Abstract

 プレートを回転させて生体サンプルを移動させる際に、回転の中心の外周側から内周側に生体サンプルを容易に移動できる生体サンプル分析プレートを提供する。  回転の中心に対して内周側に設けたチャンバー部(16)と、外周側に設けられたサンプル保持部(20)を流路(17),(18)で連結し、サンプル保持部(20)があらかじめ内部に空気を保有しており、プレートの回転による遠心力で生体サンプルがチャンバー部(16)からサンプル保持部(20)に向けて移動するときにサンプル保持部(20)内の空気がサンプル保持部(20)内に圧縮保持されるようにした。

技術分野
[0001] 本発明は、 DNAやタンパク、その他の生体サンプルを、緩衝剤中で移動させ、そ の輸送反応を検出して、生体サンプルを判別する生体サンプル分析用プレートに関 する。
背景技術
[0002] 一般的な生体サンプルを考えた場合、大きくは DNAとタンパクが存在して 、る。そ して、近年、分子生物学の急速な進展によって、様々な疾患において遺伝子の関与 力 Sかなり正確に理解されるようになり、遺伝子をターゲットにした医療に注目が集まる ようになってきている。 DNAに関して、現在 SNPs (single nucleotide polymorp hism、の略で「1塩基多型」と一般に訳されており、遺伝子における 1暗号(1塩基)の 違いの総称である。)が注目されている。その理由としては、 SNPsの分類により、多く の疾患に対する罹患率や、各個人の薬剤に対する効果や、感受性を予測でき、さら には、地球上に親子兄弟と!/、えども全く同じ SNPsを持つ人間は絶対に存在しな 、こ とから、個人の完全な特定ができると考えられて 、るからである。
[0003] 現在 SNPsを調べる方法としては、 DNAの塩基配列を端から直接読んでいくシー ケンシング (塩基配列の決定)が、最も一般的に用いられている。そして、前記シーケ ンシングを行う方法としては、いくつかの報告がある力 もっとも一般的に行われてい るのは、ジデォキシシーケリング(Sanger法)である。なお、シーケンシングは、この S anger法を含め、何れの方法においても、分離能の高い変性ポリアクリルアミドゲル 電気泳動か、キヤビラリ一電気泳動によって 1塩基長の長さの違いを分離'識別する 技術が基になって成り立つている。
[0004] また、他の方法として、ァフィ二ティリガンドキヤビラリ一電気泳動法がある。
[0005] ァフィ-ティリガンドキヤビラリ一電気泳動は、分子間親和力、とくに生態系における 特異的親和力 (酵素と基質、抗原と抗体の親和力等)を利用して分離に特異性を持 たせるものであり、具体的には、キヤビラリ一管中の泳動溶液に、塩基配列を特異的 に認識するァフィ-ティリガンドを添加しておき、試料を電気泳動させると、試料混合 物中で相互作用する分子種だけが移動速度に変化を生じることに着目して分析を行 うものである(例えば、特許文献 1参照)。
[0006] 一方、タンパク質は、細胞、組織、生体液中に存在し、生体活動の調節、細胞への エネルギー供給、重要な物質の合成、生物構造体の維持、さらには細胞間でのコミ ュ-ケーシヨンや、細胞内情報伝達に関与している。現在では、タンパク質が様々な 環境や、相互作用する他のタンパク質の存在、タンパク質が受けた修飾の程度や、 種類に応じて、複数の機能を有することが明らかになってきている。
[0007] タンパク質は、 20種類のアミノ酸力 遺伝子の指示 (配列情報)により順番につなが ることでつくられており、その種類は数千万種と言われるが、その遺伝子の配列がわ かれば、どのアミノ酸が、どういう順番でつながつてできている力、の情報を得ることが できる。生物の遺伝子 (ゲノム)から作られるタンパク質の一そろいのセットは、プロテ オームと呼ばれる力 ヒトゲノムの塩基配列解読が終わった今、プロテオームの解析 が盛んに進められている。
[0008] そして、このようなタンパク質の機能解析研究としては、同定や、キャラクタリゼーショ ンのみならず、生化学アツセィや、タンパク質間相互作用研究、タンパク質ネットヮー ク、または、細胞内外のシグナリング解明、なども行っていく必要がある。このタンパク 質機能の研究には、多方面の技術が使用され、酵素アツセィ、酵母 two— hybrid アツセィ、クロマトグラフィーによる精製、情報ツールとデータベース等がある力 特に 、電気泳動によるたんぱく質の判別は、重要な手法である。そして、電気泳動のよう に、キヤビラリ一管中のサンプル、分析物、緩衝剤、及び試薬等の液体を移動させた 際に得られる輸送反応を検出して、該サンプルの分析、判別、判定等を行う場合の、 前記液体の輸送、及び方向付けに関しては、さまざまな報告がある(例えば、特許文 献 2〜特許文献 5)。

発明の開示
発明が解決しょうとする課題
[0009] 上述したように生体サンプルの分析にぉ 、ては、キヤピラリー電気泳動装置を利用 した方法が、広く使われている。実際に輸送反応を行なう部分であるキヤビラリ一は、 外形 300ミクロン、内径 100ミクロン程度のガラス管が使用される例が多ぐ折れにくく するために、表面は、ポリイミド等でコーティングされている。
[0010] しかしながら、内部試料を検出するために、検出窓として焼いたり、薬品で溶かすな どして、コーティングを一部剥ぎ取る。この時、コーティングを剥いだ部分は、折れや すくなり、取り扱いに十分注意する必要がある。折れた場合は、なお危険である。
[0011] また、サンプルの注入においても、加圧や、吸引で行なうのが一般的な方法である 力 一定量注入する必要があり、時間で調整するものの、キヤピラリー内の緩衝剤の 粘度や、温度変化により、注入量が実験のたびに異なるという問題がある。サンプル の量は、測定結果に及ぼす影響は大きぐ大変重要な項目である。
[0012] また、このようなキヤピラリーを使用した装置の場合、構成上短い流路での電気泳動 を行なうことは困難であり、必要以上の泳動距離と、時間をかけて測定することになる
[0013] さらに、プレート上の流路を利用した場合でも、試料の分離を行うのに、例えば、特 許文献 5、特許文献 6に記載の方法では、複数のキヤビラリーチヤンネルを交差させ 、且つ少なくとも 3つの電極を設けて、該少なくとも 3つの設けられた電極のうちの 2つ の電極に印加して、前記交差部を通して前記試料を移動させているが、この方法で は、流路が交差していることから、試料を電気泳動させる際にうまく泳動しない可能性 があり、正確な測定結果が得られな 、と 、う問題がある。
[0014] また、例えば、特許文献 3,特許文献 4に記載の方法では、プラットホームに、微細 チャンネルを埋設し、該プラットホームの回転速度を変化させることで、該回転から生 じる向心力を変化させて試料を移動させている力 この方法では、試料を向心方向 にし力移動させることができないという問題にカ卩え、該微小チャンネルの形状力 かな り複雑であるという問題もある。 [0015] 本発明は、上記のような従来の課題を解決するためになされたもので、流路に充填 された緩衝剤中で、生体サンプルを移動させた際の輸送反応を検出する時に、短時 間で正確な検出結果が得られ、取り扱いも容易で、且つ小型、軽量で、安価な生体 サンプル分析用プレートを提供することを目的とする。
課題を解決するための手段
[0016] 前記課題を解決するために、本発明にかかる生体サンプル分析用プレートは、マイ クロ流路が形成され、該マイクロ流路を流れる生体サンプルを、光学的に、あるいは 電気化学的に分析する構成を有してなる生体サンプル分析用プレートにおいて、該 生体サンプル分析用プレートの回転中心に対して内周側に設けた第 1チャンバ一と 、外周側に設けた第 2チャンバ一とが、第 1マイクロ流路により連結されて、前記生体 サンプルが、該第 1マイクロ流路を介して前記第 1チャンバ一と前記第 2チャンバ一と の間で移動可能となっており、前記第 2チャンバ一は、あらかじめ内部に空気を保有 しており、該生体サンプル分析用プレートの回転により生ずる遠心力によって、前記 生体サンプルが前記第 1チャンバ一力も前記第 2チャンバ一に向けて移動するとき、 前記第 2チャンバ一内の空気は該第 2チャンバ一内に圧縮保持される、構成を有す ることを特徴とする。
[0017] さらに、本発明にかかる生体サンプル分析用プレートは、前記第 2チャンバ一の前 記第 1チャンバ一と連通する開口部は、前記第 2チャンバ一の外壁のうちの該分析用 プレートの外周側にある部分に形成されている構成を有する。
[0018] さらに、本発明に力かる生体サンプル分析用プレートは、前記第 2チャンバ一に連 結された前記第 1マイクロ流路に、さら〖こ、第 2マイクロ流路が連結されて設けられ、 前記第 2チャンバ一に保持される生体サンプルが、前記第 1マイクロ流路、および前 記第 2マイクロ流路を介して移動可能である構成を有する。
[0019] さらに、本発明に力かる生体サンプル分析用プレートは、前記第 2マイクロ流路に 連結されて、第 3チャンバ一が設けられ、該第 3チャンバ一は、前記生体サンプル分 析用プレートの回転中心から、前記第 1チャンバ一よりも近い位置に配置され、前記 生体サンプルが、前記第 2チャンバ一から前記第 2マイクロ流路を介して前記第 3チ ヤンバーに移動可能である構成を有する。 [0020] さらに、本発明に力かる生体サンプル分析用プレートは、前記第 1のマイクロ流路、 および前記第 2のマイクロ流路により形成される試料供給用流路とその一部が接し、 該接する部分でサンプル定量保持部を形成する緩衝剤供給用流路は、該生体サン プル分析用プレートの重心を中心とする円弧部分を有する構成を有する。

発明の効果
[0025] したがって、本発明によれば、生体サンプル分析用プレートを回転させることにより 、外周側に設けた保持手段が保持する生体サンプルを、遠心力を利用して、定量保 持部へ移動させることができる。
[0026] また、本発明によれば、遠心力を利用した生体サンプル判別にぉ 、て、第 2の流路 を生体サンプル分析用プレートの重心を中心とする円弧形状とすることにより、生体 サンプルを電気泳動させる第 2の流路のほぼ全体を、スキャンすることができる。
[0027] またこれにより、第 2の流路の途中や、第 2の流路の終端の、反応状況を知ることが できるようになり、反応の過程を観察することにより、極めて正確な観察結果を得るこ とが可能となる。

[0031] 本発明は、生体サンプルを緩衝剤中で移動させて、生物学的、酵素的、免疫学的 、及び化学的反応を行い、生体サンプルを容易、かつ安価に、精度よぐ短時間で、 判別することを実現するものである。
[0032] なお、本実施の形態 1では、説明を具体的にするために、生体サンプルが DNAサ ンプルで、緩衝剤が、分離用 DNAコンジュゲート、及び DNA結合制御剤を含むも のであるとし、本生体サンプル分析用プレートが、流路中に充填させた分離用 DNA コンジュゲート中に、定量された DNAサンプルを添カ卩して電気泳動させ、流路中の 蛍光度、あるいは吸光度を検出して、 DNAサンプルの SNPs (—塩基多型)の有無 を判別するものとする。

[0049] また、フィルム 37が、生体サンプル分析用プレート 100の全面に貼られているのに 対して、フィルム 27は、電極部 30の近傍のみに、それぞれ貼られている。
[0050] それでは、以下、前述した DNAサンプルの SNPs (—塩基多型)の有無を判別する までの具体的操作、および動作の一例を、図 2を用いて説明する。
[0051] まず、検体となる DNAサンプルを準備する。
[0052] 本来 DNAは、 2本鎖の螺旋構造をしたものである力 本実施の形態 1にお!/、ては、 判別した 、SNPs部位を含む約 60塩基長の 1本鎖 DNAを準備する。

[0054] 次に、緩衝剤として、 DN Aコンジュゲートを準備する。
[0055] DNAコンジュゲートとは、 6〜12塩基長1本鎖DNAの5'末端に、高分子のリニア ポリマーが共有結合したものである。さらに DNAは、正常型に対しては相補であるが 、変異型に対しては相補ではない配列であり、正常型 DNAに対しての結合力が強く 、変異型 DNAに対しての結合力が弱い特性がある。また、電気泳動した場合、 5'末 端に結合したリニアポリマーがおもりとなり、泳動速度がかなり遅いという特性もある。
[0056] 以後に記述する「DNAコンジュゲート」とは、電解質の役目もする pH緩衝剤、およ び MgC12などの DNA結合力制御剤を含んだ物性とする。
[0057] 試料の準備が終わったところで、 DNAコンジュゲート、および DNAサンプルを、プ レート内へ注入する。 DNAコンジュゲートは、ピぺッター等により、定量を、緩衝剤注 入口 6から緩衝剤注入部 7へ分注する。 DNAサンプルも、同様に、定量を、サンプル 注入口 8よりサンプル注入部 9へ分注する。
[0058] 分注量としては、パターンのスケールにより異なる力 本実施の形態 1においては、 DNAコンジュゲートは、 18マイクロリットル、 DNAサンプルは、 2マイクロリットルとす る。
[0059] 次に、モータ等に、生体サンプル分析用プレート 100を固定し、重心 5を軸に、回 転させる。この時、分注された DNAコンジュゲートと、 DNAサンプルは、遠心力によ り、外周方向へと移動する。
[0060] 緩衝剤注入部 7内の DNAコンジュゲートは、流路 10と、流路 11を通り、正電極部 1 2と、負電極部 13へ等分され、正電極部 12へ移動した DNAコンジュゲートは、さら に、流路 14を通り、サンプル定量部 23まで移動する。負電極部 13へ移動した DNA コンジュゲートも、同様に、さらに、流路 14を通り、サンプル定量部 23まで移動する。 [0061] 回転開始から 2分後、 DNAコンジュゲートの移動が停止した状態を、図 6 (a)に示 す。また、サンプル定量部 23周辺を拡大した図力 図 6 (b)である。
[0062] DNAコンジュゲート 31は、正電極部 12、および負電極部 13の、 7割程度まで充填 されており、流路 14を満たし、図 6 (b)に示すように、サンプル定量部 23まで達してい る。
[0063] 正電極部 12と、負電極部 13、内に存在する DNAコンジュゲートの液面高さと、サ ンプル定量部 23の液面高さは、重心 5を中心とする同一円周上となる。
[0064] 次に、注入された DNAサンプルの移動について、図 2、および図 7を用いて説明す る。
[0065] サンプル注入部 9内の DNAサンプルは、流路 15を通り、チャンバ一部 16、さらに は、流路 17、流路 18を通り、外周側に位置するサンプル保持部 20にまで達する。こ のとき、チャンバ一部 16内にあった空気は流路 50, 19を通って抜ける。しかしながら 、図 7に示すように、流路 18は、サンプル保持部 20の外周側に接続され、かつサン プル保持部 20には、空気抜き用の穴が存在しない。そのため、サンプル保持部 20 内に残った空気は、抜けることはなぐ加圧された状態となる。
[0066] 図 7が、回転開始から 2分後の DNAサンプル 32と、 DN Aサンプル 33の状態を示 すものである。空気 34は、圧縮されており、回転中のみ、このような遠心力と、加圧力 とが、平衡となった状態を維持する。
[0067] 本実施の形態 1では、回転数は、 4000rpmである。
[0068] それでは、次の動作を説明する。
[0069] 生体サンプル分析用プレート 100を、一定時間回転させ、 DNAコンジュゲートと、 DNAサンプルの移動が停止した状態で、回転を急停止させる。例えば、 2秒で、 40 OOrpmより、停止までに、至らしめる。
[0070] サンプル保持部 20内に存在していた DNAサンプル 32は、遠心力が無くなり、空 気 34の圧力により、サンプル保持部 20から流路 18へ、逆流しょうとする。
[0071] さらに、 DNAサンプル 32は、サンプル保持部 20内の空気 34が大気圧となるまで 流路 17、流路 19へと移動しょうとするが、この時、流路 17は、流路 19にくらべ細く形 成されているため、 DNAサンプル 32は、流路 17よりも、流路 19へ多く流れようとする [0072] そこで、急停止した直後の状態を、図 8に示す。
[0073] サンプル保持部 20内の DNAサンプル 32は、空気 34の膨張により、流路 18、流路 19を通り、サンプル定量部 23、およびバッファ部 21にまで達する。
[0074] 特に、サンプル定量部 23では、充填された DNAコンジュゲート 31と接することにな る。このとき、流路 17を通り、チャンバ一部 16へ移動する DNAサンプル 32も、多少 存在する。
[0075] 次に、生体サンプル分析用プレート 100を、中速にて数秒間もう一度回転させる。こ の時減速を緩やかにすることが重要である。停止後の状態を、図 9に示す。
[0076] 流路 19に充填されていた DNAサンプル 32は、チャンバ一部 16へと流れるが、サ ンプル定量部 23〖こは、 DNAサンプル 32が微量残存する。残存した DNAサンプル 3 2aは、他の DNAサンプル 32b, 32cに対して、電気的にも絶縁された状態である。
[0077] また、このように、 2回目の回転時に、 1回目の回転時と異なる速度で回転をさせる 理由は、その 2回目の回転速度を、遅ぐかつ減速度を緩やかにすることによって、サ ンプル保持部 20内の空気 34が強く圧縮されることがないようにするためである。した がって、サンプル保持部 20内の DNAサンプルが逆流して、流路 19が再度充填され ることちない。
[0078] 以上の動作を行ったのちに、サンプル定量部 23に残存したサンプル DNA32が、 SNPsの判別を行なう最終試料となる。
[0079] 次に、電気泳動を行なわせる。電気泳動は、正電極部 12に正電極、負電極部 13 に負電極を挿入し、数百 Vの電圧を印加する。すると、流路 14、さらにはサンプル定 量部 23にお!/、て電界が発生し、サンプル定量部 23に一定量残存した DNAサンプ ル 32は、流路 14中を、正電極側(図 9中、 A方向)へ泳動する。
[0080] 流路 14中には、 DNAコンジュゲートが充填されており、 DNAサンプル 32は、 DN Aコンジュゲートとの結合を繰り返しながら電気泳動する。この時、上述したように、 D NAサンプル 32中の正常型 DNAは DNAコンジュゲートとの結合力が強いため、泳 動速度が遅くなり、変異型 DNAはその結合力が弱いため、正常型に比べ泳動速度 は速くなる。つまり DNAサンプル中に、正常型、変異型の両方が存在した場合は、 正常型の DNAと、変異型の DNAとが分離していくこととなり、 SNPsの判別を行うこ とがでさる。
[0081] 図 11は、図 10に示す流路 14の円弧部分 40を、 DNAサンプルが電気泳動中に、 該 DNAサンプル 32をスキャンしたグラフを示す図である。
[0082] DNAの検出は、蛍光標識(FITC)を修飾した DNAを、 470nmの光で励起し、 52
Onm付近の光検出を行うことにより行なった。ここで、 DNAの検出は、 260nmの吸 光度により行なってもよい。
[0083] 図 11において、横軸が、円弧部分 40の位置を表し、 DNAは、左から右へ泳動す る。つまり、左側がサンプル定量部 23で、右側が正電極部 12側である。縦軸は、蛍 光強度で、上から 1分毎に計時変化した波形を表す。徐々に、 2つの山が分離してい く様子がわかる。この場合、 DNAサンプル 32中には、同量の正常型 DNAと、変異 型 DNAが存在して ヽることが判定された。
[0084] グラフ右側の山力 泳動速度が速 、ので変異型、左側が、遅く泳動して!/、るので正 常型となる。
[0085] 以上のように、本実施の形態 1による生体サンプル分析用プレート 100によれば、 生体サンプルを緩衝剤中で移動させた際に得られる輸送反応を検出して生体サン プルの判別を行う生体サンプル分析用プレート 100にお 、て、緩衝剤が流れる第 2 の流路 90と生体サンプルが流れる第 1の流路 80との合流部分に、一定体積の生体 サンプルを保持するサンプル定量部 23を設け、該サンプル定量部 23に生体サンプ ルを供給するようにするとともに、該サンプル定量部 23への生体サンプル供給用の 通路 18に、その開口部 20aを、該生体サンプル分析用プレート 100の外周側に有す るサンプル保持部 20を、該通路 18に接続して設けることによって、回転動作のみに よって、生体サンプル分析用プレート 100上のサンプル定量部 23に DNAサンプル 3 2を一定量保持し、これを、正電極部 12と負電極部 13と流路 14とによって電気泳動 させることができ、さらには、緩衝剤が流れる第 2の流路 90である電気泳動する上記 流路 14の一部を円弧形状としたことにより、細胞や血液等力も特定の DNAを取り出 した DNAサンプルを、本生体サンプル分析用プレートにおいて測定する際に、極め て精度の良い検出結果を得ることが可能となる効果がある。 産業上の利用可能性
本発明に力かる生体サンプル分析用プレートは、 DNAサンプル等の生体サンプル の判別を、安価で、且つ簡便に行えるようにするものとして、非常に有用である。

Claims

請求の範囲
[1] マイクロ流路が形成され、該マイクロ流路を流れる生体サンプルを、光学的に、ある いは電気化学的に分析する構成を有してなる生体サンプル分析用プレートにおいて 該生体サンプル分析用プレートの回転中心に対して内周側に設けた第 1チャンバ 一と、外周側に設けた第 2チャンバ一とが、第 1マイクロ流路により連結されて、前記 生体サンプルが、該第 1マイクロ流路を介して前記第 1チャンバ一と前記第 2チャンバ 一との間で移動可能となっており、
前記第 2チャンバ一は、あらかじめ内部に空気を保有しており、該生体サンプル分 析用プレートの回転により生ずる遠心力によって、前記生体サンプルが前記第 1チヤ ンバーから前記第 2チャンバ一に向けて移動するとき、前記第 2チャンバ一内の空気 は該第 2チャンバ一内に圧縮保持される、
ことを特徴とする生体サンプル分析用プレート。
[2] 請求項 1に記載の生体サンプル分析用プレートにお 、て、
前記第 2チャンバ一の前記第 1チャンバ一と連通する開口部は、前記第 2チャンバ 一の外壁のうちの該分析用プレートの外周側にある部分に形成されている、 ことを特徴とする生体サンプル分析用プレート。
[3] 請求項 1に記載の生体サンプル分析用プレートにお 、て、
前記第 2チャンバ一に連結された前記第 1マイクロ流路に、さらに、第 2マイクロ流路 が連結されて設けられ、
前記第 2チャンバ一に保持される生体サンプルが、前記第 1マイクロ流路、および 前記第 2マイクロ流路を介して移動可能である、
ことを特徴とする生体サンプル分析用プレート。
[4] 請求項 2に記載の生体サンプル分析用プレートにお 、て、
前記第 2チャンバ一に連結された前記第 1マイクロ流路に、さらに、第 2マイクロ流路 が連結されて設けられ、
前記第 2チャンバ一に保持される生体サンプルが、前記第 1マイクロ流路、および 前記第 2マイクロ流路を介して移動可能である、 ことを特徴とする生体サンプル分析用プレート。
[5] 請求項 4に記載の生体サンプル分析用プレートにお 、て、
前記第 2マイクロ流路に連結されて、第 3チャンバ一が設けられ、
該第 3チャンバ一は、前記生体サンプル分析用プレートの回転中心から、前記第 1 チャンバ一よりも近 、位置に配置され、
前記生体サンプルが、前記第 2チャンバ一から前記第 2マイクロ流路を介して前記 第 3チャンバ一に移動可能である、
ことを特徴とする生体サンプル分析用プレート。
[6] 請求項 3ないし 5のいずれかに記載の生体サンプル分析用プレートにおいて、 前記第 1のマイクロ流路、および前記第 2のマイクロ流路により形成される試料供給 用流路とその一部が接し、該接する部分でサンプル定量保持部を形成する緩衝剤 供給用流路は、該生体サンプル分析用プレートの重心を中心とする円弧部分を有す る、
ことを特徴とする生体サンプル分析用プレート。
[7] 請求項 1ないし 6のいずれかに記載の生体サンプル分析用プレートであって、 該生体サンプル分析用プレートは、抗原抗体反応分析用のものである、 ことを特徴とする生体サンプル分析用プレート。
[8] 請求項 1ないし 6のいずれかに記載の生体サンプル分析用プレートであって、 該生体サンプル分析用プレートは、血液成分分析用のものである、
ことを特徴とする生体サンプル分析用プレート。
[9] 請求項 1ないし 6のいずれかに記載の生体サンプル分析用プレートであって、 該生体サンプル分析用プレートは、遺伝子分析用のものである、
