Definitions
- the present invention relates to a cognitive function improving agent and a method for improving cognitive function.
- MCI Mild Cognitive Impairment
- MCI Mild Cognitive Impairment
- MCI is a symptom in the middle stage (gray zone) between dementia and healthy people, and there is a problem with one of cognitive functions (memory, decision, reasoning, execution, etc.) It means that there is no obstacle to daily life. It is said that if MCI is left unattended, cognitive function continues to decline, and about half of the stages progress to dementia. Because dementia is almost impossible to cure at this time, it is extremely important to provide appropriate prevention at the MCI stage to prevent progression of symptoms.
- Alzheimer-type dementia There are four types of dementia: Alzheimer-type dementia, Lewy body dementia, vascular dementia, and frontotemporal dementia.
- Alzheimer's disease is the most common dementia, accounting for more than half of dementia in old age. Alzheimer's disease is a disease characterized by progressive dementia that occurs from the early age to the old age. Currently, the number of patients in Japan is said to be over 3 million. Clinical symptoms of Alzheimer's disease include memory impairment, higher brain dysfunction (aphasia, agnosia, aphasia, constitutive aphasia, etc.). Alzheimer's disease is thought to be caused by the accumulation of amyloid ⁇ protein and tau protein in the brain, which causes neuronal cell death.
- ATP adenosine triphosphate
- ATP production declines year after year, and it is said that when 40 years old is 100, it is about 60% at 80 years old.
- Decreasing ATP levels promotes cognitive decline (ie, predisposition to developing dementia), but four anti-dementia drugs increase acetylcholine in the brain but cannot increase ATP levels.
- a cerebral circulation / metabolism improving agent can be used to promote ATP production, it currently has a drawback that it can be used only after an organic disorder such as cerebral infarction.
- Non-patent Document 1-5 it has been reported that a ginkgo biloba extract combined with phosphatidylserine may be used as an active ingredient of food or medicine for improving dementia and Alzheimer's disease.
- Patent Document 1-5 it has been reported that supplements containing vitamin B, vitamin C, vitamin E, ⁇ -carotene, folic acid, etc. alone or in combination (two to three types) are not effective in improving cognitive function.
- the object of the present invention is to provide a cognitive function improving agent having an effect of improving cognitive function, and a method for improving cognitive function.
- ATP production cannot be increased.
- ATP is produced through seven steps of the TCA cycle, but vitamins play an important role as coenzymes in each reaction. In other words, adequate supplementation with vitamins may promote ATP production.
- the present inventor has found that vitamin A, vitamin B group (B 1 , B 2 , B 6 and B 12 ), vitamin C, vitamin D, vitamin E, zinc, magnesium
- the present inventors have found that the above object can be achieved by a supplement containing pantothenic acid, biotin, and folic acid.
- the present invention has been completed through further studies based on these findings, and provides the following cognitive function improving agents and the like.

- the cognitive function improving agent of the present invention is used for the purpose of improving cognitive function.
- cognitive function means a function in which a disorder is observed in dementia.
- cognitive functions include memory, attention, language functions, functions to perform a series of movements that have been acquired, functions to grasp the surrounding situation through the five senses, performance functions, and orientation.
- “Dementia” is a state in which intelligence once developed normally is irreversibly lowered due to an acquired organic disorder of the brain.
- “improvement” includes prevention of further deterioration of symptoms, that is, maintenance.
- the subject of application of the cognitive function improving agent of the present invention includes patients with dementia (particularly Alzheimer's disease), patients with MCI, patients with pre-MCI, etc. Elderly people who do not have any problems are also included in the target audience.
- the cognitive function improving agent of the present invention is applied to mammals including humans.
- the cognitive function-improving agent of the present invention can be expected to be effective even when used alone, but it effectively delays the progression of dementia when used in combination with an anti-dementia drug (donepezil, memantine, galantamine, rivastigmine, etc.) Functional improvement can be expected. Furthermore, the cognitive function-improving agent of the present invention can be expected to maintain MDL and ADL (daily life movement) of patients with dementia.
- the test for cognitive function improving agent of the present invention was conducted in 10 test examples .
- 7 were subjective forgetfulness and easy-fatigue pre-MCI patients, and 3 cases (No2, No3, No10) were subjective forgetfulness and easy-fatigue .
- the patient was also diagnosed with Pre-MCI, MCI, or dementia on 1 and imaging.
- Pre-MCI Pre-MCI
- MCI MCI
- dementia dementia
- capsules having the compositions shown in Table 1 below were administered to the subjects. * 1
- the MMSE test is widely used worldwide as a screening test for dementia. It consists of 11 questions and 30 points when all questions are answered correctly.
- each subject received a doctor's examination or interview before administration of the capsule and after administration for a certain period.
