샘플 전처리 시스템 및 그 제어방법이 개시된다. 본 발명에 따른 샘플 전처리 시스템 및 그 제어방법은 작업자가 직접 수작업으로 진행하는 경우에 발생할 수 있는 오류를 최소화하고, 샘플의 전처리 및 테스트 결과에 있어서 그 정확성과 균일성을 확보할 수 있으며, 샘플의 혼합과 배출을 간편하게 수행함으로써 작업의 편의성을 높이고 사용자 친화적인 실험환경을 제공할 수 있다. 그리고 챔버 내의 급격한 압력 변화에도 샘플이 터져 나오는 것을 방지할 수 있도록 챔버 내 압력을 균일하게 유지 및 조절할 수 있고, 챔버 내에 수용된 샘플에 대한 열전달력을 높임으로써 짧은 시간 내에 원하는 온도로 가열하여 샘플의 혼합 및 반응효율을 높일 수 있다. 또한, 자력을 이용하여 샘플의 믹싱 효과를 증가시키고 기계적인 구동을 최소화할 수 있으며, 샘플의 전처리 이후에 이를 정량 토출시킬 수 있다.

Description

샘플 전처리 시스템 및 그 제어방법
본 발명은 샘플 전처리 시스템 및 그 제어방법에 관한 것으로, 보다 상세하게는 샘플 전처리 과정에서의 작업자의 오류를 줄여 시험 결과의 신뢰성을 확보하고, 전처리 과정을 간편하고 손쉽게 수행할 수 있으며, 전처리 된 샘플의 정량 토출이 가능한 샘플 전처리 시스템 및 그 제어방법에 관한 것이다.
일반적으로, 유체시료의 분석은 화학 및 생명공학 분야 외에도 환자로부터 채취한 혈액, 체액의 분석을 통한 진단 분야 등에서 광범위하게 이용되고 있다.
근래에는 이러한 유체시료의 분석을 좀 더 간편하고 효율적으로 수행하기 위하여, 소형화된 다양한 종류의 분석 및 진단 장비들과 기술들이 개발되고 있다.
한편, 이러한 유체시료를 분석하는 방법에 있어서 중요한 것 중 하나는 유체시료를 전처리하는 것이다.
여기서, 유체시료의 전처리란 유체시료의 분석 전에 원하는 양의 샘플을 추출하여 예를 들어, 희석버퍼 등에 이를 적정한 비율로 정확하게 처리하거나, 고체, 액체 상태의 반응시약과의 혼합, 또는 충진 또는 지지체를 활용하여 분리 및 정제하는 것을 말한다.
통상적으로 이를 위해 피펫이나 스포이트를 사용하여 유체시료를 전처리하지만, 랩온어칩(lab-on-a-chip) 또는 랩온어팁(lab-on-a-tip) 단위의 샘플 분석에 있어서, 전처리에 사용되는 시료의 양은 극히 소량이고 또한 아주 정확하게 처리되어야 하기 때문에, 작업자가 직접 피펫이나 스포이트를 이용하여 원하는 극소량의 샘플을 정확하게 전처리하는 것은 쉬운 일이 아니다.
아울러, 현장 검사 기법에서는 채취한 시료를 전처리하고 이를 다시 측정기기로 투입하는 단계를 거치게 되는데, 투입단계에서 다시 한번 인가하는 시료 양의 오차 문제가 발생한다.
따라서, 극소량의 혈액 내지 기타 시료를 전처리함에 있어서 작업자의 오류를 최소화하고, 전처리 과정을 용이하게 간편하고 손쉽게 수행할 수 있으며, 전처리 된 샘플의 정량 토출이 가능한 샘플 전처리 시스템의 필요성이 대두되고 있다.
본 발명의 실시예들은 작업자가 직접 수작업으로 진행하는 경우에 발생할 수 있는 오류를 최소화하고, 샘플의 전처리 및 테스트 결과에 있어서 그 정확성과 균일성을 확보하고자 한다.
또한, 샘플의 혼합과 배출 및 진단 카트리지에의 로딩의 모든 과정을 자동으로 간편하게 수행함으로써 작업의 편의성을 높이고 사용자 친화적인 실험환경을 제공하고자 한다.
또한, 샘플 전처리 및 로딩에 있어서 모듈과 카트리지를 자동으로 가열하여 원하는 시간만큼 원하는 온도로 유지함으로써 믹싱효율과 반응효율을 향상시키고자 한다.
또한, 챔버 내의 급격한 압력 변화에도 샘플이 터져 나오는 것을 방지할 수 있도록 챔버 내 압력을 균일하게 유지 및 조절하고자 한다.
또한, 챔버 내에 수용된 샘플에 대한 열전달력을 높임으로써 짧은 시간 내에 원하는 온도로 가열하여 샘플의 혼합 및 반응효율을 높이고자 한다.
또한, 자력을 이용하여 샘플의 믹싱 효과를 증가시키고 기계적인 구동을 최소화하고자 한다.
또한, 샘플의 전처리와 정량 토출 및 로딩에 대한 원스탑(one-stop) 서비스가 가능한 샘플 전처리 시스템을 제공하고자 한다.
본 발명의 일측면에 따르면, 샘플 전처리 모듈이 안착되는 모듈 홀더가 구비되는 홀더 출납부; 상기 샘플 전처리 모듈의 챔버에 수용된 샘플이 배출되어 로딩(loading)되는 카트리지가 수납되는 카트리지 수납부; 상기 샘플 전처리 모듈 내에 구비된 영구자석을 회전시키도록 자력을 생성하는 자력 생성부; 및, 상기 샘플 전처리 모듈의 관통막을 관통시키고, 캡의 이동부를 가압하여 상기 샘플을 배출시키는 관통 및 배출 실행부;를 포함하는 샘플 전처리 시스템이 제공될 수 있다.
상기 모듈 홀더는, 상기 모듈 홀더 외측을 둘러싸도록 구비되며, 상기 샘플 전처리 모듈을 가열하는 모듈 히터;를 포함하여 이루어질 수 있다.
상기 관통 및 배출 실행부는, 상기 샘플 전처리 모듈의 캡 테두리부를 가압하여 상기 샘플 전처리 모듈 내부의 관통막을 뚫는 테두리 가압부와, 상기 이동부를 가압하여 상기 샘플을 배출시키는 이동부 가압부를 포함하여 이루어질 수 있다.
상기 관통 및 배출 실행부는, 상기 테두리 가압부가 연결되는 제1 이동바와, 상기 이동부 가압부와 연결되는 제2 이동바 및, 상기 제1 이동바와 제2 이동바를 구동시키는 제1 모터를 더 포함하며, 상기 제1 이동바와 제2 이동바는 제1 모터의 구동에 따라 서로 멀어지는 방향 또는 서로 가까워지는 방향으로 동시에 이동하도록 구성될 수 있다.
상기 관통 및 배출 실행부는, 상기 테두리 가압부 중앙부에 형성되는 통공을 더 포함하며, 상기 이동부 가압부는 상기 통공을 통과하여 이동하면서 상기 이동부를 가압하도록 구성될 수 있다.
상기 자력 생성부는, 상기 모듈 홀더 일측에 회전 가능하게 설치되는 볼텍싱(vortexing) 자석과, 상기 볼텍싱 자석을 회전시키는 제2 모터를 포함하여 이루어질 수 있다.
상기 카트리지 수납부는, 상기 카트리지가 안착되며, 상기 카트리지를 로딩 또는 언로딩하는 카트리지 홀더와, 상기 카트리지 홀더를 로딩 또는 언로딩 위치로 이동시키기 위한 구동력을 제공하는 제3 모터를 포함하여 이루어질 수 있다.
상기 카드리지 수납부는 로딩된 상기 카트리지를 가열하는 카트리지 히터를 더 포함하며, 상기 카트리지 히터는 상기 카트리지의 로딩 또는 언로딩에 따라 상승 또는 하강하도록 구성될 수 있다.
본 발명에 따른 샘플 전처리 시스템은 상기 카트리지 히터의 적어도 일부를 둘러싸고, 상기 카트리지에 밀착하는 히터 연결부를 더 포함하며, 상기 히터 연결부는 상기 카트리지의 반응영역보다 더 큰 면적을 갖도록 구비될 수 있다.
상기 카트리지 히터는 상기 카트리지 반응영역을 커버하는 히터 연결부의 영역을 포함하는 면적을 갖도록 구비될 수 있다.
상기 히터 연결부는 상기 카트리지 반응영역에 밀착하도록 평평한 형상으로 이루어지는 것이 바람직하다.
상기 카트리지 히터는 상기 카트리지와 상기 히터 연결부와의 접촉면에 상응하도록 평평한 형상으로 이루어질 수 있다.
본 발명에 따른 샘플 전처리 시스템은 상기 홀더 출납부는 상기 모듈 홀더를 이동하여 상기 모듈 홀더에 안착된 샘플 전처리 모듈을 로딩 또는 언로딩하는 구동력을 제공하는 제4 모터를 더 포함하여 이루어질 수 있다.
본 발명에 따른 샘플 전처리 시스템은 상기 샘플이 배출되는 드랍(drop) 횟수를 감지하는 카운팅 센서를 더 포함하여 이루어질 수 있다.
본 발명의 다른 측면에 의하면, 카트리지를 카트리지 홀더에 안착시킨 후 카트리지 수납부로 로딩하는 단계; 챔버 내에 샘플을 수용한 샘플 전처리 모듈을 모듈 홀더에 안착시킨 후 로딩하는 단계; 상기 모듈 홀더 일측에 위치한 볼텍싱 자석을 회전시킴으로써 상기 챔버 내에 구비된 영구자석을 회전시키는 단계; 상기 샘플 전처리 모듈의 캡 테두리부를 가압하여 상기 샘플 전처리 모듈 내부의 관통막을 뚫는 단계; 및, 상기 샘플 전처리 모듈의 캡의 이동부를 가압하여 상기 샘플을 배출시킴으로써 상기 카트리지에 샘플을 로딩하는 단계;를 포함하는 샘플 전처리 시스템의 제어방법이 제공될 수 있다.
본 발명에 따른 샘플 전처리 시스템의 제어방법은 상기 카트리지를 가열하여 일정 온도로 유지하는 단계를 더 포함하여 이루어질 수 있다.
본 발명에 따른 샘플 전처리 시스템의 제어방법은 상기 모듈 홀더에 안착된 샘플 전처리 모듈을 가열하여 일정 온도로 일정 시간 동안 유지하는 단계를 더 포함하여 이루어질 수 있다.

도 1 내지 도 7을 참조하면, 본 발명의 일 실시예에 따른 샘플 전처리 모듈(100)은 크게 내부에 샘플을 수용하는 챔버(112)를 갖는 몸체(110)와, 상기 몸체(110) 일단에 결합되는 캡(120)과, 적어도 일부에 시약이 도팅(dotting)되고, 상기 챔버(112) 내부로 삽입되는 도팅 부재(dotting substrate, 130) 및, 상기 몸체(110) 타단에 이동 가능하게 결합되며, 상기 챔버(112)에 수용된 샘플을 배출하는 배출팁(140)을 포함하여 이루어질 수 있다.
본 실시예에서 상기 몸체(110)는 일정 높이를 갖는 원통형으로 이루어질 수 있으며, 상기 몸체(110) 내에는 역시 원통형으로 이루어진 일정 공간을 형성하는 챔버(112)가 구비될 수 있다. 물론 상기 몸체(110)와 챔버(112)의 형태는 원통형에 한정되는 것은 아니며, 필요에 따라 다양한 형태로 변형 실시될 수 있다.
상기 몸체(110)는 상기 챔버(112)에 수용된 샘플을 가열할 때, 열전달력을 높일 수 있도록 그 외벽을 가능한 얇게 형성하는 것이 바람직하다.
상기 몸체(110)의 일측에는 유입구(114)가 구비되어 샘플이나 버퍼가 챔버(112)로 주입될 수 있는데, 상기 도팅 부재(130)도 상기 유입구(114)를 통해 챔버(112)로 삽입되어 수용될 수 있다.
상기 몸체(110) 타측에는 배출팁(140)이 결합되며, 챔버(112)와 배출팁(140) 사이에는 관통막(116)이 구비될 수 있다. 상기 관통막(116)은 후술할 관통부(146)에 의해 관통되기 전까지 배출팁(140)과의 연통을 차단함으로써 샘플이 챔버(112) 내에 수용될 수 있다.
상기 관통막(116)에는 샘플 주입 전에 챔버(112) 내로 도팅(dotting)된 버퍼가 도포된 상태로 드라잉(drying)되며, 이후 주입된 샘플이 버퍼와 믹싱(mixing)되어 희석 또는 혼합액을 형성하는 전처리 과정을 거치게 된다.
이때 관통막(116)뿐만 아니라 챔버(112) 내벽 등 챔버(112) 내부에 전처리 물질을 적용하는 것도 가능하다.
상기 몸체(110)와 배출팁(140), 캡(120) 및 관통막(116) 등은 탄성재질의 합성수지로 제조될 수 있는데, 예를 들어 PS(Polystyrene), PP(Polypropylene) 또는 PE(Polyethylene)나 그 외 탄성을 가진 다른 재질도 사용 가능하며, 탄성 재질을 이용한 사출성형에 의해 제조될 수 있다.
특히, 버퍼로서 비타민 D(Vitamin D)가 사용되는 경우에는 상기 배출팁(140)의 수지 재질을 PP로 제작하는 것이 바람직하다. 이는 용액의 점성을 고려한 것으로서 Vitamin D의 전처리 용액은 점성이 낮기 때문에 배출팁(140)을 소수성으로 만들어서 샘플이 임의로 의도치 않게 흘러나오는 것을 방지하고 정량으로 조절되어 배출될 수 있도록 하기 위함이다.
반대로 전처리 용액의 점성이 높을 경우에는 상기 배출팁(140)을 친수성을 띤 재질로 제조하면 버퍼와 혼합된 샘플의 배출이 원활하게 이루어지도록 구성할 수 있다.
상기 도팅 부재(130)는 샘플과 함께 상기 챔버(112) 내에 수용되며, 상기 도팅 부재(130)의 적어도 일측에는 소정의 시약이 도팅되어 삽입됨으로써 수용된 샘플이 시약과 반응하거나 혼합될 수 있다. 상기 도팅 부재(130)에는 시약뿐만 아니라, 추가 샘플이나 전처리 물질이 도팅되어 삽입될 수 있다.
상기 도팅 부재(130)는 도팅 부재 몸체(134)와, 상기 도팅 부재 몸체(134)의 일측으로 연장형성되는 적어도 하나의 제1 연장부(136)와, 상기 도팅 부재 몸체(134)의 타측으로 연장형성되는 적어도 하나의 제2 연장부(138)를 포함하여 이루어질 수 있다.
상기 도팅 부재 몸체(134)는 대략 사각의 다면체 형상으로 이루어지며, 전술한 시약은 상기 제2 연장부(138)에 주로 도팅될 수 있다. 물론 이에 한정되는 것은 아니며, 상기 몸체(134)와 제1 연장부(136)에도 시약이 도팅되어 챔버(112) 내로 삽입될 수 있다.
본 실시예에서 상기 제1 연장부(136)는 두 개가 상기 도팅 부재 몸체(134) 양측으로부터 상방을 향해 연장형성될 수 있다. 본 실시예에서 상기 제1 연장부(136)를 두 개로 분리하여 연장형성하는 이유는 샘플 주입공간 확보를 위한 것으로서, 파이펫(pipette)이 챔버(112) 입구에 들어갈 공간을 마련하기 위한 것이다. 그리고 상기 도팅 부재(130)를 최대한 챔버(112) 외벽에 밀착하도록 설계하였는데 오차가 발생하더라도 유동적으로 끼워넣어질 수 있도록 하기 위함이다.
상기 제2 연장부(138)는 상기 도팅 부재 몸체(134) 하부로부터 하방을 향해 연장형성될 수 있다. 이때 상기 제2 연장부(138)는 상기 도팅 부재 몸체(134)와 단차를 이루면서 하방으로 연장될 수 있다.
원래 도팅되는 시약은 샘플이 접촉하는 면적 내에 존재해야 하는데, 시약을 제2 연장부(138) 표면에 떨어뜨렸을 때 확산되면서 건조되기 때문에 도팅 면적을 국소적으로 조절하기가 어려운 문제점이 있다. 그러나 본 실시예에는 도팅 부재 몸체(134)와 제2 연장부(138)의 사이에 단차가 형성되므로 시약이 단차를 벗어나 확산되지 않고 도팅될 수 있다.
그리고 제2 연장부(138)의 길이는 챔버(112) 내에 수용되는 샘플의 수위만큼(또는 그 이하) 연장형성됨으로써 샘플과 시약이 완전히 반응할 수 있도록 할 수 있다. 즉, 주입되는 샘플양에 따라 상기 제2 연장부(138)의 면적 또는 연장되는 길이가 조절될 수 있다.
물론, 상기 도팅 부재(130)의 형상은 도 4에 도시된 바에 한정되는 것은 아니며, 필요에 따라 다양한 형상으로 변경 실시될 수 있다.
상기 도팅 부재(130)와 함께 영구자석(132)이 챔버(112) 내부로 함께 삽입될 수 있다. 본 실시예에서 상기 영구자석(132)은 원통형으로 형성되며, 외부에서 가해지는 자기장의 변화에 따라 작용하는 자기력에 의해 회전함으로써 상기 샘플을 믹싱(mixing)하는 역할을 수행한다. 이때, 상기 영구자석(132)은 제2 연장부(138)에 의해 형성된 공간에 안착되어 샘플의 믹싱을 수행할 수 있다.
상기 영구자석(132)은 상하방향을 따라 N-S 또는 S-N으로 착자된 것으로서, 상기 챔버(112) 주변에서 전자석(M, 도 29 참조)을 회전시키면 상기 영구자석(132) 또한 원주방향을 따라 회전하게 된다.
이때, 상기 전자석(M)의 회전축과 상기 영구자석(132)의 회전축은 서로 수직을 이룬다. 만일 상기 전자석(M)이 상기 샘플 전처리 모듈(100)의 수직 방향 상부나 하부에 위치하는 경우에는 상기 전자석(M)의 회전축과 상기 영구자석(132)의 회전축은 서로 수평을 이루게 된다.
전술한 바와 같이, 상기 도팅 부재(130)의 제2 연장부(138)에는 추가 샘플이 도팅 및 드라잉되어 제공될 수 있다. 예를 들어 상기 제2 연장부(138)에는 골드 나노 파티클(gold nano particle, G, 도 25 참조)이 도팅(dotting) 및 드라잉(drying)되어 챔버(112) 내에서 샘플과 함께 혼합될 수 있다.
상기 골드 나노 파티클(G) 외에도 다양한 전처리 물질이 사용될 수 있는데 도팅 부재(130)에 전처리 물질을 적용하는 방법은 전술한 도팅 및 건조뿐만 아니라, 동결건조 방법 등이 선택적으로 적용될 수 있다.
그리고 전처리 물질은 관통막(116)이나 챔버(112) 내벽 등 챔버(112) 내부에도 전술한 방법을 이용하여 수용될 수 있다.
상기 도팅 부재(130) 전체의 길이는 상기 챔버(112)의 길이와 일치하도록 구성될 수 있다. 따라서 상기 도팅 부재(130)는 챔버(112) 내에 삽입되었을 때 챔버(112)의 형상을 유지하는 프레임 역할도 함께 수행할 수 있다.
상기 몸체(110)의 유입구(114) 측에는 캡(120)이 결합될 수 있다. 상기 캡(120)은 캡 연결부(128)에 의해 상기 몸체(110) 일측에 연결된 상태로 제공될 수 있다. 여기서 상기 캡(120)은 상기 챔버(112)와 연통되는 중공부(122)와, 상기 중공부(122) 내에 이동 가능하게 구비되어 그 이동에 따라 상기 챔버(112) 내 샘플을 가압하여 외부로 배출하는 이동부(124)를 포함하여 이루어질 수 있다. 상기 이동부(124)는 예를 들어 고무 재질의 패킹(packing)으로 이루어질 수 있다.
상기 중공부(122)는 상기 캡(120)이 상기 몸체(110)와 결합될 때, 상기 챔버(112)와 이어져 연통될 수 있다. 이를 위해 상기 캡(120)은 상기 중공부(122)로부터 연장 형성되는 챔버 연통부(126)를 구비하며, 상기 챔버 연통부(126)가 상기 유입구(114)에 끼워짐으로써 상기 캡(120)이 몸체(110)에 결합되는 동시에 상기 챔버(112)와 중공부(122)가 서로 연통될 수 있다.
그리고 상기 중공부(122) 내에 삽입된 이동부(124)를 가압하면, 상기 이동부(124)가 챔버(112) 방향으로 이동하면서 상기 챔버(112) 내부로 압력이 전달되어 샘플이 외부로 배출될 수 있다.
그런데 전술한 바와 같이 상기 챔버(112)는 관통막(116)에 의해 막혀 있는 상태이므로, 전처리 된 샘플이 배출되기 위해서는 상기 관통막(116)을 먼저 뚫어야 하는 구조이다. 이를 위해 상기 배출팁(140)이 챔버(112)측으로 이동함에 따라 상기 관통막(116)을 관통함으로써 상기 챔버(112) 내 샘플을 배출할 수 있는 배출유로(149)를 형성하는 관통부(146)가 구비될 수 있다.
도 6에 도시된 바와 같이 상기 배출팁(140)은 일측의 배출부(142)와 타측의 관통부(146)가 배출유로(149)에 의해 연통된 double-pointed 구조로 이루어진다. 구체적으로, 상기 배출팁(140)은 상기 몸체(110)에 형성된 삽입공(118)에 삽입되는 삽입몸체(144)를 구비하며, 상기 관통부(146)는 상기 삽입몸체(144)의 상단으로부터 연장 형성된다. 물론 상기 관통부(146)는 이동하면서 상기 관통막(116)을 관통할 수 있도록 첨단이 가늘어져 뾰족한 형상으로 이루어진다.
본 실시예에서 상기 관통부(146)는 중앙부가 뾰족해지도록 양측이 대칭을 이루며 경사지면서 가늘어지는 형상으로 이루어질 수 있다.
상기 삽입공(118)의 내주측으로는 원주방향을 따라 볼록한 형상을 이루는 걸림턱(119)이 형성되고, 상기 삽입몸체(144)의 상부에는 초기 조립 시 상기 걸림턱(119)에 걸려 배출팁(140)의 추가 이동을 제한하는 걸림부(145)가 형성될 수 있다.
따라서 상기 몸체(110)와 배출팁(140)을 초기에 조립할 때, 일단 상기 걸림턱(119)에 걸림부(145)가 걸릴 정도까지만 삽입되며, 나중에 샘플의 희석 또는 혼합과정이 마무리되고 샘플을 배출할 때 추가로 외력을 가하여 밀어넣음으로써 관통부(146)가 관통막(116)을 뚫게 된다.
이때, 상기 관통막(116)에는, 상기 관통부(146)가 관통막(116)의 일정한 위치를 관통할 수 있도록 안내하는 관통가이드(116a)가 형성될 수 있다. 도 2와 도 3에 도시된 것처럼, 상기 관통가이드(116a)는 관통막(116)의 중앙부가 관통될 수 있도록 관통부(146)를 안내하게 된다. 그리고 상기 관통가이드(116a)는 상기 몸체(110)와 배출팁(140)이 초기에 조립되었을 때, 관통부(146)가 관통막(116)의 임의의 위치를 뚫어서 샘플이 샐 수 있는 가능성을 억제하는 역할도 함께 수행할 수 있다.
상기 삽입몸체(144)의 하부에는 플랜지부(143)가 형성된다. 상기 플랜지부(143)가 상기 몸체(110)의 삽입공(118) 테두리에 걸리기 때문에 상기 관통막(116)이 관통막(116)을 관통한 후 배출팁(140)의 추가적인 전진을 제한할 수 있다.
이와 같이 상기 배출팁(140)의 관통부(146)가 관통막(116)을 관통한 상태에서 챔버(112) 내 샘플이 배출될 수 있는 배출유로(149)가 형성되며, 이 상태에서 전술한 바와 같이 캡(120)의 중공부(122)에 구비된 이동부(124)를 눌러 가압하면 샘플이 배출부(142)를 통해 토출될 수 있다.
여기서 토출되는 샘플의 양은 상기 이동부(124)의 이동거리 및 속도에 의해 좌우되는데, 이동부(124)를 작업자가 직접 누를 수도 있지만 일정한 속도와 거리를 가해줄 수 있는 장치를 구성하여 적용함으로써 토출되는 양을 일정하게 정량으로 유지할 수 있다.
한편, 동일한 속도와 이동거리를 적용했을 때 토출되는 샘플의 양은 배출부(142)의 크기를 달리함으로써 조절할 수 있다. 상기 배출팁(140)은 상기 몸체(110)에 탈착 가능하게 결합되므로, 상기 배출부(142)는 샘플의 종류 및 배출되는 양에 적당한 지름을 가지는 배출부(142)로 교환이 가능하다.
또한, 상기 배출부(142)의 크기에 따라서 토출되는 샘플의 방울 부피가 변할 수 있으며, 상기 배출부(142)의 지름에 따라 토출되는 부피의 조절이 가능해 지게 된다.
도 7에서 보는 바와 같이, 상기 배출부(142)는 상기 챔버(112)에서 전처리된 샘플의 종류 및 배출되도록 하는 양 등에 따라 대응되는 지름을 가지도록 형성됨이 바람직하다.
이를 위해, 상기 배출부(142)의 지름은 구의 절단부(spherical cap)의 크기로 구해질 수 있다. 예를 들면, 상기 배출부(142)의 반지름을 a, 배출되는 샘플 방울(d)의 반지름을 r, 구의 절단부의 높이를 h로 했을 때, 상기 샘플 방울(d)의 부피(V)는 V=4/3πr3 이고, r=(a2+h2)/2h 라는 식으로부터 상기 배출부(142)의 지름을 구할 수 있다.
또한, 상기 샘플 방울(d)의 표면장력으로 구할 수도 있다. 예를 들면, W=2πrγ (여기서, W=반응된 검체 방울의 무게, r=토출로의 반지름, γ=표면장력)의 식으로부터 상기 배출부(142)의 지름을 구할 수도 있다.
한편, 상기 관통부(146)가 관통막(116)을 뚫을 때, 챔버(112) 내 압력이 급격히 증가하여 챔버(112)에 수용된 샘플이 터져 나올 수 있다. 그리고 상기 몸체(110) 외주측으로 의도하지 않은 외력이 가해짐에 따라 챔버(112) 내 압력이 상승하는 경우가 발생할 수 있다.
이와 같은 상황에 대비하여 상기 캡(120)의 중공부(122) 내벽에는 상기 챔버(112) 내 압력을 조절하는 밴팅라인(125)이 형성될 수 있다. 상기 밴팅라인(125)은 상기 중공부(122) 내벽에 수직방향을 따라 일정 길이로 형성되며 챔버(112) 내 상승한 압력을 외부로 배출함으로써 챔버(112) 내 압력을 조절하는 역할을 수행할 수 있다.
본 실시예에서 상기 밴팅라인(125)은 상기 중공부(122) 내벽의 일측과 타측에 두 개가 형성됨으로써 하나의 밴팅라인(125)이 막히는 경우에도 챔버(112) 내 압력을 조절하는 역할을 계속해서 수행할 수 있다.
도 8은 본 발명의 일 실시예에 따른 샘플 전처리 시스템의 사시도이고, 도 9는 본 발명의 일 실시예에 따른 샘플 전처리 시스템의 홀더 스테이지가 인출된 상태에서 샘플 전처리 모듈을 안착시킨 상태를 도시한 사시도이며, 도 10은 본 발명의 일 실시예에 따른 샘플 전처리 시스템의 모듈 홀더를 도시한 사시도이다. 도 11은 본 발명의 일 실시예에 따른 샘플 전처리 시스템의 홀더 스테이지가 인출된 상태를 도시한 사시도이고, 도 12는 본 발명의 일 실시예에 따른 샘플 전처리 시스템의 홀더 스테이지가 수납된 상태를 도시한 사시도이다.
도 1 내 도 12를 참조하면, 본 발명의 일 실시예에 따른 샘플 전처리 시스템(1000)은 전술한 샘플 전처리 모듈(100)이 안착되는 모듈 홀더(520)가 구비되는 홀더 출납부(500)와, 상기 샘플 전처리 모듈(100)의 챔버(112)에 수용된 샘플이 배출되어 로딩(loading)되는 카트리지(미도시)가 수납되는 카트리지 수납부(400) 상기 샘플 전처리 모듈(100) 내에 구비된 영구자석(132)을 회전시키도록 자력을 생성하는 자력 생성부(300) 및, 상기 샘플 전처리 모듈(100)의 관통막(116)을 관통시키고, 캡(120)의 이동부(124)를 가압하여 상기 샘플을 배출시키는 관통 및 배출 실행부(200)를 포함하여 이루어질 수 있다.
상기 홀더 출납부(500)는 전처리 대상인 샘플을 수용한 샘플 전처리 모듈(100)을 샘플 전처리 시스템(1000) 내로 로딩하는 역할을 수행한다. 도 8에서는 샘플 전처리 시스템(1000)의 내부 구성이 그대로 드러나도록 도시하였지만, 외측을 덮는 커버(미도시)가 구비될 수 있다.
상기 홀더 출납부(500)의 세부 구성을 살펴보면, 전술한 바와 같이 샘플 전처리 모듈(100)이 안착되는 모듈 홀더(520)가 구비된다. 상기 모듈 홀더(520)는 홀더 스테이지(530)상에 설치되는데, 상기 홀더 스테이지(530)는 이동 가능하게 구비됨으로써 상기 모듈 홀더(520)를 로딩 또는 언로딩 위치로 이동시킬 수 있다.
상기 홀더 스테이지(530)를 모듈 홀더(520)와 함께 이동시킬 수 있는 구동력을 제공하도록 상기 홀더 스테이지(530) 일측에는 제4 모터(510)가 구비될 수 있다. 상기 제4 모터(510)는 모터의 회전축에 연결된 출납부 피니언기어(512)를 회전운동시키며, 상기 출납부 피니언기어(512)는 상기 홀더 스테이지(530) 하부에 구비된 랙기어(미도시)에 맞물려 회전운동을 수평운동으로 변환시킴으로써 홀더 스테이지(530)를 수평방향으로 이동시킨다.
여기서 상기 홀더 스테이지(530) 하부에는 가이드 레일(532)이 구비됨으로써 상기 홀더 스테이지(530) 수평 이동을 가이드할 수 있다.
처음에 상기 샘플 전처리 모듈(100)을 안착시키는 경우에는 상기 제4 모터(510)가 일방향으로 구동하여 홀더 스테이지(530)가 가이드 레일(532)상에서 슬라이딩함으로써 외측으로 인출된다. 이때, 샘플 전처리 시스템(1000)의 전체 커버(미도시) 상에 상기 홀더 스테이지(530)가 출입할 수 있는 도어(미도시)가 구비될 수 있다.
외측으로 인출된 홀더 스테이지(530)의 모듈 홀더(520)상에 샘플 전처리 모듈(100)을 안착시킨 후에는 다시 제4 모터(510)가 반대 방향으로 구동하며, 그에 따라 홀더 스테이지(530)가 가이드 레일(532)상에서 슬라이딩하여 내측으로 수납된다.
이러한 동작은 사용자가 외측에 구비된 스윗치(미도시)를 눌러서 이루어지도록 구현할 수 있다.
상기 홀더 스테이지(530)상에 구비된 모듈 홀더(520)는 상기 샘플 전처리 모듈(100)이 안착될 수 있도록 내측으로 대략 원통형으로 이루어진 공간을 형성한다. 그리고 상기 모듈 홀더(520) 외측에는 상기 샘플 전처리 모듈(100)을 가열하는 모듈 히터(524)가 구비될 수 있다.
구체적으로, 상기 모듈 히터(524)는 도 10에서 보는 바와 같이 홀더 몸체(522)의 외측을 둘러싸는 열선으로 이루어지며, 전류가 흐름에 따라 발열함으로써 홀더 몸체(522) 내측에 안착된 샘플 전처리 모듈(100)을 가열하게 된다.
전술한 바와 같이 본 실시예에 따른 상기 샘플 전처리 모듈(100)의 몸체(110)는 얇은 두께로 이루어져 열전달력이 높기 때문에 열원인 상기 모듈 히터(524)로부터 발산되는 열을 잘 흡수하여 단시간에 원하는 온도로 샘플을 가열할 수 있다. 이때, 상기 모듈 히터(524)와 샘플 전처리 모듈(100) 사이에 개재되는 홀더 몸체(522) 또한 열전도율이 좋은 금속성 재질로 이루어지는 것이 바람직하다. 도 10에서 보는 바와 같이, 상기 모듈 히터(524)의 외측에는 히터 커버(526)를 설치하여 마감한다.
상기 샘플 전처리 모듈(100)의 온도와 유지시간은 전처리 대상인 샘플과 버퍼의 종류가 달라질 수 있다. 예를 들어 Vitamin D의 경우 49℃로 10분간 유지하며, FreeT4 및 Testosterone의 경우 37℃로 5분간 유지한다.
도 13은 본 발명의 일 실시예에 따른 샘플 전처리 시스템의 구조를 도시한 종단사시도이고, 도 14는 본 발명의 일 실시예에 따른 샘플 전처리 시스템의 테두리 가압부가 캡 테두리부를 누르기 시작하는 상태를 도시한 단면도이며, 도 15는 본 발명의 일 실시예에 따른 샘플 전처리 시스템의 테두리 가압부가 캡 테두리부를 가압하여 관통막이 관통된 상태를 도시한 단면도이다. 도 16은 본 발명의 일 실시예에 따른 샘플 전처리 시스템의 이동부 가압부가이동부를 가압하여 샘플을 토출하는 상태를 도시한 단면도이고, 도 17은 본 발명의 일 실시예에 따른 샘플 전처리 시스템의 관통 및 배출 실행부의 작동을 도시한 부분구성도이다.
도 13 내지 도 17을 참조하면, 상기 관통 및 배출 실행부(200)는, 상기 샘플 전처리 모듈(100)의 캡 테두리부(129)를 가압하여 상기 샘플 전처리 모듈(100) 내부의 관통막(116)을 뚫는 테두리 가압부(220)와, 상기 이동부(124)를 가압하여 상기 샘플을 배출시키는 이동부 가압부(230)를 포함하여 이루어질 수 있다.
명칭에서 유추할 수 있듯이 상기 관통 및 배출 실행부(200)는 전술한 샘플 전처리 모듈(100)의 관통막(116)을 관통하는 역할과 샘플이 배출될 수 있도록 가압하는 두 가지 역할을 함께 수행할 수 있다.
이러한 관통 및 배출 실행부(200)의 구성을 자세히 살펴보면 다음과 같다.
도 13 내지 도 16에서 보는 바와 같이 상기 테두리 가압부(220)가 연결 형성되는 제1 이동바(222)와, 상기 이동부 가압부(230)와 연결 형성되는 제2 이동바(232)가 각각 하부와 상부에 배치될 수 있다. 즉, 상기 제1 이동바(222) 상부에 제2 이동바(232)가 구비되는 것이다.
상기 제1 이동바(222)와 제2 이동바(232)는 고정축(218)상에서 상하로 이동 가능하게 구비되는데, 본 실시예에서는 서로 멀어지는 방향 또는 서로 가까워지는 방향으로 동시에 이동하도록 구성된다.
상기 고정축(218)과 제1 이동바(222) 사이 및 상기 고정축(218)과 제2 이동바(232) 사이에는 각각 제1 습동베어링(226)과 제2 습동베어링(236)이 구비되어 상기 제1 이동바(222)와 제2 이동부(124)의 이동이 부드럽고 원활하게 이루어지도록 하는 역할을 수행할 수 있다.
상기 제1 이동바(222)와 제2 이동바(232)가 이동하는 구동력은 일측에 구비된 제1 모터(210)가 제공할 수 있다. 상기 제1 모터(210)의 회전력은 구동기어(212, 214)에 의해 회전축(216)으로 전달되며, 그에 따라 상기 회전축(216)이 시계방향 또는 반시계방향으로 회전할 수 있다.
도 17에서 보는 바와 같이, 상기 회전축(216)이 제1 이동바(222)와 제2 이동바(232)와 접하는 부분에는 나사산(screw thread)이 형성되며, 상기 제1 이동바(222)와 제2 이동바(232)에도 그에 대응되도록 나사산이 형성될 수 있다. 이때, 나사산의 회전 방향은 제1 이동바(222)와 제2 이동바(232)가 서로 반대 방향을 이루도록 형성되는 것이 바람직하다.
이와 같이 제1 이동바(222)측 나사산과 제2 이동바(232)측 나사산이 서로 반대 방향으로 형성되면, 상기 회전축(216)이 회전할 때 상기 제1 이동바(222)와 제2 이동바(232)는 서로 반대방향으로 이동하게 된다.
예를 들어, 도 17(a)에 도시된 것처럼 상기 회전축(216)이 시계 반대 방향으로 회전하면 상기 제1 이동바(222)와 제2 이동바(232)는 서로 가까워지는 방향으로 이동한다. 그리고 도 17(b)에 도시된 것처럼 상기 회전축(216)이 시계 방향으로 회전하면 상기 제1 이동바(222)와 제2 이동바(232)는 서로 멀어지는 방향으로 이동한다.
상기 제1 이동바(222)와 제2 이동바(232)의 구동은 상기와 같은 방식으로 이루어지게 되며, 그에 따라 상기 테두리 가압부(220)와 이동부 가압부(230)가 각각의 역할을 수행한다.
상기 테두리 가압부(220)와 이동부 가압부(230)는 상기 모듈 홀더(520)가 로딩되었을 때 샘플 전처리 모듈(100)의 수직 상방에 일렬로 정렬될 수 있다.
상기 테두리 가압부(220)는 도 13 내지 도 16에 도시된 것처럼 제1 이동바(222) 단부에 형성되며, 샘플 전처리 모듈(100) 캡(120)의 상면 특히 캡 테두리부(129)를 누를 수 있는 형상으로 이루어질 수 있다.
그리고 상기 이동부 가압부(230)는 제2 이동바(232) 단부에 연결 결합되는데 하부로 길게 연장된 원통형 바(bar) 형상으로 이루어져 상기 캡(120)의 중공부(122)에 위치한 이동부(124)를 누를 수 있도록 구비된다.
이때 상기 테두리 가압부(220)와 이동부 가압부(230)가 서로 간섭하지 않도록 상기 테두리 가압부(220)의 중앙측에는 통공(224)이 형성되며, 이를 통해 상기 이동부 가압부(230)가 상기 통공(224)을 통과하여 이동하면서 상기 이동부(124)를 가압할 수 있다.
이와 같이 구성된 테두리 가압부(220)와 이동부 가압부(230)의 작동 과정을 살펴보면 먼저, 도 14에서 보는 바와 같이 샘플 전처리 모듈(100)이 모듈 홀더(520)에 안착된 상태에서 로딩되면, 샘플 전처리 모듈(100)과 테두리 가압부(220) 및 이동부 가압부(230)가 수직으로 정렬된다. 이때, 상기 샘플 전처리 모듈(100)의 배출팁(140)은 일부만 끼워진 상태로서, 배출팁(140) 상단의 관통부(146)가 관통막(116)을 관통하지 않은 상태이다.
이 상태에서 제1 모터(210)가 구동하여 회전축(216)이 일방향으로 회전하면 상기 제1 이동바(222)와 제2 이동바(232)는 서로 멀어지는 방향으로 이동하게 되고 그에 따라 상기 테두리 가압부(220)가 하부로 이동하면서 상기 샘플 전처리 모듈(100)의 캡 테두리부(129)를 가압하게 된다.
상기 테두리 가압부(220)가 캡 테두리부(129)를 누르면 샘플 전처리 모듈(100)의 몸체(110)가 하부로 내려가게 되고 상기 관통막(116)이 관통부(146)에 의해 뚫리게 된다.
그 후 상기 제1 모터(210)의 구동방향이 바뀌면 회전축(216)이 반대방향으로 회전하게 되고 그에 따라 도 16에서 보는 것과 같이, 상기 제1 이동부(124)와 제2 이동부(124)는 서로 가까워지는 방향으로 이동한다.
이때, 상기 이동부 가압부(230)는 상기 테두리 가압부(220)의 중앙부에 형성된 통공(224)을 통과하여 하방으로 이동하면서 상기 캡(120)의 중공부(122)에 위치한 이동부(124)를 누르게 된다.
그리고 상기 이동부(124)가 하방으로 이동하면서 가압하면 챔버(112) 내의 샘플이 상기 배출부(142)를 통해 배출된다. 이와 같이 배출된 샘플은 후술할 유체분석용 카트리지(10, 도 24 참조)에 드랍되어 진단 및 분석에 활용된다.
이와 같이 관통막(116)의 관통과 샘플의 배출을 자동화함으로써 사용자의 편의성을 증대할 수 있을 뿐만 아니라, 샘플의 배출 시 이동부 가압부(230)를 기계적인 제어를 통해 일정한 속도로 일정한 거리만큼 이동하여 가압하도록 구현할 수 있다.
또한, 이동부 가압부(230) 이동속도와 이동거리를 샘플의 종류나 점성 등 물성에 따라 원하는 만큼 정확하게 변경할 수 있으므로, 휴먼 에러(human error)를 최대한 배제하고 항상 정량 토출이 가능하다.
전술한 바와 같이, 토출되는 샘플의 양은 샘플 전처리 모듈(100)의 이동부(124)의 이동거리 및 속도에 의해 좌우되는데, 이동부(124)를 상기 이동부 가압부(230)를 통해 일정한 속도와 거리로 가압해줌으로써 토출되는 양을 일정하게 정량으로 유지할 수 있다.
한편, 본 발명에 따른 샘플 전처리 시스템(1000)은 상기 샘플이 배출되는 드랍(drop) 횟수를 감지하는 카운팅 센서(600)를 구비할 수 있다. 상기 카운팅 센서(600)는 도 14 내지 도 16에 도시된 것처럼 상기 샘플이 드랍되는 부분 일측에 구비될 수 있다.
본 실시예에서 상기 카운팅 센서(600)로는 예를 들어 광 화이버 센서(optical fiber sensor)가 사용될 수 있다. 광 화이버 센서는 렌즈를 제거한 앰프에 광 화이버 케이블을 조합하여 제조될 수 있는데, 빠른 응답속도를 얻을 수 있고, 초소형 물체 검출이 가능하며, 좁은 공간에 적용할 수 있을 뿐만 아니라, 고온에 적용 가능한 장점이 있다.
상기 카운팅 센서(600)는 상기 모듈 홀더(520) 하부 일측 즉, 샘플 전처리 모듈(100)로부터 샘플이 드랍되는 부분 근처에 설치될 수 있다.
상기 카운팅 센서(600)로 광 화이버 센서가 사용되는 경우 발광부에서 레이져 빔을 항상 쏘고 있는 상태에서 샘플이 드랍될 때마다 반사되는 레이져 빔을 수광부를 통해 검지함으로써 드랍 횟수를 카운팅하게 된다.
물론 상기 카운팅 센서(600)는 광 화이버 센서에 한정되는 것은 아니며, 샘플 드랍 횟수를 감지할 수 있는 여하한 센서가 적용될 수 있다.
도 18은 본 발명의 일 실시예에 따른 샘플 전처리 시스템의 자력 생성부를 도시한 사시도이다.
도 18을 참조하면, 상기 모듈 홀더(520) 일측에는 상기 샘플 전처리 모듈(100) 내에 구비된 도팅 부재(130)를 회전시키도록 자력을 생성하는 자력 생성부(300)가 구비될 수 있다.
그리고 상기 자력 생성부(300)는, 상기 모듈 홀더(520) 일측에 회전 가능하게 설치되는 볼텍싱(vortexing) 자석(320)과, 상기 볼텍싱 자석(320)을 회전시키는 제2 모터(310)를 포함하여 이루어질 수 있다.
상기 제2 모터(310)와 볼텍싱 자석(320)의 회전축은 풀리(312)와 벨트(314)에 의해 연결됨으로써 상기 제2 모터(310)의 회전력을 볼텍싱 자석(320)으로 전달할 수 있다.
전술한 바와 같이, 상기 챔버(112) 내에 수용된 영구자석(132)은 상하 방향으로 N-S 또는 S-N으로 착자되며, 상기 볼텍싱 자석(320)이 회전함에 따른 자기장 변화로 인해 영구자석(132)에 회전력이 걸려 회전한다.
이때, 상기 영구자석(132)의 회전축은 수직방향이고, 볼텍싱 자석(320)의 회전축은 수평방향이므로 서로 수직을 이루게 된다.
도 19는 본 발명의 일 실시예에 따른 샘플 전처리 시스템의 카트리지 수납부를 도시한 사시도이고, 도 20은 본 발명의 일 실시예에 따른 샘플 전처리 시스템의 카트리지 수납부의 슬라이딩 도어가 인출 또는 수납된 상태를 도시한 사시도이며, 도 21은 본 발명의 일 실시예에 따른 샘플 전처리 시스템의 카트리지 수납부의 슬라이딩 도어가 인출된 상태를 도시한 평면도이다. 도 22는 본 발명의 일 실시예에 따른 샘플 전처리 시스템의 카트리지 수납부 작동구조를 도시한 부분사시도이고, 도 23은 본 발명의 일 실시예에 따른 샘플 전처리 시스템의 카트리지 히터가 상승 또는 하강하는 작동구조를 도시한 구성도이며, 도 24는 본 발명의 일 실시예에 따른 샘플 전처리 시스템에 카트리지가 수납된 상태를 도시한 사시도이다.
도 19 내지 도 24를 참조하면, 상기 카트리지 수납부(400)는 상기 카트리지(10, 도 24 참조)가 안착되며, 상기 카트리지(10)를 로딩 또는 언로딩하는 카트리지 홀더(420)와, 상기 카트리지 홀더(420)를 로딩 또는 언로딩 위치로 이동시키기 위한 구동력을 제공하는 제3 모터(410)를 포함하여 이루어질 수 있다.
본 발명에 따른 샘플 전처리 시스템(1000)은 외측을 덮는 커버(미도시)의 도시가 생략되었는데, 샘플 전처리 시스템(1000)의 전체 커버 상에 상기 카트리지 홀더(420)가 출입할 수 있는 출입구(미도시)가 구비될 수 있다.
상기 카트리지 홀더(420)는 수납부 몸체(110) 일측 하부에 구비된 제3 모터(410)의 구동력에 의해 인출 또는 수납될 수 있다. 구체적으로 상기 제3 모터(410)의 회전력은 벨트, 풀리 및 구동기어의 조합에 의해 수납부 피니언 기어(412)로 전달된다.
상기 수납부 피니언 기어(412)는 상기 카트리지 홀더(420)에 형성된 수납부 렉기어(422)와 맞물려 동력을 전달한다. 따라서 상기 제3 모터(410)의 회전축이 일방향 또는 그 반대방향으로 회전함에 따라 상기 카트리지 홀더(420)가 인출 또는 수납 위치로 이동할 수 있다.
한편, 상기 카트리지 수납부(400)는 로딩된 상기 카트리지(10)를 가열하는 카트리지 히터(430)를 더 포함하여 이루어지는데, 상기 카트리지 히터(430)는 상기 카트리지(10)의 로딩 또는 언로딩에 따라 상승 또는 하강하도록 구성될 수 있다.
이를 위해 상기 카트리지 히터(430)는 히터 연결부(432)에 고정 설치되며, 상기 히터 연결부(432)는 승하강부(434)에 결합된다. 그리고 상기 승하강부(434)는 상기 카트리지 홀더(420) 수납 또는 인출에 따라 상승 또는 하강하는 승하강 운동을 한다.
구체적으로, 도 22에 도시된 바와 같이 렉 가이드(440)가 추가로 구비되는데, 상기 렉 가이드(440)는 일측에 구비된 가이드 돌기(444)가 상기 카트리지 홀더(420)에 형성된 가이드 라인(424)에 맞물린 상태로 제공된다.
그리고 상기 카트리지 홀더(420)가 움직이면 상기 가이드 라인(424)의 경로에 따라 상기 렉 가이드(440)가 이동하게 된다. 즉, 도 21의 평면도를 기준으로 상기 카트리지 홀더(420)가 외부로 인출되도록 이동하면 상기 가이드 라인(424)에 맞물린 가이드 돌기(444)로 인해 렉 가이드(440)가 도면상에서 위쪽으로 수평 이동한다.
상기 카트리지 홀더(420)가 내측으로 수납되도록 이동하면 상기 가이드 라인(424)에 맞물린 가이드 돌기(444)로 인해 렉 가이드(440)가 도면상에서 아래쪽으로 수평 이동한다. 이때, 상기 렉 가이드(440) 일측에 형성된 가이드 렉 기어(442)가 수납부 피니언 기어(412)에 맞물리게 되어 더욱 안정적인 동력 전달이 이루어질 수 있다.
한편, 상기 렉 가이드(440)의 수직면 일측에는 승하강 가이드 라인(446)이 형성되며, 상기 승하강부(434) 후면에는 상기 승하강 가이드 라인(446)에 끼워져 맞물릴 수 있는 승하강부 돌기(436)가 형성되어 있다.
따라서 도 23에 도시된 바와 같이 상기 렉 가이드(440)가 수평 이동하면 상기 승하강 가이드 라인(446)의 경로를 따라 승하강부 돌기(436)가 이동하게 되고 그에 의해 상기 승하강부(434)가 상승 또는 하강 운동을 하게 된다.
결국, 상기 제3 모터(410)의 구동력에 의해 상기 카트리지 홀더(420)의 수납 또는 인출을 위한 이동이 이루어지는 동시에, 상기 카트리지 히터(430)도 함께 승하강 운동을 하는 것이다.
이와 같이 구성하는 이유는 상기 카트리지(10)하부에 배치되는 카트리지 히터(430)와 카트리지 홀더(420)가 수납 또는 인출을 위해 이동할 때, 서로 간섭 내지 충돌하는 것을 방지하기 위함이다.
즉, 상기와 같이 카트리지 홀더(420)를 인출할 때에는 카트리지 히터(430)가 하강하고, 카트리지 홀더(420)를 수납할 때 카트리지 히터(430)가 상승하도록 구현하면 위와 같은 문제점을 해결할 수 있다.
한편, 도 24에 도시된 바와 같이, 상기 히터 연결부(432)는 상기 카트리지 히터(430)의 적어도 일부를 둘러싸도록 구비되며, 카트리지 히터(430) 상승 시, 상기 히터 연결부(432)가 카트리지(10) 하부에 밀착한다.
상기 히터 연결부(432)는 열전도성이 우수한 재질로 이루어지는 것이 바람직하며, 열원인 카트리지 히터(430)로부터 카트리지(10)에 열을 전달하여 카트리지(10)가 가열될 수 있도록 한다.
예를 들어, 상기 히터 연결부(432)는 열전도성이 높으면서도 경도가 높은 알루미늄 재질이 적용될 수 있다. 구리의 경우도 열전도성이 좋지만, 경도가 낮아 잘 휘어지는 문제가 있으므로 본 실시예에서는 알루미늄을 히터 연결부(432)에 적용하였다.
상기 카트리지 히터(430)는 매우 뜨겁기 때문에 카트리지(10)에 직접 밀착하는 경우 수지로 이루어진 카트리지(10)가 녹을 수 있으며, 이를 방지하기 위해 열전도성 재질로 이루어진 히터 연결부(432)를 통해 간접적으로 가열하는 것이다.
여기서, 상기 히터 연결부(432)가 커버하는 영역은 상기 카트리지(10)의 반응영역(R)으로서, 상기 히터 연결부(432)는 상기 카트리지(10)의 반응영역(R)보다 더 큰 면적을 갖도록 이루어지는 것이 바람직하다.
그리고 상기 카트리지 히터(430)는 상기 반응영역(R)을 커버하는 히터 연결부(432)의 영역을 포함하는 면적을 갖도록 이루어지는 것이 바람직하다.
상기 카트리지(10)의 샘플 반응영역(R)은 일정온도로 유지하는 것이 필요하므로 열원인 카트리지 히터(430)로부터 발산되는 열을 히터 연결부(432)가 반응영역(R)에 균일하고 충분하게 전달할 수 있는 역할을 수행해야 한다.
이때, 상기 히터 연결부(432)의 열전달력을 높이기 위해서는 반응영역(R)보다 상대적으로 큰 열용량을 가지도록 면적이 크게 제작되어야 한다. 만약 히터 연결부(432)가 반응영역(R)의 면적보다 작을 경우에는 충분한 열전달이 이루어지지 않을 뿐만 아니라 반응영역(R) 내에서 열구배가 발생하여 카트리지(10)의 채널 내 유체의 흐름에 불균형을 초래할 수 있다.
따라서 상기 히터 연결부(432)는 상기 카트리지(10)의 반응영역(R)보다 더 큰 면적을 갖도록 구성된다.
더 나아가 이러한 히터 연결부(432)가 반응영역(R)을 모두 커버하고 있더라도 카트리지 히터(430) 자체의 면적이 상기 히터 연결부(432)가 커버하는 영역보다 작거나 어긋나 있다면 알루미늄의 열전도성이 아무리 좋다 해도 히터 연결부(432) 자체에 열구배가 발생하여 유체 흐름에 악영향을 미칠 수 있다.
따라서 상기 카트리지 히터(430)는 상기 반응영역(R)을 커버하는 히터 연결부(432)의 영역을 포함하는 면적을 갖도록 구성된다.
그리고 상기 히터 연결부(432)는 평평한 형상으로 이루어지는 것이 바람직하다. 그리고 상기 카트리지 히터(430)는 히터 연결부(432)와의 접촉면에 상응하도록 평평한 형상으로 이루어지는 것이 바람직하다.
이와 같이 카트리지 히터(430)와 히터 연결부(432)를 카트리지(10)와의 접촉면에 상응하도록 평평하게 제작하는 이유는 접촉부위가 밀착되지 않으면 공기에 의한 열손실이 발생하기 때문에 공기층을 최소화할 수 있도록 밀착시키기 위한 것이다.
도 24는 본 발명의 일 실시예에 따른 샘플 전처리 시스템에 카트리지가 수납된 상태를 도시한 사시도이고, 도 25는 본 발명의 일 실시예에 따른 샘플 전처리 모듈의 도팅 부재에 골드 나노 파티클을 도팅하고 드라잉하는 과정을 도시한 구성도이며, 도 26은 본 발명의 일 실시예에 따른 샘플 전처리 모듈의 챔버 내에 버퍼를 도팅하고 드라잉하는 과정을 도시한 구성도이다. 도 27은 본 발명의 일 실시예에 따른 샘플 전처리 모듈의 각 파트를 조립한 상태를 도시한 구성도이고, 도 28은 본 발명의 일 실시예에 따른 샘플 전처리 모듈의 챔버에 샘플을 주입한 상태를 도시한 구성도이며, 도 29는 본 발명의 일 실시예에 따른 샘플 전처리 모듈의 영구자석에 자력을 가하여 샘플을 믹싱하는 과정을 도시한 구성도이다. 도 30은 본 발명의 일 실시예에 따른 샘플 전처리 모듈의 캡 테두리부를 눌러 관통막을 관통하는 과정을 도시한 구성도이고, 도 31은 본 발명의 일 실시예에 따른 샘플 전처리 모듈 캡의 이동부를 눌러 샘플을 배출하는 과정을 도시한 구성도이다.
도 1 내지 도 31을 참조하여, 본 발명의 일 실시예에 따른 샘플 전처리 시스템(1000)을 통한 샘플 전처리 과정을 살펴보면 다음과 같다.
먼저, 도 24에 도시된 바와 같이, 카트리지(10)를 카트리지 수납부(400)에 안착시킨 후 수납한다. 상기 카트리지(10)를 수납할 때 전술한 바와 같이 하강해 있던 상기 카트리지 히터(430)가 상승하여 상기 카트리지(10) 하부에 밀착하게 된다.
상기 카트리지 히터(430)에 의해 상기 카트리지(10)가 일정 온도로 가열되는데, 상기 카트리지(10)에서 항원 항체 반응이 원활하게 이루어질 수 있도록 카트리지(10)의 온도는 예를 들어 약 32℃로 유지될 수 있다.
그리고 도 25에서 보는 바와 같이, 샘플 전처리 모듈(100)에 삽입할 도팅 부재(130)의 제2 연장부(138)에 추가 샘플을 도팅한다. 본 실시예에서는 상기 제2 연장부(138)에 골드 나노 파티클(G)을 도팅하고 드라잉한다.
그 다음 도 26에 도시된 바와 같이, 샘플 전처리 모듈(100)의 챔버(112) 내 관통막(116)상에 릴리스 버퍼(release buffer)를 도팅하고 드라잉한다. 이때 vitamin D mode에서는 vitamin D가 버퍼로 사용될 수 있다.
그 후 도 27에 도시된 것처럼 샘플 전처리 모듈(100)의 각 파트를 조립한다. 즉, 상기 챔버(112) 내에 영구자석(132)과 도팅 부재(130)를 배치하고 상기 몸체(110)에 배출팁(140)을 결합한다. 이때 상기 배출팁(140)은 관통부(146)가 관통막(116)을 관통하지 않도록 일부만 끼워진 상태이다. 그리고 상기 캡(120)의 중공부(122) 내에 이동부(124)를 끼워 넣는다.
그리고 도 28에 도시된 것처럼 상기 챔버(112) 내에 샘플을 주입하고 나서 캡(120)을 닫은 후 샘플 전처리 시스템(1000)의 모듈 홀더(520)에 샘플 전처리 모듈(100)을 안착시켜 로딩한다.
이와 같이 샘플 전처리 모듈(100)을 샘플 전처리 시스템(1000)에 로딩한 후 도 29에 도시된 것처럼 볼텍싱 자석(320)을 회전시키면 상기 영구자석(132)에 자력이 작용하여 회전하며, 그에 따라 샘플의 혼합이 이루어질 수 있다.
그리고 모듈 히터(524)를 가동하여 샘플을 가열하는데, 적용되는 온도를 구체적으로 예를 들면, vitamin D mode에서는 49℃로 약 10분간 가열하며, Free T4, testosterone에서는 37℃로 약 5분간 가열한다.
이와 같이 전처리가 완료되면 도 30에 도시된 바와 같이, 상기 테두리 가압부(220)가 캡 테두리부(129)를 하부로 눌러서 관통막(116)을 관통시킨다. 상기 관통막(116)을 뚫은 후에는 도 31에 도시된 것처럼 상기 이동부 가압부(230)가 하방으로 이동하면서 상기 캡(120)의 중공부(122)에 위치한 이동부(124)를 누르게 된다.
그리고 상기 이동부(124)가 하방으로 이동하면서 가압하면 챔버(112) 내의 샘플이 상기 배출부(142)를 통해 정량 토출된다. 이때 상기 이동부(124)는 상기 이동부 가압부(230)에 의해 일정한 이동거리 및 속도로 가압되므로 토출되는 양을 일정하게 정량으로 유지할 수 있다.
이와 같이 배출된 샘플은 하부에 위치한 유체분석용 카트리지(10)에 드랍되어 진단 및 분석에 활용된다.
이때, 도 31에서 보는 바와 같이 전술한 카운팅 센서(600)를 이용하여 드랍수를 감지한다. 그리고 샘플 로딩에 필요한 정상 드랍(drop) 횟수를 미리 설정하고 정상 횟수에서 벗어나는 경우 에러 모드(error mode)로 판단할 수 있다.
예를 들어 다음과 같이 에러 코드(error code)를 설정할 수 있다.
(1) Error 1
이동부(124)가 최대로 눌러지는 위치에 도달할 때까지 정상 드랍수가 완전히 카운팅되지 않았으면 에러로 인식한다. 이 경우 정상 드랍수에 미달되는 것으로 판단한다.
에러 발생의 원인은 다음과 같다.
- 샘플 용량이 적은 경우
- 카운팅 센서(600)의 오작동
- 샘플 전처리 모듈(100)에서 샘플이 누수된 경우
(2) Error 2
관통막(116)을 뚫는 동작의 위치에서 5 드랍수 이상이 감지되면 에러로 인식한다. 상기 위치는 테두리 가압부(220)의 하단을 기준으로 위치를 좌표화하여 설정한다. 이 경우 정상 드랍수를 초과하는 것으로 판단한다.
에러 발생의 원인은 다음과 같다.
- 이동부(124)를 누르기 전에 관통부(146)가 관통막(116)을 뚫는 타이밍에 샘플이 과도하게 나옴
- 샘플 전처리 모듈(100)의 불량

정상적으로 드랍되는 시간보다 짧은 시간 안에 드랍수가 모두 카운팅되면 에러로 인식한다. 예를 들어 5 드랍수 측정 시, 10초 이내에 5 드랍이 모두 카운팅 되면 에러로 인식하도록 설정함으로써 정상보다 빠르게 드랍 되는 현상을 감지할 수 있다. 이 경우 정상 드랍수를 초과하는 것으로 판단한다.
에러 발생의 원인은 다음과 같다.
- 이동부(124)를 누르기 전에 관통부(146)가 관통막(116)을 뚫는 타이밍에 샘플이 과도하게 나옴
- 샘플 전처리 모듈(100)의 불량
(4) Error 4
정상 동작 후 이동부 가압부(230)가 초기 위치로 올라갈 때 드랍이 감지되면 에러로 판단한다. 이 경우 정상 드랍수를 초과하는 것으로 판단한다.
에러 발생의 원인은 다음과 같다.
- 정상 동작 후 배출팁(140)에 맺혀 있던 드랍이 추가적으로 떨어지는 경우
(5) Error 5
인큐베이션(incubation) 시간이 끝나고 동작하기 시작했을 때부터 정상 드랍이 완료되었을 시간 내에 드랍수가 미달인 경우 에러로 인식한다. 여기서 정상 드랍이 완전히 완료되었을 것으로 예상되는 충분한 시간을 기준 시간으로 설정하여 에러 여부를 판단한다. 예를 들어 정상 동작 시간이 약 1분 30초일 경우, 설정 시간은 3분으로 한다. 이 경우 정상 드랍수에 미달되는 것으로 판단한다.
에러 발생의 원인은 다음과 같다.
- 샘플 용량이 적은 경우
- 카운팅 센서(600)의 오작동
- 샘플 전처리 모듈(100)에서 샘플이 누수된 경우
표 1과 표 2는 본 발명의 샘플 전처리 모듈(100)을 이용하여 실제 전처리 및 정량 토출 실험을 실시한 결과로서, Plasma와 Gold particle을 혼합하고 37℃로 5분간 유지하는 조건을 적용하였다.
시험은 표 1과 표 2 모두 다음과 같은 과정을 거쳐 이루어졌다.
(1). dotting substrate에 5ul의 gold particle을 drying dotting 한다.
(2). (1)의 dotting substrate를 모듈에 삽입한다.
(3). 피펫(Reference 2,eppendorf)을 이용하여 70ul의 혈장을 덜어내, (2)의 모듈에 주입한다.
(4). (3)의 모듈을 전처리 시스템 기기에 삽입한다.
(5). (4)의 모듈을 전처리 시스템 기기에서 5분간 37℃로 가열해 준다.
(6). (5)가 이루어지고 있는 동안, magent stirring 방식으로 gold particle과 혈장을 30초간 혼합한다.
(7). 가열이 끝난 (6)의 용액을 3방울 정량 토출시킨다.
(8). 3방울 토출을 확인 후, 전자식 저울(ME204, METTLER TOLEDO)로 용량을 측정한다.

표 1과 표 2에서 보듯이 토출량이 마이크로 단위임에도 불구하고 거의 일정한 수준을 유지하고 그 표준편차가 1 미만임을 확인할 수 있다.
지금까지 설명한 본 발명의 실시예들에 따른 샘플 전처리 시스템은 작업자가 직접 수작업으로 진행하는 경우에 발생할 수 있는 오류를 최소화하고, 샘플의 전처리 및 테스트 결과에 있어서 그 정확성과 균일성을 확보할 수 있으며, 샘플의 혼합과 배출 및 진단 카트리지에의 로딩의 모든 과정을 자동으로 간편하게 수행함으로써 작업의 편의성을 높이고 사용자 친화적인 실험환경을 제공할 수 있다.
그리고 샘플 전처리 및 로딩에 있어서 모듈과 카트리지를 자동으로 가열하여 원하는 시간만큼 원하는 온도로 유지함으로써 믹싱효율과 반응효율을 향상시킬 수 있고, 챔버 내의 급격한 압력 변화에도 샘플이 터져 나오는 것을 방지할 수 있도록 챔버 내 압력을 균일하게 유지 및 조절할 수 있으며, 챔버 내에 수용된 샘플에 대한 열전달력을 높임으로써 짧은 시간 내에 원하는 온도로 가열하여 샘플의 혼합 및 반응효율을 높일 수 있다.
또한, 자력을 이용하여 샘플의 믹싱 효과를 증가시키고 기계적인 구동을 최소화할 수 있고, 샘플의 전처리와 정량 토출 및 로딩에 대한 원스탑(one-stop) 서비스가 가능하다.
상기에서는 본 발명의 일 실시예를 참조하여 설명하였지만, 해당 기술분야의 당업자는 이하에서 서술하는 특허청구범위에 기재된 본 발명의 사상 및 영역으로부터 벗어나지 않는 범위 내에서 본 발명을 다양하게 수정 및 변경 실시할 수 있을 것이다. 그러므로 변형된 실시가 기본적으로 본 발명의 특허청구범위의 구성요소를 포함한다면 모두 본 발명의 기술적 범주에 포함된다고 보아야 한다.

Claims (17)

  1. 샘플 전처리 모듈이 안착되는 모듈 홀더가 구비되는 홀더 출납부;
    상기 샘플 전처리 모듈의 챔버에 수용된 샘플이 배출되어 로딩(loading)되는 카트리지가 수납되는 카트리지 수납부;
    상기 샘플 전처리 모듈 내에 구비된 영구자석을 회전시키도록 자력을 생성하는 자력 생성부; 및
    상기 샘플 전처리 모듈의 관통막을 관통시키고, 캡의 이동부를 가압하여 상기 샘플을 배출시키는 관통 및 배출 실행부;를 포함하는 샘플 전처리 시스템.
  2. 제1항에 있어서,
    상기 모듈 홀더는,
    상기 모듈 홀더 외측을 둘러싸도록 구비되며, 상기 샘플 전처리 모듈을 가열하는 모듈 히터;를 포함하는 샘플 전처리 시스템.
  3. 제1항에 있어서,
    상기 관통 및 배출 실행부는,
    상기 샘플 전처리 모듈의 캡 테두리부를 가압하여 상기 샘플 전처리 모듈 내부의 관통막을 뚫는 테두리 가압부와,
    상기 이동부를 가압하여 상기 샘플을 배출시키는 이동부 가압부를 포함하는 샘플 전처리 시스템.
  4. 제3항에 있어서,
    상기 관통 및 배출 실행부는,
    상기 테두리 가압부가 연결되는 제1 이동바와, 상기 이동부 가압부와 연결되는 제2 이동바 및, 상기 제1 이동바와 제2 이동바를 구동시키는 제1 모터를 더 포함하며,
    상기 제1 이동바와 제2 이동바는 제1 모터의 구동에 따라 서로 멀어지는 방향 또는 서로 가까워지는 방향으로 동시에 이동하는 것을 특징으로 하는 샘플 전처리 시스템.
  5. 제4항에 있어서,
    상기 관통 및 배출 실행부는,
    상기 테두리 가압부 중앙부에 형성되는 통공을 더 포함하며,
    상기 이동부 가압부는 상기 통공을 통과하여 이동하면서 상기 이동부를 가압하는 것을 특징으로 하는 샘플 전처리 시스템.
  6. 제1항에 있어서,
    상기 자력 생성부는,
    상기 모듈 홀더 일측에 회전 가능하게 설치되는 볼텍싱(vortexing) 자석과,
    상기 볼텍싱 자석을 회전시키는 제2 모터를 포함하는 샘플 전처리 시스템.
  7. 제1항에 있어서,
    상기 카트리지 수납부는,
    상기 카트리지가 안착되며, 상기 카트리지를 로딩 또는 언로딩하는 카트리지 홀더와,
    상기 카트리지 홀더를 로딩 또는 언로딩 위치로 이동시키기 위한 구동력을 제공하는 제3 모터를 포함하는 샘플 전처리 시스템.
  8. 제7항에 있어서,
    상기 카드리지 수납부는 로딩된 상기 카트리지를 가열하는 카트리지 히터를 더 포함하며,
    상기 카트리지 히터는 상기 카트리지의 로딩 또는 언로딩에 따라 상승 또는 하강하는 것을 특징으로 하는 샘플 전처리 시스템.
  9. 제8항에 있어서,
    상기 카트리지 히터의 적어도 일부를 둘러싸고, 상기 카트리지에 밀착하는 히터 연결부를 더 포함하며,
    상기 히터 연결부는 상기 카트리지의 반응영역보다 더 큰 면적을 갖도록 구비되는 것을 특징으로 하는 샘플 전처리 시스템.
  10. 제9항에 있어서,
    상기 카트리지 히터는 상기 카트리지 반응영역을 커버하는 히터 연결부의 영역을 포함하는 면적을 갖도록 구비되는 것을 특징으로 하는 샘플 전처리 시스템.
  11. 제9항에 있어서,
    상기 히터 연결부는 상기 카트리지 반응영역에 밀착하도록 평평한 형상으로 이루어지는 것을 특징으로 하는 샘플 전처리 시스템.
  12. 제11항에 있어서,
    상기 카트리지 히터는 상기 카트리지와 상기 히터 연결부와의 접촉면에 상응하도록 평평한 형상으로 이루어지는 것을 특징으로 하는 샘플 전처리 시스템.
  13. 제1항에 있어서,
    상기 홀더 출납부는 상기 모듈 홀더를 이동하여 상기 모듈 홀더에 안착된 샘플 전처리 모듈을 로딩 또는 언로딩하는 구동력을 제공하는 제4 모터를 더 포함하는 샘플 전처리 시스템.
  14. 제1항에 있어서,
    상기 샘플이 배출되는 드랍(drop) 횟수를 감지하는 카운팅 센서를 더 포함하는 샘플 전처리 시스템.
  15. 카트리지를 카트리지 홀더에 안착시킨 후 카트리지 수납부로 로딩하는 단계;
    챔버 내에 샘플을 수용한 샘플 전처리 모듈을 모듈 홀더에 안착시킨 후 로딩하는 단계;
    상기 모듈 홀더 일측에 위치한 볼텍싱 자석을 회전시킴으로써 상기 챔버 내에 구비된 영구자석을 회전시키는 단계;
    상기 샘플 전처리 모듈의 캡 테두리부를 가압하여 상기 샘플 전처리 모듈 내부의 관통막을 뚫는 단계; 및,
    상기 샘플 전처리 모듈의 캡의 이동부를 가압하여 상기 샘플을 배출시킴으로써 상기 카트리지에 샘플을 로딩하는 단계;를 포함하는 샘플 전처리 시스템의 제어방법.
  16. 제15항에 있어서,
    상기 카트리지를 가열하여 일정 온도로 유지하는 단계를 더 포함하는 샘플 전처리 시스템의 제어방법.
  17. 제15항에 있어서,
    상기 모듈 홀더에 안착된 샘플 전처리 모듈을 가열하여 일정 온도로 일정 시간 동안 유지하는 단계를 더 포함하는 샘플 전처리 시스템의 제어방법.
