核苷酸类似物在细胞内毋需磷酸化,因此能够克服拉米夫定 的耐药性, 而且自身不产生耐药性。 其代表性药物阿德福韦酯已 在欧美获准上市。 但是, 阿德福韦酯有一定的细胞毒性, 临床应 用能够产生肾脏毒性; 而且, 与拉米夫定相似, 阿德福韦酯治疗 在停药后, 会出现乙肝病毒复制的反跳, 引起乙肝的复发。
根据抗 HIV 药物研究的成功经验, 通过联合用药的 COCKTAIL的疗法, 能够有效克服抗药性, 加快病毒的清除。 乙肝感染的人数是 HIV感染人数的 10多倍, 但是与抗 HIV药 物相比, 临床有效的抗乙肝病毒药物还屈指可数。

本发明化合物或其药学上可接受的盐可以单独或以药物组 合物的形式给药。本发明药物组合物可根据给药途径配成各种适 宜的剂型。使用一种或多种生理学上可接受的载体, 包含赋形剂 和助剂,它们有利于将活性化合物加工成可以在药学上使用的制 剂。 适当的制剂形式取决于所选择的给药途径, 可以按照本领域 熟知的常识进行制造。
因此, 运用本领域技术人员熟悉的药物载体可以制备成含有 效剂量的本发明化合物的药物组合物。 因此本发明还提供包含与 一种或多种无毒药物可接受栽体配制在一起的本发明化合物的药 物组合物。 所述药物组合物可特别专门配制成以固体或液体形式 供口服给药、 供胃肠外注射或供直肠给药。
给药途径可以是口服、 非肠道或局部给药,优选口服和注射 形式给药。 可以口服的药物制剂包括胶嚢剂和片剂等。 病人吞咽 有困难时, 也可以釆用舌下片或者其他非吞咽的方式给药。 本发 明化合物也可以配制用于肠胃外给药或者透皮给药或者经粘膜 给药。 本领域技术人员可以理解, 本发明化合物可以采用合适的 药物释放系统 (DDS)以得到更有利的效果。 具体地, 本发明的药物组合物可通过口服、 直肠、 胃肠外、 池内、 阴道内、 腹膜内、 局部 (如通过散剂、 软膏剂或滴剂)、 口 颊给予人类和其他哺乳动物, 或者作为口腔喷雾剂或鼻腔喷雾剂 给予。 本文所用术语 "胃肠外"指包括静脉内、 肌肉内、 腹膜内、 胸骨内、 皮下和关节内注射和输液的给药方式。
适合于胃肠外注射的组合物可包括生理上可接受的无菌含水 或非水溶液剂、 分散剂、 混悬剂或乳剂, 及供重构成无菌可注射 溶液剂或分散剂的无菌散剂。 合适的含水或非水载体、 稀释剂、 溶剂或媒介物的实例包括水、 乙醇、 多元醇(丙二醇、 聚乙二醇、 甘油等)、 植物油 (如橄榄油)、 可注射有机酯如油酸乙酯及它们的 合适混合物。
这些组合物也可含有辅料, 如防腐剂、 湿润剂、 乳化剂和分 散剂。 通过各种抗细菌剂和抗真菌剂, 例如尼泊金酯类、 三氯叔 丁醇、 苯酚、 山梨酸等, 可确保防止微生物的作用。 还期望包括 等渗剂, 例如糖类、 氯化钠等。 通过使用能延迟吸收的物质, 例 如单硬脂酸铝和明胶, 可达到可注射药物形式的延长吸收。
混悬剂中除活性化合物外还可含有悬浮剂, 例如乙氧基化异 十八醇、 聚氧乙烯山梨醇和聚氧乙烯失水山梨糖醇酯、 微晶纤维 素、 偏氢氧化铝、 膨润土、 琼脂和黄蓍胶或者这些物质的混合物 等。
在一些情况下, 为延长药物的作用, 期望减慢皮下或肌内注 射药物的吸收。 这可通过使用水溶性差的晶体或无定形物盾的液 体混悬剂来实现。 这样, 药物的吸收速度取决于其溶解速度, 而 溶解速度又可取决于晶体大小和晶型。 或者, 胃肠外给药的药物 形式的延迟吸收通过将该药物溶解于或悬浮于油媒介物中来实 现。
可注射贮库制剂形式可通过在生物可降解聚合物如聚丙交酯
-聚乙交酯 (polylactide-polyglycolide)中形成药物的微胶嚢基质来 制备。 可根据药物与聚合物之比和所釆用的具体聚合物的性质, 对药物幹放速度加以控制。 其他生物可降解聚合物的实例包括聚 原酸酉旨类 (poly(orthoesters))和聚肝类 (poly(anhydrides))。 可注射 贮库制剂也可通过将药物包埋于能与身体组织相容的脂质体或微 乳中来制备。
可注射制剂可例如通过用滤菌器过滤或通过掺入无菌固体组 合物形式的灭菌剂来灭菌, 所述固体组合物可在临用前溶解或分 散于无菌水或其他无菌可注射介质。
本发明化合物或其组合物可用口服方法或非胃肠道给药方 式。 口服给药可以是片剂、 胶嚢剂、 包衣剂, 肠道外用药制剂有 注射剂和栓剂等。这些制剂是按照本领域的技术人员所熟悉的方 法制备的。 为了制造片剂、 胶嚢剂、 包衣剂所用的辅料是常规用 的辅料, 例如淀粉、 明胶、 阿拉伯胶, 硅石, 聚乙二醇, 液体剂 型所用的溶剂如水、 乙醇、 丙二醇、 植物油 (如玉米油、 花生油、 橄榄油等)。 含有本发明化合物的制剂中还有其它辅料, 例如表 面活性剂, 润滑剂, 崩解剂, 防腐剂, 矫味剂和色素等。 在片剂、 胶嚢剂、 包衣剂、 注射剂和栓剂中含有本发明式 I化合物的剂量 是以单元剂型中存在的化合物量计算的。在单元剂型中本发明式
I化合物一般含量为 l-5000mg,优选的单元剂型含有 10-500mg, 更优选的单元剂型含有 20-300mg. 具体地说, 本发明可以提供 的供口服给药的固体剂型包括胶嚢剂、 片剂、 丸剂、 散剂和颗 粒剂。 在此类固体剂型中, 活性化合物可与至少一种惰性的药 物可接受赋形剂或栽体如柠檬酸钠或磷酸二钙和 /或以下物质混 合: a)填充剂或增量剂如淀粉、 乳糖、 蔗糖、 葡萄糖、 甘露糖醇 和硅酸; b)粘合剂如羧甲基纤维素、 海藻酸盐、 明胶、 聚乙烯吡 咯烷酮、蔗糖和阿拉伯树胶; c)保湿剂如甘油; d)崩解剂如琼脂、 碳酸钙、 马铃薯或木薯淀粉、 海藻酸、 某些硅酸盐和碳酸钠; e) 溶液阻滞剂如石蜡; f)吸收加速剂如季铵化合物; g)湿润剂如鲸 蜡醇和甘油单硬脂酸酯; h)吸附剂如高岭土和膨润土以及 i)润滑 剂如滑石粉、 硬脂酸钙、 硬脂酸镁、 固体聚乙二醇、 十二烷基 硫酸钠和它们的混合物。 在胶嚢剂、 片剂和丸剂的情况下, 所 述剂型中也可包含緩冲剂。
相似类型的固体组合物使用赋形剂例如乳糖及高分子量聚乙 二醇等, 也可用作软胶嚢和硬胶嚢中的填充物。 片剂、 糖衣丸剂(dragees)、 胶嚢剂、 丸剂和颗粒剂的固体剂 型可与包衣和壳料如肠溶衣材和医药制剂领域公知的其他衣材一 起制备。 这些固^^剂型可任选含有遮光剂, 且其組成还可使其只 是或优先地在肠道的某个部位任选以延迟方式释放活性成分。 可 以使用的包埋组合物的实例包括高分子物质和蜡类。 如果适合, 活性化合物也可与一种或多种上述赋形剂配成微嚢形式。
供口服给药的液体剂型包括药学可接受的乳剂、 溶液剂、 混 悬剂、 糖浆剂和酏剂。 液体剂型除含有活性化合物外还可含有本 领域常用的惰性稀释剂, 例如水或其他溶剂, 增溶剂和乳化剂例 如乙醇、 异丙醇、 碳酸乙酯、 乙酸乙酯、 苄醇、 苯甲酸苄酯、 丙 二醇、 1,3-丁二醇、 二甲基曱酰胺、 油类 (特别是棉籽油、 花生油、 玉米油、 胚芽油、 橄榄油、 蓖麻油和芝麻油)、 甘油、 四氢糠醇 (tetrahydrofurfuryl alcohol), 聚乙二醇和脱水山梨糖醇的脂肪酸 酯及它们的混合物。 口服组合物除包含惰性稀释剂外还可包含辅 料, 例如湿润剂、 乳化和悬浮剂、 甜味剂、 矫味剂和香味剂。
供直肠或阴道给药的组合物优选是栓剂。 栓剂可通过将本发 明化合物与合适的非刺激性赋形剂或栽体例如可可脂、 聚乙二醇 或栓剂蜡混合来制备, 它们在室温下为固体, 但在体温下则为液 体, 因此可在直肠腔或阴道腔内熔化而释放出活性化合物。
本发明的化合物及其组合物还考虑用于局部给药。 供局部给 予本发明化合物的剂量形式包括散剂、 喷雾剂、软膏剂和吸入剂。 在无菌条件下将活性化合物与药学可接受的载体和任何所需的防 腐剂、 緩沖剂或推进剂混合。 眼用制剂、 眼软骨剂、 散剂和溶液 剂也被考虑在本发明范围内。
本发明化合物也可以脂质体形式给药。 如本领域所公知, 脂 质体通常用磷脂或其他脂类物质制得。 脂质体由分散于含水介质 中的单层或多层水化液晶所形成。 任何能够形成脂质体的无毒、 生理上可接受和可代谢的脂质均可使用。 脂质体形式的本发明组 合物除含有本发明化合物外, 还可含有稳定剂、 防腐剂、 赋形剂 等。 优选的脂类是天然和合成的磷脂和磷脂酰胆碱 (卵磷脂), 它 们可单独或者一起使用。 形成脂质体的方法是本领域公知的。 参 见例如 Prescott, Ed" Methods in Cell Biology, Volume XIV, Academic Press, New York, N.Y. 在 150 ml三口瓶中, 加入 84.6 g (70.5 ml, 1.05 mol)2-氯乙 醇及 31.6 g (1.09 mol)粉碎的多聚甲醛, 搅拌下通入干燥的氯化 氢气体, 持续 24小时。 停止搅拌后反应液分为两层; 分出下层, 以氯化钙干燥。 过滤后, 将滤液减压分馏, 收集沸程为 80-84 °C/28-30 mmHg的馏分, 得氯曱基 -2-氯乙基醚 74.3 g。
在 300mL反应瓶中, 加入 18 g(0.3 mol)异丙醇、 23.7 g(0.3 mol)吡啶及 100 ml石油醚,水浴冷却。剧烈搅拌下滴加 13.8 g(0.1 mol)三氯化磷于 40ml石油醚的溶液。 加完后, 于 50。C油浴中搅 拌反应 1小时, 滤去固体, 将滤液减压蒸去溶剂, 残留物减压蒸 馏,收集 106-108。C/60 mmHg的馏分,得三异丙基亚磷酸酯 15.8 g。
将 8.3 g (0.064 mol)氯曱基 -2-氯乙基醚加于 100ml三口瓶 中,在 90°C油浴中加热搅拌, 滴加 15.8 g (0.076 mol)异丙基亚磷 酸酯。 加完后, 于 125°C油浴中继续搅拌反应 4小时, 硅胶薄层 检测反应完全 (展开剂: 乙酸乙酯, Rf=0.6)。 减压蒸馏, 收集 118-122 "C/1.5 mmHg的馏分, 得 2- (二异丙基-膦酰基曱氧基) -乙 基氯 13.5 g。 实施例 2、 甲基 -2-ίί1-甲磺酰氧基)乙氧基 1甲基)膦酸 二异丙酯的制备

将 96.6g (约 0.30moL)( ?)-l-O-三苯甲基 -1,2-丙二醇粗品加 于、 500mL 无水四氢呋喃中, 磁力搅拌, 水水浴冷却。 分批加 入 11.3g(70%, 0.33moL)氢化钠, 约 40 min加完。 0.5 小时后去 掉冰水浴, 升至室温后反应约 2 小时, 再加热微回流 3 小时左 右, 至无气体产生。 降温至 5。C以下, 滴加 106.2g(0.30moL)对甲 苯磺酰基氧甲基膦酸二异丙酯的无水四氢呋喃 (200mL)溶液, 约 0.5 小时加完, 1 小时后去水水浴, 升至室温后反应约 30 小时。 反应结束后, 旋转蒸发溶剂, 加入乙酸乙酯 (250mL)和蒸镏水 (200mL)搅拌至残余固体物溶解, 转入分液漏斗分层, 水相再用 乙酸乙酯 (2x70mL)萃取, 合并有机层, 无水硫酸钠干燥。 过滤, 滤液减压除去溶剂, 残余物经柱层析分离(柱层析奈件: 硅胶 200 - 300 目; 洗脱剂为石油醚:乙酸乙酯 =10:1 到 5:1), 得

于 120mlDMF 中加入 DBU 29.3g, 2-氨基 -6-氯嘌呤 32.45g(0.191mol), 80°C搅拌 30分钟, 然后加入 2- (二异丙氧基- 膦酰基甲氧基) -乙基氯 50g (0.193mol), 100。C反应 8小时,冷却, 减压除去 DMF, 加入水和乙酸乙酯, 分层, 水相用乙酸乙酯提 取二次 ,合并有机层,干燥,用硅胶柱层析分离,用 MeOH:CH2Cl2 =1:30洗脱,得 11 ^6克。 实施例 4、 (R)-2-氨基 -6-氯 -9-{24双 (异丙氧基)膦酰基甲氧 基 丙基) -嘌呤 ί Π 2)的制备
在 500mL烧瓶 (配备 CaCl2干燥管)中加入 甲基 -2-[(1- 甲磺酰氧基)乙氧基】甲基 }膦酸二异丙酯 26.6g(80.0mmoL)、二甲 基甲酰胺 200mL、 2-氨基 -6-氯嘌呤 16.8g(96.0mmoL)和无水碳酸 钾 16.8g(120mmol), 磁力搅拌, 于 95°C油浴中加热反应。 4小时 后去掉油浴, 降至室温后过滤, 滤饼用二甲基甲酰胺 (2 <30mL) 洗涤,合并滤液,经旋转蒸发器减压蒸掉溶剂 (45°C, 2-5mmHg), 残余物冷却后加入乙酸乙酯 250mL和饱和盐水 lOOmL搅拌溶 解, 转入分液漏斗分层, 油层再用饱和盐水 (2x50mL)洗涤, 油 层经无水硫酸钠干燥, 过滤, 滤液经旋转蒸发器减压蒸掉溶剂, 粗品经柱层析分离(柱层析条件: 硅胶 200 ~ 300目; 洗脱剂二氯 甲烷:甲醇 =100:1到 50:1到 15:1), 柱层析后所得产物进一步用 乙酸乙酯-石油醚重结晶, 过滤、 千燥得白色固体产物; 母液经 旋转蒸发器减压蒸干,再次按上面精制粗品的方法得到第二批产 物。 共计得产物 27.1g, 熔点: 130 ~ 132。C, 比旋光度: [a] D 23 =_44·09(二氯甲烷, c=1.002). ^ NMR (CDCI3, 400ΜΗζ)δ: 7.95 (s, 1H), 5.21 (br s, 2H), 4.64-4.75 (m, 2H), 4.22 (dd, 1H), 4.07 (dd, 1H), 3.85-3.97 (m, 1H), 3.81 (dd, 1H), 3.78 (dd, 1H), 1.12-1.23 (m, 15H)。 实施例 5、 2-氨基 -6-(4 - 甲氧基苯硫基 )-9-ί2-膦酰基甲氧基 乙基 -嘌呤 -OV1)的制备
室温下, 于 30mlDMF 中依次加入 5g (12.8mmoI) II i , 2.6g(25.6mmol)三乙胺及 2.7g(19.2mmol)对甲氧基苯硫酚, 在充 氮气保护下, 于 70°C反应 4小时, 冷却, 减压除去 DMF, 加入 乙酸乙酯, 水搅拌, 分层, 水层用乙酸乙酯在提取两次, 合并有 机层, 干燥, 用硅胶柱层析分离, 用 MeOH:CH2Cl2=l:30洗脱, 得 2-氨基 -6-(4-甲氧基苯硫基) -9-{2- [双 (异丙氧基)膦酰基甲氧 基】 -乙基 }_嘌呤 (ΠΙι)的粘稠液体 3.98克。

本领域技术根据本发明特别是以上实施例的方法的教导以 及拥有的已有知识和参考文献,还可以合成得到本发明其它未示 例合成的式 I化合物,特别是还可以合成得到本发明上文所提出 的具体化合物或者所附权利要求书中列出的具体化合物。 实验例 1、 体外抗 HBV活性测定
用 Hep G 2.2.15细胞体外试验法测定了目标化合物对 HBV DNA的抑制作用和细胞毒性。
实验方法
通过定量实时荧光 PCR法测定目标化合物对 HBV DNA的 抑制作用: Hep G 2.2.15细胞培养于含 10%小牛血清的 DMEM 培养液中, 在 5% C02 培养箱中孵育。 然后将细胞接种于 96孔 板中, 细胞数 3X104, 继续培养, 当细胞密度达到 80%左右, 弃 去旧培养液,加入含不同浓度药物的新培养液,设置 3个平行孔; 每隔 2天更换培养液。 在给药后第 10天, 取 ΙΟΟμΙ上清, 通过 定量 PCR的方法,测定 HBV DNA的含量,计算 50%抑制浓度, 即为 IC5o值。
通过 MTT法测定目标化合物的细胞毒性: Hep G2细胞培养 于含 10%小牛血清的 DMEM培养液中, 在 5% C02 培养箱中 孵育。 然后将细胞接种于 96孔板中, 细胞数 5X104, 继续培养 3 天, 加入含不同浓度药物的新培养液, 设置 3个平行孔; 在给药 后第 3天,加入 MTT至 7.5mg/ml,继续培养 2小时,弃去上清, 加入含 10%吐温 X-100异丙醇, 120μ1/孔, 再加入 0.4μ1/孔, 用 酶联仪测定 540nm处的吸收, 计算 50%抑制浓度, 即为 CCS0 值。 结果见表 1。

本发明其它实施例化合物以及本发明上文所提出的具体化 合物或者所附权利要求书中列出的具体化合物也可以得到与上 表 1中本发明实施例化合物的 IC50值和 CC50值基本上相同的 结果。 实验例 2、 口服生物利用度的比较
SD雄性大鼠, 体重为 180至 220 g, 每组 3只。 所有动物给 药前禁食 12小时。 将待测样品配成浓度为 10mg/ml的 1%羧曱 基纤维素钠的悬浮液, 灌胃给药, 给药剂量相当于 20亳克 /公 斤的 2-氨基 -6-(4-甲氧基苯硫基) -9-(2-膦酰基甲氧基乙基) -嘌呤 (IVi , 602076); 将 602076配成 10mg/ml的生理盐水溶液, 尾静 脉注射给药, 给药剂量 10毫克 /公斤。 于给药前和给药后 0.25, 0.5, 0.75, 1.0, 1.5, 2.0, 3.0, 6.0, 12.0, 24.0小时, 于大鼠眼底静脉 丛釆血 0.5 mL, 参考文献(Clark Chan, et al.CIinical Pharmaco-kinetics of Alamifovir and Its Metabolites Antivicrob Agents Chemother, 2005, 49(5):1813-1822.)的方法, 测定不同时 间点血液中 602076的浓度。
将血药浓度-时间数据输入计算机, 采用非室模型法计算药 物动力学参数, 用梯形法计算 AUC。.oo值, 由受试大鼠分别口服 受试样品与静注 602076后所测定的 602076的平均血药浓度-时 间曲线下面积 (AUC。_),计算出大鼠口服各前体化合物后活性代 谢物 602076的绝对生物利用度 (F)。

1、 式 I所示的无环核苷膦酸酯衍生物或其药学上可接受的 盐、 异构体、 水合物或溶 合物:

各 R2 各自独立地选自 -R3或 -OR3;
各 R3 各自独立地选自 CrC8烷基、 C3-C8环烷基。
2、 权利要求 1所述的无环核苷膦酸酯衍生物或其药学上可 接受的盐、 异构体、 水合物或溶剂合物, 其中两个 R2相同; 或 者, 其中两个 R2不同。
3、 权利要求 1所述的无环核苷膦酸酯衍生物或其药学上可 接受的盐、 异构体、 水合物或溶剂合物, 其特征在于以下任一项 或多项:
i) R2为 -R3;
ii) R2为- OR3;
iii) 一个 R2为 -R3, 另一个 R2为 -OR3
4、 权利要求 1所述的无环核苷膦酸酯衍生物或其药学上可 接受的盐、 异构体、 水合物或溶剂合物, 其中每次出现的 R3各 自独立地选自 CrC6烷基或 C3-C6环烷基; 优选地, 每次出现的 R3各自独立地选自 C2-C6烷基或 C4-C6环烷基。
5、 权利要求 1所述的无环核苷膦酸酯衍生物或其药学上可 接受的盐、 异构体、 7J合物或溶剂合物, 其中每次出现的 R3各 自独立地选自甲基、 乙基、 丙基、 异丙基、 丁基、 异丁基、 叔丁 基、 特丁基、 正戊基、 异戊基、 新戊基、 环丙基、 环丁基、 环戊 基、 环己基、 -CH(CH2CH3)2等; 优选地, 每次出现的 R3各自独 立地选自乙基、 丙基、 异丙基、 丁基、 异丁基、 异戊基、 新戊基、 环戊基、 环己基、 -CH(CH2CH3)2等。
6、 权利要求 1所述的无环核苷膦酸酯衍生物或其药学上可 接受的盐、 异构体、 7J合物或溶剂合物, 其选自:

2-氨基 -6-(4-甲氧基苯硫基) -9-{2- [双 (异丁酰氧基甲氧基)膦 酰基甲氧基] -乙基 }-嘌呤;

2-氨基 -6-(4-甲氧基苯硫基) -9-{2- [双 (新戊酰氧基甲氧基)膦 酰基甲氧基] -乙基 }-嘌呤;
2-氨基 -6-(4-甲氧基苯硫基) -9-{2- [双 (环己甲酰氧基曱氧基) 膦酰基曱氧基] -乙基 }-嘌呤;

8、 制备权利要求 1所述的无环核苷膦酸酯衍生物或其药学 上可接受的盐、 异构体、 水合物或溶剂合物的方法, 其包括以下 步骤:
(i)在适宜的溶剂(例如 DMF)中,在适宜的试剂(例如 DBU(即 二环脒, Bicyclk amidine), 特别是在 R1为氢时)或碳酸钾 (特别 是在 为甲基时)存在下, 在高温下(例如 60~140°C , 例如
80-120°C, 或例如 80-100。C)下, 使式

10、权利要求 1所述的无环核苷膦酸酯衍生物或其药学上可 接受的盐、异构体、 水合物或溶剂合物或者权利要求 9所述药物 组合物在制备用于治疗和 /或预防与病毒感染相关的疾病的药物 中的用途;优选的,其中所述的病毒是肝炎病毒,例如乙肝病毒; 优选的, 其中所述的与病毒感染相关的疾病是肝炎, 例如乙型肝 炎。
11、 在有需要的哺乳动物中治疗和 /或预防与病毒感染相关 的疾病的方法, 该方法包括给有需要的哺乳动物施用治疗和 /或 预防有效量的权利要求 1 所述的无环核苷膦酸酯衍生物或其药 学上可接受的盐、异构体、 水合物或溶剂合物或者权利要求 9所 述药物组合物。
12、 用于治疗和 /或预防与病毒感染相关的疾病的化合物, 所述化合物是如权利要求 1 所述的无环核苷膦酸酯衍生物或其 药学上可接受的盐、 异构体、 水合物或溶剂合物。
