[課題]六炭糖と五炭糖の混合糖を発酵原料とした、高収率の化学品の製造方法を提供すること。 [解決手段]微生物の培養液を分離膜で濾過すること、未濾過液を培養液に保持または還流すること、発酵原料を培養液に追加すること、およびろ過液中の生産物を回収することを含む、連続発酵による化学品の製造方法であって、前記発酵原料が五炭糖および六炭糖を含み、前記微生物がペントースイソメラーゼを用いて五炭糖を代謝する経路を有する、化学品の製造方法を提供した。

Description

化学品の製造方法
 本発明は、六炭糖および五炭糖を含む発酵原料を使用した連続発酵による化学品の製造方法に関する。
 乳酸などの生分解性ポリマー原料やエタノールなどのバイオ燃料に代表されるバイオマス由来の化学品は、大気中への二酸化炭素の排出問題やエネルギー問題の顕在化と共にサスティナビリティー(持続可能性)およびライフサイクルアセスメント(LCA)対応型製品として強い注目を浴びている。これら生分解性ポリマー原料やバイオ燃料の製造方法としては、とうもろこしなどの可食性バイオマスから精製した六炭糖であるグルコースを微生物の発酵原料に使用し、発酵産物として得るのが一般的であるが、可食性バイオマスを使用すると、食料と競合することから価格高騰が引き起こされ、安定し他原料の調達ができなくなる恐れがある。そこで、稲わらなどの非可食性バイオマス由来の糖を微生物の発酵原料に使用する試みが行われている(特許文献1参照。)。
 非可食性バイオマス由来の糖を発酵原料に使用する際には、非可食バイオマスに含まれるセルロース、ヘミセルロースなどを糖化酵素によって糖に分解するが、この際にグルコースなどの六炭糖だけではなく、キシロースなどの五炭糖も同時に得られ、非可食性バイオマス由来の糖を微生物の発酵原料に使用する場合、六炭糖と五炭糖の混合糖を発酵原料として使用することになる(特許文献1参照。)。
 六炭糖と五炭糖の混合糖を使用する場合は、六炭糖の代謝経路に加えて五炭糖の代謝経路を有する微生物を使用する必要がある。五炭糖を代謝する経路はいくつか知られているが、五炭糖を代謝する経路の初段で五炭糖にペントースレダクターゼが作用してペントールが生成する経路では、ペントールが後段の代謝経路に流れにくいため培養液中に蓄積してしまう(非特許文献1参照。)。一方、五炭糖を代謝する経路の初段で五炭糖にペントースイソメラーゼが作用する経路ではペントールは生成しないため、ペントースレダクターゼが作用する経路に比べて発酵収率が高いことが知られている(非特許文献1参照。)。
 六炭糖と五炭糖の混合糖である非可食性バイオマス由来の糖を微生物の発酵原料とした発酵方法としては連続発酵が採用されうるが、実際に連続発酵した場合の発酵収率については確認されていない(特許文献2参照。)。一方で、ペントースイソメラーゼを有する微生物を、六炭糖と五炭糖の混合糖を発酵原料として連続発酵する場合、バッチ発酵よりも発酵収率が低下してしまうことも知られている(非特許文献2参照。)。したがって、六炭糖と五炭糖の混合糖をペントースイソメラーゼを有する微生物の発酵原料とした連続発酵によって発酵収率を向上させようとする場合、副産物の生合成経路を遮断するなど遺伝子改変による微生物の改良を行う必要があると考えるのがこれまでの技術常識であった。

 生分解性ポリマー原料やバイオ燃料の発酵生産において、六炭糖と五炭糖の混合糖をペントースイソメラーゼを有する微生物の発酵原料として連続発酵をした場合には、発酵収率が低下してしまうことが知られている。そこで本発明は、六炭糖と五炭糖の混合糖をペントースイソメラーゼを用いて五炭糖を代謝する経路を有する微生物の発酵原料として連続発酵する場合の発酵収率の向上を課題とするものである。
 本発明者は、上記課題を鋭意検討した結果、六炭糖と五炭糖の混合糖をペントースイソメラーゼを用いて五炭糖を代謝する経路を有する微生物の発酵原料として連続発酵する化学品の製造方法において、分離膜を用いた連続発酵を行なうことで上記課題を解決することを見出し、本発明に至った。
 すなわち、本発明は以下の(1)~(6)の通りである。
(1)微生物の培養液を分離膜で濾過すること、未濾過液を培養液に保持または還流すること、発酵原料を培養液に追加すること、およびろ過液中の生産物を回収することを含む、連続発酵による化学品の製造方法であって、前記発酵原料が五炭糖および六炭糖を含み、前記微生物がペントースイソメラーゼを用いて五炭糖を代謝する経路を有する、化学品の製造方法。
(2)酸素移動容量係数(Kla)が150h-1以下の条件で連続発酵を行う、(1)に記載の化学品の製造方法。
(3)前記発酵原料中に含まれる六炭糖と五炭糖の重量比率が1:9~9:1である、(1)または(2)に記載の化学品の製造方法。
(4)前記発酵原料がバイオマス由来の糖液を含む、(1)または(2)に記載の化学品の製造方法。
(5)ペントースイソメラーゼがキシロースイソメラーゼである、(1)~(4)のいずれかに記載の化学品の製造方法。
(6)五炭糖がキシロースである、(1)~(5)のいずれかに記載の化学品の製造方法。
 本発明によれば、六炭糖と五炭糖の混合糖をペントースイソメラーゼを用いて五炭糖を代謝する経路を有する微生物の発酵原料とするにもかかわらず、化学品を高収率で得ることができる。
 本発明は、微生物を発酵原料で培養して化学品を発酵生産する化学品の製造方法において、培養液を分離膜で濾過し未濾過液を培養液に保持または還流し、かつ、発酵原料を培養液に追加して濾過液中の生産物を回収する連続発酵する化学品の製造方法であって、ペントースイソメラーゼを用いて五炭糖を代謝する経路を有する微生物を用い、かつ、発酵原料が六炭糖および五炭糖を含むことを特徴とする。
 五炭糖とは、糖を構成する炭素の数が5つのものであり、ペントース(Pentose)とも呼ばれる。1位にアルデヒド基をもつアルドペントースと2位にケトン基をもつケトペントースがある。アルドペントースとしては、キシロース、アラビノース、リボース、リキソースが挙げられ、ケトペントースとしては、リブロース、キシルロースが挙げられる。本発明で用いる五炭糖としては、微生物が資化できるものであればいずれでも良いが、自然界での存在割合や入手のしやすさなどの観点から、好ましくはキシロース、アラビノースであり、より好ましくはキシロースである。
 六炭糖とは、糖を構成する炭素の数が6つのものであり、ヘキソース(Hexose)とも呼ばれる。1位にアルデヒド基をもつアルドースと2位にケトン基をもつケトースがある。アルドースとしては、グルコース、マンノース、ガラクトース、アロース、グロース、タロースなどが挙げられ、ケトースとしては、フルクトース、プシコース、ソルボースなどが挙げられる。本発明で用いる六炭糖としては、微生物が資化できるものであればいずれでも良いが、自然界での存在割合や入手のしやすさなどの観点から、好ましくはグルコース、マンノース、ガラクトースであり、より好ましくはグルコースである。
 本発明で使用される混合糖に関しては特に限定されないが、六炭糖と五炭糖を両方含むことが知られているセルロース含有バイオマス由来糖液が好ましく使用される。セルロース含有バイオマスには、バガス、スイッチグラス、コーンストーバー、稲わら、麦わらなど草木系バイオマスと、樹木、廃建材などの木質系バイオマスなどを例として挙げることができる。セルロース含有バイオマスは、糖が脱水縮合した多糖であるセルロースあるいはヘミセルロースを含有しており、こうした多糖を加水分解することで発酵原料として利用可能な糖液が製造される。セルロース含有バイオマス由来糖液の調製方法は、どのような方法であってもよく、こうした糖の製造方法としては、濃硫酸を使用してバイオマスを酸加水分解して糖液を製造する方法(特表平11-506934号公報、特開2005-229821号公報)、バイオマスを希硫酸で加水分解処理した後に、さらにセルラーゼなどの酵素処理することより糖液を製造する方法が開示されている(A.Adenら、“LignocellulosicBiomass to Ethanol Process Design and Economics Utilizing Co-Current Dilute Acid Prehydrolysis and Enzymatic Hydrolysis for Corn Stover”NREL Technical Report(2002))。また酸を使用しない方法として、250~500℃程度の亜臨界水を使用しバイオマスを加水分解して糖液を製造する方法(特開2003-212888号公報)、またバイオマスを亜臨界水処理した後に、さらに酵素処理することにより糖液を製造する方法(特開2001-95597号公報)、バイオマスを240~280℃の加圧熱水で加水分解処理した後に、さらに酵素処理することにより糖液を製造する方法(特許3041380号公報)が開示されている。以上のような処理の後、得られた糖液を精製してもよい。その方法は、例えばWO2010/067785に開示されている。
 混合糖に含まれる五炭糖と六炭糖の重量比率は特に制限はないが、混合糖中の五炭糖と六炭糖の重量比率として(五炭糖):(六炭糖)と表すと、1:9~9:1が好ましい。これは、混合糖としてセルロース含有バイオマス由来糖液を想定した場合の糖比率である。
 本発明に使用する発酵原料に含まれる総糖濃度は特に制限されず、微生物の化学品の生産を阻害しない範囲であれば可能な限り高い濃度であることが好ましい。具体的には、培地中の炭素源の濃度は、15~500g/lが好ましく、20~300g/lがより好ましい。総糖濃度が15g/l以下であると、五炭糖からの収率向上の効果が低下してしまうことがある。また、総糖濃度が低いと化学品の生産効率も低下してしまう。
 本発明に使用する発酵原料に含まれる六炭糖濃度は、上記の総糖濃度、五炭糖と六炭糖の割合の範囲において特に限定されないが、本発明の化学品の製造方法を用いれば、六炭糖を濃度5g/L以上含む混合糖液においても良好な収率を得ることができる。
 本発明で使用する発酵原料は、炭素源、窒素源、無機塩類、および必要に応じてアミノ酸、およびビタミンなどの有機微量栄養素を適宜含有する通常の液体培地が良い。
 本発明で使用する窒素源としては、アンモニアガス、アンモニア水、アンモニウム塩類、尿素、硝酸塩類、その他補助的に使用される有機窒素源、例えば、油粕類、大豆加水分解液、カゼイン分解物、その他のアミノ酸、ビタミン類、コーンスティープリカー、酵母または酵母エキス、肉エキス、ペプトン等のペプチド類、各種発酵菌体およびその加水分解物などが使用される。無機塩類としては、リン酸塩、マグネシウム塩、カルシウム塩、鉄塩およびマンガン塩等を適宜添加使用することができる。
 本発明で使用される微生物が生育のために特定の栄養素を必要とする場合には、その栄養物を標品もしくはそれを含有する天然物として添加する。また、消泡剤も必要に応じて使用する。本発明において、培養液とは、発酵原料に微生物が増殖した結果得られる液のことを言う。追加する発酵原料の組成は、目的とする化学品の生産性が高くなるように、培養開始時の発酵原料組成から適宜変更しても良い。
 次に、本発明に化学品の製造方法に用いることができる微生物について説明する。
 本発明は、ペントースイソメラーゼを用いて五炭糖を代謝する経路を有する微生物を用いることを特徴とする。ペントースイソメラーゼは、五炭糖(ペントース)のうち構造異性体であるアルドペントースとケトペントースの異性化を触媒する酵素として定義される。本発明に用いられるペントースイソメラーゼは五炭糖の直接的な異性化を触媒する活性を持っていれば特に限定されないが、例えば、キシロースイソメラーゼ(EC5.3.1.5)やアラビノースイソメラーゼ(EC5.3.1.3)が挙げられる。この中で、例えばキシロースイソメラーゼはD-キシロース(アルドペントース)のD-キシルロース(ケトペントース)への、かつ/またはその逆の直接的な異性化を触媒する酵素であり、D-キシロースケトイソメラーゼとしても知られる。直接的な異性化とは、ペントースレダクターゼおよびペントールデヒドロゲナーゼにより触媒される、糖アルコール中間体を経た2段階転化とは異なり、ペントースイソメラーゼにより触媒される1段階での異性化のことを指す。
 本発明に用いる微生物は、ペントースイソメラーゼを用いて五炭糖を代謝する経路を有する微生物であれば特に制限はなく、元来ペントースイソメラーゼを有する微生物を用いてもよいし、適当な宿主にペントースイソメラーゼ遺伝子を導入してペントースイソメラーゼを機能させるようにした微生物を用いてもよい。さらに、使用する微生物は、自然環境から単離されたものでもよく、突然変異や遺伝子組換えによって一部性質が改変されたものでもよい。また、ケトペントースのリン酸化を触媒する活性を有する酵素を強化した微生物を用いてもよい。ケトペントースのリン酸化を触媒する活性を有する酵素としては、例えば、キシルロキナーゼ(EC 2.7.1.17)やリブロキナーゼ(EC 2.7.1.16)が挙げられる。
 ペントースイソメラーゼを用いて五炭糖を代謝する経路を有する微生物としては、例えば、クロストリジウム(Clostridium)属、エンテロバクター(Enterobacter)属、エシェリシア(Escherichia)属、クレブシエラ(Klebsiella)属、バクテロイデス属(Bacteroides)、エルビニア(Erwinia)属などの腸内細菌類や、ラクトバチルス(Lactobacillus)属などの乳酸菌類、またアクチノプラネス(Actinoplanes)属、アルスロバクター(Arthrobacter)属、ストレプトマイセス(Streptomyces)属などの放線菌類、ピロミセス属(Piromyces)やシラマイセス属(cyllamyces)などの真菌類、あるいはバチルス(Bacillus)属、パエニバチルス(Paenibacillus)属、アエロバクター(Aerobacter)属、アンプラリエラ(Ampullariella)属、スタフィロコッカス(Staphylococcus)属、サーモアナエロバクター(Thermoanaerobacter)属、サーマス(Thermus)属に属する微生物を用いることができ、具体的にはエシェリシア属、バチルス属、パエニバチルス属に属する微生物であり、さらに具体的にはエシェリシア・コリ(Escherichia coli)、バチルス・コアギュランス(Bacillus coagulans)、パエニバチルス・ポリミクサ(Paenibacillus polymyxa)を用いることができる。これらがペントースイソメラーゼを有することは、KEGG(Kyoto Encyclopedia of Genes and Genomes)やNCBI(National Center for Biotechnology Information)等のウェブにて公開されているデータベースにて検索を行うことで簡単に知ることができる。また、データベースに情報がない微生物であっても、以下に示す酵素活性測定によって知ることができる。
 ペントースイソメラーゼ活性は、ペントース存在下で酵素反応を行い、異性化されたペントールをHPLCなどを用いて測定することで確認することができる。例えばキシロースイソメラーゼ活性は、特開2008-79564に開示される方法で測定できる。またアラビノースイソメラーゼに関しても同様の方法が採用されうる。
 適当な宿主にペントースイソメラーゼ遺伝子を導入してペントースイソメラーゼを機能させるようにした微生物としては、特表2006-525029に開示される例を挙げることができる。導入するペントースイソメラーゼ遺伝子は特に限定されることはなく、ゲノムDNAのほか、cDNA等であってもよい。また、動物、植物、真菌(酵母、カビ等)、細菌などいずれの生物に由来するものであってもよい。こうした遺伝子に関する情報は、NCBI等のウェブにて公開されているデータベースにて検索を行うことで簡単に知ることができる。
 次に、本発明において分離膜として用いられる多孔性膜について説明する。
 本発明に用いる多孔質膜については特に制限はなく、微生物の撹拌型培養器あるいは撹拌型バイオリアクターによる培養で得られた培養液を微生物から分離濾過する機能を有するものであればよく、例えば多孔質セラミック膜、多孔質ガラス膜、多孔質有機高分子膜、金属繊維編織体、不織布などを用いることができる。これらの中で特に多孔質有機高分子膜もしくはセラミック膜が好適である。
 本発明で分離膜として用いられる多孔性膜の構成について説明する。本発明で用いられる多孔性膜は、好ましくは、被処理水の水質や用途に応じた分離性能と透水性能を有するものである。
 多孔性膜は、阻止性能および透水性能や分離性能、例えば、耐汚れ性の点から、多孔質樹脂層を含む多孔性膜であることが好ましい。
 多孔質樹脂層を含む多孔性膜は、好ましくは、多孔質基材の表面に、分離機能層として作用とする多孔質樹脂層を有している。多孔質基材は、多孔質樹脂層を支持して分離膜に強度を与える。
 本発明で用いられる多孔性膜が、多孔質基材の表面に多孔質樹脂層を有している場合、多孔質基材に多孔質樹脂層が浸透していても、多孔質基材に多孔質樹脂層が浸透していなくてもどちらでも良く、用途に応じて選択される。
 多孔質基材の平均厚みは、好ましくは50μm以上3000μm以下である。
 多孔質基材の材質は、有機材料および/または無機材料等からなり、有機繊維が望ましく用いられる。好ましい多孔質基材は、セルロース繊維、セルローストリアセテート繊維、ポリエステル繊維、ポリプロピレン繊維およびポリエチレン繊維などの有機繊維なる織布や不織布であり、より好ましくは、密度の制御が比較的容易であり製造も容易で安価な不織布が用いられる。
 多孔質樹脂層は、有機高分子膜を好適に使用することができる。有機高分子膜の材質としては、例えば、ポリエチレン系樹脂、ポリプロピレン系樹脂、ポリ塩化ビニル系樹脂、ポリフッ化ビニリデン系樹脂、ポリスルホン系樹脂、ポリエーテルスルホン系樹脂、ポリアクリロニトリル系樹脂、セルロース系樹脂およびセルローストリアセテート系樹脂などが挙げられる。有機高分子膜は、これらの樹脂を主成分とする樹脂の混合物であってもよい。ここで主成分とは、その成分が50重量%以上、好ましくは60重量%以上含有することをいう。有機高分子膜の材質は、溶液による製膜が容易で物理的耐久性や耐薬品性にも優れているポリ塩化ビニル系樹脂、ポリフッ化ビニリデン系樹脂、ポリスルホン系樹脂、ポリエーテルスルホン系樹脂およびポリアクリロニトリル系樹脂が好ましく、ポリフッ化ビニリデン系樹脂またはそれを主成分とする樹脂が最も好ましく用いられる。
 ここで、ポリフッ化ビニリデン系樹脂としては、フッ化ビニリデンの単独重合体が好ましく用いられる。さらに、ポリフッ化ビニリデン系樹脂は、フッ化ビニリデンと共重合可能なビニル系単量体との共重合体も好ましく用いられる。フッ化ビニリデンと共重合可能なビニル系単量体としては、テトラフルオロエチレン、ヘキサフルオロプロピレンおよび三塩化フッ化エチレンなどが例示される。
 本発明で分離膜として使用される多孔性膜は、特に制限はなく、発酵に使用される微生物が通過できなければよいが、発酵に使用される微生物の分泌物や発酵原料中の微粒子による目詰まりを起こりにくく、かつ、濾過性能が長期間安定に継続する範囲であることが望ましい。よって、多孔性分離膜の平均細孔径が、0.01μm以上5μm未満であることが好ましい。また、さらに好ましくは、多孔性膜の平均細孔径が、0.01μm以上1μm未満であると、微生物がリークすることのない高い排除率と、高い透水性を両立させることができ、透水性を長時間保持することが、より高い精度と再現性を持って実施することができる。
 微生物の大きさに近いと、これらが直接孔を塞いでしまう場合があるので、多孔性膜の平均細孔径は1μm未満であることが好ましい。多孔性膜の平均細孔径は、微生物の漏出、すなわち排除率が低下する不具合の発生を防止するため、微生物の大きさと比較して大きすぎないことが好ましい。微生物のうち、細胞の小さい細菌などを用いる場合には、平均細孔径として0.4μm以下が好ましく、0.2μm未満であればより好適に実施することができる。
 また、微生物が所望の生産物である化学品以外の物質、例えば、タンパク質や多糖類など凝集しやすい物質を生産する場合があり、更に、発酵培養液中の微生物の一部が死滅することにより細胞の破砕物が生成する場合があり、これらの物質による多孔性膜の閉塞を回避するために、平均細孔径は0.1μm以下であることがさらに好適である。
 平均細孔径が小さすぎると多孔性膜の透水性能が低下し、膜が汚れていなくても効率的な運転ができなくなるため、本発明における多孔性膜の平均細孔径は、好ましくは0.01μm以上であり、より好ましくは0.02μm以上であり、さらに好ましくは0.04μm以上である。
 ここで、平均細孔径は、倍率10,000倍の走査型電子顕微鏡観察における、9.2μm×10.4μmの範囲内で観察できる細孔すべての直径を測定し、平均することにより求めることができる。平均細孔径は、あるいは、膜表面を走査型電子顕微鏡を用いて倍率10,000倍で写真撮影し、10個以上、好ましくは20個以上の細孔を無作為に選び、それら細孔の直径を測定し、数平均して求めることもできる。細孔が円状でない場合、画像処理装置等によって、細孔が有する面積と等しい面積を有する円(等価円)を求め、等価円直径を細孔の直径とする方法により求めることができる。
 本発明で用いられる多孔性膜の平均細孔径の標準偏差σは、0.1μm以下であることが好ましい。平均細孔径の標準偏差σは小さければ小さい方が望ましい。平均細孔径の標準偏差σは、上述の9.2μm×10.4μmの範囲内で観察できる細孔数をNとして、測定した各々の直径をXとし、細孔直径の平均をX(ave)とした下記の(式1)により算出される。

 本発明で用いられる多孔性膜においては、発酵培養液の透過性が重要な性能の一つである。多孔性膜の透過性の指標として、使用前の多孔性膜の純水透過係数を用いることができる。本発明において、多孔性膜の純水透過係数は、逆浸透膜による25℃の温度の精製水を用い、ヘッド高さ1mで透水量を測定し算出したとき、5.6×10-10/m/s/pa以上であることが好ましく、純水透過係数が、5.6×10-10/m/s/pa以上6×10-7/m/s/pa以下であれば、実用的に十分な透過水量が得られる。
 本発明で用いられる多孔性膜において、表面粗さとは、表面に対して垂直方向の高さの平均値である。膜表面粗さは、分離膜表面に付着した微生物が、撹拌や循環ポンプによる液流による膜面洗浄効果で剥離しやすくするための因子の一つである。多孔性膜の表面粗さは、特に制限はなく、膜に付着した微生物、ならびにその他の固形物が剥がれる範囲であればよいが、0.1μm以下であることが好ましい。表面粗さが0.1μm以下であると、膜に付着した微生物、ならびにその他の固形物が剥がれやすくなる。
 さらに好ましくは、多孔性膜の膜表面粗さが0.1μm以下であり、平均細孔径が0.01μm以上1μm未満であり、多孔性膜の純水透過係数が2×10-9/m/s/pa以上の多孔性膜を使用することにより、膜面洗浄に必要な動力を過度に必要としない運転が、より容易に可能であることがわかった。多孔性膜の表面粗さを、0.1μm以下とすることにより、微生物の濾過において、膜表面で発生する剪断力を低下させることができ、微生物の破壊が抑制され、多孔性膜の目詰まりも抑制されることにより、長期間安定な濾過が、より容易に可能になる。また、多孔性膜の表面粗さを、0.1μm以下とすることにより、より低い膜間差圧で連続発酵が実施可能であり、多孔性膜が目詰まりした場合でも高い膜間差圧で運転した場合に比べて、洗浄回復性が良好である。多孔性膜の目詰まりを抑えることにより、安定した連続発酵が可能になることから、多孔性膜の表面粗さは小さければ小さいほど好ましい。
 ここで、多孔性膜の膜表面粗さは、下記の原子間力顕微鏡装置(AFM)を使用して、下記の条件で測定したものである。

・試料調製 測定に際し膜サンプルは、常温でエタノールに15分浸漬後、RO水中に24時間浸漬し洗浄した後、風乾し用いた。RO水とは、濾過膜の一種である逆浸透膜(RO膜)を用いて濾過し、イオンや塩類などの不純物を排除した水を指す。RO膜の孔の大きさは、概ね2nm以下である。
 膜表面粗さdroughは、上記の原子間力顕微鏡装置(AFM)により各ポイントのZ軸方向の高さから、下記の(式2)により算出する。

 本発明で用いられる多孔性膜の形状は、好ましくは平膜である。多孔性膜の形状が平膜の場合、その平均厚みは用途に応じて選択される。多孔性膜の形状が平膜の場合の平均厚みは、好ましくは20μm以上5000μm以下であり、より好ましくは50μm以上2000μm以下である。
 また、本発明で用いられる多孔性膜の形状は、好ましくは中空糸膜である。多孔性膜が中空糸膜の場合、中空糸の内径は、好ましくは200μm以上5000μm以下であり、膜厚は、好ましくは20μm以上2000μm以下である。また、有機繊維または無機繊維を筒状にした織物や編物を中空糸の内部に含んでいても良い。
 なお、前述の多孔性膜は、例えばWO2007/097260に記載される製造方法により製造することができる。
 また、別の好ましい形態として、本発明における分離膜は、少なくともセラミックスを含む膜であり得る。本発明におけるセラミックスとは金属酸化物を含有し、高温での熱処理により焼き固められたものをさす。金属酸化物としては、アルミナ、マグネシア、チタニア、ジルコニアなどが例示できる。分離膜は金属酸化物のみから形成されてもよく、シリカや炭化珪素、シリカと金属酸化物の化合物であるムライトやコージェライトなどを含んでもよい。
 分離膜を形成するセラミックス以外の成分は、分離膜としての多孔質体をなすことができるものであれば特に限定されない。
 分離膜がセラミックスを含む場合もその形状には特に制限はなく、モノリス膜、平膜、管状膜などいずれも用いることができる。容器内への充填効率の観点から柱状構造であり、長手方向に貫通孔を有する構造のものが好ましい。充填効率が上げられる点から、好ましくはモノリス膜である。
 長手方向に貫通孔を有することが好ましい理由は以下のとおりである。柱状構造を有する分離膜をモジュール容器に収めて分離膜モジュールとして用いる場合は、分離膜は外圧式および内圧式のうちから好適な態様を選択してモジュール化、濾過することが可能となる。本発明においては、これ以降、分離膜が発酵培養液と接する側を1次側、濾過により化学品を含む濾液が得られる側を2次側とそれぞれ呼ぶこととする。
 内圧式モジュールを用いると、1次側の流路が狭いため、クロスフロー濾過を行う場合に循環ポンプの出力が節約できる。さらに、分離膜表面に堆積した濁質をモジュール外に排出する作用が強くなり、分離膜表面が清浄に保たれやすいため好ましい。ただし、この効果を得るためには、内圧式の分離膜が発酵培養液の入り口および出口を有していることが前提となる。このときの入り口および出口は一直線に配された貫通孔の状態であると、通液抵抗が小さくなるため好ましい。さらに分離膜が柱状構造で、貫通孔が長手方向に空いていることによって、分離膜を収める容器を細くすることが可能となる。分離膜モジュールが細いと、製造や取り扱いの観点から好ましく用いることが出来る。
 分離膜の気孔率は特に限定されないが、低すぎると濾過効率が悪くなり、高すぎると強度が低下してしまう。濾過効率と分離膜の強度を両立させ、繰り返し蒸気滅菌への耐性も持たせるためには、20%以上60%以下であることが好ましい。
 なお気孔率は、以下の式より決定される。
気孔率[%]=100×(湿潤膜重量[g]-乾燥膜重量[g])/水比重[g/cm]/(膜体積[cm])。
 分離膜の平均気孔径は0.01μm以上1μm以下であれば好ましく、この範囲の平均気孔径を有する膜であれば分離膜が閉塞しにくく、かつ濾過効率にも優れる。また、平均気孔径が0.02μm以上0.2μm以下の範囲であれば、タンパク質、多糖類などに例示される微生物や培養細胞による発酵の副生成物や、培養液中の微生物/培養細胞が死滅して生じる細胞破砕物といった、分離膜を閉塞させやすい物質の閉塞が起きにくくなるため特に好ましい。
 貫通孔を有する柱状構造の分離膜は、外表面が2次側となるため、濾過液を集めるためにモジュール容器を設け、モジュール容器内に分離膜を充填したモジュールとして用いることが好ましい。1本のモジュールに充填される分離膜は1本以上である。
 モジュール容器は、繰り返しの蒸気滅菌処理に耐えうる原料からなることが好ましい。蒸気滅菌処理は、たとえばステンレス鋼や、平均気孔率の低いセラミックスなどが例示される。
 このようなセラミック膜モジュールは、例えばWO2012/086763に記載される製造方法により製造することができるが、市販のものを利用することもできる。市販のものとして具体的に例示すると、MEMBRALOX精密ろ過膜(Pall)、セラミック膜フィルターセフィルトMF膜(日本ガイシ)などが挙げられる。
 次に、連続発酵について説明する。
 本発明での連続発酵は、微生物の培養液を分離膜で濾過し未濾過液を培養液に保持または還流し、かつ発酵原料を培養液に追加して濾過液中から生産物を回収する連続発酵であることを特徴とする。
 微生物の培養は、用いる微生物に適したpHおよび温度を設定すればよく、微生物が生育する範囲であれば特に限定されないが、通常、pHが4~8で温度が20~75℃の範囲で行われる。培養液のpHは、無機あるいは有機の酸、アルカリ性物質、さらには尿素、炭酸カルシウム、アンモニアガスなどによって、通常、pH4~8範囲内のあらかじめ定められた値に調節する。
 培地には、菌体増殖に必要な栄養素を添加し、菌体増殖が連続的に行われるようにすればよい。培養液中の微生物の濃度は、化学品を効率よく製造するのに好ましい濃度であればよい。培養液中の微生物の濃度は、一例として、乾燥重量として、5g/L以上に維持することで良好な生産効率が得られる。
 本発明における培養の酸素条件は、酸素移動容量係数Kla(h-1)(以下、単にKlaと略す。)が150h-1以下であることが好ましい。
 Klaとは、通気攪拌時において、単位時間に気相から液相へ酸素を移動させ溶存酸素を生成させる能力を示すものであり、以下の式(3)で定義される。(生物工学実験書、日本生物工学会編、培風館、p.310(1992))。

 ここで、C:培養液中の溶存酸素濃度DO(ppm)、C:微生物による酸素の消費がない場合の気相と平衡の溶存酸素濃度DO(ppm)、Kla:酸素移動容量係数(hr-1)である。上記式(3)より、下記式(4)が導かれるため、通気した時間に対して、C-Cの対数をプロットすることで、Klaを求めることができる。

 また、本発明におけるKlaは気体置換法(ダイナミック法)により測定される数値である。気体置換法(ダイナミック法)とは、溶存酸素濃度電極を差し込んだ通気攪拌培養装置に水あるいは使用する培地を入れ、これらの液体中の酸素を窒素ガスにより置換して該液体の酸素濃度を低下させ、次に窒素ガスを圧縮空気に切り替え、所定の通気速度、攪拌速度および温度下での溶存酸素の上昇過程を測定することによりKlaを計算する方法である。
 Klaは通気条件と攪拌条件とを組み合わせて通気攪拌培養することで適宜設定できる。その場合通気は行っても行わなくてもよく、通気する気体も培養条件によって変えることが可能である。例えば、溶存酸素濃度を低く保つには、容器を密閉して無通気状態にする、窒素ガス等の不活性ガスを通気する、炭酸ガス含有の不活性ガスを通気する等の方法を用いることができる。一方溶存酸素濃度を高く保つには、空気に酸素を加えて酸素濃度を21%以上に保つ、あるいは培養液を加圧する等の方法を用いることができる。
 本発明で規定されるKlaの範囲において、攪拌速度一定下では以下の式(5)に示すようにKlaが通気速度に比例することが知られている。よって本発明での培養に採用される所定のKlaは、例えば攪拌速度一定下で通気速度を適当に変化させてKla測定を行い、得られたKlaを通気速度に対してプロットし式(5)に当てはまる定数aおよびbを求めることで設定することができる。

ここで、V:通気速度(vvm)、a,b:定数である。
 本発明で使用することができる、ペントースイソメラーゼを用いて五炭糖を代謝する経路を有する微生物は、五炭糖からペントールへの異性化反応においてNADHなどの補酵素を必要としないため、五炭糖の代謝が酸素条件によって影響されることはない。したがって酸素を利用しない嫌気的条件下であっても五炭糖の代謝が滞ることなく化学品を製造することができる。本発明の発酵培養において採用されるKlaは特に制限されないが、具体的には、Klaが150h-1以下であることが好ましく、より好ましくは100h-1以下、さらに好ましくは60h-1以下、最も好ましくは30h-1以下である。本酸素条件でペントースイソメラーゼを有する微生物を培養することにより、目的とする化学品の生産性が顕著に向上する。
 本発明の化学品の製造方法においては、濾過時の膜間差圧は特に制限されることはなく、発酵培養液を濾過できればよい。しかし、発酵培養液を濾過するために、有機高分子膜においては150kPaより高い膜間差圧で濾過処理すると、有機高分子膜の構造が破壊される可能性が高くなり、化学品を生産する能力が低下することがある。また、0.1kPaより低い膜間差圧では、発酵培養液の透過水量が十分得られない場合があり、化学品を製造するときの生産性が低下する傾向がある。したがって、本発明の化学品の製造方法では、有機高分子膜においては、濾過圧力である膜間差圧を好ましくは0.1kPaから150kPaの範囲とすることにより、発酵培養液の透過水量が多く、膜の構造の破壊による化学品製造能力の低下もないことから、化学品を生産する能力を高く維持することが可能である。膜間差圧は、有機高分子膜においては、好ましくは0.1kPa以上50kPa以下の範囲であり、さらに好ましくは0.1kPa以上20kPa以下の範囲である。
 セラミック膜を用いる場合においても、濾過時の膜間差圧は特に制限されることはなく、発酵培養液を濾過できればよいが、500kPa以下が好ましい。500kPa以上で運転すると、膜の目詰まりが発生し、連続発酵運転に不具合を生じることがある。
 濾過の駆動力としては、発酵液と多孔性膜処理水の液位差(水頭差)を利用したサイホン、またはクロスフロー循環ポンプにより分離膜に膜間差圧を発生させることができる。また、濾過の駆動力として分離膜2次側に吸引ポンプを設置してもよい。また、クロスフロー循環ポンプを使用する場合には、吸引圧力により膜間差圧を制御することができる。更に、発酵液側の圧力を導入する気体または液体の圧力によっても膜間差圧を制御することができる。これら圧力制御を行う場合には、発酵液側の圧力と多孔性膜処理水側の圧力差をもって膜間差圧とし、膜間差圧の制御に用いることができる。
 本発明では、培養初期にバッチ培養またはフェドバッチ培養を行って微生物濃度を高くした後に、連続培養(培養液のろ過)を開始しても良い。また、高濃度の菌体をシードし、培養開始とともに連続培養を行っても良い。本発明の化学品の製造方法では、適当な時期から、培地供給および培養液の濾過を行うことが可能である。培地供給と培養物の濾過の開始時期は、必ずしも同じである必要はない。また、培地供給と培養物の濾過は連続的であってもよいし、間欠的であってもよい。
 また、連続発酵における総濾過液中の糖濃度は、生産性の観点から5g/L以下とすることが望ましい。濾過液中の糖濃度は、培地の供給速度や培養液の濾過速度、あるいは培地中の糖濃度によって調節できる。
 本発明の化学品の製造方法では、連続発酵の途中において必要に応じて、発酵槽内から微生物を含んだ培養液の一部を取り除いた上、培地で希釈することによって、培養槽内の微生物濃度を調整してもよい。例えば、発酵槽内の微生物濃度が高くなりすぎると、分離膜の閉塞が発生しやすくなることから、微生物を含んだ培養液の一部を取り除き、培地で希釈することで、分離膜の閉塞を容易に回避することができる。また、発酵槽内の微生物濃度によって化学品の生産性能が変化することがあり、生産性能を指標として微生物を引き抜くことで生産性能を維持させることも可能である。
 本発明の化学品の製造方法に従って連続発酵を行った場合、従来の培養と比較して、高い収率が得られ、極めて効率のよい連続発酵生産が可能となる。ここで、連続培養における収率は、次の式(7)で計算される。
収率(g/g)=生産物量(g)÷{投入糖(g)-未利用糖(g)}・・・(式7)。
 本発明で用いられる連続培養装置は、微生物の発酵培養液を分離膜で濾過し、濾液から生産物を回収するとともに未濾過液を前記の発酵培養液に保持または還流し、かつ、発酵原料を前記の発酵培養液に追加して濾過液中の生産物を回収する連続発酵による化学品の製造装置であれば特に制限はないが、具体例を挙げると、有機高分子膜を使用する際は、WO2007/097260に記載される装置が使用できる。また、セラミック膜を使用する際は、WO2012/086763に記載される装置が使用できる。
 本発明により製造される化学品としては、上記微生物が発酵培養液中に生産する物質であれば制限はなく、アルコール、有機酸、アミノ酸、核酸など発酵工業において大量生産されている物質を挙げることができる。例えば、アルコールとしては、エタノール、1,3-プロパンジオール、1,3-ブタンジオール、2,3-ブタンジオール1,4-ブタンジオール、グリセロール、ブタノール、イソブタノール、2-ブタノール、イソプロパノールなど、有機酸としては、酢酸、乳酸、アジピン酸、ピルビン酸、コハク酸、リンゴ酸、イタコン酸、クエン酸、核酸であれば、イノシン、グアノシンなどのヌクレオシド、イノシン酸、グアニル酸などのヌクレオチド、また、カタベリンなどのジアミン化合物を挙げることができるが、中でも有機酸やアルコールが好ましく、さらに好ましくは乳酸、エタノール、2,3-ブタンジオールである。また、本発明は、酵素、抗生物質、組み換えタンパク質のような物質の生産に適用することも可能である。これら化学品は周知の方法(膜分離、濃縮、蒸留、晶析、抽出等)により濾過液より回収することができる。
 以下、実施例を挙げて本発明を具体的に説明する。但し、本発明はこれらに限定されるものではない。
 (参考例1)グルコース、キシロース、キシルロース、エタノール、2,3-ブタンジオールの分析方法
 発酵液中のグルコース、キシロース、キシルロース、エタノールおよび2,3-ブタンジオールの濃度は、下記に示すHPLC条件で、標品との比較により定量した。

 (参考例2)乳酸の分析方法
 発酵液中の乳酸を下記に示すHPLC条件で標品との比較により定量した。

 (参考例3)Kla測定
 培養槽に溶存酸素電極(メトラートレド製)を挿入して各通気攪拌条件に対する溶存酸素濃度を測定し、窒素ガスを用いるダイナミック法によりKlaを求めた。まず、培養槽に水1.5Lを張り、水温を30℃あるいは50℃に調整しながら一定の速度で攪拌して窒素ガスを十分に水中に吹き込み、電極値が最低値となった時点で溶存酸素電極のゼロ校正を行った。そののち通気ガスを窒素ガスから所定の通気速度の空気あるいは窒素ガスに切り替えてからの溶存酸素濃度の経時変化を測定してKlaを求めた。得られた各通気攪拌条件に対するKlaを表1に示す。

 表1より、攪拌速度800rpmで空気を通気したときの、通気速度とKlaの関係をプロットして得られる式(5)の定数(a,b)は30℃および50℃でそれぞれ(284,0.49)および(257,-0.47)であった。
 (参考例4)エシェリシア・コリのキシロースイソメラーゼ活性測定
 エシェリシア・コリKO11株を5mLの前培養培地(グルコース20g/L酵母エキス10g/L、トリプトン5g/L、NaCl5g/L)に白金耳にて植菌し、30℃で一晩培養をおこなった(前培養)。翌日、表2に示す50mLのエタノール発酵培地(混合糖)入りの500mLの三角フラスコに前培養液を植え継ぎ、30℃で24時間培養を行った。回収した培養液を200mMマレイン酸緩衝液(pH5.5)にて2回洗浄した。200mMマレイン酸緩衝液(pH5.5)で菌体を再懸濁し、ビーズ式ホモジナイザー(ビーズφ0.5を0.6g、4000rpm、1分間×5回)にて破砕した後、遠心分離によって上清を回収してこれを菌体抽出液とした。その後20mM MgSO、1mM CoCl、1mM MnCl、10mM キシロースを含む200mMマレイン酸緩衝液(pH5.5)900μLに菌体抽出液100μLを添加し、35℃で2時間反応させた後、反応液をHPLCで分析した。その結果、反応前と比較してキシロースのピークが減少し、キシルロースのピークが増大しており、キシロースイソメラーゼ活性を確認することができた。

 発酵原料としてバイオマス由来の糖液を使用した発酵を行った。発酵培地は、WO2010/067785の実施例2に開示されたナノ濾過膜による調製方法にて調製したセルロース糖化液を利用し、適宜試薬により発酵培地の組成を表14に示すとおり調製した。pH調製に4N KOHを用いた他は比較例2と同様の条件で24時間バッチ発酵を行い、エタノールを製造した(表17)。

 (比較例11)バイオマス由来糖液を発酵原料にしたエシェリシア・コリの連続発酵によるエタノールの製造
 発酵原料としてバイオマス由来の糖液を使用した、分離膜を利用しない連続発酵を行った。発酵培地は、WO2010/067785の実施例2に開示されたナノ濾過膜による調製方法にて調製したセルロース糖化液を利用し、適宜試薬により前培養培地として表14に示すエタノール発酵培地(糖液)1を、また連続培養中の培地として表15に示すエタノール発酵培地(糖液)2を調製した。pH調製に4N KOHを用いた他は比較例3と同様の条件で280時間連続発酵を行い、エタノールを製造した(表17)。

 (実施例18)バイオマス由来糖液を発酵原料にしたエシェリシア・コリの分離膜利用連続発酵によるエタノールの製造
 発酵原料としてバイオマス由来の糖液を使用した、分離膜を利用した連続発酵を行った。前培養培地は比較例17と同様にとして表14に示すエタノール発酵培地(糖液)1を、また連続培養中の培地として比較例17と同様に表15に示すエタノール発酵培地(糖液)2を用い、pH調製に4N KOHを用いた他は実施例1と同様の条件で290時間連続発酵を行い、エタノールを製造した(表17)。
 (比較例12)バイオマス由来糖液を発酵原料にしたバチルス・コアギュランスのバッチ発酵によるL-乳酸の製造
 発酵原料としてバイオマス由来の糖液を使用した発酵を行った。発酵培地は、WO2010/067785の実施例2に開示されたナノ濾過膜による調製方法にて調製したセルロース糖化液を利用し、適宜試薬により発酵培地の組成を表16に示すとおり調製した。pH調製に4N KOHを用い、Klaを0(h-1)とした他は比較例8と同様の条件で70時間バッチ発酵を行い、L-乳酸を製造した(表17)。

 (比較例13)バイオマス由来糖液を発酵原料にしたバチルス・コアギュランスの連続発酵によるL-乳酸の製造
 発酵原料としてバイオマス由来の糖液を使用した、分離膜を利用しない連続発酵を行った。発酵培地は、比較例12と同様に表16記載の培地を用いた。pH調製に4N KOHを用い、Klaを0(h-1)とした他は比較例9と同様の条件で250時間連続発酵を行い、L-乳酸を製造した(表17)。
 (実施例19)バイオマス由来糖液を発酵原料にしたバチルス・コアギュランスの分離膜利用連続発酵によるL-乳酸の製造
 発酵原料としてバイオマス由来の糖液を使用した、分離膜を利用した連続発酵を行った。発酵培地は、比較例12と同様に表16記載の培地を用いた。pH調製に4N KOHを用い、Klaを0(h-1)とした他は実施例11と同様の条件で260時間連続発酵を行い、L-乳酸を製造した(表17)。
 (実施例20)混合糖を発酵原料にしたパエニバチラス・ポリミクサのセラミック製分離膜を利用した連続発酵による2,3-ブタンジオールの製造6
 パエニバチラス・ポリミクサATCC12321株を用い、セラミック製分離膜を利用した連続発酵を行った。パエニバチラス・ポリミクサATCC12321株を試験管で2mLの前培養培地(グルコース5g/L、ペプトン5g/L、酵母エキス3g/L、麦芽エキス3g/L)にて30℃で一晩培養した(前々々培養)。得られた培養液を500mL容量のひだ付三角フラスコに入った50mLの前培養培地に植菌し、一晩培養した(前々培養)。前々培養液を表5に示す組成の2,3-ブタンジオール発酵培地1(混合糖)の入った連続培養装置(WO2007/097260の図2に示す装置)に植菌し、温度調整およびpH調整を行いながら以下に示す運転条件にて30時間バッチ発酵を行った(前培養)。前培養完了後、表7に示す組成の2,3-ブタンジオール発酵培地2(混合糖)を用いて直ちに連続培養を開始し、2,3-ブタンジオールの製造を行った。濾過時の膜間差圧は500kPa以下となるよう調整しながら、260時間の連続発酵により2,3-ブタンジオールを製造した(表17)。

 以上の結果から、五炭糖と六炭糖の混合糖から化学品を高収率で製造できた。
 本発明によって、五炭糖および六炭糖を含む発酵原料を用いた種々の化学品の発酵生産の効率を大幅に向上させることができる。

Claims (6)

  1.  微生物の培養液を分離膜で濾過すること、未濾過液を培養液に保持または還流すること、発酵原料を培養液に追加すること、およびろ過液中の生産物を回収することを含む、連続発酵による化学品の製造方法であって、前記発酵原料が五炭糖および六炭糖を含み、前記微生物がペントースイソメラーゼを用いて五炭糖を代謝する経路を有する、化学品の製造方法。
  2.  酸素移動容量係数(Kla)が150h-1以下の条件で連続発酵を行う、請求項1に記載の化学品の製造方法。
  3.  前記発酵原料中に含まれる六炭糖と五炭糖の重量比率が1:9~9:1である、請求項1または2に記載の化学品の製造方法。
  4.  前記発酵原料がバイオマス由来の糖液を含む、請求項1または2に記載の化学品の製造方法。
  5.  ペントースイソメラーゼがキシロースイソメラーゼである、請求項1~4のいずれかに記載の化学品の製造方法。
  6.  五炭糖がキシロースである、請求項1~5のいずれかに記載の化学品の製造方法。
