  Verfahren zur Herstellung mikrobieller Stoffwechselprodukte mit organspezifischer Wirkung 
Es ist bekannt, dass zwischen Mikroorganismen einerseits und differenzierten Zellsystemen bei
Mensch, Tier und Pflanze anderseits mannigfaltige enge Wechselbeziehungen bestehen. Als solche Wech- selbeziehungen imponieren beim gesunden Mikroorganismus vor allem die sogenannten Symbiosen wie z. B. beim Menschen auf den einzelnen Abschnitten der Darmschleimhaut mit Enterokokken, Bact. aci- dophilum, Bact.   Coli. Bact.   Bifidum usw., auf der Vaginalschleimhaut mit Döderlein'schen Stäbchen.
Eine Störung dieser Symbiosen, sei es durch Schädigung der Mikroorganismen (z. B. durch bacterizide
Stoffe) oder aber auch durch eine Schädigung des   zugehorigen   Gewebes durch physikalische oder chemi- sche Noxen (z.

   B. radioaktive Strahlen, cytotoxische Substanzen usw.) führen zu zum Teil schweren Stö- rungen im Wirtsorganismus. Im letzteren Falle wird die Symbiose dadurch gestört, dass die zugehörigen symbiontischen Mikroorganismen nicht mehr die ihrem natürlichen Standort entsprechenden, also physiolo- gischen, Gewebsverhältnisse vorfinden und deshalb zugrunde gehen. Die Folge davon ist ein circulus vitiosus, der sich auf Grund des Fortbestehens der gestörten Relation von Gewebe zur Standortflora schäd- lich auswirken muss. So wird z. B. durch Eliminierung der normalen Darmflora die Eisenresorption inhi- biert. Es besteht also eine funktionelle physiologische Organspezifität und Organaffinität dieser Mikro- organismen. 



   Ausser dieser physiologischen   Organ-bzw. Gewebsaffinität symbicntischerMikroorganismen   zum Ge- webe ihres natürlichen Standortes gibt es aber auch zahlreiche Wechselbeziehungen zwischen differen- zierten Organen oder Geweben zu ganz bestimmten organaffinen Mikroorganismen, die direkt oder in- direkt ganz bestimmte, meist chronisch rezidivierende Organerkrankungen oder Syndrome zur Folge ha- ben. Dabei spielt die Pathogenität oder Virulenz der beteiligten Mikroorganismen keine Rolle, sondern das wesentliche Moment für das Zustandekommen pathologischer Verhältnisse besteht in der Einwirkung der lebenden Mikroorganismen auf die lebenden Zellen des betreffenden Organs oder differenzierten Ge- webes. 



   Zur Verständlichmachung des Erfindungsgedankens sei ein praktisches Beispiel erwähnt :
Wenn hämolytische Streptokokken auf den Tonsillen oder dem lymphatischen Gewebe des Waldeyerschen Rachenringes zur Vermehrung kommen, so kann-aber muss nicht-zunächst das Krankheitsbild einer akuten Angina entstehen.

   Ausserdem erfolgt aber durch Einwirkung der lebenden Mikroorganismen auf das lebende Tonsillengewebe die Bildung spezifischer Substanzen-erfindungsgemäss als mikrobielle Stoffwechselprodukte mit organspezifischer Wirkung bezeichnet-die nun ihrerseits eine ausgesprochene Affinität zum differenzierten mesenchymalen Gewebe der Gelenkflächen, der Niere und des Herzens besitzen und dort imunbiologische Veränderungen im Sinne einer allergischen   Reaktionsbereitschaft   gegen- über den oben beschriebenen mikrobiellen organspezifischen Stoffwechselprodukten hervorrufen.

   Die Beweisführung für die Richtigkeit dieser Zusammenhänge lässt sich nun wie folgt erbringen : Weder durch ein Kulturfiltrat der hämolytische Streptokokken allein oder nach Zusatz von totem Tonsillengewebe zum Nährboden noch durch supravitalen Tonsillenpresssaft aus nicht mit hämolytische Streptokokken in Kontakt gekommenen Tonsillen allein lässt sich eine allergische Gewebsreaktion auslösen. Diese erfolgt aber prompt, wenn die in maximaler Vermehrung begriffenen hämolytischen Streptokokken mit   supravitalem   Organsubstrat aus Tonsillengewebe für kurze Zeit in Kontakt gebracht wurden und das keimfreie Filtrat dieses Gemisches intrakutan oder subkutan injiziert wird. 



   Anderseits tritt aber eine derart spezifische Organsensibilisierung nicht auf, wenn sich die nach ihrem serologischen Typ und kulturellen Verhalten völlig gleichen hämolytische Streptokokken z. B. im Un-

   terhautgewebe vermehren und eine Phlegmone bilden. Mithin sind die durch die Einwirkung von lebenden organaffinen hämolytische Streptokokken auf lebendes Tonsillengewebe entstehenden mikrobiellen Stoffwechselprodukte als organspezifisch anzusehen. 



  Es wurde nun gefunden, dass derartige mikrobielle Stoffwechselprodukte geeignet sind, in kleinen Dosen intrakutan oder subkutan verabreicht, eine secundäre Organsensibilisierung zu verhindern oder ein bereits krankheitsbedingt geschädigtes Organ zu desensibilisieren. Ähnliche Wechselbeziehungen bestehen in mannigfacher Weise zwischen einer ganzen Reihe pathogener und apathogener Mikroorganismen und einzelnen Organen oder differenzierten Geweben. Klinisch äussern sich diese Wechselbeziehungen meist als chronisch rezidivierende Erkrankungen wie z. B. primär chronische Polyarthritis, chronisch rezidivierende Nephritis, Hepatitis, Myokarditis, chronisch rezidivierende Bronchitis und Sinusitis, Infektanaemie usw. 



  Besonders interessant ist jene Art der Organaffinität bestimmter Mikroorganismen zu pathologisch verändertem Gewebe wie z. B. die Affinität von Bact. prodigiosum und bestimmten Stämmen von Bact. 



  Coli zu Tumorgewebe. Es ist bekannt, dass z. B. Bact. prodigiosum auf maligne entartete Zellen einen nekrobiotischen Effekt besitzt, nicht aber gegenüber gesunden Zellen. 



  Nach den günstigen Erfahrungen mit andern erfindungsgemäss hergestellten organ-bzw. gewebsspezifischen mikrobiellen Stoffwechselprodukten erscheint es aussichtsreich, den Versuch zu unternehmen, die Abwehr des Organismus gegen maligne entartete Zellen dadurch zu steigern, dass man gewebsspezifische mikrobielle Stoffwechselprodukte verabreicht. Dabei wird man zweckmässigerweise so vorgehen, dass man gesundes differenziertes Gewebe zunächst in vivo oder in vitro solchen chemischen oder physikalischen Einwirkungen aussetzt, die erfahrungsgemäss zu einer malignen Entartung des Gewebes führen und anschliessend zur Gewinnung gewebsspezifischer mikrobieller Stoffwechselprodukte erfindungsgemäss das so vorbereitete Gewebe oder supravitale Gewebssubstrate mit in maximaler Vermehrung begriffenen Reinkulturen entsprechender gewebsaffiner Mikroorganismen zusammenbringt. 



  Um die Erfindung praktisch zu erläutern, seien folgende Beispiele gegeben, die jedoch die Erfindung nicht begrenzen sollen. 



  Beispiel l : Fakultative Saprophyten der Respirationsschleimhaut, wie z. B. vergrünende Streptokokken, Micrococcus catarrhalis, Staphylokokken, diphtheroide Stäbchen. usw. werden in Reinkulturen auf geeigneten üblichen Nährböden zu maximaler Vermehrung gebracht. Einem jungen gesunden Schlachttier werden unmittelbar nach der Tötung die Lungen unter sterilen Kautelen entnommen, die Respirationsschleimhaut abpräpariert und im sterilen Starmix zerkleinert. Zu einzelnen Portionen dieses supravitalen Zellbreies werden jeweils die oben beschriebenen Reinkulturen zugegeben und das Gemisch noch kurze Zeit bei Körpertemperatur gehalten. Anschliessend erfolgt Tiefkühlung, physikalische und fermentative Aufschliessung, Zentrifugation und keimfreie Filtration der überstehenden Flüssigkeit. 



  Die Abtrennung einzelner Fraktionen kann durch Elektrophorese, Elektrochromatographie, chemische oder physikalische Fällung erfolgen. 



  Zur Erhöhung der Haltbarkeit kann Vacuumtrocknung bei niedriger Temperatur, oder Adsorption an geeigneten Substanzen wie z. B. Aluminiumhydroxyd erfolgen. 



  Beispiel 2 : Einem jungen gesunden Schlachttier werden in maximaler Vermehrung begriffene Kulturen von Mikroorganismen mit Affinität zum Herzmuskel z. B. fakultativ saprophytäre pleomorphe Streptokokken vom serologischen Typ D intravenös injiziert. Nach einer gewissen Inkubationszeit wird das

 nert, erneut eine Kultur des ursprünglich zur Sensibilisierung verwendeten Bakterienstammes zugegeben und unter Durchspülung mit Sauerstoff kurze Zeit bei Körpertemperatur gehalten. Die weitere Aufbereitung erfolgt ähnlich wie unter Beispiel   l.   



   Beispiel 3 : Ein junges Tier oder Gewebekulturen aus menschlichen oder tierischen Organen werden in grösseren Zeitabständen radioaktiver Strahlung ausgesetzt oder mit   carcinogeen   Stoffen behandelt. 



  Die so vorbereiteten Organ- bzw. Gewebssubstrate werden unter sterilen Kautelen entnommen, im   steri- i   len Starmix fein. zerkleinert und mit organaffinen Mikroorganismen, die sich in maximaler Vermehrung befinden, zusammengebracht. Die weitere Aufbereitung ist ähnlich der wie Beispiel 1 und 2. 



   Sinngemäss können auf diese Weise durch Einwirkung von organaffinen Mikroorganismen auf supra-   vitale Organsubstrate   auch anderer differenzierter Organe und Gewebe spezifische mirkobielle Stoffwechselprodukte im Sinne der Erfindung gewonnen werden.

Verfahren zur Herstellung mikrobieller Stoffwechselprodukte durch Züchtung von organaffinen Mikroorganismen auf sterilen, Organextrakte enthaltenden Nährböden und Abtrennen der Mikroorganismen von den Stoffwechselprodukten, dadurch gekennzeichnet, dass die verwendeten Mikroorganismen zunächst auf geeigneten Nährböden zur maximalen Vermehrung gebracht und erst dann mit den entsprechenden vitalen Organsubstraten für kurze Zeit, vorzugsweise 2 bis 3 h bebrütet werden, worauf nach physikalischer oder fermentativer Aufschliessung der flüssige Anteil durch keimfreie Filtration abgesondert wird, worauf die Weiterverarbeitung, gegebenenfalls unter Aufspaltung in einzelne Fraktionen, in üblicher Weise erfolgt.
