본 발명은 히알루론산염 파이버를 활용한 히알루론산염 부직포 및 이의 제조 방법에 관한 것으로서, 더욱 상세하게는 표면이 거칠고 마이크로 직경을 갖는 히알루론산염 파이버로 제조된 고기능성 히알루론산염 부직포 및 이의 제조방법에 관한 것이다.
부직포는 천연섬유, 화학섬유, 유리섬유, 금속섬유 등 각종 섬유를 상호 간의 특성에 따라, 엉키게 하는 시트 모양의 웹(Web)을 형성하고, 이를 기계적 또는 물리 방법으로 결합시켜 만든 평면구조의 섬유구조체로서, 부직포 용도, 특성 및 기능에 따라 다양한 산업분야에서 광범위하게 사용되고 있으며, 더 나아가서는 부직포에 2차 가공 또는 다양한 기능성을 부가할 수 있는 성분을 혼합하여 다양한 부직포에 관한 연구가 진행되고 있다.
최근 여러 산업분야에서는 부직포를 활용한 제품의 소비량이 날로 증가하고 있는 추세이다. 예를 들면, 부직포를 고온 가압시켜서 하드형의 패널로도 생산하여 자동차의 마감재 또는 건축자재의 마감재로도 사용하고 있으며 의류, 위생, 토목, 건축, 자동차, 의료 등 매우 다양한 분야에서 단순한 제품의 목적으로부터 진일보하여 고기능 효과를 부여하는 고부가가치 부직포 제품으로 집중적인 연구가 이루어지고 있다. 특히, 의료 시장에 적용되는 부직포 제품은 수술용 가운, 마스크 등의 착용 제품 외에 수술용 드레이프, 패드, 드레싱, 필터 소재뿐만 아니라, 내부 장기의 재생을 위해 삽입하는 조직 지지체로도 활용되고 있다.
이러한 의료용 부직포를 제조하기 위하여, 다양한 생분해성 고분자가 이용되고 있다. 생분해성 고분자는 생체 내에서 서서히 화학적 분해가 일어나면서 그 형태와 무게가 점차 소멸되는 고분자를 총칭하는 것인 바, 대표적으로, 천연고분자로는 콜라겐 (collagen), 섬유결합소 (fibronectin), 젤라틴 (gelatin), 키토산 (chitosan), 알긴산 (alginic acid) 등이 이용되고 있으며, 합성 고분자로는 poly(lactic acid) (PLA), poly(glycolic acid) (PGA), poly(D,L-lactic-co- glycolic acid) (PLGA)와 그 유사 공중합체들과 poly(ε-caprolactone) (PCL), polyanhydrides, polyorthoesters 등이 이용되고 있다.
한국등록특허 제1329261호는 키토산을 이용한 기능성 부직포 및 그 제조방법을 개시하였고, 한국등록특허 제2002586호는 셀룰로오스 섬유에 키토산이 고르게 피복된 천연섬유가 포함된 열접착 부직포를 개시하였다.
한편, 히알루론산(Hyaluronic Acid, HA)은 진피 층에 존재하는 뮤코 다당류로서 N-아세틸 글루코사민(N-acetyl glucosamine)과 D-글루쿠론산(D-Glucuronic acid)이 β-1, 4 글루코시드 결합으로 구성된 이당류들이 히알루론산염 사슬을 형성하기 위해 β-1, 3 글루코시드 결합으로 연결되어 있는 생체 고분자 물질이다.
히알루론산은 인체의 결합조직, 상피 및 신경조직 등에 고루 분포되어 있으며 다양한 생리 활성을 지닌 생체 적합성 재료로서 피부 재생과 보습, 탄력 유지 및 주름 개선에 대한 효과가 입증됨에 따라, 최근 이러한 히알루론산염 섬유로 창상피복재, 미용패치, 유착방지피복제 등에 사용하기 위해 부직포로 제조할 필요성이 산업계에서 요구되고 있는데, 부직포 원료로 사용될 히알루론산 섬유는 그 제조방법별로 다음과 같은 문제점이 있는 실정이다.
히알루론산 나노섬유 제조방법은 나노섬유 제조 시 필수적으로 사용하는 휘발성 용매인 DMF, DMSO, 포름산 등 유기용매의 추가적인 제거 공정이 필요하며, 부직포 제조시 나노섬유 부직포 단독으로는 사용이 불편하고 물성이 취약한 단점이 있으며, 또한 단위 시간당 생산성이 매우 낮아 생산단가가 높은 문제점이 있었다.
이러한 문제점을 해결하기 위하여, 한국등록특허 제2338355호는 습식 부직포 제조법을 개시하였으나, 이 역시 물성이 취약하며, 제조공정이 다소 복잡하므로, 더욱 효율적인 제조방법에 대한 연구가 필요하다. 부직포 형태로 활용 시 일정 강도 이상의 물성이 구현되지 않을 경우 제품화를 위한 타발 등의 가공 작업이 불가능하여 상용화가 곤란하다.
이에, 본 발명자들은 상기 문제점을 해결하기 위하여 노력한 결과, 수분함량이 50∼95중량%인 히알루론산염 페이스트(paste)를 제조하고, 용융시킨 뒤 노즐을 통하여 압출시킨 다음 건조시킬 경우, 표면이 거칠고, 인장강도가 우수한 히알루론산염 파이버를 대량으로 제조할 수 있고, 이를 이용하여 니들 펀칭하여 점착력 및 인장강도가 우수한 히알루론산염 부직포를 제조할 수 있다는 것을 확인하고, 본 발명을 완성하게 되었다.
본 발명의 목적은 피부 또는 체내 활용 시 점착이 가능할 뿐만 아니라 가공을 위한 기계적 물성이 우수하여 피부 외용제용 마스크 팩과 조직 수복용 생체재료 또는 유착방지 피복재로의 활용에 효과적인 히알루론산염 부직포 및 이의 제조방법을 제공하는 것이다.
상기 목적을 달성하기 위하여, 본 발명은 하기 측정방법으로 측정 시, 휨 각도가 2~10°이고, 수분함량이 5~25중량%이며, 수분을 제외한 전체 파이버 중량에 대하여 히알루론산염 함량이 90중량% 이상인 것을 특징으로 하는 히알루론산염 파이버를 포함하는 히알루론산염 부직포를 제공한다.

본 발명에 있어서, 상기 히알루론산염 파이버의 인장강도는 3~15 kg/㎟인 것을 특징으로 한다.
본 발명에 있어서, 상기 히알루론산염 부직포의 점착력은 0.9~2.7 kPa인 것을 특징으로 한다.
본 발명에 있어서, 상기 히알루론산염 부직포의 두께는 1~10㎜인 것을 특징으로 한다.
본 발명에 있어서, 상기 히알루론산염 부직포의 인장강도는 0.3~1.5 kg/㎟인 것을 특징으로 한다.
본 발명에 있어서, 상기 히알루론산염 부직포는 피부 외용제용 마스크팩, 조직 수복용 생체재료 또는 유착방지 피복재 용도인 것을 특징으로 한다.
본 발명은 또한, (a) 히알루론산염 파이버를 제조하는 단계; (b) 제조된 히알루론산염 파이버에 컬 (Curl) 처리하는 단계; (c) 컬 (Curl)이 부여된 히알루론산염 파이버를 절삭하는 단계; (d) 절삭된 히알루론산염 파이버로 웹을 형성하는 단계; 및 (e) 형성된 히알루론산염 웹을 니들 펀칭하는 단계를 포함하는 것을 특징으로 하는 히알루론산염 부직포의 제조방법을 제공한다.
본 발명에 있어서, 상기 히알루론산염 파이버를 제조하는 단계는 (a-1) 중량평균 분자량이 100∼3,000kDa인 히알루론산염의 수분함량을 50∼95중량%로 조절하여 히알루론산염 페이스트(paste)를 제조하는 단계; (a-2) 수분함량이 조절된 히알루론산염 페이스트(paste)를 15∼100℃로 용융시킨 뒤, 노즐을 통해 압출시키는 단계; 및 (a-3) 압출된 방사유체를 건조시켜 파이버를 형성하는 단계를 포함하는 것을 특징으로 한다.
본 발명에 있어서, 상기 웹을 형성 시 콜라겐, 섬유결합소, 젤라틴, 키토산, 알긴산, 폴리락트산(PLA), 폴리글루탐산(PGA), 폴리락티드-글리콜리드 공중합체(PLGA) 및 폴리카프로락톤(PCL)로 구성된 군에서 선택되는 고분자 소재로 제조된 파이버를 혼합시키는 것을 특징으로 한다.
본 발명의 히알루론산염 부직포는 히알루론산염을 고농도로 함유하고, 고강도 히알루론산염 파이버를 니들 펀칭하여 제조되며, 우수한 점착력 및 인장강도를 갖는다.

본 발명에서는 특정 범위로 수분함량이 조절된 히알루론산염 페이스트(paste)를 일정 온도에서 노즐을 통해 방사하면서 상온이 아닌 고온에서 건조시킬 경우, 파이버 표면이 매우 거친 히알루론산염 파이버를 제조할 수 있고, 이를 니들 펀칭시킬 경우 점착력 및 기계적 강도가 우수한 부직포를 제조할 수 있다는 것을 확인하고자 하였다.
본 발명에서는 히알루론산염의 함수율을 50~95중량%로 조절하여 페이스트(paste)로 제조하고, 히알루론산염 페이스트(paste)의 온도를 15~100℃로 용융시킨 뒤 노즐을 통해 압출시킨 후, 100~150℃에서 건조하여 수분함량이 5~25중량%이고, 인장강도가 3~15 kg/㎟이며, 표면이 매우 거친 히알루론산염 파이버를 대량으로 제조하였다. 제조된 히알루론산염 파이버에 열을 가하여 컬 (Curl)를 부여 후 일정 길이로 절삭하고, 카딩과 에어레이딩 과정에 의해 웹을 형성한 다음 니들 펀칭하여 형성된 부직포를 압착시켜 점착력 및 인장강도가 우수한 부직포를 제조하였다.
따라서, 본 발명은 (a) 히알루론산염 파이버를 제조하는 단계; (b) 제조된 히알루론산염 파이버에 컬 (Curl) 처리하는 단계; (c) 컬 (Curl)이 부여된 히알루론산염 파이버를 절삭하는 단계; (d) 절삭된 히알루론산염 파이버로 웹을 형성하는 단계; 및 (e) 형성된 히알루론산염 웹을 니들 펀칭하는 단계를 포함하는 것을 특징으로 하는 히알루론산염 부직포의 제조방법에 관한 것이다.
본 발명에 있어서, 상기 히알루론산염 파이버를 제조하는 단계는 (a-1) 중량평균 분자량이 100∼3,000kDa인 히알루론산염의 수분함량을 50∼95중량%로 조절하여 히알루론산염 페이스트 (paste)를 제조하는 단계; (a-2) 수분함량이 조절된 히알루론산염 페이스트 (paste)를 15∼100℃로 용융시킨 뒤, 노즐을 통해 압출시키는 단계; 및 (a-3) 압출된 방사유체를 건조시켜 파이버를 형성하는 단계를 포함한다.
본 발명에 있어서, 히알루론산염은 히알루론산에 염이 결합되어 있는 것으로서, 히알루론산나트륨, 히알루론산칼슘, 히알루론산칼륨 등을 예시할 수 있으나 이에 한정되는 것은 아니다.
본 발명에 있어서, 히알루론산염의 분자량은 100~3,000kDa인 것이 바람직하다. 또한, 상기 히알루론산염 페이스트 (paste)의 히알루론산염의 함수량은 50~95중량%인 것을 특징으로 한다.
상기 히알루론산염 페이스트 (paste)는 히알루론산염에 용매를 가하여 제조할 수 있는데, 상기 용매는 물 또는 에탄올 수용액을 사용하며, 용매가 균일하게 혼합될 때까지 혼합시키는 것이 바람직하다.
상기 대량 방사공정을 원활하게 수행하기 위하여 히알루론산염 페이스트 (paste)의 수분 함량은 50~95중량%, 바람직하게는 88~92중량%일 수 있는데, 수분 함량이 50중량% 미만인 경우 방사 시 높은 압력이 필요하며, 수분 함량이 95중량%를 초과할 경우 페이스트 (paste) 보다는 액상의 형태이기 때문에 방사 시 실이 끊어지는 문제점이 있다.
본 발명에서는 히알루론산염 페이스트 (paste) 제조 후, 수분 평형을 위하여 0~100℃에서 6~12시간동안 방치하는 단계를 수행하는 것을 특징으로 한다. 상기 온도 범위 및 시간 범위를 벗어날 경우에는 수분 평형이 제대로 이루어지지 않는다. 상기 수분 평형은 페이스트 (paste)의 물분자와 히알루론산이 고르게 섞이도록 하기 위한 것으로서, 수분평형 없이 방사를 수행할 경우 물 분자와 히알루론산이 고르게 섞여있지 않으므로 방사 시 일정한 속도로 방사를 수행할 수 없게 된다.
본 발명에서는 수분 함량이 50∼95중량%로 조절된 히알루론산염 페이스트 (paste)를 방사 장치에 넣고, 15∼100℃로 용융시킨 후, 노즐을 통해 압출시켜 방사유체를 제조할 수 있다. 이때 온도가 15℃ 미만인 경우에는 히알루론산염 페이스트 (paste)가 용융되지 않아 압출이 되지 않고, 100℃를 초과할 경우에는 반죽이 끓어오르는 문제가 있다.
본 발명에서는 압출된 방사유체를 건조시킴으로써 최종적으로 파이버를 형성하는데, 상기 건조는 페이스트 (paste)의 수분함량에 따라 100~150℃에서 건조하는 것이 바람직하다. 건조온도가 100℃ 미만인 경우에는 제조된 파이버의 표면이 매끄럽기 때문에 웹 형성 후 니들펀칭 과정에서 엉키는 힘이 약해 부직포 형성시 어려움이 있고, 150℃를 초과할 경우에는 파이버가 갈변되고, 크랙이 발생되는 문제가 있다.
제조된 히알루론산염 파이버는 히알루론산염의 함량을 90중량% 이상으로 제조가 가능하며, 수분함량이 5~25중량%이며, 인장강도가 3~15kg/㎟이고, 표면이 매우 거칠기 때문에 이를 이용하여 부직포로 제조할 경우, 다양한 조밀도와 물성을 갖는 부직포를 제조할 수 있다.
본 발명의 히알루론산염 부직포의 제조방법은 제조된 파이버를 서로 간의 교차력 증대를 위해 열 처리 등에 의해 컬 (Curl)을 부여하는 것을 특징으로 한다. 열처리의 경우, 히알루론산염 파이버를 70℃ 이상에서 고온에서 열풍 처리함으로써, 컬 (Curl) 부여가 가능하다.
부직포 제조를 위한 웹을 형성하기 위하여, 제조된 히알루론산염 파이버를 목적하는 길이로 절삭시키고, 절삭된 히알루론산염 파이버를 공기압에 의해 분산 후 이를 공기압으로 사출시키는 카딩 및 에어레이드 공정으로 웹을 형성시킬 수 있다.
본원발명의 니들 펀칭단계는 상기 형성된 평면 시트상 웹을 니들로 펀칭하는 단계이다. 구체적으로, 니들 펀칭단계는 Needle Loom을 통과 시킬 때 니들이 붙은 침판을 상하 반복 운동시켜 2차원적인 랜덤한 섬유 배열의 일부를 3차원적인 랜덤 구조로 결합시켜 부직포를 제조할 수 있다.
본 발명의 히알루론산염 부직포의 제조방법은 웹을 형성 시 생체 적합성 소재로 제조된 파이버를 혼합시킬 수 있다. 즉, 보다 다양한 물성과 기능성 확보를 위하여 약리학적으로 허용되는 생체 적합성 소재이며 파이버 제조가 가능한 콜라겐, 섬유결합소, 젤라틴, 키토산, 알긴산, 폴리락트산(PLA), 폴리글루탐산(PGA), 폴리락티드-글리콜리드 공중합체(PLGA), 폴리카프로락톤(PCL) 등을 히알루론산 파이버와 혼합할 수 있다.
본 발명은 다른 관점에서, 하기 측정방법으로 측정 시, 휨 각도가 2~10°이고, 수분함량이 5~25중량%이며, 수분을 제외한 전체 파이버 중량에 대하여 히알루론산염 함량이 90중량% 이상인 것을 특징으로 하는 히알루론산염 파이버를 포함하는 히알루론산염 부직포에 관한 것이다.

상기 히알루론산염 파이버의 인장강도는 3~15 kg/㎟이므로, 니들 펀칭으로 부직포 제조가 가능하다. 만일 히알루론산염 파이버의 인장강도가 3kg/㎟ 미만일 경우, 파이버가 끊어질 가능이 커서 가공이 어렵고, 15kg/㎟를 초과할 경우, 재질이 딱딱하여, 부직포로 제조된 후 사용상 불편함을 초래할 수 있다.
본 발명의 히알루론산염 부직포는 두께가 1~10㎜인 것이 바람직하며, 제조된 부직포는 부직포의 두께에 따라 0.3~1.5 kg/㎟의 인장강도를 갖고, 점착력이 0.9~2.7 kPa인 것을 특징으로 한다.
본 발명의 히알루론산염 부직포는 점착력 및 인장 강도가 우수하여 피부 외용제용 마스크팩, 조직 수복용 생체재료, 유착방지 피복재 등의 용도로 사용 가능하다.
이하, 실시예를 통하여 본 발명을 더욱 상세히 설명하고자 한다. 이들 실시예는 오로지 본 발명을 예시하기 위한 것으로, 본 발명의 범위가 이들 실시예에 의해 제한되는 것으로 해석되지 않는 것은 당 업계에서 통상의 지식을 가진 자에게 있어서 자명할 것이다.
실시예 1: 히알루론산염 페이스트 제조
분자량 150kDa인 히알루론산나트륨 (Hi-AquaTM, (주)진우바이오)에 물을 가하고, 혼합 반죽하여 수분함량이 50~95중량%인 히알루론산염 페이스트 (paste)를 제조하였다. 제조된 히알루론산염 페이스트 (paste)는 4℃에서 12시간동안 냉장보관하여 수분평형을 유도하였다.
실시예 2: 방사에 의한 히알루론산염 파이버 제조
실시예 1에서 제조된 히알루론산염 페이스트(paste)를 방사 장치의 저장부에 넣고, 이를 15~100℃에서 용융시킨 뒤 가압하여 노즐을 통해 방사하고, 120℃에서 건조하여 수분함량이 5~25중량%인 히알루론산염 파이버를 제조하였다.
비교예 1: 방사에 의한 히알루론산염 파이버 제조
실시예 1에서 제조된 히알루론산염 페이스트 (paste)를 방사 장치의 저장부에 넣고, 이를 15~100℃에서 용융시킨 뒤 가압하여 노즐을 통해 방사하고, 50℃에서 건조하여 수분함량이 5~25중량%인 히알루론산염 파이버를 제조하였다.
비교예 2: 방사에 의한 히알루론산염 파이버 제조
실시예 1에서 제조된 히알루론산염 페이스트 (paste)를 방사 장치의 저장부에 넣고, 이를 15~100℃에서 용융시킨 뒤 가압하여 노즐을 통해 방사하고, 160℃에서 건조하여 히알루론산염 파이버를 제조하였다.
실험예 1: 히알루론산염 파이버 건조 온도에 따른 파이버 거칠기 평가
실시예 2, 비교예 1 및 비교예 2에서 제조된 파이버의 표면 거칠기를 현미경으로 평가하였다 (도 1).
그 결과, 50℃(비교예 1), 120℃(실시예 2) 건조 조건에서 수분함량이 5~25중량%인 파이버가 방사되었지만, 50℃에서 건조한 파이버의 경우 표면이 매끄러운 형태로 방사되었고, 120℃에서 건조한 파이버의 경우 표면이 거친 형태로 방사되었다. 건조온도가 160℃인 비교예 2는 수분함량이 1~5중량%로 파이버의 갈변과 크랙이 과도하게 형성되는 것을 확인하였다.
실험예 2: 히알루론산염 파이버의 휨 각도 측정
실시예 2에서 제조된 히알루론산염 파이버의 휨 각도를 다음의 방법으로 측정하고(도 2 참조), 그 결과를 표 1에 나타내었다.

상기 표 1로부터, 히알루론산염 파이버의 수분 함량이 25중량%를 초과할 경우 휨 각도가 10°를 초과함으로서, 웹을 형성하거나 니들 펀칭으로 부직포 제조가 쉽지 않고, 부직포로 제조되어도 물성이 떨어져 제품화를 위한 타발 등의 가공 작업이 불가하여 상용화에 문제가 있을 것으로 판단되었다.
또한, 히알루론산염 파이버의 함수율이 5중량% 미만인 경우에는 파이버의 형태는 유지하지만 단단하게 굳어있는 형태로 유연성이 없어 쉽게 부러지는 것을 확인할 수 있었다.
반면, 히알루론산염 파이버의 함수율이 5~25중량%인 경우에는 휨 각도가 2~10°로서, 니들펀칭으로 부직포 제조시 적합한 형태의 히알루론산염 파이버임을 알 수 있었다.
실험예 3: 히알루론산염 파이버의 인장강도 측정
실시예 2에서 제조된 히알루론산염 파이버의 인장강도를 측정하고, 그 결과를 표 2에 나타내었다. 인장강도는 길이가 5cm인 시료를 시험지그에 장착하고, 크로스헤드 속도를 2㎜/min로 하여 만능재료시험기를 이용하여 측정하였다.

상기 표 2로부터, 히알루론산염 파이버의 함수율이 50중량% 이상인 경우에는 인장강도가 급격히 낮아지며 쉽게 끊어지며 탄력이 없어 파이버의 기능을 제대로 수행하지 못할 것으로 판단되었다.
또한, 히알루론산염 파이버의 함수율이 5중량% 이하인 경우에는 파이버의 형태는 유지되지만 단단하게 굳어있어 쉽게 부러져 인장강도를 측정할 수 없었다.
반면, 히알루론산염 파이버의 함수율이 5~25중량%인 경우에는 인장강도가 3.24~13.38kg/㎟로서, 실의 형태와 기능을 할 수 있는 히알루론산염 파이버임을 알 수 있었다.
실시예 3: 니들 펀칭에 의한 히알루론산염 부직포 제조
1) 히알루론산염 파이버 제조
상기 실시예 2의 방법으로 휨 각도가 2~10°이고, 수분함량이 5~25중량%이며, 수분을 제외한 전체 파이버 중량에 대하여 히알루론산염 함량이 90중량% 이상인 히알루론산염 파이버(인장강도: 3~15 kg/㎟) 를 제조하였다 (도 3 A).
2) Curling 처리
제조된 히알루론산염 파이버를 70~100℃에서 열풍 처리하여 컬 (Curl)이 부여된 히알루론산염 파이버를 제조하였다 (도 3 B).
3) 절삭 (Cutting)
컬이 부여된 히알루론산염 파이버를 절삭기를 사용하여 3~7cm의 일정 길이로 절단하였다.
4) 카딩과 에어레이드에 의한 웹 형성
절삭된 히알루론산염 파이버를 공기압에 의해 분산 후 이를 공기압으로 사출시켜 웹을 형성하였다.
5) 니들 펀칭
웹으로 형성된 다중층 (Multi-Layer)의 시트를 니들 펀칭 과정에 의해 균일한 부직포(평균 두께 5㎜)를 제조하였다 (도 4).
비교예 3: 니들 펀칭에 의한 히알루론산염 부직포 제조
상기 비교예 1의 방법으로 휨 각도가 2~10°이고, 수분함량이 5~25중량%이며, 수분을 제외한 전체 파이버 중량에 대하여 히알루론산염 함량이 90중량% 이상인 히알루론산염 파이버(인장강도: 3~15 kg/㎟)를 제조하고, 이를 이용하여 실시예 3과 동일한 방법으로 히알루론산염 부직포를 제조하였다.
그 결과, 표면이 매끄러운 히알루론산염 파이버를 이용할 경우 웹 형성 후 니들 펀칭 과정에서 파이버끼리 엉키는 힘이 약해 부직포가 잘 형성되지 않는 것을 확인할 수 있었다.
실시예 4: 니들 펀칭에 의한 히알루론산염과 키토산이 혼합된 부직포 제조
1) 히알루론산염 파이버 제조
상기 실시예 2의 방법으로 휨 각도가 2~10°이고, 수분함량이 5~25중량%이며, 수분을 제외한 전체 파이버 중량에 대하여 히알루론산염 함량이 90중량% 이상인 히알루론산염 파이버(인장강도: 3~15 kg/㎟) 를 제조하였다.
2) 키토산 섬유
제조처: 새턴바이오텍, 직경 0.006~0.018mm
3) Curling 처리
히알루론산염 파이버를 70~100℃에서 고온에서 열풍 처리하여 컬 (Curl)이 부여된 히알루론산염 파이버를 제조하였다.
키토산 파이버는 100℃ 이상의 수증기로 열처리하거나 히알루론산염과 동일한 열처리 과정에 의해 컬 (Curl)이 부여된 키토산 파이버를 제조하였다.
4) 절삭 (Cutting)
컬이 부여된 히알루론산염과 키토산 파이버를 절삭기를 사용하여 3~7cm의 일정 길이로 절단하였다.
5) 카딩과 에어레이드에 의한 웹 형성 및 니들 펀칭
절삭된 히알루론산염 파이버와 키토산 파이버를 50:50 중량% 비율로 혼합하여 실시예 3과 동일하게 처리하여 최종 히알루론산염과 키토산이 혼합된 부직포(평균 두께 5㎜)를 제조하였다 (도 5).
실험예 4: 부직포의 미세 구조 확인
현미경 (CX33RTFS2, Olympus Corporation., Korea)을 사용하여 실시예 3 및 실시예 4에서 제조된 히알루론산염 부직포의 미세 구조를 확인하였다 (도 6). 파이버 간에 교차결합 된 웹 형태의 부직포 구조를 확인할 수 있었으며, 특히 키토산이 혼합된 부직포의 경우, 히알루론산염 파이버와 키토산 파이버가 잘 혼합되어있는 것을 확인하였다. 웹 형태의 부직포는 습식방사를 통한 부직포에 비해 파이버간의 교차결합 사이에 공간이 있어 보다 물성이 뛰어나 가공과 사용이 쉬운 특징이 있다.
실험예 5: 히알루론산염 부직포의 겔화도 측정
니들 펀칭에 의한 순수 히알루론산염 부직포(실시예 3)와 히알루론산염과 키토산이 각각 50중량%씩 함유된 부직포(실시예 4)의 겔화도를 다음과 같이 측정하였다.
2 x 2 cm의 순수 히알루론산염 부직포와 히알루론산염과 키토산이 각각 50%씩 함유된 부직포를 정제수에 5초간 침지 후 꺼내어 부직포 외부의 수분을 제거한 후 무게를 측정하여 겔화도를 측정하였다. 겔화도는 아래와 같이 계산하였다.

키토산이 함유된 부직포의 경우 물에 대한 용해도가 매우 낮아 순수 히알루론산염에 부직포에 비해 겔화도가 낮음을 알 수 있었다.
실험예 6: 히알루론산염 부직포의 점착력 테스트
Texture Analyser (TA) (QM100SN, Qmesys co. Ltd., Korea)를 사용하여 니들 펀칭에 의한 순수 히알루론산염 부직포(실시예 3, 수분함량 25중량% 순수 히알루론산염 부직포)와 히알루론산염과 키토산이 각각 50%씩 함유된 부직포(실시예 4, 수분함량 25중량% 히알루론산염 + 키토산 부직포, 50:50 중량% 비율)의 점착력을 다음과 같이 측정하였다.
돈피를 3 x 3 cm 직사각형으로 자른 뒤, 상온수에 24시간 동안 방치하여 수분 함량이 22.3%로 조절 후 TA 위, 아래 probe에 부착 시킨 후 아래 돈피 위에 2 x 2 cm 직사각형의 순수 히알루론산염, 키토산이 혼합된, 키토산만으로 이루어진 부직포를 올려놓고 3.2kPa로 3분간 눌러 준 뒤, 10 mm/min의 속도로 떼어내어 점착력을 측정하였다.

키토산 부직포 0
순수 키토산만으로 이루어진 부직포의 경우 점착력이 없음을 확인 할 수 있었다. 순수 히알루론산염으로 이루어진 부직포의 경우 점착력이 키토산이 혼합된 부직포에 비해 높음을 알 수 있는데 이는 겔화도가 높아 점착력이 높아지기 때문이다.
또한, 키토산이 혼합된 부작포의 경우에도 일정 점착력을 나타냄을 알 수 있었다.
이를 통해 적정량의 히알루론산염과 키토산을 혼합하여 부직포 제조를 통해 겔화도 조정을 통한 체내 지속성 조절과 점착력 부여를 통한 타켓팅 능력 부여를 통해 고물성의 부직포 제조가 가능함을 알 수 있었다.
실험예 7: 히알루론산염 부직포의 인장강도 측정
니들 펀칭에 의한 순수 히알루론산염 부직포(실시예 3, 수분함량 25중량% 순수 히알루론산염 부직포)와 히알루론산염과 키토산이 각각 50%씩 함유된 부직포(실시예 4, 수분함량 25중량% 히알루론산염 + 키토산 부직포, 50:50 중량% 비율)의 인장강도를 측정하고, 그 결과를 표 5에 나타내었다. 인장강도는 길이가 5cm인 시료를 시험지그에 장착하고, 크로스헤드 속도를 2㎜/min로 하여 만능재료시험기를 이용하여 측정하였다.

표 5로부터, 순수 히알루론산염 파이버에 키토산 섬유를 혼합했을 때, 더 높은 인장강도를 나타냄을 알 수 있었다. 부직포 형성 시, 키토산 섬유를 혼합하게 되면, 히알루론산염 부직포의 사이에 키토산 섬유가 엉키게 되어 더욱 강한 결합력을 나타냄을 알 수 있었다.
이상으로 본 발명 내용의 특정한 부분을 상세히 기술하였는바, 당업계의 통상의 지식을 가진 자에게 있어서 이러한 구체적 기술은 단지 바람직한 실시 양태일 뿐이며, 이에 의해 본 발명의 범위가 제한되는 것이 아닌 점은 명백할 것이다. 따라서 본 발명의 실질적인 범위는 첨부된 청구항들과 그것들의 등가물에 의하여 정의된다고 할 것이다.
본 발명의 히알루론산염 부직포는 우수한 점착력 및 인장강도를 가지므로, 피부 외용제용 마스크팩, 조직 수복용 생체재료, 유착방지 피복재 등 다양한 분야로 활용 가능하다.

Claims (10)

  1. 하기 측정방법으로 측정 시, 휨 각도가 2~10°이고, 수분함량이 5~25중량%이며, 수분을 제외한 전체 파이버 중량에 대하여 히알루론산염 함량이 90중량% 이상인 것을 특징으로 하는 히알루론산염 파이버를 포함하는 히알루론산염 부직포.
    <휨 각도 측정 방법>
    (a) 길이가 20㎝이고, 지름이 1㎜인 HA 파이버를 준비
    (b) 평평한 판의 윗면에 HA 파이버 한쪽 끝 3㎝를 고정
    (c) 평평한 판의 윗면 연장선(B)을 기준으로 HA 파이버 다른 쪽 끝의 연장선(T)이 이루는 각도(θ)를 측정
  2. 제1항에 있어서, 상기 히알루론산염 파이버의 인장강도는 3~15 kg/㎟인 것을 특징으로 하는 히알루론산염 부직포.
  3. 제1항에 있어서, 상기 히알루론산염 부직포의 점착력은 0.9~2.7 kPa인 것을 특징으로 하는 히알루론산염 부직포.
  4. 제1항에 있어서, 상기 히알루론산염 부직포의 두께는 1~10㎜인 것을 특징으로 하는 히알루론산염 부직포.
  5. 제1항에 있어서, 상기 히알루론산염 부직포의 인장강도는 0.3~1.5 kg/㎟인 것을 특징으로 하는 히알루론산염 부직포.
  6. 제1항에 있어서, 상기 히알루론산염 부직포는 피부 외용제용 마스크팩, 조직 수복용 생체재료 또는 유착방지 피복재 용도인 것을 특징으로 하는 히알루론산염 부직포.
  7. (a) 히알루론산염 파이버를 제조하는 단계;
    (b) 제조된 히알루론산염 파이버에 컬 (Curl) 처리하는 단계;
    (c) 컬 (Curl)이 부여된 히알루론산염 파이버를 절삭하는 단계;
    (d) 절삭된 히알루론산염 파이버로 웹을 형성하는 단계; 및
    (e) 형성된 히알루론산염 웹을 니들 펀칭하는 단계를 포함하는 것을 특징으로 하는 히알루론산염 부직포의 제조방법.
  8. 제7항에 있어서, 상기 히알루론산염 파이버를 제조하는 단계는
    (a-1) 중량평균 분자량이 100∼3,000kDa인 히알루론산염의 수분함량을 50∼95중량%로 조절하여 히알루론산염 페이스트(paste)를 제조하는 단계;
    (a-2) 수분함량이 조절된 히알루론산염 페이스트(paste)를 15∼100℃로 용융시킨 뒤, 노즐을 통해 압출시키는 단계; 및
    (a-3) 압출된 방사유체를 건조시켜 파이버를 형성하는 단계를 포함하는 것을 특징으로 하는 히알루론산염 부직포의 제조방법.
  9. 제7항에 있어서, 상기 제조된 히알루론산염 파이버의 인장강도는 3~15kg/㎟인 것을 특징으로 하는 히알루론산염 부직포의 제조방법.
  10. 제7항에 있어서, 상기 웹을 형성 시 콜라겐, 섬유결합소, 젤라틴, 키토산, 알긴산, 폴리락트산(PLA), 폴리글루탐산(PGA), 폴리락티드-글리콜리드 공중합체(PLGA) 및 폴리카프로락톤(PCL)로 구성된 군에서 선택되는 고분자 소재로 제조된 파이버를 혼합시키는 것을 특징으로 하는 히알루론산염 부직포의 제조방법.
