Abstract
本発明は、光学活性2-置換プロパナール誘導体の製造方法、より詳細には、2-置換アクロレイン誘導体の炭素-炭素二重結合を、その炭素-炭素二重結合を立体選択的に還元する能力を有する酵素源を用いて立体選択的に還元することにより、光学活性2-置換プロパナール誘導体を製造する方法に関する。
本発明によれば、医薬品や甘味料等の中間体として有用な光学活性2-置換プロパナール誘導体、とりわけ光学活性2-アルキルプロパナール誘導体を、安価で入手容易な原料から簡便な方法で製造することができる。

背景技術
[0003] 光学活性 2 置換プロパナール誘導体の製造法として、以下の様な方法が知られ ている。
1)プロリン誘導体を不斉補助基として用い、プロパナールを不斉アルキルィ匕すること により、光学純度 94%eeの (R)— 2—メチルペンタナールを合成する方法 (非特許 文献 1)。
2)ベアウベリア'スルフレセンス (Beauveria Sullurescens)を用いて 2—メチル 2—ぺ ンテナールを微生物還元して光学活性 2—メチルー 1 ペンタノールを取得し、これ を酸化して光学活性 2—メチルペンタナールを合成する方法 (非特許文献 2)。 非特許文献 1 : New Journal of Chemistry, 24(12), 973-975 (2000)

[0005] 上記に鑑み、本発明の目的は、医薬品や甘味料等の中間体として有用な光学活 性 2—置換プロパナール誘導体、中でも光学活性 2—アルキルプ口パナール誘導体 を安価で入手容易な原料から簡便に製造する方法を提供することにある。
課題を解決するための手段
[0006] 本発明者等は上記課題につき鋭意検討を行った結果、安価に入手可能な 2 置 換ァクロレイン誘導体を、その炭素 炭素二重結合を立体選択的に還元する能力を 有する酵素源を用 ヽて立体選択的に還元することにより、光学活性 2—置換プロパ ナール誘導体を簡便に製造する方法を見出した。

発明を実施するための最良の形態
[0015] 1. 2—置橼ァクロレイン謙導体 (出 物皙)
まず、本発明の実施形態に関わる化合物について説明する。本発明の実施形態と しての製造方法における出発物質は、前記式(1)で表される 2—置換ァクロレインィ匕 合物である。前記式(1)における置換基 Rとしては、置換基を有するメチル基、置換 基を有して 、ても良 、炭素数 2〜: LOのアルキル基、又は置換基を有しても良 、炭素 数 5〜 15のァラルキル基が挙げられる。
[0016] 置換基を有する場合の置換基としては、酵素源による反応に悪影響を及ぼさない 限り特に制限されず、例えばハロゲン原子、ヒドロキシル基、アルデヒド基、カルボキ シル基、シァノ基、アミノ基または-トロ基等の置換基が挙げられる。
[0017] 例えば、置換基を有するメチル基としては、クロロメチル基、ブロムメチル基、ョード メチル基、ヒドロキシメチル基、アミノメチル基、又はシァノメチル基等が挙げられる。

次に、本発明の実施形態としての光学活性 2—置換プロパナール誘導体の製造法 について説明する。
[0024] 実施形態においては、前記式(1)で表される 2 置換ァクロレイン誘導体の炭素 炭素二重結合を立体選択的に還元する活性を有する酵素源の存在下、当該 2—置 換ァクロレイン誘導体(1)の二重結合を立体選択的に還元することにより、前記式(2 )で表される光学活性 2—置換プロパナール誘導体を製造する。

ここで、「酵素源」とは、上記還元活性を有する酵素自体はもちろんのこと、上記還 元活性を有する微生物の培養物およびその処理物も含まれる。「微生物の培養物」と は、菌体を含む培養液あるいは培養菌体を意味し、また、その処理物であってもよい 。「その処理物」とは、例えば、粗抽出液、凍結乾燥菌体、アセトン乾燥菌体、または それら菌体の破砕物等を意味する。さらに上記酵素源は、公知の手段で固定化して
、固定ィ匕酵素又は固定ィ匕菌体として用いることもできる。固定ィ匕は、当業者に周知の 方法 (例えば架橋法、物理的吸着法、包括法等)で行うことができる。

また、前記式(1)で表される化合物の炭素 炭素二重結合を S選択的に還元する 活性を有する酵素源としては、ァクロモバクタ一'キシロキダンス ·サブスピーシーズ' ァ-トリフィカンス Achromobacter xvlosoxiaans subsp. denitrincans)、 /ノレ 7リクネス 'フエカリス(Alcaligenes faecalis)、アルカリゲネス *スピーシーズ(Alcaligenes sp.)、ァ ースロノくクタ一 ·クリスタロポェテス (Arthrobacter crvstallopoietes)、アースロノくクタ一 •プロトフォロマイス(Arthrobacter protophormise)、コリネバタテリゥム 'アンモニアゲ ス (Corvnebacterium ammoniagenes)又【3~ェンエリヒフ 'コリ (Escherichia coli)等の 微生物由来の酵素源が好ま U、。

また、上記還元酵素の由来となる微生物としては、野生株または変異株のいずれで もよ!/ヽ。あるいは細胞融合または遺伝子操作等の遺伝学的手法により誘導される微 生物も用いることができる。
[0030] さらにはこれら微生物由来の還元酵素を生産する能力を有する組換え微生物であ つてもよい。該酵素を生産する組換え微生物は、例えば、これらの酵素を単離及び
Zまたは精製して酵素のアミノ酸配列の一部または全部を決定する工程、このアミノ 酸配列に基づ 、て酵素をコードする DNA配列を得る工程、この DNAを他の微生物 に導入して組換え微生物を得る工程、及びこの組換え微生物を培養して、本酵素を
得る工程を含有する方法により得ることができる(国際公開第 WO98Z35025号公 報に記載の方法参照)。
[0031] 上記のような組換え微生物としては、上記還元酵素をコードする DNAを有するベタ ターで宿主微生物を形質転換して得られる形質転換微生物が挙げられる。また、宿 主微生物としては大腸菌(Escherichia coli)が好ましい より好ましくは、上記サッカロ マイセス ·セレビシェ (Saccharomvces cerevisiae)由来の NADPHデヒドロゲナーゼ( Old Yellow Enzyme 2)遺伝子(The Journal of Biological chemistry, 268, 6097-6106 (1993)参照)を有するベクターで形質転換された Escherichia coli HB101 (pTSYE2)等 が挙げられる。この Escherichia coli HB101(pTSYE2)の取得方法は、後述の実施例 7 に己載し 7こ。
[0032] また、上記酵素源として、国際生化学'分子生物学連合の酵素分類法により EC 1.6 .99に分類される酸化還元酵素を用いることもできる。 EC 1.6.99に分類される酸化還 元酵素としては、 EC 1.6.99.1 : NADPHデヒドロゲナーゼ、 EC 1.6.99.2 : NAD(P)H デヒドロゲナーゼ(キノン)、 EC 1.6.99.3 : NADHデヒドロゲナーゼ、 EC 1.6.99.5 : NA DHデヒドロゲナーゼ(キノン)、または EC 1.6.99.6 : NADPHデヒドロゲナーゼ(キノン )に分類される酵素が挙げられ、なかでも EC 1.6.99.1に分類される NADPHデヒドロ ゲナーゼ(別名: Old Yellow Enzyme)が好まし!/、。
[0033] NADPHデヒドロゲナーゼとしては、キャンディダ (Candida)属、クルイべロマイセス
(Kluweromvces)属、サッカロマイセス(Saccharomvces)属、又はシゾサッカロマイセ ス (Schizosaccharomvces)属等の酵母に広く分布して ヽることが報告されており、とり わけ、サッカロマイセス ·セレビシェ (Saccharomvces cerevisiae)由来の NADPHデヒ ドロゲナーゼ(Old Yellow Enzyme 2)が好ましい。
[0034] 酵素源として用いる微生物のための培養培地は、その微生物が増殖し得るもので ある限り特に限定されない。例えば、炭素源として、グルコース、シユークロース等の 糖質、エタノール、グリセロール等のアルコール類、ォレイン酸、ステアリン酸等の脂 肪酸及びそのエステル類、菜種油、大豆油等の油類、窒素源として、硫酸アンモ-ゥ ム、硝酸ナトリウム、ペプトン、カザミノ酸、コーンスティープリカ一、ふすま、酵母ェキ スなど、無機塩類として、硫酸マグネシウム、塩ィ匕ナトリウム、炭酸カルシウム、燐酸 1
水素カリウム、燐酸 2水素カリウムなど、他の栄養源として、麦芽エキス、肉エキス等を 含有する通常の液体培地が使用することができる。培養は好気的に行い、通常、培 養時間は 1〜5日間程度、培地の pHが 3〜9、培養温度は 10〜50°Cで行うことがで きる。
[0035] 6.還元反応
実施形態において、前記式(1)で表される化合物の炭素 炭素二重結合の還元 反応は、適当な溶媒に、基質となる前記式(1)で表される 2—置換ァクロレイン誘導 体、補酵素 NAD (P) H及び前記微生物の培養物またはその処理物等を添加し、 pH 調整下攪拌することにより行うことができる。
[0036] 上記還元反応条件は用いる酵素、微生物またはその処理物、基質濃度等によって 異なるが、通常、基質濃度は約 0. 1〜: LOO重量%、好ましくは 1〜60重量%であり、 補酵素 NAD (P) Hは基質に対して 0. 0001〜100モル%、好ましくは 0. 0001〜0 . 1モル%、反応温度は 10〜60°C、好ましくは 20〜50°Cであり、反応の pHは 4〜9 、好ましくは 5〜8であり、反応時間は 1〜120時間、好ましくは 1〜72時間で行うこと 力 Sできる。また、反応には有機系の溶媒を混合して用いてもよい。有機系溶媒として は、例えば、トルエン、酢酸ェチル、酢酸 n—ブチル、へキサン、イソプロパノール、メ タノール、ジイソプロピルエーテル、アセトン、ジメチルスルホキシド等が挙げられる。

実施形態の還元工程にぉ ヽて、一般に用いられる補酵素 NAD (P) H再生系を組 み合わせて用いてもよい。これにより、高価な補酵素の使用量を大幅に減少させるこ とができる。代表的な NAD (P) H再生系としては、例えば、グルコース脱水素酵素及 びグルコースを用いる方法が挙げられる。
[0039] 還元酵素遺伝子及びこの酵素が依存する補酵素を再生する能力を有する酵素 (例 えばグルコース脱水素酵素)の遺伝子を同一宿主微生物内に導入した形質転換微 生物、すなわち、実施形態の還元酵素をコードする DNA及び該酵素が依存する補 酵素を再生する能力を有する酵素 (例えばグルコース脱水素酵素)の遺伝子を同一
宿主微生物内に導入した形質転換微生物の培養物またはその処理物等を用いて、 上記と同様の反応を行えば、別途に補酵素の再生に必要な酵素源を調整する必要 がな 、ため、より低コストで光学活性 2—置換プロパナール誘導体を製造することが できる。
[0040] 上記のような形質転換微生物としては、上記還元酵素をコードする DNA及び該酵 素が依存する補酵素を再生する能力を有する酵素をコードする DNAの両者を有す るプラスミドで形質転換された形質転換微生物が挙げられる。ここで、補酵素を再生 する能力を有する酵素としては、グルコース脱水素酵素が好ましぐバシラス'メガテリ ゥム(Bacillus megaterium)由来のグルコース脱水素酵素がより好ましい。また、宿主 微生物としては大腸菌 (Escherichia coli)が好まし ヽ。そのような好ま ヽ形質転換微 生物として、後 i术の実施例で説明する Escherichia coli HB101 (pTSYE2Gl)が挙げら れる。
[0041] 形質転換微生物の培養は、それらが増殖する限り、通常の、炭素源、窒素源、無機 塩類、有機栄養素などを含む液体栄養培地を用いて実施できる。

実施例

[0049] (実施例 1)光学活性 2—メチルー 1一へキサナールの製造法
グルコース 5%、ペプトン 1%、酵母エキス 1%からなる液体培地 (pH6. 5)を調製し 、 500ml容坂ロフラスコに 50mlづっ分注して、 120°Cで 20分間蒸気滅菌した。これ らの液体培地に表 1に示した微生物をそれぞれ 1白金耳植菌し、 30°Cで 2〜3日間 振盪培養した。この培養液から遠心分離により菌体を集め、菌体を水洗し、氷冷ァセ トンを添加した後、減圧乾燥することによりアセトン乾燥菌体を調製した。栓付試験管 に、アセトン乾燥菌体 5mgを量り取り、 lOOmMリン酸緩衝液 (pH6. 5) 500 /z lに懸 濁させた。この溶液に、グルコース 25mg、 NAD及び NADP各 0. 5mg、グルコース 脱水素酵素(天野ェンザィム社製) 5Uを加えた。その後、基質 2—プチルァクロレイ ン 2mgを酢酸ェチル 500 1に溶解させて栓付試験管にカ卩え、 30°Cで 20時間攪拌 した。反応後、反応液を遠心分離し、酢酸ェチル層中の基質及び生成物をトリフルォ ロアセチル誘導体ィ匕してガスクロマトグラフィー(GC)により分析することで、反応変 換率、及び、生成物の光学純度を求めた。その結果を表 1に示す (反応変換率は 10 〜100%)。

サッカロマイセス.セレビジェ S288C (ATCC26108) (Saccharomyces cerevisiae S288C (ATCC26108))株由来の NADPHデヒドロゲナーゼ(Old Yellow Enzyme 2, 以後、 OYE2と呼ぶ)を大腸菌で発現させるために、以下に示した方法で発現べクタ を調製した (Appl Environ Microbiol, 69, 933 (2003)参照)。

[2] 尺が、置換基を有しても良い炭素数 2〜 10のアルキル基である請求項 1記載の製 造法。
[3] 尺が、置換基を有しても良い炭素数 2〜4のアルキル基である請求項 1記載の製造 法。
[4] 前記炭素 炭素二重結合を立体選択的に還元する能力を有する酵素源として、キ ヤンデイダ (Candida)属、クルイべロマイセス (Kluweromvces)属、ピキア (Pichia)属、 ロドトノレラ(Rhodotorula)属、サッカロマイセス (Saccharomvces)属、スポリディオボノレス
(Sporidiobolus)属、スポロボロマイセス (Spolobolomvces)属、トリゴノプシス (Trigonop ≤k)属、チゴサッカロマイセス (Zveosaccharomvces)属、 クロモノ^:クタ一 (Achromoba cter)鼠、ァシディフイリゥム (Acidiphilium)属、ァノレカリゲネス (Alcaligenes)属、アース ロバクタ一 (Arthrobacter)属、バシラス (Bacillus)属、コリネバタテリゥム (Corvnebacter ium)属、ェシエリヒア(Escherichia)属、ミクロコッカス (Micrococcus)属、シユードモナ ス(Pseudomonas)属、バエニバシラス (Paenibacillus)属、またはザントモナス(Xantho mem)属に属する微生物由来の酵素源を用いることを特徴とする請求項 ι〜3のい ずれか 1項に記載の製造法。
[5] 前記炭素 炭素二重結合を立体選択的に還元する能力を有する酵素源として、キ ヤンデイダ (Candida)属、クルイべロマイセス (Kluweromvces)属、ピキア (Pichia)属、 ロドトノレラ(Rhodotorula)属、サッカロマイセス(Saccharomvces)属、スポリディオボノレス (Sporidiobolus)属、スポロボロマイセス (Spolobolomvces)属、トリゴノプシス (Trigonop ≤k)属、チゴサッカロマイセス(Zvgosaccharomvces)属、ァシディフイリゥム(AcidiDhiliu m)属、アースロバクタ一(Arthrobacter)属、バシラス(Bacillus)属、ミクロコッカス (Micr ococcus)属、シユードモナス(Pseudomonas)属、バエニノ シラス(Paenibacillus)属、ま たはザントモナス (Xanthomonas)属に属する微生物に由来し、前記式(1)で表される 化合物の炭素 炭素二重結合を R選択的に還元する能力を有する酵素源を用いる 請求項 1〜3のいずれか 1項に記載の製造法。

[7] 前記 R選択的に還元する能力を有する酵素源力 サッカロマイセス 'セレピシェ ccharomvces cerevisiae)由来の NADPHデヒドロゲナーゼ (Old Yellow Enzyme 2)遺 伝子を有するベクターで形質転換された形質転換微生物の培養物である請求項 5記 載の製造法。
[8] 前記炭素 炭素二重結合を立体選択的に還元する能力を有する酵素源として、了 クロモパクター(Achromobacter)属、ァノレカリゲネス (Alcaligenes)属、アースロバクタ 一(Arthrobacter)属、コリネパクテリゥム(Corvnebacterium)属、またはェシエリヒア(E scherichia)属に属する微生物に由来し、前記式(1)で表される化合物の炭素 炭素 二重結合を S選択的に還元する能力を有する酵素源を用いる請求項 1〜3のいずれ 力 1項に記載の製造法。
[9] 前記 S選択的に還元する能力を有する酵素源力 ァクロモパクター 'キシロキダンス
.サブスピ ~~、ン' ~~ズヽァ-トリフィ 7ンス (Achromobacter xvlosoxidans subsp. aemtrmc ans)、アルカリゲネス'フエカリス (Alcaligenes faecalis)、アルカリゲネス 'スピーシーズ (Alcali¾enes sp.)、アースロノくクタ一'クリスタロポェテス(Arthrobacter crystalloDoiete
s) ,アースロバクタ一 ·プロトフォロミス (Arthrobacter protophormise)、コリネバクテリウ ム ·アンモニアゲネス (Corvnebacterium ammoniagenes)及びェシエリヒア ·コリ (Escher ichia coH)からなる群より選択される 1種以上の微生物由来の酵素源である請求項 8 記載の製造法。
[10] 前記炭素 炭素二重結合を立体選択的に還元する能力を有する酵素源として、 E
C番号(Enzyme Commission Number) 1.6.99に分類される酸化還元酵素を用いる請 求項 1〜3のいずれか 1項に記載の製造法。
[11] 前記 EC 1.6.99に分類される酸化還元酵素として、 EC 1.6.99.1に分類される NAD
PHデヒドロゲナーゼを用い、かつ、前記式(2)で表される化合物の R体を製造する 請求項 10記載の製造法。
[12] 前記 NADPHデヒドロゲナーゼが、サッカロマイセス ·セレビシェ (Saccharomvces c erevisiae)由来の NADPHデヒドロゲナーゼ(Old Yellow Enzyme 2)である請求項 11 記載の製造法。
