Abstract

 日本脳炎ワクチン抗原、塩基性アミノ酸及び酸を含有し、塩基性アミノ酸1モルに対する酸のモル数が、酸の価数をNとしたときに、1/(N+1)を超え2未満である、マイクロニードルコーティング用組成物が開示される。

Description

マイクロニードルコーティング用組成物及びマイクロニードルデバイス
 本発明は、マイクロニードルコーティング用組成物及びマイクロニードルデバイスに関するものである。
 薬剤を経皮投与する手段の一つとしてマイクロニードルデバイスが知られている。このようなデバイスとしては、薬物と増粘剤を含有するコーティングが形成されたものがある(例えば、特許文献1)。
 また、生物学的活性物質及び粘度向上対イオンの調製物を含んだ組成物(特許文献2)、コーティング処方物のpHがおおよそpH6未満であり、少なくとも1種類の低揮発性対イオンを含有するもの(特許文献3)が知られている。さらに、治療有効量のペプチド薬剤及び少なくとも1つの対イオンの製剤を含んだ組成物(特許文献4)、乾燥された場合に増大したpH安定性および溶解性を有する、生物学的有効成分および非揮発性の対イオンの製剤を含む組成物(特許文献5)が知られている。これらの他、生物活性物質と添加剤を含有するコーティングであって、添加材の一例として低含有量のアミノ酸を用いるものがある(特許文献6)。

 しかしながら、特許文献1及び2において、増粘剤として例示されている材料を含有させると、薬剤に含まれるタンパク質などの生理活性物質の安定性が十分でないことが見出された。
 そこで、本発明の目的は、含有する日本脳炎ワクチン抗原を安定化させることが可能なマイクロニードルコーティング用組成物、及びこの組成物から形成されるコーティング層を備えるマイクロニードルデバイスを提供することにある。
 本発明のマイクロニードルコーティング用組成物は、日本脳炎ワクチン抗原、塩基性アミノ酸及び酸を含有し、塩基性アミノ酸1モルに対する酸のモル数が、酸の価数をNとしたときに、1/(N+1)を超え2未満である。
 塩基性アミノ酸1モルに対する酸のモル数が1/(N+1)以下であると、コーティング用組成物中の酸の含有量が低く、塩基性アミノ酸を溶解させることができない。一方、塩基性アミノ酸1モルに対する酸のモル数が2以上であると、コーティング用組成物中の塩基性アミノ酸の含有量が低いため、日本脳炎ワクチン抗原の安定性が低下する。よって、塩基性アミノ酸1モルに対する酸のモル数が上記範囲であることにより、含有する日本脳炎ワクチン抗原を安定に存在させることが可能となる。
 マイクロニードルコーティング用組成物において、酸は、融点が40℃以上の酸であることが好ましく、リン酸、乳酸、安息香酸、マレイン酸、クエン酸、酒石酸、コハク酸、アスコルビン酸及びアスパラギン酸からなる群より選ばれる少なくとも1種の酸であることがより好ましく、リン酸、クエン酸及び酒石酸からなる群より選ばれる少なくとも1種の酸であることがさらに好ましい。このような酸を用いることにより、コーティング用組成物中の塩基性アミノ酸の濃度を高めることができ、含有する日本脳炎ワクチン抗原の安定性をより向上させることができる。
 マイクロニードルコーティング用組成物において、塩基性アミノ酸は、アルギニンであることが好ましい。塩基性アミノ酸としてアルギニンを用いることにより、コーティング用組成物中の日本脳炎ワクチン抗原の安定性を顕著に向上させることができる。
 マイクロニードルコーティング用組成物において、リジン又はリジン塩酸塩を含有することが好ましい。リジン又はリジン塩酸塩を含有することにより、日本脳炎ワクチン抗原の安定性をより向上させることができる。
 本発明はまた、マイクロニードル上に、上記マイクロニードルコーティング用組成物から形成されるコーティング層を備える、マイクロニードルデバイスを提供する。ここで、コーティング層は、マイクロニードルの先端部分に形成されることが好ましく、マイクロニードルの先端部分にのみ形成されていることがより好ましい。
 本発明によれば、含有する日本脳炎ワクチン抗原を安定化させることが可能なマイクロニードルコーティング用組成物及びマイクロニードルデバイスを提供することが可能となる。

 以下、図面を参照しながら、好適な実施形態を説明する。なお、図面の説明において同一要素には同一符号を付し、重複する説明を省略する。また、図面は理解を容易にするため一部を誇張して描いており、寸法比率は説明のものとは必ずしも一致しない。
 図1は、マイクロニードルデバイスの一実施形態を示す斜視図である。図1に示すマイクロニードルデバイス1は、基板2と、基板2上に二次元状に配置された複数のマイクロニードル3と、マイクロニードル3上に形成されたコーティング層5とを備えている。コーティング層5は、本発明のマイクロニードルコーティング用組成物から形成されるものであり、その揮発成分の少なくとも一部が除去されていることが好ましい。
 基板2は、マイクロニードル3を支持するための土台である。基板2の面積は、0.5~10cmであることが好ましく、より好ましくは1~5cm、さらに好ましくは1~3cmである。この基板2を数個つなげることで所望の大きさの基板を構成するようにしてもよい。
 マイクロニードル3は微小構造であり、その高さ(長さ)は、好ましくは50~600μmである。ここで、マイクロニードル3の長さを50μm以上とすることにより、マイクロニードルコーティング用組成物に含まれる日本脳炎ワクチン抗原の投与を確実になる。また、マイクロニードル3の長さを600μm以下とすることにより、マイクロニードルが神経に接触するのを回避して、痛みの可能性を確実に減少させるとともに、出血の可能性を確実に回避することができるようになる。また、マイクロニードル3の長さが500μm以下であると、皮内に入るべき量の日本脳炎ワクチン抗原を効率良く投与することができ、基底膜を穿孔させずに投与することも可能である。マイクロニードル3の長さは、300~500μmであることが特に好ましい。
 ここで、マイクロニードル3とは、凸状構造物であって、広い意味での針形状、又は針形状を含む構造物を意味する。もっとも、マイクロニードルは、鋭い先端を有する針形状のものに限定されるものではなく、先の尖っていない形状のものであってもよい。マイクロニードル3が円錐状構造である場合には、その基底における直径は50~200μm程度であることが好ましい。本実施形態ではマイクロニードル3は円錐状であるが、四角錐などの多角錐状や、別の形状のマイクロニードルであってもよい。
 マイクロニードル3は、典型的には、針の横列について1ミリメートル(mm)当たり約1~10本の密度となるように間隔を空けて設けられる。一般に、隣接する横列は横列内の針の空間に対して実質的に等しい距離だけ互いに離れており、1cm当たり100~10000本の針密度を有する。100本以上の針密度があると、効率良く皮膚を穿孔することができる。一方、10000本を超える針密度では、マイクロニードル3の強度を保つことが難しくなる。マイクロニードル3の密度は、好ましくは200~5000本、より好ましくは300~2000本、さらに好ましくは400~850本である。
 基板2又はマイクロニードル3の材質としては、シリコン、二酸化ケイ素、セラミック、金属(ステンレス、チタン、ニッケル、モリブテン、クロム、コバルト等)及び合成又は天然の樹脂素材等が挙げられるが、マイクロニードルの抗原性及び材質の単価を考慮すると、ポリ乳酸、ポリグリコリド、ポリ乳酸-co-ポリグリコリド、プルラン、カプロノラクトン、ポリウレタン、ポリ無水物等の生分解性ポリマーや、非分解性ポリマーであるポリカーボネート、ポリメタクリル酸、エチレンビニルアセテート、ポリテトラフルオロエチレン、ポリオキシメチレン等の合成又は天然の樹脂素材が特に好ましい。また、多糖類であるヒアルロン酸、ヒアルロン酸ナトリウム、プルラン、デキストラン、デキストリン又はコンドロイチン硫酸等も好適である。
 基板2又はマイクロニードル3の製法としては、シリコン基板を用いたウエットエッチング加工又はドライエッチング加工、金属若しくは樹脂を用いた精密機械加工(放電加工、レーザー加工、ダイシング加工、ホットエンボス加工、射出成型加工等)、機械切削加工等が挙げられる。これらの加工法により、基板2とマイクロニードル3は、一体に成型される。マイクロニードル3を中空にする方法としては、マイクロニードル3を作製後、レーザー等で2次加工する方法が挙げられる。
 マイクロニードルデバイス1は、マイクロニードル3上にコーティング層5を備えているが、コーティング層5は、マイクロニードルコーティング用組成物を塗付することにより形成されることが好ましい。塗付方法としては、噴霧コーティング及び浸漬コーティング等が挙げられ、浸漬コーティングが好ましい。なお、図1では、全てのマイクロニードル3にコーティング層5が形成されているが、コーティング層5は複数存在するマイクロニードル3の一部だけに形成されていてもよい。図1ではまた、コーティング層5はマイクロニードル3の先端部分だけに形成されているが、マイクロニードル3の全体を覆うように形成されていてもよい。さらには、コーティング層5は基板2の上に形成されていてもよい。
 図3(a)、(b)及び(c)は、マイクロニードルデバイス1の製造方法の一実施形態を示す図である。この方法では、まず、図3(a)に示すように、マイクロニードルコーティング用組成物10をマスク版11上でヘラ12により矢印A方向に掃引する。これにより、開口部13にマイクロニードルコーティング用組成物10が充填される。続いて、図3(b)に示すように、マスク版11の開口部13にマイクロニードル3を挿入する。その後、図3(c)に示すように、マスク版11の開口部13からマイクロニードル3を引き出す。これにより、マイクロニードル3上にマイクロニードルコーティング用組成物10を付着させる。なお、マイクロニードルコーティング用組成物10は基板2上に付着させてもよい。その後、風乾、真空乾燥、又はそれらの組み合わせの既知の方法により、マイクロニードル3上のマイクロニードルコーティング用組成物10の揮発成分を除去する。これにより、マイクロニードル3上にコーティング層5が強固に付着し、典型的にはガラス質又は固形状になり、マイクロニードルデバイスが製造される。コーティング層5の水分含有量は通常、コーティング層5の全量基準で55質量%以下、好ましくは30質量%以下、さらに好ましくは10質量%以下である。上記方法により、付着したマイクロニードルコーティング用組成物10の液ダレが防止されるが、液ダレとは、針先からコーティング組成物が垂れてくることを指し、図3(c)ではH部分が長くなることを意味する。
 マイクロニードル3上に付着しているコーティング層5の高さHは、図3(b)に示すクリアランス(ギャップ)Cで調整される。このクリアランスCは、マイクロニードル3の基底からマスク版11表面までの距離(基板2の厚みは関与しない)で定義され、マスク版11のテンションとマイクロニードル3の長さに応じて設定される。クリアランスCの距離の範囲は、好ましくは、0~500μmである。クリアランスCの距離が0の場合には、マイクロニードルコーティング用組成物10がマイクロニードル3の全体に対して塗付されることを意味する。マイクロニードル3上に付着しているマイクロニードルコーティング用組成物10の高さHはマイクロニードル3の高さによって変動するが、0~500μmとすることができ、通常10~500μmであり、好ましくは30~300μm程度で、特に好ましくは40~250μm程度である。マイクロニードルコーティング用組成物10中の日本脳炎ワクチン抗原を有効に用いるためには、マイクロニードルの一部分、すなわちニードルの先端部分に集中的に存在させた方が好ましく、また、皮膚に対する刺激及び薬物の皮膚への移行率の観点からも、先端から200μmまでに存在させることが好ましい。さらに、日本脳炎抗原ワクチンの安定性の観点から、日本脳炎ワクチン抗原の濃度を、コーティング層5の全質量を基準として、0.1~10%w/wとする場合、先端から50~150μmまでに存在させることがより好ましい。マイクロニードルコーティング用組成物10は塩基性アミノ酸を高濃度(例えば、20%w/w以上)に水系溶液中に溶解させることができ、高い粘度を有することから、マイクロニードルの一部分にコーティング層5を形成することが可能となる。このような形でマイクロニードル3上に保持されたマイクロニードルコーティング用組成物10は、マイクロニードル3を皮膚へ穿刺させたときに同時に皮内へ挿入される。
 マイクロニードル3上に付着しているコーティング層5の乾燥後の厚さは50μm未満であることが好ましく、より好ましくは40μm未満、さらに好ましくは1~30μmである。一般に、マイクロニードル上に付着しているコーティング層5の厚さは、乾燥後にマイクロニードル3の表面にわたって測定される平均の厚さである。マイクロニードル3上に付着しているコーティング層5の厚さは、マイクロニードルコーティング用組成物10の複数の被膜を適用することにより増大させること、すなわち、マイクロニードルコーティング用組成物10を付着させた後に付着工程を繰り返すことで増大させることができる。
 マイクロニードルコーティング用組成物10をマイクロニードル3に付着させる際には、装置の設置環境の温湿度は、一定に制御されることが好ましい。また、マイクロニードルコーティング用組成物10が水を含有する場合は、必要に応じて、水を充満させることもできる。これにより、マイクロニードルコーティング用組成物10中の水の蒸散を極力防ぐことができる。
 図2は図1のII-II線断面図である。図2に示すように、マイクロニードルデバイス1は、基板2と、基板2上に設けられた、マイクロニードル3と、当該マイクロニードル3上に設けられたコーティング層5と、を備えるものである。マイクロニードル上に付着しているコーティング層5は、日本脳炎ワクチン抗原、塩基性アミノ酸及び酸を含有するものであり、例えば上述した工程を経て製造することができる。
 マイクロニードルコーティング用組成物10は、日本脳炎ワクチン抗原、塩基性アミノ酸及び酸を含有し、塩基性アミノ酸1モルに対する酸のモル数が、酸の価数をNとしたときに1/(N+1)を超え2未満である。塩基性アミノ酸1モルに対する酸のモル数は、1/N以上1以下であることが好ましい。
 マイクロニードルコーティング用組成物10中の日本脳炎ワクチン抗原の濃度は、マイクロニードルコーティング用組成物10の全質量を基準として、0.01~30%w/wであることが好ましく、0.02~20%w/wであることがより好ましく、0.05~10%w/wであることがさらに好ましい。日本脳炎ワクチン抗原の濃度が0.01%w/w以上であると、皮膚への投与の際、有効量の日本脳炎ワクチン抗原を皮内に放出させることができ、十分な薬効が発揮される。また、マイクロニードルコーティング用組成物10をマイクロニードル3に塗付し、揮発成分を除去した後に得られるコーティング層5の日本脳炎ワクチン抗原の濃度は、コーティング層5の全質量を基準として、0.01~30%w/wであることが好ましく、0.03~15%w/wであることがより好ましく、0.1~10%w/wであることがさらに好ましい。
 塩基性アミノ酸としては、特に限定されないが、リジン、ヒスチジン、アルギニン、オルニチン、カルニチン等から1種又は2種以上が挙げられ、そのフリー体が好ましい。中でも、上記酸の選択性がより広範になるという点から、アルギニンであることが好ましい。
 マイクロニードルコーティング用組成物10中の塩基性アミノ酸の濃度はマイクロニードルコーティング用組成物10の全質量を基準として、粘度及び日本脳炎ワクチン抗原の安定性の点から、20%w/w以上であることが好ましく、30%w/w以上であることがより好ましい。塩基性アミノ酸の濃度が20%w/w以上であることにより、マイクロニードルコーティング用組成物10中の日本脳炎ワクチン抗原の安定性を向上させることができる。また、塩基性アミノ酸の濃度が70%w/w以下であることにより、マイクロニードル3に塗付するときの取扱いが容易となる。また、マイクロニードルコーティング用組成物10をマイクロニードル3に塗付し、揮発成分を除去した後に得られるコーティング層5の塩基性アミノ酸の濃度は、コーティング層5の全質量を基準として、50~90%w/wであることが好ましく、60~80%w/wであることがより好ましい。
 マイクロニードルコーティング用組成物10中の日本脳炎ワクチン抗原の配合量(質量)に対する塩基性アミノ酸の配合量(質量)の比は、1.8~2400であることが好ましく、3.5~700であることがより好ましく、5~500であることがさらに好ましい。日本脳炎ワクチン抗原の濃度に対する塩基性アミノ酸の濃度の比を上記範囲とすることにより、日本脳炎ワクチン抗原の安定性を向上させることができる。
 マイクロニードルコーティング用組成物10中の酸としては、融点が40℃以上の酸であることが好ましい。このような酸を使用することにより、マイクロニードルコーティング用組成物10中に塩基性アミノ酸を高濃度(例えば、20%w/w以上)で存在させることができ、マイクロニードルコーティング用組成物10中の日本脳炎ワクチン抗原の安定性を向上させることができる。中でも、リン酸、乳酸、安息香酸、マレイン酸、クエン酸、酒石酸、コハク酸、アスコルビン酸及びアスパラギン酸からなる群より選ばれる少なくとも1種の酸であることが好ましく、リン酸、クエン酸及び酒石酸からなる群より選ばれる少なくとも1種の酸であることがより好ましい。マイクロニードルコーティング用組成物10中の酸の濃度は、マイクロニードルコーティング用組成物10の全質量を基準として、5~50%w/wであることが好ましく、10~30%w/wであることがより好ましい。また、マイクロニードルコーティング用組成物10をマイクロニードル3に塗付し、揮発成分を除去した後に得られるコーティング層5の酸の濃度は、コーティング層5の全質量を基準として、5~50%w/wであることが好ましく、10~30%w/wであることがより好ましい。
 マイクロニードルコーティング用組成物10は、上記日本脳炎ワクチン抗原、塩基性アミノ酸、酸の他、精製水、生理食塩水、リン酸緩衝液、クエン酸緩衝液、酢酸緩衝液、クエン酸-リン酸緩衝液、トリス‐塩酸緩衝液、グリシン-水酸化ナトリウム緩衝液等の緩衝液などの水系溶液を含有していてもよい。これら水系溶液の含有量は、マイクロニードルコーティング用組成物10の全質量を基準として、5~75質量%が好ましい。75質量%を超えると、コーティングの際に十分な粘度が得られない傾向があり、5質量%未満であると組成物を溶解させることが困難となる傾向がある。
 マイクロニードルコーティング用組成物10は、上記塩基性アミノ酸とともにリジン又はリジンの薬学的に許容できる塩を含有していてもよい。リジン又はリジンの薬学的に許容できる塩を含有することにより、さらに日本脳炎ワクチン抗原の安定性を向上させることができる。
 リジンの薬学的に許容できる塩としては、塩酸塩が好ましい。リジン塩酸塩の濃度は、マイクロニードルコーティング用組成物10の全質量を基準として、0.1~20%w/wであってもよい。20%w/wを超えると、リジン塩酸塩が溶解できない場合があり、0.1%w/w未満であると日本脳炎ワクチン抗原の安定性が不十分となる場合がある。また、マイクロニードルコーティング用組成物10をマイクロニードル3に塗付し、揮発成分を除去した後に得られるコーティング層5のリジン塩酸塩の濃度は、コーティング層5の全質量を基準として、0.1~20%w/wであってもよい。
 また、マイクロニードルコーティング用組成物10の任意成分として、さらに、高分子担体(粘度付与剤)を含有していてもよい。高分子担体としては、ポリエチレンオキサイド、ポリヒドロキシメチルセルロース、ヒドロキシプロピルセルロース、ポリヒドロキシプロピルメチルセルロース、ポリメチルセルロース、デキストラン、ポリエチレングリコール、ポリビニルアルコール、ポリビニルピロリドン、プルラン、カルメロースナトリウム、コンドロイチン硫酸、ヒアルロン酸、デキストラン、アラビアゴム等が挙げられる。なお、高分子担体として用いられるポリエチレングリコールの重量平均分子量は、600を超え、500000以下であることが好ましい。
 高分子担体としては、日本脳炎ワクチン抗原と相容性(均一に交わる性質)の高い担体が好ましく、ヒドロキシプロピルセルロース、デキストラン、ポリビニルアルコール、プルランなどが特に好ましい。
 マイクロニードルコーティング用組成物10の高分子担体の含有量は、マイクロニードルコーティング用組成物10の全質量を基準として、0.005~30質量%であり、好ましくは0.01~20質量%であり、より好ましくは0.05~10質量%である。また、この高分子担体は、液ダレすることのないようある程度の粘性が必要である場合があり、粘度として室温(25℃)で100~100000mPa・sであることが好ましい。より好ましい粘度は、500~60000mPa・sである。
 上記の他、マイクロニードルコーティング用組成物10には、必要に応じて溶解補助剤又は吸収促進剤として、炭酸プロピレン、クロタミトン、L-メントール、ハッカ油、リモネン、ジイソプロピルアジペート等や、薬効補助剤として、サリチル酸メチル、サリチル酸グリコール、L-メントール、チモール、ハッカ油、ノニル酸ワニリルアミド、トウガラシエキス等が添加されていてもよい。
 さらに、必要に応じて、安定化剤、抗酸化剤、乳化剤、界面活性剤、塩類等の化合物が添加されても良い。界面活性剤としては、非イオン界面活性剤、イオン界面活性剤(カチオン、アニオン、両性)のいずれでもよいが、安全性の面から通常医薬品基剤に用いられる非イオン界面活性剤が望ましい。これらの化合物としては、例えば、ショ糖脂肪酸エステルなどの糖アルコール脂肪酸エステル、プロピレングリコール脂肪酸エステル、ポリオキシエチレンソルビタン脂肪酸エステル、ポリオキシエチレングリセリン脂肪酸エステル、ポリエチレングリコール脂肪酸エステル、ポリオキシエチレンヒマシ油、ポリオキシエチレン硬化ヒマシ油が挙げられる。
 他の既知の製剤補助物質は、それらがマイクロニードルコーティング用組成物10の溶解性及び粘度向上の効果、並びに乾燥されたマイクロニードル3上に付着しているマイクロニードルコーティング用組成物10の性状及び物性に有害な影響を及ぼさない限り、マイクロニードルコーティング用組成物10に添加されていてもよい。
 マイクロニードルコーティング用組成物10には、塗付後のマイクロニードル3上で液ダレすることのないよう、ある程度の粘性が必要である。粘度は、100~45000mPa・s程度であり、マイクロニードルコーティング用組成物10の粘度がこの範囲にあることにより、マイクロニードルの材質に依存することなく、所望量のマイクロニードルコーティング用組成物10を一度に付着させることが可能となる。
 マイクロニードルコーティング用組成物10の25℃における粘度が45000mPa・s以下の場合、せん断応力の上昇をもたらし物質間の剥離への抵抗を大きくする。このため、dip(浸漬)法にて薬液を塗付する場合、マイクロニードルからの解離に抵抗する個体の性質(凝集性)が強くなり、より多くのコーティング組成物をマイクロニードル上に保持することが可能になる。一方、45000mPa・sを超えると、マイクロニードル上に付着しているコーティング組成物中の日本脳炎ワクチン抗原の含量が減少に転じ、経済的に好ましくない。コーティング組成物の粘度が100mPa・s以上の場合、凝集性が強いため、多くのコーティング組成物をマイクロニードル上に保持することが可能になる。このような特徴から、マイクロニードルコーティング用組成物10の25℃における粘度は、100~45000mPa・sであることが好ましく、300~35000mPa・sであることがより好ましく、500~30000mPa・sであることがさらに好ましく、600~15000mPa・sであることが特に好ましい。
 以下、本発明の実施例を示して、本発明をさらに具体的に説明するが、本発明はこれらの実施例に限定されるものではなく、本発明の技術的思想を逸脱しない範囲での種々の変更が可能である。
<アルギニンと酸の混合物の水に対する溶解性>
 アルギニンと表1~3に示す酸(N価)を、アルギニン:酸=N:1の比率で混合し、精製水を加え20%w/wのアルギニン-酸の希薄液を調製した後、凍結乾燥によって水分を蒸発させ凍結乾燥固体を作製した。この凍結乾燥固体に、アルギニンと酸の混合物(Arg+酸):水=7:3の比率で精製水を添加して、アルギニンと酸の混合物(Arg+酸)の高濃度溶液を調製した。表中、「溶解性」はその結果を示し、完全溶解し高濃度溶液の調製が可能であったものを「○」、一部溶解したものを「△」とした。

<配合比によるアルギニンと酸の混合物の水に対する溶解性と粘度特性>
 アルギニンと酸を表4、5に示すモル比で混合し、精製水を加え20%w/wのアルギニン-酸の希薄液を調製した後、凍結乾燥によって水分を蒸発させ凍結乾燥固体を作製した。この凍結乾燥固体に、アルギニンと酸の混合物(Arg+酸):水=7:3の比率で精製水を添加して、アルギニンと酸の混合物(Arg+酸)の高濃度溶液を調製した。表中、「溶解性」はその結果を示し、完全溶解し高濃度溶液の調製が可能であったものを「○」、一部溶解したものを「△」、ほとんど溶解しなかったものを「×」とした。また、表中、「粘度」は微量サンプル粘度計(VROC、RheoSense社製)を用いて測定したものであり、単位は「mPa・s」である。
 また、上記アルギニンと酸の混合物(Arg+酸、アルギニン:リン酸=2:1、アルギニン:クエン酸=3:1、アルギニン:酒石酸=2:1)の高濃度溶液をマイクロニードル(高さ約500μm、密度640本/cm、四角錐形状)に塗付し、顕微鏡(VH-8000、KEYENCE社製)を用いて、マイクロニードルの針先を観察した。観察の結果、コーティング層がマイクロニードルの先端部分に形成されていることが確認され、アルギニンと酸の混合物が針先塗付に適したものであることが判明した。

<日本脳炎ワクチン抗原の安定性評価>
 アルギニンと酸とを配合した日本脳炎ワクチン抗原塗付液をマイクロニードル上に塗付し、安定性を評価した。
 日本脳炎ワクチン抗原のPBS(リン酸緩衝生理食塩水)溶液を原薬として用いた。この原薬を限外ろ過濃縮し、濃縮液にアルギニンと酸の混合物の凍結乾燥固体(Arg+酸)、リジン塩酸塩(Lys・HCl)を加えて混合することで、又は濃縮時にアルギニン、酸、リジン塩酸塩を混合して調製し、凍結乾燥後、再溶解させることで日本脳炎ワクチン抗原塗付液を調製した。なお、各成分の配合比(単位:質量%)及びArg+酸のモル比は表6に示すとおりである。
 得られた日本脳炎ワクチン抗原塗付液をマイクロニードル(高さ約500μm、密度640本/cm、四角錐形状)に塗付して乾燥し、処方物A~Fを得た。表6の各成分における括弧内の数値は乾燥した後の配合比(単位:質量%)を示す。処方物A~Fをモイストキャッチ(共同印刷株式会社、商品名)内にファーマキープ(三菱ガス化学株式会社、商品名)とともに封入し、40℃で1か月間保存した。
 安定性の評価は、マイクロニードルより日本脳炎ワクチン抗原を抽出液(0.01質量%Tween80/1質量%トレハロース/PBS)にて抽出後、E抗原含量測定(ELISA法)及びたん白質定量(Lowry法)を行い、たん白質含量あたりのE抗原含量を比活性とし、初期値と40℃1ヶ月後の値を比較することによって行った。

 処方物A~Fにおいて、40℃で1か月保存した検体の比活性は初期検体のものとほぼ同じ値を示し、活性の低下は見られなかった。
<リジン塩酸塩の配合効果>
 各成分の配合比(単位:質量%)及びアルギニンと酸とのモル比を表7に示す70質量%配合した日本脳炎ワクチン抗原塗付液をマイクロニードル上に塗付し、リジン塩酸塩の配合効果についてL9直交表を用いて評価した。なお、表7の各成分における括弧内の数値は揮発成分を除去した後の配合比(単位:質量%)を示す。
 日本脳炎ワクチン抗原のPBS溶液を原薬とし、これを限外ろ過濃縮したものに、所定量の添加剤を加えて混合したものをマイクロニードルの塗付液とした。これをポリ乳酸製のマイクロニードル(高さ約500μm、密度640本/cm、四角錐形状)に相対湿度80~85%の環境下にて針先に塗付した。乾燥後、モイストキャッチ(共同印刷株式会社、商品名)内にファーマキープ(三菱ガス化学株式会社、商品名)とともに封入し、各温度条件での安定性を評価した。マイクロニードルより日本脳炎ワクチン抗原を抽出液(0.01%Tween80/1%トレハロース/PBS)にて抽出後、E抗原含量測定(ELISA法)及びたん白質定量(Lowry法)を行い、たん白質含量あたりのE抗原含量を比活性として評価した。50℃2週間保存検体の比活性の値について、リジン塩酸塩の配合が比活性に及ぼす影響度を最小二乗法により解析した。図4は、リジン塩酸塩の比活性に及ぼす影響度を示すグラフである。リジン塩酸塩について、2.5又は5.0%配合した場合に、50℃2週間保存検体の比活性への影響度が高かったことから、リジン塩酸塩について安定性向上効果が確認された。

<製剤用量による日本脳炎ワクチン抗原の安定性評価>
 アルギニンと酸とを配合した日本脳炎ワクチン抗原塗付液をマイクロニードル上に塗付し、4μg製剤及び0.2μg製剤の2用量の製剤の安定性を評価した。2用量の製剤の作製は日本脳炎ワクチン抗原塗付液の抗原濃度及び塗付高さを調整して行った。
 日本脳炎ワクチン抗原のPBS(リン酸緩衝生理食塩水)溶液を原薬として用いた。この原薬を限外ろ過濃縮し、濃縮時にアルギニン、リン酸、及びリジン塩酸塩を混合して調製し、凍結乾燥後、再溶解させることで日本脳炎ワクチン抗原塗付液を調製した。なお、各成分の配合比(単位:質量%)及びArg+酸のモル比は表8に示すとおりである。
 得られた日本脳炎ワクチン抗原塗付液をマイクロニードル(高さ約500μm、密度640本/cm、四角錐形状)に塗付し、塗付高さ125μm、想定含量4μgの処方物P、塗付高さ70μm、想定含量0.2μgの処方物Q、塗付高さ120μm、想定含量8μgの処方物R、塗付高さ60μm、想定含量2μgの処方物S、塗付高さ60μm、想定含量1μgの処方物T及び塗付高さ60μm、想定含量0.032μgの処方物Uを得た。表8の各成分における括弧内の数値は乾燥した後の配合比(単位:質量%)を示す。処方物P~Uをモイストキャッチ(共同印刷株式会社、商品名)内にファーマキープ(三菱ガス化学株式会社、商品名)とともに封入し、表9に示す各温度条件で処方物P~Tについては3か月間保存し、処方物Uは4か月保存した。
 安定性の評価は、マイクロニードルより日本脳炎ワクチン抗原を抽出液(0.01質量%Tween80/1質量%トレハロース/PBS)にて抽出後、E抗原含量測定(ELISA法)及びたん白質定量(Lowry法)を行い、たん白質含量あたりのE抗原含量を比活性とすることによって行った。なお、低用量の処方物Q、UはLowry法で測定したたん白質含量の代わりに、アミノ酸含量測定値と処方物中の配合比とから計算によって比活性を求めた。アミノ酸含量はOPA(オルトフタルアルデヒド)法により行った。

 処方物P~Tにおいて、10、25、40、50℃の各温度で3か月保存した検体は初期検体に比べて、たん白質含量及びE抗原含量の低下は見られなかった。処方物Uにおいても、10、25、40、50℃の各温度で4か月保存した検体は初期検体に比べて、たん白質含量及びE抗原含量の低下は見られなかった。また、比活性もほぼ同じ値を示し、活性の低下は見られなかった。
 1…マイクロニードルデバイス、2…基板、3…マイクロニードル、5…コーティング層、10…マイクロニードルコーティング用組成物、11…マスク版、12…ヘラ、13…開口部。

Claims (7)

  1.  日本脳炎ワクチン抗原、塩基性アミノ酸及び酸を含有し、
     前記塩基性アミノ酸1モルに対する前記酸のモル数が、前記酸の価数をNとしたときに、1/(N+1)を超え2未満である、マイクロニードルコーティング用組成物。
  2.  前記酸は、融点が40℃以上の酸である、請求項1に記載のマイクロニードルコーティング用組成物。
  3.  前記酸は、リン酸、乳酸、安息香酸、マレイン酸、クエン酸、酒石酸、コハク酸、アスコルビン酸及びアスパラギン酸からなる群より選ばれる少なくとも1種の酸である、請求項1又は2に記載のマイクロニードルコーティング用組成物。
  4.  前記塩基性アミノ酸は、アルギニンである、請求項1~3のいずれか一項に記載のマイクロニードルコーティング用組成物。
  5.  リジン又はリジン塩酸塩を含有する、請求項1~4のいずれか一項に記載のマイクロニードルコーティング用組成物。
  6.  マイクロニードル上に、請求項1~5のいずれか一項に記載のマイクロニードルコーティング用組成物から形成されるコーティング層を備える、マイクロニードルデバイス。
  7.  前記コーティング層は、前記マイクロニードルの先端部分に形成されている、請求項6に記載のマイクロニードルデバイス。
