 外来遺伝子を導入することなく、エタノール産生効率の高い発酵菌を得ることを課題とする。さらに、発酵菌の増殖を妨げる有機酸等の増殖阻害物質に耐性のある発酵菌を得ることを課題とする。 育種によって五炭糖及び六炭糖から効率的にエタノール生産することができるMeyerozyma guilliermondiiに、セルフクローニングによりトランスアルドラーゼ、アルコールデヒドロゲナーゼ遺伝子を導入し、更に育種することによって、エタノール産生能を向上した菌を作成した。

Description

高効率エタノール発酵菌
 リグノセルロース系バイオマスを用いたバイオエタノール生産において、糖化溶液を発酵するための微生物に関する。
 特に、リグノセルロース系バイオマスを用いたバイオエタノール生産において、五炭糖(以下、C5ということもある。)、及び六炭糖(以下、C6ということもある。)から効率的にエタノール生産することができる微生物に関する。
 バイオエタノールは、バイオマスから産生される枯渇することのない再生可能資源として期待されている。また、バイオエタノールを燃焼させて発生する二酸化炭素はカーボンニュートラルであることから、バイオエタノールの利用が進むことによって、地球温暖化の主な原因である二酸化炭素の上昇を抑制すると考えられている。
 バイオエタノールは、バイオマスを発酵させ、蒸留してエタノールを精製する。バイオエタノールの収率を高めるために糖化溶液から多くのアルコールを生成する必要がある。バイオエタノール生産の過程で一般的に用いられている酵母は、キシロース、アラビノースなどの五炭糖をアルコールに変換できないため、発酵原料としては六炭糖のみが用いられてきた。
 原料によって異なるものの典型的なバイオマスには、35~45%のセルロース、25~40%のヘミセルロース、15~30%のリグニンが含まれていると言われている。したがって、六炭糖が重合しているセルロースだけではなく、五炭糖であるキシロース等を主として含有するヘミセルロースを基質として利用することは効率的なエタノール産生につながることになる。
 キシロースはグルコースの次にバイオマス中に多く含まれている糖であると言われており、五炭糖を効率的に利用することはバイオエタノール生産において大きな課題となっている。
 これまでに、遺伝子組換えによるキシロース利用能の付与や、キシロースを利用してエタノールを産生する微生物の利用等により、キシロースを少しでも利用する技術が開示されている。
 特許文献1には、キシローストランスポーター活性を有する遺伝子を宿主細胞に導入することによって、キシロース(C5)をキシルロースに変換し、解糖系のペントースリン酸経路に組み入れ、発酵に利用する発明が開示されている。
 特許文献2には、アラビノーストランスポーターを付与した酵母によって、アルコールを生成する技術が開示されている。特許文献1と同様にアラビノース(C5)をアラビトール、キシルロースを経て解糖系のペントースリン酸経路に組み入れ、発酵に利用するものである。
 非特許文献1には、大腸菌由来のキシロース資化遺伝子をザイモモナスに組み込むことにより、キシロース資化能を付与することが開示されている。
 非特許文献2には、ピキア属酵母が、キシロースを利用してエタノールを生産することが記載されている。

 しかしながら、特許文献1の発明は、Candida guilliermondii由来のキシローストランスポーター活性のあるタンパク質を宿主としてSaccharomyces cerevisiaeに導入している。すなわち外来遺伝子を導入することになる。
 また、特許文献2の発明もトランスポーター遺伝子は異なるものの、宿主に対して異なる種の遺伝子を導入する発明である。
 また、非特許文献1に記載の技術は、キシロース資化遺伝子を導入するものであり、上記特許文献1及び2とは技術思想は異なるが、外来遺伝子を導入することに変わりない。
 そのため、上記特許文献1及び2、非特許文献1はいずれも国連で採択された「生物の多様性に関する条約のバイオセーフティに関するカルタヘナ議定書」を実施するための封じ込め策を講じる必要がある。したがって、バイオセーフティを保証するための施設を必要とすることから、当該菌体を利用してエタノールを生産することはコストの面で不利である。
 また、非特許文献2に記載の技術によって、ピキア属酵母を利用することは、野生型のピキア属酵母のキシロース利用性が低いため、エタノール産生効率はさほど高くならない。
 本発明は、外来遺伝子を導入することなく、エタノール産生効率の高い発酵菌を得ることを課題とする。
 本発明は、五炭糖及び六炭糖から効率的にエタノールを産生するMeyerozyma guilliermondiiであって、特許微生物寄託センター(日本国 〒292-0818 千葉県木更津市かずさ鎌足2-5-8 122号室)に寄託番号NITE ABP-01976として、2014年12月4日に寄託されている菌(以下、ABP-01976株ともいう。)をコーストーバー糖液中で育種することにより、コーンストーバー糖液でのエタノール産生が向上していることを特徴とする。
 ABP-01976菌株は、稲わら由来の糖液中で育種を行い、発酵収率の高い菌を選択することによって得られた高効率エタノール発酵菌に、トランスアルドラーゼ、アルコールデヒドロゲナーゼをセルフクローニングしてさらに発酵収率を高めた菌である。
 しかしながら、稲わらで育種を行い、選択したためにコーンストーバー由来の糖液中での発酵収率はさほど高くない。そこで、コーンストーバー中で育種を行うことによって、発酵収率の向上した菌を得ることができた。
 また、本発明の高効率エタノール発酵菌は、五炭糖及び六炭糖から効率的にエタノールを産生するMeyerozyma guilliermondiiであって、特許微生物寄託センター(日本国 〒292-0818 千葉県木更津市かずさ鎌足2-5-8 122号室)に寄託番号NITE BP-01966(以下、BP-01966株ともいう。)として、2014年11月19日に寄託されていることを特徴とする。
 コーンストーバー由来の糖液中で育種することによって、コーンストーバー糖液に馴化し、発酵収率が向上した菌のうち、特に発酵収率の高いものを特許微生物寄託センターに寄託し、寄託番号NITE BP-01966を得た。
 本発明の菌は、育種とMeyerozyma guilliermondii自身の酵素遺伝子を導入した組換えによって得たものであり、カルタヘナ法に則した封じ込め策を必要としない。したがって、バイオセーフティのための特別な施設を必要とせず、従来の設備を使用することができる。

 以下、本件発明の菌株について以下に説明する。
1.菌株の単離
 Meyerozyma guilliermondiの親株(N株)を稲わら由来の糖液を用い育種を行い、エタノール産生能の高い株を選択した。熊谷産の稲わらを、等量の25%アンモニア水に80℃3時間漬け込んだ後、アンモニアを放散させた。処理したバイオマスは、pHを4に調整後、アクレモニウムセルラーゼ(Meiji Seika ファルマ社製)を添加して、50℃72時間酵素糖化を行った。作成したスラリーはフィルタープレス法にて固液分離を行い液体を回収した。この液体(以下、清澄液ともいう。)を用いて、変異剤を加えながら19ヶ月馴化培養を行い、発酵性能向上株を選抜した。発酵性能向上株は、一定時間後の生成エタノール量を基準に選抜した。発酵性能の高い菌株を独立行政法人製品評価技術基盤機構 特許微生物寄託センターに、寄託番号NITE BP-01964として、寄託した。
 ここでは示さないが、BP-01964菌株は、野生型に対して2倍以上エタノールを産生する株であり、さらにC6であるグルコースだけではなく、C5であるキシロースの資化能が向上していることを確認している。
 次に、BP-01964菌株にセルフクローニングによりトランスアルドラーゼ、アルコールデヒドロゲナーゼ遺伝子を導入して、さらにエタノール産生能の高い菌を得た。
 アンモニア処理稲わら液を用い、糖化酵素で処理して糖液を得る際に、副産物として有機酸、アルデヒド、フェノール等の発酵阻害物質が産生する。特に酢酸は約1000mg/Lと非常に多量に存在し、発酵収率を低下させる。したがって、酢酸による発酵菌の増殖阻害を抑制することができれば、さらにエタノール産生効率を向上させることができる。
 トランスアルドラーゼは、解糖系から分岐したペントースリン酸経路のセドヘプツロース-7-リン酸(S7P)及びグリセルアルデヒド3リン酸(GAP)からエリトロース-4-リン酸(E4P)及びフルクトース6リン酸(F6P)への代謝を触媒する酵素である。この酵素を導入することによって、阻害物質である酢酸存在下でのエタノール収率向上効果が確認されている。
 トランスアルドラーゼの上流にはキシロースレダクターゼのプロモーターをつなげた。キシロース資化の際に機能するキシロースレダクターゼのプロモーターを用いることによって、トランスアルドラーゼが効率良く作用すると考えられるからである。
 また、アルコールデヒドロゲナーゼはアセトアルデヒドからエタノールを産生させることができる酵素であり、稲藁糖液に阻害物質として含まれるアルデヒド類を変換し毒性を弱めることができる酵素である。
 アルコールデヒドロゲナーゼの上流にはGAPDHのプロモーターをつなげた。グリセロアルデヒド-3-リン酸脱水素酵素(GAPDH)は解糖系に存在する強力なプロモーターであることから、解糖系の酵素であるアルコールデヒドロゲナーゼのプロモーターとして使用することにより、効率よく作用すると考えられる。
 したがって、2つの酵素を遺伝子導入により導入した組換え発酵菌を作成することができれば、よりエタノール産生能の高い菌を得ることが期待できる。
 遺伝子導入は、次の手順を取った。導入したい遺伝子及びそのターミネーター部(以下、遺伝子+ターミネーター部という。)をPCR増幅する。導入に用いたプロモーター部をPCR増幅する。これらは何れも本発明で用いた菌株であるMeyerozyma guilliermondiの染色体からPCR増幅する。
 PCR増幅したDNA断片をプロモーター、遺伝子+ターミネーター部の順になるよう、インフュージョン法を用いて、大腸菌用の市販ベクターにクローニングする。クローニングされたベクターを大腸菌に形質転換し、ベクターを増幅する。増幅したベクターからプロモーター及び遺伝子+ターミネーター部を制限酵素で切出す、あるいは増幅したベクターからPCR増幅することにより、相同組換用のDNA断片を得る。
 キシロースレダクターゼのプロモーターは下記配列番号1及び配列番号2のプライマー、トランスアルドラーゼ遺伝子及びターミネーター部分は下記配列番号3及び4のプライマーを用いて増幅した。

 また、GAPDHのプロモーターは具体的には下記配列番号5及び配列番号6のプライマー、トアルコールデヒドロゲナーゼ遺伝子及びターミネーター部分は下記配列番号7及び8のプライマーを用いて増幅した。

 得られたDNA断片を菌株に相同組換えし、所望の菌株を得た。相同組換にはエレクトロポレーション法を用いた。相同組換用DNA断片は、キシロースレダクターゼのプロモーター、トランスアルドラーゼ+ターミネーター、GAPDHのプロモーター、アルコールデヒドロゲナーゼ+ターミネーターの順となっている。また、この方法により得られた菌株は、遺伝子を導入しているが、セルフクローニングであるため、カルタヘナ法上、非組換菌扱いになる範疇に属するものとなっている。
 遺伝子導入した菌株からエタノール産生能の高い菌を単離し、特許微生物寄託センターに寄託し寄託番号NITE ABP-01976を得た。
 次に、コーストーバー由来の糖液中で4ヶ月馴化培養を行い、コーストーバー由来の糖液中で発酵収率の高い菌株を得て、特許微生物寄託センターに、寄託番号BP-01966として寄託した。以下に、得られた菌株の発酵収率等性質を示す。
2.菌株の性質
2.1 発酵収率
 親株、本発明の株の発酵試験を行った。糖濃度の異なる希硫酸処理コーンストーバー由来酵素糖化溶液をpHを6に調整した溶液を複数用い、上記株の培養液を培地のOD600が2.0となるように添加し、30℃、96時間培養して得られたエタノール濃度と投入糖化溶液の糖濃度から計算した発酵収率をプロットした。結果を図1に示す。
 図1に示すように親株であるN株は発酵収率が約48%であるのに対し、育種によって得られた高エタノール産生株BP-01964は発酵収率が80%、遺伝子組換えによりトランスアルドラーゼ、アルコールデヒドロゲナーゼを導入することにより得られたABP-01976株は発酵収率が84%、さらにコーンストーバー由来糖液中で育種を行った本発明の株寄託番号BP-01966株は発酵収率がさらに高く、約86%となっていた。また、図中△で示しているのは他社の発酵菌の発酵収率であるが、これらの菌に比べても高い発酵収率を示すことは明らかである。
 さらに、エタノール産生能を比較した。図2は親株であるN株(WT)、育種により発酵収率を向上させたBP-01964菌株に対する本菌株のエタノール産生を示す。コーンストーバーを希硫酸処理した糖化液をpHを6に調整して用い、菌株の培養液を培地のOD600が2.0となるように添加した。30℃、96時間培養した後の培養液中のエタノール量を示す。糖化溶液のグルコースは、63.2g/L、キシロースは34.5g/Lであった。エタノール測定はGC-FID(ジーエルサイエンス製:GC390B)を用いた。
 図2より明らかなように、野生型に対して2.5倍以上、育種によってエタノール産生能を高めた株に対しても、約1.2倍のエタノールを産生する株を得ることができた。野生株に対してエタノール産生が向上していることから、C5であるキシロースの資化能が向上していると考えられる。そこで、この菌株のグルコース及びキシロース資化能を確認した。
2.2 キシロース資化能の検討
 次に、稲わらをアンモニア水で上記と同様に処理した後、アクレモニウムセルラーゼを添加して、50℃72時間酵素糖化を行い、作成したスラリーを用いて発酵を行った。
 スラリー発酵槽はジャケット構造になっており、温度はジャケット部に温水を循環させて調節を行った。また、底部には通気口を設けてあり、底部の通気口からは、フィルタを通した空気を所定量通気し続け、モータに連動したヘラで撹拌を行いながら発酵を行った。
 スラリー中に含まれるグルコース、キシロース、エタノールの量の経時的な変化の解析を行った。グルコース、キシロースはスラリーをサンプリングし、遠心により得た上清をHPLC(東ソー製:LC-8020)測定にかけて測定した。エタノールは上記と同様にGC-FID(ジーエルサイエンス社製、GC390B)を用いた。結果を図3に示す。
 C6であるグルコースが先に消費されるが、その後、スラリー中のグルコースの減少とともにC5であるキシロースが消費され、エタノールが産生される。得られた菌は、C5、C6ともに資化能を備えていることから、効率よくエタノールを産生することができる。したがって、工業生産的にも有用な菌株である。
2.3 スラリー発酵能、清澄液発酵能
 バイオエタノール産生を行うときに、スラリー、清澄液、どちらを用いた場合であっても効率よく発酵する菌であることが望ましい。そこで、スラリー、清澄液を用いて発酵収率を比較した。発酵収率は以下の式により計算される。
発酵収率=得られたエタノール量(g/L)/発酵開始時の糖液に含まれていたグルコース+キシロース量(g/L)/0.5114
 図4に示すように、スラリー発酵であっても、清澄液発酵であっても、同等の性能を発揮することのできる菌株を得ることができた。
 寄託番号BP-01966で示される菌は、以上示してきたように、野生型Meyerozyma guilliermondiのキシロース資化能が育種によって強化されており、バイオマスとして稲わら、コーンストーバーどちらを用いた場合であっても効率的にエタノール産生を行うことができる。
 以上示してきたように、野生型Meyerozyma guilliermondiのキシロース資化能が育種によって強化されており、さらに遺伝子組換えによってエタノール産生能が高い菌株を得ることができた。さらに、バイオマスとして稲わら、コーンストーバーどちらを用いた場合であっても効率的にエタノール産生を行うことができる。

Claims (2)

  1.  五炭糖及び六炭糖から効率的にエタノールを産生する発酵菌であって、
     特許微生物寄託センターに寄託番号ABP-01976として寄託されている菌をコーストーバー糖液中で育種することにより、
     コーンストーバー糖液でのエタノール産生が向上していることを特徴とする高効率エタノール発酵菌。
  2.  請求項1記載の高効率エタノール発酵菌において、
     特許微生物寄託センターに寄託番号BP-01966として寄託されていることを特徴とする高効率エタノール発酵菌。
