It was confirmed that more observed in the peripheral blood, it was confirmed that the lower fatty acids or salts thereof have an effect on stem cell migration ability.
- Colony-forming unit fibroblast (CFU-F) assay confirmed the enhancement of colony-forming ability of human bone marrow-derived mesenchymal stem cells (hMSC) according to lower fatty acid treatment.
- Colony forming unit fibroblast (CFU-F) analysis confirmed the increase in the cell population size of bone marrow-derived mesenchymal stem cells in mice following drinking water containing low fatty acids.
- Drinking water containing sodium acetate (200 mM) or drinking water without sodium acetate was ingested in C57BL / 6 mice for 7 days, and femurs were collected from these mice. After crushing the harvested femur with a mortar and pestle, it was treated with 0.2% collagenase (Type 1 collagenase, Worthington) to collect bone marrow cells (bone marrow cells) attached to the bone.
- the harvested cells were 2.0 ⁇ 10 5 cells / well (25T flask) in ⁇ -MEM (Sigma-Aldrich) containing 20% FBS, 1% penicillin and streptomycin (Invitrogen), 1% L-glutamine and 5% pyruvic acid. To a plate and incubated for 14 days under conditions of 5% CO 2 and 37 ° C.
- the plates were then fixed with 100% methanol, stained with a crystal violet staining solution (1% solution, Sigma-Aldrich) and the colonies counted under a stereo microscope.
- a crystal violet staining solution 1% solution, Sigma-Aldrich
- ST2 cells (3.8 ⁇ 10 5 cells) were added to 6-well plates under normal growth medium conditions. After overnight incubation, the medium was replaced with serum-free low-glucose DMEM medium without FBS and incubated for 18 hours. Cells were then treated with 10 mM sodium acetate (C2) or not (CON) for 48 hours.
- C2 10 mM sodium acetate
- RNA expression levels of VEGF and IL-10 in cells were measured using real-time polymerase chain reaction (Realtime PCR).
- the expression of IL-10 which exhibits anti-inflammatory immunosuppressive function according to sodium acetate treatment, was approximately 2.4-fold higher than that of the control group (CON) not treated with sodium acetate. It was confirmed that expression was increased about 2.5-fold. That is, it was confirmed that stem cells were activated by treatment with lower fatty acids or salts thereof to increase the synthesis of anti-inflammatory immunosuppressive and angiogenic signal-related factors.
