Definitions
- the present invention relates to a method for testing genome base editing in an extracellular, in vitro environment (cell-free, in vitro system). Specifically, target DNA mutation by base editor without using cells and without a separate complicated experimental process. This is a method that can confirm and quantify.
- Target DNA can be amplified from cells and used, but more usefully, it can be used by imitating the mutant DNA of organisms that require base editing (for example, patients with genetic diseases) through double-stranded DNA synthesis.
- adenine base editor By measuring DNA editing efficiency without a cell line or model animal carrying a specific mutation, adenine base editor, cytosine base editor, DddA-derived cytosine base editor (DdCBE), TALE-linked deaminase (TALED), zinc finger deaminase (ZFD) It can be used as a primary test method for the base editing ability of various base editors such as finger deaminase)) and CRISPR system.
- base editing is a fusion protein that connects a base-converting enzyme such as deaminase to Cas9 nickase (nCas9) or Cas9 (dCas9) with dead catalytic ability.

Description

상기 가이드 RNA는 2개의 RNA, 즉 CRISPR RNA(crRNA) 및 트랜스활성화 crRNA(transactivating crRNA, tracrRNA)로 이루어져 있는 것일 수 있으며, 또는 crRNA 및 tracrRNA의 필수적 부분의 융합에 의해 생성된 단일 사슬 RNA(single-chain RNA, sgRNA)일 수 있다.

환자유래 소변세포(UDC: Urine derived cell)는 0.1% 젤라틴(Welgene)으로 코팅한 12-웰 Clear TC-Treated Multiple Well Plates(Corning)에 Renal Epithelial cell Growth Medium Bullet kit(REGM, Lonza) 배양액으로 5% CO2에서 37℃로 유지하여 배양하였다. UDC 세포를 96-웰 Clear TC-Treated Multiple Well Plates(Corning)에 1 웰당 0.6 x 104 세포수가 되도록 시딩(seeding)해 주었다.
