미합중국특허 제5,145,684호에는 계면활성제의 존재 하에서 습식제분(wet milling)을 이용하여 난용성 약물을 분쇄하여 수백 나노미터 크기의 입자를 제조하는 방법이 개시되어 있다. 이 기술을 사용하기 위해서는, 약물을 전통적인 분쇄방법을 이용하여 100 마이크로미터 이하의 크기를 갖는 입자로 제조한 후 사용하여야 한다. 이 경우에 있어서, 입자의 크기를 목표로 하는 범위로 제조하기 위해서 소요되는 시간은 사용되는 기계적 장치에 따라 달라지는데, 볼 밀(ball mill)을 사용하면 5일 또는 그 이상의 시간이 소요되나, 고전단 매질 밀(high shear media mill)을 사용하면 1일 이내에 제조할 수 있다. 그러나 이 방법에서 얻어진 나노입자는 현탁액 제제이므로, 분말로 제조하기 위해서는 스프레이 드라이(spray dry) 또는 동결건조의 공정을 거쳐야 한다. 그러나, 이들 공정을 거치는 동안 입자들간의 상호 응집이 발생하기 때문에, 얻어진 분말을 다시 액체 내에 분산하였을 때 실질적인 나노입자로 분산되기가 어렵다. 이러한 문제를 해결하기 위해서, 미합중국특허 제5,302,401호에서는 동결건조시의 응집방지제에 대하여 기술하고 있다. 또한 미합중국특허 제 6,592,903호 B2에서는 안정제, 계면활성제 그리고 스프레이 드라이 시의 응집방지제에 대하여 기술하고 있다. 그리고 미합중국특허 2003/0185869호 A1호에서는 계면안정제로서 리소자임(lysozyme)을 사용하여 여러 난용성 약물에 대하여 습식제분(wet milling)법을 적용한 예를 기술하고 있다. 그러나 이 경우, 계면안정제가 단백질이므로, 건조에 많은 제약이 있으며, 따라서 액상 형태의 제조에 대하여만 기술하고 있다.
미합중국 공개특허 2002/0168402 A1호에서는 피스톤 갭 호모게니제이션을 이용한 나노입자 제조에 관한 방법을 기술하고 있다. 그러나 피스톤 갭 호모게니제이션을 이용하기 위해서는 제트밀 또는 햄머밀 등을 이용하여 입자를 일정한 크기로 분쇄하는 전처리 공정을 수행하여야만 한다. 또한 이 공정은 용액의 점도가 높으면 안되므로 활성물질의 농도가 낮은 상태에서 공정을 수행하여야 하는 문제가 있다.
또 다른 전통적인 방법으로는, 활성물질이 녹아 있는 용액의 환경을 변화시켜 용질을 침전 또는 결정화시킴으로써 활성물질의 미세입자를 만드는 재결정 방법이 있다. 이 재결정 방법에는 크게 두 가지 방식이 있는데, 그 하나는 치료제를 적당한 용매에 녹인 다음, 온도를 낮추어 치료제의 용해도를 변화시켜 입자를 석출시키는 방식이고, 다른 하나는 치료제가 녹아 있는 용매에 반용매(antisolvent)를 첨가하여 용매의 용해력을 낮춤으로써 입자를 석출시키는 방식이다. 그러나 이러한 재결정 방법은, 대부분 인체에 유해한 유기용매의 사용을 필요로 하게 되고, 또한, 석출된 입자를 여과한 후 젖은 상태에서 건조하는 동안 입자들끼리 서로 뭉쳐지는 현상이 자주 발생하여, 최종적으로 얻어진 입자의 크기가 일정하지 않을 수 있다.
미합중국특허 제 2003/0104068 A1호에서는 고분자를 유기용매에 녹이고 여기에 단백질 약물을 녹이거나 분산시킨 후, 초저온으로 급속 냉각하여 고형화시키고, 이를 동결건조하여 미세 분말을 제조하는 방법을 기술하고 있다. 그러나 이 경우 단백질 약물이 유기용매와의 접촉에 의해서 변성의 우려가 있으며, 또한 급속 냉각 및 동결건조의 공정을 거침으로써 공정의 경제성에도 많은 문제점이 있다.
또한, 유화를 이용하여 입자의 크기를 작게 하는 방법들이 있다. 이 방법은 화장품 분야에서 많이 사용되는 방법으로서, 난용성 물질을 열을 가하여 녹이거나 또는 유기용매를 이용하여 녹인 다음, 이를 계면활성제가 녹아있는 수용액에 첨가하면서 고속교반을 하거나, 초음파를 이용하여 분산함으로써 미세입자를 제조하는 방법이다. 그러나, 이 경우에, 미세 입자를 분말로 제조하기 위해서는 물을 제거하여야 하는데, 이 과정에서 다양한 제약이 따르게 된다. 또한, 유기용매를 사용하여 난용성 물질을 녹일 경우, 인체에 유해한 유기용매의 잔존에 대한 우려가 항상 존재한다.
미합중국특허 제2004/0067251 A1호는 활성물질을 유기용매에 녹이고, 이를 계면활성제가 녹아 있는 수용액에 분사함으로써 미세입자를 제조하는 방법을 개시하고 있다. 이 방법에서는 유기용매를 사용할 뿐만 아니라, 제조된 입자는 수용액상에 존재하기 때문에, 분말로 제조하기 위해서는 용매로 사용된 수분을 건조하여야 하는데, 수분을 건조하는 동안에 입자들간의 응집이 발생하여 재분산시에 입자들이 나노입자로 재분산되기 어렵다.
근래에 와서는, 초임계유체를 이용하여 비결정형질 또는 나노입자를 제조하고자 하는 시도들이 많이 있어 왔다. 초임계유체는 임계온도 및 임계압력보다 높은 온도 및 압력하에 존재하는 유체이며, 일반적으로는 이산화탄소가 많이 사용된다. 초임계유체를 이용하여 나노입자를 제조하고자 하는 방법 중의 하나로서 초임계 용액 급속팽창법(rapid expansion of supercritical solution, 이하 RESS)이 알려져 있다(Tom et al. (1991) Biotechnol . Prog . 7(5):403-411.; 미합중국특허 제6,316,030 B1호; 미합중국특허 제6,352,737 B1호). 이 방법에 따르면, 대상 용질을 초임계유체에 먼저 녹인 후, 그 초임계유체 용액을 비교적 낮은 압력환경으로 노즐을 통하여 급격하게 분사하게 된다. 그러면 초임계유체의 밀도가 급격하게 낮아지게 되고, 그 결과 용질에 대한 초임계유체의 용해력이 급속하게 떨어져서, 용질들이 매우 작은 입자 또는 결정으로 생성되게 된다.
초임계유체를 이용하는 다른 방법으로는 기체-반용매 재결정법(gas-antisolvent recrystallization, 이하 GAS)이 있다(Debenedetti et al. (1993) J. Control. Release 24:27-44; WO 00/37169). 이 방법에 따르면, 전통적인 유기용매에 치료약을 녹인 용액을 만들고, 이 용액을 반용매 역할을 하는 초임계유체 속으로 노즐을 통하여 분사하게 된다. 그러면 용액과 초임계유체가 서로 접촉하게 되어 급속한 부피확장이 발생하고, 그 결과 용매의 밀도 및 용해력이 떨어져서 초 과포화상태가 발생하게 되어, 용질이 핵 또는 입자를 형성하게 된다.
상기한 바와 같이 활성물질들을 나노입자화하기 위한 다양한 기술들이 개발되어 왔지만 그 적용 범위에 한계가 있었으며, 또한 경제성에도 많은 제약이 따르고 있다.
본 발명은 상기와 같은 종래 기술의 문제점을 해결하고자 한 것으로서, 밀링 공정을 이용하면서 종래 기술에서와 같은 적용의 한계 및 낮은 경제성을 극복하고, 나노입자 수준의 활성물질을 효율적으로 제조할 수 있는 방법을 제공하는 것이다.
본 발명에 따르면, (1) 활성물질, 윤활제로서 지질, 및 유리전이온도(glass transition temperature)가 80℃ 이상인 생체적합고분자를 포함하는 혼합물을 제공하는 단계; (2) 밀링 공정을 통하여 상기 (1)단계의 결과물을 분쇄하는 단계; 및 (3) 초임계유체를 사용하여 상기 (2)단계의 결과물로부터 지질을 제거하는 단계;를 포함하는, 활성물질 나노입자의 제조 방법이 제공된다.
본 발명의 일 구체예에서, 상기 (1)단계는 활성물질, 윤활제로서 지질, 및 유리전이온도가 80℃ 이상인 생체적합고분자를 물리적으로 균일하게 혼합하는 단계일 수 있다.
본 발명의 일 구체예에서, 상기 (1)단계는 활성물질과 윤활제로서 지질을 혼합하고, 이 혼합물에 유리전이온도가 80℃ 이상인 생체적합고분자를 탈염수와 함께 첨가한 후, 그 결과물을 물리적으로 균일하게 혼합하는 단계일 수 있다.

상기 (1)단계에 있어서 물리적 혼합은 v-믹서(v-mixer), 버티칼 믹서(vertical mixer), 리본 믹서(ribbon mixer), 플래너터리 믹서(planetary mixer), 롤 밀(roll mill) 등과 같이 통상적으로 사용되는 혼합기에 의하여 특별한 제한 없이 수행될 수 있다.
상기 (1)단계에서 성분들의 혼합시 온도에는 특별한 제한이 없으며, 예컨대, 40℃ 이하(예를 들어, 10~40℃), 바람직하게는 30℃ 이하(예를 들어, 10~30℃)에서 혼합이 수행될 수 있다.
본 발명의 구체예에서, 상기 (2)단계에서의 밀링 공정은 서로 역방향으로 회전하는 롤들(예컨대, 2롤 밀 또는 3롤 밀)을 사용하여 연속적으로 수행될 수 있다.
본 발명의 구체예에서, 상기 (3)단계에서의 지질 제거는 상기 (2)단계의 결과물이 담긴 반응기에 초임계유체를 연속적으로 투입 및 유출시킴으로써 수행될 수 있다.
본 발명의 구체예에서, 상기 (3)단계에서 초임계유체를 이용한 지질 제거는 50 기압 이상의 압력 조건 및 5~60 ℃의 온도 조건 하에서 수행될 수 있다.
본 발명에 따른 활성물질 나노입자 제조 방법은, 활성물질과 생체적합고분자, 및 임의로 계면활성제 및/또는 응집방지제를 포함하는 혼합물을 롤 밀(예컨대, 3롤 밀 또는 2롤 밀)을 사용하여 밀링할 때 윤활제로서 지질을 사용하여 밀링 공정이 원활하게 수행될 수 있도록 함으로써, 롤 밀을 이용한 상업적 스케일로 활성물질 나노입자의 생산을 가능하게 하며, 본 발명에 따라 제조된 활성물질 나노입자는 분산성, 흡수성, 생리적 활성 등이 매우 우수하여 의약품, 기능성 식품, 일반 식품, 화장품 등에 사용되기에 적합하다.

본 발명에 있어서, 상기 활성물질로는, 예를 들면, 의약품, 기능성 식품, 화장품 등에 있어서 생리활성을 나타내는 유기화합물, 유기금속 화합물, 천연 추출물, 단백질 및 이들의 조합으로부터 선택된 것이 사용될 수 있으나, 이에 한정되지는 않는다. 상기 활성물질에 있어서, 고체상 또는 액체상 등의 상온에서의 그 성상, 및 중성 또는 이온성 등의 그 전기적 형태에 대한 특별한 제한은 없다.

본 명세서에 있어서, 용어 "나노입자"란, 그 90% 이상이 5㎛ 이하, 바람직하게는 2㎛ 이하, 보다 바람직하게는 1㎛ 이하(예컨대, 0.9㎛ 이하, 0.8㎛ 이하, 0.7㎛ 이하, 또는 0.6㎛ 이하), 보다 더 바람직하게는 0.5㎛ 이하, 보다 더 바람직하게는 0.4㎛ 이하, 보다 더 바람직하게는 0.3㎛ 이하, 보다 더 바람직하게는 0.2㎛ 이하의 평균입경(mean size)을 가지는 입자를 의미한다. 상기 나노입자의 평균입경의 하한은 특별히 제한되지 않는다. 예컨대, 상기 나노입자의 평균입경은 1 nm 이상, 또는 5 nm 이상, 또는 10 nm 이상, 또는 50 nm 이상일 수 있으나, 이에 한정되지 않는다.
본 발명에 있어서, 윤활제로서 사용되는 상기 지질의 성상에는 특별한 제한이 없으며, 실온에서 액상 또는 고체상 등이 모두 사용 가능하다. 일 구체예에서, 바람직하게는 초임계유체에 대한 용해도가 우수한 지질이 사용될 수 있으며, 이러한 지질의 예로는, 탄소수 10~22의 포화 지방산, 탄소수 10~22의 포화 지방산 에스테르, 탄소수 10~22의 포화 지방 알코올, 탄소수 10~22의 포화지방산기를 갖는 모노-, 디- 또는 트리-글리세라이드, 탄소수 14 이상(예컨대, 14 내지 24)의 탄화수소 및 이들의 조합으로부터 선택된 것을 들 수 있으나, 이에 한정되는 것은 아니다.
본 발명의 특정 구체예에 따르면, 상기 지질은, 예컨대, 미리스틸 알코올(Myristyl alcohol), 세틸 알코올(Cetyl alcohol), 스테아릴 알코올(Stearyl alcohol) 및 라우릴 알코올(Lauryl Alcohol)과 같은 지방 알코올(fatty alcohol), 스테아릭 산(stearic acid), 팔미틱 산(palmitic acid), 미리스틱 산(myristic acid), 라우릭 산(lauric acid) 및 이들의 메틸, 에틸, 프로필 및 부틸 알코올 등의 에스테르, 스테아릴 모노글리세라이드, 팔미틸 모노글리세라이드, 미리스틸 모노글리세라이드, 라우릴 모노글리세라이드 등과 같이 지방산 모노글리세라이드, 디스테아릴 글리세라이드, 디팔미틸글리세라이드, 디미리스틸글리세라이드, 디라우릴글리세라이드 등과 같은 지방산 디글리세라이드. 테트라데칸(tetradecane), 펜타데칸(pentadecane), 헥사테칸(hexadecane), 헵타데칸(heptadecane), 옥타데칸(octadecane), 노나데칸(nonadecane), 아이코산(icosane), 헤네아이코산(heneicosane), 도코산(docosane), 트리코산(tricosane), 테트라코산(tetracosane)등과 같은 탄화수소 및 이들의 조합으로부터 선택될 수 있으나, 이에 한정되는 것은 결코 아니며, 본 발명의 방법에 따라 밀링 공정에서 윤활제로서 작용하며 초임계유체에 의하여 제거될 수 있는 어떠한 지질도 사용 가능하다.
본 발명에 있어서, 상기 지질은, 상기 활성물질 1 중량부에 대하여, 0.1 중량부 이상, 또는 0.3 중량부 이상, 또는 0.5 중량부 이상의 양으로 사용될 수 있다. 또한, 상기 지질은, 상기 활성물질 1 중량부에 대하여, 10 중량부 이하, 5 중량부 이하, 또는 3 중량부 이하, 또는 2 중량부 이하, 또는 1.5 중량부 이하의 양으로 사용될 수 있다. 상기 활성물질 대비 상기 지질의 사용량이 지나치게 적으면 후속 밀링 공정에 많은 부하가 걸려서 원활한 밀링을 수행하기 어렵거나, 활성물질이 무정형 또는 다른 결정형으로 변형되어 제조된 나노입자의 안정성이 나빠질 수 있다. 반대로, 상기 활성물질 대비 상기 지질의 사용량이 지나치게 많으면 후속 지질 제거 공정의 경제성이 현격히 낮아질 수 있다. 또한, 사용하는 지질이 상온에서 액상인 경우, 그 사용량은 활성물질 1 중량부에 대해서 1.5 중량부 이하일 수 있으며, 사용량이 너무 많으면 혼합물의 성상이 죽상으로 되어 밀링이 제대로 되지 않을 수 있다.
활성물질을 나노입자와 같은 미세입자로 제조하게 되면, 표면 에너지가 높아져 시간이 지남에 따라 큰 입자로 응집 또는 결정성장이 되는 경우가 빈번하게 발생한다. 이러한 응집 또는 결정성장을 방지하기 위해서는, 나노입자화 공정에 사용되는 생체적합고분자의 유리전이온도가 매우 중요한 요소가 된다. 나노입자 제조공정에 사용된 생체적합고분자의 유리전이온도가 80℃ 미만으로 낮으면, 제조된 나노입자의 응집 및/또는 결정성장을 충분히 방지하지 못하여, 시간이 지남에 따라 제조된 입자의 크기가 커지게 되는 문제가 발생하게 된다. 이러한 제조된 나노입자의 응집 및/또는 결정성장을 방지하기 위해서, 본 발명에서는 유리전이온도가 80℃ 이상(바람직하게는 90℃ 이상)인 생체적합고분자를 필수적으로 사용한다.
본 발명에서 사용 가능한, 유리전이온도(Tg)가 80℃ 이상(바람직하게는 90℃ 이상)인 생체적합고분자는, 예컨대, 메틸셀룰로스(methylcellulose(MC), 예컨대, Tg가 약 184~197℃인 MC), 히드록시에틸셀룰로스(hydroxyethylcellulose(HEC), 예컨대, Tg가 약 127℃인 HEC), 히드록시프로필셀룰로스(hydroxypropylcellulose(HPC), 예컨대, Tg가 약 105℃인 HPC), 히드록시프로필 메틸셀룰로스(hydroxypropyl methylcellulose(HPMC), 예컨대, Tg가 약 180℃인 HPMC), 히드록시프로필 메틸셀룰로스 아세테이트 숙시네이트(hydroxypropyl methylcellulose acetate succinate(HPMC-AS), 예컨대, Tg가 약 117℃인 HPMC-AS), 히드록시프로필 메틸셀룰로스 프탈레이트(hydroxypropyl methylcellulose phthalate(HPMC-P), 예컨대, Tg가 약 145℃인 HPMC-P), 카복시메틸 셀룰로스(carboxymethyl cellulose(CMC), 예컨대, Tg가 약 135℃인 CMC), 셀룰로스 아세테이트(cellulose acetate(CA), 예컨대, Tg가 약 180℃인 CA), 덱스트란(dextran, 예컨대, Tg가 약 200℃인 덱스트란)등과 같은, Tg가 80℃ 이상인 셀룰로스 계열의 생체적합고분자, Tg가 80℃ 이상인 폴리비닐피롤리돈(polyvinyl pyrrolidone(PVP), 예컨대, Tg가 약 138~156℃인 PVP k17, k30, k90), Tg가 80℃ 이상인 폴리비닐알코올(polyvinyl alcohol), 및 Tg가 80℃ 이상인 유드라짓(Eudragit) 계열의 생체적합고분자로부터 선택되는 1종 이상일 수 있다.
본 발명에서 필수적으로 사용되는 상기 Tg가 80℃ 이상인 생체적합고분자는 단독으로 또는 2종 이상이 혼합되어 사용될 수 있으며, 그 사용량은 활성물질 1 중량부 대비 0.01 중량부 이상인 것이 바람직하다. 보다 구체적으로, 상기 Tg가 80℃ 이상인 생체적합고분자는, 활성물질 1 중량부에 대하여, 0.05 중량부 이상, 0.1 중량부 이상, 0.15 중량부 이상, 0.2 중량부 이상, 0.25 중량부 이상 또는 0.3 중량부 이상의 양으로 사용될 수 있으며, 또한, 활성물질 1 중량부에 대하여, 5 중량부 이하, 3 중량부 이하, 2 중량부 이하, 1 중량부 이하, 또는 0.5 중량부 이하의 양으로 사용될 수 있다.
본 발명에서 임의로 사용 가능한, 유리전이온도가 80℃ 미만인 생체적합고분자, 계면활성제 및 응집방지제로부터 선택되는 하나 이상의 추가 성분은 의약품, 식품 및 화장품에 사용되는 것으로, 그 전기적 성질(예컨대, 이온성, 비이온성 등)에 특별한 제한이 없고, 또한 상온에서의 그 성상(예컨대, 액상, 왁스 또는 고체상 등)에도 특별한 제한이 없으며, 단독으로 또는 2종 이상이 혼합되어 사용될 수 있다.
본 발명에 있어서 임의로 사용 가능한 추가 성분에는 특별한 제한이 없으며, 활성물질의 나노입자화를 위해 사용 가능한 것으로 알려진 공지의 유리전이온도가 80℃ 미만인 생체적합고분자, 계면활성제, 응집방지제, 또는 신규한 것이라 하더라도 활성물질의 나노입자화를 위해 사용 가능한 것이라면 무엇이라도 본 발명에 적용될 수 있다. 이러한 임의의 추가 성분의 예로는, 젤라틴(gelatin), 카제인(casein), 아라비아고무(gum acacia), 트래거캔스 고무(tragacanth), 폴리에틸렌 글리콜(polyethylene glycols), 플록사머(poloxamers), 유리전이온도가 80℃ 미만인 유드라짓(eudragit ®), 라이소자임(lysozyme), 알부민(albumin) 등의 유리전이온도가 80℃ 미만인 생체적합고분자; 세틸 피리디움 클로라이드(cetyl pyridinium chloride), 인지질(phospholipids), 지방산(fatty acid), 벤잘코니움 클로라이드 (benzalkonium chloride), 칼슘 스테아레이트(calcium stearate), 글리세린 지방산 에스터(glycerin esters of fatty acid), 지방산 알코올(fatty alcohol), 세토마크로골(cetomacrogol), 폴리옥시에틸렌 알킬 에테르(polyoxyethylene alkyl ethers), 소르비탄 에스테르(sorbitan esters), 폴리옥시에틸렌 캐스터 오일 유도체(polyoxyethylene castor oil derivatives), 폴리옥시에틸렌 소르비탄 지방산 에스테르(polyoxyethylene sorbitan fatty acid esters), 도데실 트리메틸 암모니움 브로마이드(dodecyl trimethyl ammonium bromide), 폴리옥시에틸렌 스테아레이트(polyoxyethylene stearate), 소디움 라우릴 설페이트(sodium lauryl sulfate), 자당지방산 에스테르(sucrose fatty acid ester), PEG-콜레스테롤(PEG-cholesterol), PEG-비타민 E(PEG-vitamin E) 등의 계면활성제; 당류 등의 응집방지제를 들 수 있다. 본 발명에서 당류란, 단당류 화합물, 이당류 화합물, 다당류 화합물 및 당 알코올 등을 포함하며, 특히 포도당, 락토스, 만니톨, 슈크로즈, 자일리톨, 키토산, 녹말 섬유질 등을 포함하는 개념으로, 단독 또는 혼합물의 형태로 사용 가능하다. 이들 임의의 추가 성분은 단독으로 또는 2종 이상이 혼합되어 사용될 수 있다. 그러나 상기한 구체적인 예들로 제한되는 것은 결코 아니다.
본 발명에 있어서, 상기 임의의 추가 성분이 하나 이상 사용되는 경우, 이들 각각은, 예컨대, 활성물질 1 중량부에 대하여, 0.001 중량부 이상, 또는 0.005 중량부 이상, 또는 0.01 중량부 이상의 양으로 사용될 수 있고, 또한, 활성물질 1 중량부에 대하여, 5 중량부 이하, 또는 3 중량부 이하, 또는 2 중량부 이하, 또는 1 중량부 이하, 또는 0.5 중량부 이하의 양으로 사용될 수 있다.
본 발명에 있어서 사용 가능한 수혼화성 유기용매는, 활성물질 및 윤활제로 사용되는 지질을 잘 녹일 수 있고, 물과 잘 섞일 수 있어 물속에서 활성물질과 윤활제로 사용된 지질이 균질하게 혼합된 혼합물을 잘 제조할 수 있는 용매라면 특별한 제한이 없다.
상기 수혼화성 유기용매의 구체적인 예로는 탄소수가 8 이하(예컨대, 1 내지 8)인 모노 또는 다가 알코올, 아민, 아세톤과 같은 케톤, 디메틸 설폭사이드(dimethyl sulfoxide, DMSO), 디메틸포름아미드(dimethylformamide, DMF), 테트라하이드로퓨란(tetrahydrofurane, THF), 피리딘(Pyridine) 및 이들의 조합으로부터 선택된 것을 들 수 있으나, 이에 한정되지는 않는다.
본 발명에 있어서, 상기 수혼화성 유기용매가 사용되는 경우, 이는, 상기 활성물질 1 중량부에 대하여, 0.1 중량부 이상, 또는 0.5 중량부 이상, 또는 1 중량부 이상의 양으로 사용될 수 있다. 또한, 상기 수혼화성 유기용매는, 상기 활성물질 1 중량부에 대하여, 10 중량부 이하, 또는 6 중량부 이하, 또는 3 중량부 이하의 양으로 사용될 수 있다. 상기 활성물질 대비 상기 수혼화성 유기용매의 사용량이 지나치게 적으면 활성물질 및 윤활제인 지질을 저온(예컨대, 100℃ 이하)에서 녹이기 어려운 문제가 있을 수 있다. 반대로, 상기 활성물질 대비 상기 수혼화성 유기용매의 사용량이 지나치게 많으면 물에서 고형화할 때 활성물질의 상당량이 고형화되지 못하여 생산 수율이 낮아지는 문제가 있을 수 있다.
본 발명의 나노입자 제조방법에 있어서, 상기 (1) 단계에서는, 활성물질, 윤활제로서 지질 및 유리전이온도가 80℃ 이상인 생체적합고분자를 포함하는 혼합물이 제공된다.
일 구체예에서, 상기 (1)단계는 활성물질, 윤활제로서 지질 및 유리전이온도가 80℃ 이상인 생체적합고분자, 및 임의로 유리전이온도가 80℃ 미만인 생체적합고분자, 계면활성제 및 응집방지제로부터 선택되는 하나 이상의 추가 성분을 물리적으로 균일하게 혼합하는 단계일 수 있다.
일 구체예에서, 상기 (1)단계는 활성물질과 윤활제로서 지질을 혼합하고, 이 혼합물에 유리전이온도가 80℃ 이상인 생체적합고분자를 탈염수와 함께 첨가한 후, 그 결과물을 물리적으로 균일하게 혼합하는 단계일 수 있다.
일 구체예에서, 상기 (1)단계는 활성물질과 윤활제로서 지질을 혼합하고, 이 혼합물에 유리전이온도가 80℃ 이상인 생체적합고분자, 및 유리전이온도가 80℃ 미만인 생체적합고분자, 계면활성제 및 응집방지제로부터 선택되는 하나 이상의 추가 성분을 탈염수와 함께 첨가한 후, 그 결과물을 물리적으로 균일하게 혼합하는 단계일 수 있다.
본 발명의 일 구체예에서, 상기 (1)단계는 활성물질 및 윤활제로서 지질을 수혼화성(물과 섞이는 성질) 유기용매에 용해시켜 얻어진 용액을 물에 부어 고형화시킨 후, 이를 여과하고 건조하여 얻어진 혼합물에 유리전이온도가 80℃ 이상인 생체적합고분자를 첨가하고, 그 결과물을 물리적으로 균일하게 혼합하는 단계일 수 있다.
본 발명의 일 구체예에서, 상기 (1)단계는 활성물질 및 윤활제로서 지질을 수혼화성 유기용매에 용해시켜 얻어진 용액을 물에 부어 고형화시킨 후, 이를 여과하고 건조하여 얻어진 혼합물에 유리전이온도가 80℃ 이상인 생체적합고분자를 탈염수와 함께 첨가하고, 그 결과물을 물리적으로 균일하게 혼합하는 단계일 수 있다.

일 구체예에서, 상기 (1)단계는 활성물질, 윤활제로서 지질 및 유리전이온도가 80℃ 이상인 생체적합고분자를 수혼화성 유기용매에 용해시켜 얻어진 용액을 물에 부어 고형화시킨 후, 이를 여과하고 건조하여 얻어진 혼합물에 유리전이온도가 80℃ 미만인 생체적합고분자, 계면활성제 및 응집방지제로부터 선택되는 하나 이상의 추가 성분을 탈염수와 함께 첨가하고, 그 결과물을 물리적으로 균일하게 혼합하는 단계일 수 있다.
활성물질의 결정성을 유지해야 하거나, 활성물질이 열에 민감할 경우, 각 성분들을 분말 상태로 혼합기에 투입하고, 균일하게 혼합할 수 있다. 활성물질과 지질을 더욱 균일하게 혼합할 필요가 있을 경우, 활성물질과 지질을 반응기에 넣고, 여기에 수혼화성 유기용매를 첨가한 뒤 가열하여 혼합물들을 투명하게 녹인 후, 이 용액을 윤활제로서 사용된 지질의 녹는점보다 낮은 온도의 물, 예컨대, 40℃ 이하, 바람직하게는 30℃이하, 보다 바람직하게는 25℃, 보다 더 바람직하게는 20℃ 이하의 탈염된 증류수에 부어서 고형화하고, 얻어진 고형물을 여과하고 감압 건조하여 성분들이 균일하게 혼합된 혼합 분말을 수득할 수 있다.
상기 생체적합고분자, 계면활성제 및/또는 응집방지제는 경우에 따라서 분말 형태로 첨가되거나, 탈염수와 함께, 또는 수용액 형태로 첨가될 수도 있다.
상기 (1)단계에서 유리전이온도가 80℃이상인 생체적합고분자, 및/또는 임의의 추가 성분을 탈염수와 함께 첨가하여 물리적으로 균일하게 혼합하고자 하는 경우 사용되는 물의 양은 특별한 제한이 있는 것은 아니나, 공정의 원활성과 경제성을 위하여, 물을 포함한 혼합물의 총 무게 대비 40%(w/w) 이하(보다 구체적으로는, 35%(w/w) 이하, 30%(w/w) 이하, 25%(w/w) 이하, 20%(w/w) 이하 또는 15%(w/w) 이하)인 양의 물을 사용하는 것이 바람직하다. (2)단계인 밀링 공정에서 효과적인 분쇄가 이루어지기 위해서는 혼합물에 적당한 전단력이 전달되어야 하는데, 이 전단력은 혼합물 내의 수분 함량과 밀접한 관계가 있다. 수분의 함량이 높으면 전단력이 매우 낮아 효과적인 분쇄가 되지 않으며, 효과적인 분쇄를 가능하게 하는 전단력이 전달될 수 있는 수준까지 수분 함량이 감소하는데 많은 시간이 소요됨으로써 경제성이 나빠지게 된다.
본 발명의 나노입자 제조방법에 있어서, 상기 (2) 단계에서는, 상기 (1)단계의 결과물(즉, 활성물질, 윤활제인 지질 및 유리전이온도가 80℃ 이상인 생체적합고분자, 그리고 필요에 따라 임의의 추가 성분을 포함하는 혼합물)을 밀링 공정을 통하여 분쇄한다. 상기 밀링 공정은, 예컨대, 2 롤 밀 또는 3 롤 밀을 사용하여 연속적으로 수행될 수 있다.
롤 밀은 두 개 이상의 롤(예컨대, 2롤 밀 또는 3롤 밀 등)이 서로 역방향으로 회전하면서, 투입되는 혼합물에 압착 및 전단력을 가하여 이를 분쇄하는 장비로, 본 발명에서는 상기 (1)단계의 결과물에 압착 및 전단력을 가하여 분쇄할 수 있는 롤 밀이라면 어떠한 형태라도 사용 가능하다.
밀링 공정 동안 롤러간 간극은, 1 mm 이하, 예컨대, 500 ㎛이하, 200 ㎛ 이하, 바람직하게는 100 ㎛ 이하, 더욱 바람직하게는 50 ㎛ 이하, 더욱 더 바람직하게는 30㎛ 이하일 수 있다. 롤러간 간극이 너무 넓으면 압착 및 전단력이 낮아져서 충분한 분쇄효과를 얻기 어렵고, 간극이 너무 좁으면 투입속도가 느려지는 등의 문제가 발생할 수 있다. 이러한 간극 조절은 당 분야에 종사하는 사람이라면 생산 현장에서 쉽게 적절히 조절할 수 있을 것이다.
밀링 공정에서 롤의 온도는 사용되는 윤활제, 즉, 지질의 녹는점과 사용하는 양 및 생체적합고분자 등의 첨가시에 사용되는 물의 양에 따라 적절히 조정될 수 있다. 일 구체예에서, 사용되는 지질의 양이 활성물질 1 중량부에 대해 1 중량부 이상일 경우에는, 롤의 온도를 사용하는 지질의 녹는점 보다 낮게 유지하는 것이 바람직할 수 있다. 다른 구체예에서, 사용하는 지질의 양이 활성물질 1 중량부에 대해 1 중량부 미만일 경우에는, 롤의 온도를 사용하는 지질의 녹는점 부근(녹는점±5℃) 또는 그 이상으로 할 수도 있다. 다르게는, 예컨대, 라우릴 알코올(녹는점: 22℃)과 같이 상온에서 액상인 지질을 사용할 경우에는 롤의 온도보다는 첨가되는 지질의 양을 조절(예컨대, 활성물질 1 중량부에 대하여 0.1~1 중량부, 또는 0.2~0.8 중량부, 또는 0.3~0.5중량부로 조절)하는 것이 밀링 공정에 유리할 수도 있다.
유리전이온도가 80℃ 이상인 생체적합고분자 및/또는 임의의 추가 성분을 단순 첨가하거나, 또는 탈염수와 함께 첨가하더라도 밀링 공정 중 수분의 증발을 촉진시켜 1%(w/w) 이하의 수분 조건에서 밀링을 하고자 할 때는, 윤활제로 사용되는 지질의 녹는점 또는 그보다 낮은 온도 범위(예컨대, 지질의 녹는점 보다 0 내지 20℃ 낮은 온도 범위)에서 활성물질이 충분히 나노입자가 될 때까지 밀링 공정을 수행한다. 이때 지질이 없으면 활성물질과 생체적합고분자의 혼합물에 너무 높은 전단력이 발생하여 밀링 공정의 수행이 어려워 상업적 생산이 불가능하다.
유리전이온도가 80℃ 이상인 생체적합고분자 및/또는 임의의 추가 성분을, 탈염수와 함께 활성물질과 지질의 혼합물에 첨가하고, 수분의 감소에 따른 전단력 증가를 이용하여 분쇄를 하고자 할 경우, 롤은 35℃ 이하, 바람직하게는 25℃ 이하, 더욱 바람직하게는 20℃ 이하 더더욱 바람직하게는 17℃ 이하에서 수행하는 것이 나노입자 제조에 유리하다. 밀링 공정을 반복적으로 수행하면 혼합물 내의 수분의 함량은 점점 낮아지게 되어, 수분의 함량이 20%(w/w) 이하, 바람직하게는 15%(w/w) 이하, 더욱 바람직하게는 12%(w/w) 이하, 더 더욱 바람직하게는 10%(w/w) 이하로 되면 밀링 공정에서 전단력이 급격히 증가되어 활성물질의 나노화가 용이하게 된다. 한편 혼합물 내에 존재하는 유리전이온도가 80℃ 이상인 생체적합고분자의 유리전이온도는 혼합물 내의 수분의 함량이 낮아질수록 급격히 증가하게 되어, 윤활제로 사용되는 지질이 없으면 밀링 공정 중, 롤에 많은 부하가 발생하여 경제적인 생산이 매우 어렵게 된다.
본 발명의 나노입자 제조방법에 있어서, 상기 (3) 단계에서는, 초임계유체를 사용하여 상기 (2)단계의 결과물로부터 윤활제로서 사용된 지질을 제거한다. 일 구체예에서, 상기 지질 제거는 (2)단계의 결과물이 담긴 반응기에 초임계유체를 연속적으로 투입 및 유출시킴으로써 수행될 수 있으며, 이는, 예컨대, 50 기압 이상의 압력 조건 및 5~60 ℃의 온도 조건 하에서 수행될 수 있다.
본 명세서에 있어서, 용어 "초임계유체"란, 이산화탄소 또는 질소와 같이, 반응성이 없는 불활성이면서, 특정온도와 특정압력, 즉, 초임계온도와 초임계압력 하에서는 초임계유체(supercritical fluid)가 될 수 있는 기체 또는 액체를 말한다.
또한, 본 명세서에 있어서, 용어 "초임계압력"이란, 그 압력 이상의 압력 하에서는 초임계유체 가스가 초임계유체로 될 수 있는 특정압력을 말한다.
일 구체예에서, 상기 (2) 단계에서 얻어진 고형 혼합물을 고압 반응기에 넣고, 윤활제로 사용한 지질이 흘러 내리지 않을 온도, 예컨대, 5~60℃, 보다 구체적으로는 10~40℃로 반응기 내부를 유지하면서, 반응기에 초임계유체 가스(예컨대, 이산화탄소)를 투입하여, 예컨대, 50~400 기압, 바람직하게는 60~200 기압으로 반응기 내부를 가압한 다음, 초임계유체 투입밸브와 배출밸브를 조절하면서 초임계유체를 연속적으로 반응기 내부로 투입 및 반응기 외부로 유출시킴으로써, 윤활제로 사용된 지질을 초임계유체와 함께 반응기 외부로 내보내어 제거하게 된다. 이 때, 고압반응기 내부의 온도가 너무 높으면, 윤활제로 사용된 지질이 활성물질에 대하여 용매로 작용하게 될 수 있고, 그 결과, 나노입자 크기로 제조되었던 활성물질의 결정이 성장하게 될 수 있다. 따라서, 반응기의 온도는, (2) 단계에서 얻어진 고형 혼합물 내에 존재하는 지질의 유동성을 최대한 줄일 수 있는 범위, 바람직하게는 지질의 녹는점 이하의 온도로 유지하는 것이 바람직하며, 작업성을 고려할 경우, 10~40℃로 유지하는 것이 바람직하다.
또한, 지질을 초임계유체로 제거하는데 소요되는 시간은 사용된 지질의 종류 및 양에 좌우되며, 보다 높은 순도의 활성물질 입자를 얻기 위해서는, 가능한 충분한 시간 동안 지질을 제거하여 잔존 지질의 양을 최소화시키는 것이 바람직하다. 본 발명에서 바람직하게 사용되는 지질들은 인체에 무해하므로, 그 잔존량을 특정범위로 한정할 필요는 없지만, 얻어지는 활성물질의 순도를 고려해 볼 때, 전체 무게의 1중량% 미만으로 잔존하도록 하는 것이 바람직하다. 물론, 모노-, 디- 또는 트리-글리세라이드 계통의 화합물과 같이 계면활성제로도 사용되는 지질을 사용한 경우에는, 그 잔존량이 10중량%를 넘어도 무방하다고 할 수 있다.
상기와 같이 하여 고형 혼합물 분말로부터 제거된 지질은 별도의 반응기에 수거하여, 다음 생산 공정에서 계속 사용할 수 있다.
이하 실시예 및 비교예에 의해 본 발명을 상세히 설명하나, 이에 의해 본 발명이 한정되는 것은 아니다.
[ 실시예 ]
실시예 1~5
활성물질로서 닐로티닙(Nilotinib; free base) 1 g을 윤활제로서 하기 표 1에 나타낸 지질 2 g과 함께 비이커에 넣고, 주걱(spatula)으로 잘 혼합한 후, 여기에 폴리비닐피롤리돈(polyvinylpyrrolidone, PVP k30) 0.3 g 및 소듐 라우릴 설페이트(sodium lauryl sulfate, SLS) 0.05 g을 첨가하고 다시 잘 혼합한 후, 그 결과물에 대하여 3롤 밀(TRX-3100S; Intec system)을 이용하여 하기 표 1에 나타낸 조건으로 연속 밀링을 수행하여, 닐로티닙과 윤활제인 지질, 그리고 PVP k30 및 SLS가 균일하게 혼합된 고체 분산체형 혼합물을 수득하였다.
상기 수득된 고체분산체형 혼합물을 고압반응기에 넣고, 15~25℃, 70~100 기압에서 초임계유체인 이산화탄소를 연속적으로 흘려 보내어 윤활제로 사용된 지질을 제거하고, 닐로티닙, PVP k30 및 SLS가 혼합된 분말을 수득하였다.
수득된 분말을 1 mg/ml의 농도(닐로티닙 기준)로 탈염된 증류수에 분산한 후, DLS(Dynamic light scattering)방식의 오츠카 ELSZ-1000으로 입도를 측정하였다. 실시예 1~5에서 제조된 나노입자의 입도분포도들을 도 1에 나타내었다.

실시예 1 내지 3의 경우, 각 지질의 녹는점보다 10℃(±5℃)정도 낮은 롤러 온도(R.T)를 유지하였으며, 밀링이 원활히 되었고, 밀링 동안 롤에 많은 부하가 걸리지 않았다. 실시예 4 및 5의 경우, 지질의 녹는점 대비 롤러 온도가 실시예 1 내지 3 보다는 더 낮은 조건에서 밀링을 수행하였으며, 롤에 어느 정도 부하가 걸렸지만 나노입자는 잘 제조되었다.
실시예 6~7
활성물질로서 닐로티닙(free base) 1 g을 윤활제로서 라우릴 알코올(Lauryl Alcohol, LA) 0.3 g, 및 0.5 g과 각각 잘 혼합하고, 여기에 PVP k30 0.3 g, SLS 0.05 g을 첨가하여 다시 잘 혼합한 후, 그 결과물에 대하여 3롤 밀(TRX-3100S; Intec system)을 이용하여 하기 표 2에 나타낸 조건으로 연속 밀링을 수행하여, 닐로티닙과 윤활제인 지질, 그리고 PVP k30 및 SLS가 균일하게 혼합된 고체 분산체형 혼합물을 수득하였다.
상기 수득된 고체분산체형 혼합물을 고압반응기에 넣고, 15~25℃, 70~100 기압에서 초임계유체인 이산화탄소를 연속적으로 흘려 보내어 윤활제로 사용된 지질을 제거하고, 닐로티닙, PVP k30 및 SLS가 혼합된 분말을 수득하였다.
수득된 분말을 1 mg/ml의 농도(닐로티닙 기준)로 탈염된 증류수에 분산한 후, DLS(Dynamic light scattering)방식의 오츠카 ELSZ-1000으로 입도를 측정하였다. 실시예 6 및 7에서 제조된 나노입자의 입도분포도들을 도 2에 나타내었다.

실시예 8
활성물질로서 엘로티닙(Erlotinib; free base) 10 g과 윤활제로서 MA 20 g, 그리고 수혼화성 용매로서 DMSO 20 g을 비이커에 넣고 40℃로 가열하여 완전히 녹인 후, 10℃의 물 500 ml에 부어서 엘로티닙과 MA가 잘 혼합된 고체분산체를 제조하고, 이를 여과한 후, 감압 건조하였다. 이 건조된 고형 혼합물 30 g에, 히드록시프로필 셀룰로스(Hydroxypropyl cellulose, HPC; ssL) 3 g과 슈크로스 스테아레이트(Sucrose stearate, s-1670) 1 g을 탈염된 증류수 15 ml와 함께 첨가하고, 롤 밀을 수회 수행하여 잘 혼합한 후, 이 혼합물을 3롤 밀을 이용하여, 롤러 속도(R.S) 60 rpm, 롤러 온도(R.T) 25~30℃에서 40 회 연속 밀링을 진행하였다. 밀링 공정 중 혼합에 사용된 물은 모두 증발하여 잔존 수분의 함량은 1%(w/w) 이하가 되었다.
상기 수득된 고체분산체형 혼합물을 고압반응기에 넣고, 15~25℃, 70~100 기압에서 초임계유체인 이산화탄소를 연속적으로 흘려 보내어 윤활제로 사용된 지질을 제거하고, 엘로티닙, HPC ssL 및 슈크로스 스테아레이트가 혼합된 분말을 수득하였다.
수득된 분말을 1 mg/ml의 농도(엘로티닙 기준)로 탈염된 증류수에 분산한 후, DLS(Dynamic light scattering)방식의 오츠카 ELSZ-1000으로 입도를 측정하여, 평균입자크기가 341.7 ㎚임을 확인하였다. 실시예 8에서 제조된 나노입자의 입도분포도를 도 3에 나타내었다.
실시예 9
별도의 실험을 통하여, 소라페닙 토실레이트 III형(Sorafenib tosylate III)을 용매에 녹여 다시 결정화할 경우, 결정형의 변화 및 일부 토실레이트 염이 떨어진 자유 염기화합물이 생성될 우려가 있음을 확인하였다.
따라서 소라페닙 토실레이트 III형 분말 3 g을 윤활제인 MA 분말 9 g과 잘 혼합한 후, 여기에 히드록시프로필 메틸셀룰로스(Hydroxypropyl methylcellulose, HPMC; 5cp) 0.75 g, PVP(k30) 0.3 g 및 폴록사머(Poloxamer, 407) 0.09 g을 탈염 증류수 2.25 ml와 함께 첨가하고 롤 밀을 수회 수행하여 잘 혼합한 후, 이 혼합물을 3롤 밀을 이용하여, 롤러 속도(R.S) 60 rpm, 롤러 온도(R.T) 15~19℃에서 8 회 연속 밀링을 진행하였다. 이때 잔존 수분의 함량은 9%(w/w)이었으며(Karl Fischer 법으로 측정), 이를 상온(25℃)에서 감압 건조하였다. 수득된 고체분산체형 혼합물을 고압반응기에 넣고, 15~25℃, 70~100 기압에서 초임계유체인 이산화탄소를 연속적으로 흘려 보내어 윤활제로 사용된 지질을 제거하고, 소라페닙, HPMC, PVP 및 폴록사머가 혼합된 분말을 수득하였다.
수득된 분말을 1 mg/ml의 농도(소라페닙 기준)로 탈염된 증류수에 분산한 후, DLS(Dynamic light scattering)방식의 오츠카 ELSZ-1000으로 입도를 측정하여, 평균입자크기가 496.8 ㎚임을 확인하였다.
실시예 10
소라페닙 토실레이트 III형 분말 3 g을 윤활제인 MA 분말 4.5 g과 잘 혼합한 후, 여기에 HPC (ssL) 0.3 g, 폴리옥시에틸렌 40 스테아레이트(Polyoxyethylene 40 stearate, PS) 0.3 g, PVP(k30) 0.3 g, 슈크로스 스테아레이트(SS; s-1670) 0.3 g 및 폴록사머(407) 0.9 g을 탈염 증류수 4.5 ml와 함께 첨가하고 잘 혼합한 후, 이 혼합물을 3롤 밀을 이용하여, 롤러 속도(R.S) 60 rpm, 롤러 온도(R.T) 15~19℃에서 40 회 연속 밀링을 진행하였다. 이때 잔존 수분의 함량은 5%(w/w)이었으며(Karl Fischer 법으로 측정), 이를 상온(25℃)에서 감압 건조하였다. 수득된 고체분산체형 혼합물을 고압반응기에 넣고, 15~25℃, 70~100 기압에서 초임계유체인 이산화탄소를 연속적으로 흘려 보내어 윤활제로 사용된 지질을 제거하고, 소라페닙, HPC, PVP, PS, SS 및 폴록사머가 혼합된 분말을 수득하였다.
수득된 분말을 1 mg/ml의 농도(소라페닙 기준)로 탈염된 증류수에 분산한 후, DLS(Dynamic light scattering)방식의 오츠카 ELSZ-1000으로 입도를 측정하여, 평균입자크기가 446.0 ㎚임을 확인하였다. 또한 pXRD 분석을 통하여 소라페닙 토실레이트 III형의 결정형이 유지됨을 확인하였다. 실시예 10에서 제조된 나노입자의 pXRD 분석 결과를 도 4에 나타내었다.
비교예
윤활제인 MA를 첨가하지 않은 것을 제외한 모든 부형제의 첨가비율 및 밀링의 공정조건은 실시예 11과 동일하였다. 밀링 공정에서는 윤활제인 지질이 첨가되지 않아 매우 많은 부하가 걸려 많은 시간이 소요되었다. 또한 윤활제를 첨가하지 않았으므로 이산화탄소를 이용한 윤활제를 제거하기 위한 공정을 수행하지 않았다. 밀링만 수행한 후 감압 건조하여 얻어진, 소라페닙, HPC, PVP, PS, SS 및 폴록사머가 혼합된 분말을 1 mg/ml의 농도(소라페닙 기준)로 탈염된 증류수에 분산한 후 DLS(Dynamic light scattering)방식의 오츠카 ELSZ-1000으로 입도를 측정하여, 평균입자크기가 786.0 ㎚임을 확인하였다. pXRD 분석을 통하여 소라페닙 토실레이트의 결정형을 분석해 본 결과, 결정형이 I형으로 변형되었음을 확인하였다. 실시예 10과 비교예에서 각각 제조된 나노입자의 pXRD 분석 결과들을 대비하여 도 5에 나타내었다.
실시예 11
소라페닙(free base) 50 g과 윤활제로 MA 100 g을 DMF 100 ml에 넣고 40℃로 가열하여 완전히 녹인 후, 이 용액을 20℃의 차가운 물에 부어서 고형화하고, 그 결과물을 상온에서 3시간 정도 교반한 후 여과 및 감압건조하여 소라페닙과 MA의 혼합물을 수득하였다. 이 혼합물 9 g에, HPC 0.3 g, PVP (k30) 0.45 g 및 폴록사머(407) 0.45 g을 탈염수 3 ml와 함께 첨가하고 롤 밀을 수회 수행하여 잘 혼합한 후, 이 혼합물을 3롤 밀을 이용하여, 롤러 속도(R.S) 60 rpm, 롤러 온도(R.T) 27~30℃에서 50 회 연속 밀링을 진행하였다. 밀링 공정 중 혼합에 사용된 물은 모두 증발하여 잔존 수분의 함량은 1%(w/w) 이하가 되었다. 수득된 고체분산체형 혼합물을 고압반응기에 넣고, 15~25℃, 70~100 기압에서 초임계유체인 이산화탄소를 연속적으로 흘려 보내어 윤활제로 사용된 지질을 제거하고, 소라페닙, HPC, PVP(k30) 및 폴록사머(407)가 혼합된 분말을 수득하였다.
수득된 분말을 1 mg/ml의 농도(소라페닙 기준)로 탈염된 증류수에 분산한 후, DLS(Dynamic light scattering)방식의 오츠카 ELSZ-1000으로 입도를 측정하여, 평균입자크기가 196.5 ㎚임을 확인하였다. 실시예 11에서 제조된 나노입자의 입도분포도를 도 6에 나타내었다.
실시예 12
활성물질서 항진균제인 포사코나졸(Posaconazole) 40 g과 윤활제로서 MA 80 g을 DMSO 120 ml에 넣고 40℃에서 완전히 녹이고, 이 용액을 30℃의 물에 부어서 고형화한 후, 그 결과물을 3시간 정도 교반한 다음 여과 및 감압 건조하여 포사코나졸과 지질의 혼합물을 수득하였다. 이 혼합물 15 g에 HPC 1.5 g, 폴록사머(407) 0.5 g 및 PVP(k30) 0.5 g을 탈염수 5 ml와 함께 첨가하고, 20℃ 이하의 온도에서 롤 밀을 수회 수행하여 균일하게 혼합하였다. 이때 잔존 수분의 함량은 21%(w/w)이었으며(Karl Fischer 법으로 측정), 이를 상온(25℃)에서 감압 건조하였다. 수득된 고체분산체형 혼합물을 고압반응기에 넣고, 15~25℃, 70~100 기압에서 초임계유체인 이산화탄소를 연속적으로 흘려 보내어 윤활제로 사용된 지질을 제거하고, 포사코나졸, HPC, PVP(k30) 및 폴록사머(407)가 혼합된 분말을 수득하였다.
수득된 분말을 1 mg/ml의 농도(포사코나졸 기준)로 탈염된 증류수에 분산한 후, DLS(Dynamic light scattering)방식의 오츠카 ELSZ-1000으로 입도를 측정하여, 평균입자크기가 286.5 ㎚임을 확인하였다. 실시예 12에서 제조된 나노입자의 입도분포도를 도 7에 나타내었다.
실시예 13
실시예 12에서 제조한 포사코나졸과 윤활제인 MA의 혼합물 15 g에 HPC(ssL) 1.5 g, 폴록사머(407) 0.5 g 및 PVP(k30) 0.5 g을 탈염수 5 ml와 함께 첨가하고, 20℃ 이하의 온도에서 롤 밀을 수회 수행하여 균일하게 혼합하였다. 이 혼합물에 대하여, 3롤 밀을 이용하여 롤 속도 (R.S) 60 rpm, 롤러 온도 (R.T) 13~17℃에서 20 회 연속 밀링을 진행하였다. 이때 잔존 수분의 함량은 8.9%(w/w)이었으며(Karl Fischer 법으로 측정), 이를 상온(25℃)에서 감압 건조하였다. 수득된 고체분산체형 혼합물을 고압반응기에 넣고, 15~25 ℃, 70~100 기압에서 초임계유체인 이산화탄소를 연속적으로 흘려 보내어 윤활제로 사용된 지질을 제거하고, 포사코나졸, HPC, PVP(k30) 및 폴록사머(407)가 혼합된 분말을 수득하였다.
수득된 분말을 1 mg/ml의 농도(포사코나졸 기준)로 탈염된 증류수에 분산한 후, DLS(Dynamic light scattering)방식의 오츠카 ELSZ-1000으로 입도를 측정하여, 평균입자크기가 185.9 ㎚임을 확인하였다. 실시예 13에서 제조된 나노입자의 입도분포도를 도 7에 나타내었다.
실시예 14
2롤 밀을 이용한 것을 제외하고는, 실시예 13과 동일한 활성물질 대비 부형제의 비율, 밀링 공정 온도 조건 및 밀링 횟수를 사용하여 포사코나졸, HPC, PVP(k30) 및 폴록사머(407)가 혼합된 분말을 수득하고, 실시예 13과 동일한 방법으로 윤활제인 지질을 제거하여 평균입자크기가 199.2 nm인 나노입자를 제조하였다.
실시예 15~16
활성물질로 포사코나졸 1 중량부와 윤활제로서 MA 0.5 중량부 또는 1 중량부를 DMSO 2 중량부에 넣고 45℃로 가열하여 완전히 녹이고, 이 용액을 25℃ 탈염수에 부어 고형화하였다. 3 시간 교반 후, 여과 및 감압 건조하여, 포사코나졸:MA가 각각 1:0.5 및 1:1인 고형혼합물을 얻었다.
이 혼합물 각각에, 포사코나졸 1중량부에 대하여 HPC 0.3 중량부, 폴록사머(407) 0.1 중량부 및 PVP(k90) 0.1 중량부를 탈염수 1중량부와 함께 첨가하고, 20℃ 이하의 온도에서 롤 밀을 수회 수행하여 균일하게 혼합하였다. 이 혼합물에 대하여, 3롤 밀을 이용하여, 롤 속도(R.S) 60 rpm, 롤러 온도(R.T) 15~17℃에서 각각 45회 및 60회 연속 밀링을 진행하였다. 이때 잔존 수분의 함량은 각각 8.04 및 7.61%(w/w)(Karl Fischer 법으로 측정)이었으며, 이를 상온(25℃)에서 감압 건조하였다. 수득된 고체분산체형 혼합물을 고압반응기에 넣고, 15~25 ℃, 70~100 기압에서 초임계유체인 이산화탄소를 연속적으로 흘려 보내어 윤활제로 사용된 지질을 제거하고, 포사코나졸, HPC, PVP(k90) 및 폴록사머(407)가 혼합된 분말을 수득하였다.
수득된 분말을 1 mg/ml의 농도(포사코나졸 기준)로 탈염된 증류수에 분산한 후, DLS(Dynamic light scattering)방식의 오츠카 ELSZ-1000으로 입도를 측정하여, 그 평균입자크기를 하기 표 3에 나타내었다. 실시예 15 및 16에서 제조된 나노입자의 입도분포도들을 도 8에 나타내었다.

실시예 17~19
활성물질로 포사코나졸 6 g과 윤활제로서 MA 12 g 및 HPC(ssL) 1.8 g을 탈염수 6 ml와 함께 혼합하고, 이 혼합물에 대하여, 3롤 밀을 이용하여, 롤 속도(R.S) 60 rpm, 롤러 온도(R.T) 27~30℃에서 각각 30 회, 50회 및 70회 연속 밀링을 진행하였다. 밀링 공정 중 혼합에 사용된 물은 모두 증발하여 잔존 수분의 함량은 1%(w/w) 이하가 되었다. 수득된 고체분산체형 혼합물을 고압반응기에 넣고, 15~25℃, 70~100 기압에서 초임계유체인 이산화탄소를 연속적으로 흘려 보내어 윤활제로 사용된 지질을 제거하여, 포사코나졸 및 HPC(ssL) 가 혼합된 분말을 수득하였다.
수득된 분말을 1 mg/ml의 농도(포사코나졸 기준)로 탈염된 증류수에 분산한 후, DLS(Dynamic light scattering)방식의 오츠카 ELSZ-1000으로 입도를 측정하여, 그 평균입자크기를 하기 표 4에 나타내었다. 실시예 17 내지 19에서 제조된 나노입자의 입도분포도들을 도 9에 나타내었다.

활성성분: 포사코나졸
실시예 20
활성물질로 포사코나졸 6 g과 윤활제로서 MA 12 g 및 HPC(ssL) 1.8 g을 탈염수 6 ml와 함께 혼합하고, 이 혼합물에 대하여, 3롤 밀을 이용하여, 롤 속도(R.S) 60 rpm, 롤러 온도(R.T) 27~30℃에서 60회 연속 밀링을 진행하였다. 밀링 공정 중 혼합에 사용된 물은 모두 증발하여 잔존 수분의 함량은 1%(w/w) 이하가 되었다. 얻어진 고체분산체에 폴록사머(407) 0.6 g 및 PVP(k30) 0.6 g을 탈염수 6 ml와 함께 첨가하고, 20℃ 이하의 온도에서 롤 밀을 수회 수행하여 균일하게 혼합하였다. 이 혼합물에 대하여, 3롤밀을 이용하여, 롤 속도(R.S) 60 rpm, 롤러 온도(R.T) 13~15℃에서 24 회 연속 밀링을 진행하였다. 이때 잔존 수분의 함량은 8.48%(w/w)(Karl Fischer 법으로 측정)이었으며, 이를 상온(25℃)에서 감압 건조하였다. 수득된 고체분산체형 혼합물을 고압반응기에 넣고, 15~25 ℃, 70~100 기압에서 초임계유체인 이산화탄소를 연속적으로 흘려 보내어 윤활제로 사용된 지질을 제거하고, 포사코나졸, HPC, PVP(k30) 및 폴록사머(407)가 혼합된 분말을 수득하였다.
수득된 분말을 1 mg/ml의 농도(포사코나졸 기준)로 탈염된 증류수에 분산한 후, DLS(Dynamic light scattering)방식의 오츠카 ELSZ-1000으로 입도를 측정하여, 그 평균입자크기가 254.4 ㎚임을 확인하였다.

Claims (18)

  1. (1) 활성물질, 윤활제로서 지질, 및 유리전이온도(glass transition temperature)가 80℃ 이상인 생체적합고분자를 포함하는 혼합물을 제공하는 단계;
    (2) 밀링 공정을 통하여 상기 (1)단계의 결과물을 분쇄하는 단계; 및
    (3) 초임계유체를 사용하여 상기 (2)단계의 결과물로부터 지질을 제거하는 단계;를 포함하는,
    활성물질 나노입자의 제조 방법.
  2. 제1항에 있어서, 상기 (1)단계가 활성물질, 윤활제로서 지질, 및 유리전이온도가 80℃ 이상인 생체적합고분자를 물리적으로 균일하게 혼합하는 단계인, 활성물질 나노입자의 제조 방법.
  3. 제1항에 있어서, 상기 (1)단계가 활성물질과 윤활제로서 지질을 혼합하고, 이 혼합물에 유리전이온도가 80℃ 이상인 생체적합고분자를 탈염수와 함께 첨가한 후, 그 결과물을 물리적으로 균일하게 혼합하는 단계인, 활성물질 나노입자의 제조 방법.
  4. 제1항에 있어서, 상기 (1)단계가 활성물질, 윤활제로서 지질 및 유리전이온도가 80℃ 이상인 생체적합고분자, 및 유리전이온도가 80℃ 미만인 생체적합고분자, 계면활성제 및 응집방지제로부터 선택되는 하나 이상의 추가 성분을 물리적으로 균일하게 혼합하는 단계인, 활성물질 나노입자의 제조 방법.
  5. 제1항에 있어서, 상기 (1)단계가 활성물질과 윤활제로서 지질을 혼합하고, 이 혼합물에 유리전이온도가 80℃ 이상인 생체적합고분자 및 유리전이온도가 80℃ 미만인 생체적합고분자, 계면활성제 및 응집방지제로부터 선택되는 하나 이상의 추가 성분을 탈염수와 함께 첨가한 후, 그 결과물을 물리적으로 균일하게 혼합하는 단계인, 활성물질 나노입자의 제조 방법.
  6. 제1항에 있어서, 상기 (1)단계가 활성물질 및 윤활제로서 지질을 수혼화성 유기용매에 용해시켜 얻어진 용액을 물에 부어 고형화시킨 후, 이를 여과하고 건조하여 얻어진 혼합 분말에 유리전이온도가 80℃ 이상인 생체적합고분자를 첨가하고, 그 결과물을 물리적으로 균일하게 혼합하는 단계인, 활성물질 나노입자의 제조 방법.
  7. 제1항에 있어서, 상기 (1)단계가 활성물질 및 윤활제로서 지질을 수혼화성 유기용매에 용해시켜 얻어진 용액을 물에 부어 고형화시킨 후, 이를 여과하고 건조하여 얻어진 혼합 분말에 유리전이온도가 80℃ 이상인 생체적합고분자를 탈염수와 함께 첨가하고, 그 결과물을 물리적으로 균일하게 혼합하는 단계인, 활성물질 나노입자의 제조 방법.
  8. 제1항에 있어서, 상기 (1)단계가 활성물질 및 윤활제로서 지질을 수혼화성 유기용매에 용해시켜 얻어진 용액을 물에 부어 고형화시킨 후, 이를 여과하고 건조하여 얻어진 혼합 분말에 유리전이온도가 80℃ 이상인 생체적합고분자 및 유리전이온도가 80℃ 미만인 생체적합고분자, 계면활성제 및 응집방지제로부터 선택되는 하나 이상의 추가 성분을 첨가하고, 그 결과물을 물리적으로 균일하게 혼합하는 단계인, 활성물질 나노입자의 제조 방법.
  9. 제1항에 있어서, 상기 (1)단계가 활성물질 및 윤활제로서 지질을 수혼화성 유기용매에 용해시켜 얻어진 용액을 물에 부어 고형화시킨 후, 이를 여과하고 건조하여 얻어진 혼합 분말에 유리전이온도가 80℃ 이상인 생체적합고분자 및 유리전이온도가 80℃ 미만인 생체적합고분자, 계면활성제 및 응집방지제로부터 선택되는 하나 이상의 추가 성분을 탈염수와 함께 첨가하고, 그 결과물을 물리적으로 균일하게 혼합하는 단계인, 활성물질 나노입자의 제조 방법.
  10. 제1항에 있어서, 상기 (1)단계가 활성물질, 윤활제로서 지질 및 유리전이온도가 80℃ 이상인 생체적합고분자를 수혼화성 유기용매에 용해시켜 얻어진 용액을 물에 부어 고형화시킨 후, 이를 여과하고 건조하는 단계인, 활성물질 나노입자의 제조 방법.
  11. 제1항에 있어서, 상기 (1)단계가 활성물질, 윤활제로서 지질 및 유리전이온도가 80℃ 이상인 생체적합고분자를 수혼화성 유기용매에 용해시켜 얻어진 용액을 물에 부어 고형화시킨 후, 이를 여과하고 건조하여 얻어진 혼합 분말에 유리전이온도가 80℃ 미만인 생체적합고분자, 계면활성제 및 응집방지제로부터 선택되는 하나 이상의 추가 성분을 첨가하고, 그 결과물을 물리적으로 균일하게 혼합하는 단계인, 활성물질 나노입자의 제조 방법.
  12. 제1항에 있어서, 상기 (1)단계가 활성물질, 윤활제로서 지질 및 유리전이온도가 80℃ 이상인 생체적합고분자를 수혼화성 유기용매에 용해시켜 얻어진 용액을 물에 부어 고형화시킨 후, 이를 여과하고 건조하여 얻어진 혼합 분말에 유리전이온도가 80℃ 미만인 생체적합고분자, 계면활성제 및 응집방지제로부터 선택되는 하나 이상의 추가 성분을 탈염수와 함께 첨가하고, 그 결과물을 물리적으로 균일하게 혼합하는 단계인, 활성물질 나노입자의 제조 방법.
  13. 제1항에 있어서, 상기 (2)단계에서의 밀링 공정이 서로 역방향으로 회전하는 롤들을 사용하여 연속적으로 수행되는, 활성물질 나노입자의 제조 방법.
  14. 제1항에 있어서, 상기 (3)단계에서의 지질 제거가, 상기 (2)단계의 결과물이 담긴 반응기에 초임계유체를 연속적으로 투입 및 유출시킴으로써 수행되는, 활성물질 나노입자의 제조 방법.
  15. 제1항에 있어서, 상기 (3)단계에서 초임계유체를 이용한 지질 제거가 50 기압 이상의 압력 조건 및 5~60 ℃의 온도 조건 하에서 수행되는, 활성물질 나노입자의 제조 방법.
  16. 제1항 내지 제15항 중 어느 한 항에 있어서, 상기 활성물질이 유기화합물, 유기금속 화합물, 천연 추출물, 단백질 및 이들의 조합으로부터 선택되는, 활성물질 나노입자의 제조 방법.
  17. 제1항 내지 제15항 중 어느 한 항에 있어서, 상기 지질이 탄소수 10~22의 포화 지방산, 탄소수 10~22의 포화 지방산 에스테르, 탄소수 10~22의 포화 지방 알코올, 탄소수 10~22의 포화지방산기를 갖는 모노-, 디- 또는 트리-글리세라이드, 탄소수 14 이상의 탄화수소 및 이들의 조합으로부터 선택되는, 활성물질 나노입자의 제조 방법.
  18. 제1항 내지 제15항 중 어느 한 항에 있어서, 상기 유리전이온도가 80℃ 이상인 생체적합고분자가, Tg가 80℃ 이상인 셀룰로스 계열의 생체적합고분자, Tg가 80℃ 이상인 폴리비닐피롤리돈, Tg가 80℃ 이상인 폴리비닐알코올 및 Tg가 80℃ 이상인 유드라짓 계열의 생체적합고분자로부터 선택되는 1종 이상인, 활성물질 나노입자의 제조 방법.
