生産期間中のVCDの平均値は、生産期間中、毎日培養槽内の培養液を抜き取り、Beckman Coulter社のCell Viability Analyzer Vi-cell XR等を用い測定したVCDの値を、生産期間中の培養日数分足し算し、それを生産期間中の培養日数で除すことにより求めることができる。
生産期間中のVCVの平均値は、生産期間中、毎日培養槽内の培養液を抜き取り、Beckman Coulter社のCell Viability Analyzer Vi-cell XR等を用い測定したVCDと細胞の平均径Cdの値から、VCV=VCD×(π/6×Cd3)の式によりVCV[×106μm3/ml]を計算し、そのVCVの値を生産期間中の培養日数分足し算し、それを生産期間中の培養日数で除すことにより求めることができる。

一本鎖抗体(scFv)は、VL及びVH領域がリンカー(例えば、アミノ酸残基の合成配列)を介して接合して、連続したタンパク質鎖を形成する抗体であり、ここでリンカーは、タンパク質鎖をそれ自身に折り重ね、一価抗原結合部位を形成させるのに十分な長さである。
IgG1をコードする核酸配列を含むベクターを構築し、構築したベクターをCHO-DG44細胞へ導入することにより、IgG1を発現させるCHO-DG44細胞(IgG1細胞)を作製した。ベクターの構築及び細胞への導入は、特表2016-517691号公報の実施例2に準じて行った。上記により、モノクローナル抗体を産生するCHO細胞を準備し、以下の実験において使用した。
実施例及び比較例においては、図1に示す細胞培養装置を用いて実験を行った。
Sartorius社製の全容3Lの培養槽(Biostat(登録商標))に1.3Lの培地を投入した。上記の培地に0.5×106 cells/mLになるようにCHO細胞を播種した。撹拌回転数160rpmで、上面からは、空気38mL/分とCO2 2mL/分を、下面からは培養液中の酸素濃度が80%になるように培養槽に設置されたスパージャーからO2を自動制御し供給した。
更に5日目に灌流比1.5vvdに変更した。
その後、細胞密度が120×106cells/mLに到達したら、細胞密度が120×106cells/mLで維持されるよう培養液の一部を引き抜きながら、セルブリードを行った。
このときに使用する培養条件(培地、セルブリード条件等)を、表1に記載の通りに設定して、実施例1~6並びに比較例1及び2を行った。なお、表1に記載の培地A~Cはそれぞれ市販の培地に、グルタミンおよびグルコースを表1に記載の量になるように添加した培地である。
以降、10日間以上にわたり培養を継続し、後記する評価を行った。
セルブリードを自動制御で行う場合は、ABER社製のFUTURAセンサーを用いて、培養液の静電容量を監視しながら連続的にセルブリードを自動で行った。具体的には以下の方法で実施した。
(1)ターゲットとなる静電容量を設定する。
(2)培養槽の重量制御値を設定する。
(3)培養液の静電容量を0.1s以下の周期で測定する。
(4)培養液の静電容量がターゲットを越えたら、培養液を引き抜くポンプを0.3vvd~3vvdの速度で運転する。
(5)培養液の液量は概ね一定に保たれるよう、培養槽の重量を測定しながら、培養槽内に培地が自動供給される。
(6)培養液の静電容量がターゲットを0.01%以上下回ったら、培養液を引き抜くポンプを停止する。
(1)生細胞密度、生細胞の平均径およびバイアビリティの測定
培養槽内の培養液を抜き取り、Beckman Coulter社のCell Viability Analyzer Vi-cell XRを用い測定した。なおVi-cellソフトはVi-cell XR2.04を用い、測定時のパラメータは以下のように設定した。
Min diameter:6μm
Max diameter:50μm
Dilution:細胞密度が100×105cells/mL以下ではサンプルは希釈せず、Dilutionは1で測定した。細胞密度が100×105cells/mL以上ではサンプルを10倍希釈し、Dilutionは10で測定した。
Cell brightness:85%
Cell sharpness:100
Viable cell spоt brightness:75%
Viable cell spоt area:5%
Minimum circularity:0
Decluster degree:Medium
培養液を抜き取り、Cytiva社のWhatman (Pore Size0.2μm,25mm Diameter)で細胞を濾過した後、濾液をNova biomedical社のBioprofile FLEX2で測定した。
上記方法で1日1回測定した。

実施例および比較例の評価基準として抗体のマンノース5の評価を行った。マンノース5を含むN型糖鎖プロファイルの測定は以下の方法で行った。得られた抗体回収液の抗体をペプチド-N-グリコシダーゼFで消化処理することでN型糖鎖を切り出した。その後、切り出された糖鎖を、2-アミノピリジンにより蛍光標識後、逆相HPLC(高速液体クロマトグラフィー)でマンノース5を定量分析し、全ピーク面積に対するマンノース5のピークを算出した。
マンノース5が4%未満である場合を評価A、4~7%である場合を評価B、7%を越える場合を評価Cとした。
各実施例及び各比較例の測定結果を表2及び表3に示す。

Claims (12)
- 培養容器内で細胞を培養することを含む、生産物の生産方法であって、
前記培養が灌流培養方式であり、
目標となる生細胞密度に到達した後の生産物の生産期間において、式(1)~(5)を満たし、
前記生産期間中のVCDの変動が、前記生産期間中の平均生細胞密度に対して±20%以下であり、
前記生産期間中のVCVの変動が、前記生産期間中のVCVの平均値に対して±20%以下である、
生産物の生産方法。

VCDは、生細胞密度[×106cells/mL]を示し、
VCVは、VCD×π/6×Cd3[×106μm3/mL]を示し、
Cdは、生細胞の平均径[μm]を示す。 - 前記生産期間中のVCDの変動が、前記生産期間中の平均生細胞密度に対して±20%以下となるように、培養液をブリーディングする、請求項1に記載の生産物の生産方法。
- 前記ブリーディングにおいて、培養液の静電容量をインライン計測しながら培養液をブリーディングする、請求項2に記載の生産物の生産方法。
- 前記ブリーディングが自動で制御されている、請求項3に記載の生産物の生産方法。
- 前記インライン計測される培養液の静電容量の変動が、前記生産期間中の静電容量の平均値に対して±20%以下である、請求項3又は4に記載の生産物の生産方法。
- 前記ブリーディングが、
(i)ターゲットとなる静電容量を設定すること;
(ii)培養容器の重量制御値を設定すること;
(iii)培養液の静電容量を0.1秒以下の周期で測定すること;
(iv)培養液の静電容量が前記ターゲットを越えたら、培養液を引き抜くポンプを0.3vvd~3vvdの速度で運転すること(ここで、vvdは、引き抜かれる培養液の体積/ 培養液の体積/dayを意味する);
(v)培養液の液量の変動が10%以内になるよう、培養液の質量を測定しながら、培養容器に培地を自動供給すること;および
(vi)培養液の静電容量が前記ターゲットを0.01%以上下回ったら、培養液を引き抜くポンプを停止すること、
を含む、請求項3から5の何れか一項に記載の生産物の生産方法。 - 前記生産期間において、式(6)を満たす、請求項1から6のいずれか一項に記載の生産物の生産方法。
