Definitions
- the present invention relates to a fusion protein for antagonizing a neonatal inducing factor and its use.
- the present invention relates to fusion proteins that inhibit a variety of angiogenic factors and uses thereof. More specifically, the present invention relates to fusion proteins of VEGF receptors and FGF receptors and their use in the treatment of diseases associated with angiogenesis.
- Angiogenesis is one of the basic factors leading to the growth and metastasis of malignant tumors [1].
- the angiogenesis process is regulated by a variety of factors, some of which promote angiogenesis, and some factors inhibit angiogenesis. Therefore, regulation of angiogenesis is a very complex dynamic equilibrium process [2].
- Antiangiogenic therapy aims to control tumor growth by blocking angiogenesis stimulating factors or by angiogenesis in the angiogenesis inhibitor pHoh tumor.
- VEGF vascular endothelial growth factor
- FGF Fibroblast growth factor
- HGF Hepatocyte growth factor
- VEGF is the most specific and effective growth factor known to angiogenesis.
- VEGF vascular endothelial growth factor
- Many animals have tested that VEGF can prevent angiogenesis and inhibit tumor growth. Because of this, VEGF and its receptors are the most important targets for anti-angiogenic drugs.
- the anti-angiogenic drugs that have achieved remarkable results in clinical trials are Bevacizumab (trade name Avastin), which can directly block VEGF, which inhibits tumor angiogenesis, was approved by the US FDA in 2004 as a first-line drug for colorectal cancer. It is the first new drug approved to be marketed to inhibit angiogenesis.
- Avastin is a humanized anti-VEGF monoclonal antibody produced by Genentech, a well-known biotechnology company in the United States.
- Avastin combined with chemotherapy can significantly prolong the survival of a variety of cancer patients, including rectal cancer, lung Cancer, breast cancer, kidney cancer, etc. [5, 6].
- the clinical success of Avastin is epoch-making. It demonstrates that anti-angiogenesis therapy targeting tumor vasculature is a clinically effective treatment and opens up a whole new approach to the treatment of multiple tumors. In Western countries, Avastin has been widely used in cancer treatment and is currently the world's largest drug in sales.
- Anti-VEGF drugs have made significant advances in the clinical treatment of tumors, but clinical trials have also shown considerable limitations in anti-VEGF therapy. From the therapeutic effect of tumors, Avastin can prolong the survival time of patients with rectal cancer by about 3-4 months [9, 10], and the survival time for breast cancer patients can be extended by about 7-8 months [11], Therefore, Avastin does not effectively inhibit tumor blood vessel growth for a long time. Therefore, how to further improve the clinical therapeutic effect of anti-angiogenesis is a problem that tumor researchers need to solve, and it is also the focus of research and development of the next generation of anti-angiogenic drugs.
- the underlying cause of failure or resistance to anti-VEGF therapy may be that tumor angiogenesis is controlled by a variety of factors, and although VEGF plays an important role in angiogenesis, it is not the only angiogenic stimulator. At the same time, due to the heterogeneity of tumor cells and the complexity of the tumor microenvironment and the compensatory mechanism of the body, when the activity of VEGF is inhibited for a long time, other angiogenic stimulating factors will be expressed [12], thus The growth of tumor blood vessels is no longer dependent on the VEGF signaling pathway.
- Fibroblast growth factor is a family of growth factors that bind heparin, which has 22 family members (FGF 1-14, 16-23) in mammals. FGF plays an important role in a variety of biological functions, such as cell proliferation, differentiation, migration, angiogenesis, and tumorigenesis. It exerts its diverse biological functions by binding to and activating the cell surface FGF receptor (FGFR).
- FGF fibroblast growth factor receptor
- the fibroblast growth factor receptor (FGFR) is a receptor that binds to members of the fibroblast growth factor family. A portion of the fibroblast growth factor receptor is involved in the disease process.
- the fibroblast growth factor receptor consists of an extracellular domain, a transmembrane domain, and an intracellular domain.
- the extracellular regions comprise three immunoglobulin-like (Ig-like) domains: a first Ig-like domain, a second Ig-like domain, and a third Ig-like domain, in the first and second Ig-like domains
- the sequence between the Ig-like domain and the second Ig-like domain is referred to as the FGFR Ig-like domain intermediate functional sequence region.
- the intermediate functional sequence region comprises an acidic amino acid segment called acid box (AB)
- the VEGF and FGF dual antagonist fusion proteins will use the fully human protein sequence, combined with the excellent pharmacokinetic properties of the Fc fusion protein, and will have good drug-forming conditions and potential for clinical application.
- the affinity of the fusion protein for each target depends on the correct protein configuration.
- the receptor sheet often loses its ability to bind to the ligand, so it is efficient and specific.
- FGF-2 fusion protein drugs of VEGF and FGF
- fusion proteins that inhibit angiogenesis have been reported, such as FGFR-based fusion proteins (WO/2008/065543), Notch3-based fusion proteins (WO/2010/021729), VEGFR-based fusion proteins (WO/ 2010/105573), LK8-based fusion protein (WO/2008/075833), etc., but these fusion proteins are fused to a single target to fuse a portion from a single angiogenesis inhibitor to an immunoglobulin Fc fragment.
