Abstract

Die vorliegende Erfindung betrifft ein Verfahren zum Screenen einer Sammlung von Nukleinsäuremolekülen auf eine gewünschte Eigenschaft der Nukleinsäure oder eines davon kodierten (Poly)peptids, umfassend die Schritte (a) automatisiertes Picken einer Sammlung von Zellen, die die Sammlung von Nukleinsäuremolekülen enthalten, mittels eines ersten Roboters; (b) automatisierte Lyse der Zellenmittels eines zweiten Roboters; (c) automatisierte Abtrennung der Zell-DNA vom Zelldebris mittels des zweiten Roboters; (d) optional automatisierte Abtrennung von Endotoxinen von der DNA mittels des zweiten Roboters sofern die Zellen Bakterien sind; (e) automatisierte Transfektion von Zellen mit der in Schritt (c) oder, sofern die Zellen Bakterien sind, in Schritt (d) erhaltenen DNA mittels eines dritten Roboters; und (f) automatisiertes Screenen auf die gewünschte Eigenschaft mittels eines vierten Roboters. Fernerbetrifft die Erfindung Verfahren zur Verbesserung der Bindungseigenschaften des (Poly)peptids, das durch die in dem erfindungsgemässen Screeningverfahren identifiziert oder von der identifizierten oder isolierten DNA kodiert wird, sowie ein Verfahren zur Herstellung eines Arzneimittels auf der Basis von mit dem erfindungsgemässen Verfahren erhältlichen (Poly)peptiden und ferner die Formulierung des erhaltenen Stoffes mit einem pharmazeutisch verträglichen Träger oder Verdünnungsmittel.
