- variable regions are encoded by various genes that rearrange to form antibodies specific for foreign antigens, Contains an antigen binding domain.
- the variable regions can be further modified by a number of single amino acid changes (somatic mutations or hypermutations) to increase the affinity of the antibody for the foreign antigen.
- the constant region will change upon further response to antigen (ie isotype switch).

However, the term “human antibody” does not include antibodies in which CDR sequences derived from the germline of another mammalian species, such as a mouse, have been grafted onto human framework sequences. The terms "human” antibody and "fully human” antibody are used synonymously.
- humanized antibody refers to an antibody in which some, most or all of the amino acids outside the CDR domains of a non-human antibody are substituted with corresponding amino acids derived from human immunoglobulin. Additions, deletions, insertions, substitutions or modifications of amino acids are permitted as long as they do not abrogate the ability of the antibody to bind a particular antigen. A "humanized" antibody retains an antigenic specificity similar to that of the original antibody.
- chimeric antibody refers to an antibody in which the variable regions are derived from one species and the constant regions are derived from another species, such as an antibody in which the variable regions are derived from a mouse antibody and the constant regions are derived from a human antibody. .

- Cells were pretreated with NN2101 (10 ⁇ g/ml) for 6 hours and then treated with 50 ⁇ M CoCl 2 .
- 10 ⁇ mol/L of MG132 or CHx was added, respectively, for the indicated period.
- the total white matter expression level of HIF-1 ⁇ in cells was analyzed by Western blot. The density of each protein band was determined with an image analysis system and normalized to that of the corresponding ⁇ -tubulin.
- HUVEC Longza, Walkersville, MD, USA
- HUVEC cells were overexpressed using expression vectors encoding three isotypes of PHD, ie, GFP-PHD1, GFP-PHD2, and GFP-PHD3, to which Green Fluorescent Protein (GFP) was attached.
- Transfected cells were incubated under hypoxic or normoxic conditions simultaneously with treatment with 10 ⁇ g NN2101 as indicated for 12 hours, respectively.
- Cells were treated with or without 10 ⁇ mol/L of MG132 for 3 hours before harvesting.
- HIF-1 ⁇ was selectively immunoprecipitated (IP) from whole cell lysates, followed by analysis of HIF-1 ⁇ bound to overexpressed PHD (HIF-1 ⁇ -PHD) by Western blotting (FIG. 9).
- Example 10 NN2101 reduces the upregulation of pro-angiogenesis/inflammation factors, including HIF target genes, under hypoxic conditions in primary retinal cells.
- HRMEC Human retinal microvascular endothelial cell
- ARPE19 spontaneous retinal pigment epithelial cell
- MIO-M1 Moorfields/Institute of Ophthalmology-Mueller 1; spontaneoulsy immortalized human Mueller glia cell line

- HRMEC were incubated overnight with EBM containing 0.5% FBS, and the cells were pretreated with NN2101 antibody or Eylea (10 ⁇ g/ml) for 6 hours and further treated with 100 ⁇ M CoCl 2 for 18 hours. Culture supernatants were analyzed with a proteome profiler and quantified. As a result of the above analysis, it was confirmed that NN2101 regulates angiogenic/inflammatory factors that are increased in HRMEC under hypoxic conditions (FIG. 10).
- ARPE19 cells were cultured overnight in DMEM/F12 containing 0.5% FBS and cells were pretreated with NN2101 mAb or Eylea (10 ⁇ g/ml) for 6 hours, followed by further treatment with 100 ⁇ M CoCl 2 . The 18-hour culture supernatant was analyzed with a proteome profiler and quantified. As a result of the above analysis, it was confirmed that NN2101 regulates angiogenic/inflammatory factors that are increased in RPE under hypoxic conditions (FIG. 11).
- MIO-M1 cells were cultured overnight with DMEM containing 0.5% FBS, cells were pretreated with NN2101 mAb or Eylea (10 ⁇ g/ml) for 6 hours, and then treated with 100 ⁇ M CoCl 2 . further processed. The 18-hour culture supernatant was analyzed with a proteome profiler and quantified. As a result of the above analysis, it was confirmed that NN2101 regulates angiogenic/inflammatory factors that are increased in MIO-M1 cells under hypoxic conditions (FIG. 12).
- Y79 cells were incubated overnight with RPMI1640 containing 0.5% FBS and cells were pretreated with NN2101 mAb or Eylea (10 ⁇ g/ml) for 6 hours, then further treated with 100 ⁇ M CoCl 2 for 18 hours . Culture supernatants were analyzed with a proteome profiler and quantified. - NN2101 regulates angiogenic/inflammatory factors derived from hypoxic conditions in each cell (FIGS. 10 to 13).
- NN2101 simultaneously reduced Angiopoietin-2 and VEGF and regulated other angiogenic factors, suggesting that faricimab, which simultaneously targets Angiopoietin-2 and VEGF, will have an effect equivalent to or better than that.
