Abstract

 【課題】 本発明の課題は、簡略化され、かつ効率が改善されたDNA組換え方法を提供することにある。 【解決手段】 上記課題は、ベクターDNAに目的DNA断片を挿入する組換えDNAの調製方法であって、(a)DNA認識部位とDNA切断部位とが異なる制限酵素の存在下、該ベクターDNAの目的DNA断片挿入部位と該目的DNA断片を含むDNAとを同時に切断する反応と、(b)DNA連結酵素の存在下、該目的DNA断片を該ベクターDNAに挿入する反応を、同一の反応場所において実質的に同時に行う工程を含む、組換えDNAの調製方法によって解決される。
