本発明のシステイン誘導体は一般式(I)で表されるシステイン誘導体またはその塩である。
X及びYは、それぞれ独立して、OR1、NHR2(式中、R1及びR2は、それぞれ独立して、水素原子またはC1-22アルキル基を示す。)、であり;
Zは、水素原子またはC1-22アルキル基を示し;
Wは、C1-22アルキル基、C1-22アルコキシ基またはC1-22アルキルアミノ基を示す。]
「C1-22アルキル基」とは、炭素数1~22個の直鎖または分枝鎖状の炭化水素基を意味し、例えば、メチル基、エチル基、イソプロピル基、プロピル基、ブチル基、イソブチル基、sec-ブチル基、tert-ブチル基、ペンチル基、sec-ペンチル基、tert-ペンチル基、イソペンチル基、ヘキシル基、ヘプチル基、オクチル基、2-エチルヘキシル基、tert-オクチル基、ノニル基、イソノニル基、デシル基、イソデシル基、ウンデシル基、ドデシル基、トリデシル基、イソトリデシル基、テトラデシル基、ペンタデシル基、ヘキサデシル基、イソヘキサデシル基、ヘプタデシル基、オクタデシル基、イソオクタデシル基、ベヘニル基等が挙げられる。
「C1-16アルキル基」としては、メチル基、エチル基、イソプロピル基、プロピル基、ブチル基、イソブチル基、sec-ブチル基、tert-ブチル基、ペンチル基、sec-ペンチル基、tert-ペンチル基、イソペンチル基、ヘキシル基、ヘプチル基、オクチル基、2-エチルヘキシル基、tert-オクチル基、ノニル基、イソノニル基、デシル基、イソデシル基、ウンデシル基、ドデシル基、トリデシル基、イソトリデシル基、テトラデシル基、ペンタデシル基、ヘキサデシル基、イソヘキサデシル基等が挙げられる。
「C1-6アルキル基」としては、メチル基、エチル基、イソプロピル基、プロピル基、ブチル基、イソブチル基、sec-ブチル基、tert-ブチル基、ペンチル基、sec-ペンチル基、tert-ペンチル基、イソペンチル基、ヘキシル基等が挙げられる。
「C1-6アルコキシ基」としては、メトキシ基、エトキシ基、プロポキシ基、イソプロポキシ基、ブトキシ基、イソブトキシ基、tert-ブトキシ基、ペンチルオキシ基、ヘキシルオキシ基等が挙げられる。
「C1-6アルキルアミノ基」としては、メチルアミノ基、エチルアミノ基、プロピルアミノ基、イソプロピルアミノ基、ブチルアミノ基、イソブチルアミノ基、tert-ブチルアミノ基、ペンチルアミノ基、ヘキシルアミノ基が挙げられる。
Zで示される「C1-22アルキル基」としては、好ましくはC1-6アルキル基であり、より好ましくはメチル基である。Zは、好ましくは水素原子またはC1-6アルキル基であり、より好ましくは水素原子またはメチル基である。
Wで示される「C1-22アルキル基」としては、好ましくはC1-16アルキル基であり、より好ましくは、メチル基、ノニル基、ペンタデシル基であり、さらに好ましくはメチル基である。
Wで示される「C1-22アルコキシ基」としては、好ましくはC1-6アルコキシ基であり、より好ましくはtert-ブトキシ基である。
Wで示される「C1-22アルキルアミノ基」としては、好ましくはC1-6アルキルアミノ基である。
本発明のシステイン誘導体としては、(2R,4S)体、(2S,4R)体、またはその混合物が好ましく、より好ましくは、(2S,4R)体である。
無機塩基との塩として、例えば、ナトリウム塩、カリウム塩、カルシウム塩、マグネシウム塩、亜鉛塩、銀塩、アンモニウム塩等が挙げられる。
有機塩基との塩として、例えば、メチルアミン、ジエチルアミン、トリメチルアミン、トリエチルアミン、エタノールアミン、ジエタノールアミン、トリエタノールアミン、エチレンジアミン、トリス(ヒドロキシメチル)メチルアミン、ジシクロヘキシルアミン、N,N’-ジベンジルエチレンジアミン、グアニジン、ピリジン、ピコリン、コリン、シンコニン、メグルミン、アミノ酸(アルギニン、リジン等)等との塩が挙げられる。
自体公知の方法に従って、本発明のシステイン誘導体と、無機塩基または有機塩基とを反応させることにより、各々の塩を得ることができる。
システイン、または、あらかじめシステインをエステル化もしくはアミド化して得た一般式(II)で表される化合物(以下、化合物(II)と略記し、他の式で表される化合物についても同様である。)を、化合物(III)と反応させて環構築し、化合物(IV)を得る方法
化合物(IV)は、水またはメタノール、エタノールなどのアルコール中で化合物(II)と化合物(III)とを5~24時間反応させることによって得られる。化合物(II)の内、システインエチルエステルは例えば、システインをエチルアルコール中で、塩酸または塩化チオニル存在下、室温で5~24時間程度反応させることにより得ることができる。化合物(II)の内、システインアミドは保護システインとアミンをEDCI・HCl(1-エチル-3-(3-ジメチルアミノプロピル)-カルボジイミド塩酸塩)のような脱水縮合剤の存在下、塩化メチレン、N,N-ジメチルホルムアミド(DMF)のような溶媒中で、室温で5~24時間反応して得られた化合物を、脱保護することによって得られる。
化合物(IV)を溶媒の存在下または非存在下、塩基の存在下または非存在下に化合物(V)または化合物(V’)と反応させて、システイン誘導体(I)を合成することができる。溶媒としては、THF(テトラヒドロフラン)、酢酸エチル、酢酸イソプロピル、アセトニトリル、アセトン、エタノール、メタノール、ジクロロメタン、水、あるいはそれらの混合物等が挙げられ、好ましくはTHF、酢酸エチル、酢酸イソプロピル、アセトニトリル、アセトン、ジクロロメタン、水、あるいはそれらの混合物である。塩基としては、トリエチルアミン、ジイソプロピルエチルアミン、ピリジン、N-メチルモルホリン等の有機塩基、水酸化リチウム、水酸化ナトリウム、水酸化カリウム、炭酸ナトリウム、炭酸カリウム、酢酸ナトリウム、酢酸カリウム等の無機塩基が挙げられ、好ましくはトリエチルアミン、ジイソプロピルエチルアミン、ピリジン、N-メチルモルホリン、炭酸カリウム、炭酸ナトリウムである。
化合物(V)または化合物(V’)の使用量は、化合物(IV)1モルに対し、1.0~5.0モルであり、好ましくは1.2~3.0モルである。塩基を使用する場合には、塩基の使用量は、化合物(IV)1モルに対し、1.0~5.0モルであり、好ましくは1.2~4.0モルである。反応温度は、-10~100℃であり、好ましくは0~90℃である。反応時間は、1時間~48時間であり、好ましくは3時間~20時間である。
更にこれをエステル化、アミド化、加水分解または酸無水物化などによって他のシステイン誘導体(I)に変換しても良い。
本発明の(B)成分である美白剤としては、例えば、ハイドロキノンおよびその誘導体、アスコルビン酸およびその誘導体、コウジ酸およびその誘導体、ニコチン酸誘導体、セロトニン誘導体、サリチル酸誘導体、アスタキサンチン、α-リポ酸、パンテテインスルホン酸カルシウム、美白効果を有するエキス、美白効果を有する植物抽出物、美白効果を有するポリフェノール、美白効果を有する不飽和脂肪酸、美白効果を有するペプチド、美白効果を有するアミノ酸、美白効果を有する核酸誘導体、および美白効果を有するアルキルフェノールから選択される1種以上が挙げられる。
(B)成分の美白剤は単独で使用しても、2種以上を混合して使用してもよい。
高級アルコールとしては、例えば、セチルアルコール、ラウリルアルコール、イソステアリルアルコール、ステアリルアルコール、オレイルアルコール、ミリスチルアルコール、オクチルドデカノール、ベヘニルアルコール、ラノリンアルコール、セトステアリルアルコール、デシルテトラデカノール、ヘキシルデカノール、コレステロール、フィトステロールなどが挙げられる。

L-システイン塩酸塩一水和物(50g、285mmol)を水(100ml)に溶解後、6N水酸化ナトリウム水溶液を加え、pHを5.15に調整した。反応混合物を40℃に加熱し、ピルビン酸エチルエステル(38ml、342mmol)を徐々に添加し、40℃にて4時間撹拌した。水を加え、室温まで冷却し結晶を析出させろ過した。得られた結晶を水で洗浄し、減圧下50℃にて乾燥し2-メチルチアゾリジン-2,4-ジカルボン酸-2-エチルエステル(トランス体:シス体=88:12、12.1g、収率19%)を得た。合成した2-メチルチアゾリジン-2,4-ジカルボン酸-2-エチルエステル(5g、22.8mmol)を酢酸エチル(40ml)に溶解後、アルゴン下にてトリエチルアミン(6.4ml、45.9mmol)を加え、塩化アセチル(2.4ml、33.8mmol)をゆっくりと滴下後、室温で終夜撹拌した。反応終了後、水(15ml)を加え、さらにHClでpHを1.0に調整した。水層を分離後、有機層を水(10ml)で洗浄し、飽和食塩水で洗浄後、無水硫酸マグネシウムで乾燥した。得られた酢酸エチル溶液を濃縮し、結晶を析出させ、ろ過後、ヘプタン/酢酸エチル=1/1(10ml)で洗浄し、減圧下50℃にて乾燥し、(2S,4R)-N-アセチル-2-メチルチアゾリジン-2,4-ジカルボン酸-2-エチルエステルの結晶を得た(3.3g、収率55%)。

B16メラノーマをDMEM(Dulbecco’s Modified Eagle Medium)(高グルコース、血清10%含有)にて培養した。コンフルエント後、6ウェルプレートに播種した。翌日、各評価サンプルを所定評価濃度(表1)にて添加したDMEM(チロシンを300μM添加)にて各ウェル培地を交換し、3日間培養した。培地にて溶解しないものはジメチルスルホキシドにて溶解し、培地に添加した(培地中のジメチルスルホキシドは0.1%)。培地を回収し、10000rpmで10分間遠心分離した上清の450nmにおける吸光度を分光光度計にて測定した。培地を回収した後、細胞をPBS(phosphate-buffered saline)で1回リンスした。ウェルに4%水酸化ナトリウム水溶液とジメチルスルホキシド(9:1)の混和溶液を加え、10分間室温で静置した。細胞を溶解及び回収後、3500rpmで10分間遠心分離し、上清の吸光度(450nm)を分光光度計にて測定した。培地中及び細胞内のメラニン量の合計を、合成メラニンを用いて作成した検量線により算出した。コントロール(サンプル添加なし)の算出値を100%と規定した場合の、各サンプル所定濃度添加3日後の算出値を相対%にすることにより、コントロール中のメラニン量を100%とした場合の、各サンプルのメラニン産生率を算出した。ジメチルスルホキシドにて溶解し添加したサンプルは、0.1%ジメチルスルホキシド含有したものをコントロール(サンプル添加なし)とした。また、メラニン産生率は試験例2で得られた細胞蛋白量より規格化した。
尚、本メラニン産生抑制試験にて使用した分光光度計は、DU800、BECKMAN COULTER社製である。
B16メラノーマをDMEM(Dulbecco’s Modified Eagle Medium)(高グルコース、血清10%含有)にて培養した。コンフルエント後、6ウェルプレートに播種した。翌日、各評価サンプルを所定評価濃度(表1)にて添加したDMEM(チロシンを300μM添加)にて各ウェル培地を交換し、3日間培養した。培地を回収した後、細胞をPBS(phosphate-buffered saline)で1回リンスした。ウェルに4%水酸化ナトリウム水溶液とジメチルスルホキシド(9:1)の混和溶液を加え、10分間室温で静置した。細胞を溶解及び回収後、3500rpmで10分間遠心分離し、上清をミリQ水にて3倍に希釈した。希釈溶液(20μl)にA溶液(DC Protein Assay;BIO RAD社)100μlを添加しボルテックスにて混合後、B溶液(DC Protein Assay;BIO RAD社)800μlを添加及び混合後、室温にて15分間静置した。静置後、吸光度(750nm)を分光光度計にて測定した。また、アルブミンにて検量線を求め、上記溶液の蛋白量を算出し、相対比率を生存率とした。
尚、本メラニン産生抑制試験にて使用した分光光度計は、DU800、BECKMAN COULTER社製である。

<メラニン産生率:美白併用効果、しみ改善>
各サンプルについて、単独で使用した場合および併用して使用した場合のメラニン産生率を測定し、その併用効果を、理論値を使用して以下の基準で評価した。
なお、理論値は、単独で(A)成分を使用した場合のメラニン産生率(%)×単独で成分(B)を使用した場合のメラニン産生率(%)/100、とした。
○○:理論値-実測値が10%以上30%未満。
○:理論値-実測値が-10%以上10%未満。
△:理論値-実測値が-10%未満であるが、併用した場合、それぞれ単独で使用した場合より優れている。
×:理論値-実測値が-10%未満であり、かつ併用しても単独で使用した場合より劣っている。
各サンプルについて、単独で使用した場合および併用して使用した場合の細胞生存率を測定し、その併用効果を、毒性緩和度を使用して以下の基準で評価した。
なお、毒性緩和度は、(A)成分と(B)成分を併用した場合の細胞生存率(%)-単独で(B)成分を使用した場合の細胞生存率(%)、とした。
○○:毒性緩和度が、6以上9未満。
○:毒性緩和度が、3以上6未満。
△:毒性緩和度が、0以上3未満。
×:毒性緩和度が、0未満。
また、(A)N-Ac-CPと、(B)成分としてアルブチン、グルタチオン、カミツレエキスまたはアスコルビン酸セチルエーテルを組み合わせると、メラニン産生抑制効果が向上することが確認された。なかでも、(B)成分としてグルタチオンまたはアスコルビン酸セチルエーテルを組み合わせると、美白効果が顕著に向上し、そのなかでも、(B)成分としてグルタチオンを組み合わせると、美白効果が極めて顕著に向上することがわかった。
また、(B)成分としてアルブチン、グルタチオンまたはカミツレエキスに(A)N-Ac-CPを組み合わせると、安全性が向上することがわかった。なかでも、グルタチオンまたはカミツレエキスについては安全性が顕著に向上することがわかった。
合成例1及び合成例3のシステイン誘導体について、美白効果を比較すべくメラニン産生率測定を行った。B16メラノーマをDMEM(Dulbecco’s Modified Eagle Medium)(高グルコース、血清10%含有)にて培養した。コンフルエント後、96ウェルプレートに播種した。翌日、各評価サンプルを5~25mMに調製し添加したDMEMにて各ウェル培地を交換し、3日間培養した。450nmにおける吸光度をマイクロプレートリーダーにて測定した。コントロール(サンプル添加なし)の測定値を100%と規定した場合の、各サンプル所定濃度添加3日後の測定値を相対%にすることにより、コントロール中のメラニン量を100%とした場合の、各サンプルのメラニン産生率を算出した。
合成例1及び合成例3のシステイン誘導体のそれぞれについて、メラニン産生率を50%に抑制したときの算出濃度を表2に示す。
尚、本メラニン産生抑制試験にて使用したマイクロプレートリーダーは、MULTISKAN FC、 サーモフィッシャーサイエンティフィック社製である。
Claims (17)
- (A)システイン誘導体またはその塩が、N-アセチル-2-メチルチアゾリジン-2,4-ジカルボン酸、N-アセチル-2-メチルチアゾリジン-2,4-ジカルボン酸-2-エチルエステル、およびそれらの塩から選択される1種以上である、請求項1記載の化粧料。
- (B)美白剤が、ハイドロキノンおよびその誘導体、アスコルビン酸およびその誘導体、コウジ酸およびその誘導体、ニコチン酸誘導体、セロトニン誘導体、サリチル酸誘導体、アスタキサンチン、α-リポ酸、パンテテインスルホン酸カルシウム、美白効果を有するエキス、美白効果を有する植物抽出物、美白効果を有するポリフェノール、美白効果を有する不飽和脂肪酸、美白効果を有するペプチド、美白効果を有するアミノ酸、美白効果を有する核酸誘導体、および美白効果を有するアルキルフェノールから選択される1種以上である請求項1または2に記載の化粧料。
- (B)美白剤が、アルブチン、アスコルビン酸セチルエーテル、コウジ酸、4-メトキシサリチル酸、カミツレエキス、エラグ酸、リノール酸、グルタチオン、トラネキサム酸、、およびルシノールから選択される1種以上である請求項1または2に記載の化粧料。
- (A)システイン誘導体またはその塩がN-アセチル-2-メチルチアゾリジン-2,4-ジカルボン酸-2-エチルエステルまたはその塩であり、(B)美白剤がコウジ酸である請求項1記載の化粧料。
- (A)システイン誘導体またはその塩がN-アセチル-2-メチルチアゾリジン-2,4-ジカルボン酸-2-エチルエステルまたはその塩であり、(B)美白剤がトラネキサム酸である請求項1記載の化粧料。
- (A)システイン誘導体またはその塩がN-アセチル-2-メチルチアゾリジン-2,4-ジカルボン酸-2-エチルエステルまたはその塩であり、(B)美白剤がカミツレエキスである請求項1記載の化粧料。
- (A)システイン誘導体またはその塩がN-アセチル-2-メチルチアゾリジン-2,4-ジカルボン酸-2-エチルエステルまたはその塩であり、(B)美白剤がアルブチンである請求項1記載の化粧料。
- (A)システイン誘導体またはその塩と(B)美白剤との配合比が、(A)/(B)=1/20~5000/1(g/g)である請求項1~8のいずれかに記載の化粧料。
- (A)一般式(I)で表されるシステイン誘導体が、(2R,4S)体、(2S,4R)体、およびそれらの混合物から選択される1種以上である、請求項1~9のいずれかに記載の化粧料。
- (A)一般式(I)で表されるシステイン誘導体が、(2S,4R)体である、請求項1~9のいずれかに記載の化粧料。
- (A)システイン誘導体またはその塩が、N-アセチル-2-メチルチアゾリジン-2,4-ジカルボン酸、N-アセチル-2-メチルチアゾリジン-2,4-ジカルボン酸-2-エチルエステル、およびそれらの塩から選択される1種以上である、請求項12記載のメラニン産生抑制剤。
- (B)美白剤が、ハイドロキノンおよびその誘導体、アスコルビン酸およびその誘導体、コウジ酸およびその誘導体、ニコチン酸誘導体、セロトニン誘導体、サリチル酸誘導体、アスタキサンチン、α-リポ酸、パンテテインスルホン酸カルシウム、美白効果を有するエキス、美白効果を有する植物抽出物、美白効果を有するポリフェノール、美白効果を有する不飽和脂肪酸、美白効果を有するペプチド、美白効果を有するアミノ酸、美白効果を有する核酸誘導体、および美白効果を有するアルキルフェノールから選択される1種以上である請求項12または13に記載のメラニン産生抑制剤。
- (B)美白剤が、アルブチン、アスコルビン酸セチルエーテル、コウジ酸、4-メトキシサリチル酸、カミツレエキス、エラグ酸、リノール酸、グルタチオン、トラネキサム酸、およびルシノールから選択される1種以上である請求項12~14のいずれかに記載のメラニン産生抑制剤。
- (A)一般式(I)で表されるシステイン誘導体が、(2R,4S)体、(2S,4R)体、およびそれらの混合物から選択される1種以上である請求項12~15のいずれかに記載のメラニン産生抑制剤。
- (A)一般式(I)で表されるシステイン誘導体が、(2S,4R)体である、請求項12~15のいずれかに記載のメラニン産生抑制剤。
