技術分野
本発明は、 Very Late Antigen— 4 (VLA— 4)に対する優れた阻害作用と安全 性を有する化合物の製造中間体として有用な化合物の製造方法に関するものである 背景技術
[0002] VLA— 4は、単球、リンパ球、好酸球および好塩基球に発現して!/ヽる細胞接着関連 分子であり、血管細胞接着分子ー1 (Vascular cell adhesion molecule— 1 ;VC AM— 1)等に対する受容体として働 、て 、ることが知られて 、る。
[0003] 近年、 VLA— 4と VCAM— 1によって介在される接着の選択的な阻害力 自己免疫 疾患およびアレルギー性炎症疾患治療の解決手段となり得ることが報告されている。
[0004] 特許文献 1の一般式 (I)に記載の化合物、例えば、下記の式(1)で表される化合物 は、優れた VLA - 4阻害作用に基づく抗炎症作用を示し、かつ高い安全性を有する 医薬ィ匕合物として期待されている。(特許文献 1参照)。

[0013] この上記式(2)の化合物のうち、 R2が前記アルコキシ基またはァラルキルォキシ基 である化合物は、置換、無置換のニトロフ ニル酢酸のカルボン酸部位をエステル化 した後、ニトロ基を還元してァミノ基に変換し、さらに、 R1を部分構造に有するカルボ ン酸またはエステルを縮合させてアミド化することにより製造できることが知られている (特許文献 1、 2参照)。
しかし、特許文献 1および 2に記載の製造方法では、ニトロ基の還元反応において 、鉄金属が用いられていたため、操作性、反応性において問題があった。また、上記 工程中のアミドィ匕反応は、一般的な塩基性条件で行われていたため、副反応によつ て、 R1を部分構造に有するカルボン酸の二量体が生成し、それにともなって主反応 の収率低下が生じるという問題があった。さらには、生じた二量体を除去するための カラクロマトグラフィーによる精製が必要であった。
また、式 (2)の化合物のうち、 R2が式 (a)である化合物も、 R1を部分構造に有する力 ルボン酸またはエステルとァミノフエ-ル酢酸アミドとを縮合させて製造されて 、る。こ のアミドィ匕反応もまた、一般的な塩基性条件で行われていたため、上記と同様の問 題があった。
[0014] また、上記式(13)で表される化合物のうちで、 R5が水素原子であり X1および Y1が 塩素原子である化合物については、 2, 5—ジクロロー-トロトルエンのメチル基の臭素 ィ匕、次いで 2, 5—ジクロロー 4—-トロベンジルブ口ミドをシァノ化して 2, 5—ジクロロー 4 -トロフエ-ルァセトニトリルに導き、最後にシァノ基の加水分解で 2, 5—ジクロロー 4 一二トロフ ニル酢酸に導く方法による製造例が報告されているが、本製造方法は多 工程を要する問題を有している (非特許文献 1参照)。
[0015] 上記式(13)で表される化合物に類似する化合物として、 1, 4ージクロロー 2 フルォ 口ベンゼンを-トロ化し、次いでマロン酸と反応させて、 2, 5—ジクロロー 4—-トロフエ ニルマロン酸エステル誘導体を製造する方法が報告されて ヽる (特許文献 3参照)。 しかし、このマロン酸と反応させる方法は、多工程を有し、高価な 1, 4—ジクロロ— 2— フルォロベンゼンを用い、工業規模における操作が困難な水素化リチウムを用いると いう問題を有している。

[0017] ニトロ基に対してパラ位に目的の置換基を導入する-トロベンゼンの反応として、種 々の-トロアレーンと種々の 2—クロ口プロピオネートエステルとを反応させて、 -トロア リールプロピオネートを合成する方法が記載されている(非特許文献 4参照)。この反 応には、置換されていないかまたはハロゲン原子等で 1箇所置換された-トロベンゼ ンを用い、ハロゲン原子で 2箇所置換された-トロベンゼンの反応に関する記載はな い。

発明が解決しょうとする課題
[0018] 本発明は、優れた VLA— 4阻害作用と安全性を有する化合物の製造中間体として 重要な化合物である、上記式(2)および(13)に示した化合物を得るために工業的に 有利な製造方法を提供するものである。
課題を解決するための手段
[0019] 本発明者は鋭意研究を進めた結果、上記式 (2)で表される化合物の製造工程に おいて、ニトロ基の還元反応をマイルドな白金系触媒下で行うことにより、ベンゼン環 上のハロゲンを還元することなくニトロ基のみを選択的に還元できることを見出した。 さらには、還元物を塩酸塩の結晶として得ることにより該還元物の安定性を向上でき ることを見出した。
また、アミド化反応において、通常の塩基を添加した条件下ではなぐ酸性条件下 において、酸クロライドを生成し得る化合物とアミンを添加して反応させることで、アミ ド化が定量的に進み、化合物(2)が高収率で得られることを見出した。
[0020] さらにまた、上記式(13)で表される化合物の製造工程において、容易に入手可能 な置換ニトロベンゼンに塩基存在下、エステル誘導体を導入することによって、 VLA 4ZVCAM— 1相互作用による細胞接着を阻害する化合物の中間体( 13)をーェ 程で安価に得る方法を見出した。さらに驚くべきことに、生成化合物のうちの九割とい う高い選択性で、ニトロベンゼン環の窒素基に対してパラ位に置換基が導入できるこ とを見出した。

[0087] 本発明者は、 R2bに対応するアルコール溶媒中、白金系還元触媒下に水素化する ことによって、式 (4)の化合物の-トロ基のみを選択的に還元できることを見出した。 これによつて、操作性、反応性に問題があった鉄粉の使用が回避できるとともに、塩 素の脱離反応が抑えられて、高収率で式 (5— 1)で表される化合物を得ることが可能 となった。
反応は、出発物質である式 (4)で表される化合物または式(5 - 2)で表される化合 物を式 (4)中の R2bに対応するアルコール溶媒に溶解し、白金系還元触媒下で行う。 白金系還元触媒としては白金、 Pt— S—炭素等が挙げられ、 Pt— S—炭素が特に好ま しい。 Pt— S—炭素は、式 (4)で表される化合物の重量に対し 5%— 50%の重量で添 加することが好ましぐ 10%— 20%で添加するのがさらに好ましい。

加水分解試薬としては、酸、アルカリ共に使用できるが、ナトリウム、カリウム、リチウ ム等のアルカリ金属水酸化物またはマグネシウム、カルシウム等のアルカリ土類水酸 化物が挙げられる。この中で、水酸化ナトリウム、水酸ィ匕カリウムが好ましい。
反応温度は室温一溶媒の沸点の範囲が望まし 、。
反応時間は、 30分一 5時間で良ぐ通常は 1時間一 2時間で完結する。

(1)フラスコに、 (4 アミノー 2, 5—ジクロロフヱ-ル)酢酸 ェチルエステル 塩酸塩(1 0g, 35. lmmol)を入れ、メタノール(30mL、 3vZw)及び 2規定水酸化ナトリウム 水溶液(37.5mL、 3.75vZw)をカ卩え、約 70°Cで 1時間撹拌した。 HPLCにて原 料消失を確認し反応液を放冷した。
濃塩酸(10mL、 lvZw)を加え結晶を析出させ、 1時間氷冷した後、析出した結晶 を吸引濾過した。結晶を水 30mLで洗浄した後、 40°Cで減圧乾燥し、 4 アミノー 2, 5 ージクロロフ ニル酢酸(7.3g、収率 95%)を得た。

[0190] (2—1)フラスコに、 (4 アミノー 2, 5—ジクロ口フエ-ル)酢酸(6g、 27.3mmol)、トラ ンスー 4 [ (2S, 4S)— 4ーメトキシピロリジン 2 ィル)メトキシシクロへキサン— 1 カル ボン酸ェチルエステル(14.4g、 1.02モル当量)、 1ーヒドロキシベンゾトリアゾール( 4. 43g、 1. 2モル当量)、ァセトニトリル(120mL、 20vZw)を加え、撹拌下トリェチ ルァミン(5. 3mL、 1. 4モル当量)を室温にて添カ卩した。溶解を確認後、 1ーェチルー 3—(3—ジメチルァミノプロピル)カルボジイミド塩酸塩(7. 33g、 1. 4モル当量)をカロえ て撹拌した。 HPLCにて反応終了確認後、水(80mL、 15vZw)を添加し撹拌した。 種晶を加え結晶を析出させ、しばらく撹拌し氷冷した。
析出した結晶を吸引濾過し、結晶を水:ァセトニトリル混液(1 : 1) 10mL、水 20mL で順次洗浄し 40°Cにて減圧乾燥を行い、式(26)の化合物 1晶を 10. 5g (収率 79% 、品質 99%)で得た。
濾液を減圧下濃縮し、酢酸ェチルで抽出する。抽出液を減圧下濃縮乾固し、ァセト 二トリル 25mL加えて撹拌し、水 20mLを添カ卩し種晶をカ卩ぇ結晶を析出させ、しばらく 撹拌し氷冷する。析出した結晶を吸引濾過し、結晶を水:ァセトニトリル混液(1: 1) 2 mL、水 4mLで順次洗浄し 40°Cにて減圧乾燥を行い、式(26)の化合物 2晶を 1. 8g (収率 14%、品質 96%)を得た。

Olg, 31. 9mol)及びブタノール(4. 52g, 33. 4mmol)のジメチ ル酢酸(75mL)懸濁液にトリェチルァミン(6. 66mL, 47. 8mmol)を加え、反応液 が均一形になった後に 1ーェチルー 3— (3—ジメチルァミノプロピル)カルポジイミド塩 酸塩(6. 37g, 33. 2mmol)を加えた。室温で 4時間攪拌した後に水(225mL)を徐 々に滴下し、さらに室温で 3時間攪拌した。析出している結晶を濾取し、 75%含水ジ メチル酢酸(50mL)で洗浄した後に減圧乾燥し、式(26)の化合物(13. 90g,純度 97. 1%,純度換算単離収率 95. 6%)を微黄色結晶として得た。

Ommol)のァセトニトリル( 70mL)溶液に DMF (70mg)及び塩化チォ -ル(3. 07mL, 42. 7mmol)を加え、 室温で 30分攪拌した後に減圧下外温 50°Cで溶媒を留去した。濃縮残渣にァセトニ トリル(75mL)及びトランス 4— [ (2S, 4S)— 1 (4 アミノー 2, 5—ジクロ口フエ-ル) ァセチルー 4ーメトキシピロリジシ 2 ィル]メトキシシクロへキサン 1一力ルボン酸 ェ チルエステル(13. Og, 26. 7mmol)を加えて内温 65°Cで 2時間攪拌した。室温に 冷却してァセトニトリル(50mL)及び水(130mL)を徐々に滴下し、室温で終夜攪拌 した。析出している結晶を濾取し、 50%含水ァセトニトリル(50mL)で洗浄した後に 減圧乾燥し、式(24)の化合物(15. 24g,純度 94. 3%,純度換算単離収率 86. 1 %)を白色結晶として得た。

[0196] 第三級ブドキシナトリウム(76. 6g、 0. 797mol)を N, N—ジメチルァセトアミド(250 mL)で室温下溶解後、— 10°Cに冷却した。この溶液に、 2, 5—ジクロ口-トロベンゼン (50g、 0. 260mol)とクロ口酢酸ェチル(42. 5mL、 0. 399mol)の N, N—ジメチル ァセトアミド(lOOmL)混合溶液を滴下し、滴下終了後 1時間撹拌を行った。反応終 了後、酢酸ェチルエステル(500mL)で希釈し、 2規定の塩酸 (400mL)をカ卩えて分 液、酢酸ェチルエステル(250mL)にて抽出した。有機層を合わせ、 2規定の塩酸( 200mL)、水(200mL)、飽和食塩水(200mL)の順で洗浄後、減圧濃縮により暗 褐色油状物 87. Ogを得た。この暗褐色油状物は、化合物(30— a)と化合物(30— b) の混合物であった (化合物(30— b)を約 10%含有)。得られた残渣に 2—プロパノー ル(105mL)と水(35mL)を加えた。一晩撹拌後、析出した結晶をろ過し、化合物(3 0— a) (44. 7g、 0. 161mol、収率 59. 6%、 HPLC純度 100%)を黄色の結晶として 得た。

[0198] ナトリウム tert—ブトキシド(2. 88g)をジメチルァセトアミド(18ml)に氷冷下溶解し 、 2, 5—ジクロ口-トロベンゼン(1. 92g)、クロ口酢酸 tert—ブチルエステル(2. 26g) のジメチルァセトアミド(18ml)溶液を滴下し、内温 5°C以下に保ったまま 2時間攪拌 した。該溶液に氷冷下、 2N塩酸水溶液 23mlを加え、 tert—ブチルメチルエーテル 2 Omlにて 3回抽出し、抽出液を飽和食塩水で洗浄後、硫酸マグネシウムで乾燥しろ 過した後、溶媒を留去した。得られた残渣をシリカゲルカラムクロマトグラフィーに付し 、油状物質として式(31)の化合物を 2. 65g (86%)得た。

リチウム tert—ブトキシド(2. 4g)をジメチルァセトアミド(18ml)に氷冷下溶解し、 2, 5—ジクロロ-トロベンゼン(1. 92g)、クロ口酢酸 tert—ブチルエステル(2. 26g)のジ メチルァセトアミド(18ml)溶液を滴下し、内温 5°C以下に保ったまま 2時間攪拌した。 実施例 1と同様の後処理をした結果、標題化合物 2. 71g (88%)を得た。
