Abstract

Verfahren zur fluoreszenzbasierenden kinetischen Messung von Transglutaminase-Aktivität, dadurch gekennzeichnet, dass die Transglutaminate mit einem modifizierten Peptidsubstrat, bei dem ein Fluorophor direkt an den N-Terminus des Peptids gebunden ist und ein Glutaminsäurerest in 5-Position einen Fluoreszenzquencher trägt, welcher über einen Spacer, der eine Aminfunktion enthält, unter Ausbildung einer Amidbindung an die 5-Position gebunden ist, umgesetzt wird, so dass durch die Aktivität der Transglutaminase der Fluoreszenzquencher abgespalten wird, und die resultierende Fluoreszenz des Restmoleküls gemessen wird.
