粒子検出装置及び粒子検出方法
 本技術は、粒子を光学的に検出する粒子検出装置及び粒子検出方法に関する。より詳しくは、光照射された粒子からの散乱光を検出する技術に関する。
 細胞、微生物及びリポソームなどの生体関連微小粒子の識別には、フローサイトメトリー(フローサイトメーター)を用いた光学的検出方法が利用されている。フローサイトメトリーは、流路内を1列になって通流する粒子に特定波長のレーザ光を照射し、各粒子から発せられた蛍光や散乱光を検出することで、複数の粒子を1個ずつ識別する分析手法である。
 例えば対象粒子が細胞である場合、前方散乱光(Forward Scatter:FS)を検出すると、その形、大きさ、表面状態、核の形成や有無などの情報を得ることができる。一般に、対象粒子からの前方散乱光は、フォトダイオードなどの光検出器により検出されるが、この光検出器には、照射光(以下、0次光ともいう。)などの前方散乱光以外の光も入射する。
 目的とする散乱光以外の光の影響を低減する技術としては、例えば、受光手段を複数の光電素子で形成すると共に、これら複数の光電素子の出力を加算処理する加算処理手段を設けた光散乱式粒子検出器がある(特許文献1参照)。また、散乱光の光路上に遮光マスクを配置し、不要な光を除去する技術もある。更に、従来、散乱光の検出効率向上のため、遮光マスクの代わりに光学フィルタを配置した微小粒子測定装置も提案されている(特許文献2参照)。

 そして、光学フィルタで遮断できなかった前方散乱光以外の光成分(漏れ光)は、光検出器に直接入射するため、検出信号のS/N比を低下させる原因となる。特に、検出対象の粒子の大きさが小さくなると、前方散乱光の信号レベルも小さくなるため、漏れ光の影響が更に大きくなる。
 そこで、本開示は、精度よく散乱光を検出することが可能な粒子検出装置及び粒子検出方法を提供することを主目的とする。
 本開示に係る粒子検出装置は、粒子に光を照射する光照射部と、光照射された粒子から発せられる散乱光を検出する散乱光検出部と、を有し、前記散乱光検出部は、前記散乱光を検出する第1検出部と、前記粒子への照射光に由来する光成分を検出する第2検出部と、前記第2検出部で検出された信号に基づいて前記第1検出部で検出された信号からノイズ成分を除去する信号処理部と、を備える。
 この粒子検出装置では、前記第1検出部と前記第2検出部とが一の光検出器に設けられていてもよい。
 その場合、前記第2検出部の周囲に前記第1検出部を設けることができる。
 また、前記光検出器は、受光面が9以上の領域に分割されていてもよい。
 更に、前記光検出器の手前に、前記照射光に由来する光成分を遮断する照射光除去部材を配置することもできる。
 一方、前記第1検出部として機能する散乱光検出器と、前記第2検出部として機能する照射光検出器と、を有していてもよい。
 その場合、前記散乱光の光路上に、前記照射光検出器と前記散乱光検出器をこの順に配置することができる。
 また、前記照射光検出器と前記散乱光検出器との間に、前記照射光に由来する光成分を遮断する照射光除去部材が配置されていてもよい。
 又は、前記照射光に由来する光成分の光路を変更するミラーを有し、前記照射光検出器を前記散乱光の光路外に配置することもできる。
 その場合、前記ミラーと前記散乱光検出器との間に、前記照射光に由来する光成分を遮断する照射光除去部材を配置することもできる。
 前記信号処理部は、前記第1検出部の検出信号から、前記第2検出部の検出信号にゲイン値を乗じた信号を減ずることにより、ノイズ成分を除去してもよい。
 また、前記散乱光は、例えば前方散乱光である。
 更に、前記光照射部は、流路を通流する粒子に光を照射してもよく、前記流路は、マイクロチップに形成されていてもよい。
 本開示に係る粒子検出方法は、粒子に光を照射する光照射工程と、第1検出部において前記粒子から発せられた散乱光を検出する散乱光検出工程と、第2検出部において前記粒子への照射光に由来する光成分を検出する照射光工程と、前記第2検出部で検出された信号に基づいて前記第1検出部で検出された信号からノイズ成分を除去する信号処理工程と、を有する。
 前記信号処理工程では、前記第1検出部の検出信号から、前記第2検出部の検出信号にゲイン値を乗じた信号を減ずることにより、ノイズ成分を除去してもよい。
 また、前記粒子が存在しない状態で光を照射したときの前記第1検出部における検出信号及び第2検出部における検出信号に基づいて前記ゲイン値を設定することもできる。
 本開示によれば、散乱光検出信号からノイズ成分を除去しているため、精度よく散乱光を検出することができる。

 以下、本開示を実施するための形態について、添付の図面を参照して詳細に説明する。なお、本開示は、以下に示す各実施形態に限定されるものではない。また、説明は、以下の順序で行う。

<1.第1の実施形態>
[装置の全体構成]
 先ず、本開示の第1の実施形態の粒子検出装置について、流路内を通流する微小粒子を検出する場合を例にして説明する。図1は本実施形態の粒子検出装置の構成を模式的に示す図である。図1に示すように、本実施形態の粒子検出装置は、微小粒子3に光を照射する光照射部1と、光照射された微小粒子3から発せられる散乱光7を検出する散乱光検出部4を備えている。
[光照射部1]
 光照射部1には、レーザ光などの励起光5を発生する光源11と、光源11で発生した励起光5を微小粒子3に向けて集光するレンズ12などが設けられている。そして、例えばマイクロチップ2内に形成された流路21内を通流する微小粒子3に励起光5を照射する。
[マイクロチップ2]
 マイクロチップ2には、微小粒子3が通流可能な流路21が設けられており、流路21内に、例えば検出対象の微小粒子3を含むサンプル液が導入される。このマイクロチップ2は、ガラスや各種プラスチック(PP,PC,COP、PDMSなど)により形成することができる。マイクロチップ2の材質は、特に限定されるものではないが、光照射部1から照射される励起光5に対して透過性を有し、光学誤差が少ない材質とすることが望ましい。
 マイクロチップ2の成形は、ガラス製基板のウェットエッチングやドライエッチングによって、またプラスチック製基板のナノインプリントや射出成型、機械加工によって行うことができる。そして、流路21などを成形した基板を、同じ材質又は異なる材質の基板で封止することでマイクロチップ2を形成することができる。
[微小粒子3]
 本実施形態の粒子検出装置で検出される「微小粒子3」には、細胞、微生物及びリボゾームなどの生体関連微小粒子、又はラテックス粒子、ゲル粒子及び工業用粒子などの合成粒子などが広く含まれる。
 生体関連微小粒子には、各種細胞を構成する染色体、リボゾーム、ミトコンドリア、オルガネラ(細胞小器官)などが含まれる。また、細胞には、植物細胞、動物細胞及び血球系細胞などが含まれる。更に、微生物には、大腸菌などの細菌類、タバコモザイクウイルスなどのウイルス類、イースト菌などの菌類などが含まれる。この生体関連微小粒子には、核酸や蛋白質、これらの複合体などの生体関連高分子も包含され得るものとする。
 工業用粒子としては、有機高分子材料、無機材料又は金属材料などで形成されたものが挙げられる。有機高分子材料としては、ポリスチレン、スチレン・ジビニルベンゼン、ポリメチルメタクリレートなどを使用することができる。また、無機材料としては、ガラス、シリカ及び磁性材料などを使用することができる。金属材料としては、例えば金コロイド及びアルミニウムなどを使用することができる。なお、これら微小粒子の形状は、一般には球形であるが、非球形であってもよく、また大きさや質量なども特に限定されない。
[散乱光検出部4]
 散乱光検出部4は、励起光5が照射された微小粒子3から発せられる散乱光7を検出するものであり、光検出器41及び信号処理部42などが設けられている。
(光検出器41)
 光検出器41は、フォトダイオードなどで構成されており、光照射された微小粒子3から発せられる前方散乱光7と共に、励起光5などの前方散乱光7以外の光成分(0次光6)を検出する。図2は光検出器41の検出領域を示す模式図である。図2に示すように、光検出器41の受光面は、0次光6を検出する0次光検出領域41bと、前方散乱光7を検出する散乱光検出領域41aとに分割されている。
 前方散乱光7は、励起光5の光軸に対して鋭角(~19°)に散乱する光であるため、光検出器41に入射する光のうち、光軸周辺の光成分は主に0次光6であり、前方散乱光7はほとんど含まれていない。そこで、光検出器41の受光面は、中心部を0次光検出領域41bとし、その周囲に散乱光検出領域41aを設ける。
 ここで、光検出器41の受光面の分割数は、特に限定されるものではないが、後述する差動効果向上の観点から、9分割(3×3)以上であることが好ましく、16分割(4×4)以上であることがより好ましい。光検出器41の受光面の分割数を多くすることにより、後述する信号処理部42での信号処理において、ゲイン値を用いなくても散乱光検出領域41aで検出された信号からノイズ成分を除去することが可能となる。
(信号処理部42)
 信号処理部42は、0次光検出領域41bで検出された信号に基づいて、散乱光検出領域41aで検出された信号からノイズ成分を除去する。ここで、光検出器41に入射する光には、光源11の駆動電流ノイズ、励起光5であるレーザ光のノイズ、光路で受けるノイズなど多種多様のノイズが含まれている。これらのノイズは、レベルは異なるが、同じ位相で、0次光(漏れ光)6及び前方散乱光7の両方に含まれている。
 そこで、本実施形態の粒子検出装置では、例えば、各領域で検出された信号に対して、下記数式1に示す処理を行い、得られた信号を前方散乱光信号とする。なお、下記数式1におけるゲイン値(Gain)は、微小粒子3を通流させずに励起光5を照射したときの散乱光検出領域41aの検出信号及び0次光検出領域41bの検出信号に基づいて設定した値である。

 なお、本実施形態の粒子検出装置には、必要に応じて、側方散乱光を検出する他の散乱光検出部や、微小粒子3から発せられた蛍光を検出する蛍光検出部などが設けられていてもよい。
[動作]
 次に、本実施形態の粒子検出装置の動作、即ち、本実施形態の粒子分検出装置を使用して微小粒子3を検出する方法について説明する。本実施形態の粒子検出装置では、マイクロチップ2に設けられた流路21内に、例えば検出対象の微小粒子3を含むサンプル液を導入する。そして、光源11から出射した励起光5をレンズ12などで集光して、マイクロチップ2の流路21内を通流する微小粒子3に照射する。
 そして、光検出器41により、励起光5が照射された微小粒子3から発せられた前方散乱光7を検出する。このとき、光検出器41には、前方散乱光7の他に、励起光5が漏れ込んだ0次光6も入射する。そこで、本実施形態の粒子検出装置では、光検出器41の受光面に、0次光検出領域41bと散乱光検出領域41aを設け、その両方を検出する。
 図3Aは散乱光検出領域41aで検出された信号を示す図であり、図3Bは0次光検出領域41bで検出された信号を示す図である。図3Aに示すように、散乱光検出領域41aでは、0次光6に含まれるノイズと、前方散乱光7の信号が検出される。一方、図3Bに示すように、0次光検出領域41bでは、前方散乱光7の信号はほとんど検出されず、主に0次光6に含まれるノイズ成分が検出される。
 ここで、散乱光検出領域41aで検出される励起光5に起因するノイズと、0次光検出領域41bで検出される0次光6に含まれるノイズは、レベルの違いはあるが、同位相である。そこで、本実施形態の粒子検出装置では、信号処理部42において、0次光検出領域41bで検出された信号に基づいて、散乱光検出領域41aで検出された信号からノイズ成分を除去する。
 具体的には、上記数式1に示す散乱光検出領域41aの検出信号から、0次光検出領域41bの検出信号にゲイン値を乗じた信号を減ずる処理を行う。図4は信号処理部42でノイズ成分が除去された信号を示す図である。これにより、図4に示すように、ほぼ前方散乱光7に由来する信号の波形のみが得られる。
 ここで、信号処理部42で行う処理で用いるゲイン値は、前方散乱光7と0次光6のノイズが0になるように調整された値である。例えば、流路21に、微小粒子3を含有しないサンプル液を流し、その状態でゲイン値を変化させながら信号に含まれるノイズレベルの変化を検出し、ノイズがなくなった時の値とする。
 以上詳述したように、本実施形態の粒子検出装置では、0次光検出領域41bで検出された信号に基づいて、散乱光検出領域41aで検出された信号からノイズ成分を除去しているため、前方散乱光の検出精度を向上させることができる。また、本実施形態の粒子検出装置は、散乱光検出領域41aに入射する漏れ光(0次光6)に含まれる低周波及び高周波のノイズを除去することができるため、微小粒子3からの前方散乱光7が微小であっても、検出することが可能となる。これにより、散乱光の検出能力が向上し、理想的な散乱光信号のみの検出を実現することが可能となる。
 フローサイトメータのように流路内を通流する微小粒子の散乱光を検出する手法では、0次光と散乱光とを区別することが難しいが、本技術を適用することにより、これらを分離し、精度よく前方散乱光を検出することが可能となる。なお、本開示は、マイクロチップ2を用いて、微小粒子3を検出する場合に限定されるものではなく、種々の粒子検出装置に適用することが可能である。
 また、本技術は、前方散乱光だけでなく、他の散乱光の検出に適用することも可能であり、その場合も同様の効果が得られる。
<2.第1の実施形態の変形例>
 次に、本開示の第1の実施形態の変形例に係る粒子検出装置について説明する。図5は本変形例の粒子検出装置の構成を模式的に示す図であり、図6は光検出器41の検出領域と0次光除去部材43の位置関係を示す図である。なお、図5及び図6では、前述した第1の実施形態の粒子検出装置の構成要素と同じものには、同じ符号を付し、その詳細な説明は省略する。
 図5及び図6に示すように、本変形例の粒子検出装置では、光検出器41の手前に、0次光6を遮断する0次光除去部材43が配置されている。ここで、0次光除去部材43には、マスクや特定光を選択的に遮断する光学フィルタなどを使用することができるが、これらに限定されるものではなく、0次光6を遮断可能な光学部材であればよい。
 散乱光検出領域41aに漏れ込む0次光6の信号強度が高い場合、信号処理に用いるゲイン値の調整が難しくなることがあるが、本変形例のように0次光除去部材43を配置することで、0次光6の強度を下げることができる。これにより、0次光6の信号強度が高い場合でも、ノイズ成分を除去し、精度よく前方散乱光を検出することができる。
 なお、本変形例における上記以外の構成、動作及び効果は、前述した第1の実施形態と同様である。
<3.第2の実施形態>
 次に、本開示の第2の実施形態に係る粒子検出装置について説明する。図7は本実施形態の粒子検出装置の構成を模式的に示す図であり、図8は散乱光検出器44の検出領域と0次光検出器45の位置関係を示す図である。なお、図7及び図8では、前述した第1の実施形態の粒子検出装置の構成要素と同じものには、同じ符号を付し、その詳細な説明は省略する。
 図7及び図8に示すように、本実施形態の粒子検出装置では、光路上に、0次光6を検出する0次光検出器45と、散乱光7を検出する散乱光検出器44とが、この順に配置されている。ここで、0次光検出器45及び散乱光検出器44には、フォトダイオードなどの一般に用いられている光検出器を用いることができる。
 本実施形態の粒子検出装置では、信号処理部42において、0次光検出器45で検出された信号に基づき、散乱光検出器44で検出された信号からノイズ成分を除去する。これにより、前述した第1の実施形態と同様に、前方散乱光の検出精度を向上させることができる。
 なお、本実施形態における上記以外の構成、動作及び効果は、前述した第1の実施形態と同様である。
<4.第2の実施形態の第1変形例>
 次に、本開示の第2の実施形態の第1変形例に係る粒子検出装置について説明する。図9は本変形例の粒子検出装置の構成を模式的に示す図であり、図10は散乱光検出器44の検出領域と0次光検出器45と0次光除去部材43の位置関係を示す図である。なお、図9及び図10では、前述した第1の実施形態及び第2の実施形態の粒子検出装置の構成要素と同じものには、同じ符号を付し、その詳細な説明は省略する。
 図9及び図10に示すように、本変形例の粒子検出装置では、0次光検出器45と散乱光検出器44との間に、0次光6を遮断する0次光除去部材43が配置されている。0次光検出器45と共に0次光除去部材43を配置することにより、ノイズ除去効果を更に高め、散乱光の検出精度を向上させることができる。なお、本変形例における上記以外の構成、動作及び効果は、前述した第1の実施形態、その変形例及び第2の実施形態と同様である。
<5.第2の実施形態の第2変形例>
 図7に示す第2の実施形態では、光路上に、0次光検出器45と散乱光検出器44とを配置しているが、本開示はこれに限定されるものではなく、0次光検出器45を光路外に配置することもできる。図11は本変形例の粒子検出装置の散乱光検出部4の構成を模式的に示す図である。
 図11に示すように、本変形例の粒子検出装置では、0次光検出器45を光路外に配置し、光路上に配置したミラー46により、0次光6の光路を変更し、0次光検出器45に導光する。この場合、0次光6の漏れ込みを低減するため、ミラー46と散乱光検出器44との間に、0次光除去部材43を配置することが好ましい。
 本変形例の粒子検出装置においても、信号処理部42において、0次光検出器45で検出された信号に基づき、散乱光検出器44で検出された信号からノイズ成分を除去する。これにより、前述した第変形例と同様に、高精度で前方散乱光を検出することが可能となる。
 また、本開示は、以下のような構成をとることもできる。

  1.  粒子に光を照射する光照射部と、
     光照射された粒子から発せられる散乱光を検出する散乱光検出部と、
    を有し、
     前記散乱光検出部は、
      前記散乱光を検出する第1検出部と、
      前記粒子への照射光に由来する光成分を検出する第2検出部と、
      前記第2検出部で検出された信号に基づいて前記第1検出部で検出された信号からノイズ成分を除去する信号処理部と、
    を備える粒子検出装置。
  2.  前記第1検出部と前記第2検出部とが一の光検出器に設けられている請求項1に記載の粒子検出装置。
  3.  前記第2検出部の周囲に前記第1検出部が設けられている請求項2に記載の粒子検出装置。

  16.  前記信号処理工程は、前記第1検出部の検出信号から、前記第2検出部の検出信号にゲイン値を乗じた信号を減ずることにより、ノイズ成分を除去する請求項15に記載の粒子検出方法。
  17.  前記粒子が存在しない状態で光を照射したときの前記第1検出部における検出信号及び第2検出部における検出信号に基づいて前記ゲイン値を設定する請求項16に記載の粒子検出方法。
