본 발명은 탈세포화된 세포외 기질 및 가교제를 포함하는 조직공학용 구조체 형성용액 및 심장전구세포를 포함하는 제1 바이오프린팅용 조성물과, 상기 조직공학용 구조체 형성용액, 중간엽 줄기세포 및 혈관내피성장인자를 포함하는 제2 바이오프린팅용 조성물로 인쇄 및 가교화하여 제1 바이오프린트층 및 제2 바이오프린트층을 교대로 배치하여 3차원 구조체를 형성하는 단계; 및 상기 가교된 3차원 구조체를 열적겔화하여 가교-겔화 3차원 구조체를 얻는 단계;를 포함하는 심근조직 재생용 3차원 구조체의 제조방법 및 심근조직 재생용 3차원 구조체를 제공하며, 본 발명에 따른 제조방법은 심장전구세포를 구조체 내에 균등하게 위치시킬 뿐만 아니라, 구조체 내에 혈관세포로 이루어진 혈관 네트워크를 구현함으로써, 세포의 생존능을 장기간 유지시킬 수 있어 심근내부로의 세포전달 효율을 현저하게 향상시킬 수 있다.

본 발명은 심근조직 재생용 3차원 구조체 및 이의 제조방법에 관한 것이다. 더욱 상세하게는, 본 발명은 본 발명은 탈세포화된 세포외 기질 및 가교제를 포함하는 조직공학용 구조체 형성용액 및 심장전구세포를 포함하는 제 1 바이오프린팅용 조성물과, .상기 조직공학용 구조체 형성용액, 증간엽 줄기세포 및 혈관내피성장인자를 포함하는 제 2 바이오프린팅용 조성물로 인쇄 및 가교화하여 제 1 바이오프린트층 및 제 2 바이오프린트층을 교대로 배치하여 3차원 구조체를 형성하는 단계; 및 상기 가교된 3차원 구조체를 열적겔화하여 가교 -겔화 3차원 구조체를 얻는 단계;를 포함하는 심근조직 재생용 3차원 구조체의 제조방법 및 심근조직 재생용 3차원 구조체에 관한 것이다.
【발명의 배경이 되는기술】
3차원 프린팅은 복잡한 형상을 가진 조직 (tissue)이나 장기 (organ)의 의료 데이터로부터 얻은 임의 형상 정보를 G-code로 변환한 후 적층 공정 (layer-by-layer process)을 통하여 복잡한 골격 구조를 구축하는 것을 말한다. 이러한 3차원 프링팅은 또한 '3차원 바이오프린팅 (three-dimensional bioprinting, 3D bioprinting)'으로도 지칭된다. 예를 들어, '다축 조직 장기 프린팅 시스템 '은 대표적인 3차원 프린팅 기술 중 하나로서, 재료를 공압으로 분사하는 공압식 시린지 2개와 스텝 모터 (step motor)를 이용하여 나노리터 단위로 정밀하게 분사하는 피스톤식 시린지 2개로 이루어져 있으며, 다양한 재료를 동시에 이용할 수 있다.
통상, PLA(Polylactic Acid), PGA(Poly-glycolic Acid), PLGA(Poly-lactic-co-glycolid Acid), PCL(Polycaprolactone), 흑은 이들의 흔합물과 같은 열가소성 생체적합성 고분자를 공압식 시린지에 장착하여 구조체를 제작한다. 또한, 콜라겐, 히알루론산, 젤라틴, 알기네이트, 키토산 또는 피브린 (fibrin) 등으로 이루어진 하이드로겔을 피스톤식 시린지를 이용하여 3차원 구조체를 제작한다. 바이오프린팅은 세포 -함유 매체의 분배를 필요로 하기 때문에, 바이오프린팅의 중요한 측면은 프린팅 공정이 세포적합성 (cytocompatible)이어야 한다는 것이다. 이러한 제한은, 수성 또는 수성 겔 환경에서 수행하여야 할 필요성 때문에, 재료의 선택을 감소시킨다. 따라서, 젤라틴, 젤라틴 /키토산, 젤라틴 /알기네이트, 젤라틴 /피프로넥틴, 루트롤 F127(Lutrol F127)/알기네이트, 및 알기네이트 등의 재료를 사용한 하이드로겔이 간에서 골에 이르기까지 다양한 조직을 제조하기 위한 바이오프린팅에 사용되고 있다. 바이오프린팅에 사용되는 상기 하이드로겔 또는 하이드로겔과 세포의 흔합물 등은 '바이오잉크 (bioink)'로도 지칭된다. 일반적으로, 세포는 전체 배양 기간 동안 원래의 놓여진 위치에 특이적으로 계속 위치해 있으며, 이는 세포가 주위의 알기네이트 겔 매트릭스를 유지하거나 분해할 수 없기 때문이다 (Fedorovich, N. E. et al. Tissue Eng. 13, 1905-1925 (2007)). 따라서, 세포-프린트 구조체의 바이오프린팅에 관한 몇 가지 성공 보고가 있을 지라도, 최소한의 세포 -재료 상호작용 (cell-material interactions) 및 열등한 (inferior) 조직 형성은 가장 증요한 문제이다. 실제로, 이들 재료는 천연의 세포외 기질 (extracellular matrices, ECMs)의 복잡성을 나타낼 수 없으며, 이에 따라 생체 조직의 전형적인 세포 -세포 연결 (cell-cell connections) 및 3차원 (three-dimensional, 3D) 세포 구성을 갖는 미세환경을 재현하기에 충분하지 않다.
따라서 상기 하이드로겔 중의 세포는 생체 내에서 살아있는 조직의 고유한 형태 및 기능을 나타낼 수 없다ᅳ 따라서, 세포의 모 조직 (parent tissue)과 유사한 천연의 미세환경을 세포에 제공된다면 이상적일 것이다. 탈세포화된 세포외 기질 (decellularized extracellular matrix, dECM)은 이렇게 하기 위한 최선의 선택이며, 이는 어떠한 천연의 혹은 인공 재료도 천연의 세포외 기질의 특성 모두를 재현할 수 없기 때문이다.
더욱이, 각 조직의 ECM은 조성 (composition) 및 국소해부학 (topology) 관점에서 독특하며, 상기 조성 및 해부학 특성은 상주하는 세포 (resident cells)와 미세환경 사이의 역동적이고 상호적인 상호작용을 통해 생성된다. 조직 및 장기로부터 분리된 세포와 ECM의 최근 연구는 선택된 세포 기능 및 표현형 (phenotype)을 보존하기 위하여 조직 특이성 (tissue specificity)의 필요성을 강조하고 있다 (Sellaro, T. L. et al. Tissue Eng. Part A 16, 1075-1082 (2010); Petersen, T. H. et al. Science 329, 538-541 (2010); Uygun, B. E. et al. Nat. Med. 16, 814-821 (2010); Ott, H. C. et al. Nat. Med. 16, 927-933 (2010); Flynn, L. E. Biomaterials 31, 4715-4724 (2010)).
수확된 dECM 재료는 전형적으로 피부, 소장 점막하조직 (small intestinal submucosa)을 포함한 다양한 조직으로부터 2차원 (two-dimensional, 2D) 스캐폴드로서 가공되며, 초기 단계에서 침투성 또는 시딩된 세포는 지지성 혈관 네트워크 (supporting vascular network)가 생길 때까지 생존을 위해 산소 및 영양소의 확산에 의존한다.
그러나, 프린팅된 조직 유사체 구조는 영양소의 흐름을 허용하도록 개방된 다공성 3D 구조를 고안하는 제조방법을 필요로 한다. 본 발땅자들은 3D 구조 조직의 성장에 적합한 최적화된 미세환경을 제공할 수 있는 dECM 바이오잉크를 사용한 세포 -담지 (cell-laden) 구조의 프린팅을 위한 3차원 프린팅' 방법을 개발한 바 있으며, 상기 세포 -담지 구조는 고유의 세포 형태 및 기능을 재현할 수 있다 (Falguni Pati, et al., Nat Commun. 5,3935 (2014)).
한편, 허혈성 심장질환은 전세계적으로 매우 높은 유병률을 보이고 있으며, 단일질환 전체 사망원인 증 1위를 차지하고 있다. 한국 역시 사회 경제적인 발전과 서구화된 생활습관으로 인하여 허혈성 심장질환 환자들이 급증하고 있다. 더욱이 중증의 말기 심부전 환자의 경우, 심장이식술이나 기계적 좌심실 보조장치 이의에는 완치법이 없는 실정이다. 하지만 공여장기의 부족, 장기확보의 어려움, 높은 사망률 및 고가의 치료비용 등의 문제점을 가지고 있다.
줄기세포의 다분화능력 (multipotency)을 이용한 치료법은 손상된 조직재생을 근본적으로 치료할 수 있을 것으로 기대되고 있다. 최근에는 성체줄기세포를 혈관내피세포 및 기타 간질세포와 배합하거나 다양한 하이드로젤과 흔합하여 주입함으로써, 심근세포로의 분화를 촉진하고 세포가 병변으로부터 이탈하는 현상을 방지하여, 심장 질환 치료의 효율을 높이는 연구가 다양하게 진행되고 있다. 또한, 줄기세포 또는 줄기세포와 다양한 성체세포를 이용하여 세포 시트 (cell sheets)를 제작함으로써 줄기세포의 생체 환경을 모사하고 세포간의 상호작용을 극대화 하는 연구도 활발히 진행 중이다.
【발명의 내용】
【해결하고자 하는 과제】 본 발명의 일 예는 탈세포화된 세포외 기질 및 가교제를 포함하는 조직공학용 구조체 형성용액, 및 심장전구세포를 포함하는 계 1 바이오프린팅용 조성물과, 상기 조직공학용 구조체 형성용액, 중간엽 줄기세포 및 혈관내피성장인자를 포함하는 제 2 바이오프린팅용 조성물을 인쇄 및 가교화하여 제 1 바이오프린트층 및 제 2 바이오프린트층을 교대로 배치하여 3차원 구조체를 형성하는 단계; 및 상기 가교된 3차원 구조체를 열적겔화하여 가교 -겔화 3차원 구조체를 얻는 단계를 포함하는, 심근조직 재생용 3차원 구조체의 제조방법에 관한 것이다.
본 발명의 또 다른 일 예는 계 1 바이오프린팅용 조성물과, 저 12 바이오프린팅용 조성물을 인쇄하여 제조되는 제 1바이오프린트층 및 제 2바이오프린트층을 가교화하고, 열적겔화하여 형성된 심근조직 재생용 3차원 구조체에 관한 것이다.
【과제의 해결 수단】
본 발명자들은 심근조직의 미세환경 (microenvironment)이 구현된 3차원 구조체의 제조방법을 개발하고자 다양한 연구를 수행하였다. 이에, 본 발명자들은 탈세포화된 세포외 기질 및 심장전구세포 (cardiac progenitor cells)를 포함하는 바이오프린팅용 조성물과 탈세포화된 세포외 기질, 중간엽 줄기세포 (mesenchymal stem cells), 및 혈관내피성장인자 (vascular endothelial growth factor, VEGF)를 포함하는 바이오프린팅용 조성물을 교대로 사용한 척층 공정을 통한 3차원 프린팅을 수행하였을 때, 심근조직의 미세환경을 효과적으로 구현할 수 있다는 것을 발견하였다. 특히, 이러한 3차원 프린팅 방법은 심장전구세포를 구조체 내에 균등하게 위치시킬 뿐만 아니라, 구조체 내에 혈관세포로 이루어진 혈관 네트워크를 구현함으로써, 세포의 생존능을 장기간 유지시킬 수 있어 심근내부로의 세포전달 효율을 현저하게 향상시킬 수 있다는 것을 발견하였다.
이에 본 발명의 일 예는,
(a) 탈세포화된 세포외 기질 및 가교제를 포함하는 조직공학용 구조체 형성용액 및 심장전구세포를 포함하는 계 1 바이오프린팅용 조성물과, 상기 조직공학용 구조체 형성용액, 중간엽 줄기세포 및 혈관내피성장인자를 포함하는 제 2 바이오프린팅용 조성물로 인쇄 및 가교화하여 제 1 바이오프린트층 및 제 2 바이오프린트층을 교대로 배치하여 3차원 구조체를 형성하는 단계; 및
(b) 상기 가교된 3차원 구조체를 열적겔화하여 가교 -겔화 3차원 구조체를 얻는 단계;
를 포함 하는 심근조직 재생용 3차원 구조체의 제조방법에 관한 것이다.
상기와 같이 탈세포화된 세포외 기질 및 심장전구세포를 포함하는 제 1 바이오프린팅용 조성물과; 탈세포화된 세포외 기질, 증간엽 줄기세포 및 혈관내피성장인자를 포함하는 게 2 바이오프린팅용 조성물을 교대로 사용하는 프린팅을 통한 적층 공정을 수행하여 제조되는 3차원 구조체는, 심장전구세포가 구조체 내에 균등하게 위치하고 있을 뿐만 아니라, 구조체 내에 혈관세포로 이루어진 혈관 네트워크를 구현함으로써, 심근조직의 미세환경을 효과적으로 구현할 수 있어 세포의 생존능을 장기간 유지시킬 수 있으므로 심근내부로의 세포전달 효율을 현저하게 향상시킬 수 있는 효과가 있다.
상기 탈세포화된 세포외 기질 (decellularized extracellular matrix)은 심장 조직에서 유래된 것일 수 있다. 구체적으로는 체외로 배출된 심장조직을 탈세포화함으로써 얻어질 수 있으며, 인간, 돼지, 소, 토끼, 개, 염소, 양, 닭, 말 등의 포유동물로부터 배출된 조직을 탈세포화함으로써 얻어질 수 있으며, 바람직하게는 돼지로부터 얻어진 조직, 더욱 바람직하게는 돼지로부터 얻어진 심장 조직을 탈세포화함으로써 얻어질 수 있으나, 이에 한정되는 것은 아니다.
상기 탈세포화는 공지의 방법, 예를 들어 Ott, H. C. et al. Nat. Med. 14, 213-221 (2008), Yang, Z. et al. Tissue Eng. Part C Methods 16, 865-876 (2010) 등에 개시된 방법을 사용하여 혹은 약간 변형하여 수행될 있다. 바람직하게는, 본 발명자들이 보고한 탈세포화 방법, 즉 Falguni Pati, et al., Nat Commun. 5, 3935 (2014)에 개시된 탈세포화 방법을 사용하여 수행될 수 있다. 상기 탈세포화된 세포외 기질은 통상 동결 건조된 분말형태로 보관될 수 있다.
상기 탈세포화된 세포외 기질의 사용량은 특별히 제한되는 것은 아니나, 예를 들어 조성물 총 중량에 대하여 1 내지 4 중량0 /0, 1 내지 3 중량0 /0, 2 내지 4 중량 %, 바람직하게는 2 내지 3 중량 %의 함량으로 사용될 수 있다. 예를 들어, 제 1 바이오프린팅용 조성물 중의 상기 탈세포화된 세포외 기질은, 제 1 바이오프린팅용 조성물 총 중량에 대하여, 1 내지 4 중량%의 함량으로 존재할 수 있으며; 제 2 바이오프린팅용 조성물 중의 상기 탈세포화된 세포외 기질은, 제 2 바이오프린팅용 조성물 총 중량에 대하여, 1 내지 4 중량 %의 함량으로 존재할 수 있다. 상기 범위를 벗어나 함량이 적은 경우에는 재료가 적층 되지 않는 문제점이 발생하며, 함량이 높은 경우에는 내부에 봉입 된 세포가사멸하는 문제점이 발생한다.
상기 심장전구세포 및 혈관내피세포는 사람을 포함한 동물로부터 유래된 세포로서 상업적으로 구입가능하며 , 공지의 방법에 의해 배양, 증식시킬 수 있다.
상기 심장전구세포는 제 1 바이오프린팅용 조성물 중 105 내지 108 cells/ml, 106 내지 108 cells/ml, 105 내지 5 X 107 cells/ml, 바람직하게는 106 내지 5 X 107 cells/ml의 범위로 존재할 수 있다. 상기 범위를 벗어나 함량이 적은 경우에는 효능이 낮은 문제점이 발생하며, 함량이 높은 경우에는 세포를 감싸고 있는 바이오잉크-탈세포화 된 세포외 기질을 가교시켜도 탈세포화된 세포외 기질이 가교되지 못하여 3차원 형상을 유지시키지 못하는 문제점이 발생한다.
상기 중간엽 줄기세포는 제 2 바이오프린팅용 조성물 중 105 내지 108 cells/ml, 106 내지 108 cells/ml, 105 내지 5 X 107 cells/ml, 바람직하게는 106 내지 5 X 107 cells/ml 의 범위로 존재할 수 있다. 상기 범위를 벗어나 함량이 적은 경우에는 효능이 낮은 문제점이 발생하며, 함량이 높은 경우에는 세포를 감싸고 있는 바이오잉크-탈세포화 된 세포외 기질을 가교시켜도 탈세포화된 세포외 기질이 가교되지 못하여 3차원 형상을 유지시키지 못하는 문제점이 발생한다.
또한, 상기 혈관내피성장인자는 제 2 바이오프린팅용 조성물 중 50 내지 l OOOng/ml, 50 내지 900ng/ml, 50 내지 800ng/ml, 50 내지 700ng/ml, 50 내지 600ng/ml, 50 내지 500ng/ml, 50 내지 400ng/ml, 50 내지 300ng/ml, 50 내지 200ng/ml, 100 내지 1000ng/ml, 100 내지 900ng/ml, 100 내지 8()Ong/ml, 100 내지 700ng/ml, 100 내지 600ng/ml, 100 내지 500ng/ml, 100 내지 400ng/ml, 100 내지 300ng/ml, 바람직하게는 100 내지 200 ng/ml의 범위로 존재할 수 있다. 상기 범위를 벗어나 함량이 적은 경우에는 효능이 낮은 문제점이 발생하며, 함량이 높은 경우에는 과다한 혈관형성으로 인한 문제점이 발생한다.
저 U 바이오프린팅용 조성물 및 계 2 바이오프린팅용 조성물은, 효율적인 3차원 프린팅을 위하여, pH 6.5 내지 7.5의 범위를 갖는 점탄성의 균일한 용액의 형태를 가지는 것이 바람직하다.
따라서, 상기 조직공학용 구조체 형성용액은 산; 단백질 분해효소; 및 pH 조절제를 추가로 포함할 수 있다. 상기 산, 단백질 분해효소, 및 pH 조절제는 수성 매질 중에 포함된 것일 수 있으나, 아에 한정되는 것은 아니다.

제 1 바이오프린팅용 조성물 및 제 2 바이오프린팅용 조성물은 전단율이 증가할수록 점도가 낮아지는 점탄성 물질인 것이 바람직하며, 예를 들어 , 약 15 °C에서 측정될 때 전단율 (shear rate) 1 에서의 점도가 1 내지 30 Pa-S의 범위인 것이 바람직하다. 상기 점도는 수성 매질 (예를 들어, 물, 증류수, PBS, 생리식염수등)의 양을 적절히 조절함으로써 조절될 수 있다. 상기 게 1 바이오프린팅용. 조성물과 제 2 바이오프린팅용 조성물을 교대로 사용하는 프린팅은 공지의 3차원 프린팅 방법 (예를 들어, '다축 조직 장기 프린팅 시스템 '을 사용한 프린팅 방법)을 사용하여, Falguni Pati, et aL, zt Commun. 5, 3935 (2014) 등에 개시된 방법에 따라 수행될 수 있으며, 예를 들어, 다축 조직 장기 프린팅 시스템의 2개의 시린지를 이용하여 상기 프린팅을 수행할 수 있다.
즉, 폴리카프로락톤 프레임웍 (polycaprolactone (PCL) framework)을 시린지에 로딩하고, 약 80 °C로 가열하여 중합체를 용융시킨다. 상기한 프리-겔 형태의 3차원 프린팅용 조성물을 다른 시린지에 로딩하고, 온도를 약 15 °C 이하, 바람직하게는 약 4 내지 10°C로 유지시킨다. PCL 프레임웍의 제작 (fabrication)을 위해 400 내지 650 kPa 범위로 공기압 (pneumatic pressure)을 가한다. 상기 프리-겔 형태의 조성물을 플런저-계 저용량 분사 시스템 (plunger-based low-dosage dispensing system)을 사용하여 분사한다.
또한, 상기 제 1 바이오프린팅용 조성물과 제 2 바이오프린팅용 조성물을 교대로 사용하는 프린팅은 폴리카프로락톤 프레임웍의 사용 없이 상기 프리-겔 형태의 조성물만을 플런저-계 저용량 분사 시스템 (plunger-based low-dosage dispensing system)을 사용하여 분사함으로써 수행될 수도 있다. 일 구현예에서 상기 계 1 바이오프린팅용 조성물과 제 2 바이오프린팅용 조성물을 교대로 사용하는 적층은 제 1 바이오프린팅용 조성물과 제 2 바이오프린탕용 조성물을 서로 일정 교차각을 갖도록 배열되게 적층할 수 있다.
또한, 상기 제 1 바이오프린팅용 조성물과 제 2 바이오프린팅용 조성물은 섬유, 테이프, 또는 직물 형태로 프린팅될 수 있다.
일 예로서 상기 제 1 바이오프린팅용 조성물 및 제 2 바이오프린팅용 조성물은 각각 섬유 형태로 하나의 층에 프린팅되고, 상기 하나의 층은 제 1 바이오프린팅용 조성물이 프린팅된 섬유와 제 2 바이오프린팅용 조성물이 프린팅된 섬유가 서로 교대된 일방향 섬유 형태로 제조된 층일 수 있으며, 또 다른 층은 상기 층과 일정 교차각을 갖도록 적층된 것일 수 있다. 즉,' 0도 방향 1층 90도 방향 1층을 겹쳐 적층한 구조일 수 있으며, 이러한 적층 형태의 일 예는 도 3에 나타낸다.
일 구현예에서 상기 가교제는 리보플라빈일 수 있다. 상기 리보폴라빈은, 제 1 바이오프린팅용 조성물 또는 게 2 바이오프린팅용 조성물 총 중량에 대하여, 0.001 내지 0.1 중량0 /0, 바람직하게는 0.01 내지 0.1 증량 %의 함량으로 존재할 수 있다. 상기 범위를 설정한 이유는 심장조직과 유사한 기계적 특성을 나타내는 효과가 있기 때문이다.
또한, 상기 각각의 적층 공정은 상기 프린팅 및 UVA 하에서의 가교화를 통하여 수행되는 것일 수 있다. 상기 프린팅 및 UVA 하에서의 가교화를 반복적으로 수행함으로써, 즉 적층 공정 (layer-by-layer process))을 수행함으로써 , 3차원 구조체 형상이 형성된다.
높은 안전성을 갖는 린보플라빈을 가교쎄로서 사용하여 3차원 프린팅 및 UVA 하에서의 가교화를 통한 적층 공정을 수행하여 3차원 구조체 형상을 제작한 다음, 얻어진 3차원 구조체 형상을 열적 겔화함으로써 높은 기계적 강도를 균일하게 갖는 3차원 구조체를 제조할 수 있는 효과가 있다. 상기 UVA하에서의 가교화는 315 내지 400 nm, 325 내지 390 nm, 335 내지 380 nm, 345 내지 370 nm, 355 내지 365 nm, 바람직하게는 약 360 nm 파장의 UVA를 사용할 수 있다.
또한, 상기 UVA 하에서의 가교화는 1 내지 10분, 1 내지 9분, 1 내지 8분, 1 내지 7분, 1 내지 6분, 1 내지 5분, 1 내지 4분, 2 내지 10분, 2 내지 9분, 2 내지 8분, 2 내지 7분, 2 내지 6분, 2 내지 5분, 2 내지 4분, 바람직하게는 약 3분 동안 수행될 수 있다. 즉, 리보플라빈 사용을 포함하는 가교 -열적 겔화 방법 (crosslinking-thermal gelation)에 의해 높은 기계적 강도를 균일하게 갖는 3차원 구조체를 제조할 수 있다는 것이 본 발명에 의해 밝혀졌다. 따라서, 리보플라빈을 사용하는 상기 가교 -열적 겔화 방법에 의해 높은 기계적 강도를 제공할 수 있을 뿐만 아니라, 글루타르알데히드 (glutaraldehyde)와 같은 독성 가교제의 사용을 필요로 하고 또한 3차원 구조체 내부에는 불균일한 가교를 야기하는 프린트-후 가교 (post-print crosslinking)의 문제점을 회피할 수 있다.
따라서, 상기 3차원 구조체는 1 rad/s에서의 모들러스가 1 내지 100 kPa, 1 내지 90 kPa, 1 내지 80 kPa, 1 내지 70 kPa, 1 내지 60 kPa, 1 내지 50 kPa, 1 내지 40 kPa, 1 내지 30 kPa, 1 내지 20 kPa, 예를 들어, 10.58 kPa인 것일 수 있다. 상기 범위를 설정한 이유는 심장 근육과 유사한 기계적 강도로 주변 환경을 모사하는 효과가 있기 때문이다.
일 구현예에서, 제 1 바이오프린팅용 조성물 및 제 2 바이오프린팅용 조성물은 독립적으로, 각각의 조성물 총 중량에 대하여, 리보플라빈 0.001 내지 0.1 증량0 /0; 아세트산 및 염산으로 이루진 군으로부터 1종 이상 선택된 산 0.03 내지 30 중량0 /。; 펩신 및 매트릭스 메탈로프로티나아제로 이루어진 군으로부터 1종 이상 선택된 단백질 분해효소 으1 내지 0.4 중량0 /0; 및 ρΗ 조절제를 추가로 포함할 수 있다.
상기 구현예에 따른 게 1 바이오프린팅용 조성물 및 저 12 바이오프린팅용 조성물은,
아세트산 및 염산으로 이루진 군으로부터 선택된 1종 이상의 산 용액에 탈세포화된 세포외 기질을 첨가하는 단계;
펩신 및 매트릭스 메탈로프로티나아제로 이루어진 군으로부터 선택된 1종 이상의 단백질 분해효소를 첨가하는 단계; 및
리보플라빈, ρΗ 조절제 및 심장전구세포 (제 1 바이오프린팅 조성물의 경우), 또는 리보플라빈, ρΗ 조절제, 중간엽 줄기세포 및 혈관내피성장인자 (제 2 바이오프린팅 조성물의 경우)를 첨가하는 단계;
를 포함하는 제조 방법에 의해 제조될 수 있다.
상기 단백질 분해효소를 가한 용액을 얻는 단계는 단백질 분해효소를 가한 후 교반을 수행할 수 있으며, 상기 교반은 탈세포화된 세포외 기질의 완전한 용해를 달성할 때까지 수행될 수 있으며, 예를 들어, 통상 24 내지 48 시간 동안 수행될 수 있으나, 이에 제한되는 것은 아니다.
상기 인쇄 및 가교화는 겔화를 방지하기 위하여 15°C 이하에서 수행되는 것일 수 있으며, 예를 들어, 4 내지 15°C, ·바람작하게는 4 내지 10°C의 저온에서 수행되는 것이 바람직하다.
또한, 상기 열적겔화는 15°C 초과에서 수행되는 것일 수 있으며, 예를 들어, 20°C 이상, 25°C 이상, 바람직하게는, 20°C 내지 50°C, 20°C 내지 40°C, 20°C 내지 3TC, 25 °C 내지 50°C, 25 °C 내지 40°C, 더욱 바람직하게는, 25°C 내지 37°C의 온도에서 수행되는 것일 수 있다.
상기 제조 방법에 의해 제조되는 게 1 바이오프린팅용 조성물 및 저 12 바이오프린팅용 조성물은 pH-조절된 프리-겔 (pH-adjusted pre-gel) 형태로서, 약 4°C에서 넁장 보관하는 것이 바람직하다.
또한, 상기 열적겔화는 15°C 초과에서 수행되는 것일 수 있으며, 예를 들어, 20°C 이상, 25°C 이상, 바람직하게는 , 20°C 내지 50°C, 20°C 내자 40°C, 20°C 내지 37°C, 25 °C 내지 50°C, 251 내지 40°C, 더욱 바람직하게는, 25°C 내지 37°C의 온도에서 수행되는 것일 수 있다. 상기 범위를 설정한 이유는 탈세포화 된 세포외 기질을 물리적으로 가교하는 효과가 있기 때문이다. 상기 열적 겔화는 인큐베이터 (humid incubator) 내에서 수행 할 수 았다.
또한, 상기 열적 겔화는 5 내지 60 분, 10 내지 50 분, 15 내지 40 분, 바람직하게는 20 내지 30 분 동안 수행하는 것일 수 있다. 상기 범위를 설정한 이유는 물리적 가교화 반웅이 일어나는데 요구되는 범위이기 때문이다.

상기 심근조직 재생용 3차원 구조체는, 탈세포화된 세포외 기질 및 가교제를 포함하는 조직공학용 구조체 형성용액, 및 심장전구세포를 포함하는 제 1 바이오프린팅용 조성물과, 상기 조직공학용 구조체 형성용액, 중간엽 줄기세포 및 혈관내피성장인자를 포함하는 제 2 바이오프린팅용 조성물을 인쇄 및 가교화에 의해 교대로 적층된 구조를 1 이상 포함하는 것일 수 있다.
제 1 바이오프린팅용 조성물, 제 2 바이오프린팅용 조성물, 탈세포화된 세포외 기질, 심장전구세포, 중간엽 줄기세포, 혈관내피성장인자에 관한 사항은 상기 기술한 바와 동일하다.

【발명의 효과】
본 발명은 특정 세포, 즉 심장전구세포 (cardiac progenitor cells) 및 /또는 중간엽 줄기세포 (mesenchymal stem cells)와 혈관내피성장인자 (vascular endothelial growth factor, VEGF)를 포함하는 바이오프린팅용 조성물 (들)를 사용하여 3차원 프린팅을 수행함으로써, 심근조직의 미세환경을 효과적으로 구현할 수 있다.
또한, 본 발명의 제조방법은 심장전구세포를 구조체 내에 균등하게 위치시킬 뿐만 아니라, 구조체 내에 혈관세포로 이루어진 혈관 네트워크를 구현함으로써 세포의 생존능을 장기간 유지시킬 수 있어, 심근내부로의 세포전달 효율을 현저하게 향상시킬 수 있다. 따라서, 본 발명의 제조방법은 허혈성 심장질환을 포함한 다양한 심장질환의 치료를 위한 3차원 구조체의 제조에 유용하게 적용될 수 있다. 【도면의 간단한 설명】

1-1: 탈세포화된 세포외 기질 제조
탈세포화된 세포외 기질은 돼지의 심장 조직을 사용하여 Falguni Pati, et al., Nat Commun. 5, 3935 (2014)에 개시된 방법에 따라 제조하였다 (이하, 'hdECM'). 상기 제조된 hdECM은 최종적으로 동결 건조하여 사용 전까지 냉동 보관하였다. 상기 제조된 hdECM의 광학 현미경 사진 및 조직염색 사진을 도 2에 나타내었다.
1-2: 프리-겔 (pre-gel)상태의 조직공학용 구조체 제조 용액 제조 상기 얻어진 동결 건조된 hdECM에 액체 질소를 붓고 모르타르 및 막자로 분쇄하였다ᅳ 상기 얻어진 hdECM 분말 (330 mg)을 0.5M 아세트산 수용액 (10 ml)에 첨가한 후 펩신 (33 mg)(P7125, Sigma-Aldrich)을 가한 다음 상온에서 48시간 동안 교반하였다. 얻어진 용액의 온도를 KTC 이하로 유지하면서, 리보플라빈 (2 mg)을 가하고 10°C 이하로 넁각시킨 10M NaOH 용액을 적가하여 pH를 약 pH 7로 조절하였다. 상기 얻어진 프리-겔 (pre-gel) 상태의 조직공학용 구조체 제조 용액은 약 4°C에서 넁장 보관하였다.
1- 3: 프리-겔 (pre-gel) 상태의 조직공학용 구조체 제조 용액과 세포를 포함하는프린팅 조성물의 제조
프린팅 공정 수행 직전에 상기 프리-겔 상태의 조직공학용 구조체 제조 용액에 심장전구세포 5 X 106 cells/ml를 첨가하여 제 1 바이오프린팅용 조성물을 제조하였다. 상기 심장전구세포는 인간 심근조직 유래 심장전구세포, 부산대학교 의학전문대학원, 생리학 교실에서 얻은 것이다. 제조예 2: 제 2바이오프린팅용조성물의 제조
2- 1: 탈세포화된 세포외 기질 제조
탈세포화된 세포외 기질은 돼지의 심장 조직을 사용하여 Falguni Pati, et al., Nat Commun. 5, 3935 (2014)에 개시된 방법에 따라 제조하였다 (이하, •hdECM'). 상기 제조된 hdECM은 최종적으로 동결 건조하여 사용 전까지 냉동 보관하였다. 상기 제조된 hdECM의 광학 현미경 사진 및 조직염색 사진을 도 2에 나타내었다.
2-2: 프리-겔 (pre-gel)상태의 조직공학용구조체 제조 용액 제조 상기 얻어진 동결 건조된 hdECM에 액제 질소를 붓고 모르타르 및 막자로 분쇄하였다. 0.5M 아세트산 수용액 (10 ml)에 hdECM 분말 (330 mg)을 첨가한 후, 펩신 (33 mg)(P7125, Sigma-Aldrich)을 가한 다음 상온에서 48시간 동안 교반하였다. 얻어진 용액의 온도를 10 °C 이하로 유지하면서, 리보플라빈 (2 mg)을 가하고 KTC 이하로 냉각시킨 10M NaOH 용액을 적가하여 pH를 약 pH 7로 조절하였다. 상기 얻어진 프리-겔 (pre-gel) 상태의 조직공학용 구조체 용액은 약 4 °C에서 넁장 보관하였다.
2-3: 프리-겔 (pre-gel) 상태의 조직공학용 구조체 제조 용액과 세포를 포함하는프린팅 조성물의 제조
프린팅 공정 수행 직전에 상기 프리-겔 상태의 조직공학용 구조체 용액에 중간엽 줄기세포 5 X 106 cells/ml와 혈관내피 성장인자 (제품번호: 293-VE-lO, R&D systems사 )(100 ng/ml)를 각각 첨가하여 제 2 바이오프린팅용 조성물을 제조하였다. 상기 중간엽 줄기세포는 (하비갑개 (inferior turbinate) 유래 중간엽 줄기세포, 가를릭대학교 이비인후과학 교실에서 얻은 것이다. 비교예 1:조직공학용 3차원 구조체의 제조
제조예 1에서 얻어진 계 1 바이오프린팅용 조성물을 사용하여 Falguni Pati, et al., Nat Commun. 5, 3935 (2014)에 개시된 방법에 따라 3차원 구조체를 제작하였다.
구체적으로, PCL 프레임웍 (polycaprolactone (PCL) framework)을 다축 조직 장기 프린팅 시스템 (Jin-Hyung Shim et al., J. Micromech. Microeng. 22 085014 (2012))의 시린지 (제 1 시린지)에 로딩하고, 약 80°C로 가열하여 중합체를 용융시켰다. 제조예 1에서 얻어진 프리-겔 상태의 제 1 바이오프린팅용 조성물을 다른 시린지 (제 2 시린지)에 로딩하고, 온도를 약 10°C 이하로 유지시켰다. 제 1 시린지에 약 600 kPa의 공기압을 가하여 약 100 urn 이하의 선폭을 가지고, 약 300 urn 의 공극을 지닌 120 urn두께의 얇은 PCL 프레임웍을 제작하고, PCL 프레임웍 위에 제 2 시린지의 내용물을 분사한 후, 약 360 nm의 UVA를 3분 동안 조사하여 가교화하였다. 이후, 제 2 시린지의 내용물 분사 및 상기 가교화를 통한 적층 공정 (kyer-by-layer process)을 수행하여 3차원 구조체 형상을 형성하였다. 얻어진 3차원 구조체 형상을 약 37 °C의 인큐베이터 (humid incubator)에 넣고 30분 동안 유지시켜 열적 겔화시켜 3차원 구조체 ('CPC printed'라 칭함)를 제조하였다. 얻어진 3차원 구조체는 약 300 내지 400 um두께를 가진다. 실시예 1:조직공학용 3차원 구조체의 제조
제조예 1 및 2에서 얻어진 계 1 바이오프린팅용 조성물 및 제 2 바이오프린팅용 조성물을 사용하여 3차원 구조체를 제작하였다.
구체적으로, PCL 프레임웍 (polycaprolactone (PCL) framework)을 다축 조직 장기 프린팅 시스템 (Jin-Hyung Shim et al., J. Micromech. Microeng. 22 085014 (2012))의 시린지 (제 1 시린지)에 로딩하고, 약 8( C로 가열하여 중합체를 용융시켰다. 제조예 1에서 얻어진 프리-겔 형태의 제 1 바이오프린팅용 조성물 및 제조예 2에서 얻어진 게 2 바이오프린팅용 조성물을 다른 시린지 (각각, 제 2, 3 시린지)에 로딩하고, 온도를 약 10 °C 이하로 유지시켰다. 제 1 시린지에 약 600 kPa의 공기압을 가하여 약 100 um 이하의 선폭을 가지고, 약 300 um의 공극을 지닌 120 um 두께의 얇은 PCL 프레임웍을 제작하고, PCL 프레임웍 위에 제 2 시린지의 내용물을 분사한 후, 약 360 nm의 UVA를 3분 동안 조사하여 가교화하였다. 이후, 제 2, 3 시린지의 내용물 분사 및 상기 가교화를 통한 적층 공정 (layer-by-layer process)을 수행하여 3차원 구조체 형상을 형성하였다.
얻어진 3차원 구조체 형상을 약 37°C의 인큐베이터 (humid incubator)에 넣고 30분 동안 유지하여 열적 겔화시켜 조직공학용 3차원 구조체 ('CPC/MSC printed'라 칭함)를 제조하였다ᅳ 얻어진 3차원 구조체는 약 300 내지 400 urn 두께를 가지며, 그 형상은 도 3과 같다. 시험예 1. 리보플라빈 첨가 유무에 따른 모듈러스 (complex modulus) 측정
제조예 1에서 얻어진 프리-겔 형태의 용액을 약 360 nm의 UVA : 3분 동안 조사하여 가교화한 후, 약 37°C의 인큐베이터 (humid incubator)에 넣고 30분동안 유지하여 열적 겔화를 유도하여 하이드로겔을 형성시켰다 (가교 하이드로겔 A). 또한, 리보플라빈을 사용하지 않은 것을 제외하고는 제조예 1과 동일한 방법으로 제조한 프리-겔 상태의 조직공학용 구조체 제조용액을 약 37°C의 인큐베이터 (humid incubator)에 넣고 30분 동안 유지하여 열적 겔화를 유도하여 하이드로겔을 '형성시켰다 (하이드로겔 B).
그 다음, 얻어진 각각의 하이드로겔에 대하여, 빈도 (frequency) 1 rad/s에서의 복소 모들러스 (complex modulus)를 측정하였으며, 그 결과는 하기 표 1과 같다.

상기 표 1의 결과로부터 알 수 있는 바와 같이, 본 발명에 따른 가교 하이드로겔 A는 1 rad/s에서의 모들러스가 10.58 kPa로서, 가교 없이 겔화 처리만 수행된 하이드로겔 B에 비해서 가교화에 의해 약 30배 이상의 강도 향상을 나타냄을 알 수 있었다. 시험예 2. 4 내지 5 주령의 랫트 (무게: 300±30g)의 흉부를 절개하고, 좌전하행동맥 (Left Anterior Descending artery, LAD)의 영구 결찰 (permanent ligation)을 시행하여 심근경색을 유발하였다.
7일 후에, 비교예 1에서 제조한 3차원 구조체 (CPC printed) 및 실시예 3에서 제조한 3차원 구조체 (CPC/MSC printed)를 각각 8 ram 직경의 크기로 잘라, 심근경색 부위에 이식한 후 봉합하였다 (각각 n=7). 이식 후 4주 및 8주차에 심장초음파를 촬영하여 좌심실 내경 (left ventricle diameter, LV diameter)을 확인하여 그 결과를 도 . 5에 나타내었으며, 심근의 기능을 보여주는 좌심실 박출 계수 (left ventricle ejection fraction, LVEF) 및 구획 단축률 (fractional shortening, FS)을 계산하여 student T-test에 의한 실험군 간의 유의성 평가를 수행하여 그 결과를 도 6에 나타내었다.
도 5 및 6로부터 확인할 수 있는 바와 같이, 실시예 3의 본 발명에 따른 3차원 구조체 이식 4주 및 8주 후 아무것도 처리하지 않은 실험군 (대조군) 대비 좌심실 내경이 현저히 줄어들었다. 또한 CPC 그룹에 비하여 CPC/MSC 그룹에서 좌심실 수축 기능이 유의하게 증가한 것을 관찰할 수 있으며, 이는 구조체에 의해 심장 리모델링 (cardiac remodeling)이 저해됨을 나타낸다.
이식 8주후 쥐의 심장을 적출하여 개체마다 같은 위치 (심장의apex에서 base 방향으로 약 1/3 지점)에서 조직을 획득한 후 (3um 두께) Masson's trichrome 염색을 실시하였다. 해당 염색법을 이용하여 심장 근육 (빨간색)과 심근경색 부위의 반흔 조직 (파란색)을 구별하여, 그 결과를 도 7에 나타내었다.
도 7로부터 확인할 수 있는 바와 같이, 본 발명에 따라 제조한 3차원 구조체 이식 8주후의 좌심실 내벽의 두께가 대조군에 비하여 두껍게 유지되었으며 심근경색 부위의 반흔 조직 (scar tissue)의 형성이 적었다. 또한, 비교예 1과 같이 제 1 바이오프린팅용 조성물 단독으로 형성시킨 3차원 구조체 (CPC printed)에 비하여, 실시예 3과 같이 제 1 바이오프린팅용 조성물 및 제 2 바이오프린팅용 조성물로 형성시킨 3차원 구조체 (CPC/MSC printed)를 이식하였을 때 반흔 조직 형성이 더욱 효과적으로 억제되었고, 좌심실 내벽이 두께가 얇아지지 않고 유지되었다.
도 8로부터 확인할 수 있는 바와 같이, 본 발명에 따라 제조한 3차원 구조체를 이식한 심근경색 부위에 심근조직의 수축성을 대표하는 베타 마이오신 중쇄 (β-myosin heavy chain, pMHC)와 혈관형성을 대표하는 분화클러스터 31 (CD31)의 발현양이 현저히 증가하였고, 특히 저 U 바이오프린팅용 조성물 및 계 2 바이오프린팅용 조성물로 형성시킨 3차원 구조체 (CPC/MSC printed)(실시예 3)를 이식하였을 때 그 효과가 더욱 극대화되었다.
따라서, 본 발명의 제조방법에 의해 제조된 3차원 구조체는 구조체 내에 혈관 네트워크를 구현함으로써 (즉, 심근조직의 미세환경을 효과적으로 구현함으로써), 세포의 생존등을 장기간 유지시킬 수 있어 심근내부로의 세포전달 효율을 현저하게 향상시킬 수 있음을 확인하였다.

Claims

【특허청구범위】
【청구항 1】
(a) 탈세포화된 세포외 기질 및 가교제를 포함하는 조직공학용 구조체 형성용액, 및 심장전구세포를 포함하는 게 1 바이오프린팅용 조성물과: 상기 조직공학용 구조체 형성용액, 중간엽 줄기세포 및 혈관내피성장인자를 포함하는 제 2 바이오프린팅용 조성물을 인쇄 및 가교화하여 제 1 바이오프린트층 및 제 2 바이오프린트층을 교대로 배치하여 3차원 구조체를 형성하는 단계; 및
(b) 상기 가교된 3차원 구조체를 열적겔화하여 가교 -겔화 3차원 구조체를 얻는 단계를 포함하며,
상기 (a)단계는 열적겔화가 일어나지 않는 온도에서 수행하고, 상기 (b)단계에서 열적겔화를 일으키는 은도에서 수행하는 것인, 심근조직 재생용 3차원 구조체의 제조방법.
【청구항 2】
제 1항에 있어서,
상기 탈세포화된 세포외 기질은 심장 조직에서 유래된 것인, 제조방법.
【청구항 3】
제 1항에 있어서,
상기 제 1 바이오프린팅용 조성물 중의 탈세포화된 세포외 기질은, 제 1 바이오프린팅용 조성물 총 중량에 대하여, 1 내지 4 중량%의 함량으로 존재하는 것인, 제조방법.
【청구항 4】
제 1항에 있어서,
상기 제 2 바이오프린팅용 조성물 중의 탈세포화된 세포외 기질은, 제 2 바이오프린팅용 조성물 총 중량에 대하여, 1 내지 4 중량 %의 함량으로 존재하는 것인, 제조방법.
【청구항 5】
제 1항에 있어서,
상기 심장전구세포는, 제 1 바이오프린팅용 조성물 증 105 내지 108 cells/ml의 범위로 존재하는 것인, 제조방법.
【청구항 6】 제 1항에 있어서,
상기 중간엽 줄기세포는 제 2 바이오프린팅용 조성물 중 105 내지 108 cells/ml로 존재하는 것인, 제조방법.
【청구항 7]
제 1항에 있어서,
상기 혈관내피성장인자는 제 2 바이오프린팅용 조성물 중 50 내지 1000ng/ml의 범위로 존재하는 것인, 제조방법.

제 1항에 있어서,
상기 조직공학용 구조체 형성용액은 pH 6.5 내지 7.5의 범위를 가지며 산 및 단백질 분해효소를 추가로 포함하는 것인, 제조방법.

제 8항에 있어서, 상기 조직공학용 구조체 형성용액은,
바이오프린팅 조성물 총 중량에 대하여,
산 0.03 내지 30 중량0 /0; 및 단백질 분해효소 0.1 내지 0.4중량0 /0; 를 포함하는 것인, 제조방법.

제 8항에 있어서,
상기 조직공학용 구조체 형성용액은,
아세트산 및 염산으로 이루진 군으로부터 선택된 1종 이상의 산; 펩신 및 매트릭스 메탈로프로티나아제 (MMP)로 이루어진 군으로부터 선택된 1종 이상의 단백질 분해효소; 및
pH 조절제;를 추가로 포함하는 것인, 제조방법.
【청구항 11】
제 1항에 있어서,
제 1 바이오프린팅용 조성물 및 제 2 바이오프린팅용 조성물은 독립적으로,
혈관내피전구세포 (endothelial progenitor cells), 혈관내피세포 (endothelial cells) 및 심근세포 (cardiomyocytes)로 이루어진 세포 군에서 선택된 1종 이상의 세포; 및
섬유아세포 성장인자 (fibroblast growth factor, FGF), 혈소판유래 성장인자 (platelet-derived growth factor, PDGF), 안지오포이에틴 -l(angiopoietin-l), 형질전환성장인자 -베타 (transforming growth factor beta, TGF-β), 에리스로포에틴 (erythropoietin, EPO), 줄기세포인자 (stem cell factor, SCF), 표피성장인자 (epidermal growth factor, EGF) 및 콜로니자극인자 (colony stimulating facor, CSF)로 이루어진 성장인자 군에서 선택된 1종 이상의 성장인자;
를 추가로 포함하는 것인, 제조방법.
【청구항 12】
제 11항에 있어서,
제 1 바이오프린팅용 조성물 및 제 2 바이오프린팅용 조성물은 독립적으로,
기질금속단백분해효소 (matrix metalloproteinase, MMP) 및 기질금속단백분해저해효소 (tissue inhibitor matrix metalloproteinase, TIMP)로 이루어진 효소 군에서 선택된 1종 이상의 효소를 추가로 더 포함하는 것인, 제조방법.

제 1항에 있어서,
제 1 바이오프린팅용 조성물 및 제 2 바이오프린팅용 조성물은 독립적으로, 15 °C에서 측정된 전단율 (shear rate) 1 s-1 에서의 점도가 1 내지 30 Pa'S의 범위인 것인, 제조방법.

상기 제 1 바이오프린팅용 조성물은 탈세포화된 세포의 기질 및 가교제를 포함하는 조직공학용 구조체 형성용액 및 심장전구세포를 포함하고,
상기 제 2 바이오프린팅용 조성물은 상기 조직공학용.구조체 형성용액, 중간엽 줄기세포 및 혈관내피성장인자를 포함하는 것인, 심근조직 재생용 3차원 구조체.

제 20항에 있어서, 상기 게 1 바이오프린트층, 제 2 바이오프린트층 또는 이들 모두는 섬유, 테이프, 또는 직물 형태인 것인, 심근조직 재생용 3차원 구조체.
