Abstract

本発明の目的は、新規フィルター材およびそれを用いたフィルターを提供することである。具体的には、高い細胞分離性能を維持したまま、フィルターの目詰まりがなく、短時間で処理することができるフィルター材およびそれを用いたフィルターを提供する。さらに具体的には、繊維の塊(フロック)が均一に分布したフィルター材およびそれを用いたフィルターを提供する。本発明により、本発明に係る出願時以前に知られているフィルターの設計事項に関係なく、優れた細胞分離性能と通液性能を有する新規なフィルター材、および前記フィルター材を積層した新規フィルターを用いて効率的な細胞分離方法を提供する。

Description

新規細胞分離フィルター材およびそれを積層したフィルター
本発明は、細胞含有液から細胞を分離するための新規フィルター材およびそれを積層したフィルターに関する。
血液等の細胞含有液から細胞を分離する方法としては、細胞の比重差を利用した遠心分離法と、多孔質体を濾材とするフィルター法の2種類があるが、細胞分離性能が高く、操作が簡便で、コストが安いフィルター法が広く一般的に用いられている。現在、市場として更に高い細胞分離性能を有するフィルターに対する要求がある。しかしながら、従来技術を用いたフィルターでは、市場の要求を充たすほどの高い細胞分離性能を、単なるフィルター材の増量で達成することは困難であった。この問題点を解決するために、不織布をフィルター材として用いる場合の様々な改良手段が報告されている。たとえば、平均繊維直径を小さくする方法、充填密度を高くする方法、より繊維直径分布または孔径を均一化する方法、太さの異なる繊維が混在している孔径分布のバラつきを減少させる方法、或いは繊維直径と目付の両方を均一化する方法等が挙げられる(特許文献1~4)。しかしながら、上記方法で実際に細胞分離性能を高めようとすると、細胞分離性能の向上に伴って細胞含有液を通過させる時のフィルター部分の圧力損失が増大してしまい、処理したい細胞含有液量を濾過し終える前に、処理速度が極端に低下するという問題がある。
さらに繊維直径や平均孔径に代表される単一の物性要素の均一化以外にもフィルター材全体の繊維密度の標準偏差に関する検討がなされており、フィルター材全体の繊維密度の大小の差を小さくすること、つまり地合指数が小さいフィルター材について報告されている(特許文献5)。しかしながら、地合指数が小さなフィルター材であっても、細胞含有液の片流れが起こることで、細胞分離性能が十分に得られない場合があり、また細胞含有液の流れがフィルターの一部箇所に集中することで目詰まりが起こり、長い処理時間を要すという問題がある。
上述した通り、細胞を分離するためのフィルターは多く検討されているものの、細胞分離性能を改善した場合に発生する処理速度の低下およびフィルターへの目詰まり等の課題は未だ解決されていない。

本発明の目的は、上記課題を解決する新規フィルター材およびそれを用いたフィルターを提供することである。具体的には、高い細胞分離性能を維持したまま、フィルターの目詰まりがなく、短時間で処理することができるフィルター材およびそれを用いたフィルターを提供することである。さらに具体的には、繊維の塊(フロック)が均一に分布したフィルター材およびそれを用いたフィルターを提供することである。
本発明者らは、上記課題に対して鋭意研究を重ねた結果、新たに繊維の塊(フロック)の分布が均一なフィルター材を用いることで、細胞分離性能が高く、かつ目詰まりをせずに短時間で処理できる細胞分離フィルターが得られることを見出し、本発明を完成するに至った。
すなわち本発明は、平均繊維直径が0.3~5.0μmであり、厚さが0.10~0.60mmであり、フロックサイズ指数平均値が1.0~4.0であることを特徴とするフィルター材に関する。
前記フィルター材において、フロックサイズ指数比が0.78~1.02であることが好ましい。
前記フィルター材において、厚さ0.30mm相当のフロックサイズ指数平均値が1.0~3.0であることが好ましい。
前記フィルター材において、厚さ0.30mm相当のフロックサイズ指数比が0.84~1.02であることが好ましい。
前記フィルター材の少なくとも表面に親水性ポリマーを有することが好ましい。
前記フィルター材が不織布であることが好ましい。
また、本発明は、平均繊維直径が0.3~30μm、目付けが10~100g/m2であるフィルター材からなり、前記フィルター材中の少なくとも一部にフロックサイズ指数平均値が1.0~4.0であるフィルター材を有する液の導入口および導出口を備えたフィルターに関する。
前記フィルターにおいて、フィルター材中の少なくとも一部にフロックサイズ指数比が0.78~1.02であるフィルター材を有することが好ましい。

本発明により、本発明に係る出願時以前に知られているフィルターの設計事項に関係なく、優れた細胞分離性能と通液性能を有する新規なフィルター材、および前記フィルター材を積層した新規フィルターを用いて効率的な細胞分離方法を提供できる。

本発明について、以下具体的に説明する。本発明のフィルター材とは、細胞含有液から細胞を選択的に分離するためのフィルター材であり、本発明に係る出願時以前に知られているフィルターの設計事項に関係なく、優れた細胞分離性能と通液性能を有する新規なフィルター材、および前記フィルター材を積層した新規フィルターを意味する。例えば、地合指数に関係なく、優れた細胞分離能と通液性能を有する新規なフィルター材である。
本発明の細胞含有液とは、細胞を含む体液や合成液体、および培養細胞液を総称するものであり、例えば、全血、赤血球濃厚液、洗浄赤血球浮遊液、解凍赤血球濃厚液、合成血、乏血小板血漿(PPP)、多血小板血漿(PRP)、血漿、凍結血漿、血小板濃厚液およびバフィーコート(BC)などの、全血および全血から調製して得られる単一もしくは複数種類の血液成分からなる液体、またはそれらの液体に抗凝固剤や保存液などが添加された溶液、全血製剤、赤血球製剤、血小板製剤、血漿製剤、骨髄液、臍帯血、生体組織の処理液、またはES細胞、iPS細胞、成体幹細胞、体細胞などを培養した溶液等が挙げられる。
本発明においては、前記細胞含有液をフィルター材に通過させることにより、細胞種を選択的に分離することができる。選択的に分離される細胞種としては、ES細胞、iPS細胞、成体幹細胞(間葉系幹細胞、造血幹細胞等)、体細胞(白血球、赤血球、血小板、単球、単核球、リンパ球、上皮細胞、循環腫瘍細胞)等が挙げられ、特に前記細胞種のなかでも白血球、血小板、造血幹細胞、循環腫瘍細胞は本発明のフィルター材によって分離しやすいため好ましい。
本発明のフィルター材において、繊維の塊(フロック)の大きさを示す指数であるフロックサイズ指数平均値と、方向性を評価するフロックサイズ指数比が特定の範囲にある。
本発明でいうフロックサイズ指数平均値とは、フィルター材の下から光を当て、その透過光を電荷結合素子カメラ(以下CCDカメラと略す)で検知し、CCDカメラの各画素が検知した吸光度を用いて図1のような二値化像にした後、二値化後の全画素を巾方法(CD)および流れ方向(MD)に走査して、連続する黒点の数を平均したものであり、フロックサイズ指数平均値により、フィルター材の繊維の塊(フロック)の大きさと分布状況を把握できる。
本発明でいうフロックサイズ指数比とは、巾方向のフロックサイズ指数平均値を流れ方向のフロックサイズ指数平均値で除した値であり、フィルター材の繊維の方向性を把握できる。
具体的な算出方法は、以下の算出式(1)~(6)のとおりである。
(1)CCDカメラにより、各画素の透過率、
透過率=(点灯時の光量-消灯時の光量)/(サンプルがない状態での点灯時の光量-サンプルがない状態での消灯時の光量)×100(%)
を求める。
(2)平均透過率(全画素で測定された透過率の平均値)を求める。
(3)平均透過率より高い透過率の画素を白点、低い画素を黒点として二値化像にする。
(4)二値化後の全画素を巾方向(CD)および流れ方向(MD)に走査して、連続する黒点の数を平均し、巾方向の平均、流れ方向の平均および両方向の平均を求める。
(5)以下の式よりフロックサイズ指数平均値を求める。
フロックサイズ指数平均値=(巾方向のフロックサイズ指数平均値+流れ方向のフロックサイズ指数平均値)/2
(6)以下の式より、巾方向と流れ方向の指数比(縦横比)を求める。

フロックサイズ指数平均値およびフロックサイズ指数比はフォーメーションテスターFMT-MIII(野村商事株式会社製)にて測定する。測定機器の基本的な設定は工場出荷時から変更せず、CCDカメラの総画素数は約3400にて測定を行う。本発明でのフロックサイズ指数平均値およびフロックサイズ指数比の測定は、総画素数が約3400となるように測定サイズを4.6cm×4.6cm(1画素サイズ=0.79mm×0.79mm)にして測定を行うが、サンプルの形状に合わせて総画素数が等しくなるように測定サイズを変更しても良い。
本発明において、フロックサイズ指数平均値およびフロックサイズ指数比は、以下の方法により算出する。まず、フィルター材の材質、繊維直径、充填率、厚みが同一であるフィルター材を3枚用意し、それぞれのフロックサイズ指数平均値、フロックサイズ指数比を上記方法で測定する。次に測定したフィルター材3枚のフロックサイズ指数平均値、フロックサイズ指数比の平均値をフィルター材のフロックサイズ指数平均値、フロックサイズ指数比とする。同一フィルター内からフィルター材が測定必要数量得られなければ、同一種類のフィルターのフィルター材を組み合わせて測定しても良い。
本発明において、フロックサイズ指数平均値が高いほど、フィルター材の繊維の塊(フロック)が大きいことを表しており、フィルター材の繊維の分布が偏っていることを意味する。このフィルター材に細胞含有液を流すと、繊維密度が小さく通血抵抗の低い部分に流れが集中し、繊維密度が大きく通血抵抗の高い部分が十分に活用されない。その結果、フィルター材の全面を有効に活用できないため、フィルターの細胞分離性能が低下する。更に、細胞含有液の流れがフィルターの一部箇所に集中することで目詰まりが起き、フィルターの通液速度が低下する。反対に、フロックサイズ指数平均値が低いほど、フィルター材の繊維の塊(フロック)が小さいことを表しており、フィルター材の繊維が均等に分布していることを意味する。このフィルター材に細胞含有液を流すと、細胞含有液がフィルター材を均等に流れる。その結果、フィルター材の全面が有効に活用されるためフィルターの細胞分離性能は高くなる。更に、細胞含有液の流れが分散し、フィルターの目詰まりが発生しづらくなるため、フィルターの通液性能は高くなる。
本発明のフィルター材は、フロックサイズ指数平均値が1.0~4.0であることを特徴とする。フロックサイズ指数平均値が1.0より小さいフィルター材は安定して製造することが難しいため、好ましいフロックサイズ指数平均値の下限値は1.0以上であり、より好ましくは2.3以上であり、更に好ましくは2.6以上であり、特に好ましくは2.7以上であり、より更に好ましくは2.8以上である。フロックサイズ指数平均値が4.0より大きいと、細胞含有液がフィルター材を均等に流れないため、細胞分離性能が十分に得られず、また実質の通液面積の減少や目詰まりが起こりやすくなり、処理速度が低下する。よって、好ましいフロックサイズ指数平均値の上限値は4.0以下であり、より好ましくは3.4以下であり、更に好ましくは3.1以下である。
本発明において、フロックサイズ指数比が1前後であれば方向性がなく、その他の場合は指数の大きな方向に流れがあることを表している。フィルター材に方向性がある場合は、細胞含有液はフィルター材の方向に沿って流れるため、片流れが起きる。その結果、フィルター材の全面が十分に活用されないため、フィルターの細胞分離性能が低下する。更に、細胞含有液の流れがフィルター材の一部箇所に集中することで目詰まりが起き、フィルターの通液速度が低下する。反対にフィルター材に方向性がない場合は、細胞含有液がフィルター材を均等に流れる。その結果、フィルター材の全面が有効に活用されるためフィルターの細胞分離性能は高くなる。更に、細胞含有液の流れが分散し、フィルターの目詰まりが発生しづらくなるため、フィルターの通液性能は高くなる。
本発明のフィルター材は、フロックサイズ指数比が0.78~1.02であることを特徴とする。フロックサイズ指数比が1.02より大きいと、フィルター材の巾方向に方向性が存在し、細胞含有液の流れが巾方向に集中することにより通液面積の減少や目詰まりが発生しやすくなるため、細胞分離性能が十分に得られず、また通液面積の減少や目詰まりが起こりやすくなり、処理速度が低下する。よって、好ましいフロックサイズ指数比の上限値は1.02である。一方、フロックサイズ指数比が0.78より小さいとフィルター材の流れ方向に方向性が存在し、細胞含有液の流れが流れ方向に集中することにより通液面積の減少や目詰まりが発生しやすくなるため、細胞分離性能が十分に得られず、また通液面積の減少や目詰まりが起こりやすくなり、処理速度が低下する。よって、好ましいフロックサイズ指数比の下限値は0.78であり、より好ましくは0.87以上であり、更に好ましくは0.90以上であり、特に好ましくは0.98以上である。
本発明のフィルター材は、厚さ0.10~0.60mmであることを特徴とする。厚さの下限値は0.10mm以上であり、好ましくは0.12mm以上、更に好ましくは0.14mm以上である。厚み0.10未満では、安定してフィルター材を製造することが難しい傾向があるため好ましくない。一方、厚みの上限値は0.60mm以下であり、好ましくは0.55mm以下であり、更に好ましくは0.50mm以下である。厚みが0.60mmを超えるものは、フィルター材の目付が不均一になる傾向があるため好ましくない。
なお、本発明における厚みとは、以下の手順に従って求められる値をいう。フィルター材を3枚用意し、それぞれの厚さを測定する。厚さの測定は厚み計(OZAKI MFG.CO.,LTD.、PEACOCK MODEL H)を用いて4点測定し、フィルター材3枚の平均値をフィルター材の厚さとした。
上記の通り、フィルター材の厚みが大きくなるとフィルター材の目付が不均一になるため、フロックサイズ指数平均値およびフロックサイズ指数比は0.10mm~0.60mmの厚みを有するフィルター材で算出することが好ましく、厚みの上限と下限の中間値である0.30mm相当のフロックサイズ指数平均値およびフロックサイズ指数比を算出することがさらに好ましい。ここで、0.30mm相当のフロックサイズ指数平均値とは、厚さとフロックサイズ指数平均値から求めた回帰直線式より算出した厚さ0.30mmのフロックサイズ指数平均値を意味する。また、0.30mm相当のフロックサイズ指数比とは、厚さとフロックサイズ指数比から求めた回帰直線式より算出した厚さ0.30mmのフロックサイズ指数比を意味する。
以下の方法により厚さ0.30mm相当のフロックサイズ指数平均値およびフロックサイズ指数比も算出した。まず、フィルター材の材質、繊維直径、充填率等が同一であり、且つ厚みが均一であるフィルター材を3枚用意し、それぞれのフロックサイズ指数平均値、フロックサイズ指数比、そして厚さを測定する。厚さの測定は厚み計(OZAKI MFG.CO.,LTD.、PEACOCK MODEL H)を用いて4点測定し、その平均をフィルター材の厚さとした。次に測定したフィルター材3枚のうち2枚を重ねてフロックサイズ指数平均値、フロックサイズ指数比、そして厚さを測定する。全3通りの組合せについてフロックサイズ指数平均値、フロックサイズ指数比の測定を終了した後、厚さとフロックサイズ指数平均値、および厚さとフロックサイズ指数比の回帰直線式を求め、その式から厚さ0.30mmのフロックサイズ指数平均値、およびフロックサイズ指数比を求めた。同一フィルター内からフィルター材が測定必要数量得られなければ、同一種類のフィルターのフィルター材を組み合わせて測定しても良い。
本発明のフィルター材は、厚さ0.30mm相当のフロックサイズ指数平均値が1.0~3.0であることを特徴とする。厚さ0.30mm相当のフロックサイズ指数平均値が1.0より小さいフィルター材は安定して製造することが難しいため、好ましい厚さ0.30mm相当のフロックサイズ指数平均値の下限値は1.0以上、より好ましくは2.4以上である。厚さ0.30mm相当のフロックサイズ指数平均値が3.0より大きいと、フィルター材の構造が濾過面方向に対して不均一であり、細胞含有液がフィルター材を均等に流れないため、細胞分離性能が十分に得られず、また実質の通液面積の減少や目詰まりが起こりやすくなり、処理速度が低下する。よって、好ましい厚さ0.30mm相当のフロックサイズ指数平均値の上限値は3.0以下、より好ましくは2.8以下、更に好ましくは2.7以下である。
本発明のフィルター材は、厚さ0.30mm相当のフロックサイズ指数比が0.84~1.02であることを特徴とする。厚さ0.30mm相当のフロックサイズ指数比が1.02より大きいと、フィルター材の巾方向に方向性が存在し、細胞含有液の流れが巾方向に集中することにより実質の通液面積の減少や目詰まりが発生しやすくなるため、細胞分離性能が十分に得られず、また実質の通液面積の減少や目詰まりが起こりやすくなり、処理速度が低下する。よって、好ましい厚さ0.30mm相当のフロックサイズ指数比の上限値は1.02である。反対に、厚さ0.30mm相当のフロックサイズ指数比が0.84より小さいとフィルター材の流れ方向に方向性が存在し、細胞含有液の流れが流れ方向に集中することにより実質の通液面積の減少や目詰まりが発生しやすくなるため、細胞分離性能が十分に得られず、また実質の通液面積の減少や目詰まりが起こりやすくなり、処理速度が低下する。よって、好ましい厚さ0.30mm相当のフロックサイズ指数比の下限値は0.84以上、より好ましくは0.85以上、更に好ましくは0.90以上、特に好ましくは0.98以上である。
本発明のフィルター材の形状は、細胞含有液との接触を行うために接触頻度の観点から表面積が大きいことが望ましい。例えば、織布、不織布、繊維状、綿状などの繊維状構造体が挙げられる。特に細胞の吸着性、分離材としての取り扱い性からみて、織布、不織布が好ましく、中でも細胞との多点的な接触が可能である点で不織布が最も好ましい。なお、ここで不織布とは、編織によらずに繊維或いは繊維の集合体が、化学的、熱的、または機械的に結合された布状のものをいう。繊維と繊維とが互いに接触することによる摩擦により、或いは互いにもつれ合うことなどにより一定の形状を保っている場合も、機械的に結合されたことに含める。
フィルター材を構成するフィルター支持基材は、細胞にダメージを与えにくいものであれば特に制限はなく、例えば、ポリエステル、ポリオレフィン、ポリアクリロニトリル、ポリアミド、ポリスチレン、ポリアルキル(メタ)アクリレート、ポリ塩化ビニル、ポリクロロプレン、ポリウレタン、ポリビニルアルコール、ポリビニルアセテート、ポリスルホン、ポリエーテルスルホン、ポリブタジエン、ブタジエン-アクリロニトリル共重合体、スチレン-ブタジエン共重合体、エチレン-ビニルアルコール共重合体、セルロースジアセテート、エチルセルロース等が挙げられる。好ましくはポリエステル、ポリオレフィンで、特に好ましくはポリエステルである。更にポリエステルでもポリエチレンテレフタレート、ポリブチレンテレフタレートが好ましく、特に好ましくはポリエチレンテレフタレートである。
前記不織布は、湿式法、乾式法のいずれによっても製造することができるが、極細繊維が得られる点では、特にメルトブロー法やフラッシュ紡糸法あるいは抄造法などが好ましい。前記不織布の製造方法として、メルトブロー法の一例を以下に示す。
押出機で溶融した熱可塑性樹脂をメルトブローダイから高温・高速の空気流で糸状に吹き出し、繊維状に延伸された樹脂をコンベアー上で集積することで、繊維同士の絡み合いおよび融着が起こりノーバインダーの自己接着型極細繊維の不織布が得られる。この際、樹脂粘度、溶融温度、吐出量、熱風温度、風圧、DCD(紡糸口金と表面からコンベアーまでの距離)などを調整することにより、不織布の繊維直径、目付、繊維配向、繊維分散性を制御することができる。更に、熱プレス加工やテンター加工等により、不織布の厚み、平均孔径の制御を行うことが可能である。
本発明のフィルター材は、平均繊維直径が0.3~5.0μmであることを特徴とする。平均繊維直径の下限値は0.3μm以上であり、より好ましくは0.4μm以上、更に好ましくは0.5μm以上、とりわけ好ましくは1.0μm以上、特に好ましくは1.2以上である。平均繊維直径が0.3μm未満のものは、安定してフィルター材を製造することが難しく、細胞含有液の粘性抵抗も高くなりすぎる傾向があるため好ましくない。一方、平均繊維直径の上限値は5.0μm以下であり、好ましくは4.0μm以下である。より好ましくは3.0μm以下であり、更に好ましくは2.0μm以下である。平均繊維直径が5.0μmを超えるものは、細胞分離性能が低いため好ましくない。ここで、細胞含有液の粘性抵抗は、例えば、フィルターの圧力損失によって評価可能であり、細胞分離性能は、例えば、細胞分離率によって評価可能である。
なお、本発明における平均繊維直径とは、以下の手順に従って求められる値をいう。即ち、フィルター材の一部をサンプリングし、走査電子顕微鏡写真を基に、無作為に選択した100本以上の繊維の直径を計測し、それらを数平均して得られる。
本発明のフィルター材の平均孔径は、下限値として4.0μm以上が好ましく、より好ましくは4.5μm以上であり、更に好ましくは5.0μm以上である。平均孔径が4.0μm未満のものは、通液性能が低くなる傾向があるため好ましくない。一方、平均孔径の上限値は30.0μm以下が好ましく、より好ましくは20.0μm以下であり、更に好ましくは15.0μm以下であり、特に好ましくは10.0μm以下である。平均孔径が30.0μmを超えるものは、細胞分離性能が低くなる傾向があるため好ましくない。
ここで、通液性能は、例えば、濾過時間によって評価可能である。なお、本発明におけるフィルター材の平均孔径は、パームポロメーター(PMI社製)により測定したミーン・フロー・ポアサイズを意味する。
本発明のフィルター材の嵩密度は、下限値として0.05g/cm3以上が好ましく、より好ましくは0.06g/cm3以上であり、更に好ましくは0.07g/cm3以上である。嵩密度が0.05g/cm3より小さい場合は、細胞が漏れやすくなり、細胞分離性能が低下するため好ましくない。一方、嵩密度の上限値は0.40g/cm3以下が好ましく、より好ましくは0.35g/cm3以下であり、更に好ましくは0.30g/cm3以下である。嵩密度が0.40g/cm3を越えるものは、繊維間隔が緻密になって、通血性能が低くなる傾向があるため好ましくない。なお、本発明における嵩密度とは、フィルター材1cm3当たりの重さを測定した値を言う。
本発明のフィルター材の表面は、グラフト重合、ポリマーコーティング、アルカリ、酸等の薬品処理、プラズマ処理等で改質されていることが好ましい。中でもポリマーコーティングは高分子材料表面を好ましい構造に改質するための簡便で優れた改質法であるため、好ましい。
本発明のフィルター材は、少なくとも表面に親水性ポリマーを有することが好ましく、表面コーティングに用いるポリマーは、親水性ポリマーであれば、細胞への負荷が特に大きいものでない限り特に限定しないが、親水性官能基を有するヒドロキシエチル(メタ)アクリレートなどのモノマーと塩基性官能基を有するジメチルアミノエチル(メタ)アクリレートやジエチルアミノエチル(メタ)アクリレートとの共重合体や、ポリビニルピロリドンなどは、素材表面を親水化することにより濾材の濡れ性を改善することに加えて、荷電性官能基の導入により細胞の捕捉性能を向上できるため、特に好ましい。
上記ポリマーをフィルター支持基材にコーティングする方法としては、フィルター支持基材の細孔を著しく閉塞することなく、かつ、フィルター支持基材表面がある程度の範囲において均一にコーティングできるものであれば、特に制限はなく各種の方法を用いることができる。例えば、ポリマーを溶かした溶液にフィルター支持基材を含浸させる方法、ポリマーを溶かした溶液をフィルター支持基材に吹き付ける方法、ポリマーを溶かした溶液をグラビアロール等を用いフィルター支持基材に塗布・転写する方法、などが挙げられるが、中でもポリマーを溶かした溶液にフィルター支持基材を含浸させる方法が連続生産性に優れ、コストも低いことから好ましい。
上記ポリマーを溶かす溶剤としては、フィルター支持基材を著しく溶解させないものであれば特に限定はなく、例えば、N,N-ジメチルホルムアミド、N,N-ジメチルアセトアミドなどのアミド類、ジメチルスルホキシドなどのスルホキシド類、メタノール、エタノール、プロパノール、ブタノールなどのアルコール類、アセトン、メチルエチルケトンなどのケトン類、酢酸メチル、酢酸エチルなどのエステル類、トルエン、シクロヘキサンなどの炭化水素類、クロロホルム、ジクロロメタンなどのハロゲン化炭化水素類、水、および上記の複数の溶剤の可溶な範囲での混合物などが挙げられるが、好ましくはメタノール、エタノール、プロパノール、ブタノールなどのアルコール類で、特に好ましくはメタノール、エタノールである。
本発明のフィルターは、平均繊維直径が0.3~30μm、目付けが10~100g/m2であるフィルター材からなり、前記フィルター材中の少なくとも一部にフロックサイズ指数平均値が1.0~4.0であるフィルター材を有する液の導入口および導出口を備えたフィルターであることを特徴とする。
フィルターに含まれるフィルター材の平均繊維直径の下限値は0.3μm以上であり、より好ましくは0.4μm以上、更に好ましくは0.5μm以上、とりわけ好ましくは1.0μm以上、特に好ましくは1.2μm以上である。平均繊維直径が0.3μm未満のものは、安定してフィルター材を製造することが難しく、細胞含有液の粘性抵抗も高くなりすぎる傾向があるため好ましくない。一方、平均繊維直径の上限値は30μm以下であり、好ましくは25μm以下、より好ましくは20μm以下、特に好ましくは15μm以下である。平均繊維直径が30μmを超えるものは、細胞分離性能が低いため好ましくない。
フィルターに含まれるフィルター材の目付けの下限値は10g/m2以上であり、好ましくは20g/m2以上、より好ましくは30g/m2以上である。目付けが10g/m2未満のものは、安定してフィルター材を製造することが難しいため好ましくない。一方、目付けの上限値は100g/m2以下であり、好ましくは90g/m2以下、より好ましくは80g/m2以下、さらに好ましくは70g/m2以下である。目付けが100g/m2を超えるものは、表面の凹凸差が大きくなりやすくなるため均一なフィルター材を製造することが難しいため好ましくない。
本発明において、「フィルター材中の少なくとも一部に」とは、フロックサイズ指数平均値、フロックサイズ指数比、厚さ0.30mm相当のフロックサイズ指数平均値、厚さ0.30mm相当のフロックサイズ指数比のいずれかが、本発明における特定の数値範囲を満たすフィルター材を1枚以上有していることを示す。より具体的には、フィルター材中に1%以上含んでおり、好ましくはフィルター材中に5%以上、より好ましくはフィルター材中に6.2%以上、さらに好ましくはフィルター材中に10%以上、とりわけ好ましくはフィルター材中に50%以上、よりさらに好ましくはフィルター材中に75%、特に好ましくはフィルター材中に81%以上、最も好ましくはフィルター材中に85%以上である。
フィルターに含まれる一部のフィルター材のフロックサイズ指数平均値の下限値は1.0以上であり、より好ましくは2.3以上であり、更に好ましくは2.6以上であり、特に好ましくは2.7以上であり、より更に好ましくは2.8以上である。全てのフィルター材のフロックサイズ指数平均値が1.0より小さいフィルター材は安定して製造することが難しいため好ましくない。一方、フロックサイズ指数平均値の上限値は4.0以下であり、より好ましくは3.4以下であり、更に好ましくは3.1以下である。全てのフィルター材のフロックサイズ指数平均値が4.0より大きいと、細胞含有液がフィルターを均等に流れないため、細胞分離性能が十分に得られず、また実質の通液面積の減少や目詰まりが起こりやすくなり、処理速度が低下するため好ましくない。
フィルター容器は、球、コンテナ、カセット、バッグ、チューブ、カラム等、任意の形態をとりうるが、例えば、容量0.1~1000mL程度、直径0.1~15cm程度の透明または半透明の円柱状容器、あるいは一片の長さ0.1~20cm程度の正方形あるいは長方形で、厚みが0.1~5cm程度の四角柱状の形態等が好ましい。
本発明のフィルターは、フィルター材中の少なくとも一部にフロックサイズ指数比が0.78~1.02であるフィルター材を有することを特徴とする。
フィルターに含まれる全てのフィルター材のフロックサイズ指数比が1.02より大きいと、フィルターの巾方向に方向性が存在し、細胞含有液の流れが巾方向に集中することによりの通液面積の減少や目詰まりが発生しやすくなるため、細胞分離性能が十分に得られず、また通液面積の減少や目詰まりが起こりやすくなり、処理速度が低下する。よって、好ましいフロックサイズ指数比の上限値は1.02である。一方、全てのフィルター材のフロックサイズ指数比が0.78より小さいとフィルターの流れ方向に方向性が存在し、細胞含有液の流れが流れ方向に集中することにより通液面積の減少や目詰まりが発生しやすくなるため、細胞分離性能が十分に得られず、また通液面積の減少や目詰まりが起こりやすくなり、処理速度が低下する。よって、好ましいフロックサイズ指数比の下限値は0.78であり、より好ましくは0.87以上である。
本発明のフィルターは、フィルター材中の少なくとも一部に厚さ0.30mm相当のフロックサイズ指数平均値が1.0~3.0であるフィルター材を有することを特徴とする。
全てのフィルター材の厚さ0.30mm相当のフロックサイズ指数平均値が1.0より小さいフィルター材は安定して製造することが難しいため、フィルターに含まれる一部のフィルター材の好ましい厚さ0.30mm相当のフロックサイズ指数平均値の下限値は1.0以上、より好ましくは2.4以上である。
一方、全てのフィルター材の厚さ0.30mm相当のフロックサイズ指数平均値が3.0より大きいと、フィルターの構造が濾過面方向に対して不均一であり、細胞含有液がフィルターを均等に流れないため、細胞分離性能が十分に得られず、また実質の通液面積の減少や目詰まりが起こりやすくなり、処理速度が低下する。よって、好ましい厚さ0.30mm相当のフロックサイズ指数平均値の上限値は3.0以下、より好ましくは2.8以下、更に好ましくは2.7以下である。
本発明のフィルターは、フィルター材中の少なくとも一部に厚さ0.30mm相当のフロックサイズ指数比が0.84~1.02であるフィルター材を有することを特徴とする。
全てのフィルター材の厚さ0.30mm相当のフロックサイズ指数比が1.02より大きいと、フィルターの巾方向に方向性が存在し、細胞含有液の流れが巾方向に集中することにより実質の通液面積の減少や目詰まりが発生しやすくなるため、細胞分離性能が十分に得られず、また実質の通液面積の減少や目詰まりが起こりやすくなり、処理速度が低下する。よって、好ましい厚さ0.30mm相当のフロックサイズ指数比の上限値は1.02である。
一方、全てのフィルター材の厚さ0.30mm相当のフロックサイズ指数比が0.84より小さいとフィルターの流れ方向に方向性が存在し、細胞含有液の流れが流れ方向に集中することにより実質の通液面積の減少や目詰まりが発生しやすくなるため、細胞分離性能が十分に得られず、また実質の通液面積の減少や目詰まりが起こりやすくなり、処理速度が低下する。よって、好ましい厚さ0.30mm相当のフロックサイズ指数比の下限値は0.84以上、より好ましくは0.85以上、更に好ましくは0.90以上、特に好ましくは0.98以上である。
本発明のフィルターは、フィルター材を、液の流れ方向に積層して容器内に充填してもよい。フィルター材の積層枚数の下限値は1枚であるが、細胞分離性能の点から好ましくは5枚以上、より好ましくは10枚である。一方、フィルター材の積層枚数の上限値は100枚以下が好ましく、より好ましくは70枚以下であり、更に好ましくは50枚以下である。フィルター材の積層枚数が100枚を超えると通液性能が低下する問題が生じる。
本発明のフィルターは、フィルターの導入口側および/または導出口側に、異なる種類のフィルター材をさらに含んでもよい。例えば、主フィルター材よりも平均繊維直径の大きなフィルター材を導入口側に配置することで、通血性能がさらに向上する等の効果が期待される。
本発明のフィルターはフィルターの導入口側に平均繊維直径が大きなフィルター材を配置し、フィルターの導出口側に平均繊維直径が小さなフィルター材を配置してもよい。その場合、導出口側のフィルター材を第1のフィルター材、導入口側のフィルター材を第2のフィルター材とする。
第1のフィルター材の平均繊維直径が0.3~5.0μmであり、第1のフィルター材の導入口側に平均繊維直径が1.0~10.0μmであり、かつ平均繊維直径が第1のフィルター材よりも大きな第2のフィルター材が配置されると、通液性能がさらに向上する効果が得られる。
第2のフィルター材の平均繊維直径の下限値は1.0μm以上であり、より好ましくは1.2μm以上、更に好ましくは1.5μm以上である。平均繊維直径が1.0μm未満のものは、通液性能の向上度合いが低いため好ましくない。一方、平均繊維直径の上限値は10.0μm以下であり、より好ましくは8.0μm以下である。より好ましくは5.0μm以下であり、更に好ましくは3.0μm以下である。平均繊維直径が10.0μmを超えるものは、通液性能の向上度合いが低いため好ましくない。
本発明のフィルターは、フィルターの導入口側に微小凝集物除去のためのプレフィルター材を含んでいてもよい。細胞含有液には微小凝集物が含まれている場合が多いため、プレフィルターを使用して除去することが好ましい。プレフィルターとしては、例えば、平均繊維直径が3~50μmの繊維の集合体や平均孔径20~200μmの細孔を有する連続多孔質体などが好ましい。
本発明のフィルターシステムとは、フィルターの導入口側に採血バッグが接続され、該フィルターの導出口側に少なくとも1つの血液バッグが接続されていることを特徴とする。前記採血バッグは、例えば、採血針を備える採血チューブを連結し、内部にはACD(アシッドサイトレートデキストロース)、CPD(サイトレートフォスフェートデキストロース)等の抗凝固剤を封入したバッグを意味するが、一般の採血に使用する採血バッグを使用することもできる。また、前記血液バッグは、全血、濃厚赤血球、濃縮血小板血漿、血漿等を保存するためのバッグであり、一般的に用いられている血液バッグを用いることができる。
以下、実施例に基づいて、本発明を更に詳細に説明するが、本発明はこれらに限定されるものではない。
(実施例1)
<不織布の作製>
メルトブロー法で作製した以下のポリエチレンテレフタレート(以下PETと略す)不織布を使用した。
・プレフィルター材:平均繊維直径15μm、目付け30g/m2のPET不織布
・第2のフィルター材:平均繊維直径1.8μm、目付け70g/m2、平均孔径7.7μm、嵩密度0.20g/cm3、厚み0.35mm、フロックサイズ指数平均値2.6、フロックサイズ指数比0.98、厚さ0.30mm相当のフロックサイズ指数平均値2.7、厚さ0.30mm相当のフロックサイズ指数比0.98のPET不織布
・第1のフィルター材:平均繊維直径1.4μm、目付け40g/m2、平均孔径4.9μm、嵩密度0.18g/cm3、厚み0.22mm、フロックサイズ指数平均値3.1、フロックサイズ指数比0.98、厚さ0.30mm相当のフロックサイズ指数平均値2.8、厚さ0.30mm相当のフロックサイズ指数比0.98のPET不織布
<コーティング溶液の調製工程>
特級エタノール中に2-ヒドロキシエチルメタクリレートを0.95モル/L、2-ジメチルアミノエチルメタクリレートを0.05モル/Lの濃度になるように加え、全量を300mLとした。重合開始剤として2,2’-アゾビス(2,4-ジメチルバレロニトリル)(V-65)を0.005モル/Lの濃度になるように加え、窒素雰囲気下、45℃にて15時間重合させた後、過剰のn-ヘキサンに注いでポリマーを析出させて回収した。得られたポリマーをエタノールに再溶解させ、n-ヘキサンに注いで析出したポリマーを75℃にて4時間乾燥し、2-ヒドロキシエチルメタクリレートとN,N-ジメチルアミノエチルメタクリレートから成る共重合体(以下HEDMと略す)を得た。HEDMを濃度が1.0g/Lになるようにエタノールに溶解し、HEDMコーティング溶液を調製した。
<不織布のコーティング工程>
第2のフィルター材および第1のフィルター材をHEDMコーティング溶液に20℃にて5分間浸漬した後、ステンレス製バスケットに入れ、50℃にて1.5時間乾燥した。続いて、各不織布を水洗した後、ステンレス製バスケットに入れ、50℃にて3時間乾燥した。
<不織布ディスク作製工程>
ポンチを用いて、各不織布を直径18mmの円形に打抜き、ディスク状のプレフィルター材、第2のフィルター材、第1のフィルター材を作製した。
 <フィルター作製工程>
内径18mmの円柱ハウジング内に導入口側から導出口側へ順に、プレフィルター材を6枚、第2のフィルター材を2枚(フィルター材中の6.25%を占める)、第1のフィルター材を24枚挿入し(フィルター材中の75%を占める)、フィルターを作製した。フィルターの導入口と血液貯留容器を長さ60cmの塩化ビニル製チューブ(外径5mm、内径3mm)で接続し、クランプでチューブを閉じた。
<フィルターの性能評価>
ウシ血液に対して抗凝固剤として5%クエン酸三ナトリウム水溶液を10%添加して血液試料を調製した。恒温槽を用いて血液試料を28℃にした後、その血液試料24mLを血液貯留容器へ入れ、落差60cmの自然落下濾過を実施し、受器で濾過血液を22mL回収した。なお、濾過を開始してから血液貯留容器が空になるまでの時間を濾過時間とした。濾過前の白血球濃度、濾過前後の赤血球濃度、血小板濃度は血球カウンター(シスメックス(株)製、K-4500)を用いて測定し、濾過後の白血球濃度はLeucoCOUNTキットおよびFACSCalibur(共にベクトン・ディッキンソン社)を用いて、フローサイトメトリー法にて測定した。
白血球分離性能(-Log)、赤血球回収率(%)、および血小板分離率(%)は、
a=濾過前血液の白血球濃度、b=濾過後血液の白血球濃度、c=濾過前血液の赤血球濃度、d=濾過後血液の赤血球濃度、e=濾過前血液の血小板濃度、f=濾過後血液の血小板濃度とするとき、それぞれ、
白血球分離性能=-Log(b/a)
赤血球回収率=d/c×100(%)
血小板分離率=(e-f)/e×100(%)
で表される式によって求めた。結果を表1に示した。
(実施例2)
平均繊維直径1.3μm、目付け40g/m2、平均孔径6.8μm、嵩密度0.13g/cm3、厚み0.30mm、フロックサイズ指数平均値2.7、フロックサイズ指数比1.02、厚さ0.30mm相当のフロックサイズ指数平均値2.7、厚さ0.30mm相当のフロックサイズ指数比1.02のPET不織布を第1のフィルター材として24枚用いた以外は実施例1と同様にして、フィルターを作製した。作製したフィルターについて実施例1と同様の評価を行い、結果を表1に示した。
(実施例3)
平均繊維直径1.3μm、目付け40g/m2、平均孔径7.0μm、嵩密度0.11g/cm3、厚み0.35mm、フロックサイズ指数平均値2.7、フロックサイズ指数比0.99、厚さ0.30mm相当のフロックサイズ指数平均値2.7、厚さ0.30mm相当のフロックサイズ指数比0.99のPET不織布を第1のフィルター材として24枚用いた以外は実施例1と同様にして、フィルターを作製した。作製したフィルターについて実施例1と同様の評価を行い、結果を表1に示した。
(実施例4)
平均繊維直径1.3μm、目付け40g/m2、平均孔径6.0μm、嵩密度0.10g/cm3、厚み0.40mm、フロックサイズ指数平均値2.8、フロックサイズ指数比0.98、厚さ0.30mm相当のフロックサイズ指数平均値3.0、厚さ0.30mm相当のフロックサイズ指数比0.98のPET不織布を第1のフィルター材として24枚用いた以外は実施例1と同様にして、フィルターを作製した。作製したフィルターについて実施例1と同様の評価を行い、結果を表1に示した。
(実施例5)
平均繊維直径1.5μm、目付け40g/m2、平均孔径5.2μm、嵩密度0.11g/cm3、厚み0.38mm、フロックサイズ指数平均値2.3、フロックサイズ指数比0.87、厚さ0.30mm相当のフロックサイズ指数平均値2.4、厚さ0.30mm相当のフロックサイズ指数比0.85のPET不織布を第1のフィルター材として24枚用いた以外は実施例1と同様にして、フィルターを作製した。作製したフィルターについて実施例1と同様の評価を行い、結果を表1に示した。
(実施例6)
平均繊維直径1.5μm、目付け40g/m2、平均孔径4.7μm、嵩密度0.08g/cm3、厚み0.55mm、フロックサイズ指数平均値2.6、フロックサイズ指数比0.90、厚さ0.30mm相当のフロックサイズ指数平均値2.7、厚さ0.30mm相当のフロックサイズ指数比0.90のPET不織布を第1のフィルター材として24枚用いた以外は実施例1と同様にして、フィルターを作製した。作製したフィルターについて実施例1と同様の評価を行い、結果を表1に示した。
(実施例7)
平均繊維直径1.2μm、目付け40g/m2、平均孔径5.1μm、嵩密度0.19g/cm3、厚み0.21mm、フロックサイズ指数平均値3.4、フロックサイズ指数比0.78、厚さ0.30mm相当のフロックサイズ指数平均値3.0、厚さ0.30mm相当のフロックサイズ指数比0.84のPET不織布を第1のフィルター材として24枚用いた以外は実施例1と同様にして、フィルターを作製した。作製したフィルターについて実施例1と同様の評価を行い、結果を表1に示した。
(比較例1)
平均繊維直径1.3μm、目付け40g/m2、平均孔径4.9μm、嵩密度0.20g/cm3、厚み0.20mm、フロックサイズ指数平均値4.1、フロックサイズ指数比1.13、厚さ0.30mm相当のフロックサイズ指数平均値3.4、厚さ0.30mm相当のフロックサイズ指数比1.11のPET不織布を第1のフィルター材として24枚用いた以外は実施例1と同様にして、フィルターを作製した。
(比較例2)
平均繊維直径1.1μm、目付け30g/m2、平均孔径5.1μm、嵩密度0.20g/cm3、厚み0.15mm、フロックサイズ指数平均値4.6、フロックサイズ指数比1.03、厚さ0.30mm相当のフロックサイズ指数平均値4.4、厚さ0.30mm相当のフロックサイズ指数比1.02のPET不織布を第1のフィルター材として32枚用いた以外は実施例1と同様にして、フィルターを作製した。
(比較例3)
平均繊維直径1.3μm、目付け50g/m2、平均孔径4.7μm、嵩密度0.21g/cm3、厚み0.23mm、フロックサイズ指数平均値4.3、フロックサイズ指数比0.97、厚さ0.30mm相当のフロックサイズ指数平均値3.9、厚さ0.30mm相当のフロックサイズ指数比0.98のPET不織布を第1のフィルター材として19枚用いた以外は実施例1と同様にして、フィルターを作製した。

表1の結果から、フロックサイズ指数平均値が小さく、フロックサイズ指数比が1.00に近いフィルター材が高い細胞分離性能と高い通液性能を併せ持つことが分かる。
(実施例8)大容量血液の濾過評価
<不織布の作製>
メルトブロー法で作製した以下のPET不織布を使用した。
・プレフィルター材:平均繊維直径15μm、目付け30g/m2のPET不織布
・第2のフィルター材:平均繊維直径1.8μm、目付け70g/m2、平均孔径7.7μm、嵩密度0.20g/cm3、厚み0.35mm、フロックサイズ指数平均値2.6、フロックサイズ指数比0.98、厚さ0.30mm相当のフロックサイズ指数平均値2.7、厚さ0.30mm相当のフロックサイズ指数比0.98のPET不織布
・第1のフィルター材:平均繊維直径1.4μm、目付け40g/m2、平均孔径4.9μm、嵩密度0.18g/cm3、厚み0.22mm、フロックサイズ指数平均値3.1、フロックサイズ指数比0.98、厚さ0.30mm相当のフロックサイズ指数平均値2.8、厚さ0.30mm相当のフロックサイズ指数比0.98のPET不織布
<不織布のコーティング工程>
第2のフィルター材および第1のフィルター材をHEDMコーティング溶液に20℃にて5分間浸漬した後、ステンレス製バスケットに入れ、50℃にて1.5時間乾燥した。続いて、各不織布を水洗した後、ステンレス製バスケットに入れ、50℃にて3時間乾燥した。
<不織布ディスク作製工程>
ポンチを用いて、各不織布を7.2cm×7.2cmの正方形に打抜き、ディスク状のプレフィルター材、第2のフィルター材、第1のフィルター材を作製した。
<フィルター作製工程>
7.2cm×7.2cmの正方形ハウジング内に導入口側から導出口側へ順に、プレフィルター材を6枚、第2のフィルター材を2枚、第1のフィルター材を32枚挿入し、フィルターを作製した。フィルターの導入口と血液バッグを長さ60cmの塩化ビニル製チューブ(外径5mm、内径3mm)で接続し、クランプでチューブを閉じた。
<フィルターの性能評価>
ウシ全血に対して抗凝固剤として5%クエン酸三ナトリウム水溶液を10%添加して血液試料を調製した。血液試料456mLを血液バッグに入れ、恒温槽を用いて血液試料を28℃にした後、上記のフィルターで落差60cmの自然落下濾過を実施した。その結果、白血球分離性能(-Log)は5.29、赤血球回収率は100%、血小板分離率は86.6%、濾過時間は14.5分であった。
(実施例9)ヒト血液を用いた評価
第1のフィルター材を32枚用いた以外は実施例1と同様にして、フィルターを作製した。
<フィルターの性能評価>
ヒト全血200mLを抗凝固剤CPD液28mLが入った血液バッグ(テルモ製血液バッグCPD、組成:クエン酸ナトリウム水和物2.63w/v%、クエン酸水和物0.327w/v%、ブドウ糖2.32w/v%、リン酸二水素ナトリウム0.251w/v%)に採血、混和し、血液試料を調製した。恒温槽を用いて血液試料を28℃にした後、その血液試料24mLを血液貯留容器へ入れ、落差60cmの自然落下濾過を実施し、受器で濾過血液を22mL回収した。その結果、白血球分離性能(-Log)は5.27、赤血球回収率は100%、血小板分離率は100%、濾過時間は26.3分であった。

Claims (15)

  1. 平均繊維直径が0.3~5.0μmであり、厚さが0.10~0.60mmであり、フロックサイズ指数平均値が1.0~4.0であることを特徴とするフィルター材。
  2. フロックサイズ指数比が0.78~1.02であることを特徴とする請求項1に記載のフィルター材。
  3. 厚さ0.30mm相当のフロックサイズ指数平均値が1.0~3.0であることを特徴とする請求項1または2に記載のフィルター材。
  4. 厚さ0.30mm相当のフロックサイズ指数比が0.84~1.02であることを特徴とする請求項1~3のいずれかに記載のフィルター材。
  5. フィルター材の少なくとも表面に親水性ポリマーを有することを特徴とする請求項1~4のいずれかに記載のフィルター材。
  6. 不織布であることを特徴とする請求項1~5のいずれかに記載のフィルター材。
  7. 平均繊維直径が0.3~30μm、目付けが10~100g/m2であるフィルター材からなり、前記フィルター材中の少なくとも一部にフロックサイズ指数平均値が1.0~4.0であるフィルター材を有する液の導入口および導出口を備えたフィルター。
  8. フィルター材中の少なくとも一部にフロックサイズ指数比が0.78~1.02であるフィルター材を有する請求項7に記載のフィルター。
  9. フィルター材中の少なくとも一部に厚さ0.30mm相当のフロックサイズ指数平均値が1.0~3.0であるフィルター材を有する請求項7または8に記載のフィルター。
  10. フィルター材中の少なくとも一部に厚さ0.30mm相当のフロックサイズ指数比が0.84~1.02であるフィルター材を有する請求項7~9のいずれかに記載のフィルター。
  11. フィルター材が、液の流れ方向に積層されていることを特徴とする請求項7~10のいずれかに記載のフィルター。
  12. フィルターの導入口側および/または導出口側に、異なる種類のフィルター材を含む請求項7~11のいずれかに記載のフィルター。
  13. フィルターの導入口側に平均繊維直径が大きなフィルター材を配置し、フィルターの導出口側に平均繊維直径が小さなフィルター材を配置した請求項7~12のいずれかに記載のフィルター。
  14. フィルターの導入口側に微小凝集物除去のためのプレフィルター材を含む請求項7~13のいずれかに記載のフィルター。
  15. 請求項7~14のいずれかに記載のフィルターの導入口側に採血バッグが接続され、該フィルターの導出口側に少なくとも1つの血液バッグが接続されてなるフィルターシステム。
