Abstract

 ドナー分子及びアクセプター分子からなるサンプルについて、短時間に多数のサンプルのFRET検出を行うとき、まず、周波数f+Δfで強度変調したドナー分子励起用の第1のレーザ光をドナー分子に照射するとともに、周波数fで強度変調したアクセプター分子励起用の第2のレーザ光をアクセプター分子に照射し、アクセプター分子が発する蛍光を受光する。受光した蛍光の蛍光信号から、FRETによりアクセプター分子が発する蛍光の第1の信号成分と、第2のレーザ光の照射により励起したアクセプター分子が発する蛍光の第2の信号成分とを抽出する。抽出した第1の信号成分の位相遅れと、抽出した第2の信号成分の位相遅れとを算出し、これらの位相遅れに基づいて、FRETの発生の有無を判定する。
