明 細 書 藻類由来プロモーター、 イントロンおょぴターミネータ一 技術分野
本発明は、 遺伝子組換え技術を用いた海藻の育種や海藻の細胞を用いた工業生 産に用いるための遺伝子発現用のプロモーター、 ベクターに関する。 背景技術
従来、 産業上有用な海藻を得るためには、 海藻の収集や選抜に頼ってきた。 ま た、 交配育種法によって海藻をさらに有用なものに改変する試みも行われている 力 交配選抜するには、 その培養、 栽培が長期におよび、 また非常に手間がかか り、 十分な成果は挙げられていない。
陸上植物においては、 遺伝子組換えによる植物の改変が頻繁に行われ、 成果が あがってきている。 しかしながら、 海洋の大型藻類においては、 遺伝子組換えに よる改変はおろ力 \ 遺伝子を導入し発現させることすら成功した例は知られてな い。
生体内で遺伝子を発現させる場合、 その宿主に適したプロモーターが必須であ る。 即ち、 藻類で遺伝子を発現させる場合、 藻類で働くプロモーターが必須であ る。 しかしながら、 現在まで、 藻類に由来するプロモーターは全く知られておら ず、 藻類における遺伝子の発現は不可能であつた。 発明の目的
本発明の目的は、 上記従来技術を鑑みて行われたものであり、 藻類において外 来有用遺伝子を効率よく発現させ、 藻類の遺伝子工学的育種に有用なプロモータ 一、 イントロン、 ターミネータ一を提供するものである。 発明の概要
本発明者らは、 鋭意研究した結果、 広く食用に供されているスサビノリの遺伝 子を角军析し、 2種類のェロンゲーシヨンファクター (e 1 o n g a t i o n f a c t o r ; EF) 遺伝子が生活環の全てのステージにおいて高努現しているこ とを見出し、 該遺伝子の 5, 上流領域を単離し、 プロモーター活性を有すること を見出した。 さらに、 本発明者らは、 現在まで未知であった 1種類の EF遺伝子 のゲノムの配列をも明らかにし、 該配列中にイントロン配列、 ターミネータ一配 列を見出し、 これらの配列が導入遺伝子の発現効率を上昇させる活性を有するこ とを明らかにし、 本発明を完成させた。
即ち、 本発明を概説すれば、 本発明の第 1の発明は、 下記 (a) 〜 (c) 力 選択される DNAであって、 かつ、 藻類においてプロモーター活性を示すことを 特敫とする、 単離された DNAに関する:
( a ) 配列表の配列番号 9に示される塩基配列を有する DNAまたはその一部、
(b) (a) の DNAまたはその相補鎖と厳密な条件下でハイブリダィズし得る DNA、
(c) (a) の DNAに、 1以上の塩基の置換、 欠失、 挿入、 付加を有する DN A。
本発明の第 2の発明は、 下記 (d) 〜 (f ) に記載の DNAから選択され、 か つ、 藻類における遺伝子の転写効率を上昇させる活性を示すことを特徴とする、 単離された DN Aに関する:
(d) 配列表の配列番号 10に示される塩基配列を有する DNAまたはその一部、 (e) (d) の DNAまたはその相補鎖と厳密な条件下でハイブリダィズし得る

(f ) (d) の DNAに、 1以上の塩基の置換、 欠失、 挿入、 付加を有する DN A。
本発明の第 3の発明は、 本発明の第 1の発明の単離された D N Aの下流に隣接 して、 もしくは:!〜 10000塩基の DN A断片を介した位置に、 本発明の第 2 の発明の DNAを有し、 かつ、 藻類においてプロモーター活性を示すことを特徴 とする、 単離された DNAに関する。 本発明の第 3の発明において、 単離された D N Aとしては、 配列表の配列番号 3の 87— 1367番目の塩基配列が例示さ れる。 本発明の第 4の発明は、 下記 (g) 〜 (i) に記載の DNAから選択され、 か つ、 藻類においてターミネータ一活性を示すことを特徴とする、 単離された DN Aに関する :
( g ) 配列表の配列番号 1 1に示される塩基配列を有する D N Aまたはその一部、 (h) (g) の DNAまたはその相補鎖と厳密な条件下でハイブリダィズし得る

本発明の第 5の発明は、 本発明の第 1〜 4の発明のいずれかの DN Aに所望の 遺伝子が発現可能な状態で連結されていることを特徴とする、 組換え DNAに関 する。 本発明の第 5の発明において、 所望の遺伝子としては、 例えばタンパク質 をコードする核酸、 アンチセンス核酸をコードする核酸、 デコイをコードする核 酸おょぴリポザィムをコ一ドする核酸が例示される。
本発明の第 6の発明は、 本発明の第 1〜5の発明の DN Aを含有するベクター に関する。 本発明の第 6の発明において、 ベクターとしては、 プラスミドベクタ 一またはウィルスべクターが例示される。
本発明の第 7の発明は、 本発明の第 5の発明の DNAが導入された、 もしくは 本発明の第 6の発明のベクターで形質転換された藻類に関する。
本発明の第 8の発明は、 下記の (j ) 〜 (1) に記載の DNAから選択され、 力つ、 ェロンゲーションファクター (e l o n g a t i o n f a c t o r) 活 性を有するポリペプチドをコードすることを特徴とする、 単離された D N Aに関 する:
( j ) 配列表の配列番号 1に示される塩基配列を有する DNAまたはその一部、 (k) ( j ) の DNAまたはその相補鎖と厳密な条件下でハイブリダィズし得る DNA、

(m) 配列表の配列番号 2に示される塩基配列を有する D NAまたはその一部、 ( n ) (m) の D N Aまたはその相補鎮と厳密な条件下でハイプリダイズし得る D NA、
( o ) (m) の D NAに、 1以上の塩基の置換、 欠失、 揷入、 付加を有する D N A。
本発明のプロモータ一活性を示す D N A、 転写効率を上昇させる活性を示す D NA、 および Zまたはターミネータ一活性を示す D NAを用いることにより、 海 藻の遺伝子組換え用のベクターが提供され、 さらには、 海藻の遺伝子組換えによ つて、 海藻の育種の道を切り開くと同時に、 その結果、 新規な産業上有用な海藻 を提供することができる。
また本発明は、 海藻由来のェロンゲーションファクター活性を有するポリぺプ チドの遺伝子工学的製造を可能とするものである。 図面の簡単な説明

以下に本発明について詳細に説明する。
本明細書でいう 「プロモーター」 とは、 遺伝子の転写、 即ち D NAを錶型とし た mR NAの合成を開始させる機能を有する D NAを意味する。 本発明のプロモ 一ターは前記機能に限定されるものではなく、 転写開始点 (+ 1 ) から 2 0〜3 0塩基対上流にあって、 R NAポリメラーゼに正確な位置から転写を開始させる 機能を担う T A T Aボックスまたは T A T Aボックス類似の領域や発現調節のた めに必要な他の領域を含んで 、ても良!/、。
本明細書で言う 「プロモーター活性」 とは、 プロモーターの下流に発現可能な 状態で所望の遺伝子を連結し、 宿主 (藻類) に導入した際、 宿主内または宿主外 において所望の遺伝子の遺伝子産物を生産する能力および機能を有することを示 す。
一般的に、 プロモーターの下流に、 定量が容易に行なえるタンパク質をコード する遺伝子 (レポーター遺伝子) を発現可能な状態で連結させ、 宿主に導入し、 これらタンパク質の発現量を測定することでプロモーターの有無や強弱をプロモ 一ターの活 1"生として表わす。 このように、 プロモーターの下流に発現可能な状態 で所望の遺伝子の発現が確認された場合、 そのプロモーターは導入した宿主にお いて活性を有することになる。
本明細書で言う 「イントロン」 とは、 遺伝子またはその転写物の内部にあって、 その遺伝子から作られる最終 RNA産物に含まれない配列を言う。
イントロンはァミノ酸配列情報を持たなレ、塩基配列であるが、 外来遺伝子を宿 主に導入する際、 イントロン配列を含む導入遺伝子を用いることにより、 転写効 率が高まり、 導入遺伝子にコードされた目的のタンパク質が高生産される例があ る。 例えば、 高等植物において知られている 〔タナカ A. (T a n a k a A. ) ら;ヌクレイック ァシッズ リサーチ (Nu c l e i c Ac i d s Re s. ) 、 第 18巻、 第 23号、 第 6767〜 6770頁 (1990年) 〕 。 転写効率は、 例えば転写された R N A量をノザンハイブリダイゼーション法、 RT-PC R法等の公知の方法で測定することで調べることが出来る。
本発明で言う 「ターミネータ一」 とは、 RNAの転写を終結させる活性を有す るものを指す。 ターミネータ一は、 例えば転写された RNAのサイズをノザンハ イブリダイゼーション法等の公知の方法で測定することでその存在を調べること が出来る。
本発明で言う 「藻類」 には特に限定はなく、 例えばスサビノリ等の紅藻、 コン プ、 ワカメ、 ヒジキ等の褐藻、 ァォノリ等の緑藻等、 任意の真核生物に属する藻 類が挙げられる。 本発明で言う 「所望の遺伝子」 には特に限定はなく、 例えば藻類において発現 可能なタンパク質をコードする核酸、 アンチセンス RNAをコードする核酸、 デ コィをコードする核酸、 リボザィムをコードする核酸等が挙げられる。
また、 本発明に開示されたプロモーター活性を有する DNA、 転写効率を上昇 させる活性を有する DNA、 ターミネータ一活性を有する DNAと本来は機能的 に連結されていない 「異種の遺伝子」 を本発明に使用することができる。
藻類において発現可能なタンパク質をコードする核酸としては、 例えば藻類由 来のものが挙げられるが、 本発明においてはこれに限定にされるものではなく、 藻類において発現可能であれば、 細菌類;酵母類;放線菌類;糸状菌類;子嚢菌 類;担子菌類等の微生物由来、 植物由来、 動物由来、 またはウィルス由来の核酸 も、 本明細書で言う所望の遺伝子に含まれる。 前記タンパク質をコードする核酸 としては、 酵素、 レセプター、 サイト力イン類、 成長因子、 遺伝子発現やその制 御に関連する因子類、 構造タンパク質等をコードする核酸が挙げられるが、 これ らに限定されるものではなレ、。
本発明のプロモーター活性を有する DNA (以下、 プロモーターと記載する) は、 藻類においてプロモーター活性を有するものであり、 例えば、 スサビノリ
(P o r p hy r a y e z o e n s i s Ue d a) 由来 EF 1— a l p h a A 2遺伝子のプロモーター、 例えば、 配列表の配列番号 3に示される塩基配列に おいて、 87〜 859番目の塩基配列、 すなわち配列表の配列番号 9に記載の塩 基配列を有する DNAまたはその一部を含む DNAが挙げられる。 即ち、 本発明 のプロモーターは、 藻類においてプロモーター活性を有する範囲において、 配列 表の配列番号 9に示される塩基配列中の 、かなる D N Aであってもよい。
また、 これらの DNAまたはその相補鎖と厳密な条件下でハイブリダイス、する ことができる DNAであって、 藻類においてプロモーター活 1"生を有するものも、 本発明のプロモーターに含まれる。 ハイプリダイゼーシヨンは公知の方法、 例え ば、 1989年、 コールド ·スプリング ·ハーバー ·ラボラトリ一発行、 T. マ ニァテイス (T. Ma n i a t i s) ら編集、 モレキュラー ' クロー-ング: ァ ·ラボラトリー 'マニュアル第 2版 (Mo l e c u l a r C l o n i n g : A La b o r a t o r y Ma nu a l , 2n d e d. ) に記載の方法によ り実施することが出来る。 上記の厳密な条件としては、 例えば、 6 X S SC
(1 X S S Cの組成: 15 OmM 塩化ナトリウム、 15 mM クェン酸 3ナ トリウム二水和物、 pH7. 0) 、 0. 5%SDS、 5 Xデンハルト (De n h a r d t' s) 、 10 Omg/m 1 二シン精子 DNAを含む溶液中、 プロ一 ブとともに 65°Cで一晩保温するという条件が挙げられる。
さらに、 これらの DN Aに 1以上の塩基の置換、 欠失、 揷入、 付加を有する D
NAであって、 かつ、 藻類においてプロモーター活性を有する DNAも本発明の プロモーターに含まれる。
本発明のプロモータ一の下流に所望の遺伝子を発現可能な状態で連結した組換 え DNAを藻類に導入することにより、 藻類において所望の遺伝子を発現させる ことができる。 このことは、 藻類の遺伝子工学的育種、 藻類を宿主とした有用物 質の生産等を可能とするものである。
本発明の藻類における転写効率を上昇させる活性を有する D N Aは、 例えば、 スサピノリ由来 E F 1 - a 1 p h a A 2遺伝子のィントロン、 例えば、 配列表 の配列番号 3に示される塩基配列にお!/、て、 878〜 1331番目の塩基配列、 すなわち配列表の配列番号 10の塩基配列を有する D N Aまたはその一部を含む ものが挙げられる。 即ち、 前記の本発明の DN Aは、 藻類において転写効率を上 昇させる活' I·生を有する範囲において、 配列表の配列番号 10に示される塩基配列 中のいかなる DN Aであってもよい。
また、 これらの DNA断片またはその相補鎖と厳密な条件下でハイプリダイズ することができる DNAであって、 藻類における転写効率を上昇させる活性を有 するものも、 本発明の DNAに含まれる。 ハイブリダィゼーシヨンの条件は、 例 えば、 前記のものが挙げられる。
さらに、 これらの DNAに 1以上の塩基の置換、 欠失、 揷入、 付加を有する D NAであって、 かつ、 藻類における転写効率を上昇させる活性を示す DNAも本 発明の転写効率を上昇させる活性を有する DN Aに含まれる。
上記した本発明のプロモータ一活性を有する D N Aと所望の遺伝子を発現可能 な状態で連結した組換え DN Aに、 さらに本発明の藻類における転写効率を上昇 させる活性を有する DNAを適当な位置に揷入することにより、 転写効率を上昇 させることが出来る。 ここで、 適当な位置としては、 例えばプロモーター活性を 有する DNAの下流に,接する、 もしくは 1〜10000塩基の DNA断片を介 した位置が例示される。
本発明のターミネータ一活性を有する DNA (以下、 ターミネータ一と記載す る) は、 藻類においてターミネータ一活性を有するものであり、 例えば、 スサビ ノリ由来 E F 1 - a 1 p h a A 2遺伝子のターミネータ一、例えば、 配列表の 配列番号 3に示される塩基配列において、 2718〜 3764番目の塩基配列、 すなわち配列表の配列番号 1 1の塩基配列を有する DN Aまたはその一部を含む ものが挙げられる。 即ち、 本発明の DNAは、 配列表の配列番号 1 1に示される 塩基配列中のいかなる DNAであってもよい。
また、 これらの DNA断片またはその相補鎖と厳密な条件下でハイプリダイズ することができる DNAであって、 藻類においてターミネーター活 1·生を有するも のも、 本発明のターミネータ一に含まれる。 ハイブリダィゼーシヨンの条件は、 例えば、 前記のものが挙げられる。
さらに、 これらの DN Aに 1以上の塩基の置換、 欠失、 挿入、 付加を有する D
NAであって、 かつ、 藻類においてターミネータ一活性を有する DNAも本発明 のターミネータ一に含まれる。
上記した本発明のプロモータ一活性を有する D N Aと所望の遺伝子を発現可能 な状態で連結した組換え DN Aの下流に、 さらに本発明のターミネータ一活性を 有する断片を連結することにより、 所望の遺伝子の発現効率を上昇させることが 出来る。 また、 所望により、 さらに本発明の藻類における遺伝子の転写効率を上 昇させる活性を示す DNAを導入することにより、 さらに所望の遺伝子の発現効 率を上昇させることできる。
本発明のェロンゲーションファクター (e l o n g a t i on f a c t o r) 活性を有するポリペプチドをコードする DNA (以下、 EF遺伝子と記載す る) としては、 例えば、 配列表の配列番号 1に示される塩基配列を有する DNA またはその一部、 配列表の配列番号 2に示される塩基配列を有する D N Aまたは その一部、 配列表の配列番号 3に示される塩基配列を有する DN Aまたはその一 部、 あるいは配列表の配列番号 8に示されるァミノ酸配列を有するポリぺプチド 又はその一部をコードする DN Aが挙げられる。 好適な本発明の E F遺伝子の例 としては、 配列表の配列番号 1に示される塩基配列の 79〜: 1425番目の塩基 配列を有する DNA、 配列表の配列番号 2に示される塩基配列の 65〜1401 番目の塩基配列を有する DN Aが挙げられる。 また、 配列表の配列番号 3に示さ れる塩基配列の 1368〜2714番目の塩基配列を有する DNAが挙げられる。 さらに、 配列表の配列番号 8に示されるアミノ酸配列中のいかなるポリべプチド をコードする DNAであっても良い。
また、 これらの D N A断片またはその相補鎖と厳密な条件下でハイプリダイズ することができる DNAであって、 EF活性を有するポリペプチドをコードする ものも、 本発明の DNAに含まれる。 ハイプリダイゼーシヨンの条件は、 例えば、 前記のものが挙げられる。
さらに、 これらの DN Aに 1以上の塩基の置換、 欠失、 揷入、 付加を有する D NAであって、 かつ、 EF活性を有するポリペプチドをコードするものも本発明 の EF活性を有するポリペプチドをコードする DNAに含まれる。
これらの D N Aまたはその相補鎖と厳密な条件下でハイプリダイズすることが できる DNA、 またはこれらの DNAに 1以上の塩基の置換、 欠失、 揷入、 付加 を有する DNAとしては、 上記した DNAと例えば 90%以上、 好ましくは 9 5%以上、 さらに好ましくは 97%を超える相同性を有する DNAが挙げられる。 なお EF活性は、 例えば、 シャラタ V. F. (Sh a l a k V.
F. ) ら;ゥクラインスキー ビォヒミチュスキー ジュルナル (Uk r. B i o kh im. Z h. ) 、 第 69巻、 第 104〜: L 09頁 (1997年) に記 載の方法で測定することができる。
本発明のプロモーターと、 藻類における転写効率を上昇させる活性を有する D NA、 あるいはターミネータ一活性を有する DNAをそれぞれ単独でもしくは組 合わせて所望の遺伝子を発現可能な状態で連結した組換え DN Aは、 ベクターを 介してあるいはベクターを介さずに藻類に導入することができる。 ベクターとし ては、 プラスミドベクターまたはウィルスベクターが好適に使用される。
DNAが導入される藻類の形態に特に限定はなく、 糸状体、 胞子、 葉状体、 培 養細胞、 プロトプラスト等、 任意の形態の藻類を用いることが出来る。 藻類への DN Aの導入方法に特に限定はなく、 公知の方法、 例えばパーテイク ルガンによる導入方法が使用される。
本発明のプロモーターは、 例えば、 スサビノリより、 以下のようにして単離す ることが出来る。
スサビノリ (P o r p hy r a y e z o e n s i s Ue d a) は、 紅藻植 物門 (Rh o d o p h y t a) 紅藻綱 (Rh o d o p h y c e a e) 原始紅藻亜 糸岡 (P r o t o f l o r i d e o p h y c i d a e) ヮシケノリ白 (B a n g i a l e s) ゥシケノリ科 (B a n g i a c e a e) に属する海藻であり、 北海道 西部、 南部、 本州北部、 および朝鮮半島などに自生している。
このスサビノリは広く食用に供されている海藻であり、 盛んに養殖されており、 広く市販されている。 本発明の目的には、 海中で自生しているものを採取したも の、 養殖されているもの、 あるいは市販されているもののいずれも使用すること が出来る。 また、 公知の方法 〔クヮノ K. (Kuwa n o K) . ら;プラン ト サイエンス (P l a n t S c i . ) 、 第 1 1 6巻、 第 1集、 第 1 1 7〜1 24頁 (1 996年) 〕 によって株ィ匕されたものも同様に本発明の目的に使用さ れる。
<プロモータ一を単離するための候補遺伝子の選定〉
外来遺伝子を高発現させるための遺伝子発現プ口モーターを発見するためには、 宿主海藻において高発現している遺伝子を知り、 そのゲノム配列を単離し、 その 構造を解析する必要がある。
スサビノリの E S T酉己歹 (J (e x p r e s s e d s e q u e n c e t a g) は、 かずさ DNA研究所のホームページ 〔: t t p : //www. k a z u s a . o r. j p/e n/p 1 a n t/p o r p h y r a/EST ] にて公開されて いる。
この配列情報をもとに ESTクラスタリング解析 (EST c l u s t e r i n g a n a 1 y s i s) を、 例えば PARACEL CLUSTER I NG PACKAGEソフトウェア (パラセルネ ±®) を用いて行なう。 クラスタリング 解析の結果、 同一クラスタに属する EST配列の数が多いものが、 高発現してい る可能性のある遺伝子と考えられるので、 それらの遺伝子のプ口モーターを単離 するための配列の候補とすることが出来る。
例えば、 その中の 2つの遺伝子 [P o r p hy r a y e z o e n s i s e l on g a t i on f a c t o r EF 1— a l p h a A丄退伝ナぉょぴ P o r p h y r a y e z o e n s i s e l on g a t i o n f a c t o r EF l-a l p h a A2遺伝子 (本明細書では以下それぞれ A 1遺伝子、 A 2 遺伝子と呼ぶ) 〕 を選ぴ、 本発明のプロモーターを単離するための材料とするこ とが出来る。
上記候補遺伝子の c DNAのクローニングは、 上記クラスタリング解析におい て選定された候補遺伝子が属するクラスタに含まれる E ST配列、 アセンブル (a s s emb l e) してできたコンセンサス酉己歹 (c o n s e n s u s s e q u e n c e) またはそれらの一部をプローブとして行なうことが出来る。 例え ば、 A 1遺伝子に属する E S T配列をアセンブルしてできたコンセンサス配列か らプライマーを設計し、 このプライマーを使用した P C Rで増幅した A 1遺伝子 断片をプローブとして用いて行なうことが出来る。
このようにして、 本発明において単離された新規候補遺伝子の c DNAの一例 が配列表の配列番号 1に示される A 1遺伝子の c D N Aである。
ぐ海藻のすべての生活環で高発現している遺伝子力否かの確認 >
候補遺伝子が海藻のすべての生活環で高発現している遺伝子力、否かの確認は、 海藻の生活環のさまざまなステージにおける m R N Aの発現量を、 候補遺伝子の D N A配列をプロープに用いたノーザンハイプリダイゼーションで測定すること により調べることができる。
海藻の糸且織から全 RNAを抽出するには、 公知の方法、 例えば、 グァニジンチ オシァネート法ゃフエノール/クロ口ホルム法などが用いられる。 このように抽 出された全 R N Aを、 目的の m R N Aの発現量を測定するために使用することが 出来る。
例えば、 海藻スサビノリの生活環ステージのうち、
(1) 単胞子未形成ステージ、
(2) 単胞子形成ステージ、
(3) 単胞子形成後の栄養成長配偶体ステージ、 (4) 糸状体ステージ、
からそれぞれ改変フエノール Zクロ口ホルム法を用いて、 全 RNAを抽出して、 ァガロースゲル電気泳動を行つた後、 候補遺伝子の D N Aをプローブにしてノザ ンハイブリダイゼーシ 3ンを行うことができる。
また、 別の方法として RT— PC Rによっても発現様式を解析することができ る。 この場合、 特異的なプライマーを設計することにより、 より厳密にファミリ 一遺伝子と区別した解析が可能である。
例えば、 候補遺伝子の c DNAの非翻訳領域にプライマーと翻訳領域内のブラ イマ一を用いて、 上記の各ステージから、 例えば、 グァニジンチオシァネート法 を用いて抽出した全 RNAを鎵型にして、 T aKa Ra RNA PCR K i t Ve r . 2. 1 (宝酒造社製) 等を用いて R T— P C Rを行い、 各ステージ における遺伝子の発現量を測定することができる。
このようにして、 各ステージの海藻で高発現している遺伝子を探し出すことが できる。
くプロモーターを含有する領域のクローニング〉
上記のようにして選択された各ステージで高発現している遺伝子のプロモータ 一を含有する領域は、 例えば、 ゲノムライブラリーより該遺伝子のゲノム DN A をスクリーニングすることにより単離することが出来る。
海藻で高発現する遺伝子のプ口モーターのクローニングは、 ゲノムライブラリ 一を用いたプラークハイブリダィゼーシヨン法、 ゲノム DNAと既知の配列をも とに未知の配列を増幅する P C R法などがあるが、 これらに限定されるわけでは なレ、。
本発明において単離された、 配列表の配列番号 3に示される新規な e l o n g a t i o n f a c t o r EF 1— a l p h a A 2遺伝子を含むゲノム DN A断片は、 各ステージで高発現している遺伝子の一例である。 この A 2遺伝子
(配列番号 3 ) と配列番号 1の cDNAおよびクラスタリング解析した E S T配 列を比較すること、 転写因子検索、 コード領域検索、 イントロン 'ェキソン検索 をすることにより、 この配列には、 プロモーター配列、 イントロン配列、 ターミ ネーター配列が含まれることがわかった。 即ち、 配列表の配列番号 3に示される塩基配列は、 e l o n g a t i on f a c t o r EF l-a l p h a A 2遺伝子のゲノムの全長をカバーする配列 であり、 該配列中の 1368〜 2717番目の塩基配列が A 2ポリぺプチドをコ 一ドする配列 (終止コドンを含む) 、 878〜 1331番目の塩基配列が第 1ィ ントロン配列、 1〜 859番目の塩基配列がプロモーター配列、 2718〜 37
64番目の塩基配列がターミネーター配列である。
また、 この配列番号 3で表される塩基配列と配列番号 1の c DNAおよぴクラ スタリング解析した E S T配列を比較することにより、 A 2遺伝子の c D N A配 列が明らかとなった。 該配列を配列表の配列番号 2に示す。 該配列中、 配列表の 配列番号 3の塩基配列に含有されている領域が EF l_a l p h a A2遺伝子 のェキソン領域である。
本発明で単離された上記のプロモーターは、 藻類においてプロモーター活性を 有する DN Aであり、 所望の遺伝子を藻類において発現させるために使用するこ とが出来る。 また、 該配列に厳密な条件下でハイブリダィズし、 プロモーター活 性を有する DNA、 該配列にプロモーター活性を失わない範囲で塩基の置換、 欠 失、 挿入、 付加がなされた DNAも上記目的のために使用することが出来る。 本 明で単離された上記のターミネーターは、 藻類にぉレ、てターミネーター活 性を有する DN Aであり、 所望の遺伝子を藻類において発現させる際、 その発現 効率を上昇させる目的で使用することが出来る。 また、 該配列に厳密な条件下で ハイブリダィズし、 ターミネータ一活性を有する DNA、 該配列にターミネータ 一活性を失わない範囲で塩基の置換、 欠失、 挿入、 付加がなされた DNAも上記 目的のために使用することが出来る。
高等植物では、 イントロン配列を含む導入遺伝子を使うことにより転写効率が 高まり導入遺伝子の目的タンパクが高生産される例が知られている。 本発明で単 離された上記第 1イントロンは、 所望の遺伝子を藻類において発現させる際、 そ の転写効率を上昇させる目的で使用することが出来る。 また、 前記の転写効率上 昇活性を有するものであれば、 該配列に厳密な条件下でハイブリダイズする D N A、 該配列に塩基の置換、 欠失、 揷入、 付加がなされた DN Aも上記目的のため に使用することが出来る。 本発明で単離された上記ェキソン領域は、 藻類の新規なェ口ンゲーシヨン フ アクター (EF) である EF 1— a l p h a A 2ポリペプチドをコードする配 列である。 該配列は E F 1— a l p h a A2ポリぺプチドの遺伝子工学的生産 に使用でき、 また、 さらに新規な EF遺伝子のスクリーニング、 特に藻類由来の 新規 EF遺伝子のスクリーニングのプローブ、 プライマーとして使用できる。 本願明細書に開示された本発明の DNAは、 上記のようにして単離することが 出来る。 また本明細書に開示された配列情報をもとに化学的に合成することも出 来る。
本発明で、 藻類においてプロモーター活性を有する DNAが初めて単離された ことにより、 現在まで全く不可能であった、 藻類での所望の遺伝子の人為的な発 現が初めて可能となった。 さらに、 藻類由来のターミネータ一活性を有する DN A、 イントロンが初めて単離されたことにより、 藻類での所望の遺伝子の発現の 効率を高めることも可能となった。 このことは、 藻類における有用物質の遺伝子 工学的生産を可能とするのみならず、 従来、 甚だ効率の悪い交配法に頼るしかな かった藻類の育種を、 遺伝子工学的に効率よく行なうことをも可能にするもので ある。
本発明で、 藻類由来の新規エロンゲーションファクタ一遺伝子が初めて単離さ れたことにより、 現在まで採取あるいは培養した藻類からしか取得することが出 来なかった藻類由来のエロンゲーションファクターを遺伝子工学的に生産するこ とが可能となった。 実施例
以下に実施例をもつて本発明を示すが、 本発明はこれらの実施例の範囲のみに 限定されるものではない。
実施例 1 スサビノリの各ステージからの mRNAの調製
1. 材料
クヮノ K. (Kuwa n o K. ) ら 〔プラント サイエンス (P l a n t S c i . ) 、 第 116卷、 第 1集、 第 1 17〜 124頁 (1996年) 〕 により 確立された、 スサビノリの純系株である、 P o r phy r a y e z o e n s i s Ue d a TU— 1株の葉状体の配偶子おょぴ糸状の胞子体を用いた。
2. 培養条件
葉状体の配偶子は 1 5°C、 10時間明期 〔光量 80 E/ (m2 · s) ;白色 蛍光灯〕 、 14時間暗期のもと、 ESL培地 〔3. 5% (w/v) SEAL I F E p owe r (マリン テック (Ma r i n e Te c h) 社製) 、 1 % (v
/v) ES S2〕 の入ったガラス製の 1リットル容枝付き平底フラスコ (パイレ ックス (PYREX) 社製) 中で通気 (フィルター濾過滅菌した空気、 0. 12 リットル/ m i n) し、 還流浮遊状態で培養した。
糸状の胞子体は 25 °C、 24時間明期 〔光量 17 E/ (m2 · s) ;白色蛍 光灯〕 のもと、 ESL培地の入ったガラス製の 500ml容三角フラスコ中で静 置培養した。
3. 全 RNA抽出
スサビノリ TU— 1株の各ステージの全 RN Aは、 単胞子が未形成で葉長 1〜 2 mmの葉状の配偶体、 単胞子の放出の誘導を 30分間かけた葉長 2 mmの葉状 の配偶体、 葉長 1〜 2 c mの葉状の配偶体、 およぴ糸状の胞子体からそれぞれ抽 出した。
全 RNAの抽出は、 アプト K. E. (A t K. E. ) らのフエノー ル /ク口口ホルム法を一部改変して行つた 〔モレキュラー アンド ジェネラル ジエネテイクス (Mo 1. Ge n. Ge n e t. ) 、 第 246巻、 第 4号、 第 455〜464頁 (1995年) 〕 。
藻体から全 RN Aを抽出する手順を以下に示す。
培養した藻体の水分を紙タオルで除いた後、 重量を測定した。 1. 2 gの藻体 を液体窒素中で乳鉢と乳棒を用いて破砕し、 液体窒素で予冷した薬さじを用いて、 16. 8m 1の抽出バッファー 〔10 OmM Tr i s— HC 1 ( H 8. 0) 、 1. 5M Na C l、 2 OmM EDTA、 2 OmM DTT (ジチオト レイトール) 、 2%CTAB (セチルトリメチルアンモニゥムプロミド) 〕 を入 れた 5 Oml容の遠沈管に移した。 これを振盪機 (NR_1、 タイテック (TA I TEC) 社製) を用いて 1 10 r pm/m i n、 室温で 15分間混合した後、 等量のクロ口ホルムを加えて攪拌した。 この混合液を遠心して上層を集め、 ここに 1 Z 3量のエタノールを一滴ずつゆ つくりと加え混合した。 これを遠心して沈澱 (主に多糖類が含まれている。 ) を 除去した後、 上清に等量のクロ口ホルムを加えて攪拌した。 これを遠心して上清 を集めた。 この操作を上清の色が抜けるまで繰り返した。
上清を回収し、 上清の 1Z 2量の 9M L i C 1と上清の 1/100量の ]3— メルカプトエタノールを加え混合し、 一 20°Cでー晚静置した。 生じた沈澱を遠 心分離により回収し、 沈澱を 50 μ 1の DE P C (ジェチルピロカーボネート) 処理水に溶かして、 等量のフエノール/クロ口ホルム (1 : 1) による抽出を行 なった。 この抽出操作を中間層がなくなるまで繰り返した。
上層を回収し、 上層の lZl 0量の 3Μ 酢酸ナトリウムと 2倍量のエタノー ルを加え混合した。 生じた沈澱を遠心分離により回収し、 75 %エタノールでリ ンスした後、 風乾した。
沈澱物を 90 μ 1の DE PC処理水に溶かし、 これに lm 1の I SOGEN (和光純薬社製) を加えて混合した後、 5分間室温で保持した。 これに 2m 1のクロ口ホルムを加えて攪拌した後、 2〜 3分間室温で保持した。
これを遠心分離し、 得られた上清に 0. 5mlのイソプロパノールを加え、 生 じた沈澱を遠心分離により回収した。 沈澱を 75 %ェタノールでリンスした後、 風乾し、 400 μ 1の DE PC処理水に溶かした。
さらに、 残存している多糖類を除くため、 200μ 1の高塩沈澱溶液 (1. 2 M Na C l、 0. 8Μ クェン酸 3ナトリウム二水和物、 p H未調整) ならび に 200 μ 1のイソプロパノールを加え、 遠心した。 得られた沈澱を 75%エタ ノールでリンスした後、 風乾し、 100 1の DEP C処理水に溶かした。 このようにして抽出された全 RNAの濃度および純度は、 分光光度計 (UV— 2200, 島津製作所製) により測定した。 また、 1. 0%ァガロースゲルで電 気泳動し、 全 RNAが分解されていないことを確認した。
mRNAは、 上記の方法で抽出された全 RNAから O 1 i g o t e xT M _d T 30 s u p e r (ロシュ (R o c h e) を用いて分離した。 操作手順 は付属の説明書に従った。
実施例 2 スサビノリ E S T配列のクラスタリング解析 解析に用いるスサピノリ E ST配列は、 かずさ DN A研究所のホームページ L t t p : / / www. k a z u s a . o r. j p e n/p l a n t/ p o r p h y r a/E ST/〕 よりダウンロードした。 ダウンロードしたスサビノリ E ST配列の全 10154配列について、 パラセル社の PARACEL CLU STER I NG PACKAGEを用いて、 配列のクラスタリングを行った。 その結果、 8243個の配列が 1140のクラスタに分類された。 また、 どの クラスタにも属さない酉己列が 1911個あった。
以下の表 1〜4に、 各クラスタに属する配列の数が多い上位 50位のクラスタ I D (c l u s t e r I D) をリスト化したものを示す。 同一のクラスタに属 する配列の数 (s e qu e n c e c o un t) が多いほど高発現の遺伝子と考 えられる。
さらに、 各クラスタに属する配列をアセンブルしてできたコンテイダ (Co n t i g) のコンセンサス配列を NCB I (Na t i o n a l Ce n t e r f o r B i o t e c hn o l o gy 丄 n i o rma t i o ru [h t t p : / /www. n c b i. n i h. g o/] のヌクレオチドデータべース (n o n— r e d u n d u n tな核酸のデータベース) に対して、 BLASTプログラム 〔アルトシュル S. F. (Al t s c hu l S. F. ) ら;ヌクレイツ ク ァシッズ リサーチ (Nu c l e i c Ac i d s Re s. ) 、 第 25巻、 第 17号、 第 3389〜 3402頁 (1997年) 〕 にて、 ホモロジ一検索を行 つた。
その結果のうち、 各クラスタのコンティグ (C o n t i g) がもっとも高いス コアでヒットしたコンティグ I D (c o n t i g I D) 、 配列の数 (s e q u e n c e c o un t) およぴヒットした G e nB a n k登録番号 (G e n B a n k a c c e s s i o n No) を、 表 1〜4の Co n t i g I D、 S e q u e n c e Co un t、 Ge nBa nk Ac c e s s i o n Noの欄に 号 (No) とともに示す。 また、 その時のスコア (S c o r e) と E値 (E v a l u e) を、 表:!〜 4の S c o r e、 E v a l u eの欄に示す。 表 1 スサビノリ E S T配列のクラスタリング

* :表 8の N oは、 表 4の N oに対応している c ヒットした遺伝子タイトルに対して、 ゲノム配列やその断片のクローン等では なく、 mRNAまたは cDNAであるヒット、 あるいは P o r p h y r aの語を 含むヒットを検索したところ、 スサビノリの近縁の遺伝子が見出された。
さらに、 高発現している遺伝子のある配列を探索したところ、 上記 50のクラ スタ中、 スサビノリの近縁の mRNAまたは c DN A配列として、 クラスタ I D 13番 (表 1および表 5の No 2) 、 クラスタ I D9番 (表 3およぴ表 7の No 32) の 2種類が見出された。
見出された 2種類の mRNAまたは cDNA配列のうち、 クラスタ I D 13番 は 18 S r RNAとホモロジ一が高いものであった。 一方、 クラスタ I D 9番 のコンティグ (Co n t i g) 1のコンセンサス配列はホモロジ一検索の結果、 G e n B a n k登録番号 (Ge nB a nk Ac c e s s i on No. ) U0 8844の Po r p hy r a p u r p u r e a e l o n g a t i o n f a c t o r EF l-a l p h a (t e f — c) mRNAにヒットし、 本発明 のための候補として最も適しているものの 1つと考えられた。
また、 クラスタ I D 9番のコンティグ (Con t i g) 1のアセンブル結果を 見ると、 ミスマッチがあり、 2つのグループに分かれること、 特に 5, 末端の非 翻訳領域の配列が明らかに異なることが判明した。
これより、 この 2つのグノレープをそれぞれ、 P o r p hy r a y e z o e n s i s e l o n g a t i o n l a c t o r n, e 1— a 1 p h a Al、 e
1 o n g a t i o n f a c t o r EF l-a l p h a A 2と命名した (以 下、 それぞれ A 1遺伝子、 A 2遺伝子と呼ぶ) 。
実施例 3 A 1遺伝子の全長 c DN Aクロー-ング、 シークェンシング、 およ ぴ配歹 IJ解析
1. 材料
c DNAクローニングの材料として、 クフノ K. (Kuwa n o K) . ら 〔プラント サイエンス (P l a n t S c i. ) 、 第 116卷、 第 1集、 第 1 17〜124頁 (1996年) 〕 により確立された、 スサピノリ TU— 1株の葉 状の配偶体単藻培養株を用いた。 2. 培養条件
スサビノリ TU—l株は、 15°C、 10時間明期 〔光量 80/ EZ (m2 · s) ; 白色蛍光灯〕 、 14時間暗期の短日条件で培養した。 ESL培地が入った 1リットル容の枝付き平底フラスコ (パイレックス (PYREX) 社製) 中で通 気 (ポアサイズ 0. 2 μπιのフィルター (アドバンテック (ADVANTEC) 社製) でろ過滅菌した空気、 0. 25リツトル Zm i n) し、 還流浮遊状態で培 養した。 培地は 7日ごとに交換した。
3. RNA抽出
スサピノリ TU— 1株の葉状配偶体単藻培養株の 1. 2 gの藻体より、 実施例 1と同様の方法で全 RN Aを抽出した。
4. c DN Aライブラリースクリーニングのためのプローブ作製
スサピノリ TU— 1株の DNA抽出をキタデ (K i t a d e) ら 〔ジャーナル ォブ フィコロジー (J. P h y c o 1. ) 、 第 32卷、 第 496〜498頁 (1996年) 〕 による方法に従って行った。
この DNAを錶型とし、 Amp 1 i T a q Go l d (アプライド パイオシ ステムズ (Ap p l i e d B i o s y s t ems) ネ環) と 2種類のプライマ 一 [PYEF 1 (配列番号 7) および PYEFR 1 (配列番号 5) 〕 を用いて P CRを行った。 これらのプライマーは、 実施例 2の E STのクラスタリングの結 果をもとに設計したものであり、 e l o n g a t i o n f a c t o r EF 1 -a 1 p h a遺伝子におけるモチーフを含む、 保存性の高い配列を増幅するブラ ィマーである。 P C Rは 94 で 2分間の予温後、 94 で 1分間、 53。Cで 1 分間、 72°Cで 2分間を 1サイクルとし、 このサイクルを 30回繰り返し、 最後 に 72 °Cで 10分間反応させる条件で行つた。
得られた P C R産物 (約 170 b p ) を 1. 5 %ァガロースゲルで電気泳動し た。 目的のパンドを、 Ge n e c l e a n I I k i t (バイオ 101 (B i o 101) ^±M) を用いてゲルから回収し、 DNA L i g a t i o n K i t Ve r . 2 (宝酒造社製) を用いて T Aクロー-ング用ベクターである pT 7B l u eベクター (ノパジェン (No v a g e n) †± ) に連結した。 これを常法 (マニアテイス (Ma n i a t i s ) ら編、 モレキュラー クローニング, ァ ラボラトリー マ二ユアノレ (Mo l e c u l a r C l on i n g, a L a b o r a t o r y Ma nu a l 〔コーノレド スフリンク ノヽーノ ー (Co l d S p r i n g Ha r b o r) 社、 1989年刊〕 ) によりコンビテント細胞 大腸菌 JM109 (宝酒造社製) に導入し、 アンピシリン含有 LB寒天培地で培 養することによつて大腸菌の形質転換体のコ口ニーを得た。
その後、 形質転換されたコロニーを無作為に選び、 該コロニーが保持するブラ スミド中のインサートの長さを、 pT7B 1 u eベクター内の配列を有する T 7 プロモータープライマー (ノパジェン (No v a g e n) 社製) 、 M4プライマ 一 (宝酒造ネ ±¾) による PCR増幅産物を電気泳動することによって確認し、 目 的の長さの D N A断片を持つ大腸菌のコ口ニーを選定した。
選定した大腸菌の形質転換体のコ口ニーを液体培養した後、 Q I A G E N P l a smi d Mi n i K i t (キアゲン (Q i a g e n) 社製) を用いてプ ラスミド抽出を行った。
このプラスミドを鍀型とし、 dRh o d ami n e Te rm i n a t o r Cy c l e S e qu e n c i n g F S Re a dy Re a c t i o n K i t (アプライド バイオシステムズ (Ap p l i e d B i o s y s t em s) 社製) を用いたサイクルシークェンシング法にて塩基配列の決定を行なった。 サ.イタルシークェンシング反応は、 96°Cで 2分間の予温後、 96°Cで 10秒間、 50°Cで 5秒間、 60°Cで 4分間を 1サイクルとし、 このサイクルを 25回繰り 返すことによって行なった。
得られた反応産物を、 簡易型のエタノール沈澱を利用した方法により精製した 後、 AB I PR I SM 310 Ge n e t i c An a l y z e r (ァプラ ィド バイオシステムズ (Ap p l i e d B i o s y s t ems) 社製) によ り塩基配列を決定した。 プラスミド抽出および塩基配列決定の手順は付属の説明 書に従った。
塩基配列を決定した後、 得られた配列が目的の配列であることを確かめるため、 NCB Iが公開している BLASTプログラム 〔アルトシュル S. F. (A l t s c hu l S. F. ) ら;ヌクレイック ァシッズ リサーチ (Nu c l e i c Ac i d s Re s. ) 、 第 25卷、 第 17号、 第 3389〜 340 2頁 (1997年) 〕 を用いて、 核酸配列データベースに対し相同性検索を行つ た。 検索の結果、 得られた配列は、 クラスタリング、 アツセブリングから予測し た A 1遺伝子の配列に最も強くマッチすることが明らかとなった。
このようにして配列が決定されたプラスミ ドを铸型とし、 プライマー PYEF 1 (配列番号 7) および PYEFR1 (配列番号 5) を用い、 上述した条件で再 度 PCRを行った。 この PCR産物を PCR D I G La b e l i n g Mi x (ロシュ (Ro c h e) 社製) を用いてジゴキシゲニンで標識した。 標識反応 後、 未反応の基質を除くため標識した産物を Qu i c k S p i n C o 1 urn n s T M G— 50 (ロシュ (Ro c h e) ネ環) を用いて精製し、 スクリー二 ング用のプローブとした。 標識および精製の手順は付属の説明書に従った。
5. c DNAライプラリーの作製
c D N Aライブラリーの作製に使用した c D N Aは、 実施例 3— 4で調製した ポリ (A) + mRNAから T i m e S a V e r c DNA Syn t h e s i s K i t (アマシャム ファノレマシァ バイォテク (Am e r s h am P h a r ma c i a B i o t e c h) 社製) を用いて合成した。 c DNA合成の際に用 レ、る逆転写酵素には、 EFLCp u r eT M C l o n e d Mu r i n e R e v e r s e T r a n s c r i p t a s e (アマシャム ファノレマシノ バイ ォテク (Am e r s h am Ph a rma c i a B i o t e c h) ネ i^) を用 レ、、 反応温度は 44 °Cで行った。
合成された c DNAは; L Z AP I Iベクター (ストラタジーン (S t r a t a g e n e) 社製) に連結し、 連結反応産物は G i g a p a c k I I I Go 1 d P a c k a g i n g E t r a c t (ストラタジーン (S t r a t a g e n e) 社製) を用いてパッケージング (p a c k a g i n g) した。 以下、 この パッケージングして得られたファージ液を cDNAライプラリーとして使用した。 6. 一次スクリーニング
上記のパッケージングして得られたファージ液を ZAP— c DNA s y n t h e s i s K i t (ストラタジーン (S t r a t a g e n e) ^±M) 付属の説 明書に従い、 1プレートにっきプラーク数が約 1 X 105 となるように、 あら かじめボトムプレートを作製、 保温しておいた 24 X 24 cm角型シャーレ (MS - 12450、 住友べ一クライト (Sumi t omo Ba k e l i t e) 社製) にまいた。 常法に従い、 プレートに生じたファージプラークをニトロ セルロースメンブレン (N I TROP LUS 2000、 フナコシ (F u n a k o s h i) ネ ±¾) に写しとつた。
このメンブレンをアルカリ変性液 (0. 9M NaOH、 1. 5M Na C
1 ) に 2〜5分間、 次に中和液 (1. 5M Tr i s—HC l (pH7. 5) 、 1. 5M N a C 1 ) に 2〜 5分間浸した。 次いで、 このメンブレンを 6 X S SCで洗い、 乾燥させた後、 乾熱滅菌機 (SP— 650、 アドバンテク (ADV ANTEC) 社製) にて 80°Cで 2時間加熱した。
このメンプレンを、 終濃度 100 μ g/m 1の変性サケ精子 DN Aを含むハイ ブリダィゼーション溶液 〔 5 Xデンハルト (De nh a r d t' s) 溶液、 4. 1 mM EDTA、 0. 5%SDS、 6 X S S C〕 中で 65°C、 2時間インキ ュベートすることでプレノヽィブリダイゼーシヨンを行つた。
次に、 メンブレンを、 終濃度 1◦ 0 μ gZm 1の変性サケ精子 DN Aならびに 熱変性させ 1本鎖にした実施例 3— 4で調製したプローブを終濃度 10 n g /m 1で含むハイブリダイゼーション溶液中で 60 °Cでー晚 ( 12時間以上) ィンキ ュベートすることでハイブリダイゼーションを行った。
ハイプリダイゼーション後、 メンブレンを、 洗浄溶液 ( 2 X S S C、 0. 1%SDS) 中で室温、 5分間保持し、 結合していないプローブ、 ハイブリダィ ゼーション溶液などを洗!/、落とした。 この洗浄を 2回繰り返した。
次に、 メンプレンを、 あらかじめ 60°Cに保温しておいた洗浄溶液 (0. 5 X S S C、 0. 1 % S D S ) 中で 60 °C、 20分間保持し、 非特異的結合をし ているプローブを解離させた。 この洗浄を 2回繰り返した。
次いで、 メンプレンを、 あらかじめ 60°Cに保温しておいた洗浄溶液 (0. 1 XSSC、 0. 1%SDS) 中で 60°C、 20分間保持し、 非特異的結合をし ているプローブを解離させた。 この洗浄を 2回繰り返した。
ジゴキシゲニンの検出は、 D I G DNA La b e l i n g a nd D e t e c t i o n K i t (ロシュ (R o c h e) 社製) 付属の説明書に従い、 発 色反応で行った。 検出結果の確認は、 写真用ライトボックス (L I GHT BO X NEW 5000 INVERTER, フジ写真フィルム株式会社 (F u j i Co 1 o r) 製) を用いて行った。 以上の手順で一次スクリーニングを行つ た。
7. 二次スクリーユング
上記の一次スクリーニングで陽性となったファージを回収し、 ライプラリーの 増幅を真壁 〔真壁和裕;バイォ実験ィラストレイテッド ( 4 ) 苦労なしのクロ 一ユング 秀澗社 (1997年) 〕 の方法に従って行った。
二次スクリーエングは、 増幅されたファージ液を、 1プレートにっきプラーク 数が 50〜100となるように、 あらかじめボトムプレートを作製、 保温してお いた 9 cmシャーレ (高さ 1. 5 cm、 直径 9 cm) にまき、 得られたプラーク について二次スクリーニングを行なった。 二次スクリーニングの操作は、 一次ス クリーニングと同様の条件で行った。
上記の方法に従って、 葉状配偶体に由来する c DNAライプラリーを作成し、 一次スクリーニングを行ない、 分離された陽性クローンにっき、 増幅を行った。 増幅されたファージ液より得られたプラーク約 1 X 105 をスクリーニングす ることによって、 5個の陽1生クローンが分離された。 二次スクリーニングで陽性 と判断されたプラークについては、 ベクター配列に特異的な 2種類のプライマー 〔T 3プロモータープライマー (ノバジェン (No v a g e n) 社製) および T 7プロモータープライマー (ノバジェン (No v a g e n) ¾ )〕 を用いて P CRを行い、 増幅産物を 1. 0%ァガロースゲル電気泳動に供することにより揷 入断片の長さを確認した。
ファージベクター中の挿入断片のプラスミ ドベクターへのサブクローニングは、 i n V i V oエキシジョンにより行った。 操作手順は ZAP— c DNA S y n t h e s i s K i t s (ストラタジーン (S t r a t a g e n e) ^± ) 付 属の説明書に従った。
i n v i v oエキシジョンによって得られたコロニーを無作為に選び、 上述 のプライマー対 (T 3プロモータープライマーおよび T 7プロモータープライマ 一) を用いて PCRを行い、 増幅産物を 1. 0%ァガロースゲル電気泳動に供す ることにより揷入断片の長さを確認した。 8. 塩基配歹の解析
最も長い cDNAが揷入されたプラスミドを大腸菌に導入し、 得られた形質転 換体を液体培養した後、 Q I AGEN P l a smi d Mi n i K i t (キ ァゲン (Q i a g e n) 社製) を用いてプラスミドを抽出し、 このプラスミドを 用いて挿入 c DNA領域の塩基配列をプライマーウォーキング法により決定した。 決定された c DN A配列を配列表の配列番号 1に示す。 全長 1644b pの c DNA配列中に1350 b ρの長さの読み取り枠 (o p e n r e a d i n g f r ame、 ORF) が見いだされ (塩基番号 79〜 1428 ) (終止コドンを 含む) 、 この ORFは、 449個のアミノ酸からなるタンパク質をコードしてい た。 該 ORFにコードされるタンパク質のァミノ酸配列を配列表の配列番号 8に 示す。 該 ORFにコードされるタンパク質の予測される分子量は 49. 4 kD a であり、 等電点は 9. 18である。
開始コドンを含む塩基番号 76〜82の領域にコザック規則 [Ko z a k M. ; C e 1 1, 第 44卷, 第 283頁〜第 292頁 (1986年) 〕 と似た 配列 (ATCATGG) が見られた。
一方、 cDNA配列中にポリ (A) 配列付加信号 (AATAAA) およびポリ (A) 尾部は見つからなかったが、 出芽酵母 (S a c c h a r omy c e s c e r e v i s i a e) 由来の CYC 1遺伝子において報告されている、 ポリ (A) 付加配列 (TACATA) 〔プラウフット N. (P r o u d f o o t N. ) ;セル (C e 1 1) 、 第 64卷、 第 4号、 第 671〜674頁 (1991 年) 〕 に似た配列 (TACTATT) が塩基番号 1579〜1585の領域に見 られた。
得られた配列が EF—1ひをコードする遺伝子であることを確かめるため、 相 同性検索ならぴにモチーフ検索を行った。
相同性検索には NCB Iが公開している B LAST 2. 0 ADVANCE
D BLAST SEARCHの BLASTX2. 0. 10プログラム 〔アルト シュル S. F. (Al t s c hu l S. F. ) ら;ヌクレイック ァシ ッズ リサーチ (Nu c l e i c Ac i d s Re s. ) 、 第 25卷、 第 17 号、 第 3389〜 3402頁 (1997年) 〕 を用いて行った。 モチ1 ~フ検索は、 I n t e r P r o 〔h t t p : ZZ www. e b i . a c . u kZi n t e r p r o/〕 が公開しているプログラム I n t e r P r o R e 1 e a s e 4. 0 〔ァプヴアイラー R. (A p w e i 1 e r R. ) ら;ジ ヤーナル ォブ バイオサイェンシズ (J. B i o s c i . ) 、 第 26卷、 第 2号、 第 277〜 284頁 (2001年) 〕 を用いて、 PROS I TEのモチー ファーグベ1 ~ス し h t t p : /Zwww. e X p a s y . c hz p r o s i t e /〕 に対して検索を行った。
BLASTX (核酸の配列をァミノ酸の配列のデータベースに対して検索す る) プログラムによる相同性検索の結果、 一番初めに的中したのは類似性の評点 力 S887の P o r p hy r a p u r p u r e aの EF— l a t e f — cであ つた。 E値 (E v a l u e) (対象データベースと同じ大きさの任意の配列ラ ィブラリ一を無作為に検索したときに、 類似性の評点以上の評点を持つものが的 中する個数の期待値) は、 有意に低い数値を示したので、 偶然に的中したのでは なく、 本実施例で決定された上記配列は P o r phy r a p u r p u r e a由 来 EF— l a t e f — cと類似する配列であることは明らかである。
I n t e r P r oによるアミノ酸のモチーフ検索の結果は、 次のとおりであつ た。
PROS I TEのデータベースに対して、 配列表の配列番号 8に示されるアミ ノ酸配列のアミノ酸番号 14から 21の領域 (GHVDSGKS) に、 ATP/ GTP— b i n d i n g s i t e mo t i f A (P- 1 o o p) [PRO
S I TE登録番号 (PROS I TE a c c e s s i o n numb e r) PS 00017〕 力 また、 配列表の配列番号 8に示されるアミノ酸配列のアミノ酸 番号 61から 76の領域 (DKLKAERERG I T I D IA) に、 GTP— b 1 n d 1 n g e l o n g a t i o n f a c t o r s s i gn a t u r e 〔PROS I TE登録番号 (PROS I TE a c c e s s i o n numb e r ) P S 00301] が見つかった。
このことより、 本実施例で得られた DN A断片は e l o n g a t i o n f a c t o rとして機能するアミノ酸をコードする新規の DNA断片と考えられる。 実施例 4 スサビノリのライフサイク^/の各ステージにおける A 1遺伝子およ ぴ A 2遺伝子の発現様式の RT— PCRによる解析
1. A1遺伝子
(1) プライマー合成
A 1遺伝子の発現様式を調べるための RT— PCR用プライマーとして、 プラ イマ一 PYEFRT1 (配列番号 4) およぴプライマー P Y E F R 1 (配列番号 5) を合成した。
プライマー PYEFRT1の配列は、 実施例 3で明らかにされた配列番号 1で 示される A1遺伝子の 5' 末端非翻訳領域の配列を、 実施例 2でクラスタリング した E S T配列のうち A 2遺伝子のグループに属し、 最も 5, 末端非翻訳領域の 配列が長い配列 A V 435982とを比較し、 明らかに配列が異なる領域を選択 し、 設定した酉己列である。
即ち、 プライマー PYEFRT 1の配列は、 配列番号 1で示される A 1遺伝子 の 5, 末端非翻訳領域中の配列で、 かつ A 2遺伝子の 5, 末端非翻訳領域の配列 とホモロジ一を持たなレ、配列である。
また、 プライマー PYEFRT 1の配列は、 実施例 2でクラスタリングした E
S T配列のうち、 A 2遺伝子のグループに属する配列とも、 共通にホモロジ一の 高い、 P o r p hy r a p u r p u r e a e l o n g a t i o n r a c t o r EF 1— a l p h a (t e f — c) nxRNA中の配列である。
(2) RT— PCR
スサビノリ TU— 1株の各ステージの全 RNAは、 単胞子が未形成で葉長 1〜
2 mmの葉状の配偶体、 単胞子の放出の誘導を 30分間かけた葉長 2 mmの葉状 の配偶体、 葉長:!〜 2 cmの葉状の配偶体、 そして糸状の胞子体から、 実施例 1 と同様の方法でそれぞれ抽出した。
得られたそれぞれ 200 n gのポリ (A) + RNAから RNA PCR K i t (AMV) Ve r . 2. 1 (宝酒造ネ ± ) を用いて c DNAを合成した。 逆転写反応は、 O l i g o dT— Ad a p t o r P r ime r (宝酒造社 製) をプライマ一として用いて 53°Cで行った。 操作手順は付属の説明書に従つ た。
得られた 5 n gの cDNAを錄型とし、 Ge n e Ta q NT (和光純薬社 製) と 2種の合成プライマー 〔PYEFRT 1 (配列番号 4) および PYEFR 1 (配列番号 5 ) 〕 を用いて P C Rを行った。 P C Rは 94 °Cで 2分間の予温後、 94 °Cで 30秒間、 53でで 30秒間、 72でで 1分 30秒間を 1サイクノレとし、 このサイクノレを 30回繰り返し、 最後に 72 °Cで 7分間反応させる条件で行つた。 ネガティブコントロールとして、 逆転写反応を行なわない各ステージのポリ
(A) + RNA鎵型として上述と同様の条件で PCRを行った。 PCR産物は 1· 5 %ァガロースゲルで電気泳動した。
ァガロースゲル電気泳動の結果を図 1に示す。

いずれのステージにおいても 391 b pの増幅産物が確認され、 A 1遺伝子は スサビノリのライフサイクルのどのステージにおいても強く発現していると考え られた。
2. A 2遺伝子
(1) プライマー合成
A 2遺伝子の発現様式を調べるための RT— PC R用プライマーとして、 プラ イマ一 PYEFRT4 (配列番号 6) を以下のようにして設計した。
プライマー PYEFRT4の酉己列は、 前記した A 2遺伝子のグループに属し、 最も 5 ' 末端非翻訳領域の配列が長い配列 A V 435982を、 実施例 3で明ら かにされた配列番号 1で示される A 1遺伝子の 5 ' 末端非翻訳領域の配列と比較 し、 明らかに配列が異なる領域を選択し、 設定した。
即ち、 プライマー PYEFRT4の配列は、 A 2遺伝子の 5, 末端非翻訳領域 中の配列で、 力つ A 1遺伝子の 5, 末端非翻訳領域の配列とホモロジ一を持たな い配列である。
このプライマー PYEFRT4と、 上述のプライマー PYEFR 1 (配列番号 5) とを用いて RT—PCRを行なった。
(2) RT-PCR
スサビノリ TU— 1株の各ステージの全 RNAは、 単胞子が未形成で葉長 1〜
2 mmの葉状の配偶体、 単胞子の放出の誘導を 30分間かけた葉長 2 mmの葉状 の配偶体、 葉長 1〜 2 c mの葉状の配偶体、 そして糸状の胞子体から、 実施例 1 と同様の方法でそれぞれ抽出した。
得られたそれぞれ 200 n gのポリ (A) + RNAから RNA PCR K i t (AMV) V e r . 2. 1 (宝酒造社製) を用いて c D N Aを合成した。 逆転写反応は、 O l i g o dT— Ad a p t o r P r ime r (宝酒造社 製) をプライマーとして用いて 53°Cで行った。 操作手順は付属の説明書に従つ た。
得られた 5 n gの cDNAを錶型とし、 Ge n e T a q NT (和光純薬社 製) と 2種の合成プライマー PYEFRT4 (配列番号 6) と PYEFR1 (配 列番号 5 ) を用いて P C Rを行つた。 P C Rは 94 °Cで 2分間の予温後、 94°C で 30秒間、 53 °Cで 30秒間、 72 で 1分 30秒間を 1サイクルとし、 この サイクルを 30回繰り返し、 最後に 72 °Cで 7分間反応させる条件で行つた。 ネガティブコントロールとして、 逆転写反応を行なわない各ステージのポリ (A) + RN A錶型として上述と同様の条件で PC Rを行った。 PCR産物は 1.
5 %ァガ口ースゲルで電気泳動した。

いずれのステージにおいても 369 b pの増幅産物が確認され、 A 2遺伝子は スサビノリのライフサイクルのどのステージにおいても強く発現していると考え られた。
実施例 5 ゲノムライプラリーからの A 2遺伝子の単離、 配列決定とプロモー ター配列の単離
1. ゲノム DN Aの調製
スサビノリ TU— 1株のゲノム DNAの抽出は、 Nu c l e o n Phy t o Pu r e, P l a n t a n d f un g a l DNA e x t r a c t i o n k i t s (アマシャム ファノレマシア バイオテク (Am e r s h am P h a r ma c i a B i o t e c h) 社製) を用いて、 キットに付属の説明書の 方法を一部改変して行つた。
葉状体ステージの藻体からゲノム DNAを抽出する場合の手順を以下に示す。 培養した葉状体ステージの藻体の水分を紙タオルで除いた後, 重量を測定した。 2. 5 gの藻体を液体窒素中で乳鉢と乳棒を用いて破碎し、 液体窒素で予冷した 薬さじを用いて、 1 1. 5mlの上記キットに添付の R e a g e n t 1緩衝液を 入れた 5 Oml容の遠沈管に移し、 終濃度 20 μ g/mlとなるよう RNa s e Aを加え、 37 で 30分間保持した。 これに 3. 75mlの上記キットに添付 の Re a g e n t 2を加え、 均一になるまで攪拌した後、 65°Cで途中 3回引き 上げてよく混ぜながら 10分間保持した。 これを氷中で 20分間保持した後、 2 m 1のクロ口ホルムを加え、 _20°Cで 20分間保持した。 これに 500 μ 1の N u c 1 e o n Phy t o Pu r e DNA e x t r a c t i o n r e s i n (アマシャム フアルマシア バイオシステムズ社製) を加えて、 室温で 1 0分間混合した。
これを遠心した後、 上層を回収し、 ここに終濃度 3. 2 gZ3mlとなるよう 塩ィ匕セシウムを加えて溶解した後、 さらに 1Z10量のェチジゥムブ口マイド (終濃度 10 m g /m 1 ) を添加し、 超遠心チューブ ( 12 P Aシールチューブ、 日立製作所 (H I TACHI) 製) に分注し、 50, 000 r pm、 15°Cで 2 0時間、 日立超遠心機 65 P (日立製作所 (HI TACHI) 製) にて遠心分離 した。
遠心分離後、 超遠心チューブに紫外光を当てながら注射針 (22GX 1、 テ ルモ (Te r umo) 社製) を装着したシリンジ (テルモシリンジ 2. 5ml、 テルモ (Te r umo) 社製) を用いて DNA層を抜き取った。 抜き取った DN A層に等量のイソブタノールを加えて攪拌、 遠心した後、 下層を回収した (イソ ブタノール処理) 。 ェチジゥムブロマイドの色が確認できなくなるまでイソプタ ノール処理を繰り返した (2〜3回繰り返す) 。
回収した下層を透析膜 (S p e c t r a/P o r 7 Memb r a n e、 ス ぺクトラム (S p e c t r um) ネ ±B) に入れ、 1リツトルの TEバッファー (1 OmM Tr i s_HC l、 1 mM EDTA、 pH7. 5) に対して室温 で一晩透析を行なつた。 透析後、 1 Z 10量の 3 M 酢酸ナトリウムと 2倍量の エタノールを加え混合し、 生じた沈澱を遠心分離により回収し、 75%エタノー ルでリンスした後、 風乾した。
こうして得られた沈澱物を 100 1の TEバッファーに溶かした。 抽出され たゲノム DN Aの濃度および純度は、 分光光度計 (G e n e Q u a n t、 アマ シャム ファノレマシァ ノ ィォテク (Am e r s h am Ph a rma c i a B i o t e c h) 社製) により測定した。 また、 1. 0%ァガロースゲルで電気 泳動し、 ゲノム D N Aが分.角军されていないことを確認した。
2. プローブの作成 実施例 2での解析から、 プロモーターの単離候補の A 1遺伝子、 A 2遺伝子は、 G e n B a n k登録番号 (G e nB a n k Ac c e s s i o n No. ) UO 8844の P o r p hy r a p u r p u r e a e l o n g a t i o n f a c t o r EF 1— a l p h a ( t e f - c) mRN Aとホモロジ一が高!、 ことがわかっていた。
これを基に、 A 1、 A 2両方の遺伝子のゲノム配列を単離できるように、 e l o n g a t i o n f a c t o r EF 1— a l p h a遺 子におけ Oモチーフ を含む保存性の高い配列を検索した。 そして、 該モチーフを挟むようにフォヮ一 ドプライマ一 PYEF 1 (配列番号 7) およぴリパースプライマー PYEF R 1 (配列番号 5) を設計した。
両プライマーと Amp 1 i T a q Go l d (パーキンエルマ一 (P e r k i nE 1 me r) ネ環) を用い、 スサビノリ TU— 1株のゲノム DNAを鍚型と して、 Ge n e Amp PGR Sy s t em 9600 (パーキンェノレマー (P e r k i nE lme r) 社製) を使用して P CRを行った。 PCRは 95°C で 10分間の予温後、 94 °Cで 1分間、 52。Cで 1分間、 72 °Cで 2分間を 1サ イタルとし、 このサイクルを 30回繰り返し、 最後に 72 °Cで 10分間反応させ る条件で行った。 PCR産物を 2. 0%シーケム GTG (宝酒造社製) ァガロー スゲルで電気泳動し、 目的の D N A断片 (約 170 b p ) をゲルから切り出して 回収し、 Ge n e C l e a n I I (バイオ 101 (B i o 101) ネ環) を 用いて DNAを精製した。 この DNAを L i g a t i o n K i t v e r . 1
(宝酒造社製) を用いて T Aクローニング用ベクターである pT7B 1 u eべク ター (ノパジェン (No V a g e n) 社製) へ連結した。 約 16時間のライゲー ション反応後、 得られたライゲーション産物にて大腸菌 JM109コンビテント セル (宝酒造社製) を形質転換し、 該形質転換 JM109をアンピシリン添加 L B寒天培地にて培養し、 コロニーを形成させた。
コロニーが保持するプラスミド中のインサートを、 p T 7 B 1 u eベクター中 の配列を有する T 7プロモータープライマー (ノバジェン (No v a g e n) 社 製) および M4プライマー (宝酒造社製) を用いたコロニー PCRによって確認 し、 目的の DNA断片を持つプラスミドを保持する大腸菌のコロニーを選定した。 選定した大腸菌から Q i a g e n P l a sm i d Mi n i K i t (キア ゲン (Q i a g e n) 社製) を用いてプラスミドを抽出、 精製した。 操作手順は 該キットの説明書に従った。 このプラスミ ドを铸型にし、 Dy e Te rm i n a t i o n Cy c l e S e qu e n c i n g F S Re a dy Re a c t i o n K i t (パーキンェノレマー (P e r k i nE lme r) 社製) 、 AB
I P r i sm 310 Ge n e t i c a n a l y z e r レーキンエルマ 一 (P e r k i nE l me r) 社製) を使用し、 ジデォキシ法を用いたサイクル シークェンス法によって塩基配列の決定を行った。 操作手順は該キットの説明書 に疋つに。
このようにして塩基配列が決定されたプラスミドを PCR D I G ラベリン グキット (ロシュ (Ro c h e) 社製) を用いて標識し、 ゲノムライブラリーの スクリーニングを行うためのプローブを作製した。 操作手順は該キットの説明書 に従った。 標識反応後、 Qu i c k S p i n Co 1 umn sT M G— 50 (ロシュ (Ro c h e) 社製) を用いて余分な基質を除き、 精製標識産物を得た。 該精製標識産物をスクリ一二ング用のプローブとした。
3. ゲノムライプラリーの作製
実施例 5— 1で抽出したゲノム DNAを λ DASH I I/B amHI Ve c t o r K i t (ストラタジーン (S t r a t a g e n e) ± ) を用いて; ID ASH I Iベクターに連結し、 ゲノムライブラリーを作製した。 操作手順は該キ ットに付属の説明書に従った。 連結反応産物は G i g a p a c k I I I Go I d P a c k a g i n g Ex t r a c t 、ストフタンーン (S t r a t a g e n e) 社製) を用いて P a c k a g i n gし、 得られたファージ液を、 該 p a c k a g i n g e t r a c tに付属の説明書に従い、 1プレートのプラーク 数が 1 X 104 となるように、 あらかじめボトムプレートを作製、 保温してお いた 90X 2 Omm滅菌シャーレ (SH— 20、 イワキ (I wa k i) ^±M) にまいた。
4. 一次スクリーニング
プレートに生じたファージプラークを、 常法に従って二トロセルロースメンブ レン (NI TROPLUS 2000、 フナコシ (F u n a k o s h i ) に 写しとつた。
メンブレンをアルカリ変性液 (1. 5M NaC l、 0. 5M NaOH) に 5分間、 次に中和液 (1. 5M Na C l、 1. 5M Tr i s— HC 1 ( H 7. 5) ) に 5分間浸した。 メンプレンを 6 XS SCで洗い、 乾燥させた後、 乾熱滅菌器 (SP— 650、 ァドバンテク (AD VAN TEC) にて、 8
0°Cで 2時間加熱した。
このメンブレンを、 終濃度 100 μ g/m 1のサケ精子 DN Aを含むハイプリ ダイゼーション溶液 〔 5 X S S C、 50 %ホルムァミ ド、 50 mM チャーチ リン酸緩衝液 (Chu r c h p h o s p h a t e b u f f e r) (pH 7. 0) 、 10%B l o c k i n g r e a g e n t (ロシュ (Ro c h e) 社製) 、
7%SDS、 0. 1%N—ドデカノィルサルコシン酸ナトリウム〕 中で 65°C、 2時間ィンキュベートすることによりプレハイブリダイゼーションを行つた。 次に、 メンブレンを、 一本鎖に熱変成させたプローブを添加した終濃度 100 μ g/m 1のサケ精子 DN Aを含むハイプリダイゼーション溶液で 50°Cでー晚 インキュベートすることによりハイブリダィゼーションを行なった。
ハイブリダイゼーション後、 メンプレンを洗浄溶液 1 ( 2 X S S C、 0.
1%SDS) 中で室温、 5分間保持し、 結合していないプローブ、 ハイブリダィ ゼーション溶液などを洗レ、落とした。 この洗浄を 2回繰り返した。
次に、 あらかじめ加温しておいた洗浄溶液 2 (0. 1 XSSC、 0. 1%S DS) で 68°C、 15分間保持し、 非特異的結合をしているプローブを解離させ た。 この洗浄を 2回繰り返した。
ジゴキシゲニンの検出は, D I G DNA La b e l i n g a n d D e t e c t i on K i t (ロシュ (Ro c h e) 社製) 付属の説明書に従い、 発 色反応で行った。
5. 二次スクリーニング
上記の一次スクリーニングにて陽性となったファージを回収し、 常法に従って ファージ液を調製し、 二次スクリーニングを行なった。
二次スクリーニングは、 ファージ液を 1プレートにっきプラーク数が 50〜1 00となるように、 あらかじめボトムプレートを作製、 保温しておいた 90 X 2 Omm滅菌シャーレにまいた。 他は一次スクリーニングと同様に行った。
その結果、 2つの陽性クローン、 No. 20および No. 47が得られた。 こ れらの陽性と判断されたプラークについては、 杉浦の方法 〔杉浦昌弘:クロー二 ングとシークェンス、 農村文化社、 東京、 第 152頁〜第 156頁 (1989 年) 〕 に従って、 λファージの増幅と; ファージ DNAの抽出を行なった。
6. えファージ DN Αのサザンハイブリダィゼーション
二次スクリーニングの結果得られた陽性クローン No. 20、 No. 47につ き、 ゲノムライブラリーのスクリーニングに使用したプローブを用いたサザンハ イブリダイゼーションを行なつた。
サザンハイブリダィゼーシヨンは、 マユァテイス (Ma n i a t i s) ら編、 モレキュラー クローニング, ァ ラボラト リー マエユアノレ (Mo 1 e c u 1 a r C l o n i n g, a La b o r a t o r y Ma nu a lノ 、 第 9 、 第 31〜58頁 〔コールド スプリング ハーパー (Co I d S p r i n g Ha r b o r) 社、 1989年刊〕 に記載の方法に準じて行った。
すなわち、 陽性クローンの No. 20および No. 47より抽出した DNAを 種々の制限酵素にて切断を行なった後、 1%ァガロースゲル電気泳動に供し、 泳 動後、 ゲルをアルカリ変性、 中和し、 常法に従い、 DNAをナイロンメンプレン (ハイボンド一 N (Hyb o n d— N) 、 アマシャム フアルマシア パイォテ ク (Am e r s h am Ph a rma c i a B i o t e c h) にー晚力、 けてブロッテイングした。 プロッティング後、 メンブレンに紫外線トランスィル ミネ一ターにて紫外線 (254 nm) を 5分間照射し、 DNAをメンブレンに固 疋した。
このメンプレンを、 2 Om 1の D I G— E a s y— Hy b . Gr a nu l e s (ロシュ (Ro c h e) ネ ± ) 中で 42°C、 2時間インキュベートすることによ り、 プレハイブリダィゼーシヨンを行った。
次に、 メンプレンを 4m 1の上述のハイブリダィゼーション溶液ならびに一本 鎖に熱変成させたプローブとともに 42°Cで一晩インキュベートすることにより ハイブリダイゼーションを行なった。
ハイプリダイゼーシヨン後、 メンブレンを洗浄溶液 1 (2XSSC、 0. 1 %SDS) 中で室温、 5分間保持し、 結合していないプローブ、 ハイブリダィ ゼーション溶液などを洗!/、落とした。 この洗浄を 2回繰り返した。
次いで、 あらかじめ 68 °Cに加温しておいた洗浄溶液 2 (0. 1 XSSC、 0. 1%SDS) で 68°C、 15分間保持し、 非特異的結合をしているプローブ を解離させた。 この洗浄を 2回繰り返した。
メンブレンは、 D I G DNA La b e l i n g a n d De t e c t i o n K i t (ロシュ (Ro c h e) ^M) を用いて処理して発光させ、 乾燥さ せた後、 X線フィルム (コダック (Ko d a k) 社製) を入れたカセット内で 3 0秒感光させ、 オートラジオグラフィーをとつた。
この結果を図 3に示した。

にて、 クローン No. 20またはクローン No. 47をそれぞれ制限酵素処理し たものである。
また、 各レーンの aがクローン No. 20、 bがクローン No. 47である。 その結果、 クローン N o. 20に関しては約 4 k b pの制限酵素 H i n d I I Iと Xb a Iの 2重消化断片に、 クローン No. 47に関しては約 5. 5 k b p の制限酵素 H i n d I I I消化断片に遺伝子およびその上流域が存在する可能性 が高いと考えられた。
そこで、 クローン No. 20の DNAを制限酵素 H i n d I I Iおよび Xb a Iで完全消化した後、 1 %ァガロースゲル電気泳動し、 約 4 k b pのサイズのバ ンドをァガロースゲルから切り出し回収した。
また、 クローン No. 47の DNAを制限酵素 H i n d I I Iで完全消化した 後、 1 %ァガロースゲル電気泳動し、 約 5. 5 k b pのサイズのバンドをァガロ ースゲルから切り出し回収した。
回収したそれぞれ DN A断片をベクタープラスミド pB l u e s c r i p t l I (東洋紡 (T o Y o B o ) 據) に TaKaRa L i g a t i o n K i t v e r . 1 (宝酒造社製) を用いてライゲーシヨンした。 約 30分のライゲーシ ョン後、 得られたライゲーシヨン産物で大腸菌 J Ml 09コンビテントセル (宝 酒造社製) を形質転換し、 該形質転換 JM10.9をアンピシリン添カ卩 L B寒天培 地にて培養し、 コロニーを形成させた。
コロニーが保持するプラスミド中のインサートを、 pB l u e s c r i p t l Iベクター中の配列を有する RVプライマー (宝酒造社製) および M4プライマ 一 (宝酒造社製) でコロニー PCRを行うことによって確認し、 目的の DNA断 片を持つ大腸菌のコロニーを選定した。 選定された大腸菌から Q i a g e n P 1 a s m i d Mi n i K i t (キアゲン (Q i a g e n) 社製) を用いて、 プラスミドを抽出、 精製した。 操作手順は該キットの説明書に従った。 調製した プラスミドは、 それぞれ p EF 20、 pEF47と命名した。
プラスミド p EF 20、 p EF47をそれぞれ用いて大腸菌 J M 109株を形 質転換した。 得られた形質転換体を E s c h e r i c h i a c o l i JM1 09/pEF 20、 E s c h e r i c h i a c o l i J M 109/p E F 4 7 (FERM P— 19160) と命名、 表示した。 これは、 独立行政法人産業 技術総合研究所特許生物寄託センター [日本国茨城県つくば市東 1丁目 1番地 1、 中央第 6 (郵便番号 305-8566) ] に平成 14年 (2002年) 12月 1 8日 (原寄託日) より FERM P— 19160として寄託され、 前記独立行政 法人産業技術総合研究所特許生物寄託センターに FERM BP— 8285 [国 際寄託への移管請求日 :平成 15年 (2003年) 1月 29日] として寄託され ている。
これらのプラスミド p EF 20、 pEF47を羅とし、 Dy e Te rmi n a t i o n Cy c l e S e qu e n c i n g F S Re a dy R e a c t i o n K i t (パーキンエルマ一 (P e r k i nE l me r) ^±M) 、 A B I P r i sm 310 Ge n e t i c a n a l y z e r (パーキンエル マー (P e r k i nE lme r) ネ環) を使用し、 ジデォキシ法を用いたサイク ルシークェンス法によって塩基配列の決定を行った。 操作手順は, 該キットの説 明書に従った。
その結果、 クローン No. 20ファージ DNAには、 A 2遺伝子の翻訳開始点 から 173 b p以降が含まれていることが分かった。 即ち、 ゲノム DNAの上流 の配列が含まれていない為、 プロモーター配列は含まれていないと言える。
—方、 クローン No. 47ファージ DNAには、 A2遺伝子の翻訳開始点から 約 1. 3 k b上流までの配列が含まれていた。 該配列を、 配列表の配列番号 3に 示す。
また、 決定したクローン N o . 47の塩基配列、 即ち配列番号 3と、 配列番号
1の c DNAおよぴクラスタリング解析した E S T配列を比較し、 転写因子検索、 コード領域検索、 イントロン 'ェキソン検索を行なったところ、 クローン No. 47の配列には、 プロモーター配列、 イントロン配列、 ターミネータ一配列が含 まれることが明らかとなった。
即ち、 全長 3764 b pの DNA配列中、 塩基番号 87〜 859の配列が 5 ' 隣接領域 (5, f l a nk i n g r e g i on) 、 塩基番号 860〜 877番 目の配列が第 1ェキソン、 塩基番号 878〜 1331番目の配列が第 1イントロ ン、 塩基番号 1332〜2961番目の配列が第 2ェキソン、 塩基番号 2962 〜 3764番目の配列が 3 ' P舞接領域 (3' f l a nk i n g r e g i o n) であることが明らかになった。 また、 5 ' 隣接領域がプロモーター配列、 3' 隣 接領域がターミネータ一配列であることが明らかになった。
開始コドンを含む塩基番号 1365〜1371の領域にコザック規則 [Ko z a k Μ. ; C e 1 1 , 第 44卷, 第 283頁〜第 292頁 (1986年) 〕 と似た配列 (ATCATGG) が見られた。 一方、 高等植物の核遺伝子のポリ (Α) 配列付加信号 CAYTG 〔ジヨシ C. Ρ. ; Nu c l. Ac i d s
Re s. , 第 15卷, 第 9627〜9640頁 (1987年) 〕 が塩基番号 29 51〜2965の領域に見られ (CATTG) 、 また他のポリ (A) 配列付加信 号 A AT A A Aに類似の配列 A AT AG Aが塩基番号 2897〜2902の領域 に見られた。 また、 上記の検索から、 配列番号 3の配列中の塩基番号 860〜 8 77、 1 3 32〜 29 6 1の配列が A 2遺伝子の c D N A配列であることが明ら力 となった。 該配列を配列表の配列番号 2に示す。 該配列中、 塩基番号 65〜: 1404番目の 配列が A 2ポリぺプチドをコードする配列 (終止コドンを含む) である。 該配列 がコードする 499アミノ酸からなるポリペプチドのアミノ酸配列は、 A1遺伝 子の ORFがコードするポリペプチドのアミノ酸配列 (配列番号 8) と同一であ つた。 このことより、 本実施例で得られた DNA断片もまたェロンゲーシヨンフ アクター (e l o n g a t i o n f a c t o r) として機能するアミノ酸をコ 一ドする DN A断片と考えられる。
実施例 6 プロモータ一配列の活性の確認
(1) プロモーター活性測定用のプラスミドの作製
β一ダルクロニダーゼ遺伝子の一過性発現における GU S活性をみることによ りプロモータ一活性を測定できるように、 以下のプロモータ一活性測定用プラス ミ ドベクターを構築した。
比較コントロールのプラスミドベクターは ρ Β I 22 1 (クロンテック社製) とし、 このベクターの 35 Sプロモーター部分 (H i n d i I Iサイ トから Xb a Iサイトまでの断片) を p EF 47の H i n d I I Iサイトから EF— 1 a A 2遺伝子のィントロンを含まない転写開始直前までのプロモーター配列 (配列 表の配列番号 9の配列) と入れ替えることにより、 プロモータ一配列に β—ダル クロニダーゼ遺伝子を連結したベクターを作製した。 つまり、 配列番号 9の配列 は、 p EF 47プラスミドを铸型に Ml 3 RVプライマー (タカラバイオ製) と 5, 側に X b a Iサイトを持つように合成した配列表の配列番号 1 2に記載の EF 47 X 2プライマーを用いて PC R増幅することにより得られた約 0. 8 k b pのフラグメントを制限酵素 H i n d I I Iと Xb a Iで完全消化し、 約 0. 8 k b pのフラグメントをァガロースゲル電気泳動により切り出して精製し、 入 手した。 この DNA断片を p B I 221から同様に制限酵素 H i n d I I Iと X b a Iで完全消化し 35 Sプロモーター部分をのぞいたベクター断片にライゲー シヨンした後、 E. c o l i JM109に形質転換した。 このプラスミ ドは、 p 47GUS— I Lと命名し、 p 47GUS— I Lで形質転換された E. c o l i JM109は、 E s c h e r i c h i a c o l i JM109/p 47 G US— I Lと命名した。 この p 47GUS— I Lは、 PCRおよび塩基配列解析 により H i n d I I Iサイトから転写開始直前までのプロモーター配列 (配列表 の配列番号 9の配列) をもつことを確認した。
比較コントロールのプラスミドベクター pB I 221の 35 Sプロモーター部 分 (H i n d I I Iサイトから Xb a Iサイトまでの断片) を、 上記のプロモー ター配列 +第 1ェキソン +第 1イントロン +第 2ェキソンの AT G直前までの配 歹 IJ (配列番号 3の 87〜1367番目の配列) と入れ替えることにより、 プロモ 一ター配列 (配列表の配列番号 9の配列) とその下流に第 1イントロン (配列表 の配列番号 10 ) を含む配列に、 β -ダルクロニダーゼ遺伝子を連結したべクタ 一を作製した。 つまり、 上記のプロモーター配列 +第 1ェキソン +第 1イントロ ン +第 2ェキソンの ATG直前までの配列 (配列番号 3の 87〜1367番目の 配列) は、 pEF47プラスミドを鎳型に Ml 3 RVプライマー (タカラバイ ォ製) と 5 '側に X b a Iサイトを持つように合成した配列表の配列番号 13に 記載の EF47Xプライマーを用いて PC R増幅することにより得られた約 1.
3 k b pのフラグメントを制限酵素 H i n d I I Iと X b a Iで完全消化し、 約 1. 3 k b pのフラグメントをァガロースゲノレ電気泳動により切り出して精製し、 入手した。 この DNA断片を p B I 221から同様に制限酵素 H i n d I I Iと Xb a Iで完全消化し 35 Sプロモーター部分を除いたベクター断片にライゲー シヨンした後、 E. c o l i J Ml 09に形質転換した。 このプラスミドは、
P 47GUSと命名し、 p 47GUSで形質転換された E. c o l i JM10 9は、 E s c h e r i c h i a c o l i J M 109Zp 47 GU Sと命名し た。 この p 47 G U Sは、 P C Rおよび塩基配列解析により配列番号 3の 87〜 1367番目の配列をもつことを確認した。
(2) プロモーター活性の確認
パーティクルガンにより p B I 221、 p 47GUS— I L、 p47GUSベ クタ一のスサビノリ TU— 1株への導入を行い、 β一ダルクロニダーゼ遺伝子の 一過性発現における GU S活性を組織化学的ァッセィにより細胞内に蓄積される 青色のィンジゴチン色素の有無を正立顕微鏡で観察することにより、 プロモータ 一活性を測定した。
ベクターを細胞に導入するための金粒子に D N Aをコーティングする手順を以 下に示した。 金粒子 6 Omgを微量遠心チューブ ( 1. 5m l容量) に測り取つ た。 これに 1 m 1の新しく調製された 7 0 %エタノ一ルを加え試験管ミキサー (VORTEX GEN I E 2、 S c i e n t i f i c I n d u s t r i e s ) を用いて 5分間撹拌を行い、 1 5分間室温に放置した。 卓上遠心機 (チビタ ン、 ミリポア) で 5秒間遠心し、 上清を除いた。 これに lm 1の滅菌水を加え試 験管ミキサーで 1分間撹拌した後、 1分間静置して金粒子を沈殿させ、 卓上遠心 機で 2秒間遠心した後、 上清を除いた。 これを 3回繰り返した。 これに金粒子の 終濃度が 6 OmgZm 1となるように 5 0%グリセロールを加えた。 5 0%グリ セロール中の金粒子 (6 Omg/m 1 ) の再懸濁と凝集の破壊のために、 試験管 ミキサ一で 5分間徼しく撹拌した。 この溶液 5 0 μ 1 (金粒子が約 3 m g相当含 まれる) を微量遠心チューブ (1. 5m l容量) に測り取り、 激しく試験管ミキ サ一で撹拌しながら 1 0 μ 1の DNA (1 μ g/μ 1 ) と 5 0 μ 1の 2 · 5Μ C a C 1„ と 20 μ 1の 0. 1M スペルミジンを加え、 さらに 3分間撹拌した。
1分間静置して金粒子を沈ませた後、 卓上遠心機で 2秒間遠心し上清を除いた。 ペレツトを崩さないように 1 40〃 1の 7 0%エタノールを加え、 上清を除き、 次にペレツトを崩さないように 1 40 μ 1の 1 0 0%エタノールを加え、 上清を 除いた。 これに 4 8 μ 1の 1 00%エタノールを加え混合した。 1回の導入には 8 β 1を使用した。
スサビノリの TU— 1株の葉状の配偶体単藻培養株 (以下スサピノリ TU— 1 株とする) を、 1 5°C、 1 0時間明期 (光量 8 0 μ E/m2 / s ;白色蛍光 灯) · 1 4時間暗期の短日条件で、 E S L培地が入った容量 2 Lの簡易式培 養容器 (藤森工業) 中で通気 (ポアサイズ 0. 2 mのフィルターでろ過滅菌 した空気、 0. 2 5 l Zm i n) し、 還流浮遊状態で培養した。 培地は 7日ご とに交換した。 葉状の配偶体を約 2 cm2 の大きさに切断し、 E S L培地で広げ ながら 4分の 1の G 1 a s s M i c r o f i b r e (GF/C : 4 7mm、 W h a tma n) 上に貼り付けた。 これをブフナー漏斗 (適合ろ紙 7 Omm用、 M T) にろ紙 (9 0mm、 Wh a t ma n) を載せて、 E S L培地で湿らせた上に 載せ、 ろ過びんと吸引機を用いて吸引し、 余分の水分を除きつつ吸着させた。 こ れを E S L寒天培地上に置き、 パーティクルガン (G I E— I I I、 タナカ) で DN Aをコーティングした金粒子の導入を行った。 パーティクルガンの操作手順 は、 付属の説明書に従った。 打込み条件は、 発射口と試料との距離は 7〜9 cm、 発射時の圧力は 0. 5〜6. 0k g f /cm2 の範囲で設定し、 発射時間は 0.
015秒で打込みを行い、 1条件ごとに試料 3つ準備しそれぞれに導入を行った。 導入を行った後の試料は、 G l a s s Mi c r o f i b r eから剥がし取り、 導入したベクター別に発射口と試料との距離の条件ごとに集め、 簡易式培養容器 で 15°C、 14時間明期 · 10時間暗期の長日条件で 2日間培養した。 その後、 固定液 〔2% P a r a f o rma l d e hy d e、 0. 1M Na -p h o s p h a t e (p H 7. 0) 、 1 mM EDTA、 1. 5M D—ソルビトー ル〕 で 1時間固定し、 洗浄液 〔0. 1M リン酸ナトリウム (Na_ph o s p h a t e) (pH7. 0) 、 1. 5M D—ソルビトール〕 で 3回洗浄した。 こ れを細胞 ·組織全体が浸るように X_G 1 u c 溶液 〔 2 mM X— G 1 u c (5—ブロモー 4—クロロー 3—インドリル一 j8— D—ダルクロニド、 50mM リン酸ナトリウム (pH 7. 0) 、 1 OmM EDTA、 0. 5 mM K4 F e (CN) 6、 0. 5mM K3 F e (CN) 6、 1. 5M D—ソルビトール、 0. 5% Tr i t on X- 100) を加え、 適当な容器に入れ密封した後、 37°Cィンキュベータ (M I R— 253、 三洋) 中でー晚ィンキュベートした。 —ダルク口-ダーゼ活性 (GUS活性) は細胞内に蓄積される青色のインジゴ チン色素の有無を正立顕微鏡で観察することにより確認し、 これをプロモーター 活性とした。
その結果、 ρΒ I 221ベクターを導入した試料について発射口と試料との距 離が 9 cmの条件の中で 2つ GUS活性が確認された。 同様に p 47GUS— I Lベクターについては、 発射口と試料との距離が 8 cmと 9 c mの条件でそれぞ れ 1つの合計 2つ GUS活性が確認された。 したがって、 このベクターのプロモ 一ター (配列表の配列番号 9 ) がプロモータ一活性があることが確認された。 p 47 GUSベクターについては、 発射口と試料との距離が 7 cmの条件で 1 つ、 8 c mと 9 c mでそれぞれ 3つの合計 7つの G U S活性が確認された。 また、 GUS活 I"生が見られた試料について、 細胞の色素を脱色したところ、 発 射口と試料の距離が 9 cmで p 47 GUSベクターを一過性に導入した 12の圧 力条件 (0. 5〜6. OKg/cm2) の 1つが、 約 1週間経過しても青色のィ ンジゴチン色素の蓄積が残っていた (図 4) 。 図 4において、 Aは脱色前、 Bは 脱色 7日後の結果を示す。 したがって、 このベクターのプロモーター (配列表の 配列番号 9) はその下流に第 1イントロン (配列表の配列番号 10) を含むこと により効率よく遺伝子発現が可能でありかつ非常に高い発現の誘導が可能と考え られた。 産業上の利用の可能性
本発明により、 藻類に由来するプロモーターが初めて提供され、 該プロモータ 一を利用した藻類における遺伝子発現が可能となった。 さらに、 本発明により、 藻類に由来するターミネータ一、 イントロンが初めて提供され、 これらを利用し て藻類における遺伝子発現効率の向上が可能となった。 即ち、 本発明は、 藻類に おける有用物質の遺伝子工学的生産を可能とするのみならず、 藻類の遺伝子工学 的育種を可能とするものである。
また、 本発明により、 藻類に由来する新規ェロンゲーシヨンファクターの遺伝 子が提供され、 該遺伝子を利用して藻類に由来するェロンゲーシヨン

( j ) 配列表の配列番号 1に示される塩基配列を有する DNAまたはその一部、 (k) ( j ) の DNAまたはその相補鎖と厳密な条件下でハイブリダィズし得る

17. 配列番号 8のァミノ酸配列をコ一ドする D N A。
18. 請求項 17に記載の DNAに、 1以上の塩基の置換、 欠失、 挿入、 付加 を有し、 力つ、 エロンゲーションファクター (e l on g a t i on f a c t o r) 活性を有するポリペプチドをコードすることを特徴とする DNA。
