本発明の第1の局面は蛋白質を脱アミド化する方法(蛋白質脱アミド方法)に関する。本発明では蛋白質原料を塩類及び/又は多糖類の存在下、蛋白質脱アミド酵素で処理することによって蛋白質脱アミド反応の促進を図る。
(1)蛋白質中のグルタミン残基を脱アミド化し、グルタミン酸に変換する酵素(例えばプロテイングルタミナーゼ)。
(2)蛋白質中のアスパラギン残基を脱アミド化し、アスパラギン酸に変換する酵素(例えばプロテインアスパラギナーゼ)。
(3)蛋白質中のアルギニン残基を脱イミド化し、シトルリンに変換する酵素(例えば、アルギニンデイミナーゼ、プロテインアルギニンデイミナーゼ、ペプチジルアルギニンデイミナーゼ)。
<活性測定法>
10mmol/l Z-Gln-Glyを含む176mmol/lリン酸緩衝液(pH6.5)100μlに酵素溶液10μlを添加して、37℃、60分間インキュベートした後、12%トリクロロ酢酸溶液100μlを加えて反応を停止する。遠心分離(15000rpm、4℃、5分間)した後、上清について以下のようにAmmonia Assay Kit(Sigma-Aldrich社製)を用いて測定する(A1)。別に酵素溶液の代わりに水を用いて同様にして測定する(A2)。F-kit ammonia 100μl試薬2に上清10μlと水190μlを加え室温で5分間放置後100μlを用いての340nmの吸光度(E1)を測定する。残りの200μlに、1.0μlの試薬3(グルタメートデヒドロゲナーゼ)を加えた後、更に20分間室温に放置した後に残りの200μlの340nmの吸光度(E2)を測定する。上記条件下で1分間あたり1μmolのアンモニアを遊離する酵素量を1単位とし、以下の式に従って求める。

本発明の蛋白質脱アミド方法を用いれば脱アミド蛋白質、又はそれを含む食品や医薬品を製造することができる。そこで本発明は、脱アミド蛋白質の製造方法と、脱アミド蛋白質を含む食品又は医薬品の製造方法も提供する。脱アミド蛋白質の製造方法の典型的な一態様は、以下の工程(1)及び(2)を含む。尚、工程(2)の後に、蛋白質脱アミド酵素の失活工程を追加しても良い。以下の工程(1)及び(2)の具体的条件については、前記「1.1.蛋白質の脱アミド方法」の欄に記載の通りである。
(1)蛋白質、並びに塩類及び/又は多糖類を含む溶液を用意する工程、
(2)用意した溶液を蛋白質脱アミド酵素で処理する工程。
(1)蛋白質を含む食品原料又は医薬原料、並びに塩類及び/又は多糖類を含む溶液を用意する工程、
(2)用意した溶液を蛋白質脱アミド酵素で処理する工程。
本発明は更に、蛋白質の脱アミド化に使用できる蛋白質改良剤も提供する。本発明の蛋白質改良剤は、典型的には、本発明の蛋白質脱アミド方法、脱アミド蛋白質の製造方法、或いは食品又は医薬の製造方法に利用される。本発明の蛋白質改良剤は、蛋白質脱アミド酵素に加え、塩類及び/又は多糖類を有効成分として含む。蛋白質脱アミド酵素、塩類及び多糖類の詳細は上記の通りであるため、その説明を省略する。この例に限定されるわけではないが、蛋白質脱アミド酵素の含有量としては、製剤1g当たりに1~2000U、好ましくは10~1500U、より好ましくは50~1000Uである。塩類の含有量としては、蛋白質脱アミド酵素1U当たりに0.1~10g、好ましくは0.2~9g、より好ましくは0.5~8gである。多糖類量は、蛋白質脱アミド酵素1U当たりに0.01~10mg、好ましくは0.05~5mg、より好ましくは0.1~1mgである。また、蛋白質改良剤の最終的な形態は液体状であっても固体状(粉体状を含む)であってもよい。蛋白質改良剤は、有効成分の他、賦形剤、緩衝剤、懸濁剤、安定剤、保存剤、防腐剤、生理食塩水などを含有していてもよい。
プロテイングルタミナーゼの脱アミド化反応が塩類の存在によって促進されるか否かを検討するため、エンドウ蛋白質(製品名:NOW(登録商標) Sports Organic Pea Protein社製)を基質とした実験を用いた。精製水中に、エンドウ蛋白質10重量%(終濃度)、プロテイングルタミナーゼ(製品名:プロテイングルタミナーゼ「アマノ」500、天野エンザイム株式会社製)、クエン酸ナトリウム0.5%(W/V%)(終濃度)を混合し、塩酸又は水酸化ナトリウムを用いてpH7.5に調整後、52.5℃で1.5時間インキュベートし、反応させた。反応終了後、プロテイングルタミナーゼによって遊離したアンモニアを以下のように定量した。反応溶液を85℃、10分処理することで反応を停止させ、反応液1mLを遠心分離(13,000rpm, 10min)し、得られた上清を超純水にて100倍希釈したものについてAmmonia Assay Kit (Sigma-Aldrich)を用いてアンモニアを定量した。酵素量については、基質蛋白質1g当たり7.5Uのプロテイングルタミナーゼ(終濃度)とした。脱アミド化率はプロテイングルタミナーゼによって遊離したエンドウ蛋白質1gあたりのアンモニア量をエンドウ蛋白質1gの酸分解(6N塩酸(終濃度)中、110℃、24時間反応)によって遊離したアンモニア量で除算して算出した。また、比較のため、クエン酸ナトリウムを添加しなかったこと以外は、前記と同条件で酵素反応を行って脱アミド化率を測定した。
プロテイングルタミナーゼの脱アミド化反応が無機塩類の存在によって促進されるか否かを検討した。クエン酸ナトリウム0.5%(W/V%)(終濃度)の代わりにリン酸水素二カリウム(DKP)1.0%(W/V%)(終濃度)を用いたこと以外は、実施例1と同様の方法にて反応及び評価を行った。また、比較のため、リン酸水素二カリウムを添加しなかったこと以外は、前記と同条件で酵素反応を行って脱アミド化率を測定した。
無機塩類による脱アミド化反応の促進効果に関して、処理pHと無機塩類濃度の影響を検討した。エンドウ蛋白質を変更したことと(本実施例では、製品名:PURIS Pea Protein 870、PURIS Foods社製を使用)、処理pH及びリン酸水素二カリウム(DKP)の濃度を変更したこと以外は、実施例2と同様の方法にて反応及び評価を行った。なお、pHの調整は、塩酸又は水酸化ナトリウムを用いて行った。
プロテイングルタミナーゼの脱アミド化反応が多糖類の存在によって促進されるか否かを検討した。エンドウ蛋白質を変更したことと(本実施例では、製品名:PURIS Pea Protein 870、PURIS Foods社製を使用)、クエン酸ナトリウム0.5%(W/V%)(終濃度)の代わりにゲランガム0.02%(W/V%)(終濃度)を用いたこと以外、実施例1と同様の方法にて反応及び評価を行った。また、比較のため、ゲランガムを添加しなかったこと以外は、前記と同条件で酵素反応を行って脱アミド化率を測定した。
プロテイングルタミナーゼの脱アミド化反応が多糖類及び無機塩類の共存によって促進されるか否かを検討した。ゲランガム0.02%(W/V%)(終濃度)に加えてリン酸水素二カリウム(DKP)1.0%(W/V%)(終濃度)も用いたこと以外、実施例4と同様の方法にて反応及び評価を行った。また、比較のため、ゲランガム及びDKPの双方を添加しなかったこと以外は、前記と同条件で酵素反応を行って脱アミド化率を測定した。
プロテイングルタミナーゼの脱アミド化反応が多糖類及びポリリン酸の共存によって促進されるか否かを検討した。リン酸水素二カリウム(DKP)1.0%(W/V%)(終濃度)の代わりにポリリン酸ナトリウム(ヘキサメタリン酸ナトリウム)(SPP)0.5%(W/V%)(終濃度)を用いたこと以外、実施例5と同様の方法にて反応及び評価を行った。また、比較のため、ゲランガム及びSPPの双方を添加しなかったこと以外は、前記と同条件で酵素反応を行って脱アミド化率を測定した。
リン酸水素二ナトリウムを含む溶液に、リン酸二水素ナトリウム又は炭酸ナトリウムを補充添加した溶媒を用いて、Z-Gln-Gly(Benzyloxycarbonyl-L-glutaminylglycine)とプロテイングルタミナーゼの反応を行い、リン酸二水素ナトリウム又は炭酸ナトリウムの補充添加が脱アミド化の反応効率に及ぼす影響を評価した。具体的には、マッキルバイン緩衝液(pH7.0、16.5mMのリン酸水素二ナトリウム及び1.8mMのクエン酸含有)に、Z-Gln-Gly(終濃度60 mM)を加え、更にリン酸二水素ナトリウム又は炭酸ナトリウム(追加分の終濃度0~1.0M)を補充し、pHを7.0に調整した溶液を準備した。この溶液に、プロテイングルタミナーゼ(製品名:プロテイングルタミナーゼ「アマノ」500、天野エンザイム株式会社製)を0.13U/ml含む酵素液0.1mlを添加し、37℃で20分間反応させた。その後、0.4 Mトリクロロ酢酸溶液1.0mlを添加し、反応を停止させた。次いで、プロテイングルタミナーゼによって遊離したアンモニア量をアンモニア測定キット(製品名:アンモニアテストワコー、富士フイルム和光純薬株式会社製)を用いて測定した。リン酸二水素ナトリウム又は炭酸ナトリウムの補充添加していない条件(即ち、塩類として16.5mMのリン酸水素二ナトリウムのみを含む場合)において遊離したアンモニア量を100%として、リン酸二水素ナトリウム又は炭酸ナトリウムを補充添加した際の遊離したアンモニア量の相対値を求めた。
リン酸水素二ナトリウムを含む溶液に、炭酸ナトリウムを補充添加した溶媒を用いて、各種基質蛋白質とプロテイングルタミナーゼの反応を行い、脱アミド化の反応効率を評価した。具体的には、マッキルバイン緩衝液(pH7.0、16.5mMのリン酸水素二ナトリウム及び1.8mMのクエン酸含有)に、卵アルブミン、ミルクカゼイン、ひよこ豆蛋白、エンドウ蛋白、大豆蛋白、又は小麦グルテン(終濃度1.0 W/V%)を加え、更に炭酸ナトリウム(終濃度0.5 M)を補充添加し、pHを7.0に調整した溶液を準備した。この溶液に、プロテイングルタミナーゼ(製品名:プロテイングルタミナーゼ「アマノ」500、天野エンザイム株式会社製)を0.13U/ml含む酵素液0.1mlを添加し、37℃で20分間反応させた。その後、0.4 Mトリクロロ酢酸溶液1.0mlを添加し、反応を停止させた。次いで、プロテイングルタミナーゼによって遊離したアンモニア量をアンモニア測定キット(製品名:アンモニアテストワコー、富士フイルム和光純薬株式会社製)を用いて測定した。炭酸ナトリウムの補充添加していない条件(即ち、塩類として16.5mMのリン酸水素二ナトリウムのみを含む場合)でも同様の測定を行い、当該条件で遊離したアンモニア量を100として、炭酸ナトリウムを補充添加した際の遊離したアンモニア量の相対値を求めた。
リン酸水素二ナトリウムを含む溶液に、炭酸ナトリウムを補充添加した溶媒を用いて、プロテイングルタミナーゼの熱安定性を評価した。具体的には、マッキルバイン緩衝液(pH7.0、41.2mMのリン酸水素二ナトリウム及び4.4mMのクエン酸含有)に、炭酸ナトリウム(終濃度1.0 M)を補充添加し、pHを7.0に調整した溶液に、プロテイングルタミナーゼ(製品名:プロテイングルタミナーゼ「アマノ」500、天野エンザイム株式会社製)を0.13U/mlとなるように添加し、50℃で120分間、又は60℃で60分間インキュベートし、プロテイングルタミナーゼの活性を経時的に測定した。プロテイングルタミナーゼの活性は、前述する方法で行った。インキュベート前のプロテイングルタミナーゼの活性値を100%として、プロテイングルタミナーゼの残存活性値を求めた。また、比較のために、1.0 Mの炭酸ナトリウムを添加しなかったこと以外は、前記と同条件で試験を行い、プロテイングルタミナーゼの活性を経時的に測定した。
