明 細 書 喉ケア剤 技術分野
本発明は、 唾液の分泌を促し、 喉を殺菌し、 洗浄し、 咽頭粘膜への細菌の感染 を防止し爽快に保つ喉ケア剤に関する。 背景技術 '
口腔内のケア剤として、 口腔乾燥症患者の唾液の分泌を、 キシリトールを含有 する可食組成物で促進し、 口腔内の症状を緩和するもの (特開平 3— 8 3 9 2 0 号公報) や、 シソ科植物等由来の口腔用消臭剤とァォギリ科植物等の唾液分泌促 進剤とを併用して口臭を抑制するもの (特開平 1 1一 1 8 0 8 3 9号公報) 等が 提案されている。
しかしながら、 これらは口腔内乾燥の治療又は口臭抑制効果の増強、 持続時間 の延長等を目的としたもので、 風邪症状群等の原因である、 喉に付着した病原菌 に対して防御や症状改善等を目的としたものではない。
また、 鎮咳去痰成分、 殺菌剤、 抗炎症剤等の喉に作用して喉の炎症やせきを防 止するための喉飴が数多く用いられている。 しかしながら、 これらの喉飴は、 飴 の中に配合したヒトにとつては外来性の成分により薬効を発揮させようとするも のであり、 その効果には限界がある。
本発明の目的は、 風邪ウィルス感染及び二次性細菌感染等から喉を防御する、 即ち、 喉の殺菌、 洗浄を行い、 生体の防御反応を賦活させて喉をより効率よくケ ァし、 また、 症状改善効果を有する喉ケア剤を提供することにある。 発明の開示 本発明者は、 唾液の生体防御機能に着目して検討してきたところ、 唾液には、 ムチンが多量含まれており、 このムチンは喉に悪影響を与える起炎菌を捕獲 (包 接) したり、 喉に付着した起炎菌を排除する効果があることを見出した。
更に本発明者は、 風邪ウィルスの増殖サイクルが口腔粘膜ではなく気道の粘膜 上皮のみであって (細胞工学、 第 1 9卷第 1号、 3 3〜3 8頁)、 更にその際ィ ンフルェンザ菌、 緑膿菌、 黄色ブドウ球菌等の病原菌が産生するプロテアーゼが 風邪ウィルスの増殖活性化を補強する (Modern Phys ici an第 1 8卷第 1 1号、 1 3 3 9〜 1 3 4 2頁) ことに着目し、 喉への病原菌の付着抑制、 除去を検討し たところ、 唾液が、 これら病原菌の喉への付着抑制、 除去作用を示し、 不快感を 除くことを見出した。
本発明は、 唾液分泌促進成分からなる喉ケア剤を提供するものである。 また、 本発明は、 更に多糖類を含有する喉ケア剤を提供するものである。 また、 本発明 は、 更に喉作用成分を含有する喉ケア剤を提供するものである。
また、 本発明は、 唾液分泌促進成分を、 摂取することを特徴とする喉ケア方法 を提供するものである。
また、 本発明は、 唾液分泌促進成分の喉ケア剤製造のための使用を提供するも のである。
更には、 これらの喉ケア剤をキャンディ又は飲料の形態としたものを提供する ものである。 図面の簡単な説明

本発明で用いる唾液分泌促進成分としては、 味覚刺激成分、 口内粘膜刺激成分、 唾液腺活性化成分、 物理的刺激成分等が挙げられる。 ここで、 唾液分泌促進成分 とは、 健常人が起床後から食物を口にしない状態で 3 0分以内に安静状態で測定 した唾液分泌速度に対する、 同じ安静状態で本唾液分泌促進成分を食した後、 味 覚刺激物質及び口内粘膜刺激物質の場合はその直後、 唾液腺活性物質の場合 は 6 0〜1 8 0分後、 物理的刺激成分の場合は直後に測定した唾液分泌速度の比 が 1 . 2以上、 好ましくは 2以上、 より好ましくは 5以上のものをいう。
味覚刺激成分としては、 有機酸類、 旨味物質等が挙げられる。
有機酸類としては、 クェン酸、 イソクェン酸、 リンゴ酸、 酢酸、 蓚酸、 マロン 酸、 コハク酸、 ダルタール酸、 マレイン酸、 フマル酸、 アコニット酸、 乳酸、 酒 石酸、 ピルビン酸、 ァスコルビン酸、 アルドン酸、 ゥロン酸等の有機酸の他に、 これらを含有する梅酢、 リンゴ酢及びレモン、 オレンジ、 ュズ、 夏ミカン等の柑 橘類のチップ、 粉末、 エキス等が挙げられる。
旨味物質としては、 アミノ酸 (塩)、 核酸、 ジペプチド、 トリペプチド、 オリ ゴペプチド等が挙げられ、 具体的には、 グルタミン酸ナトリウム、 イノシン酸、 グァニル酸等が挙げられ、 特にグルタミン酸ナトリウム、 イノシン酸が好ましい 口内粘膜刺激成分としては、 イソチオシァネート類、 アミド系辛味成分、 バニ リルケトン類等が挙げられ、 具体的には、 ァリルイソチオシァネート、 カブサイ シン、 ピペリン、 サンショオール、 ジンゲロール等が挙げられる。
唾液腺活性化成分としては、 副交感神経興奮剤、 コリンエステラーゼ阻害剤、 カルシウムイオン遊離放出剤等が挙げられ、 具体的にはピロカルビン、 ムスカリ ン、 アセチルコリン、 リアノジン、 カフェイン等、 また植物のコラ ·ァクミナタ, コラ '二ティダ、 ャポランジ、 シラカバ、 スイカズラ、 セィヨウニンジン、 ドク ダミ、 ニンニク、 ハイビスカス、 ホップ、 マタタビ、 リンデン、 ローズヒップ等 が挙げられる。
なお、 これらの成分のうち成分自体が揮発性を有するもの、 例えばァリルイソ チオシァネート、 サンショオール等の場合、 嗅覚を刺激し、 唾液分泌を促す作用 fcある。
物理的刺激成分としては、 発泡剤又は高圧充填した二酸化炭素ガス、 窒素ガス 若しくは不活性ガス等が挙げられる。
発泡剤としては、 有機酸と炭酸塩又は重炭酸塩との組み合せが好ましい。 ここ で有機酸としては、 クェン酸、 酒石酸、 ァスコルビン酸、 リンゴ酸、 フマル酸、 コハク酸、 マロン酸等が挙げられる。 炭酸塩又は重炭酸塩としては炭酸ナトリウ ム、 炭酸水素ナトリウム、 炭酸カリウム、 炭酸水素カリウム、 炭酸アンモニゥム、 セスキ炭酸ナトリウム等が挙げられ、 このうち炭酸ナトリウム、 炭酸水素ナトリ ゥム、 炭酸カリウムが特に好ましい。 また、 高圧充填とは、 水溶液にガスを溶解 させたもの及び固体中の空隙に封入したものをいう。
不活性ガスとしては、 アルゴン、 ネオン、 クリプトン等が挙げられる。
唾液分泌促進成分としては、 酒石酸、 ァリルイソチオシァネート、 カフェイン、 酒石酸と炭酸水素ナトリゥムの発泡剤、 クェン酸と炭酸水素ナトリゥムの発泡剤 等が好ましい。
これらの唾液分泌促進剤は、 2種以上併用してもよい。
本発明の喉ケア剤は、 唾液分泌促進成分を多糖類と併用すると、 更に効果が優 れたものとなり好ましい。
多糖類としては、 大豆食物繊維、 グァガム、 タラガム、 トラガントガム、 ァラ ピアガム、 キサンタンガム、 カラャガム、 ジエランガム、 コ一ン食物繊維、 アル ギン酸、 サクランポガム、 メスキッドガム、 ローカストビーンガム、 寒天、 リン 酸多糖が挙げられ、 大豆食物繊維、 グァガム、 タラガム、 口一カストビーンガム、 リン酸多糖が好ましい。 リン酸多糖としてはリン酸マンナンが特に好ましい。 また、 多糖類は 2種以上併用してもよく、 例えば、 リン酸マンナンと大豆食物 繊維、 大豆食物繊維とタラガム、 リン酸マンナンとタラガム等の組み合せが好ま しい。
唾液分泌促進剤と多糖類の含有重量比率は、 唾液分泌促進剤 多糖類 = 1 0 0 ノ 1〜 1 / 1 0 0、 特に 1 0 / 1〜 1 / 1 0であるのが好ましい。
本発明の喉ケア剤は、 唾液分泌促進成分を喉作用性成分と併用すると、 更に喉 の不快感の緩和、 喉の清涼感等が改善される。
喉作用性成分としては、 ハーブ、 鎮晐去痰剤、 キチン、 キトサン、 ハチミツ、 プロポリス又は唾液含有成分等が挙げられる。
ハーブは、 香草、 葉、 花弁又は茎が香味をもち、 食用、 薬用に使用される植物 であって、 具体的には、 スペアミント、 ペパーミント、 ウィンターグリーン、 サ ッサフラス、 丁子、 セージ、 ユーカリ、 マョナラ、 肉桂、 タイム、 ティーツリー、 紫蘇、 ェキナセァ、 甜茶、 ピーパーミー、 カリン、 高麗人参、 霊芝、 キンカン、 ダイコン、 ョモギ、 サポニン、 ビヮ、 ォォバコ、 ウイキヨゥ、 生姜、 イチヨウ、 サフラン、 ナツメ等及びこれらのチップ、 粉末、 汁、 抽出物等が挙げられる。 ま た、 メントール、 シネオール、 ァネトール、 カルボン等のこれらハーブ中の成分 を用いてもよい。 この場合メントール、 シネオ一ルが好ましい。
鎮晐去痰薬としては、 デキストロメトルファンフエノールフタリン塩、 塩酸メ チルエフェドリン、 グァヤコ一ルスルホン酸力リゥム、 グァイフェネシン、 クレ ゾ一ルスルホン酸カリウム、 塩化セチルピリジニゥム、 塩化デカリニゥム、 塩酸 クロルへキシジン、 ノス力ピンが特に好ましい。
唾液含有成分としては、 ムチン、 分泌型 I g A、 分泌型 I g M、 リゾチーム、 ラク卜フェリン、 ペルォキシダーゼ、 カリクレイン、 フコースリッチ糖タンパク、 フイブロネクチン、 プロリンリッチ糖タンパク、 一アミラーゼ、 スタセリン、 ヒスチジンリッチタンパク、 インベルタ一ゼ、 デキストラナーゼ、 グリコシダー ゼ、 エストロゲン、 プロゲスティン、 アンドロゲン、 コルチコステロイド、 チロ キシン、 メラトニン等が挙げられる。
これらの喉作用性成分は、 2種以上を併用してもよい。
喉作用性成分としては、 八一ブ、 ハーブ抽出物、 キトサン、 蜂蜜、 プロポリス、 シネオ一ル等が、 喉の清涼感及び爽快感の点で好ましい。
唾液分泌促進成分と喉作用性成分とを併用する場合、 本発明の喉ケア剤中に、 唾液分泌促進成分は、 0 . 0 5〜 2 5重量%、 好ましくは 1〜 1 5重量%、 喉作 用性成分は、 0 . 0 1〜5重量%、 好ましくは 0 . 5〜2重量%含有するのが好 ましい。
本発明の喉ケア剤は、 喉に長く残留させた方が、 喉作用性成分の持続性及び唾 液の分泌がより効果的に促進され、 シロップ、 ゼリー飲料、 清涼飲料等、 特に粘 性のある飲料としたものが効果的に優れる。
この場合咽下のしゃすさの点で、 2 0 °Cにおいて B型粘度計を用いて測定した 粘度が 2〜 3 0 0 O iPa- s, 好ましくは 5 0〜1 8 0 O mPa- s, 特に 1 5 0〜4 0 O mPa - s であるのが好ましい。 粘度を調整する増粘成分としては、 水飴、 蔗糖、 ぺクチン、 カラギーナン、 口一カストビーンガム、 グァガム、 ゼラチン等が挙げ られる。 この場合基剤として、 特に水を使用するのが好ましい。
また、 本発明の喉ケア剤は、 ドロップ、 飴等のキャンディの形態であるのが好 ましく、 砂糖及び/又は水飴を主原料とするもの、 特に高温 (1 4 0〜 1 8 0 °C) で煮つめたものが好ましいが、 糖アルコールを主原料としてノンシュガー あるいは砂糖不使用としてもよい。
本発明喉キャンディ中に発泡成分は略均一に含有していてもよいが、 発泡成分 含有部と非含有部とで構成され、 当該発泡成分含有部が先に溶解終了するように 処方されていてもよい。
発泡成分含有部と非含有部とを区分する手段としては、 例えば上層と下層、 上 層、 中層、 下層などのように積層構造としてもよい。 また、 内層とその周囲を覆 う外層を有する被覆型キャンディとしてもよい。 更にはブロック状、 柱状、 球状 等の部分を一部に含む構造の飴としてもよい。
発泡成分含有部と非含有部との割合は、 含有部:非含有部 (重量比) が 0 . 1 : 1 ~ 1 : 0 . 1、 特に 0 . 4 : 1〜 1 : 0 . 4であることが好ましい。 また、 発泡成分含有部と非含有部に分けた場合、 発泡成分含有部中の発泡成分の含有量 は、 発泡性の点から有機酸と炭酸塩又は重炭酸塩との合計で 1〜8 0重量%、 更 に 5〜6 0重量%、 特に 1 0〜4 0重量%が好ましい。 また発泡成分含有部と非 含有部に分けた場合、 前記喉作用性成分は、 その両方の部分に配合してもよいし、 いずれか一方にのみ配合してもよい。
本発明喉ケア用キヤンディの製造法は略均一構造の場合は常法により、 また積 層構造の場合には発泡成分含有部と非含有部とをそれぞれ調製後積層するのが好 ましい。 また、 被覆型キャンディの場合には、 発泡成分非含有部を調製後その上 に発泡成分含有部を被覆するのが好ましい。
本発明喉ケア用キャンディは、 前記の如く優れた発泡性と唾液分泌促進効果を 有するので好ましい。
本発明の喉ケア剤は、 唾液分泌促進剤としてクェン酸と炭酸水素ナトリゥム発 泡剤を用いた場合、 喉ケアキャンディとした場合、 成人 (体重 6 0 kg) が 1日当 り 1 0 g以上摂取するのが好ましい。 喉ケアキャンディとした場合は 5 g Z個の 喉ケアキャンディを 1日 2回以上摂取するのが病原菌の喉への付着を抑制し、 除 去をして喉を洗浄して爽快に保つ点から好ましい。
唾液による殺菌洗浄効果が優れており、 これに喉作用性成分を配合した場合に は、 唾液含有成分と他の成分との相互作用により更に優れた喉ケア効果が得られ る。 実施例
唾液による菌除去を次の試験により測定した。
試験 1 . 病原菌に対するトラップ効果 黄色ブドウ球菌、 インフルエンザ菌についてそれぞれ菌数を 108/mL の PB S菌溶液を作成する。 各菌液と等量の試験液すなわち唾液を 30分間混合する。 一方、 ヒトの咽頭上皮細胞を PBS溶液で洗浄し、 細胞数が 2. 5 X 10 mL となる PBS溶液を作成する。 次に、 試験液と混合した菌液 0. 02mL と細胞 液 0. 98mL を混合し 37 °Cで 30分間反応させる。 その後、 この液を 60 Or /min、 10分間遠心分離し、 P B S溶液での洗浄を 3回繰り返し、 プレート上 に散布、 グラム染色を行ない、 光学顕微鏡にて、 設定倍率での顕微鏡視野内の菌 数を測定し、 対照条件での菌数を 100 %とした場合の細胞への菌付着率を測定 する。 なお、 対照条件は、 試験液を PBS溶液とした。 その結果、 唾液のトラッ プ効果は黄色ブドウ球菌、 インフルエンザ菌についてそれぞれ 10%、 5%を示 した。
試験 2. 病原菌に対するコーティング効果
黄色ブドウ球菌、 インフルエンザ菌についてそれぞれ菌数を 108/mL の PB S菌溶液を作成する。 一方、 ヒトの咽頭上皮細胞を PBS溶液で洗浄し、 細胞数 が 2. 5X 10 VmL となるように P B S溶液で調製し、 この溶液に等量の試験 液すなわち唾液を混合、 反応させた後、 この混合液を 600r/min、 10分間遠 心分離し、 PBS溶液での洗浄を 3回繰り返し細胞液を得る。 次に各菌液 0. 0 2mL と細胞液 0. 98mL を混合し 37 °Cで 30分間反応させる。 その後、 この 液を 600rZmin、 10分間遠心分離し、 P B S溶液での洗浄を 3回繰り返し、 プレート上に散布、 グラム染色を行ない、 光学顕微鏡にて、 設定倍率での顕微鏡 視野内の菌数を測定し、 対照条件での菌数を 100%とした場合の細胞への菌付 着率を測定する。 なお、 対照条件は、 試験液を PBS溶液とした。 その結果、 唾 液のコーティング効果は黄色ブドウ球菌、 インフルエンザ菌についてそれぞれ 2 0 %、 10 %を示した。
試験 3. ムチンの病原菌に対するトラップ効果、 コ一ティング効果
ヒト由来のムチン溶液 (0. 001 %) を用いて、 これを試験液として試験 l .、 2 . を行なったところ、 トラップ効果は黄色ブドウ球菌、 インフルエンザ 菌についてそれぞれ 1 2 %、 5 %を示した。 コーティング効果は黄色ブドウ球菌、 インフルエンザ菌についてそれぞれ 2 2 %、 1 0 %を示した。
実施例 1
表 1に示す組成の喉ケア剤を調製した。

表 1に示す喉ケア剤を飲用したときの唾液分泌流量を次法に従って測定した。 唾液分泌流量の測定
1 0人の健康な成人に表 1の喉ケア剤 1 0 gを服用し、 本発明品 1、 2、 3、 4、 6及び比較品 1、 2は服用直後、 本発明品 5は服用 9 0分後に、 各人の口中 の唾液を脱脂綿でよく拭き取り、 その後 5分間に分泌する唾液の全量を吐き出さ せ、 その重量を唾液分泌流量とした。 1 0人の算術平均値をとり評価した。
図 1に結果を示すが、 本発明品 1〜6はいずれも唾液分泌流量が、 比較品に比 して 1 0倍以上の値を示し、 唾液の分泌が促進された。
実施例 2
表 1に示す本発明品 2〜 6及び比較品 2を、 風邪による喉の不快感を持つ 1 0 人の成人に 1 0 g服用し、 その直後から喉に爽快感が残っている時間を測定した 10人の算術平均値で評価した。
図 2に結果を示すが、 本発明品 2〜 6はいずれも喉の爽快感が長時間持続し、 粘度の高い本発明品 3は約 24分間持続した。
実施例 3
次の飴を製造した。
クェン酸 12. 5重量%
炭酸水素ナトリウム 12. 5
水飴 (固形分) 15
パラチニット 59. 5
メントール 0. 5
この飴 5 gを服用し、 飴がなくなるまでの唾液を全て吐出させ、 その重量から 飴の重量を差し引いた値を唾液分泌量とした。 成人 10人の算術平均で評価した。 本発明の飴の唾液分泌量は 52 gであった。
次いで、 次の試験方法で病原菌トラップ効果及び病原菌コーティング効果を測 定した。
試験方法:
1. 病原菌トラップ効果
黄色ブドウ球菌、 インフルエンザ菌についてそれぞれ菌数を 1 OVmL の PB
S菌溶液を作成する。 各菌液と等量の試験液を 30分間混合する。 一方、 ヒトの 咽頭上皮細胞を PBS溶液 (リン酸緩衝溶液) で洗浄し、 細胞数が 2. 5X 10 VmL となる PBS溶液を作成する。 次に、 試験液と混合した菌液 0. 02mL と 細胞液 0. 98mL を混合し 37°Cで 30分間反応させる。 この後、 この液を 6
0 Or/minで 10分間遠心分離し、 P B S溶液での洗浄を 3回繰り返し、 プレー ト上に散布、 グラム染色を行い、 光学顕微鏡にて、 設定倍率での顕微鏡視野内の 菌数を測定し、 対照条件での菌数を 100とした場合の細胞への菌付着率を測定 する。 なお、 対照条件は、 試験液を PBS溶液とした。 2. 病原菌コーティング効果
黄色ブドウ球菌、 インフルエンザ菌についてそれぞれ菌数を 108/mL の PB S菌溶液を作成する。 一方、 ヒトの咽頭上皮細胞を PBS溶液で洗浄し、 細胞数 が 2. 5 X 1 OVniL となるように PB S溶液で調製し、 この溶液に等量の試験 液を混合、 反応させた後、 この混合液を 60 Or/min で 10分間遠心分離し、 P BS溶液での洗浄を 3回繰り返し細胞液を得る。 次に各菌液 0. 02mL と細胞 液 0. 98niL を混合し、 37 °Cで 30分間反応させる。 その後、 この液を 60 Or/minで 10分間遠心分離し、 P B S溶液での洗浄を 3回繰り返し、 プレート 上に散布、 グラム染色を行い、 光学顕微鏡にて、 設定倍率での顕微鏡視野内の菌 数を測定し、 対照条件での菌数を 100とした場合の細胞への菌付着率を測定す る。 なお、 対照条件は、 試験液を PBS溶液とした。
本発明の飴をなめて分泌した唾液及び溶解した飴の混合液の黄色ブドウ球菌の トラップ効果は 10%、 コーティング効果は 20 %であった。 またインフルェン ザ菌のトラップは 5 %、 コーティング効果は 10%であった。
実施例 4
次の飴を製造した。

実施例 3と同様に測定した。 本発明の飴の唾液分泌量は 52 gであった。 一方、 水飴 20重量%、 パラチニット 79. 5重量%及びメントール 0. 5S量%を含 有する飴 (比較) の場合は、 唾液分泌量 5 gであった。
実施例 3と同様にして測定した本発明の飴をなめて分泌した唾液と飴の溶解し た液の混合液の病原菌トラップ効果は 0 %、 病原菌コーティング効果は 0 %であ つた。 一方、 飴 (比較) の場合は、 病原菌トラップ効果 100%、 病原菌コ一テ ィング効果は 100 %であった。
実施例 5
次の外層/内層の重量比が 7Z3の 2層タイプの飴を製造した。
外層:酒石酸 12. 5重量%
炭酸水素 12. 5
水飴 (固形分) 16
パラチニッ卜 59
内層:水飴 (固形分) 20重量%
パラチニッ卜 78. 5
大豆食物繊維
メントール 0. 5
実施例 3と同様に評価した。 唾液分泌量は 43 g、 病原菌トラップ効果は 0%. 病原菌コーティング効果は 0%であった。
実施例 6
次のシロップを製造した。
水飴 95. 5重量%
メントール
大豆食物繊維 1
グァガム 1
カフェイン 0. 5
ポリグリセリン脂肪酸エステル 1
シロップ 5 g服用後 90分後、 口中の唾液を脱脂綿でよく拭き取り、 その後 5 分間に分泌する唾液の全量を吐出させ、 その重量を唾液分泌量とした。 成人 10 人の算術平均で評価した。 唾液分泌量は 17 gであった。 また、 実施例 3と同様に評価した病原菌トラッ プ効果は 0 %、 病原菌コ一ティング効果は 0 %であった。
一方、 水飴 98重量%、 メントール 1重量%及びポリグリセリン脂肪酸エステ ル 1重量%を含有する飲料 (比較) の場合は、 唾液分泌量 2 g、 病原菌トラップ 効果 100%、 病原菌コーティング効果 100%であった。
実施例 7
次の飲料を製造した。
水 95. 5重量%
メントール 1
大豆食物繊維 1
グァガム 1
カフェイン 0. 5
ポリグリセリン脂肪酸エステル 1
実施例 6と同様に評価した唾液分泌量は 12 g、 実施例 3と同様に評価した病 原菌トラップ効果 0 %、 病原菌コーティング効果 0 %であった。
実施例 8
次のキャンディを製造した。
キャンディ (A):
砂糖 46 g、 水飴 29 gに水を 20 gを加え混合し、 150°Cまで煮詰め、 水 分を完全に蒸発させた後、 70°C程度まで冷却し酒石酸 12 g、 重曹 12 g、 メ ントール 0. 5 g、 シネオ一ル 0. 5 gを添加、 混合した後、 5 gを 20匪 φの 型に入れて成形、 冷却する。
キャンディ (B):
(1) 第 2層の作成
砂糖 36 g、 水飴 22 g、 水 7. 2 gを混ぜ、 150°Cで煮詰め、 水分を完全 に蒸発させ、 70°C程度まで冷却し、 メントール 1 gのシネオ一ル 1 gを添加混 合する。
(2) 第 1層の作成と第 2層との接合
第 1層は、 キャンディ (A) で、 メントール、 シネオールを加えず、 混合、 煮 めを行い、 そこから 3. 5 gを取り、 第 2層 1. 5 gとともに 2 Οπυηφの型 入れて成形する。 その際、 第 2層 1. 5 gが第 1層の中心に位置するように封入 して成形する。
キャンディ (C):
砂糖 46 g、 水飴 30 gに水を 15 gを加え混合し、 150°Cまで煮詰め、 水 分を完全に蒸発させた後、 7 O :程度まで冷却し酒石酸 12 g、 重曹 12 gを添 加、 混合した後、 5 gを 20mm φの型に入れて成形、 冷却する。
比較キャンディ 1
砂糖 60 g、 水飴 39 g、 水 20 gを混ぜ、 150°Cで煮詰め、 水分を完全に 蒸発させ、 70°C程度まで冷却しメン] ^一ル 0. 5 g、 シネオール 0. 5 gを添 加、 混合した後; 5 gを 20匪 ψの型に入れて成形、 冷却する。
キャンディ (A) 〜 (C) 及び比較キャンディ 1の飴を用いて、 唾液分泌量、 ムチン量、 唾液中の殺菌成分量 ( I g A及びリゾチーム)、 及び口内唾液の pHを 次の如くして測定した。
(1) 唾液分泌量
成人 10人に対して、 試験前に口中の唾液を脱脂綿でよくふきとり、 キャンデ ィ (A) 〜 (C) 及び比較キャンディ 1を 5 gなめさせ、 分泌する唾液をすベて 吐き出させ、 その重量を測定した。'その重量から発泡量を考慮したサンプル重量 を減じた後、 10人に対する平均値を唾液分泌量とした。
唾液分泌量 = {吐き出させた重さ一 (キャンディの重さ一発生する C02計算重
(2) ムチン量
(1) で採取した唾液を用いて Blyscan Assay法により定量を行った。 ( 3 ) 唾液中の I g A及びリゾチーム量
( 1 ) で採取した唾液を用いて、 分泌型 I gAは、 Enzyme- 1 inked immunosorbent Assay (ELISA)法により、 リツチームは、 Micrococcus lysodeikt icus cells を用いた混濁度測定法により定量を行った。
(4) 口内唾液の pH
(1) で採取した唾液に対して半導体センサーを用いた pH メーターで測定し た。
得られた結果を図 3〜図 6に示す。 ,
図 3及び 4が示すように、 発泡成分あるいは酸を含むキャンディ (キャンディ (A) 〜 (C)) は、 酒石酸を含まない比較キャンディ 1に比べて唾液量、 ムチ ン量が多いのが分かる。 よって、 本発明のキャンディ (A) 〜 (C) は、 口内唾 液を健康な pH に保ちつつ、 多くの唾液量、 ムチン量を出す能力があることがわ かる。
また、 図 5が示すように、 発泡成分を含有した本発明のキャンディ (A) 〜 (C) は、 比較キャンディ 1に比べて分泌型 I gA、 リゾチームの分泌量が多く、 殺菌力が高い唾液が出ているのが分かる。
口内唾液 pH を低下させることなく、 唾液量、 ムチン量、 I gA量及びリゾチ 一ム量を増加させるのは、 本発明のキャンディ (A) 〜 (C) だけであることが わかる。
喉に不快感を感じるパネラー 20人に実施例、 比較例の飴を充分時間間隔をお いて各 1個、 かまずにゆっくりと口中でなめてもらい、 喉への効果感を評価した。 その結果を表 2に示す。 表 2

表 2から本発明のキャンディ (A)、 (B ) は、 比較キャンディ 1に比べて、 喉 への効果感 (不快感除去) が高い。
実施例 8 飲料
2剤を等量混合して飲用する次の飲料を製造した。

発泡により唾液の分泌が促進され、 爽快感が持続した 産業上の利用可能性
本発明の喉ケア剤は、 唾液の分泌を促進し唾液中の殺菌成分であるムチンが大 量利用に分泌促進され、 病原菌の喉への付着を抑制し、 除去をして喉を洗浄し爽 快に保つ。

Claims

請求の範囲
1 . 唾液分泌促進成分からなる喉ケア剤。
2 . 唾液分泌促進成分が、 味覚刺激成分、 口内粘膜刺激成分、 唾液腺活性化成分 又は物理的刺激成分である請求項 1記載の喉ケア剤。
3 . 味覚刺激成分が、 クェン酸、 イソクェン酸、 リンゴ酸、 酢酸、 蓚酸、 マロン 酸、 コハク酸、 ダルタール酸、 マレイン酸、 フマル酸、 アコニット酸、 乳酸、 酒 石酸、 ピルビン酸、 ァスコルビン酸、 アルドン酸、 ゥロン酸、 グルタミン酸ナト リゥム又はィノシン酸である請求項 2記載の喉ケア剤。
4. 口内粘膜刺激成分が、 ァリルイソチオシァネート、 カブサイシン、 ピペリン、 サンショオール又はジンゲロールである請求項 2記載の喉ケア剤。
5 . 唾液腺活性化成分が、 ピロカルビン、 ムスカリン、 アセチルコリン、 カフェ イン、 コラ 'ァクミナ夕、 コラ '二ティダ、 ャポランジ、 シラカバ、 スイカズラ、 セィヨウニンジン、 ドクダミ、 ニンニク、 ハイビスカス、 ホップ、 マタタビ、 リ ンデン又は口一ズヒップである請求項 2記載の喉ケア剤。
6 . 物理的刺激成分が、 発泡剤又は高圧充填した二酸化炭素ガス、 窒素ガスもし くは不活性ガスである請求項 2記載の喉ケア剤。
7 . 更に多糖類を含有する請求項 1〜 6のいずれか 1項記載の喉ケア剤。
8 . 多糖類が、 大豆食物繊維、 グァガム、 タラガム、 口一カストビーンガム又は リン酸多糖である請求項 7記載の喉ケア剤。
9 . 更に、 喉作用成分を含有する請求項 1〜8のいずれか 1項記載の喉ケア剤。
10. 喉作用成分が、 ハーブ、 鎮晐去痰剤、 キチン、 キトサン、 ハチミツ、 プロボ リス、 唾液含有成分、 メントール又はシネオ一ルである請求項 9記載の喉ケア剤。
11. キャンディの形態をした請求項 1〜1 0のいずれか 1項記載の喉ケア剤。
12. 飲料の形態をした請求項 1〜1 0のいずれか 1項記載の喉ケア剤。
13. 粘度が、 2〜3 0 0 O niPa ' sである請求項 1 2記載の喉ケア剤。
4. 唾液分泌促進成分を摂取することを特徴とする喉ケア方法。5. 唾液分泌促進成分の喉ケア剤製造のための使用。
