Abstract

 検出感度を低下させることなく展開時間を短くし、かつ展開成分の液戻りを低減することのできる免疫クロマト分析方法を提供することを課題とし、グラスファイバーからなる吸収部を含む免疫クロマト分析装置を用いて検体に含まれる被検出物質を検出する方法であって、非イオン性界面活性剤存在下で検体および標識物質を移動相としてクロマトグラフ媒体部に展開させ、被検出物質を検出部で検出する方法を提供する。

Description

免疫クロマト分析方法
 免疫クロマト分析方法、免疫クロマト分析装置および免疫クロマト分析キットに関する。
 近年、検体の前処理を行う必要の無い、免疫クロマトグラフィーによるイムノアッセイは、抗体の持つ特異的反応性を利用して、試料液中の抗原を検出する簡便な体外診断キットもしくは携帯用診断装置として重要性が高まっている。特に、ウィルスや細菌等の病原体検査キットは、一般の病院やクリニックでも汎用されている身近な免疫クロマトグラフィー装置である。
 免疫クロマトグラフィー装置の最も簡単な構造としては、試料添加部、標識物質保持部、検出部が担持されたクロマトグラフ媒体、および吸収部が相互に繋がった構造である。
 この中で吸収部には、従来、セルロース繊維、もしくはパルプ繊維などからなる綿、不織物またはろ紙等が用いられている。しかし、これらの素材のみで構成される吸収材は、いったん吸収した展開成分を保持する力に欠け、その結果、吸収材からの展開成分の逆流が発生するという問題があった。
 従来、このような吸収材からの展開成分の逆流を防止するために、高吸水性繊維を用いて吸収材を構成することが試みられている(特許文献1)。また、吸水性ポリマーを含有した吸収材が提案されている(特許文献2)。

 しかし、特許文献1に記載の高吸水性繊維を含む吸収材は、展開成分の吸収速度が遅く、規定された反応時間(10~15分間)内に展開成分が十分に吸収材に吸収されず、クロマトグラフ媒体上に残存した過剰な液体試料により検出部に現れる陽性シグナルが滲んだり、有色の標識物質が十分に回収されないためにバックグラウンドシグナルが上昇するという問題があった。
 また、特許文献2に記載の吸水性ポリマーを含有した吸収材は、上記問題点は解決しているものの、製造時に吸水性ポリマーが零れ落ちやすいという問題があった。
 また、展開成分の展開時間を短くするため展開速度を上昇させると、吸収部で展開成分の吸収効率が悪いことにより、吸収部で展開成分を吸収しきれず、展開成分の液戻りが発生し、偽陽性が生じてしまうという問題があった。
 したがって、本発明の目的は、検出感度を低下させることなく展開時間を短くし、かつ展開成分の液戻りを低減することのできる免疫クロマト分析方法を提供することにある。
 本発明は、以下の通りである。
1.試料添加部、標識物質保持部、検出部が担持されたクロマトグラフ媒体部およびグラスファイバーからなる吸収部を含む免疫クロマト分析装置を用いて検体に含まれる被検出物質を検出する方法であって、以下の工程(1)~(4)を含む免疫クロマト分析方法。
(1)検体希釈液により検体を希釈した検体含有液を試料添加部に添加する工程
(2)標識物質保持部に保持されている標識物質により被検出物質を認識させる工程
(3)非イオン性界面活性剤存在下で検体および標識物質を移動相としてクロマトグラフ媒体部に展開させる工程
(4)展開された移動相中の被検出物質を検出部で検出する工程
2.前記試料添加部および前記検体希釈液の少なくとも一方に非イオン性界面活性剤を含有する前項1に記載の方法。
3.前記試料添加部における非イオン性界面活性剤の含有量が1cmあたり0.05~5mgである前項2に記載の方法。
4.前記検体希釈液における非イオン性界面活性剤の含有量が0.05~10質量%である前項2に記載の方法。
5.前記非イオン性界面活性剤のHLB値が10以上である前項1~4のいずれか1項に記載の方法。
6.前記クロマトグラフ媒体部が陰イオン性界面活性剤を含む前項1~5のいずれか1項に記載の方法。
7.前記クロマトグラフ媒体部における陰イオン性界面活性剤の含有量が、0.02~4質量%である前項6に記載の方法。
8.前記クロマトグラフ媒体部がニトロセルロースからなる前項1~7のいずれか1項に記載の方法。
9.前記吸収部の濾水時間が20秒~110秒であり、かつ密度が150~500mg/mである前項1~8のいずれか1項に記載の方法。
10.試料添加部、標識物質保持部、検出部が担持されたクロマトグラフ媒体部および吸収部を順次含む免疫クロマト分析装置であって、該試料添加部は非イオン性界面活性剤を乾燥保持し、該吸収部はグラスファイバーからなる免疫クロマト分析装置。
11.試料添加部、標識物質保持部、検出部が担持されたクロマトグラフ媒体部および吸収部を順次含む免疫クロマト装置と、検体に含まれる被検出物質を希釈する検体希釈液とを含む免疫クロマト分析キットであって、該吸収部がグラスファイバーからなり、該検体希釈液が非イオン性界面活性剤を含む免疫クロマト分析キット。
 本発明の免疫クロマト分析方法は、非イオン性界面活性剤存在下で、検体および標識物質を移動相としてクロマトグラフ媒体部に展開させる免疫クロマト分析装置を用いることで、検出感度を低下させることなく、且つ展開時間を短くすることができる。
 従来の免疫クロマト分析方法においては、展開時間が短くなることによって、吸収部で展開成分を吸収しきれず、展開成分の液戻りが発生してしまうが、本発明の免疫クロマト分析方法は、吸収部にグラスファイバーを用いることで、展開成分の液戻りを低減し、偽陽性を回避することができる。
図1は、免疫クロマト分析装置の構造を示す。
 以下に、本発明を実施するための形態を説明する。
 本発明の免疫クロマト分析方法は、試料添加部、標識物質保持部、検出部が担持されたクロマトグラフ媒体部およびグラスファイバーからなる吸収部を含む免疫クロマト分析装置を用いて、非イオン性界面活性剤の存在下で検体に含まれる被検出物質を検出する方法である。
 本発明で用いることのできる免疫クロマト分析装置およびその使用方法について、以下に説明をする。
 免疫クロマト分析装置は図1に示すように、試料添加部(サンプルパッドともいう)(1)、標識物質保持部(コンジュゲートパッドともいう)(2)、クロマトグラフ媒体部(3)、検出部(4)、吸収部(5)を順次含んでおり、さらにバッキングシート(6)を有している。順次とは、本発明の免疫クロマト分析装置において試料が展開する各部位の順番を示す。それらの各部位の構造、仕様および態様は以下のとおりである。
 試料添加部(1)は、免疫クロマト分析装置において、サンプルを滴下する部位である。通常免疫クロマトグラフに使用される素材であればどのようなものでもよい。通常、試料添加部は試料を吸収保持するグラスファイバーまたはセルロースの膜が使用される。
 試料添加部には、非イオン性界面活性剤を含有させておくことが好ましい。このことにより、検体が試料部の添加部材に吸着し検出感度が低下することを防ぐ。加えて、試料の表面張力を下げることにより、展開時間が短くなる。
 試料添加部における非イオン性界面活性剤の含有量は、0.05~5mgであることが好ましく、より好ましくは0.2mg~5mgであり、さらに好ましくは0.2mg~2.5mgである。
 試料添加部における非イオン性界面活性剤の含有量が0.05mg以上であることによって、試料の表面張力を有意に下げることができ、5mg以下であることによって、非イオン性界面活性剤による試料への変性作用を抑制できるからである。
 非イオン性界面活性剤としては、例えば、ポリオキシエチレンアルキルエーテル、ポリオキシエチレン/ポリオキシプロピレンアルキルエーテル、ポリオキシエチレンソルビタン脂肪酸エステル(商品名「Tween」シリーズ)、ポリオキシエチレンp-t-オクチルフェニルエーテル(商品名「Triton」シリーズ)、ポリオキシエチレンp-t-ノニルフェニルエーテル(商品名「TritonN」シリーズ)、アルキルポリグルコシド、脂肪酸ジエタノールアミド、アルキルモノグリセリルエーテル、NP-40(シグマアルドリッチ製)、IGAPEL CA-520(シグマアルドリッチ製)等を挙げることができる。
 非イオン性界面活性剤のHLB値は、10以上であることが好ましく、より好ましくは11.0以上であり、さらに好ましくは12.4以上である。上限値については特に限定されないが、好ましくは18以下である。
 非イオン性界面活性剤のHLB値が、10以上であることによって、疎水性相互作用による試料への変性効果を抑制できる。非イオン性界面活性剤のHLB値が、18以下であることによって、疎水性相互作用による試料への変性効果を抑制できる。
 非イオン性界面活性剤のCMC値は5μmol~60mmolであることが好ましく、より好ましくは10μmol~60mmolであり、さらに好ましくは12μmol~60mmolである。
 非イオン性界面活性剤のCMC値が、5μmol以上であることにより添加の効果を発揮することができる。また、60mmol以下であることにより使用量を少なくすることができ、経済的である。
 試料添加部に含有させる場合の非イオン性界面活性剤は、0.05~10質量%の濃度の水溶液とし、この溶液に試料添加部に使用する部材を浸漬し、その後真空乾燥又は自然乾燥させて、非イオン性界面活性剤を含有した試料添加部とする。
 標識物質保持部(2)は、あらかじめ被検出物質と結合する抗体に結合した、後述する標識物質(マーカー物質)が含有されている。標識物質保持部内を被検出物質が移動する際に抗体と結合し、標識化される。標識物質保持部は、例えば、グラスファイバー不織布またはセルロース膜等からなっている。
 クロマトグラフ媒体部(3)は、クロマトグラフの展開部位である。クロマトグラフ媒体部は、毛管現象を示す微細多孔性物質からなる不活性の膜である。クロマトグラフで使用される検出試薬、固定化試薬または被検出物質などと反応性を有しないものとして、例えば、ニトロセルロース製のメンブレン(以下「ニトロセルロースメンブレン」という場合がある)等が挙げられる。
 ニトロセルロース製のメンブレンとしては、ニトロセルロースが主体で含まれていればよく、純品またはニトロセルロース混合品などニトロセルロースを主材とするメンブレンを使用すると良いが、その他の材料でも何ら問題はない。その他の材料としては、例えば、セルロース類メンブレン、ナイロンメンブレン及び多孔質プラスチック布類(ポリエチレン、ポリプロピレン)などが挙げられる。
 前記ニトロセルロースメンブレンは、さらに毛細管現象を促進させる物質を含有させることもできる。該物質としては、膜面の表面張力を低下させ、親水性をもたらす物質が好ましい。例えば、糖類、アミノ酸の誘導体、脂肪酸エステル、各種合成界面活性剤またはアルコール等の両親媒性の作用を有する物質であって、免疫クロマトグラフ上での被検出物質の移動に影響がなく、マーカー物質(例えば金コロイドなど)の発色に影響を及ぼさない物質が好ましい。
 前記物質としては、例えば、陰イオン性界面活性剤、両イオン性界面活性剤、非イオン性界面活性剤、グリセリンが挙げられ、中でも陰イオン性界面活性剤が好ましい。陰イオン性界面活性剤としては、例えば、ドデシルベンゼンスルホン酸ナトリウム(SDBS)、ドデシルスルホン酸ナトリウム(SDS)、C12~C18のアルキルのスルホン酸ナトリウム(アルカンスルホン酸ナトリウム)、ジアルキルスルホコハク酸ナトリウムおよびコール酸ナトリウム等が挙げられる。
 特に好ましくは、ドデシルスルホン酸ナトリウム(SDS)またはC12~C18のアルキルのスルホン酸ナトリウム(アルカンスルホン酸ナトリウム)である。
 クロマトグラフ媒体部における陰イオン性界面活性剤の含有量は0.02~4質量%が好ましく、より好ましくは0.02~1質量%であり、さらに好ましくは0.1~1質量%である。
 クロマトグラフ媒体部における陰イオン性界面活性剤の含有量が0.02質量%以上であることで、十分な感度と検出時間の短縮効果が得られ、保存安定性の低下を回避できる。クロマトグラフ媒体部における陰イオン性界面活性剤の含有量が4質量%以下であることで、陰性検体での非特異反応が生じることを防ぐことができる。
 クロマトグラフ媒体部中に含有させる陰イオン性界面活性剤は0.1~10質量%の濃度の水溶液とし、この溶液にニトロセルロースメンブレンを浸漬し、その後真空乾燥又は自然乾燥させて、陰イオン性界面活性剤を含有したクロマト媒体とする。前記方法の他、製膜されたメンブレンに対して添加剤を直接添加し乾燥させるなど、製膜されたメンブレンに対し外添により含有させられればよく、これらの方法に限られない。
 このようにして調製したニトロセルロースメンブレンは、多孔性であって、毛細管現象を示す。この毛細管現象の指標は、吸水速度(吸水時間:capillary flow time)を測ることで確認できる。吸水速度は、検出感度と検査時間に影響する。
 本発明の免疫クロマト分析方法で用いるクロマトグラフ媒体としてのニトロセルロースメンブレンの形態及び大きさは特に制限されるものではなく、実際の操作の点及び反応結果の観察の点において適切であればよい。
 さらに操作をより簡便にするためには、反応部位が表面に形成されているクロマトグラフ媒体の裏面に、プラスチックなどよりなる支持体を設けることが好ましい。この支持体の性状は特に制限されるものではないが、目視判定によって測定結果の観察を行う場合には、支持体は、標識物質によりもたらされる色彩と類似しない色彩を有するものであることが好ましく、通常、無色又は白色であることが好ましい。
 検出部(4)は、前記クロマトグラフ媒体部(3)上に形成され、すなわち、被検出物質と特異的に結合する物質、例えば抗体が固定化試薬として任意の位置に固定化された反応部位が形成される。
 固定化試薬をクロマトグラフ媒体に固定化する方法としては、固定化試薬をクロマトグラフ媒体に物理的又は化学的手段により直接固定化する方法と、固定化試薬をラテックス粒子などの微粒子に物理的又は化学的に結合し、この微粒子をクロマトグラフ媒体に捕捉して固定化する間接固定化方法がある。
 直接的に固定化する方法としては、物理吸着を利用してもよいし、共有結合によってもよい。ニトロセルロースメンブレンの場合、物理吸着を行うことができる。共有結合ではクロマトグラフ媒体の活性化には一般的に臭化シアン、グルタルアルデヒドまたはカルボジイミド等が用いられるが、いずれの方法も用いることができる。
 間接的に固定化する方法としては、不溶性微粒子に固定化試薬を結合した後に、クロマトグラフ媒体に固定化する方法がある。不溶性微粒子の粒径はクロマトグラフ媒体に捕捉されるが移動することのできないサイズのものを選択することができ、好ましくは平均粒径5μm程度以下の微粒子である。
 これらの粒子としては抗原抗体反応に使用されるものが種々知られており、本発明でもこれら公知の粒子を使用することができる。例えば、ポリスチレン、スチレン-ブタジエン共重合体、スチレン-メタクリル酸共重合体、ポリグリシジルメタクリレートもしくはアクロレイン-エチレングリコールジメタクリレート共重合体などの乳化重合法によって得られる有機高分子ラテックス粒子などの有機高分子物質の微粒子、ゼラチン、ベントナイト、アガロースもしくは架橋デキストランなどの微粒子、シリカ、シリカ-アルミナもしくはアルミナなどの無機酸化物または無機酸化物にシランカップリング処理などで官能基を導入した無機粒子等が挙げられる。
 本発明においては、感度調整の容易さ等から直接固定化の方が好ましい。また、クロマトグラフ媒体への固定化試薬の固定化には、いろいろな方法が使用できる。例えば、マイクロシリンジ、調節ポンプ付きペンまたはインキ噴射印刷等、種々の技術が使用可能である。反応部位の形態としては特に限定されないが、円形のスポット、クロマトグラフ媒体の展開方向に垂直にのびるライン、数字、文字または+、-などの記号等として固定化することもできる。
 固定化試薬を固定化した後、非特異的な吸着により分析の精度が低下することを防止するため、必要に応じて、クロマトグラフ媒体に、公知の方法でブロッキング処理を行うことができる。
 一般にブロッキング処理はウシ血清アルブミン、スキムミルク、カゼインまたはゼラチン等の蛋白質が好適に用いられる。かかるブロッキング処理後に、必要に応じて、例えば、Tween20、TritonX-100またはSDS等の界面活性剤を1つ又は2つ以上組み合わせて洗浄してもよい。
 吸収部(5)は、クロマトグラフ媒体の末端に、展開液を吸収させるために設置される。本発明の免疫クロマト分析装置において、吸収部はグラスファイバーからなる。吸収部がグラスファイバーからなることによって、展開成分の液戻りを大幅に低減することができる。
 非イオン性界面活性剤存在下において免疫クロマト分析を行うことにより、展開時間が短くなるが、吸収部で展開成分の吸収効率が悪いと、吸収部で展開成分を吸収しきれず、展開成分の液戻りが発生してしまう。
 ここで、吸収部にセルロース繊維などの汎用される部材を吸収剤として用いると、吸収スピードが遅く吸収部での展開成分や非イオン性界面活性剤などの拡散速度も遅いため、非イオン性界面活性剤による吸着抑制効果が顕在化するため展開成分の逆流が生じてしまう。
 しかし、グラスファイバーからなる吸収部は吸水スピードが速く、展開成分が吸収部で速やかに拡散し展開成分や非イオン性界面活性剤などは希薄となり逆流が抑制される。すなわち、吸収部にグラスファイバーを用いることによって、展開成分の液戻りを大幅に低減することができ、展開時間の短縮および偽陽性の低減が可能となる。
 上述した試料添加部では、体積あたりの非イオン性界面活性剤の密度が高いため部材への吸着を抑制する作用を発揮するが、吸収部では添加部より体積が大きいため非イオン性界面活性剤の吸収部体積あたりの密度が低くなり吸着抑制が緩和されるため吸収部材からの展開成分の逆流は起こりにくくなる。
 吸収部は、濾水時間が20秒~110秒であることが好ましく、より好ましくは20~105秒であり、さらに好ましくは20~100秒である。
 吸収部の濾水時間が20秒以上であることにより、反応に必要な時間を最低限確保することができ、検出感度が低下することがない。吸収部の濾水時間が110秒以下であることにより、判定時間を短くすることができる。吸水部の濾水時間は、JIS P 3801:1995に従うことにより測定することができる。
 また、使用する吸収部の密度は、150~500mg/mであることが好ましく、より好ましくは200~500mg/mであり、さらに好ましくは200~350mg/mである。密度が200~350mg/mであるグラスファイバーを用いることによって、展開成分の液戻りを大幅に軽減できる。
 バッキングシート(6)は、基材である。片面に粘着剤を塗布したり、粘着テープを貼り付けることにより、片面が粘着性を有し、該粘着面上に試料添加部(1)、標識物質保持部(2)、クロマトグラフ媒体部(3)、検出部(4)、および吸収部(5)の一部または全部が密着して設けられている。バッキングシート(6)は、粘着剤によって試料液に対して不透過性、非透湿性となるようなものであれば、基材としては、特に限定されない。
 本発明の免疫クロマト分析方法は以下の工程(1)~(4)を含み、上記の免疫クロマト分析装置を用いて検体に含まれる被検出物質を検出する。
(1)検体希釈液により検体を希釈した検体含有液を試料添加部に添加する工程
(2)標識物質保持部に保持されている標識物質により被検出物質を認識させる工程
(3)非イオン性界面活性剤存在下で検体および標識物質を移動相としてクロマトグラフ媒体部に展開させる工程
(4)展開された移動相中の被検出物質を検出部で検出する工程
 各工程について以下に説明する。
(1)検体希釈液により検体を希釈した検体含有液を試料添加部に添加する工程
 工程(1)では、第1に、検体を、測定精度を低下させることなく、免疫クロマトグラフ媒体中をスムーズに移動する程度の濃度に検体希釈液で調整または希釈して検体含有液とする。第2に、該検体含有液を試料添加部(1)上に、所定量(通常、0.1~2ml)滴下する。検体含有液が滴下されると、検体含有液は試料添加部(1)中で移動を開始する。
 検体希釈液は、非イオン性界面活性剤を含有することが好ましい。このことにより、検出感度が低下することなく、且つ展開時間が短くなる。非イオン性界面活性剤は上記と同様のものを使用できる。検体希釈液における非イオン性界面活性剤の含有量は、0.05~10質量%であることが好ましく、より好ましくは0.05~5質量%であり、さらに好ましくは0.05~3.5質量%である。
 検体希釈液における非イオン性界面活性剤の含有量が0.05質量%以上であることによって、検体が試料滴下部の部材に吸着し、検出感度が低下することを防ぎ、且つ検体希釈液の表面張力を下げることで展開時間を短くすることができる。
 検体希釈液における非イオン性界面活性剤の含有量が10質量%以下であることによって、非イオン性界面活性剤による試料の変性作用を抑制できるためである。
 被検出物質を含む検体としては、例えば、生体試料、即ち、全血、血清、血漿、尿、唾液、喀痰、鼻腔又は咽頭拭い液、髄液、羊水、乳頭分泌液、涙、汗、皮膚からの浸出液、組織や細胞及び便からの抽出液等の他、牛乳、卵、小麦、豆、牛肉、豚肉、鶏肉などやそれらを含む食品等の抽出液等が挙げられる。
(2)標識物質保持部に保持されている標識物質により被検出物質を認識させる工程
 工程(2)は、工程(1)において試料添加部に添加された検体含有液を、標識物質保持部(2)へと移動させ、標識物質保持部に保持されている標識物質により検体中の被検出物質を認識させる工程である。
 標識物質は、被検出物質と特異的に結合する物質、例えば抗体を標識化する。免疫クロマトグラフ法における検出試薬の標識には、一般に酵素等も使用されるが、被検出物質の存在を目視で判定するのに適していることから、標識物質としては不溶性担体を用いることが好ましい。検出試薬を不溶性担体に感作することにより標識化した検出試薬を調製することができる。
 標識物質としての不溶性担体には、金、銀もしくは白金のようなコロイド状金属粒子、酸化鉄のようなコロイド状金属酸化物粒子、硫黄などのコロイド状非金属粒子及び合成高分子よりなるラテックス粒子、またはその他を用いることができる。特に金コロイドが、検出が簡便で好ましい。
 不溶性担体は、被検出物質の存在を視覚的に判定するのに適した標識物質であり、目視による判定を容易にするためには有色であることが好ましい。コロイド状金属粒子及びコロイド状金属酸化物粒子は、それ自体が粒径に応じた特定の自然色を呈するものであり、その色彩を標識として利用することができる。
 標識物質として用いることのできるコロイド状金属粒子及びコロイド状金属酸化物粒子には、例えば、コロイド状金粒子、コロイド状銀粒子、コロイド状白金粒子、コロイド状酸化鉄粒子、コロイド状水酸化アルミニウム粒子などが挙げられる。
 特に、コロイド状金粒子とコロイド状銀粒子が適当な粒径において、コロイド状金粒子は赤色、コロイド状銀粒子は黄色を示す点で好ましい。
 コロイド状金属粒子として、例えばコロイド状金粒子を用いる場合には、市販のものを用いてもよい。あるいは、常法、例えば塩化金酸をクエン酸ナトリウムで還元する方法によりコロイド状金粒子を調製することができる。
 検出試薬をコロイド状金属粒子に感作する方法としては、物理吸着や化学結合などの公知の方法が使用できる。例えば、コロイド状金粒子に抗体を感作した検出試薬は、金粒子がコロイド状に分散した溶液に抗体を加えて物理吸着させた後、牛血清アルブミン溶液を添加して抗体が未結合である粒子表面をブロッキングすることにより調製する。
 実際の免疫クロマト分析の実施において、不溶性担体により標識化した検出試薬は、移動相を構成する展開液に分散して適用することもできるし、固定相を構成するクロマトグラフ媒体における移動相の展開移動経路上、すなわちクロマトグラフ媒体の移動相が適用される端部と反応部位との間の領域に存在させて適用することもできる。
 クロマトグラフ媒体上に存在させる場合、検出試薬が展開液に速やかに溶解して毛管作用によって自由に移動できるように、検出試薬を支持させるのが好ましい。支持させる部位には、検出試薬が感作された不溶性担体の再溶解性を良好にするため、サッカロース、マルトースもしくはラクトース等の糖類、またはマンニトール等の糖アルコールを添加して塗布したり、これらの物質を予めコーティングしたりしておくこともできる。
 検出試薬を塗布・乾燥等によりクロマトグラフ媒体上に存在させる際には、固定化試薬が固定化されたクロマトグラフ媒体に直接、塗布・乾燥等することもできるし、別の多孔性物質、例えばセルロース濾紙、グラスファイバー濾紙またはナイロン不織布に塗布・乾燥等して検出試薬保持部材を形成した後、固定化試薬が固定化されたクロマトグラフ媒体と毛管で繋がるように配置してもよい。
 本発明の方法により検出される被検出物質としては、それに特異的に結合する物質が存在するものであれば特に限定されず、例えば、蛋白質、ペプチド、核酸、糖(特に糖タンパク質の糖部分、糖脂質の糖部分等)、複合糖質等が挙げられる。
 本発明において「特異的に結合する」とは、生体分子が持つ親和力に基づいて結合することを意味する。このような親和力に基づく結合としては、例えば、抗原と抗体との結合、糖とレクチンとの結合、ホルモンと受容体との結合、酵素と阻害剤との結合、相補的核酸同士及び核酸と核酸結合蛋白質との結合などが挙げられる。
 従って、被検出物質が抗原性を有する場合、被検出物質に特異的に結合する物質としては、例えば、ポリクローナル抗体又はモノクローナル抗体が挙げられる。また、被検出物質が糖の場合、被検出物質に特異的に結合する物質としては例えば、レクチンタンパク質が挙げられる。
 具体的な被検出物質としては、例えば、癌胎児性抗原(CEA)、HER2タンパク、前立腺特異抗原(PSA)、CA19-9、α-フェトプロテイン(AFP)、免疫抑制酸性タンパク(IPA)、CA15-3、CA125、エストロゲンレセプター、プロゲステロンレセプター、便潜血、トロポニンI、トロポニンT、CK-MB、CRP、ヒト絨毛性ゴナドトロピン(hCG)、黄体形成ホルモン(LH)、卵胞刺激ホルモン(FSH)、梅毒抗体、インフルエンザウィルス、ヒトヘモグロビン、クラミジア抗原、A群β溶連菌抗原、HBs抗体、HBs抗原、ロタウィルス、アデノウィルス、アルブミンまたは糖化アルブミン等が挙げられるが、これらに限定されるものではない。
(3)非イオン性界面活性剤存在下で検体および標識物質を移動相としてクロマトグラフ媒体部に展開させる工程
 工程(3)は、工程(2)において被検出物質が標識物質に認識された後、検体および標識物質を、非イオン性界面活性剤存在下でクロマトグラフ媒体部(3)上を移動相として通過させる工程である。
 非イオン性界面活性剤は、検体および標識物質を移動相としてクロマトグラフ媒体部に展開させる過程において存在させておく。上記したように試料添加部に含有させておくか、または検体希釈液に含有させることが好ましい。
 非イオン性界面活性剤を含有させておくことによって検体が試料滴下部の部材へ吸着し、検出感度が低下することを防ぐことができ、試料溶液の表面張力を下げることで判定時間を短くすることができるからである。より好ましくは検体希釈液に非イオン性界面活性剤を含有させる。
 検体希釈液に非イオン性界面活性剤を含有させることによって、試料中に成分を予め均一化することができ、部材に含有させるよりもより部材への吸着を抑制する効果が期待できる。加えて、試料滴下部での非イオン性界面活性剤の溶出時間がないため、より判定時間が短くすることができるためである。非イオン性界面活性剤は、試料添加部と検体希釈液の両方に含有させておいてもよい。
 本発明の免疫クロマト分析方法においては、必要であれば展開液を用いてもよい。展開液は、免疫クロマトグラフ法において移動相を構成する液体であり、固定相であるクロマトグラフ媒体上を、被検出物質を含む試料及び標識化した検出試薬と共に移動する。このような展開液であれば、どのようなものであってもよい。
(4)展開された移動相中の被検出物質を検出部で検出する工程
 工程(4)は、クロマトグラフ媒体部(3)上を移動相として通過した検体中の被検出物質が、抗原・抗体の特異的結合反応により、検出部(4)に保持、即ち、担持固定されている抗体と標識試薬とによってサンドイッチ状に挟まれるように特異的に反応結合して、検出部(4)が着色する工程である。
 被検出物質が存在しない場合には、試料の水分に溶解した標識試薬は、クロマトグラフ媒体部(3)上の検出部(4)を通過しても特異的結合反応が起こらないので、検出部(4)が着色しない。
 最後に、検体含有液の水分は、吸収部(5)へと移動する。
 このように、検体に含まれる被検出物質、例えば、検体含有液中のインフルエンザウィルスの有無を検査することにより、インフルエンザウィルスの感染の有無を正確に判定することができる。
 以下、本発明を実施例および比較例によりさらに説明するが、本発明は下記実施例に制限されるものではない。
 [実施例1~4]
 (1)クロマトグラフ媒体上への検出部の作製
 メンブレンとしてニトロセルロースからなるシート(ミリポア社製、商品名:HF120、300mm×25mm)を用いた。前処理としてメンブレンを0.25質量%の表1に記載の陰イオン性界面活性剤の水溶液に浸漬した後、30℃24時間で乾燥させた。
 次に、5質量%のイソプロピルアルコールを含むリン酸緩衝液(pH7.4)で1.0mg/mlの濃度になるようにマウス由来抗インフルエンザAモノクローナル抗体(第一抗体)を希釈した溶液150μLを、陰イオン性界面活性剤を含有させ乾燥されたメンブレン上の検出部位(検出ライン)に1mmの幅で塗布し、金ナノ粒子標識試薬の展開の有無や展開速度を確認するために検出部位の下流に、金ナノ粒子標識物質や被検出物質である動物肉タンパク質などと広く親和性を有するヤギ由来抗血清をリン酸緩衝液(pH7.4)で希釈した液をコントロール部位(コントロールライン)に塗布した。その後、50℃で30分間乾燥させ、室温で一晩乾燥させ、クロマトグラフ媒体を作製した。
 (2)標識物質溶液の作製
 金コロイド懸濁液(田中貴金属工業社製:LC40nm)0.5mlに、リン酸緩衝液(pH7.4)で0.05mg/mlの濃度になるように希釈したマウス由来抗インフルエンザAモノクローナル抗体(第二抗体)を0.1ml加え、室温で10分間静置した。
 次いで、1質量%の牛血清アルブミン(BSA)を含むリン酸緩衝液(pH7.4)を0.1ml加え、更に室温で10分間静置した。その後、十分撹拌した後、8000×gで15分間遠心分離を行い、上清を除去した後、1質量%のBSAを含むリン酸緩衝液(pH7.4)を0.1ml加えた。以上の手順で標識物質溶液を作製した。
 (3)試料添加部の作製
 試料添加部としてグラスファイバーからなる不織布(ミリポア社製:300mm×30mm)を、2質量%の表1に示す非イオン性界面活性剤水溶液に浸漬した後、30℃24時間で乾燥し試料添加部を作製した。作製された試料添加部中に含まれる、非イオン性界面活性剤の種類、そのHLB値および含有量を表1に示す。表1中、Tween20とはポリオキシエチレン(20)ソルビタンモノラウレートであり、Tween80とはポリオキシエチレン(20)ソルビタンモノオーレートであり、TritonX-100とはポリオキシエチレン(10)オクチルフェニルエーテルであり、いずれも和光純薬工業株式会社製である。また、NP-40とは、(オクチルフェノキシ)ポリエトキシエタノールであり、シグマアルドリッチ社製である。
 (4)免疫クロマト分析装置の作製
 上記作製した標識物質溶液300μLに300μLの10質量%トレハロース水溶液と1.8mLの蒸留水を加えたものを15mm×300mmのグラスファイバーパッド(ミリポア社製)に均一になるように添加した後、真空乾燥機にて乾燥させ、標識物質保持部材を作製した。
 次に、バッキングシートから成る基材に、上記作製したクロマトグラフ媒体、標識物質保持部材、試料を添加する部分に用いる試料添加部、展開した試料や標識物質を吸収するための吸収部としてグラスファイバー製の不織布(密度0.25g/m)を貼り合わせた。そして、裁断機で幅が5mmとなるように裁断し、免疫クロマト分析装置とした。吸収部に用いた不織物の種類および吸水部の濾水時間を表1に示す。
 濾水時間は、JIS P 3801:1995に従い測定した。
 (5)検体希釈液
 2質量%カゼイン、25mM KCl、0.095%アジ化ナトリウムを含む50mM BICINE緩衝液(pH8.5)を調製し、検体希釈液とした。
 (6)測定
 上記作製した免疫クロマトグラフィー用試験片を用いて、以下の方法で試料中のインフルエンザAウィルスの存在の有無を測定した。即ち、吸引トラップの片方の管を吸引ポンプに、もう片方の管をインフルエンザに感染していない人の鼻腔の奥部まで挿入し、吸引ポンプを陰圧にして鼻汁を採取した。採取した鼻汁を上記検体希釈液で20倍に希釈し、これを陰性検体試料とした。また、陰性検体試料に、蛋白濃度が50ng/mLとなるように市販の不活化インフルエンザAウィルスを加えたものを陽性検体試料とした。
 陰性検体試料、陽性検体試料とも150μLを免疫クロマトグラフィー用試験片の試料添加部上に載せて展開させ、10分後に目視判定をした。テストラインの赤い線を確認できるものを「+」、赤い線は確認できるが、非常に色が薄いものを「±」、赤い線を確認できないものを「-」とした。
 また、検査開始から10分の時点でのバックグラウンド着色の有無、検査開始から60分の時点での展開成分の逆流及び偽陽性の有無(陰性検体のみ)を観察した。試験の結果を表1に示す。
 バックグラウンドの着色は展開される標識物質(金ナノ粒子)に由来し、赤色の金ナノ粒子が吸収パッドに完全に吸収されると、バックグラウンドの着色が抜け、クロマトグラフ媒体本来の白色が観察されるようになる。表1では、バックグラウンドが、クロマトグラフ媒体本来の白色と同等の色であったものを○(良好)、やや赤味がかっていたものを△(やや不良)、赤味を帯びていたものを×(不良)で示す。
 クロマトグラフ媒体は乾燥状態と湿潤状態では色調が異なって見えるため、展開成分の逆流は、クロマトグラフ媒体の色調を目視で観察することにより判定した。展開成分の逆流の評価は、60分後のバックグラウンド着色が10分後よりも悪化している(赤みが増している)ものを有りと評価し、それ以外を無しと評価した。
 偽陽性の有無は、検査開始から60分の時点で、クロマトグラフ媒体の検出部に標識物質(金ナノ粒子)に由来する赤いラインが生じるか否かを観察することにより判定した。用いた試料はPCR法による検査で陰性と判定された試料であり、イムノクロマト法による検査キットを用いた試験でも、検査開始から15分の時点では、検出部に陽性を示す赤いラインは観察されなかった。しかしながら、展開成分の逆流がある場合には、検査開始から60分の時点で、検出部に赤いラインが生じることがあり、この場合に偽陽性有りと判定した。結果を表1に示す。
 [実施例5]
 (1)クロマトグラフ媒体上への検出部の作製
 検出部については、実施例1~4と同様に作製した。
 (2)標識物質溶液の作製
 標識物質溶液については、実施例1~4と同様に作製した。
 (3)試料添加部の作製
 試料添加部については、実施例1~4と同様に作製した。
 (4)免疫クロマト分析装置の作製
 免疫クロマト分析装置については、実施例1~4と同様に作製した。
 (5)検体希釈液
 2質量%カゼイン、25mM KCl、0.095%アジ化ナトリウムを含む50mM BICINE緩衝液(pH8.5)を調製し、2質量%Tween80を記載含有率となるよう混合し検体希釈液を作製した。
 (6)測定
 測定については、実施例1~4と同様に実施し、評価した。結果を表1に示す。
 [実施例6~8]
 (1)クロマトグラフ媒体上への検出部の作製
 検出部については、実施例1~5と同様に作製した。
 (2)標識物質溶液の作製
 標識物質溶液については、実施例1~5と同様に作製した。
 (3)試料添加部の作製
 試料添加部としてグラスファイバーからなる不織布(ミリポア社製:300mm×30mm)を使用した。
 (4)免疫クロマト分析装置の作製
 免疫クロマト分析装置については、実施例1~5と同様に作製した。
 (5)検体希釈液
 2質量%カゼイン、25mM KCl、0.095%アジ化ナトリウムを含む50mM BICINE緩衝液(pH8.5)を調製し、表1に記載の非イオン性界面活性剤をそれぞれ記載含有率となるよう混合し検体希釈液を作製した。表1中、IGAPEL CA-520とは、ポリオキシエチレン(5)イソオクチルフェニルエーテルであり、シグマアルドリッチ社製である。
 (6)測定
 測定については、実施例1~5と同様に実施し、評価した。結果を表1に示す。
 [比較例1、2]
 (1)クロマトグラフ媒体上への検出部の作製
 検出部については、実施例と同様に作製した。
 (2)標識物質溶液の作製
 標識物質溶液については、実施例と同様に作製した。
 (3)試料添加部の作製
 試料添加部としてグラスファイバーからなる不織布(ミリポア社製:300mm×30mm)を使用した。
 (4)免疫クロマト分析装置の作製
 上記作製した標識物質溶液300μLに300μLの10質量%トレハロース水溶液と1.8mLの蒸留水を加えたものを15mm×300mmのグラスファイバーパッド(ミリポア社製)に均一になるように添加した後、真空乾燥機にて乾燥させ、標識物質保持部材を作製した。
 次に、バッキングシートから成る基材に、上記作製したクロマトグラフ媒体、標識物質保持部材、試料を添加する部分に用いる試料添加部、展開した試料や標識物質を吸収するための吸収部として不織布を貼り合わせた。そして、裁断機で幅が5mmとなるように裁断し、免疫クロマト分析装置とした。吸収部に用いた不織物の種類および吸収部の濾水時間を表1に示す。
 濾水時間は、JIS P 3801:1995に従い測定した。
 (5)検体希釈液
 2質量%カゼイン、25mM KCl、0.095%アジ化ナトリウムを含む50mM BICINE緩衝液(pH8.5)を調製し、表1に記載の非イオン性界面活性剤をそれぞれ記載含有率となるよう混合し検体希釈液を作製した。
 (6)測定
 測定については、実施例と同様に実施し、評価した。結果を表1に示す。
 [比較例3]
 (1)クロマトグラフ媒体上への検出部の作製
 検出部については、実施例と同様に作製した。
 (2)標識物質溶液の作製
 標識物質溶液については、実施例と同様に作製した。
 (3)試料添加部の作製
 試料添加部としてグラスファイバーからなる不織布(ミリポア社製:300mm×30mm)を使用した。
 (4)免疫クロマト分析装置の作製
 免疫クロマト分析装置については、実施例と同様に作製した。
 (5)検体希釈液
 2質量%カゼイン、25mM KCl、0.095%アジ化ナトリウムを含む50mM BICINE緩衝液(pH8.5)を調製し、検体希釈液を作製した。
 (6)測定
 測定については、実施例と同様に実施し、評価した。結果を表1に示す。

 表1に示すように、実施例1~8において、吸収部にグラスファイバーを用い、非イオン性界面活性剤存在下で検体および標識物質を移動相としてクロマトグラフ媒体部に展開させたところ、試料の液戻りが確認されず、その結果、偽陽性の反応は見られなかった。
 一方、比較例1では、吸収部にコットンリンターを用いており、比較例2では、吸収部にセルロース繊維を使用しており、その結果、試料の液戻りが確認され、偽陽性の反応が見られた。
 また、比較例3では、吸収部にグラスファイバーを用いているが、非イオン性界面活性剤の非存在下で検体および標識物質を移動相としてクロマトグラフ媒体部に展開させた結果、試料の液戻りが確認されなかったが、偽陽性の反応が見られた。
 これは、非イオン性界面活性剤が存在しないため展開速度が減少し、展開成分の剥離速度も減少するため、展開成分の逆流は発生しないが、クロマトグラフ媒体上に標識試薬が留まってしまうため、10分後と60分後のバックグラウンドの着色に変化が見られず、偽陽性の反応が見られたと考えられる。
 以上の結果から、非イオン性界面活性剤存在下で検体および標識物質を移動相としてクロマトグラフ媒体部に展開させると、試料の展開速度を上昇させることができ、また、試料の展開速度が上昇したとしても、吸収部にグラスファイバーを用いることによって、展開成分の液戻りを低減し、偽陽性を回避することができることが分かった。
 本発明を特定の態様を用いて詳細に説明したが、本発明の意図と範囲を離れることなく様々な変更および変形が可能であることは、当業者にとって明らかである。なお本出願は、2014年4月4日付で出願された日本特許出願(特願2014-077861)に基づいており、その全体が引用により援用される。

Claims (11)

  1.  試料添加部、標識物質保持部、検出部が担持されたクロマトグラフ媒体部およびグラスファイバーからなる吸収部を含む免疫クロマト分析装置を用いて検体に含まれる被検出物質を検出する方法であって、以下の工程(1)~(4)を含む免疫クロマト分析方法。
    (1)検体希釈液により検体を希釈した検体含有液を試料添加部に添加する工程
    (2)標識物質保持部に保持されている標識物質により被検出物質を認識させる工程
    (3)非イオン性界面活性剤存在下で検体および標識物質を移動相としてクロマトグラフ媒体部に展開させる工程
    (4)展開された移動相中の被検出物質を検出部で検出する工程
  2.  前記試料添加部および前記検体希釈液の少なくとも一方に非イオン性界面活性剤を含有する請求項1に記載の方法。
  3.  前記試料添加部における非イオン性界面活性剤の含有量が1cmあたり0.05~5mgである請求項2に記載の方法。
  4.  前記検体希釈液における非イオン性界面活性剤の含有量が0.05~10質量%である請求項2に記載の方法。
  5.  前記非イオン性界面活性剤のHLB値が10以上である請求項1~4のいずれか1項に記載の方法。
  6.  前記クロマトグラフ媒体部が陰イオン性界面活性剤を含む請求項1~5のいずれか1項に記載の方法。
  7.  前記クロマトグラフ媒体部における陰イオン性界面活性剤の含有量が、0.02~4質量%である請求項6に記載の方法。
  8.  前記クロマトグラフ媒体部がニトロセルロースからなる請求項1~7のいずれか1項に記載の方法。
  9.  前記吸収部の濾水時間が20秒~110秒であり、かつ密度が150~500mg/mである請求項1~8のいずれか1項に記載の方法。
  10.  試料添加部、標識物質保持部、検出部が担持されたクロマトグラフ媒体部および吸収部を順次含む免疫クロマト分析装置であって、該試料添加部は非イオン性界面活性剤を乾燥保持し、該吸収部はグラスファイバーからなる免疫クロマト分析装置。
  11.  試料添加部、標識物質保持部、検出部が担持されたクロマトグラフ媒体部および吸収部を順次含む免疫クロマト装置と、検体に含まれる被検出物質を希釈する検体希釈液とを含む免疫クロマト分析キットであって、該吸収部がグラスファイバーからなり、該検体希釈液が非イオン性界面活性剤を含む免疫クロマト分析キット。
