상기 목적을 달성하기 위한 하나의 양태로서, 본 발명은 채송화(Portulaca grandiflora Hook.) 추출물 또는 이의 분획물을 유효성분으로 포함하는 신경염증 또는 퇴행성 뇌신경 질환의 예방 또는 치료용 약학적 조성물을 제공한다.
구체적으로, 본 발명의 신경염증 또는 퇴행성 뇌신경 질환의 예방 또는 치료용 약학적 조성물은 채송화 추출물 및/또는 이의 분획물을 포함할 수 있다.
본 발명에서 "채송화(Portulaca grandiflora Hook.)"는 쇠비름과에 속하는 일년생 초본식물로서, 원산지가 남아메리카이고 전 세계적으로 40여종이 분포되어 있다. 줄기는 육질이 많고 원주형으로 홍색을 띠고 옆으로 누우며 가지를 쳐서 뻗는데 큰 것은 30㎝ 가량 자란다. 잎은 살이 많고 선상이며 털이 없고 잎겨드랑이에는 흰털이 무더기로 나와 있다. 꽃은 줄기의 끝에서 한 송이 또는 두 송이 이상 달리기도 한다. 여름에 홍색 또는 백색으로 피는데 꽃받침은 2편이고 넓은 달걀 모양이며 화판은 5개이고 넓은 도란형으로 끝이 뾰족하다. 또한 채송화의 전초를 반지련이라고 하여 약으로 사용되기도 한다.
본 발명에서 "채송화 추출물"은 채송화를 추출하여 수득한 추출물이다. 상기 채송화 추출물은 채송화 분쇄물을 건조 중량의 약 2 내지 20배, 바람직하게는 약 3 내지 5배에 달하는 부피의 물, 메탄올, 에탄올 및 부탄올 등과 같은 탄소수 1(C1) 내지 4(C4)의 저급 알콜의 극성 용매 또는 이들의 약 1:0.1 내지 1:10의 혼합비를 갖는 혼합용매를 용출 용매로써 사용하고, 추출 온도는 20 내지 100℃, 바람직하게는 실온에서, 추출 기간은 약 12시간 내지 4일, 바람직하게는 3일 동안 열수 추출, 냉침 추출, 환류 냉각 추출 또는 초음파 추출 등의 추출방법을 사용하여 추출할 수 있으나, 신경염증 또는 퇴행성 뇌신경 질환의 예방 또는 치료 활성이 있는 물질을 추출하는 방법이라면 제한없이 이용될 수 있다. 바람직하게는 냉침추출로 1회 내지 5회 연속 추출하여 감압여과하고, 그 여과추출물을 진공회전농축기로 20 내지 100℃, 바람직하게는 실온에서 감압 농축하여 물, 저급 알콜 또는 이들의 혼합용매에 가용한 채송화 조추출물을 수득한 결과물이 될 수 있으나, 본 발명의 신경염증 또는 퇴행성 뇌신경 질환의 예방 또는 치료 활성을 나타낼 수 있는 한, 이에 제한되지는 않고, 추출액, 추출액의 희석액 또는 농축액, 추출액을 건조하여 얻어지는 건조물, 또는 이들 조정제물 또는 정제물을 모두 포함할 수 있다. 상기 채송화 추출물은 천연, 잡종, 변종식물의 다양한 기관으로부터 추출될 수 있고, 예를 들어, 뿌리, 지상부, 줄기, 꽃, 가지, 잎, 열매 또는 식물 조직 배양물 등으로부터 추출 가능하다.
본 발명에서 "분획물"은 다양한 구성성분을 포함하는 혼합물로부터 특정 성분 또는 특정 그룹을 분리하는 분획방법에 의하여 얻어진 결과물이다. 본 발명의 채송화 분획물은 채송화 추출물을 현탁한 후, 물, 메탄올, 에탄올 등의 극성 용매 또는 헥산, 에틸아세테이트와 같은 비극성 용매를 사용하여 분획함으로써 극성 용매 분획물과 비극성 용매 분획물을 각각 수득할 수 있다. 구체적으로, 채송화 조추출물을 증류수 등에 현탁한 후, 현탁액의 약 1 내지 100배, 바람직하게는 약 1 내지 5배 부피의 물, 탄소수 1(C1) 내지 4(C4)의 알코올, 클로로포름, 에틸아세테이트, 헥산, 부탄올 또는 이들의 혼합용매와 같은 극성 또는 비극성 용매를 가하여 1회 내지 10회, 바람직하게는 2회 내지 5회에 걸쳐 극성 또는 비극성 용매 가용층을 추출, 분리하여 수득할 수 있다.
또한 본 발명의 분획물은 추가로 통상의 분획 공정을 수행하여 수득한 것일 수도 있다(Harborne J.B. Plant Pathology, 1998, 3rd Ed. p6-7). 예컨대, 본 발명에 따른 채송화 추출물을 일정한 분자량 컷-오프 값을 갖는 한외 여과막을 통과시켜 얻은 분획물, 다양한 크로마토그래피 (크기, 전하, 소수성 또는 친화성에 따른 분리를 위해 제작된 것)에 의한 분리 등으로 추가적으로 실시된 다양한 정제 방법을 통해 얻어진 활성분획물도 본 발명의 채송화 분획물에 포함된다.
상기 활성분획물은 분획물로부터 보다 높은 생리활성 등을 가지는 분획을 분리한 것으로, 활성분획 또는 유효분획물이라고도 한다. 계통분획과 같은 통상의 분획과정을 통해 수득한 여러 성분이 혼합되어 있는 분획물 가운데 농도구배 컬럼 크로마토그래피 등을 통하여 활성성분의 성질에 따라 분리해냄으로써 보다 강한 활성을 갖는 특정 활성분획물을 제조할 수 있다. 상기 컬럼 크로마토그래피는 실리카겔, 세파덱스, LH-20, ODS 겔, RP-18, 폴리아미드, 도요펄(Toyopearl) 및 XAD 수지로 이루어진 군으로부터 선택된 충진제를 이용하는 컬럼 크로마토그래피를 수행하여 활성분획물을 분리 및 정제할 수 있고, 컬럼 크로마토그래피는 필요에 따라 적절한 충진제를 선택하여 수차례 실시할 수 있으나, 이에 제한되는 것은 아니다. 상기 크로마토그래피를 사용함에 있어 용출용매, 용출 속도 및 용출시간은 당업계에서 일반적으로 사용하는 용매, 속도 또는 시간을 적용할 수 있다.
본 발명의 일 실시예에서는 상기 얻어진 채송화 메탄올 추출물에 증류수를 가하여 현탁한 후 각각 에틸아세테이트, 부탄올 또는 물을 가하여 에틸아세테이트 분획물, 부탄올 분획물 또는 물 분확물을 수득하였다(실시예 1 및 실시예 2).
본 발명에 따른 채송화 추출물 또는 이의 분획물은 염증관련 인자로 알려진 일산화질소(NO), 프로스타글란딘(PGE2), iNOS 및/또는 COX-2 유전자 또는 단백질의 발현을 억제함으로써 신경염증 또는 이로부터 발병할 수 있는 퇴행성 뇌신경 질환의 예방 또는 치료용도로 사용될 수 있다.
본 발명에서 "신경염증"은 신경계, 즉 신경세포, 신경조직 등에 발생하는 염증성 반응을 총칭한다. 중추신경계에 존재하는 면역세포인 소신경교세포가 다양한 외인성, 내인성 물질로 인해 활성화되어, 염증성 사이토카인인 TNF-α 및 IL-1β, 일산화질소, 프로스타글란딘, 초과산화물 등의 물질을 생산, 방출하는 현상을 포함할 수 있다. 이러한 물질들의 생성은 단기적으로는 면역반응을 유발하지만, 그 과도한 생산이나 지속적인 생산은 근접한 신경세포들의 사멸을 유도하여 결국 신경퇴행을 유발한다고 알려져 있다.
본 발명의 일 실험예에서는 채송화의 추출물 또는 이의 분획물이 신경세포에 대한 독성을 갖지 않으면서, 염증관련 인자로 알려진 NO, PGE2, iNOS 및 COX-2의 단백질 또는 mRNA의 발현수준을 억제하였음을 확인함으로써 신경염증의 예방 또는 치료효과를 확인하였다(실험예 2).
또한, 본 발명에 따른 채송화 추출물 또는 이의 분획물은 알츠하이머 관련 효소로 알려진 β-CTF 및 베타-세크리테아제(BACE1)의 발현을 억제함으로써 퇴행성 뇌신경 질환의 예방 또는 치료용도로 사용될 수 있다.
본 발명에서 "퇴행성 뇌신경 질환"은 중추신경계의 신경세포에 퇴행성 변화가 나타나면서 여러 가지 증상을 유발하는 질환을 총칭하며, 구체적으로 인지 기능, 학습 또는 기억력이 손상되거나, 신경염증 반응을 동반하는뇌신경 질환을 포함할 수 있다. 본 발명에 따른 대표적인 퇴행성 뇌신경 질환에는 치매(dementia), 알츠하이머병(Alzheimer's disease), 파킨슨병(Parkinson's disease), 헌팅톤병(Huntington's disease), 루게릭병(amyotrophic lateral sclerosis, ALS), 크로이츠펠트 야콥병(Creutzfeldt-Jakob disease, CJD), 뇌졸중(Stroke), 다발성 경화증(Multiple sclerosis), 인지 장애, 학습 장애, 기억력 손상 등이 있다.
이 중에서 알츠하이머병(Alzheimer's disease, AD)은 노인성 치매 중에서도 가장 중요하게 대두되고 있는 질환으로, 아밀로이드베타 (amyloid beta, Aβ)의 뇌내 축적과 그로 인한 신경독성이 발병의 중요한 원인으로 알려져 있다. 상기 Aβ는 아밀로이드 전구체(amyloid precursor protein, APP)가 막단백 가수분해 효소인 베타-세크리테아제 1(BACE1)와 감마-세크리테아제의 연속적인 작용으로 만들어지는 것으로 알려져 있으며, 따라서 BACE1 단백질의 발현 억제를 통해 알츠하이머 병을 예방 또는 치료할 수 있음은 자명하다.
본 발명의 일 실험예에서는 알츠하이머 관련 유전자로 알려진 β-CTF(β-secretase-cleaved carboxyl-terminal fragment)와 BACE1(Beta-secretase 1, beta-site APP-cleaving enzyme 1)의 단백질 또는 mRNA의 발현수준 또한 억제시킴을 확인함으로써 퇴행성 뇌신경 질환이 예방 또는 치료효과를 확인하였다(실험예 3).
본 발명에서 "예방"은 상기 조성물의 투여로 신경염증 또는 퇴행성 뇌신경 질환의 발병을 억제 또는 지연시키는 모든 행위를 말하며, "치료"는 상기 조성물에 의해 신경염증 또는 퇴행성 뇌신경 질환에 의한 증세가 호전되거나 이롭게 변경되는 모든 행위를 말한다.
본 발명의 채송화 추출물 또는 이의 분획물을 포함하는 약학적 조성물은 약학적 조성물의 제조에 통상적으로 사용하는 적절한 담체, 부형체 또는 희석제를 추가로 포함할 수 있다. 이때, 상기 조성물에 포함되는 채송화 추출물 또는 이의 분획물은 특별히 이에 제한되지 않으나, 조성물 총 중량에 대하여 0.001 중량% 내지 99 중량%로, 바람직하게는 0.01 중량% 내지 50 중량%를 포함할 수 있다.
상기 약학적 조성물은 정제, 환제, 산제, 과립제, 캡슐제, 현탁제, 내용액제, 유제, 시럽제, 멸균된 수용액, 비수성용제, 현탁제, 유제, 동결 건조제 및 좌제로 이루어진 군으로부터 선택되는 어느 하나의 제형을 가질 수 있으며, 경구 또는 비경구의 여러 가지 제형일 수 있다. 제제화할 경우에는 보통 사용하는 충진제, 증량제, 결합제, 습윤제, 붕해제, 계면활성제 등의 희석제 또는 부형제를 사용하여 조제된다. 경구투여를 위한 고형제제에는 정제, 환제, 산제, 과립제, 캡슐제 등이 포함되며, 이러한 고형제제는 하나 이상의 화합물에 적어도 하나 이상의 부형제 예를 들면, 전분, 탄산칼슘, 수크로오스(sucrose) 또는 락토오스(lactose), 젤라틴 등을 섞어 조제될 수 있다. 또한, 단순한 부형제 이외에 스테아린산 마그네슘, 탈크 등과 같은 윤활제들도 사용될 수 있다. 경구투여를 위한 액상제제로는 현탁제, 내용액제, 유제, 시럽제 등이 해당되는데 흔히 사용되는 단순희석제인 물, 리퀴드 파라핀 이외에 여러 가지 부형제, 예를 들면 습윤제, 감미제, 방향제, 보존제 등이 포함될 수 있다. 비경구투여를 위한 제제에는 멸균된 수용액, 비수성용제, 현탁제, 유제, 동결건조 제제, 좌제가 포함된다. 비수성용제, 현탁용제로는 프로필렌글리콜(propylene glycol), 폴리에틸렌 글리콜, 올리브 오일과 같은 식물성 기름, 에틸올레이트와 같은 주사 가능한 에스테로 등이 사용될 수 있다. 좌제의 기제로는 위텝솔(witepsol), 마크로골, 트윈(tween) 61, 카카오지, 라우린지, 글리세로젤라틴 등이 사용될 수 있다.
또 하나의 양태로서, 본 발명은 상기 약학적 조성물을 신경염증 또는 퇴행성 뇌신경 질환의 의심 개체에 투여하는 단계를 포함하는, 신경염증 또는 퇴행성 뇌신경 질환의 예방 또는 치료방법을 제공한다.
본 발명에서 상기 신경염증 또는 퇴행성 뇌신경 질환의 의심 개체는 상기 질환이 발병하였거나 발병할 수 있는 인간을 포함한 모든 동물을 의미하며, 본 발명의 약학적 조성물을 신경염증 또는 퇴행성 뇌신경 질환의 의심 개체에 투여함으로써, 개체를 효율적으로 치료할 수 있다. 상기 약학적 조성물 및 신경염증 또는 퇴행성 뇌신경 질환에 대해서는 상기에서 설명한 바와 같다.
본 발명에서 "투여"는 어떠한 적절한 방법으로 신경염증 또는 퇴행성 뇌신경 질환의 의심 개체에게 본 발명의 약학적 조성물을 도입하는 것으로서, 투여 경로는 목적 조직에 도달할 수 있는 한 경구 또는 비경구의 다양한 경로를 통하여 투여될 수 있다.
본 발명의 약학적 조성물은 약학적으로 유효한 양으로 투여할 수 있다.
본 발명에서 "약학적으로 유효한 양"은 의학적 치료에 적용 가능한 합리적인 수혜/위험 비율로 질환을 치료하기에 충분한 양을 의미하며, 유효 용량 수준은 개체 종류 및 중증도, 연령, 성별, 질병의 종류, 약물의 활성, 약물에 대한 민감도, 투여 시간, 투여 경로 및 배출 비율, 치료기간, 동시 사용되는 약물을 포함한 요소 및 기타 의학 분야에 잘 알려진 요소에 따라 결정될 수 있다. 본 발명의 조성물은 개별 치료제로 투여하거나 다른 치료제와 병용하여 투여될 수 있고 종래의 치료제와는 순차적 또는 동시에 투여될 수 있다. 그리고 단일 또는 다중 투여될 수 있다. 상기 요소를 모두 고려하여 부작용없이 최소한의 양으로 최대 효과를 얻을 수 있는 양을 투여하는 것이 중요하며, 당업자에 의해 용이하게 결정될 수 있다.
본 발명의 약학적 조성물은 신경염증 또는 퇴행성 뇌신경 질환을 목적으로 하는 개체이면 특별히 한정되지 않고, 어떠한 것이든 적용가능하다. 예를 들면, 원숭이, 개, 고양이, 토끼, 모르모트, 랫트, 마우스, 소, 양, 돼지, 염소 등과 같은 비인간동물, 인간, 조류 및 어류 등 어느 것이나 사용할 수 있으며, 상기 약학적 조성물은 비 경구, 피하, 복강 내, 폐 내 및 비강 내로 투여될 수 있고, 국부적 치료를 위해, 필요하다면 병변 내 투여를 포함하는 적합한 방법에 의하여 투여될 수 있다. 본 발명의 상기 약학적 조성물의 바람직한 투여량은 개체의 상태 및 체중, 질병의 정도, 약물형태, 투여경로 및 기간에 따라 다르지만, 당업자에 의해 적절하게 선택될 수 있다. 예를 들어, 경구, 직장 또는 정맥, 근육, 피하, 자궁내 경막 또는 뇌혈관 내 주사에 의해 투여될 수 있으나, 이에 제한되는 것은 아니다.
적합한 총 1일 사용량은 올바른 의학적 판단범위 내에서 처치에 의해 결정될 수 있으며, 일반적으로 0.001 내지 1000 mg/kg의 양, 바람직하게는 0.05 내지 200 mg/kg, 보다 바람직하게는 0.1 내지 100 mg/kg의 양을 일일 1회 내지 수회로 나누어 투여할 수 있다.
또 다른 양태로서, 본 발명은 채송화(Portulaca grandiflora Hook.) 추출물 또는 이의 분획물을 유효성분으로 포함하는 신경염증 또는 퇴행성 뇌신경 질환의 예방 또는 개선용 식품 조성물을 제공한다.
상기 채송화, 이의 추출물, 분획물 및 신경염증 또는 퇴행성 뇌신경 질환에 대해서는 상기에서 설명한 바와 같다.
본 발명에서 "개선"은 채송화 추출물 또는 이의 분획물을 유효성분으로 포함하는 조성물을 이용하여 예방 또는 치료되는 신경염증 또는 퇴행성 뇌신경 질환과 같은 질환의 의심 및 발병 개체의 증상이 호전되거나 이롭게 되는 모든 행위를 말한다.
구체적으로, 본 발명의 채송화 추출물 또는 이의 분획물을 신경염증 또는 퇴행성 뇌신경 질환의 예방 또는 개선을 목적으로 식품 조성물에 첨가할 수 있다.
본 발명의 식품 조성물은 환제, 분말, 과립, 침제, 정제, 캡슐 또는 액제 등의 형태를 포함할 수 있으며, 본 발명의 채송화 추출물 또는 이의 분획물을 첨가할 수 있는 식품의 종류에는 별다른 제한이 없으며, 예를 들어 각종 음료, 껌, 차, 비타민 복합제, 건강보조 식품류 등이 있다.
상기 식품 조성물에는 채송화 추출물 또는 이의 분획물 이외에도 다른 성분을 추가할 수 있으며, 그 종류는 특별히 제한되지 않는다. 예를 들어, 통상의 식품과 같이 여러 가지 생약 추출물, 식품학적으로 허용가능한 식품보조첨가제 또는 천연 탄수화물 등을 추가 성분으로서 함유할 수 있으며, 이에 제한되지 않는다.
상기 "식품보조첨가제"는 식품에 보조적으로 첨가될 수 있는 구성요소로서, 각 제형의 건강기능식품을 제조하는데 첨가될 수 있으며 당업자가 적절히 선택하여 사용할 수 있다. 식품보조첨가제의 예로는 여러 가지 영양제, 비타민, 광물(전해질), 합성 풍미제 및 천연 풍미제 등의 풍미제, 착색제 및 충진제, 펙트산 및 그의 염, 알긴산 및 그의 염, 유기산, 보호성 콜로이드 증점제, pH 조절제, 안정화제, 방부제, 글리세린, 알코올, 탄산음료에 사용되는 탄산화제 등이 포함되지만, 상기 예들에 의해 본 발명의 식품보조첨가제의 종류가 제한되는 것은 아니다.
상기 천연 탄수화물의 예는 포도당, 과당 등의 단당류; 말토스, 수크로스 등의 이당류; 및 덱스트린, 시클로덱스트린 등의 다당류와, 자일리톨, 소르비톨, 에리트리톨 등의 당알콜이 있으며, 상기한 것 이외의 향미제로서 천연 향미제(타우마틴 등), 스테비아 추출물(레바우디오시드 A, 글리시르히진 등) 및 합성 향미제(사카린, 아스파르탐 등)를 유리하게 사용할 수 있다.
본 발명의 식품 조성물에는 건강기능성 식품이 포함될 수 있다. 상기 "건강기능성 식품"은 인체에 유용한 기능성을 가진 원료나 성분을 사용하여 정제, 캅셀, 분말, 과립, 액상 및 환 등의 형태로 제조 및 가공한 식품을 말한다. 여기서 기능성이라 함은 인체의 구조 및 기능에 대하여 영양소를 조절하거나 생리학적 작용 등과 같은 보건 용도에 유용한 효과를 얻는 것을 말한다. 본 발명의 건강기능성 식품은 당업계에서 통상적으로 사용되는 방법에 의하여 제조가능하며, 상기 제조시에는 당업계에서 통상적으로 첨가하는 원료 및 성분을 첨가하여 제조할 수 있다. 또한 일반 약품과는 달리 식품을 원료로 하여 약품의 장기 복용 시 발생할 수 있는 부작용 등이 없는 장점이 있고, 휴대성이 뛰어날 수 있다.
유효성분의 혼합양은 사용 목적(예방, 건강 또는 치료적 처치)에 따라 적합하게 결정될 수 있다. 일반적으로, 식품의 제조 시에 본 발명의 채송화 추출물 또는 이의 분획물은 원료 조성물 중 1 내지 50 중량%, 바람직하게는 5 내지 10 중량%의 양으로 첨가될 수 있으나, 이에 제한되는 것은 아니다. 그러나 건강 및 위생을 목적으로 하거나 또는 건강 조절을 목적으로 하는 장기간의 섭취의 경우에는 상기 양은 상기 범위 이하로도 사용될 수 있다.
상기 식품의 종류에는 특별한 제한은 없다. 상기 물질을 첨가할 수 있는 식품의 예로는 육류, 소세지, 빵, 쵸코렛, 캔디류, 스넥류, 과자류, 피자, 라면, 기타 면류, 껌류, 아이스크림류를 포함한 낙농제품, 각종 스프, 음료수, 차, 드링크제, 알코올 음료 및 비타민 복합제 등이 있으며, 통상적인 의미에서의 건강기능성 식품을 모두 포함할 수 있다.
또 하나의 양태로서, 본 발명은 채송화(Portulaca grandiflora Hook.) 추출물 또는 이의 분획물을 유효성분으로 포함하는 학습 능력 또는 기억력 증진 또는 개선용 조성물을 제공한다.
상기 채송화, 이의 추출물, 분획물에 대해서는 상기에서 설명한 바와 같다.
본 발명의 일 실험예에서는 마우스 모델을 대상으로 채송화 추출물 또는 이의 분획물을 투여한 결과, Y-미로실험에서의 작업기억 증진효과 및 수동회피실험에서의 기억력 증진효과를 확인하였다. 따라서 본 발명의 채송화 추출물 또는 이의 분획물은 학습 능력 또는 기억력 증진용도로 사용될 수 있으며, 상기 조성물은 약학적 조성물, 의약외품 조성물 또는 식품 조성물일 수 있다.
또한, 본 발명은 신경염증 또는 퇴행성 뇌신경 질환의 예방, 개선 또는 치료를 위한 약학적 조성물 또는 식품 조성물의 제조를 위한, 채송화(Portulaca grandiflora Hook.) 추출물 또는 이의 분획물의 용도를 제공한다.
또한, 본 발명은 학습 능력 또는 기억력 증진 또는 개선을 위한 식품 조성물의 제조를 위한, 채송화(Portulaca grandiflora Hook.) 추출물 또는 이의 분획물의 용도를 제공한다.
또한, 본 발명은 채송화(Portulaca grandiflora Hook.) 추출물 또는 이의 분획물을 이를 필요로 하는 개체에 투여함으로써 개체의 학습 능력 또는 기억력을 증진 또는 개선시키는 방법을 제공한다.
이하, 실시예를 통해 본 발명의 구성 및 효과를 보다 더 구체적으로 설명하고자 하나, 이들 실시예는 본 발명의 예시적인 기재일 뿐, 본 발명의 범위가 이들 실시예에만 한정되는 것은 아니다.
1. 약물 및 시약
Amyloid beta25 -35 (Aβ25 -35), Dimethyl sulfoxide (DMSO), 30% 과산화수소(H2O2), lipopolysaccharide, phosphoric acid, poly-D-lysine, 3-(4,5-dimethyl thiazol-2-yl)-2,5-diphenyl tetrazolium bromide (MTT), N-(1-naphthyl)ethylenediamine dihydrochloride), sulphanilamide, Tween-20, scopolamine 및 anti-β-actin 항체는 Sigma Chemical Co. (St. Louis, MO, USA)의 것을 사용하였다. Dulbecco's modified Eagle's medium(DMEM)는 Hyclone (Logan, UT, USA)사의 것을 사용하였다. Fetal bovine serum (FBS), 0.25% trypsin-EDTA 및 penicillin/streptomycin mixture는 GIBCO-BRL (Grand Island, NA, USA)사의 것을 사용하였다. Rabbit anti-rabbit horseradish peroxidase-linked IgG 항체들은 Cell Signaling (Boston, MA, USA)사의 것을 사용하였다. Rabbit anti-APP, rabbit anti-BACE1, rabbit anti-COX-2 그리고 rabbit anti-iNOS 항체들은 Epitomics (Burlingame, CA, USA), TRIZOL, cDNA 합성 kit는 Invitrogen (MolecularProbes, OR, USA)사의 것을 사용하였다. PGE2 ELISA kit (Cayman, MI, USA)것을 사용하였다. 그 밖에 실험에 사용된 시약들은 최상의 제품의 것을 구매하여 사용하였다.
2. 실험동물의 준비
4주령의 ICR계 수컷 마우스 (25-30 g)를 코아텍 (경기, 대한민국)에서 공급받아 성균관대학교 약학대학의 동물 사육실에서 1주일 이상 사육하여 적응시켜 사용하였으며, 물과 사료는 자유롭게 섭취하도록 하였고, 온도 (23 ± 2 ℃), 습도 (55 ± 10 %) 및 명암주기 (12 시간)는 자동으로 조절되도록 하였다.
<Aβ25 -35 유도 치매모델 제작>
25-35를 최종 농도 1 mM의 saline에 녹여 37℃에 5일 동안 활성화시킨 후, 상기 4주령의 ICR계 수컷 마우스 (25-30 g)를 엔토발로 마취 후, 입체정위기구 (stereotaxic apparatus)를 이용하여 좌측 뇌실에 6 nmol/3㎕의 Aβ25-35를 주사하였다. 주사 후, 회복을 위해서 37℃ 이상의 보온 하에 실험동물을 안정화시켰다.
3. 세포배양
인간신경아세포종 SH-SY5Y 세포, SH-SY5Y swedish 형질전환 세포 및 마우스 BV-2 소교세포는 불활성화시킨 10% 태아우혈청(fetal bovine serum, FBS)과 항생제가 함유된 DMEM(dulbecco's modified eagle's medium, Hyclone, Thermo, USA) 배지 하에서 배양하였다. 배양기를 37℃ 온도로 유지했고, 95% 공기와 5% CO2가 혼합된 기체를 계속 공급하여 세포배양의 적절한 조건을 갖추었다. 세포는 6, 24, 그리고 96-웰 플레이트에 2.5 × 104, 5 × 105, 그리고 1 × 106세포를 실험 24시간 전에 배양하였다. 과산화수소는 400 μM로 결정하였으며 LPS는 100 ng/ml로 결정하였다. 채송화 메탄올 조추출물은 100% DMSO에 녹였으며, 최종농도 0.1% 이하로 사용하였다.
4. 통계처리
모든 실험 결과는 ANOVA (one way analysis of variance)를 이용하여 통계 처리하였고, 유의성이 검증될 경우 Newman-Keuls test를 사용하여 p<0.05 수준 이하에서 유의성 검정을 실시하였다.
실시예 1: 채송화 메탄올 추출물의 제조
중국 운남성에서 채취한 채송화의 음건된 것을 (분양번호, FBM016-068) 한국 생명공학연구원 해외생물소재허브센터에서 분양하여 사용하였다. 건조된 채송화 300g를 완전히 건조시킨 후 분쇄하여 채송화 300g을 95% 메탄올 3L에 85℃ 온도에서 3번 반복하여 추출한 다음 이를 감압농축기 (EYELA사, N-1000, 일본)로 감압농축한 후, 동결 건조하여 채송화 메탄올 조추출물 90g을 수득하였다.
실시예 2: 채송화 분획물의 제조
실시예 2-1: 채송화 에틸아세테이트 분획물의 제조
상기 실시예 1과 동일한 방법으로 채송화 300 g에서 얻은 채송화 메탄올 조추출물 중 36.3 g을 물 1L에 현탁한 후 에틸아세테이트(EtAc) 1L에 2회 걸쳐 추출하여 에틸아세테이트 가용 분획물 8 g을 수득하였다.
실시예 2-2: 채송화 부탄올 분획물의 제조
상기 실시예 2-1에서 분획하고 난 물층에 다시 부탄올 (BuOH) 1L에 2회 걸쳐 추출하여 부탄올 가용 분획물 5.79 g을 수득하였다.
실시예 2-3: 채송화 물 분획물의 제조
상기 실시예 2-2에서 얻은 채송화 부탄올 가용 분획물을 분획하고 난 물층을 농축하여 물 분획물 20.55 g을 수득하였다.
상기 채송화 추출물 및 분획물들은 각각 열수 처리하여 감압 농축하여 실험에 사용하였다.
실험예 1: 채송화 추출물의 신경세포 보호 효과
채송화 추출물의 처리에 의한 신경세포(SH-SY5Y) 생존율을 측정하기 위해 MTT 환원 어세이를 사용하였다. 실험을 수행한 96-웰 플레이트에 MTT 용액을 최종농도 0.5㎎/㎖가 되도록 각 웰에 넣었다. 배양기에서 2시간 동안 반응시키고 배지와 MTT 용액을 제거한 후, DMSO를 넣어 교반하였다. 완전히 용해되었을 때 마이크로플레이트 판독기(Molecular device, USA)를 이용하여 540㎚에서 UV 흡광도를 측정하였다.
측정된 흡광도 값을 다음 수학식에 대입하여 세포 생존율을 계산하였다.

그 결과, 채송화 추출물의 처리없이 과산화수소만 처리한 세포의 생존율은 48%로 현저히 감소하였으나, 채송화 추출물을 처리한 결과 추출물의 용량 의존적으로 세포 생존율이 증가하였음을 확인하였고, 이는 채송화 추출물의 유의적인 신경보호효과를 시사한다(도 1).
실험예 2: 채송화 추출물의 항염증 효과
실험예 2-1: 채송화 추출물의 일산화질소(NO) 억제효과
채송화 추출물의 항염증 효능을 확인하기 위하여 염증매개물질인 NO의 생산량을 정량적으로 측정하였다. 구체적으로, 2.5×105개의 BV-2 소교세포를 24-웰 플레이트에 실험 24시간 전에 분주하여 신경염증 유발물질인 LPS 100 ng/ml와 상기 실시예 1에서 제조한 채송화 추출물을 농도별로 첨가한 후 배양기에서 24시간 동안 반응시키고, 각각 50 ㎕의 그리스 시약(Griess reagent; 1% sulfonilamine/0.1% N-(1-naphtyl)-ethylenediamine dihydrochloride/5% 인산)을 첨가한 다음 15분 동안 반응시켰다. 이후 마이크로플레이드 판독기(Molecular device, USA)를 이용하여 540nm 파장에서 신경염증 반응 억제 효과를 측정하였다.
그 결과, BV-2 소교세포에서 LPS 처리에 의해 NO 생성량이 현저히 증가하였고, LPS에 의해 현저하게 증가된 NO 생성량은 채송화 추출물의 용량 의존적으로 억제되었음을 확인하였다(도 2).
실험예 2-2: 채송화 추출물의 프로스타글란딘E2 ( PGE2 ) 억제효과
채송화 추출물의 항염증 효능을 확인하기 위하여 염증매개물질인 PGE2의 생산량을 정량적으로 측정하였다. 구체적으로, BV-2 소교세포는 24-웰 플레이트에 2.5 × 105 세포를 실험 24시간 전에 분주하여 신경염증 유발물질인 LPS 100 ng/ml와 상기 실시예 1에서 제조한 채송화 추출물을 농도별로 첨가한 후 배양기에서 24시간 동안 반응시키고, 24시간 뒤, 각각의 농도로 처리된 웰의 상층액을 수집하여 원심분리기를 이용하여 400 g, 3분간 침전시키고, PGE2 ELISA kit를 사용하여 마이크로플레이트 판독기(Molecular device, USA)를 이용하여 490 nm에서 UV 흡광도를 측정하였다. 측정된 흡광도 값을 standard curve의 정량 그래프를 이용하여 PGE2 생성을 계산하였다.
그 결과, BV-2 소교세포에서 LPS 처리에 의해 PGE2 생성량이 현저히 증가하였고, LPS에 의해 현저하게 증가된 PGE2 생성량은 채송화 추출물의 용량 의존적으로 억제되었음을 확인하였다(도 3).
실험예 2-3: 채송화 추출물의 iNOS 및 COX-2 단백질 및 mRNA 발현 억제효과
BV-2 소교세포에서 채송화 추출물의 신경염증 관련된 단백질 그리고 mRNA 발현의 동정실험을 위해서 웨스턴 블롯 및 RT-PCR 실험을 실시하였다. 실험을 수행한 6-웰 플레이트에 채송화 추출물을 30분 동안 전처리를 한 뒤, LPS로 처리한 뒤, 6시간 또는 24시간 동안 배양기에서 반응시켰다. 6시간 또는 24시간 뒤, 각각의 농도로 처리된 웰의 상층액을 수집하여 원심분리기를 이용하여 400 g, 3분간 침전시키고, 세포는 차가운 PBS로 세척한 뒤, Tper 용해 버퍼(Thermo, USA) 100 ㎕를 첨가하여 30분간 용해시켰다. 용해물들은 원심분리기를 이용하여, 10000 g, 4℃로 15분간 원심분리시켰으며, 원심분리 후 상층액은 실험을 위한 샘플로서 사용 전까지 70℃에서 보관하였다. 단백질은 BCA 정량분석 키트(Thermo, USA)를 이용하여 정량분석하였으며, 8% 내지 12%의 SDS 겔로 용출한 후 PVDF 막에 옮겼다. 단백질의 발현을 확인하기 위해, ECL (enhanced chmilumenescence)로 형광발색시켜 확인하는데, 먼저 5% 탈지분유로 막을 1시간 동안 반응시키며, 일차항체인 iNOS, COX-2 및 β-actin으로 4℃에서 밤새 표지시켰으며, TTBS로 5회 세척한 다음 이차항체인 HRP (horseradish peroxidase)-conjugated anti-rabbit 그리고 anti-mouse 항체를 상온에서 1시간 동안 표지시켰다. 표지 후 10분간 TTBS를 이용하여 5회 세척하였고, 세척된 막은 ECL를 이용하여, x-ray 필름을 이용하여 발색시켰으며, 농도는 정량분석법에 따라 정량분석프로그램(Fujifilm, Japan)을 이용하여 분석하였다. 또한 mRNA의 발현을 확인하기 위해 TRIZOL (Invitrogen)을 이용하여 전체 RNA를 추출하였으며, cDNA 합성 kit(Invitrogen)를 이용하여 RT-PCR을 수행하였다. 상기 RT-PCR은 iNOS, COX-2 및 β-actin의 프라이머를 사용하였고, 프라이머 서열을 하기 표 1에 나타내었다.

그 결과, LPS 처리에 의해 iNOS와 COX-2 단백질 및 mRNA의 발현 수준이 현저히 증가한 반면, 채송화 추출물을 투여한 결과 이들 단백질 및 mRNA의 발현 수준이 현저히 감소함을 확인하였다(도 4 및 도 5).
실험예 3: 채송화 추출물의 알츠하이머 관련 유전자 및 단백질 발현 억제효과
SH-SY5Y swedish 형질전환 신경세포주에서 채송화 추출물의 치매(알츠하이머 병) 관련된 단백질 및 mRNA 발현의 동정실험을 위해서 β-CTF 및 BACE1에 대하여 각각 웨스턴 블롯 및 RT-PCR 실험을 실시하였다. 실험을 수행한 6-웰 플레이트에 채송화 추출물을 24시간 동안 전처리를 한 뒤, 배양기에서 반응시켰다. 24시간 뒤, 각각의 농도로 처리된 웰의 상층액을 수집하여 원심분리기를 이용하여 400 g, 3분간 침전시키고, 세포는 차가운 PBS로 세척한 뒤, Tper 용해 버퍼(Thermo, USA) 100 ㎕를 첨가하여 30분간 용해시켰다. 용해물들은 원심분리기를 이용하여, 10000 g, 4℃로 15분간 원심분리시켰으며, 원심분리 후 상층액은 실험을 위한 샘플로서 사용 전까지 70℃에서 보관하였다.

한편, mRNA의 발현을 확인하기 위해 TRIZOL (Invitrogen)을 이용하여 전체 RNA를 추출하였으며, cDNA 합성 kit(Invitrogen)를 이용하여 RT-PCR을 수행하였다. 상기 RT-PCR은 BACE1 및 β-actin의 프라이머를 사용하였고, 프라이머 서열을 하기 표 2에 나타내었다.

그 결과, β-CTF(β-secretase-cleaved carboxyl-terminal fragment) 단백질 발현수준은 처리한 채송화 추출물의 용량 의존적으로 억제하였음을 확인하였다(도 6).
BACE1 단백질 및 mRNA 발현수준 또한 처리한 채송화 추출물의 용량 의존적으로 억제하였음을 확인하였다(도 7a 및 도 7b).
실험예 4: 채송화 추출물 또는 이의 분획물의 작업기억력 증진 효과
실험예 4-1: Y-미로실험을 통한 채송화 추출물 또는 이의 분획물의 작업기억력 증진 효과 확인
마우스를 각 군당 14 마리씩 4 군으로 나누었다. 제1군은 10% Tween 20을 포함하고 있는 증류수 투여군(대조군), 제2군 내지 제4군은 각각 채송화 추출물 또는 이의 부탄올, 에틸아세테이트, 물 가용분획물 6.25, 12.5 및 25 mg/kg 투여군(실험군)으로 하였다.
채송화 추출물 또는 이의 분획물을 10% Tween 20을 포함하고 있는 증류수에 용해시켜 6.25, 12.5 및 25 mg/kg의 용량으로 마우스에 경구투여하고, 1시간 후에 마우스를 Y-미로에 넣어 각각 A, B 및 C 가지에 마우스가 자유롭게 들어가는 것을 측정하였다. 이때, 새로운 가지에 들어가면 1점을 주게 되며, 교차행동 %는 하기 수학식 2에 의하여 계산하였다.

그 결과, 채송화 추출물 또는 이의 분획물을 투여하지 않은 제1군(대조군)의 마우스에 비하여 채송화 추출물 또는 이의 분획물을 투여한 제2군 내지 제4군(실험군)의 마우스에서 채송화 추출물 또는 이의 분획물의 농도 의존적으로 교차행동 비율을 증가시켰음을 확인하였다.
이는 Y-미로실험에서 작업기억 증진효능을 나타내는 결과로서, 상기 실험결과로부터 채송화 메탄올 조추출물 및 이의 분획물이 각각 마우스의 작업기억력을 현저히 증가시킬 수 있음을 확인하였다(도 8a 내지 도 8d).
실험예 4-2: 스코폴아민 유도 건망증모델에서의 Y-미로실험을 통한 채송화 추출물 또는 이의 분획물의 작업기억력 개선 효과 확인
마우스를 각 군당 14 마리씩 5 군으로 나누었다. 제1군은 10% Tween 20을 포함하고 있는 증류수 투여군(대조군), 제2군은 10% Tween 20을 포함하고 있는 증류수 및 스코폴라민 투여군(양성대조군), 제3군 내지 제5군은 채송화 추출물 또는 이의 부탄올, 에틸아세테이트, 물 가용분획물의 투여군(실험군)으로 하였다.
채송화 추출물 또는 이의 분획물을 각각 10% Tween 20을 포함하고 있는 증류수에 용해시켜 6.25, 12.5 및 25 mg/kg의 용량으로 마우스에 경구투여하고, 30분 후에 스코폴아민 0.5 mg/kg을 피하주사 하였다.
1시간 후에 마우스를 Y-미로에 넣어 A, B 및 C 가지에 마우스가 자유롭게 들어가는 것을 측정하였다. 이때, 새로운 가지로 들어가면 1점을 주게 되며, 교차행동 %는 하기 수학식 2에 의하여 계산하였다.

그 결과, 스코폴라민을 투여하지 않은 제1군(대조군)의 마우스에 비하여 스코폴라민을 투여한 제2군(양성대조군)에서 교차행동 비율이 낮아짐으로써 건망증이 유도되었음을 확인하였다.
한편, 채송화 추출물 또는 이의 분획물을 투여한 제3군 내지 제5군(실험군)의 마우스에서 교차행동 비율을 증가시켰음을 확인하였다. 상기 실험결과를 통하여 채송화 메탄올 추출물 또는 이의 분획물이 기억력 손상을 효과적으로 예방할 수 있음을 확인하였다(도 9a 내지 도 9d).
실험예 4-3: 25 -35 유도 치매모델에서의 Y-미로실험을 통한 채송화 추출물의 작업기억력 개선 효과 확인
마우스를 각 군당 11~12 마리씩 5 군으로 나누었다. 제1군은 10% Tween 20을 포함하고 있는 증류수 투여군(대조군), 제2군은 10% Tween 20을 포함하고 있는 증류수 및 Aβ25 -35 투여군(양성대조군), 제3군 내지 제5군은 농도별 채송화 추출물의 투여군(실험군)으로 하였다.
채송화 추출물을 각각 10% Tween 20을 포함하고 있는 증류수에 용해시켜 6.25, 12.5 및 25 mg/kg의 용량으로 마우스에 경구투여하고, 1시간 후에 Aβ25 -35 6nmol을 좌측 뇌실에 입체정위 기구를 사용하여 주사하였다. 5일 동안 각각의 군들에 10% Tween 20을 포함하여 채송화 추출물 6.25, 12.5 및 25 mg/kg을 투여하였다.
5일째 각각의 채송화 추출물을 투여 1시간 후에 마우스를 Y-미로에 넣어 A, B 및 C 가지에 마우스가 자유롭게 들어가는 것을 측정하였다. 이때, 새로운 가지로 들어가면 1점을 주게 되며, 교차행동 %는 하기 수학식 2에 의하여 계산하였다.

그 결과, Aβ25 - 35을 투여하지 않은 제1군(대조군)의 마우스의 교차행동 비율(67%)에 비하여 Aβ25 - 35을 투여한 제2군(양성대조군)에서 교차행동 비율(52%)이 낮아짐으로써 치매가 유도되었음을 확인하였다(p<0.01).
한편, 채송화 메탄올 추출물 6.25 mg/kg 투여군, 12.5 mg/kg 투여군 및 25 mg/kg 투여군은 각각 58%, 67%, 및 62%의 교차행동 비율을 보여 Aβ25 -35 유도 치매모델군(양성대조군)에 비하여 현저히 교차행동 비율을 증가시켰음을 확인하였다.
상기 실험결과를 통하여 채송화 추출물이 알츠하이머성 기억력 손상을 효과적으로 예방할 수 있음을 확인하였다(도 12).
실험예 5: 채송화 추출물 또는 이의 분획물의 학습 및 기억력 증진 효과
실험예 5-1: 수동회피실험을 통한 채송화 추출물 또는 이의 분획물의 학습 및 기억력 증진 효과 확인
마우스를 각 군당 14 마리씩 4 군으로 나누었다. 제1군은 10% Tween 20을 포함하고 있는 증류수 투여군(대조군), 제2군 내지 제4군은 각각 채송화 추출물 또는 이의 가용 분획물(에틸아세테이트, 부탄올, 물) 6.25, 12.5 및 25 mg/kg 투여군(실험군)으로 하였다.
채송화 추출물 또는 이의 가용 분획물(에틸아세테이트, 부탄올, 물)을 각각 10% Tween 20을 포함하고 있는 증류수에 용해시켜 6.25, 12.5 및 25 mg/kg의 용량으로 마우스에 경구투여하였다. 1시간 후에 마우스를 스텝-스루 (step-through) 장치에 넣어 어두운 상자로 들어가면 0.25mA의 전기 충격을 3초 동안 가하여 학습(training)시켰다. 24 시간 후에 마우스를 다시 밝은 상자에 넣어 검은 상자에 들어가기까지의 체재시간을 측정하여 학습 및 기억력의 지표로 하였다. 이때 마우스의 컷-오프 시간 (cut-off time)은 300초로 하여 대조군과 비교하였다.
그 결과, 채송화 추출물 또는 이의 분획물을 투여하지 않은 제1군(대조군)의 마우스에 비하여 채송화 추출물 또는 이의 분획물을 투여한 제2군 내지 제4군(실험군)의 마우스에서 체재시간이 증가함을 확인하였다.
상기 실험결과로부터 채송화 메탄올 조추출물 및 이의 분획물이 마우스의 학습 및 기억력을 현저히 증가시킬 수 있음을 확인하였다(도 10a 내지 도 10d).
실험예 5-2: 스코폴아민 유도 건망증모델에서의 수동회피실험을 통한 채송화 추출물 또는 이의 분획물의 학습 및 기억력 개선 효과 확인
마우스를 각 군당 14 마리씩 5 군으로 나누었다. 제1군은 10% Tween 20을 포함하고 있는 증류수 투여군(대조군), 제2군은 10% Tween 20을 포함하고 있는 증류수 및 스코폴라민 투여군(양성대조군), 제3군 내지 제5군은 채송화 추출물 또는 이의 가용분획물(에틸아세테이트, 부탄올, 물)의 투여군(실험군)으로 하였다.
채송화 추출물 또는 이의 분획물을 각각 10% Tween 20을 포함하고 있는 증류수에 용해시켜 6.25, 12.5 및 25 mg/kg의 용량으로 마우스에 경구투여하고, 30분 후에 스코폴아민 0.5 mg/kg을 피하주사 하였다.
30분 후에 마우스를 스텝-스루(step-through) 장치에 넣어 어두운 상자로 들어가면 0.25mA의 전기 충격을 3초 동안 가하여 학습(training)시켰다. 24 시간 후에 마우스를 다시 밝은 상자에 넣어 검은 상자에 들어가기까지의 체재시간을 측정하여 스코폴아민 유도 건망증에 대한 학습 및 기억력의 지표로 하였다. 이때 마우스의 컷-오프 시간 (cut-off time)은 300초로 하여 대조군과 비교하였다.
그 결과, 스코폴라민을 투여하지 않은 제1군(대조군)의 마우스에 비하여 스코폴라민을 투여한 제2군(양성대조군)에서 체재시간이 단축됨으로써 건망증이 유도되었음을 확인하였다.
한편, 채송화 추출물 또는 이의 에틸아세테이트, 부탄올, 물 가용분획물을 각각 투여한 제3군 내지 제5군(실험군)의 마우스에서 체재시간이 증가되었음을 확인하였다. 상기 실험결과를 통하여 채송화 메탄올 추출물 또는 이의 분획물이 기억력 손상을 효과적으로 예방할 수 있음을 확인하였다(도 11a 내지 도 11d).
실험예 5-3: 25 -35 유도 치매모델에서의 수동회피실험을 통한 채송화 추출물의 학습 및 기억력 개선 효과 확인
마우스를 각 군당 11~12 마리씩 5 군으로 나누었다. 제1군은 10% Tween 20을 포함하고 있는 증류수 투여군(대조군), 제2군은 10% Tween 20을 포함하고 있는 증류수 및 Aβ25 -35 투여군(양성대조군), 제3군 내지 제5군은 농도별 채송화 메탄올 추출물의 투여군(실험군)으로 하였다.
채송화 메탄올 추출물을 각각 10% Tween 20을 포함하고 있는 증류수에 용해시켜 6.25, 12.5 및 25 mg/kg의 용량으로 마우스에 경구투여하고, 1시간 후에 Aβ25 -35 6 nmol을 좌측 뇌실에 입체정위 기구를 사용하여 주사하였다. 5일 동안 각각의 군들에 10% Tween 20을 포함하여 채송화 추출물 6.25, 12.5 및 25 mg/kg을 투여하였다.
1시간 후에 마우스를 스텝-스루(step-through) 장치에 넣어 어두운 상자로 들어가면 0.3mA의 전기 충격을 3초 동안 가하여 학습(training)시켰다. 24 시간 후에 마우스를 다시 밝은 상자에 넣어 검은 상자에 들어가기까지의 체재시간을 측정하여 Aβ25 -35 유도 치매에 대한 학습 및 기억력의 지표로 하였다. 이때 마우스의 컷-오프 시간 (cut-off time)은 300초로 하여 대조군과 비교하였다.
그 결과, Aβ25 - 35을 투여하지 않은 제1군(대조군)의 마우스의 체재시간(218초)에 비하여 Aβ25 - 35을 투여한 제2군(양성대조군)에서 체재시간(73초)이 단축됨으로써 건망증이 현저하게 유도되었음을 확인하였다(p<0.001).
한편, 채송화 메탄올 추출물 6.25 mg/kg 투여군, 12.5 mg/kg 투여군 및 25 mg/kg 투여군은 각각 97초, 163초, 및 153초의 체재시간을 보여 Aβ25 -35 유도 치매모델군(양성대조군)에 비하여 현저한 체재시간의 증가효과를 보였다.
상기 실험결과를 통하여 채송화 추출물이 알츠하이머성 기억력 손상을 효과적으로 예방할 수 있음을 재차 확인하였다(도 13).

Claims (12)

  1. 채송화(Portulaca grandiflora Hook.) 추출물 또는 이의 분획물을 유효성분으로 포함하는 신경염증 또는 퇴행성 뇌신경 질환의 예방 또는 치료용 약학적 조성물.
  2. 제1항에 있어서, 상기 추출물은 채송화를 물, C1 내지 C4의 저급알콜 및 이들의 혼합용매로 구성된 군으로부터 선택되는 용매로 추출하여 수득한 것인 조성물.
  3. 제1항에 있어서, 상기 추출물은 채송화를 메탄올로 추출한 것인 조성물.
  4. 제1항에 있어서, 상기 분획물은 채송화 메탄올 추출물을 물, 에틸아세테이트 또는 부탄올을 사용하여 분획하여 수득한 것인 조성물.
  5. 제1항에 있어서, 상기 퇴행성 뇌신경 질환은 치매, 알츠하이머병, 파킨슨병, 헌팅턴병, 루게릭병, 크로이츠펠트야콥병, 뇌졸중, 다발성 경화증, 학습장애, 인지장애 및 기억력 손상으로 이루어진 군에서 선택된 것인 조성물.
  6. 채송화(Portulaca grandiflora Hook.) 추출물 또는 이의 분획물을 유효성분으로 포함하는 신경염증 또는 퇴행성 뇌신경 질환의 예방 또는 개선용 식품 조성물.
  7. 제6항에 있어서, 상기 추출물은 채송화를 물, C1 내지 C4의 저급알콜 및 이들의 혼합용매로 구성된 군으로부터 선택되는 용매로 추출하여 수득한 것인 조성물.
  8. 제6항에 있어서, 상기 추출물은 채송화를 메탄올로 추출한 것인 조성물.
  9. 제6항에 있어서, 상기 분획물은 채송화 메탄올 추출물을 물, 에틸아세테이트 또는 부탄올을 사용하여 분획하여 수득한 것인 조성물.
  10. 제6항에 있어서, 상기 퇴행성 뇌신경 질환은 치매, 알츠하이머병, 파킨슨병, 헌팅턴병, 루게릭병, 크로이츠펠트야콥병, 뇌졸중, 다발성 경화증, 학습장애, 인지장애 및 기억력 손상으로 이루어진 군에서 선택된 것인 조성물.
  11. 채송화(Portulaca grandiflora Hook.) 추출물 또는 이의 분획물을 유효성분으로 포함하는 학습 능력 또는 기억력 증진 또는 개선용 식품 조성물.
  12. 제1항 내지 제5항의 조성물 중 어느 한 항의 조성물을 이를 필요로 하는 개체에 투여하는 단계를 포함하는 신경염증 또는 퇴행성 뇌신경 질환의 예방 또는 치료방법.
