Abstract
一种胶原支架的制备方法及胶原支架。制备方法包括步骤:对原材料组织进行清洗;对清洗后的原材料组织进行冻融循环处理得到第一中间物;对第一中间物进行脱脂处理得到第二中间物;对第二中间物进行脱细胞处理得到第三中间物;对第三中间物进行脱核酸处理得到第四中间物;将第四中间物进行洗涤和干燥处理,得到胶原支架;采用全能核酸酶溶液进行脱核酸处理,全能核酸酶的含量为100U/L至10000U/L。采用所述制备方法制备的胶原支架,其构成的三维空间可以促进种子细胞的增殖与迁移、提高种子细胞按照其原始表型表达的稳定性,促进细胞基质的分泌从而增加支架的坚固性。
Description
本发明涉及组织工程技术领域,具体涉及一种胶原支架的制备方法及胶原支架。
组织工程(tissue engineering),是一门以细胞生物学和材料科学相结合,进行体外或体内构建组织或器官的新兴学科,其基本原理是,从机体获取少量的活体组织,用特殊的酶或其他方法将细胞(又称种子细胞)从组织中分离出来在体外进行培养扩增,然后将扩增的细胞与具有良好生物相容性、可降解性和可吸收的生物材料(支架)按一定的比例混合,使细胞黏附在生物材料(支架)上形成细胞-材料复合物;将该复合物植入机体的组织或器官病损部位,随着生物材料在体内逐渐被降解和吸收,植入的细胞在体内不断增殖并分泌细胞外基质,最终形成相应的组织或器官,从而达到修复创伤和重建功能的目的。生物材料支架所形成的三维结构为细胞获取营养、生长和代谢提供了良好的环境。
现有方法制备的生物材料支架即胶原支架的总表面积较小且结构可靠性差,另外,现有方法仅能够对单一原材料组织处理制备,适用范围小。
发明内容
基于上述现状,本发明的主要目的在于提供一种胶原支架的制备方法,以解决现有制备方法存在的上述问题。
为实现上述目的,本发明采用的技术方案如下:
本发明的第一方面提供了一种胶原支架的制备方法,所述制备方法包括步骤:
S100、对原材料组织进行清洗;
S200、对清洗后的原材料组织进行冻融循环处理得到第一中间物;
S300、对所述第一中间物进行脱脂处理得到第二中间物;
S400、对所述第二中间物进行脱细胞处理得到第三中间物;
S500、对所述第三中间物进行脱核酸处理得到第四中间物;
S600、将所述第四中间物进行洗涤和干燥处理,得到所述胶原支架;
其中,所述步骤S500中采用全能核酸酶溶液进行所述脱核酸处理,所述全能核酸酶的含量为100U/L至10000U/L,例如为100U/L、500U/L、1000U/L、1500U/L、2000U/L、2500U/L、3000U/L、3500U/L、4000U/L、4500U/L、5000U/L、5500U/L、6000U/L、6500U/L、7000U/L、7500U/L、8000U/L、8500U/L、9000U/L、9500U/L、10000U/L。
首先对原材料组织进行冻融循环处理,在冻融循环过程中形成结冰以及通过冻融循环的盐溶液的浓度变化(冻融循环的冷冻过程中随着温度下降会导致未结冰水中的盐的溶解度出现变化,盐浓度逐渐上升,产生细胞膜内外浓度差)使得细胞破裂,从而提高细胞裂解效果,获得更大的总表面积,从而更有利于组织的再生。
其次,由于不同的原材料组织的主要不同在于DNA和RNA不同,本申请的制备方法中设置有特定的脱核酸步骤,在该步骤利用全能核酸酶进行脱核酸处理,从而能够降解所有形式的DNA和RNA,通用性强,适用范围广,可以适用于不同的单一原材料组织以及各种不同原材料组织的组合处理。原材料组织可以为猪、牛、羊等哺乳动物的适于作为原材料的组织或器官,例如心脏、肝脏、肾脏、膀胱、膈肌、大网膜、肠系膜等。
优选地,在所述步骤S200中,将清洗后的原材料组织加入到含有三羟甲基氨基甲烷和苯甲基磺酰氟且PH值为7.8至8.2的缓冲液中进行所述冻融循环,PH值例如为7.8、7.9、8、8.1、8.2;
采用上述缓冲液一方面能够提高细胞裂解效果,另一方面苯甲基磺酰氟的添加能够为组织提供一个稳定的环境,避免细胞内释放的蛋白酶对原材料组织的其他蛋白质结构(构成基质材料的结构)造成破坏,从而保证生成的胶原支架的三维空间更加稳固。
优选地,所述缓冲液中,所述三羟甲基氨基甲烷的摩尔浓度为1mmol/L至20mmol/L,例如可以为1mmol/L、2mmol/L、3mmol/L、4mmol/L、5mmol/L、 6mmol/L、7mmol/L、8mmol/L、9mmol/L、10mmol/L、11mmol/L、12mmol/L、13mmol/L、14mmol/L、15mmol/L、16mmol/L、17mmol/L、18mmol/L、19mmol/L、20mmol/L,进一步优选为8mmol/L至12mmol/L,更进一步优选为10mmol/L,所述苯甲基磺酰氟的质量百分比为0.1%至1%,例如质量百分比为0.1%、0.2%、0.3%、0.4%、0.5%、0.6%、0.7%、0.8%、0.9%、1%,进一步优选为0.8%至1%,更进一步优选为1%。
优选地,所述缓冲液中还含有乙二胺四乙酸,所述乙二胺四乙酸的摩尔浓度为1mmol/L至10mmol/L,例如为1mmol/L、2mmol/L、3mmol/L、4mmol/L、5mmol/L、6mmol/L、7mmol/L、8mmol/L、9mmol/L、10mmol/L,进一步优选为8mmol/L至10mmol/L,更进一步优选为10mmol/L。
利用乙二胺四乙酸来调节缓冲液的PH值,使其PH值控制在7.8至8.2范围内,另外,由于乙二胺四乙酸具有抑制作用,采用其进行PH值调节能够避免因引入新的物质而对原材料组织的其他蛋白质结构(构成胶原支架的结构)造成破坏,从而进一步保证生成的胶原支架的三维空间更加稳固。
优选地,所述冻融循环中一次冻融循环包括:首先在-80℃至-20℃温度条件下进行1至8小时的冷冻处理,冷冻处理的温度例如可以为-80℃、-75℃、-70℃、-65℃、-60℃、-55℃、-50℃、-45℃、-40℃、-35℃、-30℃、-25℃、-20℃,冷冻处理的时长例如可以为1小时、2小时、3小时、4小时、5小时、6小时、7小时、8小时;然后在30℃至40℃温度条件下进行0.5至1小时的融化处理,融化处理的温度例如可以为30℃、31℃、32℃、33℃、34℃、35℃、36℃、37℃、38℃、39℃、40℃,融化处理的时长例如可以为0.5小时、0.6小时、0.7小时、0.8小时、0.9小时、1小时。
优选地,冻融循环的次数可以为2-3次,进一步优选为3次。
冻融循环虽然是一种现有的处理技术,但对于要求比较苛刻的胶原支架,融化温度过高会导致蛋白质变性,融化温度过低会导致融解不完全,因苯甲基磺酰氟这种蛋白酶抑制剂半衰期太短,如若融化处理时间过长,会导致细胞裂解后的细胞内蛋白酶对细胞外基质酶解,因此,现有的胶原支架的制备过程中并没有采用这一技术,本申请中,通过对温度和时长的限制,使得冻融循环能够很好的使得细胞破裂且不会对细胞外基质造成破坏。
优选地,所述步骤S300包括步骤:
S310、采用升浓度梯度的方式利用乙醇对所述第一中间物进行洗涤;
S320、采用丙酮、己烷对经所述步骤S310洗涤后的组织进行脱脂处理;
S330、采用降浓度梯度的方式利用乙醇对所述步骤S320处理后的组织进行洗涤得到所述第二中间物。
步骤S310能够使得组织脱水,采用升梯度乙醇洗涤能够避免直接使用高浓度乙醇使得细胞快速失水形成致密层而影响脱水效果,步骤S330能够使得脱脂后的组织水化,从而恢复基质原有状态,保护基质的原有性能。
优选地,所述步骤S310包括:
S311、利用浓度50%至70%的乙醇对所述第一中间物进行洗涤得到中间物Ⅰ,乙醇浓度例如为50%、55%、60%、65%、70%;
S312、利用浓度99%至100%的乙醇对所述中间物Ⅰ进行洗涤得到中间物Ⅱ,乙醇浓度例如为99%、99.2%、99.4%、99.6%、99.8%、100%;
优选地,所述步骤S311中对所述第一中间物洗涤一次,一次的洗涤时长为0.5小时至1小时,例如为0.5小时、0.6小时、0.7小时、0.8小时、0.9小时、1小时;
优选地,所述步骤S312中对所述中间物Ⅰ洗涤2至3次,每次的洗涤时长为0.5小时至1小时,例如为0.5小时、0.6小时、0.7小时、0.8小时、0.9小时、1小时。
其中,步骤S311实现脱水,步骤S312中利用高浓度乙醇进行洗涤能够尽可能把组织中的水带走,从而防止后续脱脂过程形成油包水(即溶剂包围着含水组织)而导致脱脂不完全。
优选地,所述步骤S320包括步骤:
S321、采用丙酮萃取所述中间物Ⅱ中的极性脂肪,得到中间物Ⅲ;
S322、采用己烷和丙酮的混合物萃取所述中间物Ⅲ中的非极性脂肪,得到中间物Ⅳ;
优选地,所述步骤S321中,采用浓度为99%至100%的丙酮对所述中间物Ⅱ洗涤1至3次,每次洗涤时长为0.5小时至1小时,例如为0.5小时、0.6小时、0.7小时、0.8小时、0.9小时、1小时;
优选地,所述步骤S322中,采用己烷和丙酮的混合物对所述中间物Ⅲ洗涤1至3次,每次洗涤时长为4小时至16小时,例如为4小时、5小时、6 小时、7小时、8小时、9小时、10小时、11小时、12小时、13小时、14小时、15小时、16小时;
优选地,所述步骤S322中,己烷与丙酮的质量比为1~4:1~3,例如为1:1、1:2、1:3、2:1、2:3、3:1、3:2、4:1、4:3。采用该比例范围的组合能够实现更加快速有效的非极性脂肪的萃取。
优选地,所述步骤S330包括步骤:
S331、利用浓度99%至100%的乙醇对所述中间物Ⅳ进行洗涤得到中间物Ⅴ,乙醇浓度例如为99%、99.2%、99.4%、99.6%、99.8%、100%;
S332、利用浓度50%至70%的乙醇对所述中间物Ⅴ进行洗涤得到所述第二中间物,乙醇浓度例如为50%、55%、60%、65%、70%;
步骤S331中利用高浓度乙醇能够将己烷和丙酮洗掉,以利于步骤S332中利用低浓度乙醇对组织的水化过程
优选地,所述步骤S331中,对所述中间物Ⅳ洗涤一次,一次的洗涤时长为0.5小时至1小时;
优选地,所述步骤S332中,在2℃至40℃的温度环境下,对所述中间物Ⅴ洗涤一次,一次的洗涤时长为8小时至24小时,例如为8小时、10小时、12小时、14小时、16小时、18小时、20小时、22小时、24小时。由于乙醇的穿透力强于水,因此脱水比水化过程快,因此步骤S332持续的时间较长。
优选地,所述步骤S400包括步骤:
S410、采用PH值为7至8.2的磷酸缓冲盐溶液对所述第二中间物进行洗涤得到中间物A,PH值例如为7、7.2、7.4、7.6、7.8、8、8.2,通过该步骤能够去除上一步骤残留的乙醇;
S420、采用含有三羟甲基氨基甲烷和乙二胺四乙酸的溶液对所述中间物A进行洗涤得到中间物B;
S430、采用含有十二烷基硫酸钠的溶液对所述中间物B进行洗涤得到中间物C;
S440、采用含有脱氧胆酸钠的磷酸缓冲盐溶液对所述中间物C进行洗涤得到中间物D;
S450、采用磷酸缓冲盐溶液对所述中间物D进行洗涤得到所述第三中间 物;
优选地,在所述步骤S410中,对所述第二中间物洗涤1至4次,每次持续时长为0.5小时至1小时,例如为0.5小时、0.6小时、0.7小时、0.8小时、0.9小时、1小时;
优选地,在所述步骤S420中,所述三羟甲基氨基甲烷的含量为1mmol/L至20mmol/L,例如1mmol/L、3mmol/L、5mmol/L、7mmol/L、9mmol/L、11mmol/L、13mmol/L、15mmol/L、17mmol/L、19mmol/L、20mmol/L,所述乙二胺四乙酸的含量为1mmol/L至10mmol/L,例如1mmol/L、2mmol/L、3mmol/L、4mmol/L、5mmol/L、6mmol/L、7mmol/L、8mmol/L、9mmol/L、10mmol/L,洗涤时长为0.5小时至4小时,例如为0.5小时、1小时、1.5小时、2小时、2.5小时、3小时、3.5小时、4小时;低浓度的Tris-EDTA使得组织细胞通过渗透压补水,使组织舒张更有利于后续脱细胞。
优选地,在所述步骤S430中,所述十二烷基硫酸钠的质量百分比为0.5%至1.5%,例如为0.5%、0.6%、0.7%、0.8%、0.9%、1%、1.1%、1.2%、1.3%、1.4%、1.5%,洗涤次数为2至3次,每次洗涤持续时长为8小时至24小时,例如为8小时、10小时、12小时、14小时、16小时、18小时、20小时、22小时、24小时;
优选地,在所述步骤S440中,所述脱氧胆酸钠的含量为1mmol/L至10mmol/L,例如为1mmol/L、2mmol/L、3mmol/L、4mmol/L、5mmol/L、6mmol/L、7mmol/L、8mmol/L、9mmol/L、10mmol/L,洗涤次数为1至3次,每次洗涤持续时长为0.5小时至2小时,例如为0.5小时、0.6小时、0.7小时、0.8小时、0.9小时、1小时、1.1小时、1.2小时、1.3小时、1.4小时、1.5小时、1.6小时、1.7小时、1.8小时、1.9小时、2小时;
十二烷基硫酸钠和脱氧胆酸钠能够溶解细胞和核膜,两者联用可有效地去除致密组织中的细胞质蛋白。
优选地,在所述步骤S450中,洗涤次数为2至3次,每次洗涤持续时长为0.5小时至2小时,例如为0.5小时、0.6小时、0.7小时、0.8小时、0.9小时、1小时、1.1小时、1.2小时、1.3小时、1.4小时、1.5小时、1.6小时、1.7小时、1.8小时、1.9小时、2小时。
磷酸缓冲盐溶液用于洗去残留的十二烷基硫酸钠和脱氧胆酸钠。
优选地,所述步骤S500包括步骤:

细胞核是细胞物质集中的地方,浓度很高,故采用高渗Tris溶液,全能核酸酶能降解所有形式的DNA和RNA,牛血清白蛋白(BSA)能提高酶活性、使酶切更完全,同时核酸酶的活性依赖于Mg
2+的存在。

优选地,上述的洗涤过程中,无另说明的,PBS的PH为7至8.2,例如为7、7.2、7.4、7.6、7.8、8、8.2,洗涤过程在温度0~40℃、50~200rpm的震荡箱或轨道式摇床中进行。
经实验,采用本申请提供的制备方法制备的胶原支架,其构成的三维空间可以促进种子细胞的增殖与迁移、提高种子细胞按照其原始表型表达的稳定性,促进细胞基质的分泌从而增加支架的坚固性。此外,该胶原支架还具有良好的生物相容性,良好的降解性能,与宿主整合能力强等特点。
本发明的第二方面提供了一种胶原支架,由上述的制备方法制备。

S600、将所述第四中间物进行洗涤和干燥处理,得到所述胶原支架;
其中,所述步骤S500中采用全能核酸酶进行所述脱核酸处理,所述全能核酸酶的含量为100U/L至10000U/L。
下面给出该制备方法的具体实施例:
实施例一
将新鲜猪大网膜用生理盐水洗涤3次;
将完成洗涤的大网膜加入到含10mmol/L三羟甲基氨基甲烷(Tris)、10mmol/L乙二胺四乙酸(EDTA)以及质量百分比1%苯甲基磺酰氟(PMSF) 的pH=8.0的缓冲液中冻融循环3次,其中,一次冻融循环中冷冻温度为-80℃,持续时长为1小时,融化温度为37℃,持续时长为0.5小时;
将冻融循环处理后的组织进行脱脂处理:
采用浓度50%的乙醇洗涤0.5小时;
采用浓度99%的乙醇洗涤2次,每次持续0.5小时;
采用浓度99%的丙酮洗涤1次,持续0.5小时;
采用质量比1:1的己烷和丙酮溶液洗涤1次,持续4小时;
采用浓度99%的乙醇洗涤0.5小时;
在2℃的环境下采用浓度50%的乙醇洗涤8小时。
将脱脂处理后的组织进行脱细胞处理:
采用PH值为7的磷酸缓冲盐溶液洗涤1次,持续0.5小时;
采用含有1mmol/L的三羟甲基氨基甲烷、1mmol/L的乙二胺四乙酸的溶液洗涤0.5小时;
采用十二烷基硫酸钠质量百分比0.5%的溶液洗涤1次,持续8小时;
采用含有1mmol/L的脱氧胆酸钠的磷酸缓冲盐溶液洗涤1次,持续0.5小时;
采用磷酸缓冲盐溶液洗涤1次,持续0.5小时。
将脱脂处理后的组织进行脱核酸处理:
采用含有30mmol/L三羟甲基氨基甲烷、1mmol/L镁离子的镁盐、PH值为7.8的缓冲液洗涤0.5小时;
在30℃环境下采用含有30mmol/L三羟甲基氨基甲烷、1mmol/L镁离子的镁盐、质量百分数0.01%牛血清白蛋白、100U/L全能核酸、PH值7.8的溶液中洗涤8小时。
将脱核酸处理后的组织进行洗涤冻干处理:
采用PH值7.8、含有30mmol/L三羟甲基氨基甲烷的溶液洗涤1次,持续0.5小时;
采用PH值7的磷酸缓冲盐溶液洗涤1次,持续0.5小时;
采用双蒸水洗涤1次,持续0.5小时;
冻干处理得到胶原支架。
实施例二
将新鲜猪心脏组织用生理盐水洗涤3次;
将完成洗涤的心脏组织加入到含1mmol/L三羟甲基氨基甲烷(Tris)、1mmol/L乙二胺四乙酸(EDTA)以及质量百分比0.1%苯甲基磺酰氟(PMSF)的pH=7.8的缓冲液中冻融循环3次,其中,一次冻融循环中冷冻温度为-20℃,持续时长为8小时,融化温度为30℃,持续时长为1小时;
将冻融循环处理后的组织进行脱脂处理:
采用浓度70%的乙醇洗涤1小时;
采用浓度100%的乙醇洗涤3次,每次持续1小时;
采用浓度100%的丙酮洗涤3次,持续1小时;
采用质量比4:3的己烷和丙酮溶液洗涤3次,持续16小时;
采用浓度100%的乙醇洗涤1小时;
在40℃的环境下采用浓度70%的乙醇洗涤24小时。
将脱脂处理后的组织进行脱细胞处理:
采用PH值为8.2的磷酸缓冲盐溶液洗涤4次,每次持续1小时;
采用含有20mmol/L的三羟甲基氨基甲烷、10mmol/L的乙二胺四乙酸的溶液洗涤4小时;
采用十二烷基硫酸钠质量百分比1.5%的溶液洗涤3次,每次持续24小时;
采用含有10mmol/L的脱氧胆酸钠的磷酸缓冲盐溶液洗涤3次,每次持续2小时;
采用磷酸缓冲盐溶液洗涤3次,每次持续2小时。
将脱脂处理后的组织进行脱核酸处理:
采用含有60mmol/L三羟甲基氨基甲烷、10mmol/L镁离子的镁盐、PH值为8.2的缓冲液洗涤2小时;
在42℃环境下采用含有60mmol/L三羟甲基氨基甲烷、10mmol/L镁离子的镁盐、质量百分数1%牛血清白蛋白、10000U/L全能核酸、PH值8.2溶液中洗涤48小时。
将脱核酸处理后的组织进行洗涤冻干处理:
采用PH值8.2、含有60mmol/L三羟甲基氨基甲烷的溶液洗涤3次,每次持续2小时;
采用PH值8.2的磷酸缓冲盐溶液洗涤3次,每次持续2小时;
采用双蒸水洗涤3次,每次持续2小时;
冻干处理得到胶原支架。
实施例三
将新鲜猪膀胱组织用生理盐水洗涤3次;
将完成洗涤的膀胱组织加入到含20mmol/L三羟甲基氨基甲烷(Tris)、8mmol/L乙二胺四乙酸(EDTA)以及质量百分比0.8%苯甲基磺酰氟(PMSF)的pH=8.2的缓冲液中冻融循环3次,其中,一次冻融循环中冷冻温度为-50℃,持续时长为5小时,融化温度为40℃,持续时长为0.8小时;
将冻融循环处理后的组织进行脱脂处理:
采用浓度60%的乙醇洗涤0.8小时;
采用浓度99.5%的乙醇洗涤2次,每次持续0.8小时;
采用浓度99.5%的丙酮洗涤2次,持续0.8小时;
采用质量比2:3的己烷和丙酮溶液洗涤2次,持续10小时;
采用浓度99.5%的乙醇洗涤0.8小时;
在20℃的环境下采用浓度60%的乙醇洗涤12小时。
将脱脂处理后的组织进行脱细胞处理:
采用PH值为8的磷酸缓冲盐溶液洗涤2次,每次持续0.8小时;
采用含有10mmol/L的三羟甲基氨基甲烷、8mmol/L的乙二胺四乙酸的溶液洗涤2小时;
采用十二烷基硫酸钠质量百分比1%的溶液洗涤2次,每次持续12小时;
采用含有8mmol/L的脱氧胆酸钠的磷酸缓冲盐溶液洗涤2次,每次持续1小时;
采用磷酸缓冲盐溶液洗涤2次,每次持续1小时。
将脱脂处理后的组织进行脱核酸处理:
采用含有45mmol/L三羟甲基氨基甲烷、8mmol/L镁离子的镁盐、PH值为8的缓冲液洗涤1小时;
在38℃环境下采用含有45mmol/L三羟甲基氨基甲烷、8mmol/L镁离子的镁盐、质量百分数0.1%牛血清白蛋白、5000U/L全能核酸、PH值8溶液中洗涤25小时。
将脱核酸处理后的组织进行洗涤冻干处理:
采用PH值8、含有45mmol/L三羟甲基氨基甲烷的溶液洗涤2次,每次持续1小时;
采用PH值8的磷酸缓冲盐溶液洗涤2次,每次持续1小时;
采用双蒸水洗涤2次,每次持续1小时;
冻干处理得到胶原支架。
本领域的技术人员能够理解的是,在不冲突的前提下,上述各优选方案可以自由地组合、叠加。
应当理解,上述的实施方式仅是示例性的,而非限制性的,在不偏离本发明的基本原理的情况下,本领域的技术人员可以针对上述细节做出的各种明显的或等同的修改或替换,都将包含于本发明的权利要求范围内。
Claims (10)
- 一种胶原支架的制备方法,其特征在于,所述制备方法包括步骤:S100、对原材料组织进行清洗;S200、对清洗后的原材料组织进行冻融循环处理得到第一中间物;S300、对所述第一中间物进行脱脂处理得到第二中间物;S400、对所述第二中间物进行脱细胞处理得到第三中间物;S500、对所述第三中间物进行脱核酸处理得到第四中间物;S600、将所述第四中间物进行洗涤和干燥处理,得到所述胶原支架;其中,所述步骤S500中采用全能核酸酶溶液进行所述脱核酸处理,所述全能核酸酶的含量为100U/L至10000U/L。
- 根据权利要求1所述的胶原支架的制备方法,其特征在于,在所述步骤S200中,将清洗后的原材料组织加入到含有三羟甲基氨基甲烷和苯甲基磺酰氟且PH值为7.8至8.2的缓冲液中进行所述冻融循环。
- 根据权利要求1所述的胶原支架的制备方法,其特征在于,所述步骤S300包括步骤:S310、采用升浓度梯度的方式利用乙醇对所述第一中间物进行洗涤;S320、采用丙酮、己烷对经所述步骤S310洗涤后的组织进行脱脂处理;S330、采用降浓度梯度的方式利用乙醇对所述步骤S320处理后的组织进行洗涤得到所述第二中间物。
- 根据权利要求3所述的胶原支架的制备方法,其特征在于,所述步骤S310包括:S311、利用浓度50%至70%的乙醇对所述第一中间物进行洗涤得到中间物Ⅰ;S312、利用浓度99%至100%的乙醇对所述中间物Ⅰ进行洗涤得到中间物Ⅱ。
- 根据权利要求4所述的胶原支架的制备方法,其特征在于,所述步骤S320包括步骤:S321、采用丙酮萃取所述中间物Ⅱ中的极性脂肪,得到中间物Ⅲ;S322、采用己烷和丙酮的混合物萃取所述中间物Ⅲ中的非极性脂肪,得到 中间物Ⅳ。
- 根据权利要求5所述的胶原支架的制备方法,其特征在于,所述步骤S330包括步骤:S331、利用浓度99%至100%的乙醇对所述中间物Ⅳ进行洗涤得到中间物Ⅴ;S332、利用浓度50%至70%的乙醇对所述中间物Ⅴ进行洗涤得到所述第二中间物。
- 根据权利要求1所述的胶原支架的制备方法,其特征在于,所述步骤S400包括步骤:S410、采用PH值为7至8.2的磷酸缓冲盐溶液对所述第二中间物进行洗涤得到中间物A;S420、采用含有三羟甲基氨基甲烷和乙二胺四乙酸的溶液对所述中间物A进行洗涤得到中间物B;S430、采用含有十二烷基硫酸钠的溶液对所述中间物B进行洗涤得到中间物C;S440、采用含有脱氧胆酸钠的磷酸缓冲盐溶液对所述中间物C进行洗涤得到中间物D;S450、采用磷酸缓冲盐溶液对所述中间物D进行洗涤得到所述第三中间物。
- 根据权利要求1所述的胶原支架的制备方法,其特征在于,所述步骤S500包括步骤:S510、采用含有三羟甲基氨基甲烷和镁盐且PH值为7.8至8.2的缓冲液对所述第三中间物进行洗涤得到中间物a;S520、在30℃至42℃的环境下,采用含有三羟甲基氨基甲烷、镁盐、牛血清白蛋白和全能核酸酶且PH值为7.8至8.2的溶液中对所述中间物a进行洗涤得到所述第四中间物。
- 根据权利要求1所述的胶原支架的制备方法,其特征在于,所述步骤S600包括步骤:S610、采用含有三羟甲基氨基甲烷、PH值为7.8至8.2的溶液对所述第四中间物进行清洗得到中间物b;S620、采用PH值为7至8.2的磷酸缓冲盐溶液对所述中间物b进行洗涤 得到中间物c;S630、采用双蒸水对所述中间物c进行洗涤得到中间物d;S640、对所述中间物d进行冻干处理得到所述胶原支架。
- 一种胶原支架,其特征在于,由如权利要求1至9任一项所述的制备方法制备。
