Abstract

検査用サンプル液の調製システムは、微生物分離装置(3)を有し、サンプル原液から検査用サンプル液を調製する。微生物分離装置(3)は、流体力分離装置(4)と、サンプル原液を流体力分離装置(4)へ供給する送液部(5)とを有する。流体力分離装置(4)は、矩形断面を有する湾曲流路を有し、湾曲流路を液体が流れることによって生じる渦流れを利用して、液体を第1画分と第2画分とに分離する。サンプル原液に含まれる粒子は、第1画分に濃縮される。効率的に微生物検査等の検査を行え、微生物の培養に供することが可能な検査用サンプル液を供給可能である。

Description

検査用サンプル液の調製システム及び検査用サンプル液の調製方法
 本開示は、液体に含まれる微生物等の検査を目的として、サンプル原液から検査に適した状態の検査用サンプル液を得るための検査用サンプル液の調製システム及び検査用サンプル液の調製方法に関する。
 一般に、医薬品や飲食品の製造においては、製品の安全性や品質を管理するために、微生物検査が行われ、酵母、カビなどの菌類や、大腸菌、レジオネラ菌などの細菌類の混入の有無が調べられる。十分な適正な製品管理を行うには、多数のサンプルを検査する必要がある。
 従来、微生物検査では培養法が用いられ、液体サンプルを用いて寒天培地による培養を行い、培地上のコロニー数を生存細菌数として計測している。しかし、培養に長時間を要する培養法では、迅速な計測は難しい。これに関して、近年、生菌の迅速な計測を目的として、様々な計測方法が開発され、その一つとして、流体中に分散する粒子を光学的に分析することで生菌数を計測する方法が提案されている(下記非特許文献1参照)。
 また、注射剤等の液体状の医薬製品については、液中に含まれる異物や微粒子の検査が行われており、日本薬局方において、光遮断粒子計測法又は顕微鏡係数法での検査が規定されている。注射剤などのように高価な液体の場合、検査で生じる製品ロスは問題であるため、これを削減するために、検査前のサンプルに含まれ得る粒子を濃縮して液体を回収することが考えられる。しかし、このような作業については、サンプルに加えられる遠心力や剪断力による粒子の破壊、乱流や泡立ちによる成分の変質などが生じる可能性が考えられる(下記非特許文献2参照)。
 流体に含まれる粒子の分離に関する文献として、下記特許文献1があり、湾曲チャネルを有する流体力学的分離デバイスが記載される。このデバイスでは、粒子を含む流体を湾曲チャネルに供給して、湾曲チャネルを流れる流体に作用する力を利用して粒子を分離する。

 非特許文献1に記載されるような計測方法においては、効率よく計測するために、測定の障害となる夾雑物の除去や、サンプルに含まれる菌体の濃縮を目的として、前処理を行う必要がある。例えば、飲食品に関するサンプルでは、原料由来の固形粒子等が含まれる場合は、これを除去する必要がある。このような前処理において、菌体に影響を与えることは好ましくなく、また、前処理中の微生物汚染は避けなければならない。従って、前処理は、無菌的に行うことが望まれる。異物検査のサンプルについても、粒子破壊や成分の変質を生じるような前処理では、正しい検査結果が得られなくなり、液体の回収も困難になる。
 このように、医薬品や飲食品等の製造においては、微生物検査や異物検査に供するサンプルに関するコスト増や効率低下の問題を解消する必要があり、検査用サンプルの前処理は重要な要素である。この点に関し、特許文献1に記載される分離技術は、固体粒子の分離に関するものであるが、微生物検査を目的とするサンプルを取得する場合には、微生物への影響については検討する必要がある。
 従って、効率的に検査を進行可能なサンプルを調製することは重要であり、製品製造の管理を行う検査に分離技術を適用するには、適正なサンプルが得られるかについて、十分に検討を重ねる必要がある。
 本開示は、効率的に検査を実施可能であり、微生物への影響の点でも問題がない検査用サンプル液を供給することができる検査用サンプル液の調製システム及び検査用サンプル液の調製方法を提供することを課題とする。
 上記課題を解決するために、サンプル原液中に粒子状に分散する微生物や固形物を分離する際に微生物に及ぶ影響について検討し、流体力分離技術を利用して、サンプル原液に含まれる微生物又は粒子状固形物を粒子の大きさによって効率的に分離して濃縮することにより、好適な検査用サンプル液を得ることが可能であることを見出した。
 本開示の一態様によれば、検査用サンプル液の調製システムは、微生物分離装置を有し、サンプル原液から検査用サンプル液を調製する検査用サンプル液の調製システムであって、前記微生物分離装置は、矩形断面を有する湾曲流路を有し、前記湾曲流路を液体が流れることによって生じる渦流れを利用して、前記液体を第1画分と第2画分とに分離する流体力分離装置と、前記サンプル原液を前記流体力分離装置へ供給する送液部とを有し、前記流体力分離装置によって、前記サンプル原液に含まれる粒子は第1画分に濃縮される。
 前記サンプル原液は、飲食品又は医療用薬品の製造における液状原料、液状中間物、又は、液状最終製品であってよい。
 前記送液部は、製造ラインから分岐して前記流体力分離装置に接続される供給路と、前記流体力分離装置へ前記サンプル原液を供給するための流動圧を前記サンプル原液に付勢するポンプとを有するとよく、製造ラインを流通する液状原料、液状中間物、又は、液状最終製品の一部が前記サンプル原液として前記流体力分離装置へ供給されるように構成することができる。前記送液部は、更に、前記第1画分及び前記第2画分のうちの一方を製造ラインへ返送する返送路を有してよい。或いは、前記送液部は、前記サンプル原液を収容する容器と、前記容器を前記流体力分離装置に接続する供給路と、前記流体力分離装置へ前記サンプル原液を供給するための流動圧を前記サンプル原液に付勢するポンプとを有してもよい。そして、前記送液部は、更に、前記第1画分及び前記第2画分のうちの一方を前記容器に返送する返送路を有すると好適である。前記送液部は、更に、前記第1画分及び前記第2画分のうちの他方の一部を前記供給路へ還流するための還流路を有してよい。
 上記検査用サンプル液の調製システムは、更に、追加の流体力分離装置を有し、前記送液部は、更に、前記流体力分離装置で分離した前記第1画分及び前記第2画分のうちの他方の残部を前記追加の流体力分離装置へ供給するための流路を有するように構成することができる。また、上記検査用サンプル液の調製システムは、更に、検査装置と、前記流体力分離装置を前記検査装置と接続する流路と、前記検査装置を製造ラインに接続する流路を有するように構成することができ、前記第1画分及び前記第2画分の他方は、前記検査装置を経て製造ラインに返送されるようにしてもよい。
 又、本開示の一態様によれば、検査用サンプル液の調製方法は、微生物分離処理を含み、サンプル原液から検査用サンプル液を調製する検査用サンプル液の調製方法であって、前記微生物分離処理は、矩形断面を有する湾曲流路を液体が流れることによって生じる渦流れを利用して、前記液体を第1画分と第2画分とに分離する流体力分離工程と、前記サンプル原液を前記流体力分離工程へ供給する送液工程とを有し、前記流体力分離工程によって、前記サンプル原液に含まれる粒子は第1画分に濃縮される。
 前記粒子は、ウイルス、真正細菌、古細菌、菌類、粘菌類、藻類及び原生動物のうちの少なくとも一種の微生物であってよく、前記第1画分は検査用サンプル液として供給され、返送路によって、前記第2画分を前記流体力分離装置から返送することができる。また、前記粒子は、微生物より大きい粒子状固形物であってもよく、前記第2画分は検査用サンプル液として提供するとよい。その際、前記第1画分は、返送路によって返送されるか、又は、システム外に排出するとよい。
 本開示によれば、液体中に粒子状に分散する微生物又は粒子状固形物を効率的に分離すると共に、分離後の液体を検査用サンプル液として用いて、サンプル原液に含まれ得る微生物等の検査を効率的に行うことができる。また、連続的且つ無菌的に分離操作を行えるので、検査用サンプル液の調製における汚染が防止される。故に、効率的に検査用サンプル液の調製を実施可能な検査用サンプル液の調製システム及び検査用サンプル液の調製方法が提供され、製品の安全性や品質の管理を効率的に実施することが可能である。

 本開示の実施形態について、図面を参照して、以下に詳細に説明する。尚、実施形態において示す寸法、材料、その他の具体的な数値等は、内容の理解を容易とするための記載であって、特に断る場合を除き、本開示を限定するものではない。又、本願明細書及び図面において、実質的に同一の機能及び構成を有する要素については、同一の符号を付することにより重複説明を省略し、本開示に直接関係のない要素は、図示を省略する。
 飲食品又は医療用薬品の製造においては、製品の安全性や品質を管理するために、微生物検査や異物検査などが実施される。微生物検査においては、サンプル液に含まれる微生物数の計測によって微生物汚染の有無を確認する。微生物数の計測は、流体中に分散する粒子を光学的に分析することで、生菌数を迅速に計測することが可能になっている。このような計測を効率的に行うには、サンプル原液中に含まれ得る微生物が濃縮されたサンプル液を調製することが有効である。また、サンプル原液に、微生物より大きい粒子状固形物が含まれる場合、このようなものを夾雑物として除去した検査用サンプル液を調製すると、正確な計測を行う上で有効である。更に、異物検査においては、液中に含まれ得る粒子が濃縮されるようなサンプル液の調製を行うと、残部のサンプル原液は回収できるので、異物検査において異常がない場合には再利用が可能になる。従って、液体中に含まれる粒子の濃縮、及び、大きさによる分離は、検査用サンプル液の調製において重要である。
 液体中に含まれる粒子を分離する分離技術の一つに、流体に発生するディーン渦の作用を利用するものがある(前記特許文献1参照。以下、この技術を流体力分離と称する)。これは、流れ方向に垂直な断面が矩形であって一側に湾曲した湾曲流路を流れる液体にディーン渦が発生することによって、液体中の粒子の分布に偏りが生じることを利用した分離技術である。湾曲流路を流れる粒子は、その大きさによって流路における分布が異なる変化をする(前記特許文献1参照)。具体的には、流路の断面において輪を描くような粒子分布が形成されて、粒子は螺旋状に流路を流れ、この際、相対的に大きい粒子が輪の外側に、相対的に小さい粒子が内側に位置する。更に、所定の分離条件に設定することにより、粒子の分布形態は更に変化し、一定の大きさを超える粒子が流路の外周側へ収束する。
 本開示においては、液体に含まれる粒子、つまり、粒子状の微生物又は固形物の分離に上述の流体力分離を適用し、所定の大きさ以上の微生物粒子又は固形物粒子が湾曲流路の外周側へ収束する分離形態を利用する。これにより、製品製造における液状原料、液状中間物、又は、液状最終製品をサンプル原液として、検査用サンプル液を調製する。本開示の検査用サンプル液の調製システムにおいては、比較的高い流速で液体を湾曲流路に供給する。供給される液体が湾曲流路を流れる間に、相対的に小さい粒子は、前述したように、流路の断面において輪を描くように分布するが、相対的に大きい粒子は、湾曲流路の外側(外周側)に遍在するように収束する。つまり、相対的に大きい粒子が濃縮した液体を第1画分として分取することができる。従って、サンプル原液に微生物又は固形物の粒子が含まれる時は、粒子が集中する第1画分と残部の第2画分とに分離することによって、第1画分を微生物検査や異物検査のサンプル液として利用することができる。検査において異常が認められなければ、残部の第2画分を回収して再利用することができる。従って、注射液等の高価な医療用薬品の製造におけるサンプル原液について、検査によるロスの削減が可能になる。流体力分離において、微生物は、粒子としての大きさによって分離される。つまり、微生物は、粒子状の微生物として分離され、微生物の単一細胞、及び、複数の細胞が凝集した粒子の何れでもよい。固形物粒子は、有機物粒子及び無機物粒子の何れであってもよく、プラスチック、エラストマー、繊維、セラミックス、金属等の固形物粒子を好適に分離することができる。
 飲食品製造におけるサンプル原液は、検査対象である微生物より大きい粒子状固形物を含む場合がある。つまり、サンプル原液に含まれる粒子は、相対的に大きい粒子状固形物と、相対的に小さい微生物粒子とを含み得る。例えば、液状原料として、しょうゆなどの調味液を用いる場合、原料由来の微細粒子や成分の凝集物が含まれ得る。製造途中の液状中間物についても同様である。液状最終製品である茶飲料や果実飲料等には、茶葉粒子や果実粒子等の粒子状固形物が含まれる。このような場合、流体力分離において、相対的に大きい粒子である粒子状固形物は、第1画分に濃縮され、残部の第2画分には、相対的に小さい粒子の大部分が含まれる。従って、粒子状固形物が除去された第2画分を検査用サンプル液として微生物検査に利用して、相対的に小さい粒子である微生物粒子を検出することができる。検査において異常が認められなければ、第1画分は、回収及び再利用してもよい。この場合の第2画分は、サンプル原液に微生物が含まれる時は、死細胞片(デブリ)等を含み得る。検査用サンプル液からデブリ等を除去するには、分離状態を調整して更に流体力分離を繰り返すとよい。これにより、相対的に大きな粒子である微生物と相対的に小さい粒子であるデブリとを分別することが可能である。流体力分離に供給する液体の流速(流量)、及び、湾曲流路における断面の大きさの設定によって、濃縮する粒子の大きさを調整することができる。
 微生物は、活性が高い状態においては大きく生育するが、活性が低下すると、比較的小さい状態で死滅し分解する。微生物において相対的に小さい粒子の多くは、微生物の死骸又は死骸片であり、DNA合成途中の活性な細胞が含まれる割合は少ない。流体力分離によって分離される粒子の大きさは、分離条件(液体の供給流量)を変更することによって調整することができるので、異なる分離条件で流体力分離を繰り返して活性な微生物とデブリ等を分離できる。分離条件を変更しながら流体力分離を繰り返す多段階の流体力分離を行うと、異なる大きさの粒子を各段階で分取することができる。或いは、一定の分離条件で流体力分離を繰り返すことによって濃縮度及び分離精度を高めることができる。
 流体力分離は、流路内の流れが層流であり、液体に含まれる粒子の破壊は比較的生じ難い。従って、異物検査用のサンプル液の調製において非常に有効である。但し、効率的な微生物検査を継続するために、また、培養の必要が生じた場合への対応などを考慮すると、サンプル液の調製中の微生物へのダメージの有無は検討すべき点である。この点について検討したところ、酵母や菌等の微生物は、比較的高圧下でも耐性を有し、例えば、1MPa程度の加圧供給においても生存率は維持される。更に、微生物は、圧力変動に対してもかなり耐久性が高い。培養した動物細胞の場合は、分離中に細胞に加わる圧力の変動(圧力低下)による影響を受けて細胞の生存率が減少し易いが、微生物は、流体力分離においてかなりの圧力変動を受けても生存率を維持することができる。このため、微生物粒子の流体力分離において、圧力環境の制御は特に行わなくてよい。従って、流体力分離装置への液体供給に使用するポンプ等の駆動力を変更して流動圧を変動させることも許容されるので、流動圧に基づいて液体の流量を任意に調整できる。つまり、ポンプの駆動制御によって分離条件の設定変更を容易に行うことができるので、システム構成の簡素化が容易である。また、流体力分離は、液体の無菌保持を行い易く、操作者の熟練度に関係なく、再現性の高い分離が可能であるという利点もある。
 検査対象とする微生物について圧力環境による影響が懸念される場合は、流体力分離において微生物に加わる圧力の変動を制限するとよい。その場合、分離条件として、分離中に細胞に加わる圧力の変動(圧力低下)が一定レベルを超えないように、湾曲流路へ液体を供給する圧力環境が制御される。つまり、分離中に微生物に加わる圧力の変動(入口圧力と出口圧力との差)が所定値以下となるように液体の供給が制御される。具体的には、圧力変動(圧力差)が0.60MPa未満になるように制御すると適切であり、好ましくは0.45MPa以下、より好ましくは0.40MPa以下となるように設定するとよい。上記のように圧力変動が抑制された分離条件においては、微生物のダメージは少なく、微生物の生存率は98%程度以上に維持することができる。従って、濃縮分離された大きい微生物粒子を分別し、増殖能等を維持した状態で、粒径分布の測定や、微生物の培養及び検査等に供給することができる。
 以下に、検査用サンプル液の調製システムの構成について、図面に示す実施形態を参照して説明する。図1の検査用サンプル液の調製システム1は、微生物分離装置3を有し、これを用いて、サンプル原液Lから検査用サンプル液が調製される。微生物分離装置3は、流体力分離装置4と、サンプル原液Lを流体力分離装置4へ供給する送液部5とを有する。流体力分離装置4は、矩形断面を有する湾曲流路を有し、湾曲流路を液体が流れることによって生じる渦流れを利用して、液体を第1画分と第2画分とに分離する。流体力分離装置4によって、サンプル原液に含まれる粒子は、第1画分に濃縮される。
 図1の実施形態において、サンプル原液Lは、製造ラインPLから分取される。つまり、送液部5は、製造ラインPLから分岐して流体力分離装置4に接続される配管で構成される供給路6と、ポンプ10とを有する。ポンプ10は、流体力分離装置4へサンプル原液Lを供給するための流動圧をサンプル原液Lに付勢する。製造ラインPLを流通する液状原料、液状中間物、又は、液状最終製品の一部が、サンプル原液Lとして流体力分離装置4へ供給される。
 流体力分離装置4は、流れ方向に垂直な一定の矩形断面を有する湾曲流路を内部に有する。湾曲流路は、一端においてサンプル原液Lを取り入れる単一の導入口41を有し、湾曲流路の他端は、サンプル原液を分離して排出する少なくとも2つの導出口42,43を有する。流体力分離装置4は、液体が湾曲流路を流れることによって一方向に旋回する液体に生じる渦流れを利用して、サンプル原液Lに含まれる粒子から相対的に大きい粒子を流路の外側(外周側)に遍在させる。従って、湾曲流路から排出される液体を、外側の画分と内側の画分とに分割することによって、外側の画分として、相対的に大きい粒子が濃縮された液体を分離することができる。1つの導出口42から相対的に大きい粒子が濃縮して含まれる液体La(第1画分)が、導出口42に接続される流路7を通じて排出される。もう1つの導出口43に接続される流路8から、相対的に小さい微生物粒子が含まれる残部の液体Lb(第2画分)が排出される。
 従って、サンプル原液Lが、検査対象の微生物より大きい粒子を含まないものであると、微生物粒子を第1画分に濃縮して、液体Laとして導出口42から得ることができる。これを検査用サンプル液として、微生物検査に使用して微生物の有無を検査することができる。必要に応じて、微生物粒子の個数計測、粒径分布、粒子形状の計測などを実施することができる。
 湾曲流路の湾曲形状は、略円周状、略円弧(部分円周)状、螺旋状等が挙げられ、これらの形状の何れであってもよい。流体力分離装置4は、1つの湾曲流路を有する流路ユニットを構成単位として設計することができる。具体的には、流路ユニットとして、内部に1つの湾曲流路が形成された平層状の成形体をプラスチック等で形成し、その際に、湾曲流路の両末端が成形体の端面に開口して1つの導入口と少なくとも2つの導出口を有するように構成する。このような成形体を流路ユニットとして用いて、1つの流路ユニット、又は、複数の流路ユニットの組み合わせによって流体力分離装置を構成することができる。複数の流路ユニットを積層して並列状の流路を構成することによって、サンプル原液Lの処理流量を高めることができる。
 流体力分離装置4における粒子の分離効率は、湾曲流路に供給される液体におけるDe数及び圧力によって変化し、好適な分離が可能なDe数及び圧力の適正な範囲が存在する。ディーン数は、式:De=Re(D/2Rc)1/2、によって表される(Re:レイノルズ数(-)、D:代表長さ(m)、Rc:流路の旋回半径(m))。De数は、液体の流速に比例するので、流体力分離装置に供給される液体の流速を適正に制御することによって、好適な粒子の分離が実施される。概して、De数が30以上且つ100以下であることが好ましく、50~80程度であると更に好ましい。従って、このような範囲のDe数になるように液体の流速(流量)が設定される。分離する粒子の大きさに対応して、流体力分離装置4へ供給される液体の流量を調整するとよい。
 検査用サンプル液の調製システム1の送液部5は、流体力分離装置4へサンプル原液Lを供給するための流動圧をサンプル原液Lに付勢する付勢装置、具体的には、ポンプ10を有する。ポンプ10が付与する流動圧によって、サンプル原液Lが流体力分離装置4へ供給される流量及び流圧は変化する。微生物の検査を行う上で、微生物の生存率は重要な要素である。この点に関して、流体力分離装置4による分離に対して、微生物は耐久性を有し、生存率を維持することができる。但し、圧力変動による影響を排除することが望まれる場合には、例えば、ポンプ10の駆動制御によって、好適な圧力環境に調整することができる。分離中の微生物における生存率の低下やダメージを抑制するには、圧力差が0.60MPa未満になるように圧力環境を制御するとよく、好ましくは、0.45MPa以下、より好ましくは0.40MPa以下になるように制御される。圧力制御弁及び流量調整弁を設けると、サンプル原液の供給圧力及び供給流量を任意に調整することができ、圧力計及び流量計を用いて圧力及び流量を監視するとよい。
 図1の検査用サンプル液の調製システム1においては、流量調整弁を用いずにポンプ10の駆動制御によって、流体力分離装置4へ供給する液体の流量を適正に調整する。液体の供給流量及び圧力は、流体力分離装置4における処理能力(流路断面の大きさ及び流路数)に基づいて供給路6及び流路7,8の寸法を適正に設計することによって、ある程度規制できる。従って、図1の検査用サンプル液の調製システム1のような簡略な構成でも、微生物検査に対応した粒子の分離によって検査用サンプルを効率的かつ継続的に調製することができる。
 検査用サンプル液の調製において、微生物への影響を可能な限り取り除く場合、微生物に剪断力を加えない方式のポンプを使用するとよい。具体的には、往復動又は回転動による容積変化を利用して一定容積の液を押し出す容積式ポンプを使用すると好適である。容積式ポンプとしては、例えば、ピストンポンプ、プランジャポンプ、ダイヤフラムポンプ、ウィングポンプ等の往復ポンプ、歯車ポンプ、ベーンポンプ、ねじポンプ等の回転ポンプが挙げられる。一実施形態として、コンプレッサを付設した加圧タンクを利用するものが挙げられ、加圧タンクに収容した液体にコンプレッサで加圧することによって、加圧タンクから流体力分離装置へ液体を圧送することができる。
 検査用サンプル液の調製システムの送液部は、更に、第1画分及び前記第2画分のうちの一方を製造ラインへ返送する返送路を有してもよい。図1の検査用サンプル液の調製システム1においては、流路8から分岐して製造ラインに接続される返送路8aを有し、導出口43から第2画分として排出される残部の液体Lbを製造ラインに返送可能なように構成される。返送路8aの分岐点には切り替え弁9が設けられ、第2画分について、系外への排出又は製造ラインへの返送の何れかを選択できる。従って、これを利用した回収によって、検査によるロスを削減することができる。第1画分に微生物が含まれる場合、流体力分離によって分取される第2画分は、相対的に小さい微細物細胞やデブリ等を含んでいる可能性があり、検査で異常が認められる時の第2画分は廃棄するとよい。サンプル原液に高価な成分や希少な物質が含まれる場合は、フィルターによってデブリや凝集物を除去し、精製処理を行うことによって所望の成分を回収することができる。フィルターは、除去対象に応じて、適切な孔径のものを選択すればよく、例えば、精密濾過膜、限外濾過膜等が挙げられる。
 検査用サンプル液の調製システム1において、サンプル原液から検査用サンプル液を調製する検査用サンプル液の調製方法が実施され、この調整方法は、微生物分離処理を含む。微生物分離処理は、流体力分離工程と、サンプル原液を前記流体力分離工程へ供給する送液工程とを有する。流体力分離工程では、矩形断面を有する湾曲流路を液体が流れることによって生じる渦流れを利用して、液体を第1画分と第2画分とに分離する。これにより、サンプル原液に含まれる粒子は、第1画分に濃縮される。サンプル原液として、検査対象の微生物より大きい粒子を含まないものを用いることにより、第1画分として、微生物が濃縮された液体を得ることができる。これを検査用サンプル液として、微生物検査に使用することができ、微生物の有無を確認できる。必要に応じて、微生物粒子の個数計測、粒径分布、粒子形状の計測などを併せて実施することができる。
 つまり、上記の流体力分離における分離対象となる粒子は、微生物粒子であり、第1画分は検査用サンプル液として供給され、第2画分は、流体力分離装置から系外に排出されるか、或いは、返送路を通じて製造ラインPLに返送される。0.1μm程度~数mm程度の微生物粒子を好適に分離でき、分離条件によって、これを超えるものの分離も可能である。ウイルス、真正細菌、古細菌、菌類、粘菌類、藻類及び原生動物のうちの少なくとも一種の微生物を検査対象とすることができる。
 流体力分離工程において、サンプル原液は、矩形断面を有する湾曲流路へ単一の導入口から導入され、均一な状態で湾曲流路に供給される。流体力分離装置の湾曲流路は、流れ方向に垂直な断面(径方向断面)が矩形の流路である。均一な液体が湾曲流路を流れる間に、ディーン渦に載って、矩形断面において輪を描くように分布が変化する一方、粒子には、流路の外周側に滞留する揚力が作用して、分布が外周側に集中する。湾曲流路の末端出口は、外周側に位置する導出口42と内周側の導出口43に二分割される。外周側の導出口42から、粒子が濃縮して含まれる液体が排出され、内周側の導出口43から、残部の液体が排出される。流体力分離工程における分離条件は、送液工程における流量の調製によって変更することも可能であり、分離対象の粒子の大きさに応じて適切な流量を設定するとよい。
 図2の検査用サンプル液の調製システム1Aは、流体力分離によって得られる第2画分を検査用サンプル液として供給するように図1の検査用サンプル液の調製システムを変形した一例である。従って、製造ラインPLに液体を返送する返送路7aは、第1画分を排出する流路7から分岐して製造ラインPLに接続するように構成される。返送路7aの分岐点には、同様に、切り替え弁9が設けられ、系外への排出と製造ラインPLへの返送の何れかを選択できる。
 図1では、サンプル原液に含まれ得る粒子は、微生物粒子である。これに対し、図2の検査用サンプル液の調製システム1Aにおいて実施される検査用サンプル液の調製方法では、サンプル原液として、微生物より大きい粒子状固形物を含有するものが用いられる。つまり、流体力分離における分離対象となる粒子は、微生物より大きい粒子状固形物であり、第1画分として分離される。第1画分は、返送路7aによって返送されるか、又は、システム外に排出される。例えば、粒子状固形物が、検査の障害となる夾雑物やごみなどである場合は、粒子状固形物を含む液体Laは、流路7から排出される。粒子状固形物が、飲食品の原料粒子など、製品として許容される成分であれば、製造ラインPLへ返送してもよい。サンプル原液に微生物が含まれる時は、相対的に小さい粒子である微生物の大部分は、第2画分に含まれる。従って、粒子状固形物が除去された第2画分を検査用サンプル液として用いれば、微生物が検出される。
 図3の検査用サンプル液の調製システム1Bは、容器に収容されるサンプル原液を用いて検査用サンプル液を調製するように図1の検査用サンプル液の調製システムを変形した一例である。つまり、検査用サンプル液の調製システム1Bの送液部5は、サンプル原液Lを収容する容器2と、容器2を流体力分離装置4に接続する供給路6と、流体力分離装置4へサンプル原液Lを供給するための流動圧をサンプル原液に付勢するポンプ10とを有する。サンプル原液Lが、検査対象の微生物より大きい粒子を含まないものであれば、微生物粒子は、第1画分に濃縮され、液体Laとして導出口42及び流路7を通じて得られる。従って、液体Laを検査用サンプル液として、微生物検査に使用することで、微生物の有無を確認できる。必要に応じて、微生物粒子の個数計測、粒径分布、粒子形状の計測などを実施することができる。
 検査用サンプル液の調製システム1Bの送液部5は、更に、第1画分及び第2画分のうちの一方を容器2に返送する返送路を有してもよい。図3の実施形態においては、流路8から分岐する返送路8aが容器2と接続される。つまり、導出口43から第2画分として排出される残部の液体Lbを容器2に返送可能なように構成される。返送路8aの分岐点には切り替え弁9が設けられ、第2画分について、系外への排出又は容器2への返送の何れかを選択できる。従って、第2画分の回収及び再利用が可能であり、これを利用して、検査によるロスを削減することができる。
 容器2は、微生物汚染を防止可能な容器であり、必要に応じて、ヒーター又は冷却器、及び、温度調節機能が装備されたものを使用してもよい。内部の液体は、サンプル原液の保管に適した温度に維持される。容器2は、液体を均一化するための攪拌装置を備えてもよく、微生物を損傷しない適切な速度で攪拌することができる。又、必要に応じて、微生物に適した環境に調整するために、酸素/二酸化炭素/空気の量、pH、導電率、光量等の調整機能を備えたものを利用してもよい。
 図3の検査用サンプル液の調製システム1Bにおいて実施可能な検査用サンプル液の調製方法は、製造ラインPLではなく容器2へ第2画分を返送する点以外は、図1の検査用サンプル液の調製システム1と同様である。つまり、サンプル原液として、検査対象の微生物より大きい粒子を含まないものを用い、微生物が存在する時は、第1画分に濃縮される。従って、第1画分を検査用サンプル液として、微生物検査に使用することができ、微生物の有無を確認できる。必要に応じて、微生物粒子の個数計測、粒径分布、粒子形状の計測などを実施することができる。ウイルス、真正細菌、古細菌、菌類、粘菌類、藻類及び原生動物のうちの少なくとも一種の微生物を検査対象とすることができる。第2画分は、流体力分離装置から系外に排出されるか、或いは、返送路を通じて容器2に返送される。検査対象が、ウイルスなどの非常に小さい粒子である場合、容器2中のサンプル原液に凝集剤を添加して、流体力分離を実施し易い大きさに凝集させてもよい。その場合、得られる第1画分における凝集を解消したものを、検査用サンプル液として供給するとよい。
 容器2からサンプル原液Lを供給する構成は、製造ラインから直接抜き取ったものについて、希釈又はろ過分離等を行って使用する場合に適している。また、培養した微生物を用いた対照試験等として検査を行う場合に利用して、サンプル液を調製してもよい。
 図4の検査用サンプル液の調製システム1Cは、流体力分離によって得られる第2画分を検査用サンプル液として供給するように図3の検査用サンプル液の調製システムを変形した一例である。従って、容器2に液体を返送する返送路7aは、第1画分を排出する流路7から分岐して容器2に接続するように構成される。返送路7aの分岐点には、同様に、切り替え弁9が設けられ、系外への排出と製造ラインPLへの返送の何れかを選択できる。
 図4の検査用サンプル液の調製システム1Cにおいて実施される検査用サンプル液の調製方法は、図2の検査用サンプル液の調製システム1Bと同様である。サンプル原液として、検査対象の微生物より大きい粒子状固形物を含有するものが用いられる。従って、流体力分離における分離対象となる粒子は、相対的に大きい粒子、つまり、微生物より大きい粒子状固形物であり、第1画分として分離される。第1画分は、導出口42から返送路7aを通じて返送されるか、又は、システム外に排出される。例えば、粒子状固形物が検査の障害となる夾雑物やごみなどである場合は、粒子状固形物を含む液体Laは、流路7から排出される。この場合、容器2中のサンプル原液に凝集剤を添加して、粒子状固形物を凝集させてもよく、これにより、流体力分離を行い易くすることができる。或いは、粒子固形物が、飲食品の原料粒子など、製品として許容される成分であれば、容器2へ返送して利用してもよい。相対的に小さい粒子である微生物の大部分は、第2画分に含まれるので、流体力分離装置4の導出口43から流路8を通じて排出される第2画分は、検査用サンプル液として提供される。
 図1の検査用サンプル液の調製システム1において、第1画分及び前記第2画分のうちの一方(つまり、第2画分)は、製造ラインPLに返送可能であり、他方はそのまま検査用サンプル液として回収される。これに対し、図5の検査用サンプル液の調製システム1Dは、更に、第1画分及び前記第2画分のうちの他方(つまり、第1画分)を供給路6へ還流するための還流路11を有するように構成した一例である。還流路11は、流路7から分岐して、供給路6におけるポンプ10の上流側に接続される。従って、第1画分の一部は、供給路6に返送されて流体力分離が繰り返されるので、流路7から排出される第1画分における粒子の濃縮が次第に増加する。つまり、粒子の濃縮度を高めることができる。従って、サンプル原液に含まれる粒子が極めて少なく、検査において計測誤差との区別が難しいような時に、濃縮度を高めて計測をし易くすることができる。つまり、検査における検出感度の向上に寄与する。図5の検査用サンプル液の調製システム1Dの構成は、図3の検査用サンプル液の調製システム1Bに適用してもよく、容器に収容されるサンプル原液から得られる第1画分を部分的に還流して濃縮度を高めることができる。
 図6の検査用サンプル液の調製システム1Eは、図5の検査用サンプル液の調製システム1Dが、更に、追加の流体力分離装置4aを有するように変形した一例である。つまり、二段階の流体力分離を実施可能なように構成される。図5では、第1画分及び前記第2画分のうちの他方(つまり、第1画分)の一部を供給路6へ還流するが、その残部は、そのまま検査用サンプル液として供給される。これに対し、図6においては、流路7は、追加の流体力分離装置4aに接続され、第1画分の残部を追加の流体力分離装置へ供給する。つまり、追加の流体力分離装置4aを用いて二段目の流体力分離を行うので、第1画分における粒子の濃縮度及び分離精度を高めるのに有効である。流体力分離装置4aの導出口42aから第1画分として得られる液体Lcは、流路12を通じて検査用サンプル液として供給される。
 検査用サンプル液の調製システム1Eにおいて、流体力分離装置4aの導出口43aに接続される流路13は、第2画分として液体Ldを排出する。更に、流路13から分岐して製造ラインPLに接続する返送路13aが設けられ、第2画分の液体Ldを製造ラインPLへ返送可能なように構成される。返送路13aの分岐点には、切り替え弁9aが設けられ、同様に、系外への排出と製造ラインPLへの返送の何れかを選択できる。図6の検査用サンプル液の調製システム1Eの構成は、図3の検査用サンプル液の調製システム1Bに適用して、容器に収容されるサンプル原液から得られる第1画分について、多段階の流体力分離を行うようにしてもよい。
 図6の検査用サンプル液の調製システム1Eでは、2段階の分離を行うことによって、液体に含まれる成分が3つに分割される。2段目の流体力分離装置4aにおける分離条件は、流路7を通じて供給される液体Laの供給流量の制御によって調整可能であるので、流路7にポンプを設けて分離条件を調整してもよい。2つの流体力分離装置4,4aにおける分離条件を適切に設定することによって、粒子の分離精度の改善及び濃縮度の向上が可能である。尚、1段目の流体力分離装置における2つの画分の分割比率に応じて、2段目の流体力分離装置の処理能力が適切に設定される。
 尚、図6の検査用サンプル液の調製システム1Eで実施されるような多段階の流体力分離は、図5の検査用サンプル液の調製システム1Dを用いて実施することも可能である。この場合、還流路11の分岐点及び還流点に切り替え弁を設けて、第1画分の全量について、系外への排出又は還流の何れかに切り替えられるように構成する。その際、一時収容用の容器を還流路11に設けるとよい。流体力分離装置4から排出される第1画分を容器に収容して流体力分離の分離条件を変更し、第1画分を再び流体力分離装置4に供給することによって、2段目の流体力分離を行うことができる。分離条件の変更は、例えば、ポンプの駆動力の調整によって可能である。このように、分離条件を段階的に変更しながら、分離操作を繰り返すことによって、多段階分離を行うことができる。
 図7の検査用サンプル液の調製システム1Fは、図5の検査用サンプル液の調製システム1Eを検査装置に接続した例である。つまり、検査用サンプル液を、直接検査装置に供給可能である。流体力分離装置によって分離される第1画分及び前記第2画分は、一方を製造ラインPLへ返送し、他方を検査用サンプル液として供給することができるが、図7においては、第1画分を検査装置に供給する。更に、検査装置を経て製造ラインに返送するように構成されている。
 具体的には、検査用サンプル液の調製システム1Fは、検査装置14と、流体力分離装置4を検査装置14に接続する流路7とを有する。流路7は、流体力分離装置4の導出口42と検査装置14とを接続し、第1画分として排出される液体が、検査用サンプル液として検査装置14に供給される。検査用サンプル液の調製システム1Fは、更に、検査装置14を製造ラインPLに接続する流路を有する。つまり、検査装置から検査後のサンプル液を排出する流路15は分岐して、返送路15aが設けられ、その分岐点に切り替え弁16が設けられる。従って、検査後のサンプル液について、系外への排出と製造ラインPLへの返送の何れかを選択でき、切り替え弁16によって変更できる。
 図7のように、流体力分離装置を検査装置と配管で直接接続することによって、サンプル原液から検査用サンプル液を調製して検査に供給するまでの汚染を防止することができる。従って、図1~図6の検査用サンプル液の調製システムも、同様に、流体力分離装置を検査装置と接続することによって、検査用サンプル液の調製及び検査を無菌的に行うことができる。
 図7のシステムを用いて、製造ラインからのサンプル原液の抜き取り及び検査を連続的に行うことができる。検査において異常が認められない時は、サンプル原液由来の液体を製造ラインへ返送することができる。異常が認められる時は、サンプル原液由来の液体を全て系外に排出するように切り替え弁9,16を制御するとよい。また、製造ラインPLから分岐する供給路6に開閉弁を設けて、サンプル原液の抜き取りを任意に行えるように構成してもよく、これは、断続的な抜き取りによる定期的な検査を実施する場合に好適である。
 前述の式で示したように、De数は、湾曲流路の旋回半径Rc及び流路の断面寸法によって変化する(前述の式における代表長さDは、湾曲流路の幅と見なすことができる)。従って、湾曲流路の設計に基づいてDe数が好適な値になるように調整することができ、それによって、流体力分離装置は、粒子の濃縮分離を良好な分離効率で実施することができる。又、流体の流量は、湾曲流路の断面の幅(径方向)及び高さの何れかの設定によって調節可能であるので、湾曲流路の設計に基づいて、所望の流量で粒子の分離を実施可能なように流体力分離装置を構成することができる。従って、湾曲流路の設計は、分離対象の条件(微生物の寸法分布、液体の粘度等)に応じて、好適な分離が可能なように適宜変更可能である。微生物粒子の分離効率の観点においては、湾曲流路が、アスペクト比(幅/高さ)が10以上の長方形の断面を有する流路であると好適である。このような湾曲流路に、液体を100~500mL/分程度の流量で供給することによって、分離が良好に進行し、50億~250億細胞/分程度の効率で微生物粒子の濃縮分離を行うことができる。この場合、粒径が10μm程度以上の微生物粒子を、外周側の画分として濃縮分取することができる。5μm程度以下の相対的に小さい微生物粒子は、輪状に分布して、その大部分は内周側の画分として分取される。湾曲流路の末端出口の設計(導出口の分割位置)によって、外周側の画分に含まれる粒子の大きさ及び分離精度を調整することができるので、外周側の画分に濃縮される微生物粒子の大きさの下限を10μmより小さくすることも可能である。図5~図7のように、必要に応じて流体力分離工程を繰り返すことで、粒子の濃縮度及び分離精度を高めることができる。
 流体力分離装置における粒子の分離状態(濃縮する粒子の大きさ、分取比率)は、出口における導出口の分割位置によって異なる。概して、外周側画分/内周側画分の分割比率(容積比)が、10/90~70/30程度になるように流路出口における断面積比を設計すると、上述のような粒子の濃縮分離に好適である。図1~図7の検査用サンプル液の調製システムにおいて、流体力分離装置は、湾曲流路の出口として、2つの導出口を有するが、3つ以上の導出口に分割してもよい。末端出口を3つ以上の導出口に分割した場合、分取する画分の量を状況に応じて変更可能なように構成してもよい。
 上述のような検査用サンプル液の調製システムを用いて、様々なサンプル原液から検査用差プル益を調製することができる。サンプル原液として使用可能なものとして、具体的には、医薬医療品や化学製品の製造における液状原料、液状中間物、及び、液状最終製品、微生物又は細胞の培養液、飲料水、製造用水、食品から採取した食品サンプルを含む液等が挙げられる。製造用水には、医療医薬品製造用水、飲食品製造用水、産業製品の製造で使用される洗浄水などが含まれる。液状最終製品には、例えば、清涼飲料水、乳酸菌飲料、乳、ビールや日本酒、蒸留酒等の酒類が挙げられ、産業製品の洗浄水として、電子基板、半導体、精密機器、医療機器などの製造工程で使用されるものがある。
 本開示の検査用サンプル液の調製システムでは、流体力分離において液体に含まれる成分への影響は小さいので、変質し易い物質の検査に適用することも可能である。例えば、細胞培養を行った液体中に含まれる抗体等のような糖タンパクは、脆弱な長鎖高分子物質であるが、流体力分離を利用して検査用サンプル液を調製することができる。具体的には、培養液に含まれる細胞を、流体力分離によって第1画分に濃縮して分離除去し、第2画分について検査や測定を行うことができる。従って、例えば図4の検査用サンプル液の調製システムを用いて、好適に検査用サンプル液を調製できる。
 微生物には、原核生物(真正細菌及び古細菌)、真核生物(藻類、原生生物、菌類、粘菌類)及びウイルスがある。具体的には、原核生物としては、大腸菌、枯草菌、黄色ブドウ球菌等の細菌や、放線菌、ラン藻、メタン生成菌等の枯草菌が挙げられる。真核生物としては、アオミドロ、ワカメ等の藻類、ゾウリムシ等の原生生物、カビ、酵母、キノコの真菌類などが挙げられる。また、原生生物としては、アメーバなどもあり、ワムシなどの輪形動物や、ボツリオコッカス・ブラウニーなどの微細藻類も微生物に含まれる。
 流体力分離は、微生物を培養した液体培地に適用可能である。液体培地は、合成培地、半合成培地、天然培地の何れの液体培地であってよく、栄養素、検査を目的とする鑑別剤(pH指示薬、酵素基質、糖類等)、目的外微生物の発育を抑制する選択剤などを含んでいてもよい。流体力分離を効率的に進行させるために、粘性があまり高くない液体培地を使用すると好ましい。また、ウイルス等の粒径が小さい微生物粒子について、流体分離を行い易い大きさの粒子を形成するために、凝集剤を添加してもよい。凝集剤として、例えば、高分子凝集剤、及び、ポリ塩化アルミニウム、硫酸アルミニウム、ポリシリカ鉄等の無機凝集剤が挙げられる。
 検査装置14が微生物の検査装置である場合、微生物粒子の粒子個数、粒径分布及び微生物の生存率のうちの少なくとも一種を測定する手段を有するとよく、光学的分析による検査装置では、連続的な検査を効率的に行うことができる。また、検査装置14における計測は、培養、顕微鏡による観察、試薬による染色や発光、蛍光の検出、電気泳動、赤外等の電磁波を用いた吸収スペクトルの計測などの操作を含んでもよい。また、特定の成分や構造、遺伝子等に選択的に反応するマーカーによる標識を利用する操作であってもよい。微生物の計測は、フローサイトメトリー等の光学的分析などの操作によって行ってもよい。このような操作を実施可能な装置として、例えば、Thermo Fisher Scientifc社製のフローサイトメーター(製品名:Attune NxT)、Beckmann Coulter社製の細胞アナライザー(製品名:Vi-Cell)、Perkin Elmer社製のプレートリーダー(製品名:ViewLux)などが挙げられる。
 検査用サンプル液を用いて、液体に含まれる微生物の培養を行ってもよい。微生物の培養は、その微生物について判明している培養条件に基づいて、常法に従って培養を行ってよい。培養する微生物に適した方法を適宜選択して使用してよい。概して、特定の菌種を増殖させるように処方された選択増菌培地や選択分離培地が好適に使用される。市販の培地から適宜選択して使用しても、或いは、既知の処方に従って栄養素及び精製水等を用いて調製してもよく、検査を目的とする鑑別剤(pH指示薬、酵素基質、糖類等)や、目的外微生物の発育を抑制する選択剤などを必要に応じて添加してよい。
 図8は、流体力分離装置における分離効率とDe数との関係を調べた結果を示すグラフである。詳細には、流路の寸法が異なる5種類の流体力分離装置(装置A1~A5)の何れかと、ポリマー粒子(スチレン-ジビニルベンゼン共重合体)又はCHO細胞(チャイニーズハムスターの卵巣細胞)の何れかの分離対象を用いて、流体力分離装置による分離を行った。何れの分離対象も平均粒子径が14~18μmの範囲にある。分離効率は、[1-(x/X)]×100(%)として計算した値であり、計算式中のXは、分離前の液体に含まれる分離対象の濃度、xは、分離後の内周側の画分に含まれる分離対象の濃度を示す。何れの分離対象も粒径分布が狭いので、粒子又は細胞の全量が外周側の画分に濃縮される状態の分離効率を100%と見なした評価である。グラフから理解されるように、ポリマー粒子及び動物細胞の何れにおいても、De数が70前後において最も分離効率が高くなり、概して、De数が50~80の範囲になる条件において高い分離効率を達成することができる。従って、粒子の組成に係らず、流体力分離によって粒子の濃縮分離が可能であることが理解される。
 図9は、流体力分離装置へ液体培地を送るポンプの種類による動物細胞の生存率への影響を調べた結果を示す。細胞を培養した液体培地を、ポンプを用いて0.3MPaの吐出圧で流体力分離装置へ供給し、分離装置から排出される液体培地の2つの画分を纏めて収集し、少量をサンプリングした。細胞計測装置(ベックマンコールター社製、製品名:Vi-Cell)によりサンプルの細胞の生存率(%、全細胞中の生細胞の割合)を計測した結果である。グラフ中の「ガス圧送」は、コンプレッサが接続された加圧タンクから液体培地を圧縮空気で圧送する形態であり、容積式ポンプとして分類することができる。図9から、遠心ポンプにおいては細胞における損傷が大きく、容積式ポンプに分類される他のポンプでは、生存率の低下が防止されることが判る。微生物粒子は、動物細胞より耐久性を有するので、ポンプの種類によって図9のような著しい影響は見られていないが、濃縮する粒子の耐久性が不確実であるような場合には、適宜、ポンプを選択して用いるとよい。
 このように、流体力分離技術を利用して、サンプル原液に含まれる粒子を濃縮分離して検査用サンプル液を調製することができ、調製の際に、固形物粒子の破壊や、微生物への影響を防止することができる。流体力分離は、遠心分離より極めて効率的に粒子成分の濃縮分離を進めることができ、フィルター分離等に比べて、微生物を損傷せずに高い生存率で濃縮分離することができる。また、高価な成分や希少な物質を回収することができる。流体力分離において、液体を比較的高い速度で湾曲流路へ供給するので、本開示の検査用サンプル液の調製システムは、粒子の濃縮分離を行う処理能力が高く、検査用サンプル液の調製を効率的に行うことができる。また、製品製造における品質管理及び安全性の管理に十分適用可能である。
 前記特許文献1に記載される粒子分離器を参照して、円弧状の湾曲流路(旋回直径:約115mm、流路の径方向断面:長方形、流路幅:約3mm)が内部に形成された平板状の樹脂製成形体を作製した。この成形体を流路ユニットとして用いて流体力分離装置を構成した。液体を供給するためのポンプ10としてシリンジポンプ(プランジャを繰り返し押し引きして流体を圧送する)を使用し、上述の流体力分離装置を用いて、図3の検査用サンプル液の調製システム1を構成した。但し、返送路8aは設けず、第1画分及び第2画分を各々回収するようにした。
 サンプル原液として、酵母を水に加えて分散させ、酵母粒子を含んだ酵母水性液を用意し、容器2に収容した。シリンジポンプを用いて、容器2の酵母水性液を流体力分離装置の湾曲流路に110mL/minの一定流量で圧送した。流体力分離装置の導出口42,43から排出される外周側の第1画分及び内周側の第2画分を、各々、回収容器に回収した。サンプル原液及び第1画分の各々について、液に含まれる酵母の数及び生存率(全酵母中の生酵母の割合)を、画像式粒子計測装置(ベックマンコールター株式会社製、製品名:Vi-Cell)を用いて計測した。この操作及び計測を6回行った。各回における結果を表1に示す。尚、表1中の濃縮率は、液体の単位容積当たりの酵母の粒子数を酵母濃度として、サンプル原液の酵母濃度を基準とした第1画分の酵母濃度の百分率(%)として表記している。

 表1から分かるように、6回の操作の何れにおいても、第1画分に酵母が濃縮されている。流体力分離の前後で酵母の生存率の明らかな低下は見られないことから、分離操作による酵母へのダメージは殆どないと言える。また、分離前の酵母水性液と比較して、第1画分における酵母の生存率が有意に増加している。これは、第1画分に含まれる相対的に小さい粒子の割合が減少することによって、死菌体、デブリ及び不純物の割合が減少したために、生存率が増加したと見なすことができる。従って、流体力分離によって、酵母の生存率を低下させることなく、酵母が濃縮した第1画分を回収可能であり、検査用サンプル液として提供可能であることが明らかである。
 1.5Lの液体培地(GEヘルスケア社製、製品名:SH30934.01 HyCell CHO Medium)にアミノ酸(L-アラニル-L-グルタミン)及び抗生物質(ペニシリン、ストレプトマイシン、アムホテリシンB)を適量添加した。温度を36.5~37.0℃に維持した液体培地中で、CHO細胞株(ATCC CRL-12445 Cricetulus griseus/CHO DP-12 clone#1932)を培養した。培養中、pHが6.70未満にならないように液体培地のpHを管理した。得られた培養液には、細胞が分泌したIgG抗体が含まれており、その濃度を測定した。
 実施例1と同様にして検査用サンプル液の調製システムを構成し、上記で得られた細胞培養液を、サンプル原液として、容器2に収容した。シリンジポンプを用いて、容器2の細胞培養液を流体力分離装置の湾曲流路に一定流量で圧送した。流体力分離装置の導出口42,43から排出される外周側の第1画分及び内周側の第2画分を、各々、回収容器に回収した。第2画分について、液に含まれる細胞数を、画像式粒子計測装置(ベックマンコールター株式会社製、製品名:Vi-Cell)を用いて計測した。また、第2画分に含まれるIgG抗体の濃度を測定した。
 上記の操作及び測定を3回行った。各回における結果を表2に示す。尚、表2中の細胞分離効率は、[1-(x/X)]×100(%)として計算した値であり、計算式中のXは、サンプル原液の細胞濃度、xは、分離後の第2画分(内周側の画分)の細胞濃度を示す。

 表2から分かるように、3回の操作の何れにおいても分離効率は90%以上であり、第1画分に細胞が濃縮されている。また、流体力分離の前後で、抗体濃度において有意な変化は見られないことから、抗体タンパク質のような脆弱な長鎖タンパク質であっても、組成変化を生じることなく、検査に供給することができる。つまり、液中成分を変化させることなく、微生物検査等や液中の成分分析を行うことができ、液体の品質を確認することができる。従って、流体力分離を利用して検査用サンプル液を調製することによって、固体成分の混入に起因する分析品質の低下を防止し、高精度の検査を行うことができる。
 液体に含まれる成分に影響を及ぼすことなく、液体中に含まれる粒子を濃縮して分取し、検査用サンプル液を効率的に調製することができる。医療医薬製品や飲食品の製造における各種検査に利用して、製造される製品の品質管理及び安全管理を効率的に実施できるので、経済性、品質の向上に寄与する。

Claims (12)

  1.  微生物分離装置を有し、サンプル原液から検査用サンプル液を調製する検査用サンプル液の調製システムであって、前記微生物分離装置は、
     矩形断面を有する湾曲流路を有し、前記湾曲流路を液体が流れることによって生じる渦流れを利用して、前記液体を第1画分と第2画分とに分離する流体力分離装置と、
     前記サンプル原液を前記流体力分離装置へ供給する送液部と
     を有し、前記流体力分離装置によって、前記サンプル原液に含まれる粒子は第1画分に濃縮される検査用サンプル液の調製システム。
  2.  前記サンプル原液は、飲食品又は医療用薬品の製造における液状原料、液状中間物、又は、液状最終製品である請求項1に記載の検査用サンプル液の調製システム。
  3.  前記送液部は、製造ラインから分岐して前記流体力分離装置に接続される供給路と、前記流体力分離装置へ前記サンプル原液を供給するための流動圧を前記サンプル原液に付勢するポンプとを有し、製造ラインを流通する液状原料、液状中間物、又は、液状最終製品の一部が前記サンプル原液として前記流体力分離装置へ供給される請求項1又は2に記載の検査用サンプル液の調製システム。
  4.  前記送液部は、更に、前記第1画分及び前記第2画分のうちの一方を製造ラインへ返送する返送路を有する請求項3に記載の検査用サンプル液の調製システム。
  5.  前記送液部は、前記サンプル原液を収容する容器と、前記容器を前記流体力分離装置に接続する供給路と、前記流体力分離装置へ前記サンプル原液を供給するための流動圧を前記サンプル原液に付勢するポンプとを有する請求項1又は2に記載の検査用サンプル液の調製システム。
  6.  前記送液部は、更に、前記第1画分及び前記第2画分のうちの一方を前記容器に返送する返送路を有する請求項5に記載の検査用サンプル液の調製システム。
  7.  前記送液部は、更に、前記第1画分及び前記第2画分のうちの他方の一部を前記供給路へ還流するための還流路を有する請求項4に記載の検査用サンプル液の調製システム。
  8.  更に、追加の流体力分離装置を有し、前記送液部は、更に、前記流体力分離装置で分離した前記第1画分及び前記第2画分のうちの他方の残部を前記追加の流体力分離装置へ供給するための流路を有する請求項7に記載の検査用サンプル液の調製システム。
  9.  更に、検査装置と、前記流体力分離装置を前記検査装置と接続する流路と、前記検査装置を製造ラインに接続する流路を有し、前記第1画分及び前記第2画分の他方は、前記検査装置を経て製造ラインに返送される請求項4に記載の検査用サンプル液の調製システム。
  10.  前記粒子は、ウイルス、真正細菌、古細菌、菌類、粘菌類、藻類及び原生動物のうちの少なくとも一種の微生物であり、前記第1画分は検査用サンプル液として供給され、前記返送路は、前記第2画分を前記流体力分離装置から返送する請求項4又は6に記載の検査用サンプル液の調製システム。
  11.  前記粒子は、微生物より大きい粒子状固形物であり、前記第2画分は検査用サンプル液として提供され、前記第1画分は、前記返送路によって返送されるか、又は、システム外に排出される請求項4又は6に記載の検査用サンプル液の調製システム。
  12.  微生物分離処理を含み、サンプル原液から検査用サンプル液を調製する検査用サンプル液の調製方法であって、前記微生物分離処理は、
     矩形断面を有する湾曲流路を液体が流れることによって生じる渦流れを利用して、前記液体を第1画分と第2画分とに分離する流体力分離工程と、
     前記サンプル原液を前記流体力分離工程へ供給する送液工程と
     を有し、前記流体力分離工程によって、前記サンプル原液に含まれる粒子は第1画分に濃縮される検査用サンプル液の調製方法。
