par l'oxydation dans chaque fragment Fab', produisant ainsi des fragments individuels Fab' ayant chacun un groupe sulfhydryle réactif unique. Le procédé de détection quantitative consiste également, de manière plus générale, à utiliser n'importe quel agent biospécifique pour former un complexe avec l'analyte cible.
