Definitions
- the present invention relates to a sample analyzer that measures the amount of a sample contained in a sample.
- the sample analyzer is a device that measures the amount of sample contained in the sample.
- dirt on the sample that adheres to the side surface of the needle that sucks the sample dirt that adheres to the flow path, dirt that has entered the gap created between the connectors that connect the flow path, or dirt that has stuck to the flow path, etc. It may be detected when analyzing the sample. This phenomenon is called carryover. As a result, it is known that the measured value exceeds the original value and the measurement accuracy deteriorates.
- Patent Document 1 describes a method of measuring the presence or absence of carryover by cleaning the needle and the flow path after measuring the sample and then performing blank measurement in order to avoid carryover.
- Patent Document 2 discloses a method of detecting carryover by detecting a high value abnormality of a measured value of a measurement item.
- Patent Document 2 Since the method described in Patent Document 2 detects carryover using the measured values of the measurement items, erroneous detection of carryover may occur depending on the concentration of the measurement items in the sample, and a washing step that is not originally necessary may be performed. It may be necessary to remeasure the carryover as it is added. This has the problem of reducing the sample processing capacity.
- the present invention has been made in view of the above problems, and an object of the present invention is to provide a sample analyzer capable of measuring carryover without reducing the sample processing capacity.
- the sample analyzer measures a first sample containing a first sample and a first internal standard substance, then measures a second sample containing a second sample and a second internal standard substance, and the first sample is described.
- the second contained in the second sample using the amount of the first internal standard substance measured when the sample was measured and the amount of the second internal standard substance measured when the second sample was measured. Calculate the amount of sample.

It is a table explaining the process of measuring the carryover when measuring a plurality of measurement items. It is a conceptual diagram which shows the correspondence example of the measurement item and the internal standard substance at the time of measuring the carryover for a large number of measurement items.
- FIG. 1 is a block diagram showing the overall configuration of the sample analyzer 100 according to the first embodiment of the present invention.

- the measurement conditions include a list of measurement items and device control values corresponding to various measurement items.
- the control value includes the voltage value of each electrode, the measurement time, and the like. Since the mass spectrometer separates substances by the ratio of mass number to charge m / z, m / z is also input as a control value. m / z has a one-to-one correspondence with the measurement item.
- the mass spectrometer can switch the measurement item in milliseconds by switching the voltage at high speed and changing the m / z to be measured. By repeatedly performing this switching during one measurement, a plurality of different measurement items can be measured at the same time. These measurement conditions can be arbitrarily changed by the user from the input device 146 via the input interface 145.
- the detection signal of the photodetector 123a processed by the digital signal processing circuit 123d is input to the control unit 140, and the CPU 141 (Central Processing Unit) processes the detection signal.
- the control unit 140 records the signal amounts of various measurement items on the hard disk 142 together with the measurement time.

- FIG. 5 is a table showing a carryover measurement process using a conventional blank sample.
- the internal standard substance to be added is referred to as IS.
- the "measurement order" indicates a time series, and measurements are performed from a small number to a large number. In each side-hand order, the sample measurement sequence shown in FIG. 4 is carried out, and the total signal amount of the specified measurement item (integral value of the signal amount peak illustrated in FIG. 3) can be obtained as measurement data.
- the total signal amount for the measurement item A in the sequence i be Int (i, A).
- the effect of the carryover appears as an increase in the total signal amount in the subsequent measurement sequence.
- the increased signal amount be the carryover signal amount ⁇ (1, A).
- the ratio of the signal amount of the measurement order of the carryover destination to the signal amount of the measurement order of the carryover source is defined as the carryover ratio CO (1, A).
- the carryover signal amount and the carryover ratio of the measurement item A with respect to the order 1 to the order 2 are shown by the following formula 1 because the sample in the order 2 is blank. Will be done.
- the carryover ratio CO (i, A) for the measurement item A in the sequence i is the total signal amount Int (i, i,) of the measurement item A at the time of sample measurement in the sequence i. It is given as a ratio between A) and the total signal amount of measurement item A at the time of blank sample measurement in order i + 1. That is, the carryover value is calculated by the following equation 2. i indicates the order at the time of sample measurement.
- FIG. 6 is a table showing the carryover measurement process in the first embodiment. Similarly to FIG. 5, a case where one measurement item A is continuously measured is shown.
- the two types of internal standard substances to be added are referred to as IS1 and IS2.
- the carryover amount for the measurement item A is measured by using the total signal amount of the internal standard substance. Assuming that the carryover ratio of the measurement item A with respect to the order 1 to the order 2 measured in the first embodiment is CO_NEW (1, A), CO_NEW (1, A) is represented by the following formula 3.
- the carryover signal amount ⁇ (1, A) can be calculated by the following equation 4.
- JA and IS1 represent the correlation coefficient between the carryover value of the measurement item A and the carryover ratio of the internal standard substance IS1.
- the carry-over amount of the measurement item and the carry-over amount of the internal standard substance are considered to be correlated. Therefore, it is assumed that the carry-over ratio of the measurement item and the carry-over ratio of the internal standard substance have a similar correlation. In the first embodiment, this relationship is expressed as a correlation coefficient.
- the correlation coefficient is expressed as the ratio of the carryover ratio of the measurement item to the carryover ratio of the internal standard substance.
- JA and IS1 are represented by the following equation 5.
- the carry-over amount of the measurement item A is calculated by measuring the carry-over amount of the internal standard substance through the above correlation coefficient.
- the internal standard substance is substantially in the order 2. The same measurement as the blank measurement of IS1 is carried out.
- FIG. 7 is a table illustrating a procedure for obtaining a correlation coefficient.
- the correlation coefficients JA and X IS1, IS2
- the correlation coefficients JA and X can be obtained by measuring the carryover amount of the measurement item A and the carryover amount of each internal standard substance through the conventional carryover measurement procedure as shown in FIG. .. Since the correlation coefficients JA and X are defined as the ratio of the two carryover ratios as shown in Equation 5, the dependence on the measurement order is removed. That is, the correlation coefficient is a constant that does not depend on the measurement order. Therefore, it is sufficient to measure the correlation coefficients JA and X once.
- the correlation coefficient does not depend on the measurement order.
- the carry-over amount of the internal standard substance IS1 with respect to the measurement order 1 to 3 is considered to be smaller than that for the measurement order 1 to 2. This is because the amount of carryover generally decreases over time.
- the carry-over amount of the measurement item A with respect to the measurement order 1 to the measurement order 3 also decreases with time. Therefore, in the present invention, it is assumed that the carry-over ratio CO (i, IS1) of the internal standard substance IS and the carry-over ratio CO (i, A) of the measurement item A always change with time at the same ratio. Under this assumption, the ratio of the two is considered to always be the same regardless of the measurement order. Therefore, by using Equation 5, the same correlation coefficient can be used regardless of the measurement order. In other words, the dependence on the measurement order can be removed.
- the correlation coefficient is significant because it does not depend on the measurement order, the correlation coefficient may be obtained by a calculation formula or function other than the formula 5 as long as it has the same function. On the contrary, if the correlation coefficient depends on the measurement order, the correlation coefficient may be obtained for each combination of measurement orders and the correlation coefficient corresponding to the measurement order may be used.
- the numerical value of the correlation coefficient is supplemented.
- the values of JA and X are small, there is no correlation between the internal standard substance and the measurement item, and the carryover amount can hardly be measured by the method according to the first embodiment. Therefore, it is necessary to select an appropriate substance with a large correlation coefficient as the internal standard substance.
- two values of the signal increase amount ⁇ (i, A) and CO_NEW (i, A), which is the ratio from the previous measurement amount, are measured as the carryover measurement values. Can be sought. Regarding the presence or absence of carryover, the threshold value and the measured value are compared as in the conventional method, and if the measured value exceeds the threshold value, it can be considered that the carryover amount exceeds the allowable range. In that case, either ⁇ (i, A) or CO_NEW (i, A) may be compared with the threshold value. When the carryover amount is considered to exceed the permissible range by comparing these measured values with the threshold value, the control unit 140 can output an alert or the like to that effect.
- the sample analyzer 100 measures the first sample containing the measurement item A and the internal standard substance IS1, and then measures the second sample containing the measurement item A and the internal standard substance IS2.
- the carryover amount of the measurement item A is measured by using the signal amount of the internal standard substance IS1 in.
- the sample analyzer 100 has a correlation coefficient representing the relationship between the carry-over ratio CO (1, A) of the measurement item A and the carry-over ratio CO (1, IS1) of the internal standard substance IS1.
- the carryover amount of measurement item A is measured using JA and IS1 . Thereby, the carryover amount of the measurement item A can be measured by the correlation coefficient measured once without depending on the measurement order. If the carryover amount is known, the accurate amount of the measurement item A can be calculated by subtracting this from the measurement result.
- Embodiment 2 The carryover measurement procedure described in the first embodiment can also be used to measure the carryover of each measurement item when measuring a plurality of measurement items. A specific example thereof will be described in Embodiment 2 of the present invention.
- the configuration of the sample analyzer 100 is the same as that of the first embodiment.
- FIG. 8 is a table illustrating the process of measuring carryover when measuring a plurality of measurement items.
- the measurement items are A, B, and C, and the internal standard substances to be added are referred to as IS (A), IS (B), and IS (C), respectively.
- the sample includes all the measurement items ABC, and one ABC is measured in each measurement sequence.
- the method of measuring the carryover may be to multiply the ratio of the signal amounts of the internal standard substances by the correlation coefficient between the order i and the order i + 1. That is, the carryover ratio of the measurement item A between the order 1 and the order 2 is expressed by the following equation 6.
- the control unit 140 calculates the carryover ratio of the measurement item B according to the equations 7 and 8, and compares this with the threshold value to determine whether or not the carryover of the measurement item B is within the allowable range. Can be determined. If the carryover of B is within the permissible range, the measurement result (signal amount) of B in the measurement sequence 2 may be adopted as it is. If the permissible range is exceeded, for example, an alert to that effect can be output from the control unit 140 to prompt cleaning or remeasurement.
- the carryover ratio can be obtained for item C as well.
- the carryover ratio for all measurement items can be obtained in any order.
- the sample analyzer 100 has a carryover ratio CO (1, IS (A)) of the internal standard substance IS (A) to measurements 1 to 2 and a carryover of measurement item B to measurements 1 to 2.
- the carryover ratio (Equation 7) of the measurement item B is calculated according to the correlation (Equation 8) with the ratio CO (1, B).
- the method according to the second embodiment can be applied to the general public when there are two or more types of measurement items.
- the internal standard substance is not limited to three types, and two or more types of internal standard substances that can be measured alternately as in the first embodiment may be used according to the number of measurement items.
- FIG. 9 is a conceptual diagram showing an example of the correspondence between the measurement items and the internal standard substance when measuring the carryover for a large number of measurement items.
- the same type of measurement items are classified as a measurement item group. Furthermore, two corresponding internal standard substances were assigned to each measurement item group.
- the internal standard substance assigned to the measurement item group is added to the sample.
- the two internal standard samples corresponding to the measurement item group are alternately added as in the first embodiment.
- the internal standard substance IS A1 or A2
- the internal standard substance IS B1 or B2
- the measurement item group and the internal standard substance are classified so that the correlation coefficient for each measurement item group is sufficiently large and close to each other. To do. By classifying in this way, carryover can be measured efficiently, and the types of internal standard substances used can be reduced.
- the carryover is measured using the internal standard substance, but the internal standard substance can also be used in combination with this for other purposes.
- the signal amount of the measurement item 301 can be corrected by using the signal amount of the internal standard substance 302.
- the signal amount of the measurement item 301 can be corrected by using the signal amount of the internal standard substance 302.
- step S403 may be performed after the quantitative suction of step S402, or an internal standard substance may be mixed with the sample in advance as a pre-step of step S401. In this case, step S403 is not necessary.
- the measurement item and the internal standard substance may be any as long as they can be measured by the ion detection unit 123.
- hormones such as Testosterone, Estradiol, and Cortisol can be used.
