Abstract

Die vorliegende Erfindung beschreibt ein Verfahren zum Nachweis von Nukleinsäuresequenzen, dadurch gekennzeich-net, dass folgende Schritte ausgeführt werden: a) mindestens eine Nukleinsäure wird auf einer Festphase gebunden; b) Sondenmoleküle werden sequenzspezifisch an die Nuk-leinsäure hybridisiert, wobei die Sondenmoleküle mit ei-ner spaltbaren Bindung und einer Massenmarkierung verse-hen sind, welche für das Sondenmolekül spezifisch ist; c) Entfernung der nicht hybridisierten Sondenmoleküle; d) Kontaktieren der hybridisierten Sondenmoleküle mit ei-ner Matrix, die besagte spaltbaren Bindungen spaltet und zugleich als Matrix in einem MALDI-Massenspektrometer dient; e) Nachweis der Massenmarkierung an den Positionen, an denen die Nukleinsäure gebunden wurde.
