- Example 11 To 1 kg of ginseng powder, 0.53 L of sulfuric acid (96% manufactured by Wako Pure Chemical Industries, Ltd.) and 9.47 L of water (0.96 mol / L sulfuric acid hydrolyzate) were added for 6 hours. It heated at 80 degreeC and the hydrolysis process was performed. Subsequently, a 5M sodium hydroxide solution was added to the obtained hydrolyzed solution to adjust the pH to 7.0, followed by suction filtration. The obtained residue was dried by heating and ventilation to obtain 147.5 g of a powder (sapogenin-containing composition). Table 4 shows each processing condition.
- Example 12 2. 1 kg of ginseng powder, 0.53 L of sulfuric acid (96% manufactured by Wako Pure Chemical Industries, Ltd.), 6.47 L of water, and 99.5% ethanol (manufactured by Wako Pure Chemical Industries, Ltd.) 0 L (0.96 mol / L sulfuric acid hydrolyzate) was added and heated at 80 ° C. for 6 hours for hydrolysis treatment. Subsequently, 5M sodium hydroxide solution was added to the obtained liquid after hydrolysis treatment to adjust the pH to 7.0, 90 L of distilled water was added, and the mixture was stirred and suction filtered. The obtained residue was dried by heating and ventilation to obtain 156.8 g of a powder (sapogenin-containing composition). Table 4 shows each processing condition.

(Manufactured) 3.0 L was added (1.92 mol / L hydrochloric acid hydrolyzate), and the mixture was heated at 80 ° C. for 6 hours for hydrolysis treatment. Subsequently, 5M sodium hydroxide solution was added to the obtained liquid after hydrolysis treatment to adjust the pH to 7.0, 90 L of distilled water was added, and the mixture was stirred and suction filtered. The obtained residue was dried by heating and ventilation to obtain 453.0 g of a powder (sapogenin-containing composition). Table 4 shows each processing condition.

Abstract
Description
<1> サポニン含有植物に、0.01mol/L~4mol/L濃度の強酸水溶液を作用させて加水分解処理を施し、得られた加水分解処理後の液を中和後、濾過し、残渣を乾燥することを特徴とするサポゲニン高含有組成物の製造方法である。
<2> 加水分解処理を、低級アルコールの存在下で行う前記<1>に記載のサポゲニン高含有組成物の製造方法である。
<3> 低級アルコールの使用量が、加水分解液総量に対して1容量%以上80容量%以下である前記<2>に記載のサポゲニン高含有組成物の製造方法である。

本発明のサポゲニン高含有組成物の製造方法は、サポニン含有植物に、所定の濃度の強酸水溶液を作用させて加水分解処理を施し(加水分解処理工程)、得られた加水分解処理後の液を中和後(中和工程)、濾過し(濾過工程)、残渣を乾燥する(乾燥工程)ことを特徴とする。なお、前記サポゲニン高含有組成物の製造方法は、前記各工程以外のその他の工程を有していてもよい。
本発明において、原料として使用される前記サポニン含有植物としては、サポニンが含まれる天然物であれば特に制限はなく、目的に応じて適宜選択することができ、例えば、ウコギ科トチバニンジン属田七人参の根、ウコギ科トチバニンジン属御種人参の根、ウコギ科トチバニンジン属トチバニンジンの根(チクセツニンジン)、ウコギ科ウコギ属エゾウコギの根、ウコギ科タラノキ属タラノキの根、ウコギ科タラノキ属ウドの根、セリ科ミシマサイコ属ミシマサイコの根(サイコ)、ヒメハギ科ヒメハギ属イトヒメハギの根(オンジ)、ヒメハギ科ヒメハギ属ヒロハセネガの根(セネガ)、キキョウ科キキョウ属キキョウの根、ウリ科アマチャヅル属アマチャヅルの全草、マメ科カンゾウ属カンゾウの根、ヒユ科イノコズチ属ヒナタイノコズチの根(ゴシツ)、アケビ科アケビ属ミツバアケビの茎(モクツウ)、クロウメモドキ科ナツメ属ナツメの実(タイソウ)、ユリ科ハナスゲ属ハナスゲの根茎(チモ)、ユリ科ジャノヒゲ属ジャノヒゲの根(バクモンドウ)、ヤマノイモ科ヤマノイモ属オニドコロの根茎(ヒカイ)などが挙げられる。
中でも、ウコギ科トチバニンジン属田七人参の根、ウコギ科トチバニンジン属御種人参の根、ウコギ科トチバニンジン属トチバニンジンの根(チクセツニンジン)、ウコギ科ウコギ属エゾウコギの根、ウコギ科タラノキ属タラノキの根、ウコギ科タラノキ属ウドの根、ウリ科アマチャヅル属アマチャヅルの全草が好ましく、最も高いサポゲニン収量が得られるウコギ科トチバニンジン属田七人参の根が特に好ましい。
前記加水分解処理工程では、前記サポニン含有植物に所定の濃度の強酸水溶液を作用させ、前記植物中のサポニンを加水分解し、サポニンよりも体内吸収性に優れたサポゲニンを生成させる。
なお、前記加水分解処理は、低級アルコールの存在下で行うことがより好ましい。前記低級アルコールを使用することにより、前記サポニン含有植物と、前記強酸水溶液との親和性を向上させ、効率よく加水分解を進めることが可能となる。また、前記低級アルコールを使用することにより、得られるサポゲニン含有植物の、味や取扱い性を高めることができる点でも、有利である。前記低級アルコールとしては、特に制限はなく、目的に応じて適宜選択することができるが、中でも、メタノール、エタノール、プロパノールが好ましく、安全性の点からエタノールが特に好適である。
前記中和工程では、前記加水分解処理後、得られた加水分解処理後の液を中和する。
前記中和は、特に制限はなく、公知の手法により行うことができ、例えば、前記加水分解処理後の液に、水酸化ナトリウム、水酸化カリウム等の強塩基水溶液を適宜加えることにより、行うことができる。なお、前記中和後のpHは、5~8とすることが好ましい。
前記濾過工程では、前記中和工程後の加水分解処理後の液を濾過し、濾液と残渣とに分離する。
前記濾過は、特に制限はなく、公知の手法により行うことができる。なお、濾過後は、更に塩がなくなるまで水洗を繰り返してもよい。
前記加水分解処理工程で低級アルコールを使用しなかった場合は、中和後、そのまま前記濾過工程に進むことができるが、低級アルコールを使用した場合は、濾過前に、生成されたサポゲニンの残渣への残留を促す目的で、水を加えて加水分解処理後の液中の低級アルコール濃度を下げることが好ましい。この場合に添加する水は多いほど良いが、加水分解処理後の液中の低級アルコール濃度は、低いほど好ましく、具体的には、50容量%以下となるように添加することが好ましく、30容量%以下となるように添加することがより好ましく、10容量%以下となるように添加することが更に好ましい。前記加水分解処理後の液中の低級アルコール濃度が、50容量%を超えたまま濾過に供すると、生成されたサポゲニンが低級アルコールに溶解して濾液として排出されてしまい、残渣中のサポゲニン含有量が減少してしまう点で不利となる。一方、前記加水分解処理後の液中の低級アルコール濃度を、前記更に好ましい範囲内とすると、より残渣中のサポゲニン含有率を高めることができる点で、有利である。
なお、サポニンは、水溶性であるが、加水分解処理により得られるサポゲニンは、水不溶性である。
また、濾過前に、生成されたサポゲニンの残渣への残留を促す目的で、減圧濃縮により低級アルコールを留去することで、加水分解処理後の液中の低級アルコール濃度を下げることができる。この場合、濃縮温度は70℃以下が好ましく、40℃~50℃がより好ましい。低級アルコール濃度は50容量%以下となるように留去することが好ましく、30容量%以下となるように留去することがより好ましく、10容量%以下となるように留去することが更に好ましい。前記加水分解処理後の液中の低級アルコール濃度が、50容量%を超えたまま濾過に供すると、生成されたサポゲニンが低級アルコールに溶解して濾液として排出されてしまい、残渣中のサポゲニン含有量が減少してしまう点で不利となる。一方、前記加水分解処理後の液中の低級アルコール濃度を、前記更に好ましい範囲内とすると、より残渣中のサポゲニン含有率を高めることができる点で、有利である。
また、前記減圧濃縮と、前記加水濾過とは、それぞれ単独の工程として行ってもよいが、一連の工程として行ってもよい。この場合、前記減圧濃縮後の液に対して水を加え、前記加水濾過を行う。
前記乾燥工程では、前記濾過工程後の残渣を乾燥し、サポゲニン高含有組成物を得る。
前記乾燥は、特に制限はなく、公知の手法により行うことができ、例えば、凍結乾燥、通風乾燥、加熱乾燥、減圧乾燥など通常の方法が利用できる。
本発明では、前記したような簡便な手法により、原料であるサポニン含有植物中に含まれるサポニンからサポゲニンを生成し、サポゲニンを高い含有率で含有する植物組成物(サポゲニン高含有組成物)を得ることができる。なお、得られたサポゲニン含有植物中のサポゲニン含有率は、3%以上が好ましく、5%以上がより好ましく、10%以上が更に好ましい。なお、前記サポゲニン含有率は、後述する実施例に記載の方法により測定した値である。
サポゲニンはサポニンよりも体内吸収性に優れるため、得られたサポゲニン含有高含有組成物は原料であるサポニン含有植物よりも強い生理活性(糖代謝改善作用等)が期待できる。また、前記のような方法により得られたサポゲニン含有組成物は、安全性が高く、また、味や取扱い性の点でも優れるため、そのまま、或いは、適宜処理を施した後に、健康食品等の有効成分として好適に利用可能である。
田七人参粉末1kgに、塩酸(35.0~37.0%和光純薬工業(株)製)0.0083L、及び、水9.9917Lを加えて(0.01mol/L濃度の塩酸加水分解液)、6時間、80℃で加熱し、加水分解処理を行った。続いて、得られた加水分解処理後の液に5Mの水酸化ナトリウム溶液を加え、pH7.0に調整した後、吸引ろ過を行った。得られた残渣を加温減圧乾燥して、粉末(サポゲニン含有組成物)167.4gを得た。各処理条件を表2に示す。
田七人参粉末1kgに、塩酸(35.0~37.0%和光純薬工業(株)製)1.6L、及び、水8.4Lを加えて(1.92mol/L濃度の塩酸加水分解液)、6時間、80℃で加熱し、加水分解処理を行った。続いて、得られた加水分解処理後の液に5Mの水酸化ナトリウム溶液を加え、pH7.0に調整した後、吸引ろ過を行った。得られた残渣を加温減圧乾燥して、粉末(サポゲニン含有組成物)173.1gを得た。各処理条件を表2に示す。
田七人参粉末1kgに、塩酸(35.0~37.0%和光純薬工業(株)製)3.3L、及び、水6.7Lを加えて(4mol/L濃度の塩酸加水分解液)、6時間、80℃で加熱し、加水分解処理を行った。続いて、得られた加水分解処理後の液に5Mの水酸化ナトリウム溶液を加え、pH7.0に調整した後、吸引ろ過を行った。得られた残渣を加温減圧乾燥して、粉末(サポゲニン含有組成物)152.7gを得た。各処理条件を表2に示す。
田七人参粉末1kgに、塩酸(35.0~37.0%和光純薬工業(株)製)1.6L、及び、水8.3L、並びに、99.5%エタノール(和光純薬工業(株)製)0.1Lを加えて(1.92mol/L濃度の塩酸加水分解液)、6時間、80℃で加熱し、加水分解処理を行った。続いて、得られた加水分解処理後の液に5Mの水酸化ナトリウム溶液を加え、pH7.0に調整した後、蒸留水を90L添加し、攪拌して、吸引ろ過を行った。得られた残渣を凍結乾燥して、粉末(サポゲニン含有組成物)150.3gを得た。各処理条件を表2に示す。
田七人参粉末1kgに、塩酸(35.0~37.0%和光純薬工業(株)製)1.6L、及び、水5.4L、並びに、99.5%エタノール(和光純薬工業(株)製)3.0Lを加えて(1.92mol/L濃度の塩酸加水分解液)、6時間、80℃で加熱し、加水分解処理を行った。続いて、得られた加水分解処理後の液に5Mの水酸化ナトリウム溶液を加え、pH7.0に調整した後、蒸留水を90L添加し、攪拌して、吸引ろ過を行った。得られた残渣を凍結乾燥して、粉末(サポゲニン含有組成物)162.9gを得た。各処理条件を表2に示す。

三角フラスコに、水2L、田七人参粉末80g、及び、セルラーゼY-NC(ヤクルト薬品工業株式会社製)4gを入れ、緩やかに振盪しながら5日間、50℃に保温した。これに1.66mol/Lの塩酸を8mL添加し(溶液中の塩酸濃度0.0066mol/L)、120℃で40分間、加熱処理した。これを凍結乾燥し、粉末85gを得た。各処理条件を表5に示す。

実施例1~22、及び、比較例1~20で得られた各粉末について、サポゲニン含有率、並びに、味のよさを以下のようにして評価した。また、実施例1~22、及び、比較例1~20について、製法の簡便性を以下のようにして評価した。また、実施例1~22、及び、比較例1~20について、廃液処理(溶媒回収操作)の簡便性の評価を以下のようにして評価した。各評価結果を表2~10に示す。
1.5mLのエッペンチューブに、被検試料(実施例1~22、及び、比較例1~20で得られた各粉末)10mg、及び内部標準としてマグノロール(和光純薬工業(株)製)100mg/100mLエタノール溶液を0.1mL入れ、エタノールで全容量を1mLとし、1分間ボルテックスした後に5分間ソニケーション(超音波処理)を行い、サポゲニンを抽出した。これを10,000rpmで15分間遠心分離し、上清をさらにMillex(登録商標)-LGフィルター(MILLIPORE製)で濾過し、濾過液10μLを液体クロマトグラフィーにて分析し、4種のサポゲニン(プロトパナキサトリオール、パナキサトリオール、プロトパナキサジオール、パナキサジオール)のピークエリア面積を測定した。

4種のサポゲニンの検量線作成用標準溶液A、B、Cは、エタノールにそれぞれプロトパナキサトリオールがA:10mg/100mL、B:50mg/100mL、C:100mg/100mL、パナキサトリオールがA:100mg/100mL、B:25mg/100mL、C:500mg/100mL、プロトパナキサジオールがA:5mg/100mL、B:20mg/100mL、C:200mg/100mL、パナキサジオールがA:100mg/100mL、B:500mg/100mL、C:1000mg/100mL、及び内部標準マグノロールが各10mg/100mL含有されるように調製した。(4種のサポゲニンはいずれもシグマ製)。この標準溶液A、B、C、各10μLについて、液体クロマトグラフィー条件下でピークエリア面積を測定し、各サポゲニンとマグノロールのピーク面積比及び濃度比から検量線を作成した。
各検量線から被検試料中の4種のサポゲニン量を読み取り、含有率を算出した。なお、表2~10中のサポゲニン含有率は4種のサポゲニン含有率の合計で示した。
パネラー6名に各粉末を試食してもらい、以下の評点及び基準に従い評価した。

Claims (13)
- サポニン含有植物に、0.01mol/L~4mol/L濃度の強酸水溶液を作用させて加水分解処理を施し、得られた加水分解処理後の液を中和後、濾過し、残渣を乾燥することを特徴とするサポゲニン高含有組成物の製造方法。
- 加水分解処理を、低級アルコールの存在下で行う請求項1に記載のサポゲニン高含有組成物の製造方法。
- 低級アルコールの使用量が、加水分解液総量に対して1容量%以上80容量%以下である請求項2に記載のサポゲニン高含有組成物の製造方法。
- 加水分解処理後かつ濾過前に、加水分解処理後の液に水を加え、前記加水分解処理後の液中の低級アルコール濃度を50容量%以下に調整する請求項2から3のいずれかに記載のサポゲニン高含有組成物の製造方法。
- 加水分解処理後かつ濾過前に、加水分解処理後の液を減圧濃縮することにより、前記加水分解処理後の液中の低級アルコール濃度を50容量%以下に調整する請求項2から4のいずれかに記載のサポゲニン高含有組成物の製造方法。
- サポニン含有植物がウコギ科に属する植物である請求項1から5のいずれかに記載のサポゲニン高含有組成物の製造方法。
