   Verfahren zur Herstellung von neuen   Q'-Hydrazino-indolyl- (3)-carbonsäurederivaten   
Die Erfindung betrifft ein Verfahren zur Herstellung neuer   K-Hydrazino-indolyl- (3)-carbonsäurederi-   vate der allgemeinen Formel :

 in der   R., R.   und   R   Wasserstoffatome oder niedere Alkylgruppen,   R4   und   R6   Wasserstoffatome oder niedere aliphatische Acylreste und   R   ein Wasserstoffatom, einen niederen Alkylrest, eine Aralkylgruppe oder einen niederen aliphatischen Acylrest bedeuten können, sowie der nichttoxischen Salze dieser Verbindungen.

     a-Methyl- ss-3, 4-diliydroxyphenylalanin   inhibiert   Säugetier-Decarboxylase, hemmt   die Ansammlung von Norepinephrin im Herzen und Gehirn und hat sich als starkes Antihypertonicum erwiesen. 



   Es wurde nun gefunden, dass   a-Hydrazino-ss- (5-hydroxy-3-indolyl) -alkansäuren   sowie deren Ester und Acylderivate eine neue Klasse von Verbindungen darstellen, die ebenfalls starke Inhibitoren für Säugetier-Decarboxylase sind. Diese neuen Verbindungen sind ausserdem als Zwischenprodukte bei der Herstellung anderer Verbindungen und als Reagenzien für die Trennung von Ketonverbindungen von NichtKetonverbindungen verwertbar. 



   Als Zwischenprodukte können die neuen Verbindungen verwendet werden, um eine Klasse von Verbindungen herzustellen, die in ihrer Struktur dem Serotonin ähneln, sich von diesem jedoch dadurch unterscheiden, dass sie nicht Amine, sondern Hydrazine sind. Dies erfolgt durch enzymatische Decarboxylierung der erfindungsgemäss hergestellten Verbindungen, z. B. unter Verwendung von Säugetier-Decarboxylase, die durch Eindampfen eines wässerigen Extraktes von gemahlenen Rinder- oder Schweinenieren oder gemahlener Rinder- oder Schweineleber erhältlich ist. Die Hydrazinosäure wird in einem auf einen pH-Wert von 6, 8 gepufferten wässerigen Medium mit dem Enzym und dem Coenzym Pyridoxalphosphat auf 370C erwärmt, worauf man das entsprechende Hydrazin erhält. 



   Weiterhin können sowohl die auf diese Weise decarboxylierten Hydrazine als auch die Hydrazinosäuren selbst als Zwischenprodukte verwendet werden, indem man sie in dem gewöhnlich mit Phenylhydrazin durchgeführten Verfahren mit Acylessigsäureestern kondensiert und das Pyrazolonprodukt alkyliert. 



  Hiedurch erhält man Verbindungen, die in ihrer Struktur den analgetischen und antineuralgischen Mitteln von der Art des 1-Phenyl-2,3-dimethyl-pyrazolons-(5) ähneln. Die erfindungsgemäss hergestellten Hydrazinosäuren können also verwendet werden, um pyrazolonartige Verbindungen herzustellen, die als zusätzlichen Substituenten eine Carboxylfunktion aufweisen. 



   Die neuen Verbindungen eignen sich ferner zur Trennung von Ketonprodukten von nicht ketonartigen Produkten in ähnlicher Weise, wie diese Trennung mit dem Girardschen Reagenz durchgeführt wird. Da

 die Verbindungen Hydrazine sind, können sie mit Carbonylverbindungen zu Hydrazonen umgesetzt werden. Da sie ausserdem Carbonsäuren sind, können die so erhaltenen Hydrazone in wässerigem Alkali gelöst und die Carbonylverbindungen mit Säure regeneriert werden. Die Verbindungen unterscheiden sich also von den Girardschen Reagenzien P und T, welche durch quaternäre Ammoniumgruppen substituierte Hydrazine sind. 



   Die neuen Verbindungen werden erfindungsgemäss nach dem in der nachfolgenden Reaktionsfolge II

 den   N-Alkylderivaten   desselben hergestellt werden. Bei dieser Herstellung der Keton- und Aldehydausgangsverbindungen wendet man das Verfahren von Noland und Lange (Journal of the American Chemical Society, 81   [1959],   S. 1203) an, um eine ss-Nitroalkylgruppe in die 3-Stellung des Benzyloxyindols einzuführen. Dann wird das Nefsche Abbauverfahren (Nef, Annalen, 280   [1894], S. 263 ; Hass und Riley,   Chemical Review, 32 [1943],   S : 398)   angewendet, um die Nitrogruppe durch eine Carbonylgruppe zu ersetzen.

   Die Carboalkoxyester der obigen Verbindungen können leicht durch Veresterung der Hydrazinosäure mit einem niederen Alkanol in Gegenwart einer starken Säure, wie Schwefelsäure, Salzsäure, Bromwasserstoffsäure od. dgl., hergestellt werden. 



   Die 0, N, N-Acylester (d. h. die 5-Acyloxy-l-acyl-3-indolyl-verbindungen) werden leicht durch Erhitzen der Aminosäure mit dem entsprechenden aliphatischen Carbonsäureanhydrid in Gegenwart eines Protonenakzeptors, wie Pyridin, Picolin oder Dimethylanilin, gewonnen. Für diesen Zweck verwendbare niedere aliphatische Carbonsäureanhydride sind z. B. Essigsäureanhydrid, Propionsäureanhydrid, Buttersäureanhydrid u. dgl. Die Formylierung erfolgt mit Ameisensäure-Essigsäureanhydrid. 



   Die Ester beider Arten sind besonders wertvoll, weil sie im Körper viel besser absorbiert werden und man mit kleineren und weniger häufigen Dosen eine viel längere Wirkung erzielt. Besonders die Carbalkoxyester können in Form ihrer nichttoxischen Salze, wie der Hydrobromide, Hydrochloride, Sulfate u. dgl., verwendet werden. Diese Salze sind wasserlöslich und bilden sich bei der Herstellung der Ester. 



  Wenn der freie Hydrazinoester hergestellt werden soll, wird das Salz alkalisch gemacht. Auch der Ester selbst kann therapeutisch verwendet werden. 



   Die   erfindungsgemäss   erhältlichen Verbindungen können mit einem Träger kombiniert werden, der entweder ein fester Stoff oder eine sterile, parenteral applizierbare Flüssigkeit sein kann. Die Mittel können die Form von Tabletten, Pulvern, Kapseln oder andern Dosierungsformen besitzen, die sich besonders für die orale Darreichung eignen. Flüssige Verdünnungsmittel werden in sterilem Zustande für die parenterale Verabfolgung angewendet. Im allgemeinen werden die freien Säuren in Dosen von 10 bis 350 mg/ kg täglich dargereicht. Vorzugsweise werden sie oral   inDosen   im Bereich von 50 bis 150 mg/kg täglich dargereicht, vorzugsweise in häufigen kleinen Gaben, obwohl die   Verabfe'. gung   auch nur zweimal am Tag zu erfolgen braucht.

Ein Gemisch aus 923 g   1- (5-Benzyloxy-3-indolyl)-aceton, 1, 85 l   Benzol und 1 kg Natriumbisulfit in 200 1 Wasser wird mehrere Stunden bei Raumtemperatur gerührt. Das Produkt wird dreimal mit je 250 ml Wasser und dreimal mit je   230ml Äther   gewaschen, dann an der Luft und schliesslich im Vakuum bei Raumtemperatur getrocknet.

   Man erhält   5-Benzyloxy-3- (2-hydrazi-     no -2 -cyanpropyl) -indol.

   50 ml konzentrierte Salzsäure werden bei -100C mit gasförmigem Chlorwasserstoff gesättigt. Dann wird die Lösung langsam* unter starkem Rühren mit 3, 6 g des Produktes gemäss Beispiel la versetzt. Das Gemisch wird über Nacht gerührt, wobei es allmählich Raumtemperatur annimmt. Dann wird es im Vakuum zu einem Sirup eingeengt. Der Sirup wird mit 100 ml piger Bromwasserstoffsäure versetzt.    Ein Gemisch aus 53, 6 g   5-Benzyloxy-3- (2-nitropropyl)-indol   und 300 ml wässeriger Natronlauge, die 16 g NaOH enthalten, wird unter starkem Rühren zu einem eiskalten Gemisch aus 50 ml konzentrierter Schwefelsäure und 320 ml Wasser zugetropft. Dann wird das Gemisch mit weiterer Natronlauge alkalisch gemacht und mehrmals mit Chloroform extrahiert. Die Chloroformextrakte werden getrocknet und unter vermindertem Druck zu einem Rückstand aus 1- (5-Benzyloxy-3-indolyl)-aceton eingedampft. Das Rohprodukt wird von restlichen Mengen der-Nitroverbindung befreit.

     α-Methyl-ss-3, 4-di-hydroxyphenylalanine inhibits mammalian decarboxylase, inhibits the accumulation of norepinephrine in the heart and brain, and has been shown to be a potent antihypertonic.



   It has now been found that α-hydrazino-ss- (5-hydroxy-3-indolyl) alkanoic acids and their esters and acyl derivatives represent a new class of compounds which are also strong inhibitors of mammalian decarboxylase. These new compounds can also be used as intermediates in the preparation of other compounds and as reagents for the separation of ketone compounds from non-ketone compounds.



   As intermediates, the new compounds can be used to prepare a class of compounds that are similar in structure to serotonin, but differ from it in that they are not amines but hydrazines.
