Process for the preparation of 9-halo-1, 4-pregnadien-11-ols
The invention relates to a new process for the preparation of 9-halo-1, 4-pregnadien-11-ols of the general formula
EMI1.1
in which R is hydrogen or a lower alkanol radical with 1-6 carbon atoms in the molecule and X is fluorine, chlorine or iodine, which consists in the fact that a 9ss, 118-oxido-1,4-pregnadiene of the general formula
EMI1.2
in which R has the meaning given above, with a hydrogen halide of the formula HX, in which X has the above meaning, preferably in the presence of an organic solvent and at a temperature in the range from -10 to +500, after which the alkanoyl groups are optionally reacted in saponified the product thus obtained.
It is known (Journ. Amer. Chem. Soc., B. 78 [1956], p. 5693) that the effect on sodium retention is abolished by the introduction of a 16-hydroxyl group in 9a-halo-steroid compounds, in particular in 9a-fluoro-steroid compounds without reducing the glucocorticoid effect. It has now been found that these compounds are highly active in clinical use without the undesirable side effect of causing edema, as has been the case with many steroids heretofore. A chemical process for making these compounds is therefore desirable.
The preparation of the compounds of the general formula II is described elsewhere.

The method is expediently do-l, 4-pregnadiene-16a, 17cx, 21-triol-3,20-dione in which the 16c and 21-position hydroxyl groups are esterified with a corresponding alkanoyl radical, and the reaction of this ester with the hydrogen halide is used advantageously in chloroform, carbon tetrachloride, toluene, benzene, xylene and the like. dgL carried out. The reaction is complete after a period of about 1/2 hour to 24 hours. Thereafter, the desired product can be obtained by recrystallization from an organic solvent or from a mixture of solvents, e.g. B. ethyl acetate / petroleum ether or can be purified by chromatographic separation.
The following examples are intended to explain the invention in more detail:

After evaporation of the chloroform at 00, the crystallization of the solid residue from ethyl acetate / petroleum ether (90-100) yields 98 mg of substance with an F = 229-231 (decomp.).
Recrystallization from ethyl acetate / petroleum ether (90-100) increases the melting point to 234-2350

The mixture is shaken at 50 for 20 h, then poured into ice water, neutralized with sodium bicarbonate and extracted with chloroform. The washed and dried extract is evaporated to dryness and the residue is dissolved in 20 ml of pyridine, 4 ml of acetic anhydride are added and the mixture is left to stand overnight at room temperature. After adding methanol and benzene, a solution is obtained which is evaporated to dryness. The residue is dissolved in 7.5 ml of the stationary phase of the system ethyl acetate: petroleum ether (90-100): methanol: water, 4: 3: 3: 2, and mixed with 15 g of the silica preparation known under the trade name "Celite". This mixture is applied to a column (3 × 49 cm) which consists of 120 g of silica (Celite) and is moistened with 60 ml of the stationary phase of the above system.
The column is then eluted with the mobile phase and the fractions containing the desired product are combined and evaporated to dryness. The residue is crystallized from acetone / petroleum ether and gives 18.2 mg of substance, which melts between 160 and 2100 after prior softening and foaming. Recrystallization does not change the melting point. The infrared spectrum is identical to that of an authentic sample.
