核酸溶液を導入し分割するための複数の貫通孔を有する基板と、
前記基板の第1の面と、前記基板の前記第1の面とは反対側の第2の面とにおいて、前記貫通孔をふさぐために覆うオイルと、
前記基板を加熱するために前記第2の面の側に設けられた熱伝導プレートと、
前記第1の面の側から照射される励起光の反射を抑制する反射抑制機構と、
を有することを特徴とする。
計測デバイスを用いた測定方法であって、
前記計測デバイスは、
複数の貫通孔を有する基板と、
前記基板の温度を変化させるために設けられた熱伝導プレートと、
前記基板の前記熱伝導プレート側とは反対側の第1の面の側から照射される励起光の反射を抑制する反射抑制機構と、
を有し、
前記測定方法は、
前記基板に対して、核酸溶液を導入し分画する工程と、
前記基板の第1の面と、前記基板の前記第1の面とは反対側の第2の面とにおいて、前記核酸溶液が導入された前記貫通孔をふさぐために覆うオイルを導入する工程と、
前記熱伝導プレートにより前記基板の温度を変化させる工程と、
温度変化させながら前記基板の前記第1の面の側から励起光を照射し、前記第1の面の側から蛍光を検出し、前記核酸溶液を測定する工程と、
を有する。
図1は、貫通孔ウェル1を保持した従来の計測デバイスにおいて蛍光測定した際の概略図である。この計測デバイスは複数の貫通孔ウェル1を保持した基板2、ウェル内に注入した核酸と蛍光標識プローブの混合溶液3、貫通孔ウェル1を覆うオイル4、熱伝導プレート5で構成されている。
[実施例1]
以下、図3および図4を用いて、実施例1について説明する。実施例1は、熱伝導プレートを黒色処理することで気泡が観察されにくくするものである。本実施例に係る計測デバイスを用いることで、気泡の影響なく蛍光強度測定が可能になる。
以下、図5と図6を用いて、実施例2について説明する。実施例2は、貫通孔ウェル400の下部に微粒子401を注入することで、気泡402が観察されにくくするものである。本実施例に係る計測デバイスを用いることで、気泡402の影響なく蛍光強度測定が可能になる。以下、実施例1と共通する部分については、説明を省略する場合がある。
(微粒子断面積)×(注入する微粒子の個数)≦(貫通孔ウェル底面積)…(数式1)
を満たすようにすると好適である。なお、必ずしも貫通孔ウェルの底面積全てを微粒子で覆う必要はない。ある例では、ウェルの底面積の10%程度を微粒子で覆うという条件を用い、気泡の影響が低減することが確認されている。
以下、図7を用いて、実施例3について説明する。実施例3は、反射抑制機構として、計測デバイスの下部を透明な熱伝導プレート500にすることで、気泡501が観察されにくくするものである。本実施例に係る計測デバイスを用いることで、気泡501の影響なく蛍光強度測定が可能になる。以下、実施例1または2と共通する部分については、説明を省略する場合がある。
透明な熱伝導プレート500の材質の例として、ポリカーボネートなどのプラスチックや、ガラスなどが挙げられる。また、透明な熱伝導プレート500は、ガラス基板に酸化インジウムスズなどをドープした透明導電性基板などであってもよい。
以下、図8を用いて、実施例4について説明する。実施例4は、凹凸構造の熱伝導プレート600を用いることで、気泡601が観察されにくくするものである。本実施例に係る計測デバイスを用いることで、気泡601の影響なく蛍光強度測定が可能になる。以下、実施例1~3と共通する部分については、説明を省略する場合がある。
以下、図9を用いて、実施例5について説明する。実施例5は、貫通孔を覆うオイルを着色することで、気泡700が観察されにくくするものである。本実施例に係る計測デバイスを用いることで、気泡700の影響なく蛍光強度測定が可能になる。以下、実施例1~4と共通する部分については、説明を省略する場合がある。
以下、図10を用いて、実施例6について説明する。実施例6は、貫通孔ウェル800下部にインク801を導入することで、気泡802が観察されにくくするものである。本実施例に係る計測デバイスを用いることで、気泡802の影響なく蛍光強度測定が可能になる。以下、実施例1~5と共通する部分については、説明を省略する場合がある。
以下、図11と図12を用いて、実施例7について説明する。実施例7は、実施例1に係る計測デバイスを用いることで、実際に気泡が観察されなくなる効果を示したものである。
以下、図13から図15を用いて、実施例8について説明する。実施例8は、実施例1~6の計測デバイスを用いた融解曲線分析の測定方法を示したものである。本実施例に係る計測デバイスを用いることで、融解曲線分析の高精度な測定が可能となる。
以下、図16と図17を用いて、実施例9について説明する。実施例9は、実施例1の熱伝導プレートの黒色表面処理や実施例2の微粒子の貫通孔ウェルの注入により、気泡を観察されにくくする効果が得られることを確認する方法を示したものである。

- 計測デバイスであって、
核酸溶液を導入し分割するための複数の貫通孔を有する基板と、
前記基板の第1の面と、前記基板の前記第1の面とは反対側の第2の面とにおいて、前記貫通孔をふさぐために覆うオイルと、
前記基板を加熱するために前記第2の面の側に設けられた熱伝導プレートと、
前記第1の面の側から照射される励起光の反射を抑制する反射抑制機構と、
を有することを特徴とする、計測デバイス。 - 前記反射抑制機構は、前記貫通孔に導入される微粒子を備え、前記微粒子は親水性コーティングされており、前記微粒子は前記核酸溶液よりも比重が重いことを特徴とする、請求項1に記載の計測デバイス。
- 前記微粒子の前記比重は、1g/cm3以上であることを特徴とする、請求項2に記載の計測デバイス。
- 前記反射抑制機構は、前記貫通孔に導入されるインクを備え、前記インクは前記核酸溶液よりも比重が重いことを特徴とする、請求項1に記載の計測デバイス。
- 前記インクの比重は、1g/cm3以上であることを特徴とする、請求項4に記載の計測デバイス。
- 前記反射抑制機構は反射抑制膜を備え、前記反射抑制膜は前記熱伝導プレート上に配置され、前記反射抑制膜は前記励起光を吸収することを特徴とする、請求項1に記載の計測デバイス。
- 前記反射抑制機構は凹凸構造を備え、前記凹凸構造は前記熱伝導プレート上に配置され、前記凹凸構造のピッチは1μm以下であることを特徴とする、請求項1に記載の計測デバイス。
- 前記反射抑制機構は、前記熱伝導プレートの有色の表面処理構造であることを特徴とする、請求項1に記載の計測デバイス。
- 前記熱伝導プレートの材質は、金属、樹脂、ガラスのいずれかであることを特徴とする、請求項1に記載の計測デバイス。
- 前記オイルは有色であることを特徴とする、請求項1に記載の計測デバイス。
- 前記有色は、波長400nm~波長700nmにわたって反射率が10%以下であることを意味することを特徴とする、請求項8または10に記載の計測デバイス。

前記測定方法は、
前記基板に対して、核酸溶液を導入し分画する工程と、
前記基板の第1の面と、前記基板の前記第1の面とは反対側の第2の面とにおいて、前記核酸溶液が導入された前記貫通孔をふさぐために覆うオイルを導入する工程と、
前記熱伝導プレートにより前記基板の温度を変化させる工程と、
温度変化させながら前記基板の前記第1の面の側から励起光を照射し、前記第1の面の側から蛍光を検出し、前記核酸溶液を測定する工程と、
