Description
本明細書において、「ポリヌクレオチド」は、「核酸」または「核酸分子」とも換言できる。「ポリヌクレオチド」は、特に明記しない場合は、天然に存在するヌクレオチドと同様に機能することができる天然に存在するヌクレオチドの既知の類似体を含有するポリヌクレオチドを包含する。また、「塩基配列」は、「核酸配列」または「ヌクレオチド配列」とも換言できる。特に言及のない限り、「塩基配列」はデオキシリボヌクレオチドの配列またはリボヌクレオチドの配列を意図している。また、ポリヌクレオチドは、一本鎖であっても二本鎖構造であってもよく、一本鎖の場合はセンス鎖であってもアンチセンス鎖であってもよい。
本発明の一実施形態に係る核酸構築物は、Trkの細胞内領域と膜局在化配列とを含む融合ポリペプチドをコードする核酸構築物である。本発明は、この核酸構築物を細胞内で発現させることによって、神経栄養因子(ニューロトロフィン)の投与を行わずとも、Trkの下流の細胞内シグナル経路を活性化可能にするという驚くべき知見に基づきなされたものである。すなわち、本願発明は、この核酸構築物がコードしている融合ポリペプチドが持つ膜局在化配列を介して、Trkの細胞内領域を細胞膜に局在させるという、細胞活性化の新たな手法に関する。
1)対応する神経栄養因子を過剰発現又は投与することを要さない。
2)Trk全長と比較して、活性化に必要な分子サイズを大幅に小さくすることができる。これによって、分子の高発現化を可能とする。さらに、核酸構築物の設計の自由度が大幅に拡大するため、例えば、高発現化や遺伝子導入効率の向上等をより容易に実現することができる。
本実施形態に係る核酸構築物がコードしている融合ポリペプチドは、後述する、1)Trkの細胞内領域と、2)膜局在化配列と、を含むものである。膜局在化配列は、その特性に応じて、融合ポリペプチドのN末端側、又は、融合ポリペプチドのC末端側に配置されることが好ましい場合がある。Trkの細胞内領域との関係では、膜局在化配列は、当該細胞内領域のN末端側(膜貫通領域に近い側)に対して配置しても、C末端側に対して配置してもよい。なお、実施例に示す常時活性型Trkでは、膜局在化配列は、融合ポリペプチドのC末端側に配置され、かつ、Trkの細胞内領域のC末端側に対して配置されている。これにより、実施例に示す常時活性型Trkは、細胞膜に対して、Trkの細胞内領域を通常とは逆に位置させる(図1・9も参照)。
本実施形態において、Trkの細胞内領域とは、一回膜貫通型のタンパク質である、神経栄養因子の高親和性受容体(Trk)の細胞内領域に相当するポリペプチド、並びに、当該ポリペプチドと機能的に同等なポリペプチドを指す。細胞内領域は、細胞質ドメインとも称される。
1)配列番号2、6、9、12、又は14の何れかに記載のアミノ酸配列で示されるポリペプチド。なお、配列番号2はヒト由来のTrkBの細胞内領域のアミノ酸配列の一例であり、配列番号6はマウス由来のTrkAの細胞内領域のアミノ酸配列の一例であり、配列番号9はマウス由来のTrkCの細胞内領域のアミノ酸配列の一例であり、配列番号12はヒト由来のTrkAの細胞内領域のアミノ酸配列の一例であり、配列番号14はヒト由来のTrkCの細胞内領域のアミノ酸配列の一例である。
2)配列番号2、6、9、12、又は14の何れかに記載のアミノ酸配列で示されるポリペプチドと機能的に同等なポリペプチド。機能的に同等なポリペプチドとしては、例えば、配列番号2、6、又は9の何れかに記載のアミノ酸配列と90%以上の配列同一性を示すアミノ酸配列で示されるポリペプチドが挙げられる。なお、配列同一性は、91%以上、92%以上、93%以上、94%以上、95%以上、96%以上、97%以上、98%以上、又は99%以上がより好ましい。
なお、配列番号6に示すアミノ酸配列における104番目のアミノ酸(Lys)は、チロシンキナーゼ活性又はTrkAの下流の細胞内シグナル経路の活性化に不可欠なアミノ酸であるため変異が無いことが好ましい。
同様に、配列番号2に示すアミノ酸配列における134番目のアミノ酸(Lys)は、チロシンキナーゼ活性又はTrkBの下流の細胞内シグナル経路の活性化に不可欠なアミノ酸であるため、変異が無いことが好ましい。
同様に、配列番号9に示すアミノ酸配列における118番目のアミノ酸(Lys)は、チロシンキナーゼ活性又はTrkCの下流の細胞内シグナル経路の活性化に不可欠なアミノ酸であるため、変異が無いことが好ましい。
同様に、配列番号12に示すアミノ酸配列における104番目のアミノ酸(Lys)は、チロシンキナーゼ活性又はTrkAの下流の細胞内シグナル経路の活性化に不可欠なアミノ酸であるため、変異が無いことが好ましい。
同様に、配列番号14に示すアミノ酸配列における118番目のアミノ酸(Lys)は、チロシンキナーゼ活性又はTrkCの下流の細胞内シグナル経路の活性化に不可欠なアミノ酸であるため、変異が無いことが好ましい。
膜局在化配列は、上記の融合ポリペプチドを、細胞膜に局在化させるためのペプチド配列である。膜局在化配列は、好ましくは、アミノ酸の脂質修飾を誘導するものである。アミノ酸の脂質修飾の種類は特に限定されないが、例えば、1)脂肪酸アシル化、2)プレニル化、3)グリコシルホスファチジルイノシトール化、又は、4)コレステロール化、等が挙げられ、より具体的には、ミリストイル化、パルミトイル化、ファルネシル化、又は、ゲラニルゲラニル化等が挙げられ、中でも好ましくはファルネシル化、又は、ゲラニルゲラニル化が挙げられる。
1)Met-Gly-X-X-X-Ser-X-X-X(N末端側脂質修飾モチーフ(Nミリストイル化))2)CAAXモチーフ、CCモチーフ、CXCモチーフ、CCXXモチーフ(C末端側脂質修飾モチーフ)。
本実施形態に係る核酸構築物の一形態は、Trkの細胞内領域と膜局在化配列とを含む上記の融合ポリペプチドをコードする遺伝子である。その一例は、配列番号4、5、8、又は11に示す何れかの塩基配列を持つ遺伝子であり、或いは、配列番号4、5、8、又は11に示す塩基配列の何れかと85%以上、90%以上、91%以上、92%以上、93%以上、94%以上、95%以上、96%以上、97%以上、98%以上、又は99%以上の配列同一性を示す塩基配列を持つ遺伝子である。なお、上記融合ポリペプチドを構成するTrkの細胞内領域をコードする遺伝子の一例は、配列番号3、7、10、13、又は15に示す何れかの塩基配列を持つ遺伝子であり、或いは、配列番号3、7、10、13、又は15に示す塩基配列の何れかと85%以上、90%以上、91%以上、92%以上、93%以上、94%以上、95%以上、96%以上、97%以上、98%以上、又は99%以上の配列同一性を示す塩基配列を持つ遺伝子である。
本発明の一実施形態に係る医薬組成物は、上記した核酸構築物を含んでなる遺伝子治療薬である。この医薬組成物は、一例において、神経関連疾患薬であり、より具体的な一例では、神経保護又は神経再生用である。この医薬組成物は、強力な神経保護効果と軸索再生効果とを発揮し、神経関連疾患の治療及び/又は予防に用いることができる。本発明の範疇には、この医薬組成物を投与対象に有効量投与する、神経関連疾患の治療及び/又は予防の方法が含まれる。
投与対象となるヒト又は非ヒト動物は、好ましくは、ヒトを含む哺乳類からなる群より選択される何れかである。投与対象となる哺乳類の種類は特に限定されないが、マウス、ラット、ウサギ、モルモット、ヒトを除く霊長類等の実験動物;イヌ、ネコ等の愛玩動物;ウシ、ウマ等の家畜;ヒト;が挙げられ、特に好ましくはヒトである。投与対象となるヒト又は非ヒト動物は、例えば、後述する神経関連疾患を有している。
治療及び/又は予防の対象となる神経関連疾患の種類は特に限定されないが、神経系細胞の変性、及び/又は、神経系細胞の細胞死に起因して、その機能が損なわれる病態を指す。神経系細胞の変性や細胞死の原因は特に限定されず、事故等を原因とする神経系の損傷等も含まれる。
医薬組成物の投与方法は特に限定されず、注入(注射器又は注入ポンプ等を利用)、点眼投与、経皮投与、舌下投与といった手法により局所投与されてもよいし、経口投与、静脈内又は動脈内への血管内投与、腸内投与といった手法により全身投与されてもよい。好ましい一つの投与態様では、治療等の対象となる神経系の近傍に対して局所投与される。
本発明の一実施形態に係る医薬組成物は、上記の核酸構築物と、薬学的に許容される担体とを少なくとも含んで構成されていてもよい。薬学的に許容される担体は、特に限定されないが、上記の核酸構築物の機能を実質的に阻害せず、かつ、投与対象となるヒト又は非ヒト動物に対して実質的な悪影響を及ぼさないという性質を備えることが好ましい。なお、この医薬組成物は水等の液体担体を含むものであることが好ましい場合があり、一例ではリポソーム製剤であってもよい。
本発明の一実施形態に係る細胞活性化の方法は、細胞内で、Trkの細胞内領域を、膜局在化配列を介して細胞膜に局在させる工程(局在化工程)を含む方法である。この局在化工程は、典型的には、〔1.核酸構築物〕欄に記載の核酸構築物、及び、〔2.医薬組成物〕欄に記載の医薬組成物から選択される何れかを神経系細胞内で発現させることによって行うことが出来る。ここで神経系細胞の種類は特に限定されず、中枢神経系の細胞、末梢神経系の細胞、これらの細胞の前駆細胞等を包含する。神経系細胞は単離されたものであってもよく、ヒト又は非ヒト動物の体内に存在するものであってもよい。
上述の通り、本発明は、以下に示す態様を含む。
2)配列番号2、6、9、12、又は14の何れかに記載のアミノ酸配列と90%以上の同一性を示すアミノ酸配列で示されるポリペプチド。
(方法)
TrkB分子の細胞内領域を発現するAAVベクター(AAV-CA-TrkBと称する)を、次の通り作製した。CA-TrkBをコードする遺伝子断片を組み込むAAVベクターは、Applied Viromics社製のpAAV-CAG-shuttle-WPREを用いた。TrkBの細胞内領域のN末端にmyc-タグを付加し、当該領域のC末端にファルネシル(Farnecyl)化シグナル配列を付加したタンパク質をコードするDNA断片を、CA-TrkBをコードする遺伝子断片として準備した(図1も参照)。すなわち、CA-TrkBは、TrkBの細胞外領域および膜貫通領域を含まず、TrkBの細胞内領域のみを用い、当該細胞内領域にmyc-タグおよびファルネシル化シグナル配列を付加して構成している。
AAV-CA-TrkBをCos7細胞へトランスフェクションした結果を、図3及び図4に示す。図3は、AAV-CA-TrkBをCos7細胞へトランスフェクションして、24時間の培養の後に細胞を溶解し、抗myc抗体(Santa Cruz社製のc-Myc抗体(9E10))を用いてウエスタンブロットを行った結果を示す。AAV-CA-TrkBは、同様の実験を行った、全長TrkBベクターと比較して、発現量が明らかに増大することが確認された。なお、全長TrkBベクターとは、AAV-CA-TrkBの構造と比較して、CA-TrkBをコードする遺伝子断片に代えて全長のTrkBをコードする遺伝子断片が挿入されたアデノ随伴ウイルスベクターである。
(方法)
=神経保護効果の評価法=
マウス(系統名 C57BL/6:チャールズリバー社より入手)に対して、視神経挫滅の14日前に、AAV-DsRed(対照群)またはAAV-CA-TrkBを、眼球内に2μL投与した。なお、AAV-DsRedとは、AAV-CA-TrkBの構造と比較して、CA-TrkBをコードする遺伝子断片に代えて蛍光タンパク質DsRedをコードする遺伝子断片が挿入されたアデノ随伴ウイルスベクターである。
マウス(系統名C57BL/6:チャールズリバー社より入手)に対して、視神経挫滅の14日前に、AAV-DsRed(対照群)またはAAV-CA-TrkBを、眼球内に2μL投与した。
=神経保護効果=
視神経挫滅14日後に、残存する網膜神経節細胞の数を定量化した。AAV-CA-TrkB投与群(1720 ± 72cells/mm2)では、AAV-DsRed投与群(664 ± 14 cells/mm2)と比較して、残存する網膜神経節細胞数が有意に増加していた(図5)。p<0.01(one-tailed Student’s t test)である。
視神経挫滅14日後にCTBでラベルされた再生軸索を計測した結果、AAV-CA-TrkB投与群ではAAV-DsRed投与群と比較して、再生軸索数及び伸長距離の両面において、顕著な改善が認められた(図6)。なお、図6に示すグラフのX軸は視神経挫滅部位からの距離(μm)、Y軸は軸索数を指す。p<0.05(one-tailed Student’s t test)である。
(方法)
緑内障モデルマウスであるGLASTノックアウトマウス(参考論文:Harada T, et al. The potential role of glutamate transporters in the pathogenesis of normal tension glaucoma. The Journal of Clinical Investigation 117: 1763-1770, 2007.)は、3-5週齢にかけて網膜神経節細胞が消失して視機能低下を引き起こす。10日齢のGLASTノックアウトマウスに、AAV-DsRed(対照群)またはAAV-CA-TrkBを、眼球内に1μL投与した(実施例2も参照)。次いで、12週齢に達したこれらノックアウトマウスの網膜の展開標本を作成し、残存する網膜神経節細胞の数をカウントした。網膜神経節細胞の検出にはRBPMS抗体(Merck Millipore社製のanti-RBPMS antibody)による免疫染色を用いた。RBPMSは、網膜神経節細胞のマーカーである。
図7に示すように、AAV-CA-TrkB投与群では、対照群と比較して、残存する網膜神経節細胞数が有意に増加していた。P<0.05(one-tailed Student’s t test)である。
(方法)
次に、CA-TrkBの以下の変異体(図8も参照)を発現するベクターを用いて、シグナル伝達の活性を検討した。
・「Cyto-TrkB」ベクター:TrkBの細胞内領域のN末端にmyc-タグを付加したタンパク質をコードするDNA断片を、実施例1のAAVベクターに組み込んだもの。実施例1のAAV-CA-TrkBと比較して、TrkBの細胞内領域のC末端にファルネシル化シグナル配列を付加していない点のみが異なる。
・「KD-TrkB」ベクター:実施例1のAAV-CA-TrkBと比較して、コードするタンパク質が、TrkBの細胞内領域に、そのキナーゼ活性を消失させる1アミノ酸変異(134番目のアミノ酸がリジンからアスパラギンに変異)が生じている点のみが異なる。
・「iSH-TrkB」ベクター:実施例1のAAV-CA-TrkBと比較して、コードするタンパク質が、TrkBの細胞内領域のN末端とmyc-タグとの間に、さらにp85のSH2配列を挿入したものである点のみで異なる。なお、SH2配列は、PI3Kの構成因子p110と複合体を形成するために必要な配列である。iSH-TrkBをコードする遺伝子断片(AAVベクターへ挿入した遺伝子断片の全長)の塩基配列を配列番号5、及び図10として示す。
図8に示すように、「Cyto-TrkB」ベクターおよび「KD-TrkB」ベクターをトランスフェクションした場合は、pERKおよびpAKTの増大が見られず、両シグナルの活性化は認められないことが判明した。一方、「iSH-TrkB」ベクターをトランスフェクションした場合は、AAV-CA-TrkBをトランスフェクションした場合(図中ではCA-TrkBと表示)と比較してAKT活性が顕著に増加する傾向を認めた。なお、図8中のコントロールは、実施例1のコントロールと同じものである。
(方法)
TrkA分子の細胞内領域を発現するAAVベクター(AAV-CA-TrkAと称する)を、実施例1の方法に従って作製した。すなわち、TrkBの細胞内領域に代えてTrkAの細胞内領域をコードする点以外はCA-TrkBをコードする遺伝子断片(実施例1参照)と同じ構造を持つ断片を用いて、実施例1に記載の方法に従いAAV-CA-TrkAを作製した(図9も参照)。
TrkAの細胞内領域のアミノ酸配列を配列番号6、TrkAの細胞内領域をコードする遺伝子(DNA)の塩基配列を配列番号7として示す。また、CA-TrkAをコードする遺伝子断片(AAVベクターへ挿入した遺伝子断片の全長)の塩基配列を配列番号8、及び図11として示す。
TrkCの細胞内領域のアミノ酸配列を配列番号9、TrkCの細胞内領域をコードする遺伝子(DNA)の塩基配列を配列番号10として示す。また、CA-TrkCをコードする遺伝子断片(AAVベクターへ挿入した遺伝子断片の全長)の塩基配列を配列番号11、及び図12として示す。
図9に示すように、AAV-CA-TrkAおよびAAV-CA-TrkCをトランスフェクションした場合は、pERKおよびpAKTが増大しており、両シグナルが活性化していることが認められた。なお、図9中のコントロールは、実施例1のコントロールと同じものである。
(方法)
成熟マウスに対して、実施例2の方法に従ってAAV-CA-TrkB投与群を得た。その14日後に視神経を挫滅し、挫滅の14日後および28日後に、脳の上丘に電極ポートを手術的に取り付け、フラッシュ刺激による視覚誘発電位(F-VEP:Flash-Visual Evoked Potential)を測定した。
図13に示すように、挫滅2週間後では、視覚誘発電位の反応が消失したが、挫滅4週間後では視覚誘発電位の発生が見られ、視機能の部分的な回復が確認された。
Claims (13)
- Trkの細胞内領域と膜局在化配列とを含む融合ポリペプチドをコードする核酸構築物。
- Trkの細胞内領域が以下の何れかから選択される、請求項1に記載の核酸構築物。
1)配列番号2、6、9、12、又は14の何れかに記載のアミノ酸配列で示されるポリペプチド、
2)配列番号2、6、9、12、又は14の何れかに記載のアミノ酸配列と90%以上の同一性を示すアミノ酸配列で示されるポリペプチド。 - 膜局在化配列は、アミノ酸の脂質修飾を誘導するものである、請求項1又は2に記載の核酸構築物。
- 膜局在化配列はファルネシル化配列である、請求項3に記載の核酸構築物。
- ベクターである、請求項1~4の何れか一項に記載の核酸構築物。
- アデノ随伴ウイルスベクターである、請求項5に記載の核酸構築物。
- 請求項1~6の何れか一項に記載の核酸構築物を含む医薬組成物。
- 神経関連疾患薬である、請求項7に記載の医薬組成物。
- 神経保護又は神経再生用である、請求項8に記載の医薬組成物。
- 請求項1に記載の核酸構築物が発現可能に導入されている、神経系細胞。
- 細胞内で、Trkの細胞内領域を、膜局在化配列を介して細胞膜に局在させる工程を含む、細胞活性化の方法。
- 上記工程は、請求項1~6の何れか一項に記載の核酸構築物、請求項7~9の何れか一項に記載の医薬組成物から選択される何れかを神経系細胞内で発現させることによって行う、請求項11に記載の方法。
- Trkの細胞内領域と膜局在化配列とを含む融合ポリペプチド。
