Abstract
Description
本実施の形態の医療用組成物は、支持体および当該支持体上に設けられた表層を有する担体と、上記担体の表面に備えられた細胞とを含むものである。以下に、各構成について詳細に説明する。なお、本実施の形態の医療用組成物は、特に、血管新生用組成物として用いることが可能である。
上記支持体は、生体吸収性高分子によって形成されている。
本実施の形態の医療用組成物では、上述した支持体上に、ハイドロキシアパタイト(Ca10(PO4)6(OH)2)からなる表層が設けられている。そして、主として当該表層と上記支持体とによって、後述する細胞を備えさせる対象である担体が形成されている。
ここで「一次粒子」とは、焼結工程の前に、ハイドロキシアパタイト(HAp)によって形成された粒子のことを意味する。すなわち、ハイドロキシアパタイト粒子の製造工程において、初めて形成された粒子のことを意味する。また狭義には単結晶粒子のことを意味する。なお本明細書において「一次粒子」とは、非晶質(アモルファス)の状態のもの、及びその後に焼結を行なった焼結体の状態のものをも含む。
混合工程は、一次粒子と融着防止剤とを混合する工程である。つまり、上記一次粒子生成工程によって得られた一次粒子群の粒子間に、あらかじめ融着防止剤を介在させておくことによって、その後の焼結工程における一次粒子同士の融着を防止するための工程である。なお本当該混合工程によって得られた一次粒子と融着防止剤との混合物を「混合粒子」と呼ぶこととする。
焼結工程は、上記混合工程によって得られた混合粒子を焼結温度に曝して、当該混合粒子に含まれる混合粒子をセラミック粒子(焼結体粒子)にする工程である。このとき、混合粒子の粒子間には融着防止剤が介在しているので、焼結工程における高温条件に曝されても、ハイドロキシアパタイト一次粒子同士の融着を防止することができる。
除去工程は、焼結工程によって得られた焼結体粒子の粒子間に混在する融着防止剤を取り除く工程である。
表層形成工程は、上述した一連の工程によって得られたハイドロキシアパタイト粒子によって支持体をコーティングし、これによって、支持体上に表層を形成する工程である。換言すれば、表層形成工程は、支持体と表層とを有する担体を形成する工程である。
本実施の形態の医療用組成物では、上述したようにして形成された担体上に、細胞が備えられている。
本実施の形態の医療用キットは、上述した本発明の医療用組成物と、当該医療用組成物を生体(例えば、患者)に投与するための注射器とを備えるものである。なお、本実施の形態の医療用キットは、特に、血管新生用キットとして用いることが可能である。
本実施の形態の医療用組成物の製造方法は、生体吸収性高分子からなる支持体、および当該支持体上に設けられたハイドロキシアパタイトからなる表層を有する粒子状の担体と細胞とを混合する工程を含む方法である。
〔実施例〕
以下の実施例では、本願発明の医療用組成物の血管新生効果を簡便に検討するために、ポリ乳酸膜を用いた構成と、ポリ乳酸粒子を用いた構成との両方について検討した。ポリ乳酸膜を用いた構成とポリ乳酸粒子を用いた構成とは、血管新生に関しては同等の効果を示したが、投与の容易性、投与時にできる傷跡の有無、当該傷跡の治療の必要性などの観点からは、ポリ乳酸粒子を用いた構成の方が優れているといえる。この点については、以下の具体的な記載から容易に理解できるであろう。
アンモニア水にてpHを12.0に調製したCa(NO3)2水溶液(42mN、800mL)を、コンデンサーおよび半月状攪拌翼が設置された1Lフラスコに注ぎ込み、80℃に保持した。
ポリ乳酸不織布膜(厚さ140μm、目付20g/m2、引裂強力3N)をアンモニア水溶液(pH12.0)に30分間浸漬することによって、ポリ乳酸不織布の膜表面にカルボキシル基を導入した。上記処理後のポリ乳酸粒子を、水およびエタノールにて洗浄した。
ポリ乳酸粒子(粒子径100μm)を室温にてエタノールで洗浄した。次いで、実施例1において作製したハイドロキシアパタイト粒子を、2重量%の濃度にてエタノール中に分散させた。
実施例2および3において作製したハイドロキシアパタイト/ポリ乳酸膜複合体上およびハイドロキシアパタイト/ポリ乳酸粒子複合体上にて、骨髄細胞を2.5時間培養した。なお、コントロールとしては、ハイドロキシアパタイト粒子を備えないポリ乳酸膜またはポリ乳酸粒子を用いた。
実施例3において作製したハイドロキシアパタイト/ポリ乳酸粒子複合体上にて、骨髄細胞を2.5時間培養した。
下肢虚血モデルマウスを公知の大腿動脈切除法によって作製し、膜状のハイドロキシアパタイト/ポリ乳酸膜/骨髄細胞複合体、および、ハイドロキシアパタイト/ポリ乳酸膜複合体の各々を大腿動脈を切除した下肢の筋肉表面に付着させた後に、皮膚を縫合した(図3の(a)参照)。また、粒子状のハイドロキシアパタイト/ポリ乳酸粒子/骨髄細胞複合体、および、ハイドロキシアパタイト/ポリ乳酸粒子複合体の各々を、注射器を用いて筋肉内に投与した(図3の(b)参照)。投与の2週間後に、走査型顕微鏡にて血管新生の有無を確認した。
BALB/Cマウスを用いて公知の方法によって虚血肢を作製し、大腿動脈を切除した下肢の筋肉中に、ハイドロキシアパタイト/ポリ乳酸粒子/骨髄細胞複合体、ハイドロキシアパタイト/ポリ乳酸粒子複合体、骨髄細胞のみ、の各々を注射器にて筋肉内に投与した。その後、虚血下肢の予後を観察した。なお、骨髄細胞は、移植するマウスとは別のBALB/Cマウスから採取した。
Claims (10)
- 生体吸収性高分子からなる支持体、および前記支持体上に設けられたハイドロキシアパタイトからなる表層を有する粒子状の担体と、
前記担体の表面に備えられた細胞と、を含むことを特徴とする医療用組成物。 - 前記担体の粒子径は、10μm~200μmであることを特徴とする請求項1に記載の医療用組成物。
- 前記担体は、多孔質であることを特徴とする請求項1または2に記載の医療用組成物。
- 前記生体吸収性高分子は、ポリ乳酸、ポリグリコール酸、ポリエチレングリコール、プロピレングリコール、ポリヒドロキシブチレート、ポリカーボネート、ポリアミド、セルロース、キチン、キトサン、デンプン、ポリグルタミン酸、ポリジオキサノン、シアノアクリレート重合体、ポリカプロラクトン、合成ポリペプチド、ヒアルロン酸、ポリリンゴ酸、ポリコハク酸ブチレン、およびこれらの組み合わせからなる共重合体からなる群より選択される少なくとも1つであることを特徴とする請求項1~3の何れか1項に記載の医療用組成物。
- 前記細胞は、骨髄単核細胞、末梢血単核細胞、多能幹細胞、造血幹細胞、血管内皮前駆細胞、iPS細胞、ES細胞、血小板、および間葉系幹細胞からなる群より選択される少なくとも1つの細胞であることを特徴とする請求項1~4の何れか1項に記載の医療用組成物。
- 前記担体の表面に、血管新生サイトカインが備えられていることを特徴とする請求項1~5の何れか1項に記載の医療用組成物。
- 前記血管新生サイトカインは、aFGF、bFGF、VEGF、HGF、PDGF、PIGF、TNF、EGF、angiopoietin、IL、HAPO、Shh、TGF-β、G-CSF、M-CSF、SCF、EPO、TPO、およびFltからなる群より選択される少なくとも1つのサイトカインであることを特徴とする請求項6に記載の医療用組成物。
- 請求項1~7の何れか1項に記載の医療用組成物と、
前記医療用組成物を生体に投与するための注射器と、を備えることを特徴とする医療用キット。 - 生体吸収性高分子からなる支持体、および前記支持体上に設けられたハイドロキシアパタイトからなる表層を有する粒子状の担体と細胞とを混合する工程を含むことを特徴とする医療用組成物の製造方法。
- 前記工程では、前記担体1粒子あたり1×1010個以下の前記細胞を混合することを特徴とする請求項9に記載の医療用組成物の製造方法。
