Description

ビーズ封入方法、ターゲット分子を検出する方法、アレイ、キット及びターゲット分子検出装置
 本発明は、ビーズ封入方法、ターゲット分子を検出する方法、アレイ、キット及びターゲット分子検出装置に関する。
 タンパク質、核酸等の生体分子を個々識別した態様で観察することにより種々の測定を行なう方法として、一分子測定が知られている。この一分子測定を行なうための手法がいくつか知られている。
 特許文献1には、一分子酵素活性検出に用いられるマイクロチャンバが記載されている。このマイクロチャンバは、液滴を封入することができ、この液滴により充填可能な容量が1000fL(フェムトリットル)以下である容器部を有する。容器部は、第1の部材と第2の部材とを貼り合わせることにより、少なくとも第1の部材又は第2の部材が有する窪みによって構成される。この液滴中で酵素反応を行うことにより、反応生成物の分子数が極めて少数でも濃度を高くすることができるため、酵素一分子の活性を検出することができる。
 非特許文献1には、液滴がオイルで覆われており、外部から液滴に直接アクセス可能なフェムトリットルオーダーの液滴のアレイを用いて、一分子酵素アッセイを行なう方法が記載されている。このアレイでは、親水性表面に高さ17nmの疎水性領域が形成されることにより、親水性領域のパターンが形成されている。
 非特許文献2には、一分子の酵素結合免疫吸着アッセイ(ELISA)を用いてタンパク質を検出する方法が記載されている。この方法では、タンパク質特異的な抗体で覆われた微細なビーズによって微量のタンパク質を捕捉し、ビーズとタンパク質との複合体を蛍光標識させる。この複合体を含むビーズを遠心力によって反応チャンバーに導入し、タンパク質を捕捉しているビーズの数を数えることによって、タンパク質を定量測定する。

 低濃度の疾病マーカー等を検出し、疾病、感染症等の早期発見を行なうために、バイオセンシング技術のさらなる高感度化が求められている。例えば、体積1mm3の腫瘍に含まれる100万個の癌細胞からマーカータンパク質(各細胞から100分子ずつ)が5Lの血中に分泌された場合、このタンパク質の血中濃度は30aM程度である。このような非常に低濃度のターゲット分子を検出しうる技術が求められている。
 このようなターゲット分子を検出するために、上述した一分子酵素アッセイを用いて一分子単位の感度で検出する方法が考えられる。すなわち、ターゲット分子を特異的にフェムトリットルオーダーの液滴(超微小液滴)に封入した後、酵素によって標識された抗体等をターゲット分子に接続し、上述した方法で標識酵素の活性を検出する方法である。ターゲット分子を特異的に超微小液滴に封入する方法としては、ターゲット分子に特定的に結合する別の抗体等で標識されたビーズ等を用いる方法が考えられる。この方法では、ビーズにターゲット分子を結合させた後、ビーズを超微小溶液に封入する。
 ここで、溶液中に極わずかにしか存在しないターゲット分子、例えば上述したような30aM程度のターゲット分子等を効率的に検出するためには、100万個程度の非常に多数の超微小液滴アレイを用意し、このアレイにビーズを捕捉する必要がある。
 しかし、非特許文献2に記載の方法では、ビーズを強い遠心力によりアレイに導入する必要があり、時間と労力を要する。また、非特許文献2に記載の方法で用いているアレイの数は5万個程度であり、100万個程度のアレイに適用することは極めて困難である。そのため、非特許文献2に記載の方法では、多数のビーズをアレイに効率よく封入させることが困難である。また、特許文献1及び非特許文献1には、このような問題を解決する方法は記載されていない。
 本発明の目的は、多数のビーズをアレイに効率よく封入させる技術を提供することにある。
 上記の課題を解決するために、本発明に係るビーズ封入方法は、ビーズを1個のみ収容可能な複数の収容部が、疎水性の上面を有する側壁によって互いに隔てられて形成されている下層部と、当該下層部における当該収容部が形成されている面に対向している上層部との間の空間に、当該ビーズを含む親水性溶媒を導入するビーズ導入工程と、上記ビーズ導入工程の後に上記空間に疎水性溶媒を導入する疎水性溶媒導入工程とを含むことを特徴とする。
 上記の課題を解決するために、本発明に係るアレイは、複数の収容部が疎水性の上面を有する側壁によって互いに隔てられて形成されている下層部と、上記下層部における上記収容部が形成されている面に対して空間を隔てて対向している上層部とを備えていることを特徴とする。
 本発明に係るビーズ封入方法を用いれば、多数のビーズをアレイに効率よく封入させることができるので、低濃度のターゲット分子を高感度に検出可能な技術に寄与することが可能となる。

 以下、本発明の一実施形態について、詳細に説明する。
 〔ビーズ封入方法〕
 本実施形態に係るビーズ封入方法について、図1(a)~(e)を参照して説明する。図1(a)~(e)は、本発明に係るビーズ封入方法を説明するための模式図であり、アレイ1の側方断面図を示す。
 本実施形態では、下層部10と上層部20とを備えたアレイ1にビーズ41、41’を封入する場合について説明する。下層部10は、ビーズ41、41’を1個のみ収容可能な複数の収容部13が、疎水性の上面を有する側壁12によって互いに隔てられて形成されている。また、上層部20は、下層部10における収容部13が形成されている面に対向している。
 ビーズは、平均粒子径1μm~4μmであることが好ましい。これにより、アレイへの効率的な封入とアレイの高密度化とを達成することができる。なお、「平均粒子径」とは、ここでは電子顕微鏡観察又は動的光散乱法を用いて測定した数値をいう。
 本実施形態では、ターゲット分子を特異的に捕捉するビーズを用いる場合について説明するが、本発明は特にこれに限定されない。本実施形態では、封入するビーズは、ターゲット分子を捕捉していないビーズ41と、ターゲット分子を捕捉したビーズ41’との混合物である。
 ターゲット分子を特異的に捕捉するビーズとして、例えば、ターゲット分子を特異的に捕捉するための分子が結合されているビーズを用いることができる。ターゲット分子を特異的に捕捉するための分子は、ビーズの表面にある修飾基に、例えばリンカーを介して結合させてもよい。例えば、アミノ基修飾ビーズの表面にあるアミノ基に、N-ヒドロキシスクシニミド(N―hydroxysuccinimide)等を持つ架橋剤を介して共有結合させることにより結合させることができる。
 「ターゲット分子」とは、検出対象である分子(標的分子)、すなわちここではビーズに捕捉させることによって検出しようとする分子をさす。ターゲット分子としては、例えばタンパク質、核酸、糖等の生体分子、ならびにウイルス粒子そのものなどが挙げられる。
 ターゲット分子を特異的に捕捉するための分子(以下、「ターゲット捕捉分子」ともいう。)としては、ターゲット分子に応じて選択すればよく、例えばタンパク質、抗体、核酸等を用いることができる。なお、ターゲット捕捉分子は、ビーズ1個あたり10万分子以上結合されていることが好ましい。例えばターゲット捕捉分子が抗体の場合には、解離定数がnMオーダー程度であるが、上述した構成であれば、ビーズとターゲット分子とを反応させる際のターゲット捕捉分子の濃度を充分に高くすることができる(例えば、ビーズの濃度が8×106粒子/mLの場合、ターゲット捕捉分子が約1nMとなる)。
 本実施形態に係るビーズ封入方法は、ビーズ導入工程と、脱気工程と、疎水性溶媒導入工程とを含む。各工程について、以下に詳細に説明する。

 ビーズ導入工程は、下層部10と上層部20との間の空間30に、ビーズ41、41’を含む親水性溶媒42を導入する工程である。親水性溶媒42を導入する方法としては、下層部10と上層部20との間の空間30に、下層部10と上層部20とが対向する面に平行な方向に導入すればよい。例えば、上層部20及び下層部10の少なくとも一方に形成されている貫通孔(図示せず)から、空間30内に導入してもよい。
 親水性溶媒42としては、例えば、水、親水性アルコール、親水性エーテル、ケトン、ニトリル系溶媒、ジメチルスルホキシド(DMSO)、及びN,N-ジメチルホルムアミド(DMF)からなる群より選択される少なくとも1つ又はこれを含む混合物等を好適に用いることができる。親水性アルコールとしては、例えばエタノール、メタノール、プロパノール、グリセリン等が挙げられる。親水性エーテルとしては、例えばテトラヒドロフラン、ポリエチレンオキサイド、1,4-ジオキサン等が挙げられる。ケトンとしては、例えばアセトン、メチルエチルケトン等が挙げられる。ニトリル系溶媒としては、例えばアセトニトリル等が挙げられる。
 親水性溶媒42は、ビーズ41、41’の他に、例えばビーズ41’に捕捉されたターゲット分子を特異的に検出するための物質をさらに含んでいてもよい。このような物質としては、例えば、ビーズ41’に捕捉されたターゲット分子又はこれに特異的に結合した分子に結合した所定の酵素によって分解されて、蛍光物質を遊離する蛍光基質などであってもよい。ターゲット分子に特異的に結合する分子としては、例えば二次抗体、核酸等であってもよい。また、所定の酵素としては、例えばβ-ガラクトシダーゼ、ペルオキシダーゼ等が挙げられる。蛍光基質としては、例えばFluorescein-di-β-galactopyranoside(FDG)、Amplex red(登録商標)等が挙げられる。
 (脱気工程)
 脱気工程について、図1(c)を参照して説明する。
 脱気工程は、ビーズ導入工程の後であって疎水性溶媒工程より前に、下層部10と上層部20との間の空間30内を脱気する工程である。脱気する方法としては、例えば、アレイ1を減圧環境下において放置する方法等を好適に用いることができる。具体的には、約0.1気圧の減圧デシケータ内にアレイ1を約30秒放置する方法などである。
 本発明において、脱気工程は必須の工程ではないが、脱気工程を行なうことにより、収容部13内の空気が除去され、ビーズ41、41’を含む親水性溶媒42を収容部13内に効率よく導入することができる。したがって、ビーズ41、41’を収容部13内に効率よく封入させることが可能になるため、脱気工程を行なうことがより好ましい。
 (疎水性溶媒導入工程)
 疎水性溶媒導入工程について、図1(d)~(e)を参照して説明する。
 疎水性溶媒導入工程は、下層部10と上層部20との間の空間30に疎水性溶媒43を導入する工程である。疎水性溶媒導入工程は、ビーズ導入工程の後に行われる工程であり、脱気工程の後に行われることがより好ましい。
 疎水性溶媒43は、ビーズ導入工程で用いた親水性溶媒42と混ざり合いにくい溶媒であればよい。疎水性溶媒43として、例えば飽和炭化水素、不飽和炭化水素、芳香族炭化水素、シリコーンオイル、パーフルオロカーボン、ハロゲン系溶媒、及び疎水性イオン液体からなる群より選択される少なくとも1つ又はこれを含む混合物等を好適に用いることができる。飽和炭化水素としては、例えばアルカン、シクロアルカンなどが挙げられる。アルカンとしては、例えばデカン、ヘキサデカン等が挙げられる。不飽和炭化水素としては、例えばスクアレン等が挙げられる。芳香族炭化水素としては、例えばベンゼン、トルエン等が挙げられる。パーフルオロカーボンとしては、例えばフロリナート(登録商標)FC40(SIGMA社製)等が挙げられる。ハロゲン系溶媒としては、例えばクロロホルム、塩化メチレン、クロロベンゼン等が挙げられる。疎水性イオン液体とは少なくとも水中では解離しないイオン液体をさし、例えば1―Butyl―3―methylimidazolium Hexafluorophosphate等が挙げられる。イオン液体とは、室温において液体で存在する塩をさす。
 疎水性溶媒導入工程を行なうことにより、収容部13の各々に、疎水性溶媒43により覆われたドロップレット(液滴)を効率よく形成させることができる。また、各ドロップレット内にビーズ41、41’を効率よく封入することができる。
 本実施形態では、下層部10と上層部20との間の空間30を用いてビーズ41、41’を導入することにより、大きな面積(例えば1cm2以上の面積)に多数作製した収容部13の各々に、ビーズを高い効率で封入することができる。
 本実施形態によれば、多数の収容部を有する大面積のドロップレットアレイを実現することが可能である。例えば、100万個以上の収容部を有するアレイであっても、効率よくビーズ41、41’を各収容部に封入することが可能である。したがって、本実施形態であれば、ターゲット分子を高感度に検出することができるため、0.1aM程度の非常に低濃度のターゲット分子であっても検出することが可能となる。
 〔ターゲット分子を検出する方法〕
 次に、本実施形態に係るターゲット分子を検出する方法について、説明する。
 本実施形態に係るターゲット分子を検出する方法は、反応工程と、ビーズ封入工程と、検出工程とを含む。
 本実施形態では、ビーズとして、ターゲット分子を特異的に捕捉するビーズを用いる。例えば、ビーズは、ターゲット分子を特異的に捕捉するための分子が結合されているものであってもよい。なお、ビーズ、ターゲット分子、及びターゲット分子を特異的に捕捉するための分子については、本実施形態に係るビーズ封入方法において例示したものと同様のものを好適に用いることができる。
 反応工程は、ビーズとターゲット分子とを反応させる工程である。例えばビーズを含む溶液と、ターゲット分子を含む溶液とを混合させることにより反応させることができる。
 ビーズ封入工程は、反応工程において反応させたビーズを用いて上述したビーズ封入方法を行なう工程である。すなわち、ビーズ封入工程は、ビーズ導入工程と、疎水性溶媒導入工程とを含む工程、または、ビーズ導入工程と、脱気工程と、疎水性溶媒導入工程とを含む工程である。なお、ビーズ導入工程、脱気工程及び疎水性溶媒導入工程については、上述した「ビーズ封入方法」における各工程と同様に行なうことができるため、ここでは説明を省略する。
 検出工程は、ビーズ封入工程の後に、収容部13の各々にターゲット分子を捕捉したビーズ41’が収容されているか否かを検出する工程である。
 ターゲット分子を捕捉したビーズ41’が収容されているか否かを検出する方法としては、例えば抗原抗体反応、ストレプトアビジン-ビオチン反応、核酸の相補的な結合等、公知の分子認識反応を好適に用いることができる。例えば、ターゲット分子又はこれに特異的に結合した分子に結合した所定の酵素によって、蛍光基質が分解されて遊離した蛍光物質を検出する方法が挙げられる。蛍光物質の検出方法としては、蛍光顕微鏡、イメージセンサ等を用いて各収容部における蛍光強度を検出する方法などが挙げられる。
 また、検出工程においては、各収容部13にビーズ41又はビーズ41’が収容されているか否かをも検出することが好ましい。各収容部13にビーズ41又はビーズ41’が収容されているか否かを検出する方法としては、例えば顕微鏡下においてビーズ41又はビーズ41’の有無を観察することにより検出することができる。また、ビーズ41、41’の有無を検出する方法として、ビーズによる散乱光を検出する方法、電界効果トランジスタ(FET)による電位計測を利用する方法などもある。
 検出工程の結果、ビーズ41又はビーズ41’が収容されている収容部13の数と、ターゲット分子を捕捉したビーズ41’が収容されている収容部13の数とを用いて、ビーズの全数のうちターゲット分子を捕捉したビーズの数の割合を算出することができる。これにより、ターゲット分子の濃度を定量化することが可能となる。
 本実施形態であれば、多数の収容部を有する大面積のドロップレットアレイを実現させることが可能であり、100万個以上の収容部を有するアレイであっても、効率よくビーズ41、41’を各収容部に封入することが可能である。したがって、本実施形態であれば、ターゲット分子を高感度に検出することができるため、10aM程度の非常に低濃度のターゲット分子であっても検出することが可能となる。
 〔アレイ〕
 次に、本実施形態に係るアレイ1の構成について、図1(a)を参照して詳細に説明する。アレイ1は、本実施形態に係るビーズ封入方法に用いるためのアレイであってもよいし、本実施形態に係るターゲット分子を検出する方法に用いるためのアレイであってもよい。
 アレイ1は、下層部10と、上層部20とを備えている。
 下層部10は、板状部材11と疎水性の上面を有する側壁12とを備えている。下層部10には、複数の収容部13が側壁12によって互いに隔てられて形成されている。
 板状部材11は親水性表面を有していることが好ましい。「親水性表面」とは、親水性溶媒との親和性が疎水性溶媒との親和性よりも高い表面を指す。板状部材11としては、固体材料であればよいが、例えばガラス、シリコン、高分子樹脂等を用いることができる。
 側壁12は、板状部材11の表面上、好ましくは親水性表面上に設けられている、複数の収容部13の各々を隔てる構造物である。側壁12は、疎水性の上面を有している。「疎水性」とは、ここでは「親油性」と同じ意味で用いられ、疎水性溶媒との親和性が親水性溶媒との親和性よりも高いことをいう。
 なお、側壁12は、その上面、すなわち上層部20と対向する面が疎水性であればよく、側面、すなわち収容部13内の内壁は、疎水性であっても親水性であってもよい。
 例えば、側壁12は、親水性の構造物と、その上面に形成されている疎水性層とにより構成されていてもよい。親水性の構造物には、例えばガラス、シリコン、高分子樹脂等を用いることができる。疎水性層には、例えば撥水性の樹脂、フッ素系高分子樹脂等を用いることができる。フッ素系高分子樹脂としては、例えばアモルファスフッ素樹脂等が挙げられる。アモルファスフッ素樹脂は、高い疎水性を有し、かつ、生体試料に対する毒性が低いという理由で、好ましく用いられる。
 上記アモルファスフッ素樹脂としては、例えば、CYTOP(登録商標)、TEFLON(登録商標)AF2400、およびTEFLON(登録商標)AF1600から選択した少なくとも1つを好適に用いることができる。中でも、微細加工が容易であるという理由で、CYTOP(登録商標)が最も好ましい。
 また例えば、側壁12は、疎水性の材料により構成されていてもよい。側壁12として、例えばフッ素系高分子樹脂、パラキシリレン系高分子樹脂等を用いることができる。フッ素系高分子樹脂としては、例えばアモルファスフッ素樹脂等が挙げられる。アモルファスフッ素樹脂としては上述の樹脂を好適に用いることができる。
 側壁12は、板状部材11上に複数の収容部13が形成されるように構成されていればよく、例えば収容部13が形成される位置に孔が形成されている板形状の構造物であってもよい。
 板状部材11の表面からの側壁12の高さ(垂直方向の厚み)は、収容部13に収容されたビーズ41、41’が、後述する疎水性溶媒導入工程において収容部13から排出されない程度の高さであればよい。例えば収容部13に収容されたビーズ41、41’の大部分、好ましくは全てが側壁12の上面よりも下に収まる高さであってもよい。
 なお、側壁12の高さは、ビーズ41、41’を収容部13に効率よく収容するという観点から、ビーズ41、41’の平均粒子径の1倍以上であることが好ましい。また側壁12の高さは、ビーズ41、41’を1個のみ収容部13に収容させるという観点から、ビーズ41、41’の平均粒子径の1.5倍以下であることが好ましい。
 複数の収容部13の各々は、ビーズ41、41’を1個のみ収容可能な凹部であり、互いに側壁12によって隔てられている。収容部13は、板状部材11の表面の一部を底面としており、底面が親水性である。
 収容部13は、ビーズ41、41’が1個のみ収容される形状及び大きさであればよい。収容部13の底面及び側面によって囲まれた領域の形状は、例えば円柱形状、角柱形状等であってもよい。
 各収容部13の水平方向の幅A(水平方向の断面が円であればその直径、正方形であればその一辺の長さなど)は、ビーズ41、41’の平均粒子径よりも大きければよいが、例えばビーズ41、41’の平均粒子径の1.5倍~2倍であることが好ましい。各収容部13の深さは、本実施形態では側壁12の高さと同じである。なお、本発明における収容部の深さは、ビーズを収容部に効率よく収容するという観点から、ビーズの平均粒子径の1倍以上であることが好ましい。また本発明における収容部の深さは、ビーズを1個のみ収容部に収容させるという観点から、ビーズの平均粒子径の1.5倍以下であることが好ましい。
 本実施形態では、収容部13の底面が親水性であり、かつ側壁12の上面が疎水性である。これにより、後述するビーズ導入工程において、ビーズ41、41’を含む親水性溶媒42を効率よく収容部13の中に導入することができるとともに、後述する疎水性溶媒導入工程において疎水性溶媒43が収容部13に入り込むことを防止することができる。したがって、ビーズ41、41’を含む液滴を含む収容部13を、疎水性溶媒によって効率よく密閉することができる。
 上層部20は、板状部材21と、疎水性層22とを備えている。疎水性層22は、板状部材21における、下層部10に対向する面上に設けられている。板状部材21としては、例えばガラス、シリコン、高分子樹脂等を用いることができる。また、疎水性層22としては、例えば撥水性の樹脂、フッ素系高分子樹脂等を用いることができる。フッ素系高分子樹脂としては、例えばアモルファスフッ素樹脂等が挙げられる。
 上層部20は、下層部10における収容部13が形成されている面に対して空間30を隔てて対向している。すなわち、側壁12と疎水性層22との間に空間30があいている。この空間30は、流路を構成する。この構成により、アレイ1はフローセル構造となっている。
 空間30は、下層部10と上層部20との間に、下層部10と上層部20とが互いに対向する面と平行な方向に流体を流すための流路として用いられうる。
 側壁12の上面と疎水性層22板状部材21との間の距離、すなわち空間30の垂直方向の幅は、少なくともビーズ14、14’の平均粒子径より大きければよく、10~150μmであることが好ましい。
 下層部10又は上層部20には、空間30に流体を導入するための貫通孔(図示せず)が形成されていてもよい。例えば、下層部10は、収容部13が形成されている領域と、収容部13が形成されていない領域とを有していてもよい。そして、下層部10における収容部13が形成されていない領域、又は上層部20におけるこの領域と対向する部分に、貫通孔が形成されていてもよい。
 本実施形態においては、空間30の上面が疎水性層22の表面であり、空間30の下面が側壁12の上面及び収容部13である。したがって、空間30のうち、収容部13の底面以外の部分は全て疎水性となっている。これにより、後述するビーズ導入工程において、ビーズ41、41’を含む親水性溶媒42を効率よく各収容部13内に導入することができる。また、後述する疎水性溶媒導入工程において、各収容部13内に疎水性溶媒43を入り込ませることがない。したがって、疎水性溶媒43を空間30内に導入することにより、各収容部13にビーズ41、41’が封入されたドロップレットを効率よく形成させることができる。
 本実施形態に係るアレイ1の一例としては、例えば100万個以上の収容部が形成されたアレイである。このような大面積のアレイであっても、本実施形態に係るビーズ封入方法又はターゲット分子を検出する方法を用いることにより、効率よくビーズを各収容部に封入することが可能である。したがって、本実施形態であれば、ターゲット分子を高感度に検出することができるため、0.1aM程度の非常に低濃度のターゲット分子をも検出可能なアレイを提供することができる。
 〔キット〕
 次に、本実施形態に係るキットの構成について説明する。
 本実施形態に係るキットは、アレイ1とビーズ41とを少なくとも備える。アレイ1としては、上述した構成のアレイ1を好適に用いることができる。アレイ1の収容部13の各々は、このキットが備えるビーズ41を1個のみ収容可能であるように構成されている。
 このキットが備えるビーズ41は、ターゲット分子を特異的に捕捉するものであってもよく、例えばターゲット分子に特異的に結合する分子が結合したものであってもよい。ターゲット分子、及びこれに特異的に結合する分子としては、前述したものを好適に用いることができる。
 また、このキットは、ターゲット分子を特異的に検出するための物質をさらに備えていてもよい。ターゲット分子を特異的に検出するための物質としては、上述したものを好適に用いることができる。また、キットは、水溶性溶媒、疎水性溶媒等をさらに備えていてもよい。
 〔ターゲット分子検出装置〕
 次に、本実施形態に係るターゲット分子検出装置50の構成について、図2を参照して説明する。図2は、本発明に係るターゲット分子検出装置の一実施形態を模式的に示す図である。
 本実施形態に係るターゲット分子検出装置50は、アレイ1と、イメージセンサ51と、光源52とを備える。アレイ1としては、上述した構成を好適に用いることができるため、ここでは説明を省略する。
 イメージセンサ51は、収容部13の各々から、ターゲット分子を捕捉したビーズが収容された場合に発せられる光を検出するセンサである。例えば、ターゲット分子又はこれに特異的に結合した分子に結合した所定の酵素によって、蛍光基質が分解されて発せられる蛍光を検出するセンサであってもよい。イメージセンサ51としては、例えばCMOSイメージセンサ等を好適に用いることができる。
 光源52は、アレイ1に光を照射する光源である。なお、図2では、光源52はアレイ1の上に設けられているが、本発明は特にこれに限定されず、例えばアレイ1の横から光を照射するものであってもよい。
 なお、アレイ1とイメージセンサ51との間には、干渉フィルタ、ライトガイドアレイ等が設けられていてもよい。また、光源52とアレイ1との間には励起フィルタ等が設けられていてもよい。
 本実施形態であれば、アレイ1とイメージセンサ51とが直結されているため、顕微鏡等の他の装置を用いることなく、各収容部13にターゲット分子を捕捉したビーズが収容されているか否かを容易に検出することができる。したがって、簡便かつ高速に検出することが可能になる。また、安価な商品として提供することができる。
 本願には、以下の発明が含まれる。
 本発明に係るビーズ封入方法は、ビーズを1個のみ収容可能な複数の収容部が、疎水性の上面を有する側壁によって互いに隔てられて形成されている下層部と、当該下層部における当該収容部が形成されている面に対向している上層部との間の空間に、当該ビーズを含む親水性溶媒を導入するビーズ導入工程と、上記ビーズ導入工程の後に上記空間に疎水性溶媒を導入する疎水性溶媒導入工程とを含むことを特徴とする。
 また、本発明に係るビーズ封入方法では、上記ビーズ導入工程の後であって上記疎水性溶媒導入工程より前に、上記空間内を脱気する脱気工程をさらに含むことが好ましい。
 また、本発明に係るビーズ封入方法では、上記親水性溶媒は、水、親水性アルコール、親水性エーテル、ケトン、ニトリル系溶媒、ジメチルスルホキシド、及びN,N-ジメチルホルムアミドからなる群より選択される少なくとも1つ又はこれを含む混合物であることが好ましい。
 また、本発明に係るビーズ封入方法では、上記疎水性溶媒は、飽和炭化水素、不飽和炭化水素、芳香族炭化水素、シリコーンオイル、パーフルオロカーボン、ハロゲン系溶媒、及び疎水性イオン液体からなる群より選択される少なくとも1つ又はこれを含む混合物であることが好ましい。
 上記の課題を解決するために、本発明に係るターゲット分子を検出する方法は、ターゲット分子を特異的に捕捉するビーズと、当該ターゲット分子とを反応させる反応工程と、上記反応工程の後に、上記ビーズを用いて上述したいずれかのビーズ封入方法を行なうビーズ封入工程と、上記ビーズ封入工程の後に、上記収容部の各々に上記ターゲット分子を捕捉したビーズが収容されているか否かを検出する検出工程とを含むことを特徴とする。
 また、本発明に係るターゲット分子を検出する方法では、上記ビーズには、上記ターゲット分子に特異的に結合する分子が結合していることが好ましい。
 本発明に係るアレイは、複数の収容部が疎水性の上面を有する側壁によって互いに隔てられて形成されている下層部と、上記下層部における上記収容部が形成されている面に対して空間を隔てて対向している上層部とを備えていることを特徴とする。
 また、本発明に係るアレイでは、上記収容部の各々は、底面が親水性であることが好ましい。
 また、本発明に係るアレイでは、上記上層部における上記下層部に対向する面が疎水性であることが好ましい。
 また、本発明に係るアレイでは、上記上層部及び上記下層部の少なくとも一方に、上記空間に流体を導入するための貫通孔が形成されていることが好ましい。
 上記の課題を解決するために、本発明に係るキットは、上述したいずれかのアレイと、ビーズとを備え、上記収容部の各々は、上記ビーズを1個のみ収容可能であることを特徴とする。
 上記の課題を解決するために、本発明に係るターゲット分子検出装置は、上述したいずれかのアレイと、上記収容部の各々から、ターゲット分子を捕捉したビーズが収容された場合に発せられる光を検出するイメージセンサとを備えることを特徴とする。
 本発明は上述した各実施形態に限定されるものではなく、請求項に示した範囲で種々の変更が可能である。以下に実施例を示し、本発明の実施の形態についてさらに詳しく説明する。もちろん、本発明は以下の実施例に限定されるものではなく、細部については様々な態様が可能であることはいうまでもない。
 各実施例で用いた材料及び方法について説明する。
 (材料)
 各実施例では、ターゲット分子として、β-ガラクトシダーゼでラベルされたストレプトアビジン(Streptavidin、SIGMA社より購入)(以下、単に「ストレプトアビジン」ともいう。)を用いた。また、ビーズとして、平均粒子径3μmのアミノビーズ(材質:ポリスチレン;micromer-NH2-3μm;Micromod社より購入)をビオチン化したビオチン化ビーズを用いた。
 (ビオチン化ビーズの作製)
 以下の方法により、アミノビーズのアミノ基にNHS-PEO4-Biotinを反応させ、アミノビーズをビオチン化した。
 まず、アミノビーズ250μLにバッファーA(100mMリン酸バッファー,pH8.0)を750μL加えた。
 次に、10000rpm,10分,4℃で遠心してアミノビーズを集め、500μLのバッファーAに懸濁した(懸濁液A)。そして、懸濁液AにNHS-PEO4-Biotin(2μg/50μL DMSO)を50μL加え、少なくとも3時間、ゆるやかに混合しながら25℃で反応させた(end-over-endでチューブをmixした)。
 次に、得られたビオチン化ビーズを洗浄した。10000rpm,10分,4℃で遠心してビオチン化ビーズを集め、水相をピペットマンで取り除いた。このビオチン化ビーズの沈殿に1mLのバッファーAを加え、懸濁させた。これを6回繰り返し、未反応のNHS-PEO4-Biotinを除いた。そして、500μLのバッファーAに懸濁した(懸濁液B)。この懸濁液Bを4℃で保存した。
 次に、懸濁液Bにおけるビオチン化ビーズの濃度を調べた。血球計算盤を用いて、一定体積内のビオチン化ビーズの個数をカウントすることにより、ビオチン化ビーズの濃度を算出した(約3.0×108ビーズ/mL)。なお、カウントしやすくするために、例えば、懸濁液BをバッファーAにより5倍程度に希釈してカウントした。
 以上の方法により、ビオチン化ビーズを得た。
 (ストレプトアビジンの捕捉)
 次に、以下の方法により、ビオチン化ビーズを用いてストレプトアビジンを捕捉した。
 まず、ビオチン化ビーズを希釈した(8×106ビーズ/500μL)。また、β-ガラクトシダーゼでラベルされたストレプトアビジンを、バッファーB(100mMリン酸バッファー,pH8.0,0.1% TWEEN20(detergent)を含む)により目的の濃度の2倍の濃度に希釈した(全量500μL)。
 そして、ビオチン化ビーズ500μLと、希釈したストレプトアビジン500μLとをチューブ内で混合し(全量1mL)、ゆるやかにチューブを上下させ、25℃で30分反応させた。
 次に、10000rpm,10分,4℃で遠心して反応後のビーズ(ストレプトアビジンとビオチン化ビーズとの複合体と、未反応のビオチン化ビーズとの混合物)を集め、水相をピペットマンで取り除いた。このビーズの沈殿に1mLのバッファーAを加え、懸濁した。これを4回繰り返すことによって洗浄し、未反応のターゲット分子を除いた。
 次に、洗浄後のビーズの沈殿に、バッファーC(100mMリン酸バッファー,pH7.5,1mM MgCl2)を15μL加えて懸濁した。ビーズの濃度は最終的に約6.5×106ビーズ/15μL バッファーCとなった。
 (アレイの作製)
 次に、以下の方法により、図1(a)~(e)に示すアレイ1と同様のフローセル構造のアレイを作製した。以下の説明において、アレイ1における構成要素と同じ機能を有する構成要素には同じ符号を付して説明する。
 まず、親水/疎水パターンガラス(下層部10)と、上面ガラス(上層部20)(縦24mm×横26mm×厚さ5mm、SiO2、直径1mmの貫通孔あり)とを準備した。
 (親水/疎水パターンガラスの作製)
 親水/疎水パターンガラスを作製するために具体的に用いた方法を、図7を参照して説明する。図7は、本発明の一実施例において親水/疎水パターンガラスを作製する方法を説明するための図である。
 本実施例では、フォトリソグラフィとドライエッチングとにより、ガラス上に親水/疎水パターンを作製した。親水/疎水パターンの作製では、CYTOP(登録商標)塗布工程、フォトリソグラフィ工程、エッチング・レジスト除去工程の3つの工程を行なった。
 CYTOP(登録商標)塗布工程では、まず、縦24mm×横32mmのガラス(商品名:NEO MICRO COVER GLASS Thickness No.1;MATSUNAMI社製)(板状部材11)上にCYTOP(登録商標)CTL-809(商品名、ASAHI GLASS社製)を塗布し、疎水性の樹脂層61を形成させた。
 続いて、フォトリソグラフィ工程では、ポジ型のフォトレジスト62(商品名:AZ-4903、AZ Electronic Materials USA社製)を塗布した。次に、目的のパターンのフォトマスク63を介して上部からUVを露光し、アルカリによる現像処理を行なった。この現像処理によって、UVが照射された部分のみ、フォトレジスト62が溶解し、疎水性の樹脂層61の面がむき出しになった。
 その後、エッチング・レジスト除去工程では、このガラスを、一部が溶解したフォトレジスト62’を介してO2プラズマによりエッチングすることで、樹脂層61の一部を除去した。これにより、疎水性の側壁12が形成された。最後に、有機溶媒でフォトレジスト62’を溶解させることにより、親水/疎水パターンを形成した。
 さらに詳しい手順は以下の通りである。図7中、(1)~(23)で示す数字は、下記の(1)~(23)に対応する。
 <CYTOP(登録商標)塗布工程(下記プロセスで膜厚約3.3~3.5μmのCYTOP(登録商標)層を作製)>
 (1)まず、ガラス(板状部材11)の洗浄及びCYTOP(登録商標)CTL-809の塗布を行なった。
 (2)次に、このガラスを10N KOHに一晩浸しておいた。
 (3)KOHに浸したカバーガラスを脱イオン水で10回以上洗った。
 (4)180℃のホットプレートでガラスを乾燥させた。
 (5)乾燥させたガラスを室温に戻した。
 (6)ガラス上にCYTOP(登録商標)CTL-809を約70μL垂らした。
 (7)下記のプログラムAでスピンコートした。

 (8)ホットプレート上で180℃,1時間ベークした。
 以上の(6)~(8)を4回繰り返すことで、深さ3.3~3.5μmの疎水性の樹脂層61が形成された。
 <フォトリソグラフィ工程>
 続いてフォトリソグラフィを行なった。
 (9)ポジ型のフォトレジスト62(AZ-4903)を、CYTOP(登録商標)塗布工程で作製した樹脂層61上に垂らした。量は、ガラス上で垂らしたレジストが直径約8mmに広がる程度とした。
 (10)下記のプログラムBでスピンコートした。

 (11)ガラスのエッジに残ったレジストを、100%EtOHを含ませたガーゼでふき取った。
 (12)55℃ 3分ベークした。
 (13)110℃ 5分ベークした。
 (14)フォトマスク63をアセトンで洗浄し、マスクアライナー(SAN-EI ELECTORIC社製)にセットした。
 (15)フォトレジスト62を塗布したガラスをマスクアライナーの試料台にセットし、試料台をリフトアップすることにより、ガラスとフォトマスク63とをコンタクトさせた。
 (16)UVを35秒間照射した(Power=256)。
 (17)ガラスをAZ Developer(AZ Electronic Materials USA社製)に5分以上浸し、現像した。
 (18)MilliQ(蒸留水)で10分程すすいだ。
 <エッチング・レジスト除去工程>
 続いて、エッチング及びレジストの除去を行なった。
 (19)RIE-10NR(Samco社製)を用いて、所定のプロセス条件(O2 50sccm;Pressure 10Pa;Power 50W;Time 30min)によりO2プラズマエッチングを行なった。
 (20)エッチング後のガラスをアセトンに浸け、15分間ソニケーションを行なった。
 (21)アセトンを交換し、もう一度15分間ソニケーションを行なった。
 (22)EtOHで15分間ソニケーションを行なった。
 (23)MilliQ(蒸留水)で洗浄した。
 以上の方法により、ガラス上に複数のウェル(収容部13)を形成させた。各ウェルの底面及び側面によって囲まれた領域は円柱形状であった。各ウェルの水平方向の断面は、直径5μmの円形状であり、各ウェルを隔てる側壁の高さは約3.3~3.5μmであった。また、隣接する2つのウェルの間の距離Bは5μmであった。
 (上面ガラスの作製)
 また、上面ガラスを以下の方法にて作製した。上面ガラスとして、直径1mmの貫通孔が形成されている厚さ5mmのガラスを用いた。このガラスの一方の面をCYTOP(登録商標)CTL-809(商品名、ASAHI GLASS社製)約70μLで覆い、下記プログラムCによりスピンコートした。

その後、ホットプレート上で180℃,1時間ベークした。
 以上の方法により、一方の面に厚さ1μmの疎水性層を備えた上面ガラスを作製した。
 (親水/疎水パターンガラスと上面ガラスとの貼り合わせ)
 次に、パラフィルム(Peckiney Plastic Packaging社製)の裏紙に高真空グリース(DOW CORNING TORAY社製)を付し、親水/疎水パターンガラスのうち、親水/疎水パターンが形成された面側であって親水/疎水パターンが形成されていない部分に張り付けた。上面ガラスを、コーティング剤でコートされた面が親水/疎水パターンガラス側になるように、親水/疎水パターンガラスの親水/疎水パターンが形成された面上に貼り付けた。
 これにより、親水/疎水パターンガラスと上面ガラスとの間に空間が形成された。この空間の垂直方向の幅、すなわち親水/疎水パターンガラスの側壁の上面と上面ガラスとの間の距離は、約150μmであった。
 (ドロップレットへのビーズの封入)
 次に、以下の方法により、ストレプトアビジンと反応後のビーズをドロップレットに封入した。
 まず、50mM Fluorescein-di-β-galactopyranoside(FDG)(Marker Gene Technology社製)/DMSOをFDGバッファー(100mM KPi buffer(PH=7.5),1mM MgCl2,4μL/mL 2-mercaptethanol)で希釈し、4mM FDG溶液を準備した。そして、ビーズ(6.5×106ビーズ/15μL バッファーC)15μLと、4mMFDG溶液15μLとを混ぜ、ビーズ溶液を作製した。
 次に、このビーズ溶液30μLを、上面ガラスの貫通孔からイエローチップで流路内にロードした(図1(a)~(b))。
 次に、各ウェル内の空気を抜くため、脱気を1分行なった(図1(c))。脱気は、0.1気圧の減圧デシケータ内に、アレイを約30秒放置することにより行った。その後5分間静置し、ビーズをウェルの底に沈めた。
 その後、Fluorinert(登録商標)FC40(SIGMA社製)200μL~1000μLを、上面ガラスの貫通孔から流路内にロードした(図1(d)~(e))。
 その結果、ウェル部分にのみ水相がトラップされてドロップレットが形成され、ビーズがドロップレット内に封入された。
 〔実施例1〕
 上述した方法を用いて、ビオチン化ビーズと1fMのストレプトアビジンとを反応させた後、FDGとともにアレイに導入し、ビーズを各ドロップレットに封入した。アレイとしては、2744個の収容部が形成された512μm×512μmのサブアレイを20個×20個にて配置した、合計1097600個の収容部を有する1.0cm×1.0cmのアレイを用いた。このアレイを蛍光顕微鏡(IX71(OLYMPUS社製))にて観察した。
 図3は、本発明の一実施例においてビーズを封入したアレイの蛍光像を表す図である。なお、図3は、1個のサブアレイについて示す。図3に示すように、蛍光像において、輝点がいくつか検出された(一視野中に138個)。この輝点は、ストレプトアビジンを捕捉したビオチン化ビーズが封入された収容部がある位置を示す。したがって、本発明に係る方法を用いれば、1fMのストレプトアビジンを十分検出可能であることがわかった。
 〔比較例1〕
 本発明の比較例として、従来のバルク(Bulk)計測法を用いてターゲット分子を検出した。
 実施例1で用いた濃度(1fM)よりも12.5倍濃い濃度である12.5fMストレプトアビジンとFDGとを混ぜ、蛍光分光器によって蛍光を測定した。また、コントロールとして、500倍の濃度である6.3pMのストレプトアビジンを用いて、同様に実験を行なった。
 この結果を図4に示す。図4は、従来の方法によってターゲット分子を検出した場合における蛍光強度を示すグラフである。なお、図4において、グラフaは12.5fMのストレプトアビジンを用いた場合の結果を示し、グラフbは6.3pMのストレプトアビジンを用いた場合の結果を示す。
 図4に示すように、従来の方法を用いた場合には、12.5fMのストレプトアビジンを検出することができなかった。したがって、従来の方法では、12.5fMは検出限界以下であることが示された。
 〔実施例2〕
 次に、ビオチン化ビーズと、5種類の異なる濃度(1fM、100aM、10aM、1aM、0.1aM)のストレプトアビジンとをそれぞれ反応させた後、FDGとともにアレイに導入し、ビーズを各ドロップレットに封入した。アレイとしては、実施例1と同様の構成のアレイを用いた。このアレイについて、顕微鏡下、明視野像と蛍光像とを観察した。
 (検出結果)
 図5(a)~(f)は、本発明の他の実施例においてビーズを封入したアレイの顕微鏡画像を表す図である。なお図5(a)、図5(c)及び図5(e)は1個のサブアレイの明視野像を現す図である。また、図5(b)、図5(d)及び図5(f)は、それぞれ図5(a)、図5(c)及び図5(e)が示すサブアレイの蛍光像を表す図である。また、図5(a)~(b)は1fMのストレプトアビジンについての結果であり、図5(c)~(d)は100aMのストレプトアビジンについての結果であり、図5(e)~(f)は10aMのストレプトアビジンについての結果である。
 1fMのストレプトアビジンを用いた場合、1つのサブアレイに封入されたビーズの全数は1735個であり、そのうちストレプトアビジンを捕捉したビーズは138個であった。100aMのストレプトアビジンを用いた場合、1つのサブアレイに封入されたビーズの全数は2008個であり、そのうちストレプトアビジンを捕捉したビーズは6個であった。10aMのストレプトアビジンを用いた場合、1つのサブアレイに封入されたビーズの全数は1360個であり、そのうちストレプトアビジンを捕捉したビーズは1個であった。
 (理論値と実験値との比較)
 また、ストレプトアビジンの各濃度において、ビーズの全数に対する、ストレプトアビジンを捕捉したビーズ(アクティブビーズ)の数の割合(%)についての理論値と実験値とを算出した。
 理論値は、反応させたビーズの全数に対するストレプトアビジンの分子数の割合(%)として算出した。また、実験値は、アレイに収容されたビーズの数に対するストレプトアビジンを捕捉したビーズの数の割合(%)として算出した。
 図6は、本発明の他の実施例において、ストレプトアビジンの濃度と、ストレプトアビジンを捕捉したビーズの数の割合との関係を示すグラフである。図6において、グラフaは理論値を示し、丸い点は実験値を示し、点線で囲まれた丸は、実験値の平均を示す(N=2~3)。グラフbは、実験値の平均値を直線によって近似したグラフを示す。
 図6に示すように、理論値と実験値とがほぼ重なったことから、本実施例の方法は、定量性が高く、ターゲット分子の濃度を正確に測定することが可能であることが示された。以上の結果より、本実施例の方法であれば、0.1aM程度の濃度であっても十分検出可能であることが示された。
 〔実施例3〕
 実施例1において作製したフローセル構造のアレイを用いて、上述した方法によりビーズ溶液(6.5×106ビーズ/30μL)をアレイに導入(ロード)し、ドロップレットに封入(トラップ)させた。このときのトラップ効率(ロードしたビーズの数に対するトラップされたビーズの数の割合(%))を算出した。
 この結果を図8に示す。図8は、フローセル構造を有するアレイを用いた場合(実施例3)と、フローセル構造を有しないアレイを用いた場合(比較例2)との、ビーズのトラップ効率を示す図である。
 〔比較例2〕
 本比較例では、フローセル構造を有しないアレイ(上述した親水/疎水パターンガラスのみからなるアレイ)を用いて、ビーズ(Φ=3μmのアミノビーズ;micromer-NH2-3μm;micromod社製)の封入を行なった。
 下記に示す方法により、フローセル構造を有するアレイを用いた場合(実施例3)と同濃度(2.2×108ビーズ/mL=6.5×106ビーズ/30μL)に希釈したビーズ溶液を用いて、親水/疎水パターンガラスにビーズを封入した。
 (1)バッファー(100mM KPi buffer(PH=7.5),1mM MgCl2,2μL/mL 2-mercaptethanol)でビーズを2.2×108ビーズ/mLに希釈した。
 (2)ペトリディッシュ(35mmペトリディッシュ;Becton Dickinson社製)の底面に、親水/疎水パターンガラス(上述した方法で作製したもの)を接着させた。接着剤としては、アラルダイト AR-R30(NICHIBAN社製)を用いた。
 (3)親水/疎水パターンガラス上を、ビーズ溶液500μLで覆った。
 (4)脱気した後、5分間インキュベートを行なった。
 (5)親水/疎水パターンガラス上にFC40(Fluorinert(登録商標)FC40,SIGMA社製)を2mLロードし、ビーズを封入した。
 (6)ドロップレットの蒸発を防ぐために、油相上を水で覆った。
 その後、ドロップレットに閉じ込められたビーズの数をカウントし、トラップ効率(ロードしたビーズの数に対するトラップされたビーズの数の割合(%))を算出した。
 この結果を図8に示す。図8に示されるように、フローセル構造を有するアレイを用いた場合には、フローセル構造を有しないアレイを用いた場合よりもトラップ効率が25倍以上高かった。フローセル構造を有しないアレイを用いた場合、高さ方向に散乱可能な距離が増すので、ビーズが基板に近づきにくくなったと考えられる。
 また、フローセル構造を有しないアレイを用いた場合、ビーズ溶液によってアレイの基板一面を覆う必要があり、大量のビーズ溶液(フローセル構造を有するアレイを用いる場合の約16倍)が必要になる。したがって、フローセル構造を有するアレイを用いれば、少量のビーズ溶液によって効率よくビーズを封入することが可能になる。
 本発明は、低濃度のターゲット分子を検出するための方法、アレイ、装置等に好適に利用することができる。

Claims (12)

  1.  ビーズを1個のみ収容可能な複数の収容部が、疎水性の上面を有する側壁によって互いに隔てられて形成されている下層部と、当該下層部における当該収容部が形成されている面に対向している上層部との間の空間に、当該ビーズを含む親水性溶媒を導入するビーズ導入工程と、
     上記ビーズ導入工程の後に上記空間に疎水性溶媒を導入する疎水性溶媒導入工程とを含むことを特徴とするビーズ封入方法。
  2.  上記ビーズ導入工程の後であって上記疎水性溶媒導入工程より前に、上記空間内を脱気する脱気工程をさらに含むことを特徴とする請求項1に記載のビーズ封入方法。
  3.  上記親水性溶媒は、水、親水性アルコール、親水性エーテル、ケトン、ニトリル系溶媒、ジメチルスルホキシド、及びN,N-ジメチルホルムアミドからなる群より選択される少なくとも1つ又はこれを含む混合物であることを特徴とする請求項1又は2に記載のビーズ封入方法。
  4.  上記疎水性溶媒は、飽和炭化水素、不飽和炭化水素、芳香族炭化水素、シリコーンオイル、パーフルオロカーボン、ハロゲン系溶媒、及び疎水性イオン液体からなる群より選択される少なくとも1つ又はこれを含む混合物であることを特徴とする請求項1~3のいずれかに記載のビーズ封入方法。
  5.  ターゲット分子を特異的に捕捉するビーズと、当該ターゲット分子とを反応させる反応工程と、
     上記反応工程の後に、上記ビーズを用いて請求項1~4のいずれかに記載のビーズ封入方法を行なうビーズ封入工程と、
     上記ビーズ封入工程の後に、上記収容部の各々に上記ターゲット分子を捕捉したビーズが収容されているか否かを検出する検出工程とを含むことを特徴とする、ターゲット分子を検出する方法。
  6.  上記ビーズには、上記ターゲット分子に特異的に結合する分子が結合していることを特徴とする請求項5に記載のターゲット分子を検出する方法。
  7.  複数の収容部が疎水性の上面を有する側壁によって互いに隔てられて形成されている下層部と、
     上記下層部における上記収容部が形成されている面に対して空間を隔てて対向している上層部とを備えていることを特徴とするアレイ。
  8.  上記収容部の各々は、底面が親水性であることを特徴とする請求項7に記載のアレイ。
  9.  上記上層部における上記下層部に対向する面が疎水性であることを特徴とする請求項7又は8に記載のアレイ。
  10.  上記上層部及び上記下層部の少なくとも一方に、上記空間に流体を導入するための貫通孔が形成されていることを特徴とする請求項7~9のいずれかに記載のアレイ。
  11.  請求項7~10のいずれかに記載のアレイと、
     ビーズとを備え、
     上記収容部の各々は、上記ビーズを1個のみ収容可能であることを特徴とするキット。
  12.  請求項7~10のいずれかに記載のアレイと、
     上記収容部の各々から、ターゲット分子を捕捉したビーズが収容された場合に発せられる光を検出するイメージセンサとを備えることを特徴とするターゲット分子検出装置。
