Abstract

【課題】無菌状態で大規模な培養が可能であり、コンパクトな培養装置10を提供する。 【解決手段】培養装置10は、液体が流通するポート73,74を有し、細胞培養が可能な培養バッグ70と、培養容器70を保持する保持板42,43、及び回転軸48,49を有するバッグ保持部31と、回転軸48を支持して回転させる回転機構34と、培養バッグ70へ液体を供給及び排出する液体給排機構37と、回転機構34の回転を制御する回転制御部20と、を備える。回転制御部20は、第1情報に応じて、バッグ保持部31を略水平方向に沿った第1姿勢とし、第2情報に応じて、ポート74から培養バッグ70内の培地が流出されるとき、バッグ保持部31を第2姿勢とし、第3情報に応じて、少なくともポート74から培養バッグ70内の細胞懸濁液が流出されるとき、バッグ保持部31を略鉛直方向に沿った第3姿勢とする。 

Description

培養装置
 本発明は、細胞を培養する装置に関する。 
 細胞培養においては、培養容器中の細胞を他の培養容器に移し替える工程や、培養容器中の培地を交換する工程が行われる。このような細胞培養の各工程は、無菌状態で行われる必要がある。また、細胞培養は、数日から数週間に渡って行われ、この期間に前述された各工程が複数回行われる。細胞培養において人が行う作業を簡略化したり、大量の細胞を培養したりすることを目的として、細胞培養における各工程を機械により行う装置が公開されている(特許文献1,2)。

 特許文献1に記載された細胞培養装置のように、完全に密閉されていない培養容器が用いられる手法では、培養容器が置かれる培養空間が無菌状態に保持される必要があり、装置が大型化しやすいという問題がある。また、培地交換や継代のために培養容器を扱うためのロボットアームやテーブルなどが配置される空間も無菌状態に保持されるという理由によっても、装置が大型化しやすいという問題が生じる。
 バッグのように内部空間を無菌状態に保持可能な培養容器が用いられる手法では、培養容器が配置される空間が無菌状態に保持される必要はない。しかし、バッグ形状の培養容器は撓みやすいので、培地交換や継代などにおいて、バッグの内部空間に対して液体を流出入させることが難しい。
 本発明は、これらの事情に鑑みてなされたものであり、その目的は、培養容器を用いて容易に細胞培養ができる培養装置を提供することにある。
 (1) 本発明に係る培養装置は、液体が流通するポートを有しており、内部空間において細胞培養が可能な培養容器と、上記培養容器を保持する保持板、及び回転軸を有する容器保持部と、上記回転軸を支持して回転させる回転機構と、上記培養容器へ液体を供給及び排出する液体給排機構と、上記回転機構の回転を制御する回転制御部と、を備え、上記回転制御部は、上記培養容器において細胞培養が行われることを示す第1情報に応じて、上記容器保持部を上記保持板における上記培養容器を支持する支持面が略水平方向に沿った第1姿勢とし、上記培養容器において溶液交換が行われることを示す第2情報に応じて、少なくとも上記ポートから上記培養容器内の溶液が流出されるとき、上記容器保持部を上記支持面が水平方向及び鉛直方向のいずれにも沿っていない第2姿勢とする。
 第1姿勢の容器保持部に保持された培養容器において、細胞培養が行われる。第1姿勢では、容器保持部の支持面が略水平方向に沿っているので、培養容器の内面の一方が略水平方向に沿った状態に保持される。これにより、培養容器が安定して容器保持部に支持され、例えば接着性細胞が培養容器の内面に付着して培養される。
 第2姿勢の容器保持部に保持された培養容器において、培地や試薬などに代表される溶液交換が行われる。溶液交換において、例えば、培養容器に貯留された溶液の一部が流出される。第2姿勢では、培養容器の内面が水平方向及び鉛直方向のいずれにも沿っていないので、培養容器のポートが鉛直方向の下方に位置されることによって、重力により培養容器の内部空間からポートを通じて培地が流出されるが、第3姿勢よりも重力による培養容器の変形が小さい。すなわち、培養容器において、内面同士が接触するおそれが低減される。これにより、例えば、培養容器の内面から細胞が剥がれ落ちるおそれが低減される。
 (2) 好ましくは、上記回転制御部は、上記培養容器から溶液が排出されることを示す第3情報に応じて、少なくとも上記ポートから上記培養容器内の溶液が流出されるとき、上記容器保持部を上記支持面が略鉛直方向に沿った第3姿勢とする。 
 第3姿勢の容器保持部に保持された培養容器において、ポートを通じて細胞懸濁液や剥離液などの溶液が流出される。第3姿勢では、容器保持部の支持面が略鉛直方向に沿っているので、培養容器の内面が略鉛直方向に沿った状態に保持される。培養容器のポートが鉛直方向の下方に位置されることによって、重力により培養容器の内部空間から細胞懸濁液や剥離液などの溶液がポートを通じて流出される。
 (3) 好ましくは、上記液体給排機構を制御する給排制御部を更に備え、上記培養容器は、上記容器保持部に保持された状態において、上記回転軸と直交する方向の両端側にそれぞれ上記ポートを有するものであり、上記回転制御部は、上記第2情報又は上記第3情報に応じて、上記容器保持部を上記第2姿勢又は上記第3姿勢とし、上記給排制御部は、上記第2情報又は上記第3情報に応じて、上記液体給排機構に、上記第2姿勢又は上記第3姿勢の上記容器保持部に保持された上記培養容器において下方に位置するポートから上記培養容器内の溶液を流出させる。
 (4) 好ましくは、上記第1姿勢の上記容器保持部に保持された上記培養容器においていずれか一方のポートから上記培養容器内へ溶液を流入させ、上記第2姿勢又は上記第3姿勢の上記容器保持部に保持された上記培養容器において上方に位置するポートから所定量の吸引を行う。
 第2姿勢又は上記第3姿勢の容器保持部において、鉛直方向の下方に位置する培養容器のポートを通じて培養容器に貯留された培地などの溶液が流出される。その後、第1姿勢の容器保持部において、培養容器のポートを通じて内部空間に培地などの溶液が流入される。そして、第2姿勢又は第3姿勢の容器保持部において、鉛直方向の上方に位置する培養容器のポートを通じて、液体給排機構が所定量の吸引を行う。この吸引によって、培地などの溶液の交換において、培養容器の内部空間に気体が進入したとしても、第2姿勢又は第3姿勢において、鉛直方向の上方に位置する培養容器のポート付近に気体が集まり、その気体がポートを通じて培養容器から吸い出される。これにより、効率的に培養容器に対して気体又は液体が流出入される。
 (5) 好ましくは、上記給排制御部は、上記液体給排機構に、上記第3姿勢の上記容器保持部に保持された上記培養容器のポートから培地を流出させるステップと、上記第1姿勢の上記容器保持部に保持された上記培養容器に剥離液を流入させるステップと、上記第2姿勢又は上記第3姿勢の上記容器保持部に保持された上記培養容器から剥離液を流出させるステップと、上記第1姿勢の上記容器保持部に保持された上記培養容器に培地を流入させるステップと、上記第3姿勢の上記容器保持部に保持された上記培養容器から細胞懸濁液を流出させるステップと、を実行させる。
 これにより、培養容器の内面に付着した細胞が細胞懸濁液として培養容器から流出される。また、遠心分離を行うことなく培地を溶媒とした細胞懸濁液を得ることができる。
 (6) 好ましくは、上記第2姿勢は、上記支持面が水平方向に対して鋭角をなす角度が20度以上70度以下である。
 (7) 好ましくは、上記培養容器の内面は、接着性細胞を培養するに適した細胞接着性を有する。
 (8) 好ましくは、上記回転制御部は、上記第1情報に応じて、上記容器保持部を上記第1姿勢とした後、上記第1姿勢から上記支持面が180度回転された第4姿勢とする。
 第1姿勢においては、鉛直方向の上方を向く培養容器の内面に、例えば接着性細胞が付着して培養される。第4姿勢においては、第1姿勢において鉛直方向の上方を向いていた培養容器の内面が、鉛直方向の下方を向く。他方、第1姿勢において鉛直方向の下方を向いていた培養容器の内面が、鉛直方向の上方を向く。これにより、第1姿勢において接着性細胞が付着し難い内面に、接着性細胞が付着して培養される。
 (9) 好ましくは、上記回転制御部に上記第1情報、上記第2情報、及び上記第3情報を出力する培養制御部を更に具備しており、上記培養制御部は、上記第1情報を出力する培養ステップと、上記第2情報を出力する溶液交換ステップと、上記第3情報を出力する溶液排出ステップと、を実行する。
 これにより、培養ステップ、培地などの溶液を交換するステップ、及び細胞懸濁液などの溶液を排出するステップが自動的に制御されて行われる。
 (10) 好ましくは、上記培養制御部は、上記溶液交換ステップにおいて、上記液体給排機構を駆動して、上記培養容器内の溶液の一部又は全てを交換する。
 本発明に係る装置によれば、培養容器を用いた細胞培養を容易に行うことができる。

 以下、本発明の好ましい実施形態を説明する。なお、本実施形態は本発明の一実施態様にすぎず、本発明の要旨を変更しない範囲で実施態様を変更できることは言うまでもない。
[培養装置10の概要]
 図1に示されるように、培養装置10は、制御部11、冷蔵保存部12、常温保存部13及び2つの培養部14を備える。冷蔵保存部12及び常温保存部13は、培養部14の外部に設けられている。2つの培養部14は、上下に分かれて配置されている。制御部11は、ディスプレイ15を備えている。ディスプレイ15は、培養装置10の前面に配置されている。制御部11は、不図示のデータ入力部を有しており、データ入力部を通じて制御部11に細胞培養に関する種々の条件などが入力される。
 培養装置10は、制御部11に入力され保存されたプログラムに従い自動で細胞を培養する装置である。以下、培養装置10の構成要素が詳細に説明される。以下の説明において、図1における上下に沿って上下方向101が定義され、図1における左右に沿って左右方向102が定義され、上下方向101及び左右方向102と垂直な方向(図1の紙面に垂直な方向)に沿って前後方向103が定義される。
 図2,3に示されるように、制御部11は、回転制御部20、培養制御部21、及び給排制御部22を有する。回転制御部20、培養制御部21、及び給排制御部22は、それぞれの制御対象の動作を制御するための演算装置であり、予めプログラムや情報が格納されている。培養制御部21は、回転制御部20及び給排制御部22に制御情報を出力する。また、培養制御部21は、培養部14の環境温度を制御するための制御情報を出力する。回転制御部20は、回転機構34と電気的に接続されており、回転機構34の駆動を制御するための制御情報を出力する。給排制御部22は、液体給排機構37と接続されており、液体給排機構37の各駆動部、すなわち供給ポンプ91,排出ポンプ92、及びバルブV1~V18を駆動するための制御情報を出力する。
 図1に示されるように、冷蔵保存部12は、内部に試薬又は培地を貯留した容器を載置するための棚が形成された筐体である。冷蔵保存部12の前面には、筐体の前面に設けられた開口を開閉可能な扉が設けられている。冷蔵保存部12は、不図示の冷却機構を備える所謂冷蔵庫である。冷却機構により、冷蔵保存部12の内部の温度は、常温より低い任意の設定温度、例えば、約10℃や約4℃などに保たれる。常温保存部13は、内部に試薬又は培地を貯留した容器を載置するための棚を有する筐体である。
 冷蔵保存部12及び常温保存部13の内部に載置された容器は、試薬又は培地を液密に貯留可能なものである。容器としては、例えば、バッグ、ボトル、カセットなどが挙げられる。各容器は、内部の液体を流出可能にチューブなどが接続されており、液体給排機構37によって貯留された液体が流出可能である。
[培養部14]
 2つの培養部14は、装置における配置が異なる他は同様の構造なので、以下、1つの培養部14を例に詳細な構成が説明される。培養部14は、培養装置10の内部に形成された空間であり、当該空間は、培養装置10の筐体フレームと、トレイ17(図2参照)により区画されている。トレイ17は、培養装置10の前面から手前へ引き出し可能である。トレイ17が引き出されることにより、培養部14が開放され、培養部14に設けられた各構成にアクセス可能となる。
 培養部14は、所定の温度及びCO濃度に保持可能である。各図には示されていないが、培養部14には、加温装置及びCO供給装置が設けられている。また、培養部14には、温度センサ及びCO濃度センサが設けられている。培養制御部21は、温度センサ及びCO濃度センサの出力に基づいて、培養部14の内部を設定された温度及びCO濃度にすべく加温装置及びCO供給装置を駆動する。細胞培養において、培養部14は、例えば、37℃、5%COの環境に保持可能である。
 図2,3に示されるように、トレイ17には、第1バッグ保持部31、第2バッグ保持部32、及び第3バッグ保持部33が左右方向102に並んで設けられている。第1バッグ保持部31の前後方向103の後方には、回転機構34が設けられている。回転機構34によって、第1バッグ保持部31が所定の回転姿勢に回転される。同様に、第2バッグ保持部32の前後方向103の後方に回転機構34が設けられており、また、第3バッグ保持部33の前後方向103の後方に回転機構34が設けられている。トレイ17の前側には、液体給排機構37が設けられている。また、第1バッグ保持部31と同軸で回転可能に第1バッグ保持部31と連結された濃縮装置支持部104が設けられている。また、トレイ17の左右方向102の左側には、容器収納部27,28が設けられており、右側には、容器収納部29が設けられている。
[第3バッグ保持部33]
 第1バッグ保持部31、第2バッグ保持部32、及び第3バッグ保持部33は、配置及び寸法が異なる他は、基本的に同様の構成なので、以下には、第3バッグ保持部33を一例として詳細な構成が説明される。なお、第1バッグ保持部31、第2バッグ保持部32、及び第3バッグ保持部33の外形及び寸法は、保持可能な培養バッグの外形に合わせて設計されている。
 図4及び図5に示されるように、第3バッグ保持部33は、保持板42,43、ホルダ44,45、スペーサー46,47及び回転軸48,49を備える。保持板42,43は、矩形の平板である。保持板42,43には、厚み方向に貫通する孔が複数形成されている。この孔は、保持板42,43に挟まれて保持された培養バッグ90へ培養部14内の気体からの熱伝導を高めるためのものである。保持板42,43は、相互に対向して配置される。保持板42,43において相互に対向する面が、支持面65,66である。
 保持板42,43の間であって、培養バッグ90(培養容器の一例)のポート73,74が配置される一対の対向する縁部には、スペーサー46,47が配置されている。スペーサー46,47は、保持板42,43の間隔を維持するためのものである。スペーサー46,47は、それぞれが四角柱形状である。スペーサー46,47の長手方向の長さは、保持板42,43の一対の縁部の長さとほぼ同じである。また、スペーサー46,47は、断面形状が長手方向に渡って一定である。スペーサー46,47の長手方向の中央には、長手方向と直交する方向へ凹む凹部46A,47Aが形成されている。凹部46A,47Aは、培養バッグ90のチューブ99がそれぞれ挿入される空間である。なお、スペーサー46,47は、保持板42,43の一方と一体に構成されていてもよい。
 スペーサー47には、距離センサ67が配置されている。スペーサー47と当接する保持板42には、距離センサ67と対向してマグネット68が配置されている。距離センサ67は、マグネット68からの磁束密度に応じた電圧を出力するものであり、例えば、ホール素子が使用される。保持板42,43が、培養バッグを保持した通常の状態であれば、保持板42とスペーサー47との距離は一定なので、距離センサ67の出力も一定である。培養バッグに流入した液体の量が多く、培養バッグが膨らむことによって保持板42,43が互いに離れる方向に押し広げられるように撓んで保持板42とスペーサー47との距離が大きくなると、距離センサ67が検知するマグネット68の磁束密度が小さくなり、距離センサ67の出力が変化する。
 制御部11には、閾値が予め格納されている。この閾値は、保持板42とスペーサー47との距離が一定以上に大きくなったことを判定するためのものである。制御部11は、距離センサ67の出力を閾値と比較することによって、保持板42とスペーサー47との距離が一定以上に大きくなったことを判定できる。なお、距離センサ67はスペーサー47ではなく、スペーサー46に設けられていてもよく、スペーサー46,47の両方に設けられていてもよい。また、マグネット68は、距離センサ67により磁束が検知可能であれば、保持板42,43のいずれに設けられてもよい。また、距離センサ67とマグネット68との配置は相対的なものであり、距離センサ67とマグネット68との配置が入れ替わってもよいことは言うまでもない。また、保持板42,43に、距離センサ67とマグネット68とがそれぞれ対向して配置されていてもよい。
 ホルダ44,45は、スペーサー46,47を介在させた状態の保持板42,43を挟み込んで一体に保持するものである。ホルダ44,45は、断面が横向きU時形状の細長な部材であり、横向きU時形状の内側に、スペーサー46,47を介在させた状態の保持板42,43の縁部が挿入される。ホルダ44,45に挿入される保持板42,43の縁部は、スペーサー46,47が介在されない一対の縁部である。ホルダ44,45のそれぞれの長手方向の両端側には、ネジ孔が形成されており、そのネジ孔にネジ18が螺合されている。ネジ18の先端は、ホルダ44,45の横向きU時形状の内側に突出されている。ホルダ44,45の横向きU時形状の内側に挿入されたホルダ44,45の一方が、ネジ18によって押圧されることにより、ホルダ44,45がスペーサー46,47を介在させた状態に保持板42,43を保持する。この状態において、保持板42,43の支持面65,66の間に形成された空間が培養バッグを保持する空間となる。なお、ホルダ44,45は、保持板42,43の一方と一体に構成されてもよい。また、保持板42,43は、ホルダ44,45の一方により蝶番のごとく回動自在に連結されていてもよい。
 ホルダ44,45には、長手方向の中央付近から、保持板42,43が保持される側と反対向きに突出する回転軸48,49がそれぞれ設けられている。回転軸48,49は、ホルダ44,45が保持板42,43を保持した状態において、同軸に延びる。回転軸48,49が延びる方向は、保持板42,43の支持面65,66と平行である。
 図5(A)に示されるように、第3バッグ保持部33が、支持面66を支持面65より下方として、支持面65,66が水平方向に沿った状態であるときに、第3バッグ保持部33に支持されている培養バッグ90に液体が入っていない場合、培養バッグ90は主として支持面66に接触している。このとき、支持面65,66の距離は、距離D1である。図5(B)に示されるように、培養バッグ90には、仮に貯留する液体によって培養バッグ90が膨らんで支持面65,66に接触したとしても、保持板42,43を撓ませない程度の容量の液体が流入される。このような液体の量が、培養バッグ90に貯留可能な液体量として予め設定されている。図5(C)に示されるように、培養バッグ90に予め設定された量より多くの液体が流入されると、支持面65,66に接触している培養バッグ90が更に膨らんで、保持板42,43を互いに離れる方向に押し広げられるように撓ませる。保持板42,43が撓むことによって支持面65,66の距離が大きくなる(距離D2)。保持板42とスペーサー47の距離に応じた距離センサ67の検知値が前述された閾値を超えると、制御部11が支持面65,66の距離が距離D2を超えたと判定する。
 第1バッグ保持部31及び第2バッグ保持部32の詳細な構成については、図を用いて詳細には説明されないが、第3バッグ保持部33と同様に、保持板ホルダ、スペーサーを備える。   
[濃縮装置支持部104]             
 図6に示されるように、濃縮装置支持部104は、第1バッグ保持部31の前後方向103の後方に配置されている。濃縮装置支持部104は、第1バッグ保持部31と一体に連結されている。したがって、濃縮装置支持部104は、回転軸48,49周りに第1バッグ保持部31と一体に回転する。濃縮装置支持部104は、濃縮装置120を支持する。濃縮装置支持部104に支持された濃縮装置120は、流入ポート110及び第1流出ポート111が、回転軸48,49による回転軸と交差する方向(本実施形態では直交する方向)をそれぞれ向くように配置される。これにより、濃縮装置支持部104と共に濃縮装置120が回転軸48,49周りに回転すると、流入ポート110及び第1流出ポート111の上下方向101の位置が変わる。また、濃縮装置支持部104に支持された濃縮装置120の流入ポート110及び第1流出ポート111がそれぞれ延びる方向と、第1バッグ保持部31に保持された培養バッグ70のポート73,74がそれぞれ延びる方向とは、合致している。
 図2に示されるように、トレイ17において、第1バッグ保持部31、第2バッグ保持部32、第3バッグ保持部33、及び濃縮装置支持部104が配置される箇所には、回転する第1バッグ保持部31、第2バッグ保持部32、第3バッグ保持部33、及び濃縮装置支持部104との干渉を避けるために開口17Aが3箇所に形成されている。
 各開口17Aの前後方向103の前方及び後方には、一対の軸受部24がそれぞれ配置されている。一対の軸受部24は、第1バッグ保持部31、第2バッグ保持部32、第3バッグ保持部33、及び濃縮装置支持部104の各回転軸48,49を前後方向103に沿った状態でそれぞれ回転自在に支持する。これにより、第1バッグ保持部31、第2バッグ保持部32、及び第3バッグ保持部33の各保持板42,43、並びに濃縮装置支持部104は、回転軸48,49を回転中心として回転可能である。
 図8に示されるように、各軸受部24は、トレイ17上に設けられた各重量検出部23上に積載されている。第1バッグ保持部31、第2バッグ保持部32、又は第3バッグ保持部33にそれぞれ対応する一対の重量検出部23によって、対応するバッグ保持部、対応するバッグ保持部に保持されている培養バッグ、及び軸受部24などの重量が検知される。重量検出部23としては、例えば重量検出部23に加えられた力(重量)を電気的な信号に変換するロードセルなどが用いられる。
[回転機構34]
 図2及び図3に示されるように、トレイ17において、各軸受部24の前後方向103のそれぞれ後方には、回転機構34がそれぞれ設けられている。回転機構34は、回転軸支持部16及び不図示のステッピングモータを有する。回転軸支持部16は、前後方向103の後方に向かって延びており、各軸受部24に支持された回転軸49と同軸に連結されている。各図には示されていないが、ステッピングモータには、電源から電力が供給されている。また、ステッピングモータと回転軸支持部16とは、公知の減速ギアなどによって駆動伝達されている。回転制御部20から出力される制御信号に基づいた電力がステッピングモータに供給されることにより、回転軸支持部16が所定の回転角だけ回転する。なお、ステッピングモータには、原点位置、すなわち第1バッグ保持部31、第2バッグ保持部32、及び第3バッグ保持部33の保持板42,43の支持面65,66が水平方向に沿った状態となる回転位置を検出するためのセンサが設けられていてもよい。回転制御部20は、このセンサの出力に基づいて、支持面65,66が水平方向に沿った状態となるようにステッピングモータを駆動することができる。同様に、濃縮装置120の流入ポート110及び第1流出ポート111が水平方向に延びる状態となるようにステッピングモータを駆動することができる。なお、ステッピングモータは回転機構34の駆動源の一例であり、ステッピングモータに代えて、例えば回転量を検出可能なエンコーダを有する直流モータなど他の駆動源が採用されてもよい。また、駆動源の回転位置はレゾルバなどの他の検知手段により検知されてもよいし、第1バッグ保持部31などの回転位置を直接検出できるセンサが設けられてもよい。
[液体給排機構37]
 図2,3に示されるように、液体給排機構37は、供給ポンプ91、排出ポンプ92、及び複数のバルブV1~V18を有する。供給ポンプ91及び排出ポンプ92は、チューブ内の液体を送液可能な公知のものが採用される。このようなポンプとして、例えば軟質のチューブを回動するローラによって扱くことにより送液する所謂チューブポンプが挙げられる。供給ポンプ91及び排出ポンプ92には電源から電力が供給されている。給排制御部22は、供給ポンプ91及び排出ポンプ92に供給される電力を制御することにより、供給ポンプ91及び排出ポンプ92を一定時間駆動させることができる。
 複数のバルブV1~V18は、供給ポンプ91に関連するバルブV1~V11と、排出ポンプ92に関連するバルブV12~V18に大別される。供給ポンプ91及びバルブV1~V11が、液体給排機構37における液体供給機構81であり、排出ポンプ92及びバルブV12~V18が、液体給排機構37における液体排出機構82である。各バルブV1~V18は、給排制御部22から出力される制御信号に基づいてオン/オフが切り替えられる。各バルブV1~V18のオン/オフによって、培養バッグなどに連結されている各チューブ38における液体の流れを変更することができる。バルブV1~V18としては、例えば、電磁弁が採用される。
[培養回路]
 図3に示されるように、培養回路は、3つの培養バッグ70,80,90と、2つのサーババッグ39,40と、回収バッグ41と、濃縮装置120と、冷蔵保存部12又は常温保存部13に保持される貯留容器116,117と、これらを液体が流通可能に連結する複数のチューブ38と、を有する。培養バッグ70は、第1バッグ保持部31に設置されるものである。培養バッグ80は、第2バッグ保持部32に設置されるものである。培養バッグ90は、第3バッグ保持部33に設置されるものである。濃縮装置120は、濃縮装置支持部104に設置されるものである。回収バッグ41は、培養部14内に設置されるものである。培養バッグ70が、培養バッグ80,90と比べて容量が小さいほかは、各培養バッグ70,80,90は同様の構成なので、以下、培養バッグ90を例に詳細な構成が説明される。
 図4に示されるように、培養バッグ90は、合成樹脂製の2枚の矩形のシートの周縁が熱溶着などの公知の方法で張り合わされることにより、バッグ形状に形成されたものである。2枚の矩形のシートにおいて対向する一対の端部77,78の中央付近にそれぞれ合成樹脂製のチューブ99が配置されている。図9(A)、(B)に示されるように、各チューブ99において、培養バッグ90の内部空間75に位置する部分には、培養バッグ90の内部空間75に位置する端から縁87,88へ向かって切欠部105が形成されている。図10に示されるように、切欠部105は、培養バッグ90の内部空間75を画定する内面71,72と対向しない位置、すなわち、2枚の矩形のシートにおける一対の端部77,78以外の端部を向いている箇所に位置している。仮に、培養バッグ90から液体が流出されるときに、内部空間75に負圧が生じて内面71,72が相互に近づいたとしても、チューブ99付近においては内面71,72の間にチューブ99が存在することにより、内面71,72同士が接触し難くなる。他方、チューブ99において内面71,72と対向しない箇所には切欠部105が形成されているので、切欠部105を通じて、内部空間75の縁87,88付近に残存する液体は、チューブ99の内部空間へ流入可能である。
 図4に示されるように、チューブ99の内部空間を通じて、培養バッグ90の内部空間75が外部と連通されている。すなわち、各チューブ99によってポート73,74が形成されている。培養バッグ90の内部空間75を区画する縁のうち、チューブ99が配置されている縁87,88は、ポート73,74から離れるに従って縁87,88間の距離が小さくなるテーパ形状である。換言すれば、縁87,88は、チューブ99が配置されている中央へ向かって、培養バッグ90の外方へ向かうように膨らんでいる形状である。培養バッグ90が第1バッグ保持部31に保持された状態において、チューブ99は、回転軸48,49と直交する方向へ延びる。すなわち、ポート73,74は、回転軸48,49と直交する方向へ延びる。
 培養バッグ90に用いられる合成樹脂シートとしては、可撓性を有し、培地を入れたときにバッグ形状が維持できる曲げ剛性を有するものであり、例えば、低密度ポリエチレン、超高分子量ポリエチレン、環状ポリオレフィン系樹脂、またはこれら及び他の材料とのラミネート構造を有するものが挙げられる。
 培養バッグ90の内面71,72は、接着性細胞を培養するに適した細胞接着性を有する。詳細には内面71,72は、例えばプラズマ処理などによって細胞接着性官能基が露出されている。細胞接着性官能基としては、例えば、アミノ基、アミン基、水酸基、スルホン基、スルフェン基、スルフィン基、エーテル基、カルボキシル基、カルボニル基などが挙げられる。これらのうち、細胞との接着性が高いアミノ基及びカルボキシル基が好ましい。
 図3に示されるサーババッグ39,40、回収バッグ41、貯留容器116,117は、合成樹脂製のシートが張り合わされてバッグ形状に形成されたものであり、少なくとも1つのポート94~98を有する。サーババッグ39,40は、培地を貯留するためのものである。回収バッグ41は、細胞懸濁液を回収するためのものである。貯留容器116,117は、剥離液やプライミング液を貯留するためのものである。サーババッグ39,40、回収バッグ41、及び貯留容器116,117は、公知のバッグのほか、培地や細胞懸濁液を貯留可能な公知の容器が採用されうる。
 図3に示されるように、培養バッグ70,80,90、サーババッグ39,40、回収バッグ41、貯留容器116,117の各ポートには、チューブ38がそれぞれ連結されている。培養バッグ70,80,90のそれぞれの一方のポート73に接続された各チューブ38、濃縮装置120の流入ポート110に接続されたチューブ38、サーババッグ39,40の各ポート94,95に接続された各チューブ38、貯留容器116,117の各ポート97,98に接続された各チューブ38は、供給ポンプ91へ延出されている。これら各チューブ38は、供給ポンプ91へ到達する前にコネクタを介して1本のチューブ38にまとめられて液体供給回路を構成している。また、これら各チューブ38は、1本のチューブ38にまとめられる前に、それぞれがバルブV6~V11に通されており、各バルブV6~V11によってチューブ38の内部空間が液体が流通可能な開状態と、液体が流通できない閉状態とに変化可能である。
 培養バッグ70,80,90のそれぞれの他方のポート74に接続された各チューブ38、濃縮装置120の第1流出ポート111及び第2流出ポート112に接続された各チューブ38,回収バッグ41のポート96に接続されたチューブ38は、排出ポンプ92へ延出されている。これら各チューブ38は、排出ポンプ92へ到達する前にコネクタを介して1本のチューブ38にまとめられて液体排出回路を構成している。また、これら各チューブ38は、1本のチューブ38にまとめられる前に、それぞれがバルブV12及びV14~V18に通されており、各バルブV12及びV14~V18によってチューブ38の内部空間を液体が流通可能な開状態と、液体が流通できない閉状態とに変化可能である。
 供給ポンプ91に通ずるチューブ38は、再び分岐されて各々がバルブV1~V5を通じて冷蔵保存部12又は常温保存部13に置かれたバッグや容器に接続されている。これらバッグや容器には、細胞懸濁液や培地、剥離液などの試薬が貯留されている。排出ポンプ92に通ずるチューブ38は、廃液容器19へ接続されている。
[濃縮装置120]
 図7(A)に示されるように、濃縮装置120は、本体106、キャップ107、キャップ108、及び中空糸束109を有する。本体106は、概ね円筒形状である。本体106は、軸線方向(図7(A)における上下方向に沿った方向)の両側において開口している。本体106の各開口には、キャップ107及びキャップ108がそれぞれ螺合されている。キャップ107には、濃縮装置120の内部空間と外部空間とを連通する流入ポート110が、本体106の軸線方向に沿って延びるように形成されている。キャップ108には、濃縮装置120の内部空間と外部空間とを連通する第1流出ポート111が、本体106の軸線方向に沿って延びるように形成されている。
 本体106の各開口付近には、不図示の一対の中空糸支持体が設けられており、この一対の中空糸支持体に支持されて、本体106の内部空間に中空糸束109が配置されている。中空糸束109の両端は、本体106の開口付近に配置されている。中空糸支持体は、中空糸束109の両端を支持するとともに、中空糸束109の両端と、本体106の内部空間とを液密に隔てている。したがって、キャップ107の流入ポート110を通じて本体106の内部空間に流入した液体は、中空子束114の一端へ流れ込み、本体106の内部空間において一対の中空糸支持体によって区画された空間には流入しない。他方、中空糸束109の他端から流出した液体は、キャップ108の第1流出ポート111から外部へ流出し、本体106の内部空間において一対の中空糸支持体によって区画された空間から第1流出ポート111には液体が流出しない。
 本体106には、キャップ108が螺合される側に第2流出ポート112が、本体106の軸線方向(図7における上下方向に沿った方向)と直交する方向へ延びており、キャップ107が螺合される側にポート113が、本体106の軸線方向と直交する方向へ延びている。第2流出ポート112及びポート113は、本体106の内部空間において一対の中空糸支持体に区画された空間と本体106の外部とを連通する。第2排出ポート112を通じて、中空糸束109からろ過により流出した液体が外部へ流出される。なお、ポート113は、本実施形態では封止されている。また、流入ポート110、第1流出ポート111、第2流出ポート112には、前述されたチューブ38が接続されている。
 中空糸束109は、透析膜が管状に形成された中空糸の束である。中空糸はそれぞれの両端において開口している。中空糸の原料としてはトリアセテートやポリエーテルスルホン等が挙げられる。中空糸の太さや膜厚、孔径、長さ、種類等は、濃縮装置120内でろ過される細胞のサイズ等の条件に応じて適宜設定される。
[培養装置10を用いた細胞培養方法]
 以下に、培養装置10を用いた細胞培養方法が説明される。培養装置10を用いた細胞培養は、培養バッグ70,80,90のいずれか一つ又は複数を任意に選択して行うことができるが、以下には、培養バッグ70のみを用いた細胞培養方法が説明される。培養装置10を用いた細胞培養方法は、以下に示される各ステップを含む。
(1)培養バッグ70内で細胞を増幅する培養ステップ。
(2)培養バッグ70内の培地を交換する培地交換ステップ。
(3)培養バッグ70内の細胞懸濁液を回収する細胞懸濁液回収ステップ。
(4)サーババッグ40内の細胞懸濁液を濃縮する細胞懸濁液濃縮ステップ。
 培養装置10には、予め、培養回路がセットされる。詳細には、図3に示されるように、培養バッグ70,80,90が第1バッグ保持部31、第2バッグ保持部32、及び第3バッグ保持部33にそれぞれセットされ、濃縮装置120が濃縮装置支持部104にセットされ、サーババッグ39,40が容器収納部27,28にそれぞれ保持され、回収バッグ41が容器収納部29に保持される。また、貯留容器116,117が冷蔵保存部12又は常温保存部13に保持される。また、回路の各チューブ38が各バルブV1~V18、供給ポンプ91、排出ポンプ92にセットされる。
 培養制御部21には、培養ステップ、培地交換ステップ、培養ステップ、細胞懸濁液濃縮ステップ、細胞懸濁液回収ステップが順次行われるように、予めユーザが設定を行っている。培養ステップにおける培養時間や、培地交換ステップにおける培地交換量、細胞懸濁液濃縮ステップにおける濃縮時間や培地供給量、細胞懸濁液回収ステップにおける剥離液との反応時間などの各種設定も、予めユーザが設定を行う。培養制御部21は、培養ステップにおいて各種設定情報を含む第1情報を出力し、培地交換ステップにおいて各種設定情報を含む第2情報を出力し、細胞懸濁液回収ステップにおいて各種設定情報を含む第3情報を出力する。また、培養制御部21は、細胞懸濁液濃縮ステップにおける各種設定情報を出力する。回転制御部20は、培養制御部21から出力される各情報に基づいて回転機構34の駆動を制御する。給排制御部22は、培養制御部21から出力される各情報に基づいて液体給排機構37(液体供給機構81及び液体排出機構82)の駆動を制御する。
 図11に示されるように、培養装置10における培養部14は、培養制御部21が培養開始の入力を受け付けることにより、予備動作を行う。詳細には、培養制御部21からの制御情報に基づいて加温装置及びCO供給装置が駆動されることにより、培養部14の
温度が37℃に、二酸化炭素の濃度が5%に調節される(ステップS11)。続いて、回転制御部20は、第1バッグ保持部31が第1姿勢であるか否かを判定する(ステップS12)。第1バッグ保持部31が第1姿勢でない場合、すなわち回転機構34におけるステッピングモータが原点位置にないと回転制御部20が判定したときは(ステップS12:No)、回転制御部20が回転機構34のステッピングモータを原点位置となるまで回転する。これにより、図12に示されるように、第1バッグ保持部31が、支持面65,66を水平方向に沿った第1姿勢で停止する(ステップS13)。第1バッグ保持部31が第1姿勢であるときは(ステップS12:Yes)、回転制御部20は回転機構34を駆動しない。なお、第1姿勢における支持面65,66は、ほぼ水平方向に沿っていればよく、厳密に水平方向に沿っている必要はない。また、各バルブV1~V18は、チューブ38を閉状態とする。
 続いて、給排制御部22は、培養バッグ70に細胞懸濁液を供給する(ステップS14)。培養バッグ70への細胞懸濁液の供給は、液体供給ステップに従って行う。詳細には、図13に示されるように、第1バッグ保持部31が第1姿勢で停止しているときに、重量検出部23は、培養バッグ70及び第1バッグ保持部31の重量を検出する。重量検出部23から出力された重量の情報に応じて、制御部11は、第1基準値を設定する(ステップS121)。続いて、制御部11は、培養バッグ70に供給すべき細胞懸濁液の重量を第1基準値に加えた値を第1目標値として算出する。培養バッグ70に供給すべき液体の重量は、培養バッグ70に供給すべき細胞懸濁液の容量に基づいて算出することができる。なお、細胞懸濁液や培地、剥離液などの各種液体の比重を近似的に1として容量(mL)=重量(g)として扱ってもよい。なお、ステップS14において、培養バッグ70に細胞懸濁液を供給しているが、ステップS14を省略し、手動で予め培養バッグ70に細胞懸濁液を充填してもよい。
 給排制御部22は、第1バッグ保持部31が第1姿勢で停止しているときに、液体供給機構81を駆動させて、培養バッグ70に液体を供給する。詳細には、バルブV6,V9が開状態とされる(ステップS122)。続いて、供給ポンプ91が駆動される(ステップS123)。サーババッグ40には、予め培養すべき細胞を含む細胞懸濁液が貯留されている。したがってサーババッグ40から細胞懸濁液が、ポート73を通じて培養バッグ70へ供給される。培養バッグ70に細胞懸濁液が供給されている間に、すなわち供給ポンプ91が駆動されている間に、制御部11は、距離センサ67が予め設定された閾値を超えたか否かを監視する(ステップS124)。制御部11は、距離センサ67の出力値が閾値を超えたと判定すると(ステップS124:Yes)、ブザー音を発生させたり、ライトを点灯したりして警報を発する(ステップS126)。そして、給排制御部22は、供給ポンプ91を停止する(ステップS128)。
 制御部11は、供給ポンプ91が駆動されてから予め設定された時間が経過したか否かを監視する(ステップS125)。時間としては、供給ポンプ91が培養バッグ70に最大量の液体を供給するに十分な時間より長い時間が設定されている。制御部11は、供給ポンプ91が駆動されてから予め設定された時間が経過したと判定すると(ステップS125:Yes)、前述と同様にして警報を発し(ステップS126)、給排制御部22は、供給ポンプ91を停止する(ステップS128)。
 制御部11は、供給ポンプ91が駆動されている間に、重量検出部23の出力値が第1目標値に到達したか否かを監視する(ステップS127)。制御部11が、重量検出部23の出力値が第1目標値に到達したと判定すると(ステップS127:Yes)、給排制御部22は、供給ポンプ91を停止し(ステップS128)、また、バルブV6,V9を閉状態とする(ステップS129)。これにより、培養バッグ70への細胞懸濁液の供給(液体供給ステップ)が終了する(ステップS14)。
 続いて、図11に示されるように、回転制御部20は、回転機構34を駆動して、第1バッグ保持部31を反時計回りに90度回転する。これにより、図14,15に示されるように、第1バッグ保持部31は、支持面65,66が鉛直方向に沿い、かつポート74が上方にあり、ポート73が下方にある第3姿勢となる(ステップS15)。第3姿勢の第1バッグ保持部31に保持された培養バッグ70おいて、上方にあるポート74の縁88には、内部空間75に進入した気体が集まる。縁88は、ポート74へ向かうテーパ形状なので、縁88に沿って移動する気体がポート74に集まる。なお、第3姿勢における支持面65,66は、概ね鉛直方向にそっていればよく、必ずしも厳密に鉛直方向に沿っている必要はない。
 第1バッグ保持部31が第3姿勢にされた後、給排制御部22は、バルブV6,V9を閉状態とし、バルブV13,V15を開状態とする。続いて、給排制御部22は、排出ポンプ92を駆動する。これにより、培養バッグ70のポート74から液体又は気体が排出される。培養バッグ70に細胞懸濁液が供給されるときに、培養バッグ70の内部空間75に気体が残っているときにはポート74を通じて内部空間75から気体が排出される(ステップS16)。これにより、予備動作が終了する。
 予備動作が終了した後、培養制御部21は、培養ステップ(ステップS17)、培地交換ステップ(ステップS18)、培養ステップ(ステップS19)、細胞懸濁液回収ステップ(ステップS20)を順次行う。各ステップの詳細は後述される。細胞懸濁液回収ステップの後、培養制御部21に、継代を行う旨の指示が入力されていれば(ステップS21:Yes)、継代を行うべく、別の培養バッグ80,90に回収された細胞懸濁液が供給される(ステップS22)。継代を行う旨の指示が入力されていないときは(ステップS22:No)、培養制御部21は、継代を行わない。その後に細胞懸濁液濃縮ステップ(ステップS23)が行われる。
[培養ステップ]
 以下、培養ステップが説明される。培養ステップが行われるときには、培養制御部21は、第1情報を回転制御部20及び給排制御部22に出力する。図12及び図16に示されるように、回転制御部20は、回転機構34を駆動させて、第1バッグ保持部31を第1姿勢とする(ステップS24)。これにより、図17(A)に示されるように、培養バッグ70において、内面71が上方となり、内面72が下方となる。培養バッグ70の内部空間75には、培養すべき細胞と培地とを含む細胞懸濁液が貯留されている。内部空間75において、細胞は、細胞懸濁液を重力により降下して内面72に接触する。これにより、内面72に細胞が付着して培養される。
 図16に示されるように、回転制御部20は、予め設定された時間が経過するまで(ステップS25:No)、第1バッグ保持部31を第1姿勢に維持する。回転制御部20は、予め設定された時間が経過した後(ステップS25:Yes)、回転機構34を駆動させて第1バッグ保持部31を培養装置10の前方から視て時計回りに180度回転する(ステップS26)。これにより、図17(B)及び図18に示されるように、第1バッグ保持部31は、支持面65,66が水平方向に沿っており、かつ培養バッグ70の内面72が上方であり、内面71が下方となる第4姿勢となる。培養バッグ70において、内面72が上方となり、内面71が下方となると、培養バッグ70の内部空間75に貯留された細胞懸濁液に含まれており、かつ内面72に付着していない細胞は、細胞懸濁液内を重力により降下して内面71に接触する。これにより、内面71に細胞が付着して培養される。回転制御部20は、予め設定された時間が経過するまで(ステップS27:No)、第1バッグ保持部31を第4姿勢に維持する。予め設定された時間が経過すると(ステップS27:Yes)、培養ステップが終了する。
[培地交換ステップ]
 以下、培地交換ステップが説明される。培地交換ステップは、溶液交換ステップの一例である。培地交換ステップが行われるときには、培養制御部21は、第2情報を回転制御部20及び給排制御部22に出力する。冷蔵保存部12又は常温保存部13に保存された新鮮培地を貯留する貯留容器117と接続されているチューブ38が通されたバルブV1とバルブV7を開状態として、供給ポンプ91を駆動して、サーババッグ39に新鮮培地を供給する。その後、給排制御部22は、供給ポンプ91を停止し、バルブV1を閉状態とし、バルブV6を開状態とする。給排制御部22は、サーババッグ39,40と接続されているチューブ38が通されたバルブV6,V7を開状態として、供給ポンプ91を逆向きに駆動して、サーババッグ40に新鮮培地を供給する。その後、給排制御部22は、供給ポンプ91を停止し、バルブV6,V7を閉状態とする。新鮮培地は、サーババッグ40において37℃に加温されるべく保持される。
 図19に示されるように、回転制御部20は、回転機構34を駆動して、第1姿勢の第1バッグ保持部31を培養装置10の前方から視て時計回りに30度回転する。これにより、図20,21に示されるように、第1バッグ保持部31は、支持面65,66が水平方向に対して鋭角をなす角度が30度となり、かつポート73が上方にあり、ポート74が下方にある第2姿勢となる(ステップS31)。なお、第2姿勢は、支持面65,66が水平方向に対して鋭角をなす角度が30度に限定されず、支持面65,66が水平方向及び鉛直方向に沿っていなければ任意に設定されてよい。好ましくは、第2姿勢において、支持面65,66が水平方向に対して鋭角をなす角度は、20度以上70度以下である。さらに好ましくは、30度以上60度以下である。
 続いて、給排制御部22は、培養バッグ70から培地を排出する(ステップS32)。培養バッグ70からの培地の排出は、液体供給ステップに従って行う。詳細には、図22に示されるように、第1バッグ保持部31が第2姿勢で停止しているときに、重量検出部23は、培養バッグ70及び第1バッグ保持部31の重量を検出する。重量検出部23から出力された重量の情報に応じて、制御部11は、第2基準値を設定する(ステップS141)。続いて、制御部11は、第2基準値に培養バッグ70から排出すべき培地の重量を差し引いた値を第2目標値として算出する。
 給排制御部22は、第1バッグ保持部31が第2姿勢で停止しているときに、液体排出機構82を駆動させて、培養バッグ70から培地を排出する。詳細に説明すると、バルブV13,V15が開状態とされる(ステップS142)。続いて、排出ポンプ92が駆動される(ステップS143)。制御部11は、排出ポンプ92が駆動されてから予め設定された時間が経過したか否かを監視する(ステップS144)。時間としては、排出ポンプ92が培養バッグ70から半量の液体を排出するに十分な時間より長い時間が設定されている。制御部11は、排出ポンプ92が駆動されてから予め設定された時間が経過したと判定すると(ステップS144:Yes)、前述と同様にして警報を発し(ステップS145)、給排制御部22は、排出ポンプ92を停止する(ステップS147)。
 制御部11は、排出ポンプ92が駆動されている間に、重量検出部23の出力値が第2目標値に到達したか否かを監視する(ステップS146)。制御部11が、重量検出部23の出力値が第2目標値に到達したと判定すると(ステップS146:Yes)、給排制御部22は、排出ポンプ92を停止し(ステップS147)、また、バルブV13,V15を閉状態とする(ステップS148)。これにより、培養バッグ70に貯留された培地の半分が、培養バッグ70の内部空間75から排出(液体排出ステップ)される(ステップS32)。第1バッグ保持部31が第2姿勢に維持されることにより、ポート74から培地が排出されやすくなり、また培地の半分が培養バッグ70から排出されたとしても、図21に示されるように、培養バッグ70の内面71,72が接触し難い。
 図19に示されるように、培養バッグ70から半分の培地が排出された後、回転制御部20は、回転機構34を駆動して、第1バッグ保持部31を第1姿勢とする(ステップS33)。第1バッグ保持部31が第1姿勢にされた後、バルブV6,V9が開状態とされ、培養バッグ70への培地の供給が、前述された液体供給ステップと同様に行われる。これにより、サーババッグ40に貯留されている新鮮培地が、培養バッグ70へ供給される(ステップS34)。培養バッグ70へ供給される新鮮培地の量は、先に培養バッグ70から排出された量と同じ、すなわち内部空間75に貯留されていた培地の半分の量である。給排制御部22は、培養バッグ70に新鮮培地が供給された後、供給ポンプ91を停止して、バルブV6,V9を閉状態とする。
 培養バッグ70へ新鮮培地が供給された後、回転制御部20は、回転機構34を駆動して、第1姿勢の第1バッグ保持部31を、培養装置10の前方から視て反時計回りに90度回転する。これにより、図14に示されるように、第1バッグ保持部31は、支持面65,66が鉛直方向に沿っており、かつポート73が下方にあり、ポート74が上方にある第3姿勢となる(ステップS35)。第1バッグ保持部31が第3姿勢とされた後、給排制御部22は、バルブV13,V15を開状態として、排出ポンプ92を駆動する。これにより、培養バッグ70の内部空間75に気体が混入しても、ポート74を通じて培地とともに気体が内部空間75から排出される(ステップS36)。そして、排出ポンプ92が停止され、バルブV13,V15が閉状態とされて、培地交換ステップ(ステップS18)が終了する。
 培地交換ステップ(ステップS18)が終了すると、前述と同様にして培養ステップ(ステップS19)が行われる。なお、培地交換ステップ及び培養ステップは、継代後に更に繰り返されて行われてもよい。また、継代後に繰り返される培養ステップにおいて、細胞の量が増える場合には、培養バッグ70より容量が大きい培養バッグ80,90が用いられるべく、細胞懸濁液が培養バッグ70から培養バッグ80,90へ移動されてもよい。必要に応じて繰り返し行った培養ステップ(ステップS19)の後、細胞懸濁液回収ステップ(ステップS20)、細胞懸濁液濃縮ステップ(ステップS23)が行われる。
[細胞懸濁液回収ステップ]
 以下、細胞懸濁液回収ステップについて説明する。細胞懸濁液回収ステップは、溶液排出ステップの一例である。細胞懸濁液回収ステップが行われるときには、培養制御部21は、第3情報を回転制御部20及び給排制御部22に出力する。図23に示されるように、回転制御部20は、回転機構34を駆動して、第4姿勢の第1バッグ保持部31を、培養装置10の前方から視て反時計回りに90度回転する。これにより、図24及び図25に示されるように、第1バッグ保持部31は、支持面65,66が鉛直方向に沿っており、かつポート73が上方にあり、ポート74が下方にある第3姿勢となる(ステップS41)。なお、本実施形態においては、第1バッグ保持部31の支持面65,66が鉛直方向に沿っている姿勢であれば、ポート73,74のいずれが上方又は下方にあっても、第3姿勢と称している。
 図23に示されるように、第1バッグ保持部31が第3姿勢にされた後、培養バッグ70からの培地の排出が行われる。給排制御部22は、バルブV13,V15を開状態として、排出ポンプ92を駆動する。これにより、培養バッグ70に貯留された培地の全量が、ポート74と通じて内部空間75から排出される(ステップS42)。培養バッグ70から培地の全量が排出された後、回転制御部20は、回転機構34を駆動して、第3姿勢の第1バッグ保持部31を第1姿勢にする(ステップS43)。
 第1バッグ保持部31が第1姿勢にされた後、培養バッグ70への剥離液の供給が行われる。給排制御部22は、冷蔵保存部12又は常温保存部13に保存された剥離液を貯留する容器と接続されているチューブ38が通されたバルブV3~V5のいずれかと、バルブV9とを開状態として、供給ポンプ91を駆動する。これにより、培養バッグ70の内部空間75へポート73を通じて剥離液が供給される(ステップS44)。培養バッグ70へ剥離液が供給された後、給排制御部22は、供給ポンプ91を停止して、開状態としたバルブV3~V5のいずれかと、V9を閉状態とする。回転制御部20は、予め設定された時間が経過するまで(ステップS45:No)、第1バッグ保持部31を第1姿勢に維持する。剥離液によって、培養バッグ70の内面71,72に付着していた細胞の内面71,72への接着が弱まる。このような内面71,72に対して細胞の接着を弱める作用は、剥離液の種類や濃度、内面71,72との接触時間、第1バッグ保持部31などの姿勢などが適宜設定されることによって実現される。なお、培養バッグ70へ剥離液が供給された後、必要があれば、前述と同様にして培養バッグ70の内部空間75から気体が排出されてもよい。
 回転制御部20は、予め設定された時間が経過した後(ステップS45:Yes)、回転機構34を駆動して、第1バッグ保持部31を培養装置10の前方から視て反時計回りに90度回転して第3姿勢とする(ステップS46)。第1バッグ保持部31が第3姿勢にされた後、培養バッグ70からの剥離液の排出が、前述された液体排出ステップと同様に行われる。給排制御部22は、バルブV6,V9を開状態として、供給ポンプ91を逆向きに駆動する。これにより、培養バッグ70において剥離液がポート73を通じて内部空間75から廃液容器19へ排出される(ステップS47)。培養バッグ70から剥離液の全量が排出された後、回転制御部20は、回転機構34を駆動して、第1バッグ保持部31を培養装置10の前方から視て時計回りに90度回転して第1姿勢とする(ステップS48)。
 第1バッグ保持部31が第1姿勢にされた後、培養バッグ70への培地の供給が行われる。給排制御部22は、冷蔵保存部12又は常温保存部13に保存された培地を貯留する容器と接続されているチューブ38が通されたバルブV3~V5のいずれかと、バルブV9とを開状態として、供給ポンプ91を駆動する。これにより、培養バッグ70の内部空間75へポート73を通じて培地が供給される(ステップS49)。培養バッグ70へ培地が供給された後、給排制御部22は、供給ポンプ91を停止して、開状態としたバルブV3~V5のいずれかと、V9を閉状態とする。回転制御部20は、予め設定された時間が経過するまで(ステップS50)、第1バッグ保持部31を第1姿勢に維持する。培地の供給によって、培養バッグ70の内面71,72に付着していた細胞が剥離される。培地の供給による内面71,72からの細胞の剥離は、培地の流速や、第1バッグ保持部31などの姿勢、放置時間などが適宜設定されることによって実現される。なお、培養バッグ70へ培地が供給された後、必要があれば、前述と同様にして培養バッグ70の内部空間75から気体が排出されてもよい。
 回転制御部20は、予め設定された時間が経過した後(ステップS50)、回転機構34を駆動して、第1バッグ保持部31を培養装置10の前方から視て反時計回りに90度回転して第3姿勢とする(ステップS51)。第1バッグ保持部31が第3姿勢にされた後、培養バッグ70からの細胞懸濁液の排出が、前述された液体排出ステップと同様に行われる。給排制御部22は、バルブV6,V9を開状態として、供給ポンプ91を逆向きに駆動する。これにより、培養バッグ70において細胞懸濁液がポート73を通じて内部空間75から排出され(ステップS52)、サーババッグ40に回収される。これにより、細胞懸濁液回収ステップ(ステップS20)が終了する。
[細胞懸濁液濃縮ステップ]
 以下、細胞懸濁液濃縮ステップ(ステップS23)が説明される。濃縮装置120を用いた細胞懸濁液の濃縮方法は、以下に示される各ステップを含む。
(4-1)培養バッグ70からサーババッグ40に細胞懸濁液を移動する移動ステップ。
(4-2)濃縮装置120へプライミング液を供給するプライミングステップ。
(4-3)濃縮装置120へ細胞懸濁液を供給するろ過ステップ。
(4-4)濃縮装置120から細胞懸濁液を回収する回収ステップ。
 図26に示されるように、培養バッグ70からサーババッグ40に細胞懸濁液が移動される(ステップS151)。移動ステップは、前述された細胞懸濁液回収ステップにおいて実行されている。
 続いて、回転制御部20は、濃縮装置支持部104が第1状態であるか否かを判定する(ステップS152)。第1状態とは、濃縮装置120の本体106の軸線方向が鉛直方向に沿っており、流入ポート110が第1排出ポート111より下方にある状態である(図7(B)参照)。濃縮装置支持部104が第1状態でないときは(ステップS152:No)、回転制御部20は、回転機構34を駆動して、濃縮装置支持部104を第1姿勢まで回転する(ステップS153)。なお、第1状態において、濃縮装置120の本体106の軸線方向は、かならずしも鉛直方向に沿っている必要はない。
 続いて、プライミング(ステップS154)が行われる。図7(B)に示されるように、第1状態の濃縮装置支持部104に支持された濃縮装置120へプライミング液が供給される。プライミング液は、例えばリン酸緩衝液などである。給排制御部22は、冷蔵保存部12又は常温保存部13に保持されておりプライミング液を貯留する貯留容器116と接続されているバルブV2と、バルブV8,V13,V14とを開状態として、供給ポンプ91を駆動する。これにより、流入ポート110を通じてプライミング液が濃縮装置120へ供給される。また、プライミング液によって押し出されるようにして、濃縮装置120の第1流出ポート111から空気や、中空糸束109に保存のために充填されていた物質などが排出される。給排制御部22は、供給ポンプ91が駆動されてから予め設定された時間が経過したか否かを監視する。時間としては、供給ポンプ91が濃縮装置120の内部から気体を排出するに十分な時間より長い時間が設定されている。給排制御部22は、供給ポンプ91が駆動されてから予め設定された時間が経過したと判定すると、供給ポンプ91を停止し、バルブV2と、バルブV13,V14とを閉状態とする。これにより、プライミングステップが終了する。
 プライミングステップが終了した後、回転制御部20は、回転機構34を駆動して、濃縮装置支持部104を180度回転する(ステップS155)。これにより、図7(C)に示されるように、濃縮装置120が第2状態となる。第2状態とは、濃縮装置120の本体106の軸線方向が鉛直方向に沿っており、流入ポート110が第1排出ポート111より上方にある状態である。なお、第2状態において、濃縮装置120の本体106の軸線方向は、かならずしも鉛直方向に沿っている必要はない。
 続いて、ろ過ステップ(ステップS156)が行われる。給排制御部22は、バルブV6,V12とを開状態として、供給ポンプ91を駆動する。これにより、サーババッグ40から流入ポート110を通じて濃縮装置120へ細胞懸濁液が供給される。濃縮装置120へ供給された細胞懸濁液は、中空糸束109の内部を流通し、細胞懸濁液に含まれる細胞は中空糸束109の内部の下方に沈殿して、第1流出ポート111付近に溜まる。一方、細胞懸濁液に含まれる培地などの液体は中空糸束109の外側へ流出し、第2流出ポート112を通じて濃縮装置120の外部へ流出される。これにより、濃縮装置120へ流入された細胞懸濁液は、中空糸束109内であって第1流出ポート111付近において濃縮された状態に保持される。濃縮装置120へ細胞懸濁液の全量が供給された後、給排制御部22は、供給ポンプ91を停止し、バルブV6,V12を閉状態とする。これにより、ろ過ステップが終了する(ステップS156)。
 続いて、回収ステップ(ステップS157)が行われる。給排制御部22は、冷蔵保存部12又は常温保存部13に保存された新鮮培地を貯留する貯留容器117と接続されているバルブV1、及びバルブV14,V18を開状態として、供給ポンプ91を駆動する。これにより、図7(D)に示されるように、流入ポート110を通じて濃縮装置120へ新鮮培地が供給される。濃縮装置120において、中空糸束109を流通する新鮮培地は、濃縮された細胞懸濁液と共に、第1排出ポート111を通じて濃縮装置120から回収バッグ41へ流出される。これにより、回収ステップが終了する(ステップS157)。なお、濃縮された細胞懸濁液は、回収バッグ41へ流出させたが、培養バッグ70,80,90へ流出させてもよい。
[本実施形態の作用効果]
 本実施形態によれば、第1姿勢のバッグ保持部31,32,33に保持された培養バッグ70,80,90において、細胞培養が行われる。第1姿勢では、バッグ保持部31,32,33の支持面65,66が略水平方向に沿っているので、培養バッグ70,80,90の内面71,72の一方が略水平方向に沿った状態に保持される。これにより、培養バッグ70,80,90が安定してバッグ保持部31,32,33に支持され、例えば接着性細胞が培養バッグ70,80,90の内面72に付着して培養される。
 第3姿勢のバッグ保持部31,32,33に保持された培養バッグ70,80,90において、ポート74を通じて細胞懸濁液が流出される。第3姿勢では、バッグ保持部31,32,33の支持面が略鉛直方向に沿っているので、培養バッグ70,80,90の内面71,72が略鉛直方向に沿った状態に保持される。培養バッグ70,80,90のポート74が鉛直方向の下方に位置されることによって、重力により培養バッグ70,80,90の内部空間75から細胞懸濁液がポート74を通じて流出される。
 第2姿勢のバッグ保持部31,32,33に保持された培養バッグ70,80,90において、培地交換が行われる。培地交換において、例えば、培養バッグ70,80,90に貯留された培地の一部が流出される。第2姿勢では、培養バッグ70,80,90の内面71,72が水平方向及び鉛直方向のいずれにも沿っていないので、培養バッグ70,80,90のポート74が鉛直方向の下方に位置されることによって、重力により培養バッグ70,80,90の内部空間75からポート74を通じて培地が流出されるが、第3姿勢よりも重力による培養バッグ70,80,90の変形が小さい。すなわち、培養バッグ70,80,90において、内面71,72同士が接触するおそれが低減される。これにより、例えば、培養バッグ70,80,90の内面71,72から細胞が剥がれ落ちるおそれが低減される。
 また、第2姿勢のバッグ保持部31,32,33において、鉛直方向の下方に位置する培養バッグ70,80,90のポート74を通じて培養バッグ70,80,90に貯留された培地が流出される。その後、第1姿勢のバッグ保持部31,32,33において、培養バッグ70,80,90のポート73を通じて内部空間75に培地が流入される。そして、第2姿勢又は第3姿勢のバッグ保持部31,32,33において、鉛直方向の上方に位置する培養バッグ70,80,90のポート74を通じて、液体給排機構37が所定量の吸引を行う。この吸引によって、培地交換において、培養バッグ70,80,90の内部空間75に気体が進入したとしても、第2姿勢又は第3姿勢において、鉛直方向の上方に位置する培養バッグ70,80,90のポート74付近に気体が集まり、その気体がポート74を通じて培養バッグ70,80,90から吸い出される。
 また、培養バッグ70,80,90に形成されたポート73,74は、バッグ保持部31,32,33に保持された状態において、回転軸48,49と直交する方向に沿って延びているため、効率的に培養バッグ70,80,90に対して気体又は液体が流出入される。
 また、培養バッグ70,80,90の内部空間75を区画する縁であって、ポート73,74が配置されている縁87,88は、バッグ保持部31,32,33に保持された状態において、ポート73,74から離れるに従って回転軸48,49へ近づく形状である。このため、培養バッグ70,80,90のポート74が鉛直方向の上方に位置されると、培養バッグ70,80,90の内部空間75においてポート74へ向かって気体が集まりやすくなる。
 また、回転制御部20は、第1情報に応じて、バッグ保持部31,32,33を第1姿勢とした後、第1姿勢から支持面65,66が180度回転された第4姿勢とする。これにより、第1姿勢においては、鉛直方向の上方を向く培養バッグ70,80,90の内面72に、例えば接着性細胞が付着して培養される。第4姿勢においては、第1姿勢において鉛直方向の上方を向いていた培養バッグ70,80,90の内面72が、鉛直方向の下方を向く。他方、第1姿勢において鉛直方向の下方を向いていた培養バッグ70,80,90の内面71が、鉛直方向の上方を向く。これにより、第1姿勢において接着性細胞が付着し難い内面71に、接着性細胞が付着して培養される。
 また、回転制御部20は、第3情報に応じて、バッグ保持部31,32,33を第3姿勢とした後、給排制御部22は、液体給排機構37に第3姿勢のバッグ保持部31,32,33に保持された培養バッグ70,80,90のポート74を通じて培地を流出させる。培養バッグ70,80,90から培地を流出させた後、回転制御部20は、バッグ保持部31,32,33を第1姿勢とし、給排制御部22は、液体給排機構37に第1姿勢のバッグ保持部31,32,33に保持された培養バッグ70,80,90のポート73を通じて剥離液を流入させる。続いて、回転制御部20は、バッグ保持部31,32,33を第3姿勢とし、給排制御部22は、液体給排機構37に、第3姿勢のバッグ保持部31,32,33に保持された培養バッグ70,80,90のポート74を通じて剥離液を流出させる。剥離液を完全に培養バッグ70,80,90から流出させた後に、回転制御部20は、バッグ保持部31,32,33を第1姿勢とし、給排制御部22は、液体給排機構37に第1姿勢のバッグ保持部31,32,33に保持された培養バッグ70,80,90のポート73を通じて培地を流入させる。続いて、回転制御部20は、バッグ保持部31,32,33を第3姿勢とし、給排制御部22は、液体給排機構37に、第3姿勢のバッグ保持部31,32,33に保持された培養バッグ70,80,90のポート74を通じて、細胞懸濁液が培養バッグ70,80,90から流出される。これにより遠心分離を行うことなく培地を溶媒とする細胞懸濁液が回収される。
 また、回転制御部20に第1情報、第2情報、及び第3情報を出力する培養制御部21を更に具備することにより、培養制御部21は、第1情報を出力する培養ステップと、第2情報を出力する培地交換ステップと、第3情報を出力する細胞懸濁液回収ステップと、を実行する。これにより、培養ステップ、培地交換ステップ、及び細胞懸濁液回収ステップが自動的に制御されて行われる。
[変形例]
 なお、本実施形態では、培養制御部21に、培養ステップ、培地交換ステップ、培養ステップ、細胞懸濁液回収ステップが順次行われるように設定されている例が説明されたが、各ステップは任意に設定される。また、各ステップは、それぞれが個別独立して行われるように設定されていてもよい。すなわち、各ステップ毎に培養装置10の動作が停止され、次に行うべき培養装置10の動作をユーザが任意に入力するように設定されていてもよい。
 また、細胞培養において、培養バッグ70,80,90のいずれか一つ又は複数が任意に使用されることができる。また、サーババッグ39,40や回収バッグ41などは任意の構成であり、必要に応じて用いられればよい。
 また、培養バッグ70から気体を排出するときに、第1バッグ保持部31が第3姿勢にされているが、第3姿勢に替えて第2姿勢が採用されてもよい。
 なお、本実施形態では、接着性細胞が培養される例が示されているが、接着性細胞以外の細胞の培養が培養装置10によって行われてもよい。
 また、培養バッグ70には、2つのポート73,74が形成されているが、ポートは1つのみ形成されていてもよい。また、第1バッグ保持部31の支持面65,66が鉛直方向に沿っている姿勢であれば、ポート73,74のいずれかが上方又は下方にあっても、第3姿勢と称しているが、ポートが1つのみ形成されている場合において、細胞懸濁液回収ステップにおいて、培養バッグ70から細胞懸濁液を排出するときは、ポートが下方にある姿勢のみが第3姿勢とされる。
 また、本実施形態では、培養容器として培養バッグ70が用いられたが、培養容器はバッグ形状のものに限定されず、可撓性を有するボトルやカセットなどの形状のものが採用されてもよい。

Claims (10)

  1.  液体が流通するポートを有しており、内部空間において細胞培養が可能な培養容器と、
     上記培養容器を保持する保持板、及び回転軸を有する容器保持部と、
     上記回転軸を支持して回転させる回転機構と、
     上記培養容器へ液体を供給及び排出する液体給排機構と、
     上記回転機構の回転を制御する回転制御部と、を備え、
     上記回転制御部は、
     上記培養容器において細胞培養が行われることを示す第1情報に応じて、上記容器保持部を上記保持板における上記培養容器を支持する支持面が略水平方向に沿った第1姿勢とし、
     上記培養容器において溶液交換が行われることを示す第2情報に応じて、少なくとも上記ポートから上記培養容器内の溶液が流出されるとき、上記容器保持部を上記支持面が水平方向及び鉛直方向のいずれにも沿っていない第2姿勢とする培養装置。
  2.  上記回転制御部は、上記培養容器から溶液が排出されることを示す第3情報に応じて、少なくとも上記ポートから上記培養容器内の溶液が流出されるとき、上記容器保持部を上記支持面が略鉛直方向に沿った第3姿勢とする請求項1に記載の培養装置。
  3.  上記液体給排機構を制御する給排制御部を更に備え、
     上記培養容器は、上記容器保持部に保持された状態において、上記回転軸と直交する方向の両端側にそれぞれ上記ポートを有するものであり、
     上記回転制御部は、上記第2情報又は上記第3情報に応じて、上記容器保持部を上記第2姿勢又は上記第3姿勢とし、
     上記給排制御部は、上記第2情報又は上記第3情報に応じて、上記液体給排機構に、上記第2姿勢又は上記第3姿勢の上記容器保持部に保持された上記培養容器において下方に位置するポートから上記培養容器内の溶液を流出させる請求項2に記載の培養装置。
  4.  上記第1姿勢の上記容器保持部に保持された上記培養容器においていずれか一方のポートから上記培養容器内へ溶液を流入させ、上記第2姿勢又は上記第3姿勢の上記容器保持部に保持された上記培養容器において上方に位置するポートから所定量の吸引を行う請求項2又は3に記載の培養装置。
  5.  上記給排制御部は、上記液体給排機構に、上記第3姿勢の上記容器保持部に保持された上記培養容器のポートから培地を流出させるステップと、
     上記第1姿勢の上記容器保持部に保持された上記培養容器に剥離液を流入させるステップと、
     上記第2姿勢又は上記第3姿勢の上記容器保持部に保持された上記培養容器から剥離液を流出させるステップと、
     上記第1姿勢の上記容器保持部に保持された上記培養容器に培地を流入させるステップと、
     上記第3姿勢の上記容器保持部に保持された上記培養容器から細胞懸濁液を流出させるステップと、を実行させる請求項2から4のいずれかに記載の培養装置。
  6.  上記第2姿勢は、上記支持面が水平方向に対して鋭角をなす角度が20度以上70度以下である請求項1から5のいずれかに記載の培養装置。
  7.  上記培養容器の内面は、接着性細胞を培養するに適した細胞接着性を有する請求項1から6のいずれかに記載の培養装置。
  8.  上記回転制御部は、上記第1情報に応じて、上記容器保持部を上記第1姿勢とした後、
    上記第1姿勢から上記支持面が180度回転された第4姿勢とする請求項1から7のいずれかに記載の培養装置。
  9.  上記回転制御部に上記第1情報、上記第2情報、及び上記第3情報を出力する培養制御部を更に具備しており、
     上記培養制御部は、
     上記第1情報を出力する培養ステップと、
     上記第2情報を出力する溶液交換ステップと、
     上記第3情報を出力する溶液排出ステップと、を実行する請求項1から8のいずれかに記載の培養装置。
  10.  上記培養制御部は、上記溶液交換ステップにおいて、上記液体給排機構を駆動して、上記培養容器内の溶液の一部又は全てを交換する請求項9に記載の培養装置。
