BAFF抑制剤又は阻害剤のスクリーニング方法
技術分野
[0001] 本発明は、新規な BAFF抑制剤又は阻害剤のスクリーニング方法に関する。詳細 には、本発明は、市販の細胞株に対して適切な刺激条件を加え、細胞外に検出可 能な充分量の BAFFを安定して遊離する系を構築することにより、 BAFFの発現又 は産生を抑制又は阻害する物質をスクリーニングする方法に関する。さらに詳細には 、本発明は、培養ヒト T細胞に、 TPAとアイオノマイシン(ionomycin)との組み合わせ 及び Z又は抗 CD3抗体を添加して、当該細胞の BAFFの産生を誘発する方法、及 び培養ヒト T細胞に、 TPAとアイオノマイシン (ionomycin)との組み合わせ及び Z又は 抗 CD3抗体を添カ卩した BAFFの産生系に、被検物質を添カ卩して、当該 BAFF産生 系における BAFFの発現量及び Z又はその活性量を測定すること力もなる、 BAFF の発現又は活性を制御することができる物質をスクリーニングする方法、並びに TPA とアイオノマイシン (ionomycin)との組み合わせ及び Z又は抗 CD3抗体を含有してな る BAFF産生細胞に対する BAFFの産生誘発剤に関する。
背景技術
[0002] BAFF(B cell activating factor belonging to the TNF family)は、 T細胞、単球 Zマ クロファージ、榭状細胞等から産生'分泌され、 B細胞上の 3種の受容体を介して B細 胞の分化、活性化、生存等を制御することが知られている(Mooreら Science(1999)28 5,260-263)。
[0003] ヒト BAFFは、 285個のアミノ酸力もなる膜貫通型の蛋白質である。そのアミノ酸配 列中には、 46個のアミノ酸力もなる細胞質ドメインと 218個のアミノ酸力もなる細胞外 ドメインが存在し、また、 Nグリコシルイ匕部位が 2ケ所有り、 3量体を形成するといつた 構造的な特徴を有する。さらに C末端から 152アミノ酸の細胞外ドメインは、 Furinファ ミリ一のプロテアーゼによって切断され、可溶型として遊離される。ヒト BAFFのァミノ 酸配列は当初、 Neutrokine aとの名称で、国際特許出願公開 W098Z18921号公 報中の配列番号 1又は 2として開示された。この他別名称として、 Kay、 TNFSF13B 、 Blys、 TALL— 1、 THANK, zTNF4等も知られている。
[0004] BAFFの受容体としては、 BAFF— R、 TACI(transmembrane activator and calciu m modulator and cyclophilin ligana interactor)及び BCMA(B cell maturation antigen )が知られている。 BAFF— R及び BCMAは主に B細胞に発現しており、 TACIは B 細胞と活性化 T細胞に発現して ヽる。
[0005] BAFFの生理的作用は、上述のように B細胞の分化、活性化、生存等の制御であ る力 近年、病態との関わりも次第に明らかとなった。即ち、 BAFFを過剰に発現する マウスでは、末梢血 B細胞の増多、リンパ節や脾臓の肥大、血清中 IgG濃度上昇、抗 核抗体産生、腎内に免疫複合体の沈着、蛋白尿、腎炎等の、 SLE様の症状を示す ことが報告されている(Mackayら J.Exp.Med.,(1999)190,1697- 1710、 Khareら Pro Na tl.Acad.Sci.USA (2000)97,3370-3375)。さらにこのマウスは、加齢とともに唾液腺炎 、唾液腺破壊等の、 SS様の症状を示すことも判明した (Groomら J.Clin.Invest.(2002) 109,59-68)。また SLE、 RA、 SSの患者において、血清中 BAFF濃度が増加してい ることも報告されており(Zhangら J.Immunol.(2001)166,6— 10、 Cheemaら Arthritis Rhe um.(2001)44,1313- 1319、 Groomら J.Clin.Invest.(2002)109,59- 68)、さらには RA患 者の滑液中の BAFFレベルが血清中に比べて高いこと(Cheemaら Arthritis Rheum 2001)44,1313-1319)や、 SS患者の唾液腺浸潤白血球における BAFFの発現(Groo mら J.Clin.Invest.(2002)109,59-68)、 SLE患者における血清 BAFFレベルとィムノグ ロブリンゃ抗 dsDNA抗体との相関性(Zhangら J.Immunol.(2001)166,6-10)、 RA患者 における BAFFとリウマトイド因子との相関性(Cheemaら Arthritis Rheum.(2001)44,l 313-1319)等、数多くの報告がある。
[0006] これらの事実から、 BAFFの発現又は産生を抑制又は阻害することは SLE、 RA、 SS等の自己免疫疾患の予防 '治療に繋がるものということができるため、 BAFFの発 現又は産生の抑制又は阻害を的確にスクリーニングする系の確立が望まれている。 そのようなスクリーニング系の確立には、例えば、遺伝子組換え技術により、 BAFF を発現するよう構築した遺伝子ベクターを培養細胞に導入することにより、安定して B AFFを発現する細胞を得る方法が考えられる。しかし、 BAFFが発現したとしても、 分泌型として細胞外に遊離されるとは限らないし、また BAFF発現、産生において安 定な細胞株とすることができるかという問題を有する。このため、従来、上記のような 系は全く報告されて 、な 、。例えば市販の細胞株に対して適切な刺激条件を加える ことにより、細胞内における BAFF発現の誘導と細胞外に検出可能な充分量の BAF F遊離が認められる系を構築できれば、上記のような問題も解決するといえる。
発明の開示
[0007] 本発明の目的は、市販の細胞株に対して適切な刺激条件を加え、細胞外に検出 可能な充分量の BAFFを安定して遊離する系を構築することにより、 BAFFの発現 又は産生を抑制又は阻害する物質をスクリーニングする方法を提供することにある。 またそのような系を構築することにより、全身性エリテマトーデス (SLE)、慢性関節リウ マチ (RA)、シヱーダレン症候群 (SS)、自己免疫性糖尿病、 AIDS又は B細胞の活性 化を伴う自己免疫疾患等の自己免疫疾患の予防'治療剤のスクリーニング方法をも 提供できる。
[0008] そこで本発明者らは、細胞株の選定や刺激の方法などについて鋭意検討した結果 、 CD3を高発現して!/、る市販のヒト T細胞株である Loucy細胞又は Jurkat細胞に対し 、抗 CD3抗体及び Z又は TPA(12- 0- tetradecanoylphorbol 13- acetate、又は Phorb ol 12-myristate 13— acetate : PMAと表記することもある)とアイオノマイシン(ionomyci n)を組み合わせて刺激することにより、細胞において安定した BAFF発現誘導が認 められ、且つ、培養液中に検出可能な充分量の BAFFが安定して遊離されることを 見出し、本発明を完成した。

発明を実施するための最良の形態

即ち、市販のヒト T細胞を適切な培地に適切な濃度で懸濁し、培養プレート等に播 種し、 TPA (終濃度として 0. 001〜1. 、好ましくは 0. 005〜0. 1 μ & mL)及びアイオノマイシン(ionomycin) (終濃度として 0. 01〜: L0. 0 μ g/mL,好ま しくは 0. 05〜: L Ο /z gZmL)を添加して刺激し、さらに必要に応じて被験物質 (終 濃度 10_15〜10_3Mにおける、任意の濃度勾配列)を添加し、培養を開始する。培 養開始後 1時間〜 7日後、好ましくは 3時間〜 5日後に培養上清を回収し、上清中の BAFF濃度を測定し、これを対照と比較することで、本スクリーニング系における被験 物質の BAFF抑制又は阻害活性を調べることができる。また、上述の系を実施する にあたり、予め培養プレート等を PBS等で希釈した抗 CD3抗体 (終濃度として 0. 01 〜1000 μ g m 好ましくは 0. 1〜: L00 μ g/mL)にてコーティングしたものを用 いてもよい。また抗 CD3抗体にてコーティングした培養プレート等を用いる場合は、 T PAとアイオノマイシン(ionomycin)を用いなくても充分量の BAFFを検出することがで き、より簡便なスクリーニング系として有用である。
[0019] 本発明で用いるヒト T細胞株は、巿販のものが好ましぐ例えば、 Loucy細胞 (ATCC
Cat. No. CRL- 2629)、 Jurkat細胞(ATCC Cat. No. TIB- 152)、 TALL- 1細胞(JCRB 0086)が挙げられ、このうち T細胞受容体を介した細胞活性ィ匕が予想できる、 CD3を 高発現している Loucy細胞(ATCC Cat. No. CRL- 2629)、 Jurkat細胞(ATCC Cat. N o. TIB- 152)がさらに好ましい。
[0020] また、本発明で用いる抗 CD3抗体としては、当業者に公知の方法で作製された抗 ヒト CD3モノクローナル抗体、抗ヒト CD3ポリクローナル抗体が挙げられる力 このう ち抗ヒト CD3モノクローナル抗体が好ましく、マウス抗ヒト CD3モノクローナル抗体(B D Pharmingen Cat No. 555336、等)がさらに好ましい。
[0021] また、本発明で用いる TPAは、 12- 0- tetradecanoylphorbol 13- acetateの略称であ り、別名称として Phorbol 12-myristate 13-acetate (PMAと表記することもある)として 知られて!/、るホルボールエステルであるが、本発明で使用するものとしては!、ずれの 表記による化合物でもよぐまた立体異性体をも包含する。好ましい市販の TPAとし ては、例えば 4- a - Phorbol 12-myristate 13- acetate(SIGMA Cat. No.P- 148)が挙げ られる。
[0022] さらに、本発明で用いるアイオノマイシン(ionomycin)は、 Streptomyces conglobatus 由来のポリエーテル系抗生物質であり、カルシウムィオノホアとしての作用が知られ ているが、本発明で使用するアイオノマイシン (ionomycin)としては、製薬的に許容さ れる塩も含まれる。好ましい市販品のアイオノマイシン(ionomycin)としては、例えば Io nomycin, calcium salt (SIGMA Cat. No. 10634)が挙げられる。
[0023] 本発明の BAFFの産生誘発剤は、 BAFFの産生を誘発する活性成分として、 TPA とアイオノマイシン (ionomycin)との組み合わせ及び Z又は抗 CD3抗体を含有してな ることを特徴とするものである。本発明の BAFFの産生誘発剤は、前記した活性成分 のほかに緩衝液などの細胞系に添加可能な担体を含有することができ、このような細 胞系に添加可能な担体と共に BAFFの産生誘発用組成物とすることができる。 [0024] 本発明の BAFFの産生誘発剤は、 BAFFを産生する細胞、好ましくは培養細胞に 適用することができ、好ましい細胞としては、培養ヒト T細胞、より具体的には Loucy細 胞ゃ Jurkat細胞が挙げられる。
[0025] 本発明の BAFFの産生誘発剤は、活性成分として TPAとアイオノマイシン(ionomy cin)との組み合わせを用いる場合には、それぞれの活性成分が、 TPAは終濃度とし TO. 001〜1. O /z g/mL、好ましく ίま 0. 005〜0. 1 g/mLになるような量を、ァ ィオノマイシン(ionomycin)は終濃度として 0. 01 μ gZmL〜10 μ g/m 好ましく は 0. OS /z gZmL l /z gZmLになるような量を、細胞系に添カ卩して使用することが できる。また、活性成分が、抗 CD3抗体の場合には、活性成分の濃度が 0. 1 μ gZ mL〜100 μ g m 好ましくは 1 μ gZmL〜30 μ gZmLの緩衝液溶液を培養プ レートに敷いて使用することができる。
[0026] 以上のごとく得られる本発明の BAFFの産生系は、後記の実施例に示すように、培 養液中に検出可能な充分量の BAFFが安定して遊離されるという驚くべき特徴を有 するので、 BAFFの発現又は活性を制御することができる物質、好ましくは抑制又は 阻害することができる物質のスクリーニング方法はもとより、全身性エリテマトーデス (S LE)、慢性関節リウマチ (RA)、シエーダレン症候群 (SS)、自己免疫性糖尿病、 AIDS 又は B細胞の活性化を伴う自己免疫疾患等の自己免疫疾患の予防'治療剤のスクリ 一二ング方法を実現することができる。

以下に、実施例を挙げてこの発明をさらに具体的に説明するが、この発明の技術 的範囲はこれらの実施例に限定されるものではない。
実施例 1
[0028] Loucy細胞又は Jurkat細胞を抗 CD3抗体又は TPAとアイオノマイシン(ionomycin)に より刺激した時の BAFFの発現
(a) TPAとアイオノマイシン (ionomycin)による刺激培養
ATCC (American Type Culture Collection)より購入した Loucy細胞又は Jurkat細 胞を 10%FBS (Fetal bovine serum), 0. 45%D— glucose, ImMピルビン酸ナトリ ゥム、 ImMHEPESを含む RPMI1640培地に懸濁し、 1 X 106cells/wellになるよう に、 6 well培養プレートに播種した。これと同時に TP Aとアイオノマイシン(ionomycin) を最終濃度がそれぞれ 0. 05 μ g/mL、 0. 5 μ gZmL又は 0. 1 μ gZmLとなるよう に添加し、培養開始後 5時間、 24時間、 48時間、 72時間(Loucy細胞)又は 48時間 、 120時間、 168時間(Jurkat細胞)で細胞を回収した。これら回収した細胞から total RNAを抽出し、次の条件下において RT— PCR法により BAFFの mRNAの解析を 行った。 ReverTra Dashキット(TOYOBO Cat. No. PCR- 401)を用い、 30°C 10分、 42°C 20分、 99°C 5分の反応過程を経て、逆転写反応を行い、 cDNAを得た。こ の cDNAを、 Advantage 2 PCR Enzyme system(BD Biosciences Cat. No. PT3281— 1) を用い、 94°Cで 1分反応後、 94°C 30秒、 66°C 1. 5分を 35サイクル反応させ、 66 °C 5分の過程を経て、獲得した DNAを 1. 5%ァガロースゲル電気泳動により分離 し、 975bpの位置に目的のバンドが認められた。 PCR反応に使用したヒト BAFFのプ ライマー配列は次の通りである。
Forward: 5 aggccccaaccttcaaagttcaag (酉己列番号 1)
Reverse: o cttagaggtacagagaaagggagg (酉己列番号 2)
この結果を図 1に示す。図 1の(a)は Loucy細胞の場合を示し、図 1の(b)は Jurkat細 胞の場合を示す。また、 T+Iは TP Aとアイオノマイシン (ionomycin)による刺激を示し ( (a)にお!/、て ίまそれぞれ 0. 05 μ g/mL、 0. 5 g/mLであり、(b)にお!/、て ίまそ れぞれ 0. 05 μ g/mL, 0. 5 μ g/mL (向力つて右佃 J)又 ίま 0. 1 μ g/mL (向力つ て左側)である)、(一)は無刺激を示す。さらに、各々の図における G3PDHは陽性コ ントロールを示す。図 1 (a) 、 (b)に示されるように、 TPAとアイオノマイシン(ionomyc in)刺激を受けた Loucy細胞、及び Jurkat細胞において、刺激後 5時間、及び 2日に おいて BAFF mRNAの発現が認められ、 TPAとアイオノマイシン(ionomycin)による BAFF mRNAの明らかな発現誘導は培養開始 72時間、及び 7日(168時間)後ま で継続して認められた。従って、 Loucy細胞又は Jurkat細胞を TPAとアイオノマイシン (ionomycin)によって刺激培養することにより、安定した BAFFの発現誘導が認めら れることから、この培養系を用いて、 BAFF発現抑制、あるいは阻害物質のスクリー- ングが可能となることがわ力つた。
(b)抗 CD3抗体による刺激培養 PBSにより 10 μ gZmLに希釈した抗 CD3抗体を 6ゥエル培養プレートにしき、 4°C 一晩で底面吸着させた後、(a)と同様に懸濁した Loucy細胞を 1 X 106cells/wellにな るように播種した。培養開始後 5時間、 24時間、 48時間、 72時間で細胞を回収した 。これら回収した細胞から total RNAを抽出し、次の条件下において RT—PCR法に より BAFFの mRNAの解析を行った。 ReverTra Dashキット(TOYOBO Cat. No. PC R- 401)を用い、 30°C 10分、 42°C 20分、 99°C 5分の反応過程を経て、逆転写 反応を行い、 cDNAを得た。この cDNAを、 Advantage 2 PCR Enzyme system(BD Bi osciences Cat. No. PT3281- 1)を用い、 94°Cで 1分反応後、 94°C 30秒、 66°C 1. 5分を 35サイクル反応させ、 66°C 5分の過程を経て、獲得した DNAを 1. 5%ァガ ロースゲル電気泳動により分離し、 975bpの位置に目的のバンドが認められた。この 結果、図 1の (a)の右側に anti-CD3として示されるように、刺激後 5時間力も BAFF m RNAの発現が認められ、その後減衰し、培養開始 72時間後に再度発現する 2相性 を示すことが明らかとなった。このような発現様式はそれぞれに生物学的意義が異な る可能性があると考えられ、 Loucy細胞を抗 CD3抗体によって刺激培養することによ り誘導された BAFF発現を用いて、さらにユニークな BAFF発現の抑制、あるいは阻 害物質のスクリーニングが可能となると考えられる。
実施例 2
Loucy細胞を抗 CD3抗体又は TPAとアイオノマイシン(ionomycin)により刺激した時 の BAFF蛋白質の発現
PBSにより 10 μ gZmLに希釈した抗 CD3抗体を 6ゥエル培養プレートにしき、 4°C 一晩で底面吸着させた後、実施例 1と同様に懸濁した Loucy細胞を 1 X 106cells/well になるように播種した。培養開始後 24時間、 48時間、 72時間、 96時間で細胞を回 収した。これら回収した細胞を用いて、定法のウェスタンプロット法にて BAFF蛋白質 の発現を検討した。なお、 BAFF蛋白質検出抗体として、ゥサギ抗ヒト BAFFポリクロ ーナル抗体 (AB16530、 Chemicon社製)を用いた。この結果を図 2に示す。図 2の 左側の(―)は刺激を行わなかった場合をコントロールとして示し、図 2の CD3 stimula tionは抗 CD3抗体で刺激した場合を示し、 TPA+Ionomycinは TPAとアイオノマイシン (ionomycin)とで刺激した場合を示す。図 2の下段の β - actinは陽性コントロールを示 す。図 2に示されるように、抗 CD3抗体による刺激を受けた Loucy細胞おいて、刺激 後 24時間力もヒト BAFFに該当する 34kDaのバンドが確認され、この明らかな発現 誘導は、培養開始 96時間まで継続して認められた。一方、 Loucy細胞を 10%FBS ( Fetal bovine serum), 0. 45%D— glucose、 ImMピルビン酸ナトリウム、 ImMHEP ESを含む RPMI1640培地に懸濁し、 1 X 106cells/wellになるように、 6 well培養プレ ートに播種し、これと同時に TPAとアイオノマイシン (ionomycin)を最終濃度がそれぞ れ 0. 0. 5 gZmLとなるように添カ卩し、培養開始後 24時間、 48時間 、 72時間、 96時間において細胞を回収した。これら回収した細胞を用いて、定法の ウェスタンプロット法にて BAFF蛋白質の発現を検討した。この結果、図 2に示される ように、 TPAとアイオノマイシン(ionomycin)による刺激を受けた Loucy細胞おいて、 刺激後 24時間力もヒト BAFFに該当する 34kDaのバンドが確認され、この明らかな 発現誘導は、培養開始 96時間まで継続して認められた。これらの結果より、 Loucy細 胞を抗 CD3抗体又は TPAとアイオノマイシン (ionomycin)によって刺激培養すること により、安定した BAFF蛋白の発現誘導が培養開始 96時間後まで認められ、この培 養系を用いて、 BAFF蛋白質発現抑制、あるいは阻害物質のスクリーニングが可能と なることがわかった。
参考例 1
抗ヒト BAFFモノクローナル抗体(4H4)の作製
実施例 3に用いる抗ヒト BAFFモノクローナル抗体 (4H4)は、以下のようにして作製 した。即ち、配列表の配列番号 3に示した BAFFの 285個のアミノ酸から細胞外ドメイ ンの膜近傍に該当する 13アミノ酸 (配列番号 4)を選択し、 MBS法を用いて KLHとコ ンジュゲートし、抗原とした。 100 1の lmgZmL抗原ペプチド生理食塩水溶液を、 フロイント完全アジュバントでェマルジヨン化し、マウス(BalbZc, 6週齢)腹腔内に免 疫した。 2週間後に 100 1の lmgZmL抗原ペプチド生理食塩水溶液、フロイント完 全アジュバントを超音波処理によってェマルジヨンィ匕したものを追加免疫し、以降 2週 間毎に追加免疫を 2回行った。初回免疫後 2ヶ月目に脾臓を摘出し、 RPMI 1640 培地 (ペニシリン,ストレプトマイシン入り)中でリンパ球を分離した。分離したリンパ球 をポリエチレングリコール(PEG)法によりマウス骨髄腫由来のミエローマ細胞 P3U1 株と融合させ、ハイプリドーマ細胞を作製した。ハイプリドーマ細胞をフィーダ一細胞 入りの HAT培地に懸濁し 96穴プレート(Greiner)に分注し 15日間培養した。ハイブ リドーマ細胞を培養したゥェルカ 培養上清を回収し、 ELISA法(enzyme-linked im munosorbent assay)によって抗原ペプチドと反応がある抗体産生細胞を選択した。即 ち、まず 96穴プレートに 50 μ ΚΌΙΟ μ gZmL抗原ペプチドをしき、 4°C、ー晚底面に 吸着させた後、 100 1の 2%BSAZPBSで 37°C、 2時間ブロッキングさせた。ハイブ リドーマ細胞の上清 100 /z lを 4°C、 1晚反応させた後、 HRP標識抗マウス IgGを 100 0倍希釈して 37°C、 1時間反応させ、オルトフエ-レンジアミンを基質に用いて発色さ せた。 50 1の 2N硫酸で反応を停止させた後、 492nmの吸光度を測定して 1. 0以 上の値が得られたノヽイブリドーマを選択し,限界希釈法によるクローユングを行った。
[0032] 7日前、 3日前にそれぞれ 0. 5mlのプリスタンを腹腔内投与したマウス(BalbZc) に、選択したハイプリドーマ細胞を腹腔内注射し、約 10日後に腹水を採取した。採取 した腹水は室温で 30分おいた後、 4°Cでー晚静置し、 15Krpm、 10分間遠心して上 清を回収し、マウス IgG画分をプロテイン A-Sepharoseカラムにて分離精製した。 以上の方法にて、抗ヒト BAFF抗体 (4H4、アイソタイプは IgG 1)を産生するハイブリ ドーマ細胞株、並びに該抗体 (4H4)を得た。得られた 4H4、及び対照として Chemico n製ゥサギ抗ヒト BAFFポリクローナル抗体 (AB16530 :図 3中、 Chemと表記)を、常法 のウェスタンブロッテイング法にてリコンビナントのヒト BAFF(Chemicon製)の検出に 用いた結果、図 3に示すように可溶性ヒト BAFFに該当する 17KDaのバンドが確認さ れた。
実施例 3
[0033] Loucy細胞又は Jurkat細胞を TPAとアイオノマイシン(ionomycin)により刺激した時の BAFFの産生
実施例 1と同様に、懸濁した Loucy細胞又は Jurkat細胞を 1 X 106cells/wellになるよ うに、 6 well培養プレートに播種した。これと同時に TPAとァィォノマィシン(ionomyci n)を最終濃度がそれぞれ 0. 05 μ g/m 0. 5 g/mLとなるように添カ卩し、培養 開始後 5時間、 24時間、 48時間、 72時間(Loucy細胞)又は 1、 4、 7日(Jurkat細胞) で培養上清を回収した。これらの培養上清中に含有される細胞から産生された BAF Fを、市販のゥサギ抗ヒト BAFFポリクローナル抗体(Chemicon社 Cat. No. AB16530 )と上記参考例 1で作製したマウス抗ヒト BAFFモノクローナル抗体 (4H4)を用いた E LISA法にて測定した。 ELISA法は次に記載した方法で行った。即ち、 1次抗体とし てゥサギ抗ヒト BAFFポリクローナル抗体(Chemicon社 Cat. No. AB16530 )を用い、 96穴プレートに 1 gZwellで 4°Cー晚コーティングした。 0. 05%Tween20を含む P BSで 3回洗浄後 BlockAce (大日本製薬社)を 150 /z LZwellカ卩え、 37°C、 2時間反 応させた。 0. 05%Tween20を含む PBSで 3回洗浄後、検体 50 L及びピオチン 標識した 4H4を SO /z SngZmL)加え、室温にて 2時間反応させた。 0. 05%Twe en20を含む PBSで 3回洗浄後、 0. 05%Tween20を含む PBSで 1000倍に希釈し たストレプトアビジン標識 HRP (Horse RAdish peroxidase)を 50 μ L添加し、室温に て 30分反応させた。 0. 05%Tween20を含む PBSで 5回洗浄後 TMB One Solutio n(Clonetech社)を 50 μ Lカロえ、 5分間室温にて反応させた後、 IN HC1 50 μ Lを添 加後プレートリーダー (PerkinElmer製)で 450nmの吸光度を測定した。標準物質とし てリコンビナントのヒト BAFF (Chemicon社 Cat. No. GF119)を用い、標準曲線から 検体中の BAFF濃度を測定した。
[0034] Loucy細胞を用 、た場合の結果をグラフ化して図 4に示し、 Jurkat細胞を用 V、た場 合の結果を同様にグラフ化して図 5に示す。それぞれのグラフにおける縦軸は BAFF 濃度(ngZmL)を示し、それぞれの棒グラフの左側は TPAとアイオノマイシン (ionom ycin)による刺激を行わな力つた場合の BAFF濃度をコントロールとして示して 、る。 この結果、図 4及び図 5に示されるように、 TPAとアイオノマイシン (ionomycin)刺激を 受けた Loucy細胞又は Jurkat細胞において、刺激後 5時間又は 1日力 培養上清中 の BAFFが検出でき、培養開始 72時間又は 7日まで継続して認められた。これらの 結果より、 Loucy細胞又は Jurkat細胞を TPAとアイオノマイシン(ionomycin)によって 刺激培養することにより、安定した BAFF産生誘導が認められ、この培養系を用いて 、 BAFF産生抑制物質、あるいは阻害物質のスクリーニングが可能となることが判明 した。
産業上の利用可能性
[0035] 本発明は、全身性エリテマトーデス (SLE)、慢性関節リウマチ (RA)、シエーダレン症 候群 (SS)、自己免疫性糖尿病、 AIDS又は B細胞の活性ィ匕を伴う自己免疫疾患等 の自己免疫疾患との関係が明らかにされてきている BAFFの産生を培養ヒト T細胞に おいて誘発する方法を提供し、自己免疫疾患の治療や予防に有効な新しい物質を スクリーニングするための手法を提供するものであり、産業上の利用可能性がある。

Claims

請求の範囲
[I] 培養ヒト T細胞に、 ΤΡΑとアイオノマイシン (ionomycin)との組み合わせ及び Z又は 抗 CD3抗体を添加して、当該細胞の BAFFの産生を誘発する方法。
[2] 培養ヒト T細胞力 Loucy細胞である請求項 1に記載の方法。
[3] 培養ヒト T細胞力 Jurkat細胞である請求項 1に記載の方法。
[4] 培養ヒト T細胞に、 TPAとアイオノマイシン (ionomycin)との組み合わせ及び Z又は 抗 CD3抗体を添カ卩した BAFFの産生系に、被検物質を添カ卩して、当該 BAFF産生 系における BAFFの発現量及び Z又はその活性量を測定すること力もなる、 BAFF の発現又は活性を制御することができる物質をスクリーニングする方法。
[5] BAFFの発現又は活性の制御力 BAFFの発現又は活性の抑制又は阻害である 請求項 4に記載の方法。
[6] 培養ヒト T細胞力 Loucy細胞である請求項 4に記載の方法。
[7] 培養ヒト T細胞力 Jurkat細胞である請求項 4に記載の方法。
[8] 培養ヒト T細胞に、 TPAとアイオノマイシン (ionomycin)との組み合わせ及び Z又は 抗 CD3抗体を添カ卩した BAFFの産生系に、被検物質を添カ卩して、当該 BAFF産生 系における BAFFの発現量及び Z又はその活性量を測定することからなる、全身性 エリテマトーデス (SLE)、慢性関節リウマチ (RA)、シエーダレン症候群 (SS)、自己免 疫性糖尿病、 AIDS又は B細胞の活性化を伴う自己免疫疾患等の自己免疫疾患の 予防'治療剤のスクリーニング方法。
[9] 培養ヒト T細胞力 Loucy細胞である請求項 8に記載の方法。
[10] 培養ヒト T細胞力 Jurkat細胞である請求項 8に記載の方法。
