液状物に対する殺菌方法において、殺菌作用を担保しつつも、更に風味の劣化を抑制した殺菌方法及び装置を提供する。 本発明の殺菌方法は、水以外の飲料物である被処理溶液に対して、発光波長が260nm以下の紫外光を実質的に照射せずに、発光波長が280nm以上320nm以下の紫外光を照射する方法である。

Description

殺菌方法、殺菌装置
 本発明は、水以外の飲料物に対する殺菌方法、及び殺菌装置に関する。
 従来、光を用いた殺菌処理として、紫外から可視域までの光をパルス照射させるものがある。また近年では、紫外光を用いた殺菌処理方法が期待されている。
 特に、食品の殺菌用途では、紫外光を用いた殺菌(UV殺菌)は熱殺菌と比べて風味を劣化させにくい方法として考えられている。そして、発光効率の観点から、水銀ランプ(主たる発光波長は254nm)を用いて、当該ランプから放射される紫外光を対象物に対して照射することで、風味の劣化を抑えつつ殺菌処理することが知られている(下記特許文献1,2参照)
 また、下記特許文献3では、飲食物である果汁飲料やゼリー、ムースなどの殺菌処理として、紫外光を照射する装置が記載されている。この文献では、波長域として200~300nm、特に220nm~280nm(UV-C)の紫外光を用いることで、殺菌処理を行うことが記載されている。

 本発明は、液状物に対する殺菌方法において、殺菌作用を担保しつつも、更に風味の劣化を抑制した殺菌方法及び装置を提供することを目的とする。
 従来、飲料物を含む食品などの風味の劣化の分析は、官能検査により行われていた。これに対し、近年の化学分析技術の進歩により、分析装置による定量的な分析が可能となってきた。本発明者は、この分析装置によって定量的な分析を行ったところ、液状物に対して紫外光照射によって殺菌を行う場合において、従来好ましいとされていた水銀ランプからの光(波長254nm)を用いると、処理前には存在していなかった新たな物質が発生したり、特定の物質の量が大きく変化することで、味やにおい(香り)が変化してしまうという課題を新規に発見した。
 本発明に係る殺菌方法は、水以外の飲料物である被処理溶液に対して、発光波長が260nm以下の紫外光を実質的に照射せずに、発光波長が280nm以上320nm以下の紫外光を照射することを特徴とする。
 被処理溶液に対して上記波長帯(UV-B)の光を照射することで、波長254nmの光が照射される際に生じる苦味や雑味成分や悪臭成分の元となる物質の発生を抑え、風味の阻害を防ぎつつ、殺菌が可能であることが確認された。
 本発明に係る殺菌装置は、水以外の飲料物である被処理溶液が流通される流路に沿って、主たる発光波長が280nm以上320nm以下であって、発光波長が260nm以下の成分を実質的に含まない紫外光を照射する紫外光照射装置が設けられていることを特徴とする。
 本発明によれば、殺菌作用を担保しつつも、更に風味の劣化を抑制した殺菌方法及び装置が実現される。

 本発明の殺菌装置及び殺菌方法における処理対象は、水以外の飲料物である被処理溶液である。このような被処理溶液の例としては、ワイン、日本酒、ビールなどの酒類、コーヒー、ジュース、及び、液状の香料自体が含まれる。
 図1は、本発明の殺菌装置の一例における要部の構成を概略的に示す、流路に沿った断面図である。図2は、図1におけるA-A線断面図である。なお、各図面において、図面上の寸法比と実際の寸法比とは必ずしも一致しない。
 この殺菌装置は、リアクター10を備えている。リアクター10は、被処理溶液2が通流される流路3と、この流路3に沿って設けられた紫外光照射装置20とを備える。紫外光照射装置20は、主たる発光波長が280nm以上320nm以下であって、発光波長が260nm以下の成分を実質的に含まない紫外光を発する構成である。
 本明細書において、主たる発光波長が280nm以上320nm以下である光とは、発光スペクトルの半値を示す波長帯が280nm以上320nm以下の範囲内を示す光を指す。また、発光波長が260nm以下の成分を実質的に含まない光とは、発光スペクトルの半値を示す波長帯に260nm以下の成分が含まれない光を指す。
 リアクター10は、互いに同軸上に配置された円筒状の外管11及び円筒状の内管12を有する。被処理溶液2の流路3は、外管11の内周面と内管12の外周面とに挟まれた領域によって構成される。
 リアクター10における外管11を構成する材料としては、特に限定されないが、例えばステンレス鋼などの金属材料を用いることができる。リアクター10における内管12を構成する材料としては、紫外光照射装置20からの紫外光を透過するものであればよく、例えば石英ガラスなどを用いることができる。
 紫外光照射装置20は、リアクター10における内管12の内部において、リアクター10の中心軸5に沿って配置されている。上述したように、紫外光照射装置20は、主たる発光波長が280nm以上320nm以下であって、発光波長が260nm以下の成分を実質的に含まない紫外光を放射する光源を備えている。発光波長に260nm以下の成分が含まれる光(例えば、低圧水銀ランプ(254nmの輝線)からの光)が被処理溶液2に放射されると、被処理溶液2の風味が劣化してしまう。この点は、実施例を参照して後述される。
 紫外光照射装置20として利用される光源としては、例えば、XeとBrの混合ガスが放電用ガスとして封入されたXeBrエキシマランプ(ピーク波長が283nm)、Br2が放電用ガスとして封入されたエキシマランプ(ピーク波長が289nm)、XeとClの混合ガスが放電用ガスとして封入されたXeClエキシマランプ(ピーク波長が308nm)などを用いることができる。
 また、紫外光照射装置20として利用される光源として、例えば紫外光放射蛍光ランプを用いることもできる。紫外光放射蛍光ランプは、誘電体バリア放電によって生成されるエキシマから放出される光を励起光として蛍光体に照射し、その蛍光体が励起することによって得られる特定の波長範囲の紫外光を放射光として放射する。蛍光体としては、例えば、励起によってピーク波長が290nmの紫外光を放射するビスマス付活イットリウムアルミニウムホウ酸塩などを用いることができる。また、蛍光体として、励起によってピーク波長が320nm付近に半値幅の広い発光ピークを有するセリウム付活リン酸ランタンなどを用いることもできる。
 更に、紫外光照射装置20として利用される光源として、発光波長が280nm以上320nm以下であって、発光波長が260nm以下の成分を実質的に含まないように構成されたLED素子を用いることもできる。
 紫外光照射装置20による被処理溶液2に対する紫外光照射量は、例えば170mJ/cm2以上であることが好ましく、より好ましくは170~500mJ/cm2である。紫外光照射量を170mJ/cm2以上とすることで、被処理溶液2自体の味やにおいの変化を抑制しながら、殺菌処理を行うことができる。
 なお、リアクター10における流路3の径、すなわち、外管11の内周面と内管12の外周面との間の距離は、例えば0.05~1mmであることが好ましい。このような構成とすることで、仮に、被処理溶液2が、紫外光照射装置20からの紫外光の透過率が低い材料で構成されている場合(例えば被処理溶液2に照射される紫外光の光量の99%が不透過である場合)においても、被処理溶液2に対して均一に殺菌することができる。
 流路3内に流通される被処理溶液2の流量、流路3における紫外光が照射される領域の大きさ(すなわち、紫外光照射装置20に含まれる光源の、流路3の方向に係る長さ)、及びその他の条件は、紫外光照射量が上記特定の範囲内の大きさとなるように適宜設定することができる。
 上記の殺菌装置においては、処理すべき被処理溶液2が流路3内に導入されて、当該流路3内を流通される過程において、紫外光照射装置20から放射される、上記波長帯の紫外光が被処理溶液2に照射されることにより、被処理溶液2の殺菌処理が行われる。これにより、後述する実施例の結果に示されるように、飲料物としての被処理溶液2の風味の劣化を抑制しながらも、殺菌効果を示すことができる。
 以上、本発明の実施形態について説明したが、本発明は上記の実施形態に限定されるものではなく、種々の変更を加えることができる。例えば、リアクター10は、被処理溶液2が流通される流路に沿って紫外光照射装置20が設けられる構造であればよく、上記のような構造に限定されるものではない。
 以下、実施例を参照して、本発明の効果について説明する。
 図1に示す構成に従い、下記の仕様のリアクター10を製作した。
 外管11はステンレス鋼よりなり、内径がφ27mmである。内管12は石英ガラスよりなり、外径がφ26.5mm、肉厚が1.0mmである。紫外光照射装置20からの紫外光が照射される領域の長さは80mmである。外管11の内周面と内管12の外周面との間の距離、すなわち流路3の径方向の幅は、0.5mmである。
 (実施例1)
 紫外光照射装置20に含まれる光源として、ピーク波長が283nm(半値が280nm~286nm)の紫外光を放射するXeBrエキシマランプを用いた。当該XeBrエキシマランプの発光長は80mmである。
 (実施例2)
 紫外光照射装置20に含まれる光源として、ピーク波長が320nm(半値が310nm~360nm)の紫外光を放射する紫外線エキシマ蛍光ランプ(ウシオ電機株式会社製UV-XEFL320BB)を用いた。当該紫外線エキシマ蛍光ランプの発光長は80mmである。
 (実施例3)
 紫外光照射装置20に含まれる光源として、ピーク波長が290nm(半値が270nm~320nm)の紫外光を放射する紫外線エキシマ蛍光ランプ(ウシオ電機株式会社製UV-XEFL290BB)を用いた。当該紫外線エキシマ蛍光ランプの発光長は80mmである。
 (比較例1)
 紫外光照射装置20に含まれる光源として、ピーク波長が254nm(半値が251~257nm)の紫外光を放射する低圧水銀ランプを用いた。当該低圧水銀ランプの発光長は80mmである。
 (比較例2)
 紫外光照射装置20に含まれる光源として、ピーク波長が222nm(半値が215~229nm)の紫外光を放射するKrClエキシマランプを用いた。当該KrClエキシマランプの発光長は80mmである。
 図3は、実施例1~実施例3、比較例1~2の各光源の発光スペクトルを示す。
 <検証1>
 被処理溶液2として、数種類の材料を準備し、それぞれの被処理溶液2に対して、実施例1及び比較例1の紫外光を照射して、香気分析を行った。被処理溶液2として準備された材料は、コーヒー飲料、アップル果汁、レモン果汁、ワインの4種類である。
 紫外光の照射条件は、以下の通りである。
 被処理溶液流路内における試験溶液の流量:0.72ミリリットル/時
 被処理溶液流路における紫外光強度:3.6mW/cm2
 処理時間:139秒
 紫外光照射量:500mJ/cm2
 処理中の被処理溶液温度:20℃
 香気分析試験は、以下の方法で行われた。
 紫外光照射処理後の各被処理溶液2について、におい嗅ぎGC/GCMSシステム(株式会社島津製作所製のガスクロマトグラフ質量分析装置QP-2010Plus)を用いて、におい成分の分析試験を行った。より詳細には、実施例1の光源、及び比較例1の光源を用いて紫外光を照射した後の各被処理溶液2に対して分析試験を行った。この結果を、図4A、図4B、図5A、図5B、図6A、図6B、図7A、及び図7Bに示す。なお、前記図4A~図7Bの各グラフにおいて、横軸が保持時間であり、縦軸が強度である。
 (コーヒー飲料)
 図4A及び図4Bは、被処理溶液2がコーヒー飲料である場合の結果である。図4Bは、図4Aを見やすくする観点で、未処理、実施例1、及び比較例1の各グラフを、縦軸方向に平行移動させて描画し直したものである。図4A及び図4Bの結果によれば、比較例1において、未処理時及び実施例1では存在していなかった3-メチル-1-ブタノールが検出された。3-メチル-1-ブタノールは、不快な悪臭を有する物質であるところ、比較例1の光源によって処理を行うことで、処理前の状態からにおいが変化することが確認される。
 なお、図4A及び図4Bの分析結果からは、実施例1の光源による光照射後においては、コーヒー由来のにおい成分を除く、新たなにおい成分の発生は確認されなかった。また、実施例1及び比較例1の双方において、未処理時と比較して含有される物質の量にいくらかの変化が見られるものの、これらの変化は、コーヒー由来のにおいを変化させるほどのものとは認められなかった。
 (アップル果汁)
 図5A及び図5Bは、被処理溶液2がアップル果汁である場合の結果である。図5Bは、図5Aを見やすくする観点で、未処理、実施例1、及び比較例1の各グラフを、縦軸方向に平行移動させて描画し直したものである。図5A及び図5Bの結果によれば、比較例1において、未処理時及び実施例1では存在していなかったシクロヘプタノンが検出された。シクロヘプタノンは、ハッカ臭を有する物質であるところ、比較例1の光源によって処理を行うことで、このにおい成分が増加し、アップル果汁が本来有するにおいとは異なるにおいに変化することが確認される。
 なお、図5A及び図5Bの分析結果からは、実施例1の光源による光照射後においては、アップル果汁由来のにおい成分を除く、新たなにおい成分の発生は確認されなかった。また、実施例1及び比較例1の双方において、未処理時と比較して含有される物質の量にいくらかの変化が見られるものの、これらの変化は、アップル果汁本来のにおいを変化させるほどのものとは認められなかった。
 (レモン果汁)
 図6A及び図6Bは、被処理溶液2がレモン果汁である場合の結果である。図6Bは、図6Aを見やすくする観点で、未処理、実施例1、及び比較例1の各グラフを、縦軸方向に平行移動させて描画し直したものである。図6A及び図6Bの結果によれば、比較例1において、エタノールの強度が大きく上昇することが確認された。エタノールは、アルコール臭を有する物質であるところ、比較例1の光源によって処理を行うことで、このにおい成分が増加し、レモン果汁が本来有するにおいとは異なるにおいに変化することが確認される。
 また、図6A及び図6Bの結果によれば、比較例1において、未処理時及び実施例1では存在していなかったリモネン異性体が検出された。この結果に鑑みると、比較例1の光源からの紫外光がレモン果汁に照射されたことで、柑橘臭を有する物質であるリモネンの一部が破壊されて、新たにリモネン異性体が生成されたものと考えられる。これにより、未処理時及び実施例1の場合と比較して、レモン果汁本来の柑橘系のにおいが変化したものと考えられる。
 (ワイン)
 図7A及び図7Bは、被処理溶液2がワインである場合の結果である。図7Bは、図6Aを見やすくする観点で、未処理、実施例1、及び比較例1の各グラフを、縦軸方向に平行移動させて描画し直したものである。図7A及び図7Bの結果によれば、比較例1において、3-メチル-1-ブタノールの強度が大きく上昇することが確認された。3-メチル-1-ブタノールは、不快な悪臭を有する物質であるところ、比較例1の光源によって処理を行うことで、このにおい成分が増加し、ワインが本来有するにおいとは異なるにおいに変化することが確認される。
 なお、図7A及び図7Bの分析結果からは、実施例1及び比較例1の双方において、未処理時と比較して含有される物質の量にいくらかの変化が見られるものの、これらの変化は、ワインのにおいを変化させるほどのものとは認められなかった。
 <検証2>
 被処理溶液2として、コーヒー飲料を準備し、実施例1、実施例2及び比較例1の紫外光を照射して、味覚分析を行った。紫外光の照射条件は、検証1と同じである。
 味覚分析試験は、以下の方法で行われた。
 各被処理溶液2に対して、上記のリアクター10による紫外光照射処理(殺菌処理)を行った。そして、紫外光照射処理後の各被処理溶液2について、味覚センサー(株式会社インテリジェントセンサーテクノロジー製の味認識装置「TS-5000Z」)を用いて味の分析試験を行った。その結果を図5に示す。
 図8に示すチャートは、紫外光照射処理を行わない場合の被処理溶液2の本来の味の分析により得られた各項目の数値を0.0としたときの相対値で示されている。この味認識装置による味覚分析試験において、敏感な人が味の違いを検知できる閾値は±0.8程度である。なお、図8のチャートに表示されている8つの分析項目は、それぞれ以下の通りである。
 [分析項目]
 (1) 酸味(先味):クエン酸や酒石酸が呈する酸味
 (2) 苦味雑味(先味):苦味由来物質で、低濃度ではコクや隠し味などに相当するもの
 (3) 渋味刺激(先味):渋味物質による刺激味
 (4) 旨味(先味):アミノ酸、核酸などの旨味
 (5) 塩味(先味):食塩などの無機塩の塩味
 (6) 苦味(後味):一般食品に見られる味の苦味
 (7) 渋味(後味):渋味物質由来の後味の渋味
 (8) 旨味コク(後味):旨味物質が呈する持続性のあるコク味
 図8によれば、比較例1の光源を用いて処理をした場合、未処理時と比較して苦味雑味が認識できる程度に変化していることが確認される。一方、実施例1及び実施例2の光源を用いて処理をした場合には、未処理時と比較して味の違いが認識できる程度には変化していない。
 なお、被処理溶液2を、アップル果汁、レモン果汁、ワインとして同様の検証を行ったところ、比較例2は、実施例1及び実施例2と比べて味の変化を示したものの、変化量は±0.8以内に留まっていたことが確認された。
 この検証結果から、コーヒー飲料に対して紫外光を照射する場合、比較例1の光源を用いると、においのみならず、味も変化することが確認された。
 <検証3>
 被処理溶液2として、コーヒー香料、グレープ香料、及びオレンジ香料を準備し、実施例1~実施例3、比較例1~比較例2の紫外光を照射して、味覚分析の官能評価を行った。具体的には、φ71mmの透明のフラワー容器(70g)内に収容した各サンプルを3個ずつ準備し、紫外光の照射前と照射後で風味の変化が確認できるかどうかにつき、10人に対して確認させた。この官能評価の結果を表1に示す。表1において、評価Aは、風味の変化が確認できた人の割合が3%未満であることを示し、評価Bは、風味の変化が確認できた人の割合が3%以上40%未満であることを示し、評価Cは、風味の変化が確認できた人の割合が40%以上であることを示している。

 コーヒー香料については、比較例1の光源からの光を照射した後には、光照射前と比較して風味が低下したことが確認された。また、比較例2の光源からの光を照射した後には、光照射前と比較して風味の低下と共に雑味が増加したことが確認された。一方、実施例1~3の各光源からの光を照射した後においては、光照射前と比較して風味の劣化がほとんど確認されなかった。
 グレープ香料については、比較例1の光源からの光を照射した後には、光照射前と比較して風味及び後味が低下したことが確認された。また、比較例2の光源からの光を照射した後には、光照射前と比較して味が薄くなったことが確認された。一方、実施例1~3の各光源からの光を照射した後においては、光照射前と比較して風味の劣化がほとんど確認されなかった。
 オレンジ香料については、比較例1の光源からの光を照射した後には、光照射前と比較して風味が低下したことが確認された。また、比較例2の光源からの光を照射した後には、光照射前と比較して味が薄くなったことが確認された。一方、実施例1~3の各光源からの光を照射した後においては、光照射前と比較して風味の劣化がほとんど確認されなかった。
 <検証4>
 セレウス菌(JCM2152)を芽胞状態としたものを供試菌として用い、初発菌数が105CFU/mLとなるように、所定量の供試菌を被処理溶液2に懸濁させて試験溶液を調製した。ここでは、被処理溶液2としてコーヒーゼリー香料を採用した。
 紫外光照射処理(殺菌処理)を行う前の試験溶液と、実施例1の光源を用いて紫外光照射処理(殺菌処理)を上記検証1と同一の条件で行った後の試験溶液とをそれぞれ寒天培地上に塗沫し、30℃で48時間培養した後、寒天培地に発生したコロニーの数を調べた。この結果を図9に示す。図9によれば、実施例1の光源を用いた場合であっても、殺菌効果が担保できていることが確認された。なお、処理時間を適切に設定することで、実施例2及び実施例3の光源を用いた場合であっても、殺菌効果が担保できることが確認された。
 一例として、実施例1の光源を用いる場合の処理時間は、55秒以上84秒以下であり、実施例2の光源を用いる場合の処理時間は、100秒以上300秒以下である。なお、この処理時間は、肉や魚などの食品分野においてフラッシュランプを用いた殺菌処理の時間(msecオーダー)と比べて十分に長い時間である。肉や魚は多くの油を含んでおり、この油が光を吸収することで変質してしまう。このため、食品分野においては、極めて短い時間だけ光を照射することで、瞬間的に高温化して殺菌処理を行う方法が用いられることがある。
 上記の検証結果によれば、飲料物に含まれる成分(例えば、糖質、脂質、たんぱく質、炭水化物等、といった化合物)の一部が、光の作用によって変化し、風味を変えているものと考察できる。特に、コーヒー飲料、アップル果汁やレモン果汁などの果汁を含む果汁飲料、ワインなどの酒類飲料に含まれる成分には、光の作用によって変化し、風味を変えるものが含まれていることが分かる。本発明は、飲料物の分野において、所定の波長帯の紫外光を照射することでにおいや味に変化が生じることを新たに発見した上で、前記所定の波長帯を除く特定の波長帯の紫外光を照射することで、殺菌効果を担保しながらも、においや味の変化・劣化を抑制することを可能にするものである。
 ところで、光は、波長が短いほどエネルギーが高い。上述した分析によれば、ピーク波長が254nm(半値が251~257nm)である比較例1の光源を用いて処理を行った場合、被処理溶液2が本来有している物質の結合が破壊されるなどによって、新たな物質が発生したり、すでに含まれている物質の量が大きく変化したものと推察される。このことに鑑みれば、比較例1の光源よりも更に波長の短い光源の光を用いた場合には、においや味が更に変化してしまうものと考えられる。この点は、検証3において、比較例2の光源からの光を各サンプルに照射したときの結果にも、現れている。
 一方、実施例1の光源を用いて処理を行った場合、未処理時と比較してにおい及び味について大きな変化は確認されなかった。このことに鑑みると、実施例1の光源よりも更に波長の長い光源の光を用いた場合には、においや味についての変化の程度は、実施例1と同等か、それよりも抑制されるものと考えられる。この点は、検証3において、実施例2及び実施例3の光源からの光を各サンプルに照射したときの結果にも、現れている。
 そして、検証4で示したように、実施例2及び実施例3の光源を用いた場合であっても、殺菌効果が確認されたことから、発光波長が260nm以下の紫外光を実質的に照射せずに、発光波長が280nm以上320nm以下の紫外光を照射することにより、味やにおいの変化を抑制しながら、殺菌処理を行うことができることが分かる。
 図10は、同一の光量を照射したときの波長別の殺菌力の相対値を示すグラフである。図10において、横軸は波長を示し、縦軸は254nmにおける殺菌力を100%としたときの波長別の殺菌力の相対値を示す。波長280nmの光の殺菌力は、波長254nmに対して約60%である。なお、波長320nmよりも長波長の光の殺菌力は、波長254nmに対して0.001%以下となる。
 殺菌の能力は、照射光量を高めることで向上する。すなわち、光源の出力や照射時間を調整することで、実質的に殺菌能力を実用的なレベルに担保することができる。しかし、波長が320nmよりも長波長の範囲内の光については、光源の出力を極めて高くし、且つ、極めて長い照射時間を確保しなければ、殺菌の効果を十分に確保することができない。特に、図1に示すように、流路3内に被処理溶液2を流しながら、光源から光を照射して殺菌することを想定した場合、320nmよりも長波長の光を発する光源を採用する場合には、極めて長い照射時間を確保するために、被処理溶液2の流速を極めて遅くすると共に、光源の流路方向の長さを極めて長くする必要があり、実用的ではない。
 以上の点に鑑みると、発光波長が260nm以下の紫外光を実質的に照射せずに、発光波長が280nm以上320nm以下の紫外光を照射することにより、現実的な装置設計の下で、味やにおいの変化を抑制しながら、殺菌処理を行うことができることが分かる。

Claims (5)

  1.  水以外の飲料物である被処理溶液に対して、発光波長が260nm以下の紫外光を実質的に照射せずに、発光波長が280nm以上320nm以下の紫外光を照射することを特徴とする殺菌方法。
  2.  水以外の飲料物である被処理溶液が流通される流路に沿って、主たる発光波長が280nm以上320nm以下であって、発光波長が260nm以下の成分を実質的に含まない紫外光を照射する紫外光照射装置が設けられていることを特徴とする殺菌装置。
  3.  コーヒー飲料である被処理溶液に対して、主たる発光波長が280nm以上320nm以下であって、発光波長が260nm以下の成分を実質的に含まない紫外光を照射することを特徴とする殺菌方法。
  4.  果汁飲料である被処理溶液に対して、主たる発光波長が280nm以上320nm以下であって、発光波長が260nm以下の成分を実質的に含まない紫外光を照射することを特徴とする殺菌方法。
  5.  酒類飲料である被処理溶液に対して、主たる発光波長が280nm以上320nm以下であって、発光波長が260nm以下の成分を実質的に含まない紫外光を照射することを特徴とする殺菌方法。
