Description

モノクローナルIgA抗体の製造方法
 本発明は、モノクローナルIgA抗体、それを含む経口又は経腸組成物、及びそれを含む医薬組成物に関する。
 腸管内は、生体にとっては言わば『体外』に当たるため、非常に多種、且つ多数の腸内常在細菌、ウイルス、食物由来等の抗原に、常に晒された状態となっている。この様な腸内常在細菌は、腸内細菌叢(以後、本明細書において、これを「腸内細菌フローラ」と呼ぶ場合がある。)を形成している。
 この腸内細菌叢は、腸上皮細胞等から構成される粘膜面を介して、生体に対して様々な機能を果たしていることが明らかになっており、腸内細菌が存在しないと腸管免疫系が正常に発達しないことも分かってきた(非特許文献1~3)。
 もし腸内細菌叢が変化して宿主との共生関係が崩れると、腸管免疫系が過剰に刺激されてその恒常性が破綻し、延いては炎症性腸疾患、大腸がん、喘息、アレルギー、肥満等といった多くの疾患が誘発されてしまう。この様なことから、腸管免疫系は病原体等を排除するだけではなく、免疫系全体の恒常性の維持に重要な役割を担っていることが知られている(非特許文献4~7)。
 消化管の粘膜組織表面は、常在細菌、病原性微生物、アレルゲン等といった抗原の生体内への侵入経路の一つであり、これら多数の外界からの病原性微生物の侵入部位として絶えず攻撃を受け、また様々な抗原に曝されている。これらの病原性微生物をはじめとする外来性抗原に対する認識及び/又は応答システムが、粘膜免疫システムと言われるものである。消化管の粘膜組織表面の直下には、多数の粘膜免疫システムを担う細胞が存在し、これらの細胞が、病原性微生物、食物抗原、アレルゲン外来性の異種抗原等に対する免疫応答を行う、ダイナミックな免疫機構を構成している。
 粘膜免疫システムは、腸上皮細胞からなる粘膜面を介して取り込まれた外来性の異種抗原に対して、腸管由来のIgA抗体を中心とした独自の免疫システムを構成している。腸管由来のIgA抗体はパイエル板、腸管膜リンパ節、孤立リンパ濾胞等のようないわゆる免疫反応の場としての特別な組織内でT細胞依存性に産生されるだけでなく、腸管粘膜固有層に散在する抗体産生細胞によっても大量に産生される(非特許文献8、9)。
 腸管由来のIgA抗体の機能の1つは病原菌を排除する機能である。管腔側へ分泌された腸管由来のIgA抗体は腸管腔内の病原菌やその毒素と結合し中和する。あるいは腸上皮細胞内、粘膜固有層等に侵入してきた病原菌、毒素等と結合し中和すると共に、腸管由来のIgA抗体そのものが管腔側へ分泌されるときにそれと共に斯かる病原菌も排除する。腸管由来のIgA抗体の機能で特徴的な点は、一般的な抗体が関与する全身免疫系の反応と異なり、炎症反応を起こさないことである(非特許文献10~12)。
 もう一つの腸管由来のIgA抗体の機能は、腸内常在細菌と宿主の共生関係の維持である。腸管由来のIgA抗体は病原菌だけでなく常在細菌も認識してそれらと結合し、常在細菌が粘膜内へ過剰に侵入しないように防いでいる。しかも単に常在細菌を排除するのではなく、常在細菌と結合した腸管由来のIgA抗体は上皮細胞上のムチン層に担持され、常在細菌と腸管由来のIgA抗体によるバイオフィルムを形成して病原菌の上皮細胞への接触、侵入等を防いでいると考えられている(非特許文献13、14)。
 このような腸管粘膜免疫システムのうち、特に体液性免疫システムの構築において、抗体の体細胞突然変異とクラススイッチが、重要な役割を果たしていることが知られている。体細胞突然変異は、抗体を産生するB細胞の免疫グロブリン遺伝子を多様化して、高親和性抗体を産生するための、生体内に備わっている機構である。一方、抗体のクラススイッチは、抗体の可変領域は変化させずに、H鎖定常領域の構造を変化させる現象、すなわち、生体内で選択されたIgM抗体を産生する細胞において、そのIgM抗体と同じ可変領域を有するIgG抗体、IgE抗体、IgA抗体等といった、他のクラスの免疫グロブリンを産生するようにクラスを変更する現象である。これら2つの現象が組み合わさることにより、多様な抗原結合部位を有する多種類の抗体が産生され、かつ各抗原結合部位のそれぞれについて各クラスの抗体が産生されることになり、身体の免疫機能を適切に調整することができる。
 体細胞突然変異とクラススイッチには、ともにactivation-induced cytidine deaminase(AID)タンパク質が必要であることが知られている。AIDタンパク質のN末端側が体細胞突然変異に関与しており、C末端側がクラススイッチに関与していることまで知られている(非特許文献15、16)。体細胞突然変異では、抗体遺伝子に高い確率でAIDによるDNA切断が生じ、それが引き金になって抗体遺伝子に変異が導入される(非特許文献17)。
 また、このようなAIDタンパク質のN末端側の23位のグリシンをセリンに置換し変異を導入したAID G23Sを発現するマウス(AID G23Sマウス)では、クラススイッチは生じるために各クラスの抗体が十分に産生されるものの、体細胞突然変異が起こらず、抗体の抗原結合性が低下していることが明らかになっている(非特許文献18)。
 炎症性腸疾患は、治療に難渋する事が多く難病に指定されている。この疾患は、宿主免疫系と腸内細菌のバランスが崩れること、即ち腸内細菌の異常増殖や、腸内フローラの病的変化により発症する疾患の一つに挙げられるものである。この疾患の治療には、臨床現場においてステロイドや抗TNF抗体等の免疫抑制剤の全身投与が行われている。しかしながら、これらの薬剤は長期投与が必要で、その副作用が大きな問題となっている。そのため、副作用が少なく、効率的に炎症性腸疾患等といった腸内疾患の治療又は予防に有効な医薬品の開発が望まれている。

 本発明の課題は、生体における腸内環境の改善若しくは最適化、腸内腐敗の抑制、又は腸内細菌の異常増殖及び/又は腸内細菌叢の病的変化を抑制する効果を発揮する成分を提供することである。また、腸内疾患の治療に好適に用いられる有効成分を提供することも、本発明の課題である。
 本発明者らは、上述の課題を解決すべく、鋭意検討したところ、特定のアミノ酸配列を有するタンパク質の特定の部位に特異的に結合するモノクローナルIgA抗体が、腸内免疫システムにおける過剰な応答を抑制し、大腸の粘膜固有層における腺管の減少から復帰させる作用を有することを見出した。また、このようなモノクローナルIgA抗体の作用は、個体に対して経口投与することによって確認された。
 本願発明は、この様な知見に基づいて完成したものであり、以下に示す広い態様の発明を包含するものである。

〔モノクローナルIgA抗体〕
 本発明に係るモノクローナルIgA抗体は、セリンヒドロキシメチルトランスフェラーゼの11~333番目のアミノ酸配列に特異的に結合する。好ましくは、11番目又は25番目の何れかから28番目、37番目、44番目、250番目、333番目の何れかのアミノ酸配列(11~250、11~44、11~37、11~28、25~333、25~250、25~44、25~37、又は25~28番目)であり、さらに好ましくは25~44番目のアミノ酸配列であり、最も好ましくは25~28番目のアミノ酸配列である。
 セリンヒドロキシメチルトランスフェラーゼとは、45~50kDa程度の分子量を有し、その等電点は4.3~4.7程度である。具体的には、例えば大腸菌由来であれば、NCBIのウェブサイトに収載される、ACCESSION No.J01620 J01621;VERSION J01620.1 GI:146216に示されるアミノ酸配列等を有するタンパク質である。さらに具体的には、配列番号74に示すアミノ酸配列を有するタンパク質である。このタンパク質は、L-セリンをグリシンに、そして同時にテトラヒドロ葉酸を5,10-メチレンテトラヒドロ葉酸に可逆的に変換する機能を有する酵素であり、その立体構造は大腸菌から哺乳動物まで非常に保存された酵素である。

 また、表1に示すように、上記配列番号74に示すアミノ酸配列を基にしたアライメントから、大腸菌由来のセリンヒドロキシメチルトランスフェラーゼの25~26番目(RQ)及び31~44番目、中でも31~37番目(ELIASEN)に相当するアミノ酸配列が、上記の多数の種間において非常によく保存されている。これらのアミノ酸配列は、下記実施例に示すように、本発明に係るモノクローナルIgA抗体が結合する大腸菌由来のセリンヒドロキシメチルトランスフェラーゼのエピトープと考え得る最少単位のアミノ酸配列を含むものである。したがって、配列番号74に示すアミノ酸配列の25~26番目及び31~44番目のアミノ酸を含む領域が、本発明に係るモノクローナルIgA抗体のエピトープとして25~28番目のアミノ酸配列と共に含まれると考えられる。
 本発明に係るモノクローナルIgA抗体が結合するセリンヒドロキシメチルトランスフェラーゼの由来は、大腸菌以外にも、Pseudomonas fuluva等のPseudomonas属菌;Staphylococcus aureus等のブドウ球菌;Eubacterium rectale、Haemophilus influenzae、Klebsiella pneumoniae、Legionell pneumophilia、Salmonella paratyphi A、Salmonella typhimurium、Shigella flexneri、Yersinia pestis bv. Antique、Burkholderia mallei等が挙げられ特に限定はされない。
 一方で本発明に係るモノクローナルIgA抗体は、上記以外に由来するセリンヒドロキシメチルトランスフェラーゼには結合しないか、結合したとしてもすぐに解離してしまう傾向となる。この様な由来は特に限定はされないが、例えばLactobacillus casei、Bifidobacterium bifidum等の乳酸菌、Blautia coccoides、Coprococcus eutactus、Megamonas hypermegale、Bordetella parapertussis;Saccharomyces cerevisiae、Schizosaccharomyces pompe、Arabidopsis thaliana、ウサギ、マウス、ヒト等の真核生物等に由来するセリンヒドロキシメチルトランスフェラーゼが挙げられる。これらの結合の有無やその傾向については、培養した各種菌体のライセートを、本発明に係るモノクローナルIgA抗体を用いたウエスタンブロットに供して確認する。
 上述のように、本発明に係るモノクローナルIgA抗体が特異的に結合するセリンヒドロキシメチルトランスフェラーゼ中の特定の部位(エピトープ)は、例えば本願明細書の実施例に示すように、セリンヒドロキシメチルトランスフェラーゼのトランケートミュータントを作製し、これと本発明に係るモノクローナルIgAが結合するか否かを、ウエスタンブロッティング法等といった公知の免疫化学的手段で確認すればよい。
 そのほかに、上述のような本発明に係るモノクローナルIgA抗体が結合することが確認できる、例えば大腸菌に由来する野生型のセリンヒドロキシメチルトランスフェラーゼ中の一部のアミノ酸配列を、上述のような本発明に係るモノクローナルIgA抗体が結合しないか、結合してもすぐに解離してしまう、例えば乳酸菌に由来するセリンヒドロキシメチルトランスフェラーゼの上記一部のアミノ酸配列に対応するアミノ酸配列で置換した変異体を作成し、この変異体が本発明に係るモノクローナルIgA抗体に結合できないか、又は野生型のセリンヒドロキシメチルトランスフェラーゼよりも結合力が減少した場合に、上記一部のアミノ酸配列を本発明に係るモノクローナルIgA抗体が特異的に結合するセリンヒドロキシメチルトランスフェラーゼ中のアミノ酸配列と決定すればよい。なお、結合するかどうか、結合力の低下等確認する手段は、上述のような公知の免疫化学的手段を適宜採用すればよい。
 本明細書にて使用する用語『モノクローナル』とは、実質的に均一な抗体の集団から得られる抗体等の特徴を示す修飾語である。このような抗体の集団に含まれる個々の抗体は、微量に存在し得る、可能な天然に生じる突然変異を除いて同一である。
 本発明に係るモノクローナルIgA抗体とは、免疫グロブリンの一種であり、重鎖可変領域及び/又は軽鎖可変領域を有する態様が挙げられる。中でも、これらのそれぞれに、更に定常領域を含む、重鎖及び/又は軽鎖を有する態様がより好ましい。より好ましくは、重鎖および軽鎖を有する態様である。
 このような重鎖及び軽鎖は、主としてポリペプチドで構成され、それぞれ抗原を認識する可変領域と呼ばれる部位(一般的に、それぞれ重鎖可変領域及び軽鎖可変領域と呼ばれる。)を含む。
 上述の可変領域の中に、より抗原を認識する部位として特定される、それぞれCDR1~3と呼ばれる部位をアミノ末端から順に有する。なお、これらのCDR1~3は、より詳細に重鎖CDR1、重鎖CDR2、重鎖CDR3、軽鎖CDR1、軽鎖CDR2、軽鎖CDR3等とも呼ばれる。また、重鎖及び軽鎖のCDR1~3以外の領域は、それぞれアミノ末端から順に、重鎖FR1~4及び軽鎖FR1~4と呼ばれる。
 本発明に係るモノクローナルIgA抗体における可変領域には、上述のような重鎖可変領域及び軽鎖可変領域にそれぞれCDR1~3の全てを必ずしも含んでいる必要はなく、CDR1~3のうちの少なくとも1つのCDRを含んでいればよい。中でもCDR3を含んでいることが好ましい。
 このような、重鎖CDR1~3及び軽鎖CDR1~3を構成するポリペプチドのアミノ酸配列は、特に限定はされないが、例えば、
重鎖CDR1としては配列番号3、13、23、33、又は43の何れかに示すアミノ酸配列等;
重鎖CDR2としては配列番号4、14、24、34、又は44の何れかに示すアミノ酸配列等;
重鎖CDR3としては配列番号5、15、25、35、又は45の何れかに示すアミノ酸配列等;
軽鎖CDR1としては配列番号8、18、28、38、又は48の何れかに示すアミノ酸配列等;
軽鎖CDR2としては配列番号9、19、29、39、又は49の何れかに示すアミノ酸配列等;
軽鎖CDR3としては配列番号10、20、30、40、又は50の何れかに示すアミノ酸配列等が挙げられる。
 本発明に係るモノクローナルIgA抗体の重鎖可変領域は、特に限定はされないが、例えばCDR1~3を含む態様の重鎖可変領域であれば、上述の
(1)配列番号3、13、23、33、又は43の何れかに示すアミノ酸配列からなる、重鎖CDR1;
(2)配列番号4、14、24、34、又は44の何れかに示すアミノ酸配列からなる、重鎖CDR2;
(3)配列番号5、15、25、35、又は45の何れかに示すアミノ酸配列からなる、重鎖CDR3
が、アミノ末端から順に含まれている重鎖可変領域が挙げられる。
 また、例えばCDR1~3を含む態様の軽鎖可変領域も特に限定はされないが、例えば上述の
(I)配列番号8、18、28、38、又は48の何れかに示すアミノ酸配列からなる、軽鎖CDR1;
(II)配列番号9、19、29、39、又は49の何れかに示すアミノ酸配列からなる、軽鎖CDR2;
(III)配列番号10、20、30、40、又は50の何れかに示すアミノ酸配列からなる、軽鎖CDR3
が、アミノ末端から順に含まれている軽鎖可変領域が挙げられる。
 より好ましいCDR1~3を含む態様の重鎖可変領域としては、
配列番号3に示すアミノ酸配列からなる重鎖CDR1、配列番号4に示すアミノ酸配列からなる重鎖CDR2、及び配列番号5に示すアミノ酸配列からなる重鎖CDR3をアミノ末端から順に含む重鎖可変領域;
配列番号13に示すアミノ酸配列からなる重鎖CDR1、配列番号14に示すアミノ酸配列からなる重鎖CDR2、及び配列番号15に示すアミノ酸配列からなる重鎖CDR3をアミノ末端から順に含む重鎖可変領域;

 さらに好ましくは、配列番号3に示すアミノ酸配列からなる重鎖CDR1、配列番号4に示すアミノ酸配列からなる重鎖CDR2、及び配列番号5に示すアミノ酸配列からなる重鎖CDR3をアミノ末端から順に含む重鎖可変領域である。
 また、より好ましい例えばCDR1~3を含む態様の軽鎖可変領域としては、配列番号8に示すアミノ酸配列からなる軽鎖CDR1、配列番号9に示すアミノ酸配列からなる重鎖CDR2、及び配列番号10に示すアミノ酸配列からなる軽鎖CDR3をアミノN末端から順に含む軽鎖可変領域;
配列番号18に示すアミノ酸配列からなる軽鎖CDR1、配列番号19に示すアミノ酸配列からなる軽鎖CDR2、及び配列番号20に示すアミノ酸配列からなる軽鎖CDR3をアミノ末端から順に含む軽鎖可変領域;

 さらに好ましくは、配列番号8に示すアミノ酸配列からなる軽鎖CDR1、配列番号9に示すアミノ酸配列からなる重鎖CDR2、及び配列番号10に示すアミノ酸配列からなる軽鎖CDR3をアミノ末端から順に含む軽鎖可変領域である。
 上述の重鎖可変領域の中でもさらに好ましいのは、配列番号2、12、22、32、又は42に示すアミノ酸配列からなる重鎖可変領域であり、最も好ましくは配列番号2に示すアミノ酸配列からなる重鎖可変領域である。
 また、上述の軽鎖可変領域の中でもさらに好ましいのは、配列番号7、17、27、37、又は47に示すアミノ酸配列からなる軽鎖可変領域であり、最も好ましくは配列番号7に示すアミノ酸配列からなる軽鎖可変領域である。
 本発明に係るモノクローナルIgA抗体は、上述のような重鎖可変領域及び/又は軽鎖可変領域を適宜組み合わせた構成を有する態様であってもよい。このような他の態様のモノクローナルIgA抗体は、例えばF(ab’)2、Fab、Fv、scFv、scFv-Fc、minibody等と呼ばれる構成を有する。
 本発明に係るモノクローナルIgA抗体の中でも、さらに好ましい態様として、上述のような重鎖及び/又は軽鎖可変領域にさらに定常領域を含んだ重鎖及び/又は軽鎖を含む態様のモノクローナルIgA抗体が挙げられる。
 このようなモノクローナルIgAは、特に限定されるものでは無いが、例えば、配列番号1、11、21、31、又は41に示すアミノ酸配列を含む重鎖及び/又は配列番号6、16、26、36、又は46に示すアミノ酸配列を含む軽鎖を有するモノクローナルIgA抗体が挙げられる。
 より好ましくは、配列番号1、11、21、31、又は41に示すアミノ酸配列を含む重鎖及び配列番号6、16、26、36、又は46に示すアミノ酸配列を含む軽鎖を有するモノクローナルIgA抗体である。
 さらに好ましい態様として、配列番号1に示すアミノ酸配列を含む重鎖及び配列番号6に示すアミノ酸配列を含む軽鎖を有するモノクローナルIgA抗体;
配列番号11に示すアミノ酸配列を含む重鎖及び配列番号16に示すアミノ酸配列を含む軽鎖を有するモノクローナルIgA抗体;
配列番号21に示すアミノ酸配列を含む重鎖及び配列番号26に示すアミノ酸配列を含む軽鎖を有するモノクローナルIgA抗体;
配列番号31に示すアミノ酸配列を含む重鎖及び配列番号36に示すアミノ酸配列を含む軽鎖を有するモノクローナルIgA抗体;
配列番号41に示すアミノ酸配列を含む重鎖及び配列番号46に示すアミノ酸配列を含む軽鎖を有するモノクローナルIgA抗体が挙げられ、より好ましくは、
配列番号1に示すアミノ酸配列を含む重鎖及び配列番号6に示すアミノ酸配列を含む軽鎖を有するモノクローナルIgA抗体である。
 上記のモノクローナルIgA抗体は、単量体であっても多量体であってもよい。好ましくは2量体である。なお、上記のモノクローナル抗体が多量体である場合、後述するJ鎖を含んでいる。
 J鎖とは、抗原認識部位を有さないアミノ酸配列を含むペプチドであり、上記の重鎖とも軽鎖とも異なるアミノ酸配列を有するペプチドである。当該アミノ酸配列として、具体的には、マウス由来であれば、National Center of Biotechnology Information(NCBI)ウェブサイト(http://www.ncbi.nlm.nih.gov/)に収載される、ACCESSION No.AAA38673;VERSION AAA38673.1 GI:196379に示されるアミノ酸配列等が挙げられ、ヒト由来であれば、同じくNCBIウェブサイトに収載される、ACCESSION No.NP_653247;VERSION NP_653247.1 GI:21489959に示されるアミノ酸配列等が挙げられる。このようなJ鎖は、上記のモノクローナルIgA抗体モノマーの重鎖と各々ジスルフィド結合している。
 なお、上記のモノクローナルIgA抗体を規定するアミノ酸配列には、本明細書にて説明するモノクローナルIgA抗体の機能、効果等を減衰させない範囲に限り、適宜変異が施されていてもよい。具体的な変異導入の数は、共に特に限定はされないが、通常は変異前のアミノ酸配列と85%以上、好ましくは90%以上、より好ましくは95%以上、最も好ましくは99%以上の同一性を有する変異体となるような変異導入数とすればよい。
 本明細書にて使用する用語、『同一性』とは、2以上の対比可能なアミノ酸配列又は塩基配列の、お互いに対する同一のアミノ酸配列又は塩基配列の程度をいう。従って、ある2つのアミノ酸配列又は塩基配列の同一性が高いほど、それらの配列の同一性または類似性は高い。アミノ酸配列又は塩基配列の同一性のレベルは、通常は、配列分析用ツールであるFASTAを用い、デフォルトパラメーターを用いて決定される。
 若しくは、KarlinおよびAltschulによるアルゴリズムBLAST(例えば、Karlin S,Altschul SF.Proc.Natl Acad Sci USA.87:2264-2268(1990)、Karlin S,Altschul SF.Natl Acad Sci USA.90:5873-7(1993)等)を用いて決定できる。このようなBLASTのアルゴリズムに基づいたBLASTNやBLASTXと呼ばれるプログラムが開発されている(例えば、Altschul SF,GishW,Miller W,Myers EW,Lipman DJ.J Mol Biol.215:403-10(1990)等)。これらの解析方法の具体的な手法は公知であり、NCBIのウェブサイトを参照すればよい。
 上述の変異導入とは、置換、欠失、挿入等である。具体的な変異導入については、公知の方法を採用することができ、特に限定はされないが、例えば置換であれば保存的な置換技術を採用すればよい。
 本明細書にて使用する用語『保存的な置換技術』とは、アミノ酸残基が類似の側鎖を有するアミノ酸残基に置換される技術を意味する。
 例えば、リジン、アルギニン、ヒスチジンといった塩基性側鎖を有するアミノ酸残基同士で置換されることが、保存的な置換技術にあたる。その他、アスパラギン酸、グルタミン酸といった酸性側鎖を有するアミノ酸残基;グリシン、アスパラギン、グルタミン、セリン、スレオニン、チロシン、システインといった非帯電性極性側鎖を有するアミノ酸残基;アラニン、バリン、ロイシン、イソロイシン、プロリン、フェニルアラニン、メチオニン、トリプトファンといった非極性側鎖を有するアミノ酸残基;スレオニン、バリン、イソロイシンといったβ-分枝側鎖を有するアミノ酸残基、チロシン、フェニルアラニン、トリプトファン、ヒスチジンといった芳香族側鎖を有するアミノ酸残基同士での置換も同様に、保存的な置換技術にあたる。
 上記のモノクローナルIgA抗体のサブクラスは特に限定はされず、例えばIgA1であっても、IgA2であってもよい。
 上記のモノクローナルIgA抗体は分泌因子を含んでいてもよい。分泌因子とは、上述のJ鎖と同様に抗原認識部位を有さないアミノ酸配列を含むペプチドであり、通常は糖鎖が修飾された、上記の重鎖とも軽鎖とも異なるアミノ酸配列を有するペプチドである。
 当該アミノ酸配列として、具体的には、マウス由来であれば、NCBIのウェブサイトに収載される、ACCESSION No.O70570;VERSION O70570.1 GI:6225856に示されるポリイムノグロブリンレセプターのアミノ酸配列等のうちの、細胞外ドメインに相当するアミノ酸配列等が挙げられる。上記の例であれば、19~638番目のアミノ酸配列が分泌因子に相当する。
 ヒト由来であれば、同じくNCBIウェブサイトに収載される、ACCESSION No.P01833;VERSION P01833.4 GI:150421625に示されるポリイムノグロブリンレセプターのアミノ酸配列のうちの、細胞外ドメインに相当するアミノ酸配列等が挙げられる。上記の例であれば、19~645番目のアミノ酸配列が分泌因子に相当する。
 このような分泌因子を含むモノクローナルIgA抗体を製造するために、上記工程1にて得られるハイブリドーマに、更に上述したポリイムノグロブリンレセプターのSecretory Component領域をコードする塩基配列を有する核酸を導入する工程が含まれていてもよい。具体的な遺伝子導入方法は、公知の方法を適宜改変すればよく、特に限定はされない。
 上記のモノクローナルIgA抗体は同一種に由来するアミノ酸配列を有していても異種に由来するアミノ酸を有していてもよい。同一種由来の場合、例えば、ヒト由来、マウス由来、ラット由来、ハムスター由来、ウサギ由来、ヤギ由来、ロバ由来、ブタ由来、ウシ由来、ウマ由来、ニワトリ由来、サル由来、チンパンジー由来等が挙げられる。
 異種由来の場合も、特に限定はされないが、例えばヒト、マウス、ラット、ハムスター、ウサギ、ヤギ、ロバ、ブタ、ウシ、ウマ、ニワトリ、サル、チンパンジー等の何れか2つ以上に由来するモノクローナルIgA抗体が挙げられる。
 さらに具体的な異種に由来するアミノ酸配列を有するモノクローナルIgA抗体の1つの態様として、ヒトに由来するアミノ酸配列及びヒト以外の動物種に由来するアミノ酸配列を有するモノクローナルIgA抗体であって、重鎖及び軽鎖可変領域の全てがヒト以外の動物種に由来するアミノ酸配列であり、それ以外の領域がヒトに由来するアミノ酸配列を有する態様のモノクローナルIgA抗体が挙げられる。これを特にキメラ抗体と呼ぶことがある。
 本発明に係るモノクローナルIgA抗体は、少なくとも2種類の腸内細菌の成育を阻害する。
 本明細書において用いる用語『腸内細菌』とは、生体内の腸内に常在する細菌であれば特に限定はされない。この様な腸内細菌としては、生体内で腸内細菌叢を形成し、腸内免疫系の恒常性の維持に関与する細菌が挙げられる。
 具体的な腸内細菌としては、特に限定されるものでは無いが、例えば、
Prevotella属に属する細菌、
Bacteroides属に属する細菌、
Megamonas属に属する細菌、
Bifidobacterium属に属する細菌、
Faecalibacterium属に属する細菌、
Coprococcus属に属する細菌、
Ruminococcus属に属する細菌、
Blautia属に属する細菌、
Eubacterium属に属する細菌、
Roseburia属に属する細菌、
Lactobacillus属に属する細菌、
Clostridium属に属する細菌、
Escherichia属に属する細菌、
Staphylococcus属に属する細菌、
Enterococcus属に属する細菌、
Pseudomonas属に属する細菌、
Enterorhabdus属に属する細菌
等が挙げられる。
 これらの細菌の中でも、
Prevotella melaninogenica、
Bacteroidetes vulgatus、
Megamonas funiformis、
Megamonas hypermegale、
Bifidobacterium bifidum、
Faecalibacterium prausnitzii、
Coprococcus eutactus、
Ruminococcus obeum、
Blautia productus、
Blautia coccoides、
Eubacterium rectale、
Roseburia intestinalis、
Lactobacillus murinus、
Lactobacillus casei、
Clostridium difficile、
Escherichia coli、
Staphylococcus aureus、
Enterococcus faecalis、
Pseudomonas fulva、及び
Enterorhabdus mucosicola
等の細菌が挙げられる。
 また、本発明に係るモノクローナル抗体は、下記実施例に示すように大腸の粘膜固有層における腺管の減少または又は委縮を回復させる作用を有する。大腸の粘膜固有層における腺管の減少は、腸内疾患、特に腸内細菌の異常増殖及び/又は腸内細菌叢の病的変化により発症する腸内疾患に見られる病理組織学所見である。また、一般的に潰瘍性大腸炎の慢性病変としてよく見られる病変でもある。
 従って、本発明に係るモノクローナルIgA抗体は、腸内細菌の異常増殖及び/又は腸内細菌叢の病的変化を抑制する効果を発揮することが期待され、さらに炎症性腸疾患、潰瘍性大腸炎、クローン病、アレルギー、喘息、肥満、自己免疫疾患等といった、腸内細菌の異常増殖及び/又は腸内細菌叢の病的変化により発症する、例えば腸内疾患の治療に効果を発揮することが期待される。
 上述の腸内細菌の異常増殖とは、特に限定はされないが、例えば腸管内に生息する細菌数が異常に増加する状態を表す。
 上述の腸内細菌叢の病的変化とは、特に限定はされないが、例えば、腸内細菌叢を構成する腸内細菌の種類と、それぞれの腸内細菌の割合が変化することで宿主との間の共生関係の恒常性保持に異常を来す状態を表す。
 なお、このような腸内疾患の異常増殖及び腸内細菌叢の病的変化とは、必ずしも上述した腸内疾患に罹患した状態に限定されるものでは無く、腸内疾患に罹患する要因を誘起する状態を意味するものであってもよい。
 本明細書にて使用する用語『治療』とは、所望の薬理学的効果及び/又は生理学的効果を得ることを意味する。この効果は、疾病及び/又は疾病に起因する悪影響(病態、症状等)を、部分的又は完全に治癒することを含む。また、上記効果には、疾病及び/又は疾病に起因する悪影響(病態、症状等)の進行を阻止又は遅延する効果、病態や症状を緩和する(疾病、症状等の後退、又は進行の逆転を引き起こす)効果、再発を阻止する効果等が含まれる。
 また、上記効果には、疾病及び/又は疾病に起因する悪影響(病態、症状等)の素因を持ち得るが、まだ持っていると診断されていない個体において、疾病及び/又は疾病に起因する悪影響(病態、症状等)が起こることを部分的又は完全に防止する効果が含まれる。従って、『治療』なる用語には、『緩解』、『再発防止』、『予防』等の意味も含まれる。
 上述のように、本発明に係るモノクローナルIgA抗体は、腸内疾患の治療に用いる事によって、優れた効果を発揮することが期待される。この様な効果は、斯かるモノクローナル抗体が経口又は経腸的に個体に投与されることによって発揮される効果である。このことから、本発明に係るモノクローナルIgA抗体は、経口又は経腸組成物、医薬組成物の有効成分として有用である。
〔核酸〕
 本発明に係る核酸は、本発明に係るモノクローナルIgA抗体をコードする核酸である。より具体的には、配列番号1~50のいずれか1つに示すアミノ酸配列をコードする塩基配列を有する核酸である。当該核酸は、リボヌクレオチドであっても、デオキシヌクレオチドであってもよい。また、当該核酸は1本鎖の形態であっても、2本鎖の形態であってもよく特に限定はされない。
 上述のアミノ酸配列をコードする塩基配列は、1つのアミノ酸配列に対して1種類の塩基配列からなる核酸には限定されず、核酸の使用目的に応じて当該アミノ酸をコードする各種コドンを適宜選択して決定されたものであればよい。使用目的としては、例えば、当該塩基配列を用いて、本発明に係るモノクローナルIgA抗体を発現させ、それを製造すること等が挙げられる。特に製造時に採用する宿主細胞の種類に対応したコドン頻度を考慮して各種コドンを選択すればよい。
 このような塩基配列は、例えば公知のプログラムを採用すれば、インシリコにて容易に決定することができる。また、本発明に係る核酸は、配列番号1~50に対して、上記の〔本発明のモノクローナルIgA抗体〕にて詳述した変異が施されたアミノ酸配列をコードする塩基配列を有するものであってもよい。
 このような具体的な塩基配列については、特に限定はされないが、例えば配列番号51~60に示す塩基配列が挙げられる。これらの塩基配列と〔本発明のモノクローナルIgA抗体〕にて詳述した配列番号1~50に示すアミノ酸配列との対応については下記の表2に説明できる。

〔モノクローナルIgA抗体の製造方法〕
 本発明に係るモノクローナルIgA抗体の製造方法は、上述の〔モノクローナルIgA〕の製造方法であって、下記の工程1及び2を含むことを特徴とする。
(工程1)
 腸管粘膜固有層から採取されたB細胞と、それ以外の種類の細胞を混合して融合させ、ハイブリドーマを作製する工程。
(工程2)
 上記工程1にて作製したハイブリドーマを培養し、少なくとも2種類の腸内細菌と結合するIgA抗体を産生する細胞を決定し、該細胞から、前記IgA抗体を回収する工程。
 本発明に係るモノクローナルIgA抗体の製造方法には、工程2の後に、後述するような重鎖可変領域及び/又は軽鎖可変領域を必要に応じて、これらを適宜組み合わせた構成とするために、例えば、IgAに特異的なプロテアーゼ等によって処理する工程、ジスルフィド結合等の化学的結合が可能となるような、官能基を導入する工程、並びにそれに次いで前記官能基を介して前記化学的結合を形成させる工程等が含まれていてもよい。
 本発明のモノクローナルIgA抗体の製造方法における上記工程1及び工程2について、以下に詳述する。
(工程1について)
 本発明に係るモノクローナルIgA抗体の製造方法における工程1は、腸管粘膜固有層から採取されたB細胞と、それ以外の種類の細胞を混合して融合させ、ハイブリドーマを作製する工程である。
 工程1において用いるB細胞とは腸管粘膜固有層に局在するB細胞である。また、B細胞とはIgA抗体を産生する機能を発揮する細胞であればよく、必ずしもIgA抗体をコードするDNA又はmRNA含み、当該mRNAが翻訳されてIgA抗体を産生するものには限定されない。例えば、当該B細胞内でIgA抗体とは異なるサブタイプのIgG抗体、IgM抗体等といったイムノグロブリンがクラススイッチしてIgA抗体となり、当該細胞内にてIgA抗体を産生する細胞も上記B細胞に含まれる。
 腸管粘膜固有層(Lamina Propria)とは、特に限定はされないが、例えば食道、胃、小腸(十二指腸、空腸、回腸等を含む。)、大腸(盲腸、結腸、直腸等を含む。)等に存在する粘膜を構成する層の1つであって、上皮細胞を含む層と粘膜基板(Muscularis mucosae)との間に位置する層であればよい。中でも、リンパ系組織、毛細血管、リンパ管等を含んでいる、小腸に存在する腸管粘膜固有層が好ましい。
 腸管粘膜固有層からB細胞を採取する方法は、特に限定はされないが、例えば腸管を採取し、得られた腸管を洗浄後に切開して粘膜層を露出させる。次いで、適当な濃度のEDTAを含む生理食塩水中で振盪させて上皮細胞を遊離させて除去する。その後、適当な濃度のコラゲナーゼ等といった消化酵素で処理することによって、B細胞を回収する方法が挙げられる。これ以外の公知の方法を採用してもよくまた、市販の腸管粘膜固有層由来のB細胞を購入して入手してもよい。
 腸管粘膜固有層の由来、すなわち上記B細胞の由来は、特に限定はされないが、例えば、マウス、ラット、ハムスター、ウサギ、ヤギ、ヒツジ、ロバ、ブタ、ウシ、ウマ、ニワトリ、サル、チンパンジー、ヒト等が挙げられる。
 それ以外の細胞の種類(本明細書において、「他の細胞」と呼ぶことがある。)とは、前記B細胞以外の種類の細胞であって、当該B細胞と接触することによって融合してハイブリドーマを形成し、当該ハイブリドーマが上述したB細胞が発揮するIgA抗体を産生する機能を失わないものであればよい。
 この様な他の細胞として、前記B細胞と融合してハイブリドーマを形成し、当該ハイブリドーマが不死化機能を獲得できるような細胞が好ましく、具体的にはガン細胞、中でも、ミエローマ等といった骨髄種に由来する細胞等が挙げられる。好ましい他の細胞は、ミエローマである。
 他の細胞の由来は、特に限定はされないが、例えばマウス、ラット、ハムスター、ウサギ、ヤギ、ヒツジ、ロバ、ブタ、ウシ、ウマ、ニワトリ、サル、チンパンジー、ヒト等が挙げられる。
 融合とは、前記B細胞と他の細胞が一体不可分に合一することを意味する。ここで、一体不可分とは、融合した後の細胞が増殖する際の現象である細胞分裂は除外するものである。このようにして、融合した細胞が、本明細書におけるハイブリドーマの一例として挙げられる。
 混合及び融合時の条件は特に限定されず、細胞を融合する方法において通常用いられる条件を適宜採用すればよい。例えば、B細胞、又は他の細胞を培養する際に用いられる条件を適宜改変して行えばよい。このような方法として、例えば適当な培地中において、B細胞とB細胞以外の種類の細胞とを混合してポリエチレングリコール等の存在下でこれらを接触させる方法;上記混合の後に電気刺激を与える方法;センダイウイルス等のウイルスを用いる方法等の後に、これを37℃、5%の二酸化炭素の条件下でインキュベートする方法が挙げられる。
 融合にかかる時間も、特に限定されるものではなく、融合そのものが完了するまでの時間を適宜設定すればよい。融合が完了したことを確認する方法は、通常の細胞の融合を行う際に用いられる、公知の方法を適宜改変して行えばよい。このような方法として、例えば顕微鏡下にて融合の進行度合いを観察する方法が挙げられ、公知のキットを適宜選択して、その使用条件に沿って細胞を融合してもよい。
(工程2について)
 本発明に係るモノクローナルIgA抗体の製造方法における工程2は、上記工程1にて作製したハイブリドーマを培養し、少なくとも2種類の腸内細菌と結合するIgA抗体を産生する細胞を決定し、該細胞から、前記抗体を回収する工程である。
 本発明の工程2の前に、工程1にて得られるハイブリドーマを、モノクローナル抗体を製造する際に通常用いられるような限界希釈法等のサブクローニング工程等に供していてもよい。
 少なくとも2種類の腸内細菌に結合するIgA抗体を産生するハイブリドーマは、当該IgA抗体を、例えば後述する実施例に記載のような、該IgA抗体を識別しうる抗IgA抗体(必要に応じて、放射性同位体、蛍光、発色等で標識化されていてもよい。)を用いたELISA、EIA、RIA、FLISA、FIA等による手段、FACSによる手段等用いて決定すればよい。
 上記の具体的な『少なくとも2種類の腸内細菌との結合するIgA抗体を産生するバイブリドーマ』を決定する方法は、特に限定はされないが、例えば上述の抗IgA抗体を用いた以下に示す2種類の方法等が挙げられる。なお、以下の2種類の方法は、適宜組み合わせて用いてもよい。
 1つ目の決定する方法としては、
(A)上述の工程1にて得られるハイブリドーマの中から、ある1つの腸内細菌に結合するIgA抗体を産生するハイブリドーマを決定する工程;及び
(B)工程Aにて決定されたハイブリドーマの中から、それが産生するIgA抗体が、前記腸内細菌とは異なるその他の腸内細菌とも結合することを確認し、両方の腸内細菌と結合するIgA抗体を産生するハイブリドーマを決定する工程、
を含む方法が挙げられる。
 この場合、例えばELISA法を用いた手段であれば、工程Aとして、上述の工程1にて得られるハイブリドーマの培養上清をサンプリングし、斯かるサンプリング物と、ある1つ目の腸内細菌が固相化された、例えばマルチウェルプレートの1つのウェル内にて、斯かる腸内細菌と接触させて、前記サンプリング物に前記腸内細菌と結合するIgA抗体が存在することを確認することによって、IgA抗体を産生するハイブリドーマを決定すればよい。
 次いで、工程Bとして、工程Aにて決定したIgA抗体を産生するハイブリドーマの培養上清のサンプリング物をそのまま用いて、又は必要に応じて上述の決定したハイブリドーマの培養上清を再度サンプリングして、斯かるサンプリング物と、上記の腸内細菌とは異なる、その他の(2つ目の)腸内細菌が固相化された、例えばマルチウェルプレートの1つのウェル内にて、斯かる2つ目の腸内細菌と接触させて、当該サンプリング物に2つ目の腸内細菌とも結合するIgA抗体が存在することを確認することによって、少なくとも2種類の腸内細菌と結合するIgA抗体を産生するハイブリドーマを決定すればよい。
 当然、上述のその他の腸内細菌として、上述の1番目とも2番目とも異なる3番目以上の腸内細菌を用いて、工程Bに引き続く工程が繰り返し含まれていてもよい。
 なお、EIA、RIA、FLISA、FIA等の手段であれば、上記ELISAでの方法と同様に、ウェルまたはそれに類する実験器具に、1つ目、及び2つ目(必要に応じて3つ目以上)の腸内細菌をそれぞれ固相化すればよい。
 FACSによる手段を選択するのであれば、上述のサンプリング物を各種腸内細菌と混合した後に、抗IgA抗体をさらに接触させて、各種腸内細菌、それに結合するIgA抗体、更に抗IgA抗体の複合体が形成されていることを適宜検出して、腸内細菌に結合するIgA抗体を産生するハイブリドーマと決定する手段に適用すればよい。
 2つ目の方法として、異なる腸内細菌毎にIgA抗体の結合を検出する系を各々用いて、上述の工程1にて得られるハイブリドーマの中から、少なくとも2種類の腸内細菌と結合するIgA抗体を産生するハイブリドーマを特定する工程を含む方法が挙げられる。
 これについて、例えばELISA法を用いた手段であれば、上記の1つ目の方法における工程Aにおいて、異なる2種類の(必要に応じて3種類以上の)腸内細菌を、例えばマルチウェルプレートの異なる2つ(前記腸内細菌の種類数に応じて3つ以上)のウェル内に固相化し、工程1にて得られるハイブリドーマの培養上清のサンプリング物をそれぞれのウェル内にて、斯かる異なる2種類(又は3種類以上)の腸内細菌に接触させ、斯かるサンプリング物中に前記2種類(又は3種類以上)の両方の腸内細菌に結合するIgAが存在することを確認することによって、少なくとも2種類の腸内細菌に結合するIgAを産生するハイブリドーマを同時に特定する方法も挙げられる。
 なお、EIA、RIA、FLISA、FIA等の手段であれば、上記ELISAでの方法と同様に、ウェルまたはそれに類する実験器具に、1つ目及び2つ目(必要に応じて3つ目以上)の腸内細菌をそれぞれ固相化すればよい。
 FACSによる手段を選択するのであれば、それぞれのサンプリング物と各種腸内細菌と混合した後に、抗IgA抗体をさらに接触させて、各種腸内細菌、それに結合するIgA抗体、更に抗IgA抗体の複合体が形成されていることを適宜検出して、腸内細菌に結合するIgA抗体を産生するハイブリドーマを決定する手段に適用すればよい。
 また、上述の方法にて決定した少なくとも2種類の腸内細菌と結合するIgA抗体を産生するハイブリドーマを決定した後に、上述したようなモノクローナル抗体を製造する方法にて用いられる限界希釈法等を用いて、上記少なくとも2種類の腸内細菌と結合するIgA抗体を産生するハイブリドーマをさらにクローニングする工程が含まれていてもよい。
 このようなクローニングを行う際には、上述の少なくとも2種類の腸内細菌と結合するIgA抗体を産生するハイブリドーマを決定する際に採用したものと同一の特定の2種類の腸内細菌を用いて、上述のハイブリドーマの決定方法を繰り返し行えばよい。
 工程2における具体的な回収方法は、特に限定はされないが、例えばIgA抗体を産生する細胞の培養液の上清を回収する方法、前記細胞の溶解液を回収する方法が挙げられる。細胞の溶解液は、超音波破砕、フレンチプレス等といった公知の機械的手段及び/又は界面活性剤、細胞壁消化酵素、細胞膜消化酵素を用いた公知の化学的処理による方法を適宜組み合わせて細胞を溶解し、その後固液分離工程に供した後の液相画分を回収して本発明のモノクローナルIgA抗体が製造されうる。
 また、上述のハイブリドーマからIgAを回収する前に、当該ハイブリドーマを適当な培地を用いて一定期間培養した後に、上述のようにその上清を回収する方法、又は細胞の溶解液から回収する方法に供してもよい。この他にも、上述のハイブリドーマを免疫不全マウス等のハイブリドーマの由来に適応した動物個体に腹腔内投与し、斯かる個体の腹水から回収してもよい。
 なお、上述の方法で回収したモノクローナルIgA抗体は、適宜公知の精製工程に供してもよい。具体的な精製手段は、特に限定はされないが、例えばアセトン、硫酸アンモニウム等を用いた沈殿による精製手段;アフィニティ、陰イオン交換、陽イオン交換、サイズ排除、逆相等のカラムクロマトを用いた精製手段等といった、公知のタンパク質の精製手段を適宜組み合わせて採用すればよい。
 本発明のモノクローナルIgA抗体の製造方法には、以下の態様の製造方法も含まれる。例えば、上述の〔核酸〕にて詳述した核酸を、モノクローナルIgA抗体を産生し得る細胞に導入し、当該細胞を培養して、その細胞抽出液又は培養上清から、モノクローナルIgA抗体を回収し、必要に応じて精製する方法が挙げられる。
 このようなモノクローナルIgA抗体を産生し得る細胞とは、通常、哺乳類等に由来する高次構造を形成することによって機能を発揮する各種タンパク質を産生し得る細胞であれば、特に限定はされないが、例えばCOS細胞、HEK細胞、HELA細胞、CHO細胞等の哺乳類由来の細胞、Sf9等といった昆虫由来の細胞等といった公知の細胞を適宜選択すればよい。
 具体的な核酸の導入方法、当該核酸を導入した細胞の培養条件、回収方法、精製方法については、用いる細胞の種類等によって区々であり、特に限定されず、公知の方法を適宜改変し、組み合わせることで、本発明に係るモノクローナルIgA抗体を製造することができる。
 上述の本発明に係るモノクローナルIgA抗体がキメラ抗体である場合、上述の核酸に関し、重鎖可変領域及び軽鎖可変領域をコードする塩基配列、並びに異種由来の重鎖定常領域及び軽鎖定常領域をコードする塩基配列を有する核酸を作製し、作製した重鎖可変領域をコードする塩基配列と重鎖定常領域をコードする塩基配列を有する核酸を結合させ、また作製した軽鎖可変領域をコードする塩基配列と軽鎖定常領域をコードする塩基配列を有する核酸を結合させ、結合させたこれら核酸を上述のようにモノクローナルIgA抗体を産生し得る細胞に導入すればよい。
 更に、本発明に係るモノクローナルIgA抗体が、重鎖及び/又は軽鎖CDR1~3がマウスに由来するアミノ酸配列を有し、それ以外の領域がヒトに由来するアミノ酸配列を有するといった態様の抗体である場合(これを、ヒト化抗体と称することもある。)の製造方法も、上述のキメラ抗体と同様に、重鎖及び/又は軽鎖CDR1~3をコードする塩基配列、並びにそれら以外の領域をコードする塩基配列を有する核酸を作製し、重鎖および軽鎖を構成するように核酸を再結合し、これらを上述のようにモノクローナルIgA抗体を産生し得る細胞に導入すればよい。抗体の結合能の保持のため、一部の塩基を他の塩基に置換する必要がある場合もある。
 本発明に係るモノクローナルIgA抗体が、J鎖を含む場合の製造方法として、上述のようにモノクローナルIgA抗体を産生し得る細胞に、当該モノクローナルIgA抗体をコードする塩基配列を有する核酸と共に、J鎖をコードする塩基配列を含む核酸を導入すればよい。
 J鎖をコードする塩基配列は、上述の〔モノクローナルIgA抗体〕において説明した、NCBIのウェブサイトに記載されたJ鎖のアミノ酸配列を基に、公知の方法を採用して適宜決定することができる。また、斯かる核酸は、決定した塩基配列を基に、公知の方法を用いて製造することが可能である。
 本発明に係るモノクローナルIgA抗体が、分泌因子を含む場合の製造方法として、上述のようにモノクローナルIgA抗体を産生し得る細胞に、当該モノクローナルIgA抗体をコードする塩基配列を有する核酸、並びにJ鎖をコードする塩基配列を含む核酸と共に、分泌因子をコードする塩基配列を含む核酸を導入すればよい。この様な方法は、例えば、Li C,et al.Sheng Wu Gong Cheng Xue Bao.2011 Feb;27(2):219-25.等に記載されており、斯かる記載、及び必要に応じてこれを適宜改変することによって製造することが可能である。
 上述の分泌因子をコードする塩基配列を含む核酸としては、例えば、上述した〔モノクローナルIgA抗体〕に記載した、NCBIのウェブサイトに記載されたポリイムノグロブリンレセプターのアミノ酸配列を基に、公知の方法を採用して適宜決定することができる。また、斯かる核酸は、決定した塩基配列を基に、公知の方法を用いて製造することが可能である。
 〔ハイブリドーマ〕
 本発明に係るハイブリドーマは、上述の本発明に係るモノクローナルIgA抗体を産生するハイブリドーマである。
 具体的なハイブリドーマとは、上述の〔モノクローナルIgA抗体の製造方法〕における工程1で得られるものである。すなわち、上述の本発明に係るモノクローナルIgA抗体の製造方法における工程1にて記載したB細胞と、それ以外の種類、すなわちB細胞以外の種類の細胞を混合させて融合した細胞である。
 ハイブリドーマは、同一種由来のハイブリドーマであっても、異種由来のハイブリドーマであってもよい。同一種由来のハイブリドーマは、上述の〔モノクローナルIgA抗体の製造方法〕において、前記B細胞と前記B細胞以外の種類の細胞の由来種を同一にすればよく、異種由来のハイブリドーマは、前記B細胞と前記B細胞以外の種類の細胞の由来種を異なるものにすればよい。
 同一種由来のハイブリドーマとしては、例えば上述〔モノクローナルIgA抗体の製造方法〕における工程1に記載した由来のうち、B細胞及びB細胞以外の種類の細胞の由来において重複するものから適宜選択すればよい。例えば、ヒト由来、マウス由来、ラット由来、ハムスター由来、ウサギ由来、ヤギ由来、ロバ由来、ブタ由来、ウシ由来、ウマ由来、ニワトリ由来、サル由来、チンパンジー由来等の同一種由来ハイブリドーマが挙げられる。
 異種由来のハイブリドーマとしては、例えば上述の〔モノクローナルIgA抗体の製造方法〕における工程1に記載した由来のうち、B細胞及びB細胞以外の種類の細胞の由来において記載した由来から、それぞれ適宜選択したものを組み合わせればよい。例えば、ヒト、マウス、ラット、ハムスター、ウサギ、ヤギ、ロバ、ブタ、ウシ、ウマ、ニワトリ、サル、チンパンジー等の由来を適宜組み合わせた異種由来のハイブリドーマが挙げられる。
〔医薬組成物〕
 本発明に係る医薬組成物は、本発明に係るモノクローナルIgA抗体を含む。
 本発明に係る医薬組成物は、特に限定はされないが、例えば腸内疾患の治療のために好適に用いることができる。さらに好ましくは、腸内細菌の異常増殖及び/又は腸内細菌叢の病的変化により発症する腸内疾患の治療を目的として用いられる。なお、腸内細菌の異常増殖及び腸内細菌叢の病的変化とは、上述の〔本発明のモノクローナルIgA抗体〕にて説明した通りである。
 腸内細菌の異常増殖及び/又は腸内細菌叢の病的変化により発症する腸内疾患とは、特に限定はされないが、例えば、炎症性腸疾患、潰瘍性大腸炎、クローン病、アレルギー、喘息、肥満、自己免疫疾患等が挙げられ、中でも炎症性腸疾患が好ましい。
 このような本発明に係る医薬組成物は、本発明に係るモノクローナルIgA抗体を有効量含んでいればよく、例えば、医薬組成物100重量%中の本発明に係る抗体の含有割合が0.001~99.99重量%の範囲になるように、対象とする腸内疾患の種類、剤型、投与方法、投与対象、投与対象者の症状の度合い、並びに投与によって発揮される効果の程度等を勘案して、適宜設定することができる。
 本明細書にて使用する用語『有効量』とは、本発明に係るモノクローナルIgA抗体が腸内疾患に対する治療効果を発揮できる量、又は上述する所望の薬理学的効果及び/又は生理学的効果(腸内疾患治療効果)を発揮する量をいう。
 本発明に係る医薬組成物には、本発明に係るモノクローナルIgA抗体と共に、薬学的に許容可能な担体或いは添加物を配合しても良い。薬学的に許容可能な担体或いは添加物とは、任意の担体、希釈剤、賦形剤、懸濁剤、潤滑剤、アジュバント、媒体、送達システム、乳化剤、錠剤分解物質、吸収剤、保存剤、界面活性剤、着色剤、香料、または甘味料を意味し、公知のものを採用すればよい。
 本発明に係る医薬組成物は、上述の腸内疾患に罹患した個体に投与する工程を含む腸内疾患の治療方法における利用可能性を有している。また、上述のような腸内疾患の病態や症状を発症していないものの、腸内疾患の素因を持ち得る個体に投与する工程を含む腸内疾患の予防方法における利用可能性も有している。これらの個体を、本発明に係る医薬組成物の投与対象とすればよい。
 この様な投与対象となる個体は、特に限定はされないが、例えばヒト、マウス、ラット、モルモット、ウサギ、ハムスター、イヌ、ネコ、イタチ等が挙げられる。
 当該医薬組成物の投与量並びに投与方法は、投与対象となる個体が罹患する腸内疾患の種類、性別、種、年齢、全身状態、疾患の重篤度、所望する効果の程度等に区々ではある。投与量としては、通常は、0.001~100mg/kg/日の範囲で適宜設定すればよい。
 また、投与方法については、特に限定はされないが、消化管に直接投与することが好ましく、このような投与方法として、例えば経口投与、経鼻投与、経粘膜投与、経腸投与等が挙げられる。
 なお、経腸投与とは、肛門を介した投与には限られず、例えば胃瘻等の様に個体外からチューブ等を消化管に挿入して、それを経由した投与も含まれる。消化管を挿入する位置は腸に限らず、食道、胃、小腸(十二指腸、空腸、回腸等を含む。)、大腸(盲腸、結腸、直腸等を含む。)等が挙げられる。
 なお、このような本発明に係る医薬組成物の投与は、上記の量を1日に1度に投与してもよく、数回に分けて投与してもよい。また、上記疾患に対する治療効果を有する範囲において、投与間隔は、毎日、隔日、毎週、隔週、2~3週毎、毎月、隔月または2~3ヶ月毎でもよい。
〔経口又は経腸組成物〕
 本発明に係る経口又は経腸組成物は、本発明に係るモノクローナルIgA抗体を含む。
この様な経口又は経腸組成物を使用することによって、腸内細菌の異常増殖及び/又は腸内細菌叢の病的変化を抑制する効果を発揮する。なお、腸内細菌の異常増殖及び腸内細菌叢の病的変化とは、上述の〔本発明のモノクローナルIgA抗体〕にて説明した通りである。
 この様な経口又は経腸組成物における上記モノクローナルIgA抗体の配合割合は、特に限定はされず、経口又は経腸組成物の形態、用途等に応じて適宜調整すればよいが、通常は経口又は経腸組成物の総量に対して、0.001~99重量%程度とすればよい。
 本発明に係る経口又は経腸組成物の使用対象とする個体は、特に限定はされないが、例えばヒト、マウス、ラット、モルモット、ウサギ、ハムスター、イヌ、ネコ、イタチ等が挙げられる。
 上述のように、本発明に係る経口又は経腸組成物に含有される上述のモノクローナルIgA抗体は腸内細菌の異常増殖及び/又は腸内細菌叢の病的変化を抑制する効果を発揮することから、整腸組成物、腸内環境改善組成物、腸内環境最適化組成物、又は腸内腐敗防止組成物として特に有用である。
 本発明に係る経口又は経腸組成物の使用量は、上述のような経口又は経腸組成物の効果が発揮される範囲であれば、特に限定はされず、更に経口又は経腸組成物を摂取する個体の種類や、目的とする効果、その目的とする度合い、並びにその他の諸条件などに応じて設定すればよい。具体的には本発明に係るモノクローナルIgA抗体の量に換算して、通常は0.001~100mg/kg/日程度とすればよく、これを1日1回~数回に分けて摂取してもよい。
 なお、経腸とは、上述の〔医薬組成物〕にて説明したように、肛門を介した投与に限られない。例えば、本発明の経腸組成物を、公知の成分と共に配合して腸内洗浄液として使用することも可能である。
 本発明に係る経口又は経腸組成物は、腸内細菌の異常増殖及び/又は腸内細菌叢の病的変化を抑制する効果を発揮する本発明に係るモノクローナルIgA抗体を含むものであり、このような効果を発揮することを期待して食品の分野、飼料の分野に好適に用いることができる。従って、本発明の経口又は経腸組成物は、食品組成物又は飼料組成物とすることができる。
 上述の食品組成物は、本発明の経口又は経腸組成物を食品の分野に専ら好適に用いる組成物である。このような食品組成物は、整腸用、腸内環境改善用、腸内環境最適化用、腸内腐敗防止用等と表示された食品組成物として提供することができる。
 上述の食品組成物としては、一般の食品の他、条件付き特定保健用食品を含む特定保健用食品、栄養補助食品、機能性食品、病者用食品等を挙げることができる。
 上述の食品組成物の具体的な形態は特に限定はされないが、例えば、清涼飲料、炭酸飲料、栄養飲料、果実飲料、乳酸飲料、乳飲料等の飲料;アイスクリーム、アイスシャーベット、かき氷等の冷菓;飴、キャンディー、ガム、チョコレート、錠菓、スナック菓子、ビスケット、ゼリー、ジャム、クリーム、焼き菓子等の菓子類;そば、うどん、はるさめ、中華麺、即席麺等の麺類;かまぼこ、ハム、ソーセージ等の水産・畜産加工食品;加工乳、発酵乳等の乳製品;サラダ油、てんぷら油、マーガリン、マヨネーズ、ショートニング、ホイップクリーム、ドレッシング等の油脂及び油脂加工食品;ソース、たれ等の調味料;スープ、シチュー、サラダ、惣菜、ふりかけ、漬物、パン、シリアル等を例示できる。また、特定保健用食品、栄養補助食品、機能性食品等の場合であれば、粉末、顆粒、カプセル、トローチ、タブレット、シロップ等の形態が挙げられる。
 上述の飼料組成物は、本発明の飼料組成物を飼料の分野に専ら用いられる組成物である。このような飼料組成物は、整腸用、腸内環境改善用、腸内環境最適化用、腸内腐敗防止用等と表示された食品組成物として提供することができる。
 上述の飼料組成物の具体的な形態は特に限定はされないが、例えば上述した本発明に係る飼料組成物が発揮する効果が損なわれない限りにおいて、通常の飼料に混合、又は必要に応じて通常の飼料に配合可能な成分と共に混合して飼料組成物とすればよく、飼料組成物そのものを飼料としてもよい。
〔疾患の治療方法〕
 本発明に係る疾患の治療方法は、腸内疾患の治療方法であって、上述の本発明に係るモノクローナルIgA抗体、本発明に係る医薬組成物、又は本発明に係る経口組成物の有効量を、腸内疾患に罹患したヒトに投与する工程を含む方法である。
 上述の腸内疾患に罹患したヒトとは、上記〔本発明に係る医薬組成物〕にて説明した投与対象とすればよい。また、具体的な投与方法、投与量についても上記〔本発明に係る医薬組成物〕にて説明したものとすればよい。
 以下に、本発明をさらに説明する。但し、本発明が以下に示す実施例に限定されないのは言うまでもない。
実験例1:抗体の調製
〔1-1:腸管粘膜固有層細胞の分離〕
 8週齢の野生型C57BL/6マウス(日本クレア)を、自由飲食条件下で長浜バイオ大学付属実験施設にて20週齢まで飼育した。次いで飼育したマウスを、炭酸ガスにより安楽死させた後、開腹して小腸全長を摘出した。この摘出した小腸サンプルから、結合組織とパイエル板を取り除いた後、小腸を縦切開し、腸内容物をPBSで満たした10cmのdish内で洗い流した。そしてPBSをさらに3~4回交換して、小腸を十分洗浄した。
 また、野生型C57BL/6マウスと同様に、AIDタンパク質のN末端側に変異を導入したAID G23Sを発現するマウス(AID G23Sマウス)からも小腸を採取した。この様なAID G23Sマウスは、例えば非特許文献18に示す公知の方法を用いて作成することが可能である。
 次に、50mlチューブに1mMのEDTAを含有する50mlのPBSを入れ、洗浄した小腸を0.5~1cmの長さに切断して加えた。37℃で20分間振盪した後、ストレイナーに小腸断片を集め、その後PBSを廃棄した。さらに上述のチューブに小腸を入れて、10秒間激しく振盪させた後、再びストレイナーに小腸断片を集め、その後同様にPBSを廃棄した。この50mlのPBSを用いた10秒間の小腸の激しい振盪を2回繰り返した。
 次に、50ml容チューブに37℃に温めた50mlの消化酵素液(500mlのRPMI 1640、25mlのFCS(最終濃度5%)、2μlの2-メルカプトエタノール(最終濃度55μM)、0.75gのコラゲナーゼ、及び5mlジスパーゼ(最終濃度1U/ml)にて調製)を入れ、はさみでさらに切り刻んだ小腸断片を加え、これを37℃で60分間振盪した後、フラスコを10秒間静置し、小腸組織を沈めた後に、その上清を新しい50ml容チューブに移した。この消化酵素液を用いた消化工程を2回繰り返した。
 上記の消化工程において分離した上清を、1,500rpm、4℃で5分間遠心し、その上清を廃棄した。さらに得られた沈殿物を1mlの2%FCSを含有するRPMI 1640に懸濁した。懸濁液をフィルターに通して組織片を取り除き、細胞懸濁液を氷上に保存した。
 上述の2回目の酵素反応を60分間行った後、同様に細胞懸濁液を回収した。1回目の細胞懸濁液と2回目の細胞懸濁液の2本の細胞懸濁液を合わせ、これを腸管粘膜固有層の細胞とした。
〔IgA産生ハイブリドーマの作製〕
 上記〔腸管粘膜固有層細胞の分離〕において得られた腸管粘膜固有層の細胞と、マウスミエローマ細胞であるNS1細胞を融合してハイブリドーマを作成した。NS1細胞は細胞融合に先立って培養及び維持を行った。細胞融合は、STEMCELL Technologies社のClonaCell(登録商標)-HY Hybridoma Cloning Kitに従い、その試薬と手順に従って行った。
 次いで、得られたハイブリドーマを上記キットに従ってメチルセルロース含有培地内で増殖させクローニングを行った。その後、その増殖中のハイブリドーマから上清を分離した。この上清中に含まれる抗体に関して、サンドイッチELISAにより、産生される抗体がIgAであること、そして上清中のIgA抗体価を測定し、IgA産生ハイブリドーマのクローンを分離した。この結果、計37クローンが樹立されたことが確認された。
 各クローンからIsogenII(Nippon Gene Co.,Ltd.)を使用してRNAを抽出し、これを鋳型にしてcDNAを合成し、抗体のVh領域に対する7種のプライマー(MH1~7)及びCα領域特異的プライマー(IgAR)を用いてRT-PCRを行った。これらのプライマーの具体的な塩基配列を表3に示す。

 増幅されたVDJ領域PCR産物を直接シークエンスすることで、VDJ usageと突然変異の数を解析した。この解析により、独立クローンとして17種類のIgA産生ハイブリドーマを得たことが明らかになった。これらを、それぞれW1、W2、W3、W4、W6、W7、W11、W14、W24、W27、W28、W30、W32、W34、W37、W43、及びW47と呼ぶ。
 また、同様の実験によりG23Sマウスから、10種類のIgA産生ハイブリドーマの独立クローンが得られた。これらをそれぞれ、G1、G8、G9、G10、G12、G14、G15、G16、G18、及びG19と呼ぶ。
〔1-2:IgA抗体の調製〕
 上述の〔IgA産生ハイブリドーマの作製〕において得られた17クローンのIgA抗体産生ハイブリドーマを10%のFCSを含有するRPMI 1640にて大量培養し、その培養上清を遠心分離によって採取した。次いで得られた培養上清を0.22μmフィルターに通した後に、硫安沈殿とPBSに対して透析することによって、それぞれ27種類のモノクローナルIgA抗体(以下、本実施例において、〔クローン名〕モノクローナルIgA抗体と呼ぶことがある。)を粗精製した。
 もう一つの方法として、〔IgA産生ハイブリドーマの作製〕にて得られた27クローンのハイブリドーマを、免疫不全マウスに腹腔内注射し、10~21日間の育種の後、マウスから腹水を採取した。採取した腹水は硫安沈殿後PD-10カラムを使ってPBSにバッファーを置換した。これによっても、それぞれ27種類のモノクローナルIgA抗体が粗精製された。
 これらの方法で調製した27種類のモノクローナルIgA抗体を、ELISA法を用いてその濃度を測定した。ここで使用した抗体としては、固層へのコーティングにヤギ抗-マウスIgA(Southern Biotech)を2μg/ml、検出用にALP-標識化ヤギ抗-マウスIgA(Southern Biotech)を0.5μg/mlで用いた。標準検品としてマウスIgA κ(Immunology Consultants Laboratory)を用いた。
〔1-3:腸内常在細菌の準備〕
 長浜バイオ大学付属実験施設SPF室で飼育中の野生型マウス糞便をPBSに懸濁後、血液寒天培地に希釈懸濁液をぬり拡げ、48~72時間37℃で培養した。その後、コロニーから直接16S rRNA遺伝子のPCRを行った。PCR産物をシークエンスして、各コロニーの細菌の種類を同定した。この方法により、6種類のマウス腸内細細菌をクローニングした。
 クローニングされたマウス腸内常在菌は、具体的には以下の6種類である:
Enterorhabdus mucosicola;
Escherichia coli;
Staphylococcus aureus;
Lactobacillus murinus;
Enterococcus faecalis;
Pseudomonas fulva。
〔1-4:各種腸内細菌に対する結合力及び特異性の検索〕
 上述のクローニングした6種類の腸内常在菌及び市販の
Escherichia coli(DH5α株:TOYOBO社から入手);Lactobacillus casei(ATCCより入手)、
Coprococcus eutactus(ATCCより入手)、
Blautia coccoides(ATCCより入手)、
Megamonas hypermegale(ATCCより入手)、
Eubacterium rectale(ATCCより入手)、
Bifidobacterium bifidium(ATCCより入手)
の合計13種類の腸内細菌を至適条件で培養し遠心により集菌した。
 具体的には0.05MのNa2CO3バッファーに細菌を懸濁し、ELISA法用のプレートにコーティングした。公知の方法でブロッキングした後に、上述の27種類のモノクローナルIgA抗体(濃度は1.4μg/ml)をそれぞれに加え、上記13種類の腸内細菌に対するこれらのクローナルIgA抗体の結合力と特異性をそれぞれ、上述のELISA法を用いて測定した。なお、検出用の抗体は上述のものを使用した。
 この結果を図1に示す。上述の27種類のモノクローナルIgAのうち22種類のモノクローナルIgA(野生型マウス由来のW1、W3、W4、W6、W7、W11、W14、W24、W27、W30、W32、W34、W43、及びW45モノクローナルIgA抗体;G23Sマウス由来のG1、G9、G10、G12、G14、G15、G18、及びG19モノクローナルIgA抗体。)が、2種以上の腸内細菌に結合することが明らかとなった。
 次いで、これらのモノクローナルIgA抗体の使用濃度を変化させ、各種腸内細菌に対する結合力の強さを比較する実験を行った。具体的な実験方法は上述のELISA法を採用した測定方法であり、使用したモノクローナルIgA抗体は、W27、G15、G19モノクローナルIgA抗体である。また、ネガティブコントロールとしてW2クローンモノクローナルIgA抗体を用いた。
 そして、使用した腸内常在菌は、
Staphylococcus aureus、Escherichia coli、及びEnterococcus faecalisである。
 結果を図2に示す。各種腸内細菌に対して、W27モノクローナルIgA抗体クローンが濃度依存的に最も効果的に結合することが明らかとなった。
 同様の実験を、W27モノクローナルIgA抗体、これに分泌因子が結合したW27SC3F、W11、W34、W43、及びG15モノクローナルIgA抗体を用いて行った。また、ネガティブコントロールとしてW2モノクローナルIgA抗体を用いた。
 W27SC3FモノクローナルIgA抗体の製造は、以下のようにして行った。まず、野生型マウス小腸の約1cmの断片よりRNAを抽出した。これを基に、Invitrogen社のSuperScript IIIを用いて逆転写反応を行い、cDNAを得た。このcDNAを鋳型に用いて、2つプライマー(EcoRI SCF:5’-gaattcaccatgaggctctacttg-3’;配列番号69、Flag SC3R:5’-ctcgagtcacttgtcgtcatcgtctttgtagtccccgggatt-3’;配列番号70)でポリイムノグロブリンレセプターの細胞外ドメインをPCRで増幅した。その際、リバースプライマーにFlag-tagの塩基配列を付加し、細胞内での発現を抗Flag抗体で検出可能になるように設定した。増幅されたDNAが有する塩基配列は、配列番号71に示すアミノ酸配列をコードするものである。なお、C末端にはFLAGタグ配列が付加されている(mSC3F-FLAG)。
 得られたPCR産物を制限酵素処理し、pcDNA3.1(+)ベクター(Invitrogen社)にクローニングした。以下完成した発現ベクターをpcDNA3.1(+)/mSC3Fと記載する。pcDNA3.1(+)/mSC3FのプラスミドDNAを大腸菌より調製し、Nucleofector(AMAXA社)によりW27モノクローナル抗体を産生するハイブリドーマに遺伝子導入を行った。その後、G418による選択を行った。これによって選択されたハイブリドーマより、上述のようにバイブリドーマからモノクローナルIgA抗体を回収する方法に準じて、W27SC3FモノクローナルIgA抗体を得た。
 そして、使用した腸内常在菌は、
Enterococcus faecalis、Staphylococcus aureus、Escherichia coli、及びPseudomonas fulvaである。
 結果を図3に示す。この実験からも、W27モノクローナルIgA抗体が濃度依存的に最も効果的に各種腸内細菌に対して、結合することが明らかとなった。
〔1-5:各種モノクローナルIgA抗体を用いた各種細菌に対するウエスタンブロット解析〕
 腸内細菌として、
Escherichia coli(DH5α;市販品)、
Escherichia coli(マウス腸内細菌からのクローニング株)、
Pseudomonas fulva、
Staphylococcus aureus及び
Eubacterium rectale
を10mlのLB又は至適培養液中で、それぞれの菌体に適した条件下で振蕩培養を行った。遠心して集菌後、1%のNP-40を含むPBSに懸濁し氷上で超音波破砕を行った。氷上で30分間静置後、遠心分離によって上清を得た。これにSDS-buffer(2-ME含有)を加え、95℃で10分間の熱処理によりタンパク質を変性させた。8%のSDS-PAGE後、フィルターに転写し、ブロッキング後、W27、W30、及びW45モノクローナルIgA抗体(2μg/ml)、続いてGoat anti-mouse IgA(1μg/ml)(Southern Biotechnology社)、IR800 anti-goat IgG(0.2μg/ml)(Rockland社)、更に必要に応じでラベル化されたanti-Goat IgGにより反応を行うウエスタンブロッティング法に供し、Odyssey(LI-COR)により、そのシグナルを検出した。
 結果を、図4に示す。W27、W30、及びW45モノクローナルIgA抗体は、上述の全ての菌の抽出物に結合することが明らかとなった。従って、この実験結果からW27、W30、及びW45モノクローナルIgA抗体は腸内細菌の構成タンパク質1種類を抗原とすることが示唆された。さらに、この構成タンパク質の分子量が45~50kDa程度であることも明らかとなった。
 更に、Eubacterium rectale、Blautia coccoides、Coprococcus eutactus、Megamonas hypermegale、Lactobacillus casei、及び
Bifidobacterium bifidumも上記方法と同様の実験を行った。使用したモノクローナルIgA抗体は、W27である。結果を図4に合わせて示す。Eubacterium rectale以外の菌体の抽出物中のタンパク質に、W27モノクローナルIgA抗体は結合しないことが明らかとなった。
〔1-6:各種モノクローナルIgA抗体のアミノ酸配列の解析〕
 上述の方法によって得られたモノクローナルIgA抗体のうち、2種類以上の腸内細菌に結合することが確認されたW27クローン、W30クローン、W34クローン、W43クローン、及びW11モノクローナルIgA抗体の5つのモノクローナルIgA抗体のアミノ酸配列を、公知の方法によって解析した。
 アミノ酸配列を公知の方法によって解析した。具体的には5’及び3’RACE反応を行って、その全長塩基配列を得た。RACE法はIn-Fusion(登録商標)SMARTerTM Directional cDNA library Construction Kit (Clontech社)の方法に従った。Kitに付属のプライマーおよび上述したプライマーを組み合わせて重鎖に関してはシークエンス解析を行った。軽鎖に関しては、以下のプライマーを組み合わせてシークエンス解析を行った。

 結果を表4~表8に示す。なお、これらの表中の「全長配列」の下線部は可変領域を示し、「全長配列」及び「可変領域」の太字部は、前から(N末端から)順に、CDR1、CDR2及びCDR3を示す。
 また、これらのアミノ酸をコードする核酸配列は以下の通りである;
・W27モノクローナルIgA抗体;重鎖:配列番号51、軽鎖:配列番号52;
・W30モノクローナルIgA抗体;重鎖:配列番号53、軽鎖:配列番号54;

実施例2:モノクローナルIgA抗体の抗原特定実験
〔2-1:免疫沈降実験〕
 上記〔1-5〕の方法で上述の方法でEscehrichia coli(DH5α)、Pseudomonas fulva、及びStaphylococcus aureusのそれぞれ3種類の細菌より細胞抽出成分を得た後、2-ME(終濃度300mM)とSDS(終濃度1%)を加え95℃10分間で熱変性を行った。その後、溶液中の2-ME濃度を60mMにSDS濃度を0.2%に調整した。まず細胞抽出成分に洗浄済みProtein G Sepharose 4 Fast Flow(GE社)を100μl加えて、4℃で30分間の転倒混和を行い、Protein G Sepharoseへの非特異的結合成分を遠心操作により排除した。このPre-clear後の細胞抽出成分に、5μgのW27モノクローナルIgA抗体を加え氷上30分間静置、その後Goat anti-mouse IgAを5μg加えさらに氷上30分間静置した。続いて洗浄済みProtein G Sepharose 4 Fast Flow(GE社)を30μl加えて4℃で15時間の転倒混和を行った。1%のNP-40を含むPBSによる洗浄操作を4回行い、洗浄液を排除後のProtein G SepharoseにSDS-buffer(2-MEを含む)を加え、95℃10分間の熱処理によりタンパク質を変性させた(免疫沈降サンプル)。これを8%のSDS-PAGE、続いて上述のウエスタンブロッティングに供して上述の〔1-5〕にて確認したタンパク質が濃縮されたことを確認した(図5)。
〔2-2:2次元電気泳動・MS解析〕
 上述の免疫沈降サンプルに対してアセトン沈殿を行い、2次元電気泳動用のバッファーに溶解した。1次元目のゲルはAgar Gel(pH3~8;ATTO社)を使用した。2次元目は6%のSDS-PAGEを行った。毎回、サンプルを2組作成し同時に電気泳動した後は、1組目のサンプルは上述のウエスタンブロッティングを行い、標的タンパク質のゲル内の位置の確認を行った。具体的にはフィルターに転写後、まず転写タンパク質をMemCodeTM Reversible Protein Stain Kit(Thermo Scientific社)を用いて染色してその染色画像を得た(図5)。
 続いて除染してから上述の通常のウエスタンブロッティングを行い、標的タンパク質の位置を確認する画像を得た(図5)。2つの画像から標的タンパク質が2次元に展開されたタンパク質のどのスポットであるかを特定した。2組目のゲルはそのまま銀染色(シルベストステイン;Nacalai tesque社)を行い、すべてのタンパク質を可視化した。ウエスタンブロッティングから得られた位置情報をもとに標的タンパク質を特定し、ゲルから切り出した。
 そして切り出したゲルはトリプシンによるゲル内酵素消化を37℃で15時間行った。
MS解析はLCMS-IT-TOF(Shimazu社)により実施した。解析の結果、スポットのタンパク質は、セリンヒドロキシメチルトランスフェラーゼを抗原とすることが明確となった。
 次いで、C末端側にc-Mycタグを付けた配列番号74に示すアミノ酸配列に示すセリンヒドロキシメチルトランスフェラーゼの全長を大腸菌(DH5α)にて発現させ、これに対して上記の各種抗体(W11、W27、W30、W34、及びW43モノクローナルIgA抗体)との結合をウエスタンブロッティングによって確認した。結果を図9に示す。
 この結果、W27モノクローナルIgA抗体のみならず、W11、W30、W34、及びW43モノクローナルIgA抗体もセリンヒドロキシメチルトランスフェラーゼを抗原とすることが明らかとなった。
〔2-3:エピトープ特定実験〕
 次いで、セリンヒドロキシメチルトランスフェラーゼの各種変異体及び野生型を、大腸菌発現系及び293T細胞を用いて作製し、上記各種モノクローナル抗体のエピトープを検討する実験を行った。
 作成した各種変異体は、配列番号74に示すアミノ酸配列の
(1)1~417番目のアミノ酸配列(full length:野生型)、
(2)11~417番目のアミノ酸配列(△2-10)、
(3)26~417番目のアミノ酸配列(△2-25)、
(4)29~417番目のアミノ酸配列(△2-28)、
(5)1~333番目のアミノ酸配列(△334-417)及び
(6)1~250番目のアミノ酸配列(△251-417)であり、
C末端側にMycタグ(40aa)を設けた。
 これらの野生型及び変異型のセリンヒドロキシメチルトランスフェラーゼを上述の同様の方法を用いてウエスタンブロッティングに供した。抗体はW27モノクローナルIgA抗体を使用した。結果を図10に示す。
 図10に示す結果によると、W27モノクローナルIgA抗体は、△2-25変異体には結合するものの、△2-28変異体には結合しないことが明らかとなった。これは、大腸菌に由来する内在性の野生型セリンヒドロキシメチルトランスフェラーゼの影響がないと考えられる293Tの系を用いた場合の実験から明らかである。また、W27モノクローナルIgA抗体は△251-417変異体及び△334-417変異体に結合することが明らかとなった。
実施例3:W27モノクローナルIgA抗体経口投与実験
〔3-1:G23Sマウスの病態モデル解析〕
 G23Sマウスの、大腸組織切片を作成し、これをHE染色によって炎症の度合いを調べたところ、13-16週齢においても野生型マウスに比べて腺管の減少または萎縮が多く観察された(図6)。とくに40週齢以上では腺管の減少または萎縮、炎症性細胞の大腸粘膜内への浸潤が多く観察された。免疫染色ではCD11b+細胞やIL-6陽性細胞、IL-6、IL-17等の炎症性細胞の数が野生型マウスに比べて多く存在していた。
 また、TNFα、IFNγ、IL-1β、IL-17、IL-6等の炎症性サイトカインの発現量を、定量PCR法を用いた方法によって測定した結果からも同様に炎症性細胞の数が、野生型マウスよりも多く存在する結果が明らかとなった。以上のことから、G23Sマウスは、炎症性大腸炎、潰瘍性大腸炎等のモデルマウスとして用いるにふさわしいことが明らかとなった。
〔3-2:W27モノクローナルIgA抗体の効果〕
 上述のW27モノクローナルIgA抗体を、腸管免疫系過剰反応状態、即ちパイエル板胚中心過形成を示し、腸組織において炎症性サイトカインの産生が亢進しているマウスに対して経口投与を行った。
 本実験では10~12週齢のオスのG23Sマウスを用いた(バックグラウンドはBalb/c)。飼育用の飲料水中に25μg/mlの濃度になるようにW27モノクローナルIgA抗体を加え、これを28日間経口摂取させた。ネガティブコントロールとしては水のみを摂取させた実験、また野生型マウスに同様に水のみを摂取させた実験を行った。
なお、全マウス個体に対して、通常の飼育時と同様に飼料も与えた。
 投与開始一ヶ月後にマウスを安楽死させ、小腸よりパイエル板を分離した。パイエル板組織を、スライドガラスを使って単細胞に分離し、下記の抗体で表面抗原を染色した。フローサイトメトリー解析でB220+PNAhighを示した胚中心B細胞画分の細胞数の割合を求め、抗体投与の有無で胚中心そのものが退縮したかどうかを検討した。
 フローサイトメトリーで使用した抗体を以下に示した:
・PE-Cy7-labelled anti-mouse B220(eBioscience社);
・Biotinylated peanut agglutinin(VECTOR laboratories社);
・Streptavidin APC(eBioscience社)。
 パイエル板胚中心のB細胞の割合を分散分析とt検定により投与群と対照群の2群間の差が有意であるかどうかを検定した。具体的な、パイエル板胚中心のB細胞の割合及びB細胞数について解析した、結果を図7に示す。
 ネガティブコントロールのG23Sマウスでは、パイエル板の胚中心B細胞数が顕著に増加していたが、W27モノクローナルIgA抗体を経口投与することにより、胚中心のB細胞数が野生型マウスの程度にまで顕著に減少することが明らかになった。
 上述のように、G23Sマウスは炎症性大腸炎、潰瘍性大腸炎等のモデルマウスとして用いるにふさわしいことが明らかである。また、G23Sマウスでは、パイエル板の胚中心のB細胞が過剰に増殖していた。パイエル板の胚中心のB細胞は、腸内免疫システムにおけるIgA抗体の産生の場である。ここで、B細胞が過剰に増加することは、腸内免疫システムにおいてIgA抗体が過剰に産生している状態を示し、これは腸内細菌の腸管粘膜の侵入に対応しきれていない状態を意味する。この様な状態を改善すべく、腸内免疫システムにおいては補完的にT細胞が活性化され、これが上述のような炎症性サイトカインの産生を亢進し、炎症が生じる。
 このことから、W27モノクローナルIgA抗体は、パイエル板の胚中心のB細胞を減少させる効果を有することから、腸内免疫システムが正常化しており、T細胞等の働きを抑えることが可能になるため、炎症が解消する。従って、W27モノクローナルIgA抗体は、炎症性大腸炎、潰瘍性大腸炎等といった、腸内細菌の異常増殖及び/又は腸内細菌叢の病的変化により発症する疾患、延いては腸内疾患に対して有効であることが明確に示唆された。
 図11は13-15週齢の野生型マウス、および未処置のG23SマウスとW27モノクローナルIgA抗体の経口投与を4週間行ったG23Sマウスの大腸粘膜組織の腺管の減少または委縮病変を示す解析結果を示す。大腸全長の内容物を取り除き、生理食塩水で洗浄後、楊枝を軸にロールを作り、OCTコンパウンドに封入して凍結した。その後、6ミクロンの厚さの凍結切片を作製し、ヘマトキシリンーエオジン染色を行った。顕微鏡下に腺管が減少した病変部分を特定し、大腸全長に占める病変部位の広がりを数値化し、図11aのグラフにまとめた。典型的な組織像を図11bに示した。
 〔3-3:経口投与によるW27モノクローナルIgA抗体の体内での動態実験〕
 上述のW27モノクローナルIgA抗体を経口投与して、それが消化管を経由して腸にまで到達するかどうかを確認する実験を、IgAが産生されないAIDノックアウトマウスを用いて行った。
 AIDノックアウトマウスに上述の経口投与実験と同様の抗体投与量、方法を用いて、W27モノクローナルIgA抗体を経口投与した。具体的には飲料水にW27モノクローナルIgA抗体を25μg/mlになるように調製し、28日間、持続して経口摂取によって投与した。そして、2日目に糞便を回収した。回収した糞便100mgに対して0.9mlのPBSを加え懸濁後、遠心して上清を回収し上清中のIgA濃度をELISAによって測定した。
 結果を図8に示す。W27モノクローナルIgA抗体が糞便中から検出されたことから、これを経口投与しても、消化管内で分解酵素などの影響を受けることなく腸に到達していることが明らかとなった。このことは、W27モノクローナル抗体が経口組成物として十分に機能を発揮することを示唆している。
〔3-4:各種モノクローナルIgA抗体を用いた菌体増殖抑制効果〕
 大腸菌株DH5αをLB液体培地に加え、37℃で一晩震盪培養を行った。翌日、大腸菌液を適宜希釈し、LBプレートに播種し、37℃で一晩培養を行った。翌日にプレート上のコロニー数を数えることで、上記の大腸菌液中の大腸菌数を算定した。これを増殖抑制実験開始時の大腸菌数とした。
 上記のLBプレートに大腸菌液を播種すると同時に、96穴プレートにLB液体培地で希釈した50μLの大腸菌を加え、さらに終濃度が1.6mg/mLになるようにW27モノクローナルIgA抗体、W27SC3FモノクローナルIgA抗体、及びW2モノクローナルIgA抗体それぞれを加えた。
 96穴プレートを37℃で7時間静置培養を行った。7時間後に大腸菌液を適宜希釈し、これをLBプレートに播種し、37℃で一晩培養した。翌日、LBプレート上のコロニー数を数えて大腸菌数を計測し、上記の開始時大腸菌数を1とした場合に算出される細菌数をProliferation rate(図12グラフ縦軸)としてグラフにプロットした。結果を図12に示す。
 図12に示す結果から、W2モノクローナルIgA抗体とW27モノクローナルIgA抗体による大腸菌の増殖率に有意な差が見られた。詳しい機序は不明であるが、W2モノクローナルIgA抗体は大腸菌に結合しない傾向であることが上記実験例の結果から明らかであることから、W27モノクローナルIgA抗体は大腸菌に結合することで大腸菌の増殖を抑制していることが示唆される。
 さらに分泌型IgAであるW27SC3FモノクローナルIgA抗体は、最も強力な増殖抑制効果示すことから、実際に生体の腸管内に分泌されている分泌型IgA2量体が細菌を認識して結合した後に独自のエフェクター機能を発揮し、結果として大腸菌の増殖を抑制していることが示唆される。したがって、腸内細菌制御のためにモノクローナルIgA抗体を利用する際の剤型を分泌型IgAとすることが好ましいことが示唆される。

Claims (26)

  1.  セリンヒドロキシメチルトランスフェラーゼの11~333番目のアミノ酸に結合する、モノクローナルIgA抗体。
  2.  モノクローナルIgA抗体が、
    (1)配列番号3、13、23、33、又は43に示すアミノ酸配列からなる重鎖CDR1、
    (2)配列番号4、14、24、34、又は44に示すアミノ酸配列からなる重鎖CDR2、及び
    (3)配列番号5、15、25、35、又は45に示すアミノ酸配列からなる重鎖CDR3
    を含む重鎖可変領域;及び/又は
    (I)配列番号8、18、28、38、又は48に示すアミノ酸配列からなる軽鎖CDR1、
    (II)配列番号9、19、29、39、又は49に示すアミノ酸配列からなる軽鎖CDR2、及び
    (III)配列番号10、20、30、40、又は50に示すアミノ酸配列からなる軽鎖CDR3
    を含む軽鎖可変領域を含む、請求項1に記載のモノクローナルIgA抗体。
  3.  モノクローナルIgA抗体が、配列番号2、12、22、32、又は42に示すアミノ酸配列からなる重鎖可変領域;及び/又は配列番号7、17、27、37、又は47に示すアミノ酸配列からなる軽鎖可変領域を含む、請求項1又は2に記載のモノクローナルIgA抗体。
  4.  モノクローナルIgA抗体が、配列番号1、11、21、31、又は41に示すアミノ酸配列からなる重鎖;及び/又は配列番号6、16、26、36、又は46に示すアミノ酸配列からなる軽鎖を含む、請求項1~3の何れか1項に記載のモノクローナルIgA抗体。
  5.  少なくとも2種類の腸内細菌の成育を阻害する、請求項1~4の何れか1項に記載のモノクローナルIgA抗体。
  6.  腸内細菌が、
    Prevotella属に属する細菌、
    Bacteroides属に属する細菌、
    Megamonas属に属する細菌、
    Bifidobacterium属に属する細菌、
    Faecalibacterium属に属する細菌、
    Coprococcus属に属する細菌、
    Ruminococcus属に属する細菌、
    Blautia属に属する細菌、
    Eubacterium属に属する細菌、
    Roseburia属に属する細菌、
    Lactobacillus属に属する細菌、
    Clostridium属に属する細菌、
    Escherichia属に属する細菌、
    Staphylococcus属に属する細菌、
    Enterococcus属に属する細菌、
    Pseudomonas属に属する細菌、及び
    Enterorhabdus属に属する細菌
    からなる群に属する細菌より選択される少なくとも2種の細菌である、請求項1~5の何れか1項に記載の製造方法。
  7.  腸内細菌が、
    Prevotella melaninogenica、
    Bacteroidetes vulgatus、
    Megamonas funiformis、
    Megamonas hypermegale、
    Bifidobacterium bifidum、
    Faecalibacterium prausnitzii、
    Coprococcus eutactus、
    Ruminococcus obeum、
    Blautia productus、
    Blautia coccoides、
    Eubacterium rectale、
    Roseburia intestinalis、
    Lactobacillus murinus、
    Lactobacillus casei、
    Clostridium difficile、
    Escherichia coli、
    Staphylococcus aureus、
    Enterococcus faecalis、
    Pseudomonas fulva、及び
    Enterorhabdus mucosicola
    からなる群より選択される少なくとも2種の細菌である、請求項1~6の何れか1項に記載の製造方法。
  8.  モノクローナルIgA抗体が、IgA1又はIgA2である、請求項1~7の何れか1項に記載のモノクローナルIgA抗体。
  9.  モノクローナルIgA抗体が、J鎖を含む多量体である、請求項1~8の何れか1項に記載のモノクローナルIgA抗体。
  10.  モノクローナルIgA抗体が、分泌因子を含む、請求項1~9の何れか1項に記載のモノクローナルIgA抗体。
  11.  モノクローナルIgA抗体が、ヒト化抗体である、請求項1~10の何れか1項に記載の製造方法。
  12.  モノクローナルIgA抗体が、キメラ抗体である、請求項1~11の何れか1項に記載の製造方法。
  13.  以下の工程1及び2を含む、請求項1~12の何れか1項に記載のモノクローナルIgA抗体の製造方法:
    (1)腸管粘膜固有層から採取されたB細胞と、それ以外の種類の細胞を混合して融合させ、ハイブリドーマを作製する工程1、
    (2)工程1にて作製した該ハイブリドーマを培養し、少なくとも2種類の腸内細菌と結合するモノクローナルIgA抗体を産生する細胞を決定し、該細胞から、該IgA抗体を回収する工程2。
  14.  請求項1~12の何れか1項に記載のモノクローナルIgA抗体を含む、医薬組成物。
  15.  医薬組成物が、腸内疾患の治療用である、請求項14に記載の医薬組成物。
  16.  腸内疾患が、腸内細菌の異常増殖及び/又は腸内細菌叢の病的変化により発症する疾患である、請求項14又は15に記載の医薬組成物。
  17.  腸腸内細菌の異常増殖及び/又は腸内細菌叢の病的変化により発症する疾患が、炎症性腸疾患、潰瘍性大腸炎、クローン病、アレルギー、喘息、肥満、及び自己免疫疾患からなる群から選択される、請求項14~16の何れか1項に記載の医薬組成物。
  18. 請求項1~12の何れか1項に記載のモノクローナルIgA抗体を含む経口又は経腸組成物。
  19.  経口又は経腸組成物が、腸内環境改善組成物、腸内環境最適化組成物、又は腸内腐敗防止組成物である、請求項18に記載の経口又は経腸組成物。
  20.  経口又は経腸組成物が、飲食品組成物又は飼料組成物である、請求項18又は19に記載の経口又は経腸組成物。
  21.  請求項1~12の何れか1項に記載のモノクローナルIgA抗体をコードする核酸。
  22.  請求項1~12の何れか1項に記載のモノクローナルIgA抗体を産生するハイブリドーマ。
  23.  少なくとも2種類の腸内細菌に請求項1~12の何れか1項に記載の抗体を接触させる工程を含む、少なくとも2種類の腸内細菌の増殖を抑制する方法。
  24.  腸内疾患の治療方法であって、請求項1~12の何れか1項に記載の抗体の有効量を、腸内疾患に罹患したヒトに投与する工程を含む、治療方法。
  25.  腸内疾患の治療方法における使用のための、請求項1~12の何れか1項に記載のモノクローナルIgA抗体。
  26.  腸内疾患の治療するための、医薬の製造のための、請求項1~12の何れか1項に記載のモノクローナルIgA抗体の使用。
