Claims (20)
- 一种药物组合物,包含融合蛋白和缓冲剂,其中所述融合蛋白包含共价结合的IL-2与抗IL-2抗体,所述缓冲剂为组氨酸缓冲剂、醋酸盐缓冲剂或琥珀酸盐缓冲剂;优选地,所述缓冲剂为组氨酸缓冲剂或醋酸盐缓冲剂;更优选地,所述缓冲剂为组氨酸-盐酸组氨酸缓冲剂或醋酸-醋酸钠缓冲剂。
- 根据权利要求1所述的药物组合物,其中所述融合蛋白的浓度为5mg/mL至50mg/mL;优选地,所述融合蛋白的浓度为10mg/mL至30mg/mL。
- 根据权利要求1或2所述的药物组合物,其中所述药物组合物包含表面活性剂;优选地,所述表面活性剂为聚山梨酯或泊洛沙姆;更优选地,所述表面活性剂为聚山梨酯80、聚山梨酯20或泊洛沙姆188。
- 根据权利要求3所述的药物组合物,其中所述表面活性剂的浓度为0.05mg/mL至2mg/mL;优选地,所述表面活性剂的浓度为0.1mg/mL至0.8mg/mL。
- 根据权利要求1至4任一项所述的药物组合物,其中所述药物组合物包含糖;优选地,所述糖为蔗糖、甘露醇和海藻糖中的一种或多种;更优选地,所述糖为蔗糖。
- 根据权利要求5所述的药物组合物,其中所述糖的浓度为20mg/mL至100mg/mL;优选地,所述糖的浓度为30mg/mL至80mg/mL;更优选地,所述糖的浓度为30mg/mL至50mg/mL。
- 根据权利要求6所述的药物组合物,其中所述药物组合物还包含辅料;优选地,所述辅料为甘氨酸、甲硫氨酸、脯氨酸、组氨酸、苯丙氨酸、盐酸精氨酸、谷氨酸、天冬氨酸或乙二胺四乙酸二钠;更优选地,所述辅料为甘氨酸。
- 根据权利要求7所述的药物组合物,其中所述辅料的浓度为1mg/mL至30 mg/mL;优选地,辅料的浓度为5mg/mL至15mg/mL。
- 根据权利要求1至8任一项所述的药物组合物,其中所述缓冲剂的浓度为5mM至100mM;优选地,所述缓冲剂的浓度为5mM至30mM;更优选地,所述缓冲剂的浓度为5mM至10mM。
- 根据权利要求1至9任一项所述的药物组合物,其中所述缓冲剂的pH为5.0至6.5;优选地,所述缓冲剂的pH为5.5至6.0;更优选地,所述缓冲剂的pH为6.0。
- 根据权利要求1至10任一项所述的药物组合物,所述抗IL-2抗体包含重链可变区和轻链可变区,所述重链可变区具有:包含SEQ ID NO:37的氨基酸序列的HCDR1、包含SEQ ID NO:38的氨基酸序列的HCDR2和包含SEQ ID NO:39的氨基酸序列的HCDR3,所述轻链可变区具有:包含SEQ ID NO:40的氨基酸序列的LCDR1、包含SEQ ID NO:41的氨基酸序列的LCDR2和包含SEQ ID NO:42的氨基酸序列的LCDR3;优选地,所述重链可变区包含SEQ ID NO:9的氨基酸序列,所述轻链可变区包含SEQ ID NO:10的氨基酸序列;更优选地,所述抗IL-2抗体包含SEQ ID NO:165的氨基酸序列的重链和SEQ ID NO:166的氨基酸序列的轻链。
- 根据权利要求1至11任一项所述的药物组合物,所述IL-2直接或通过连接子融合至抗IL-2抗体的轻链的N端;优选地,所述IL-2的第3位氨基酸残基为E;更优选地,所述融合蛋白包含SEQ ID NO:165的氨基酸序列的重链和SEQ ID NO:189的氨基酸序列的轻链。
- 根据权利要求1至12任一项所述的药物组合物,其包含如下组分:a)10mg/mL至30mg/mL的所述融合蛋白,b)0.1mg/mL至0.8mg/mL的聚山梨酯或泊洛沙姆,c)30mg/mL至80mg/mL的蔗糖,d)1mg/mL至30mg/mL的甘氨酸,和e)5mM至10mM的组氨酸-盐酸组氨酸缓冲剂,所述组氨酸-盐酸组氨酸缓冲剂的pH为6.0;优选地,所述药物组合物包含如下组分:a)10mg/mL至30mg/mL的所述融合蛋白,b)0.1mg/mL至0.8mg/mL的聚山梨酯80,c)30mg/mL至50mg/mL的蔗糖,d)5mg/mL至15mg/mL的甘氨酸,和e)5mM至10mM的组氨酸-盐酸组氨酸缓冲剂,所述组氨酸-盐酸组氨酸缓冲剂的pH为6.0。
- 根据权利要求1至13任一项所述的药物组合物,其中所述组合物是液体制剂;优选地,所述组合物的施用途径为皮下注射、皮内注射、静脉注射或肌内注射;更优选地,所述组合物的施用途径为皮下注射。
- 一种冻干制剂,所述冻干制剂复溶后可形成权利要求1至14任一项所述的药物组合物。
- 一种制备冻干制剂的方法,其中包括将权利要求1至14中任一项所述的药物组合物进行冷冻干燥的步骤。
- 一种冻干制剂,所述制剂通过将权利要求1至14中任一项所述的药物组合物经冷冻干燥获得。
- 一种复溶溶液,所述复溶溶液是通过将权利要求15或17所述的冻干制剂复溶制备的。
- 一种制品,其包括容器,该容器中装有如权利要求1至14任一项所述的药物组合物、权利要求15或17所述的冻干制剂或权利要求18所述的复溶溶液。
- 一种治疗自身免疫疾病或延缓自身免疫疾病进展的方法,所述方法包括向有需要的受试者施用治疗有效量的权利要求1至14任一项所述的药物组合物、权利要求15或17所述的冻干制剂、权利要求18所述的复溶溶液或权利要求19所述制品;优选地,所述自身免疫疾病选自以下的任一项或组合:狼疮、移植物抗宿主病、丙型肝炎诱导的脉管炎、I型糖尿病、多发性硬化、炎性肠病、慢性GvHD、哮喘、皮炎、斑秃、急性肺损伤、败血症、反应性关节炎和类风湿性关节炎。
